背景技术
某些金属的抗微生物特性在相当长的一段时间内是已知的。这种独特的性质已经为包括农业和医疗保健在内的各个行业所利用,以试图控制感染和污染。

对于在医疗设备中防止感染和/或生物膜的建立中使用抗微生物材料的兴趣尤其突出。一个特别的挑战是预防因在相当长的一段时间内持续使用与皮肤或其他粘膜表面长时间接触的医疗设备而导致的感染,例如使用导液管、鼻胃管等。栖息在皮肤上的通常无害的细菌和其他微生物可以在这些医疗设备上生长,达到可能导致患者感染的程度。这对于接受长期护理和/或老年的患者来说尤其危险。

术语“抗微生物材料”是指具有抗微生物性质例如杀生物的或生物抑制(biostatic)性质的材料。在本发明的上下文中,术语“杀生物的”应理解为意指可以破坏、阻止、使其无害或对病原生物施加控制作用的物质,而术语“生物抑制”是指可以抑制生物体例如微生物的生长或繁殖。预期本发明将可用于对抗任何微生物,例如任何细菌、病毒和/或真菌。特别地,可预期葡萄球菌属(Staphylococcus)和克雷伯菌属(Klebsiella)中的细菌、念珠菌属(Candida)中的真菌以及冠状病毒(Coronaviridae)科的成员将对本发明记载的材料敏感。
本发明提供了具有令人惊讶的高的抗微生物活性的材料。包含本发明材料的产品,例如医疗设备,将降低败血症和感染的发生率。本发明特别适用于预防与常用医疗设备相关的感染,例如导液管、鼻胃管、套管和/或腹腔镜管,其中微生物可以通过医疗设备从外部环境传播到受试者并引起感染,或存在于皮肤或其他粘膜表面上的微生物可以定植在设备上并导致形成生物膜。这种预防在长时间使用医疗设备的情况下(因此会增加受试者发生感染的风险)尤其重要。
使用“基底”,我们意指可以将金属组分掺入到其中的任何合适的结构材料,从而提供可以将金属组分布局于其上或其中的物理介质。本发明的基底包含橡胶组分。可用于本发明的合适的橡胶品种的实例包括天然橡胶(胶乳)、氯丁橡胶、硅酮橡胶和丁腈橡胶。
优选地,橡胶组分是硅酮橡胶。硅酮,也称为聚硅氧烷,是包含由硅氧烷重复单元组成的合成化合物的任何聚合物。硅氧烷是由交替的硅原子和氧原子组成的链,与碳、氢结合,有时与其他元素结合。由于许多有利的特性,硅硅酮橡胶的使用是令人满意的;一般无反应性,稳定性高,对极端环境的抵抗力强,例如极热和极冷(例如,-55摄氏度至300摄氏度)。设想用于本发明的硅氧烷类型是液体硅酮橡胶(LSR)和高稠度硅酮橡胶(HCR或口香糖原料)。本领域技术人员将认识到要选择的硅酮橡胶类型取决于制造过程和最终应用。例如,在寻求制造具有复杂形状或薄壁的医疗设备时,使用HCR可能更合适。
可以形成基于聚合物的材料基底的基础的合适聚合物的实例包括合成的聚合物聚氨酯和聚丙烯,以及天然存在的基质聚合物胶原。基底可以优选基于聚合物的水凝胶。在水凝胶中使用的聚合物可以是本发明公开的任何聚合物。术语“基于聚合物的水凝胶”是指被水大量溶胀的聚合物网络。“基于聚合物的水凝胶”的实例,其可以用于本发明中,包括水凝胶和Nanorestore
使用“化学键合”,我们意指由于离子键、共价键或金属键而在铜和锌的原子、离子或分子之间产生的任何持久的吸引力。因此,这可以包括铜合金或铜化合物,包括但不限于铜盐和氧化物。
优选地,抗微生物材料的金属组分包含铜-锌合金。合金应理解为两种元素的混合物,其中一种是金属。在这种情况下,铜-锌合金应理解为一种替代合金,其中两种组分的原子可以在同一晶体结构内相互替换,从而产生大量的离域电子。
本领域技术人员会认识到,为了制备所需的合金,元素铜和锌在固化为新的独特化学实体之前将以它们的熔融形式混合在一起。在一个实施方案中,设想另外的金属及其化合物,例如金属的盐,可以掺入到该材料或金属组分中。这些金属包括但不限于锆、铜、锌、银、金、钯、铂、铱、铝、镍、钨、钼、钽、钛、碘。应当理解,所述后者化合物可以是所要求保护的有利于进一步增强材料的抗微生物性质的材料的附加组分。
与纯的金属或相关化合物的形式相对,与使用纯铜相比,使用合金产生了许多有利的性质。具体地,铜锌合金得益于锌的超强抗微生物性质,优异的延展性/可铸性和高的强度。
可预期金属组分的颗粒尺寸为10-80μm,优选尺寸为15-30μm内的任意值。与粗的粉末相比,精细研磨的粉末会释放更多的离子,抗微生物作用可归因于这些被释放的离子。
设想金属组分将包含至少60%的铜。该制剂具有增强的抗微生物性质。优选地,金属组分包含75-80%的铜和相应量的20-25%的锌。如上所述,金属组分可以额外地包含其他元素、其化合物以及盐。这些添加物可以赋予所要求保护的材料有益的性质。例如,添加的组分可以进一步增强抗微生物作用或允许所要求保护的产品的寿命延长。
在本发明的一个实施例中,金属组分和/或橡胶可以散布(interspersed)在整个基底中。使用“散布”,我们意指金属散落在基质材料的颗粒/分子之间。或者可以将这种结构描述为“浸渍”。金属成分可以均匀地或不均匀地分散(dispersion)在整个基底材料中。本领域技术人员会理解,散布、分散和/或浸渍的程度可以取决于在制造基底材料中使用的聚合物类型和/或将金属组分和/或橡胶施加到基底的方法。在本发明的另一个实施方案中,金属组分和/或橡胶可以作为涂层存在于基底表面上。在存在涂层的情况下,预期涂层将被布置为使得在使用中它与潜在污染的表面/伤口接触以发挥其抗微生物作用。涂层可以是任何厚度。另外,涂层应理解为存在于基底的至少一个表面上,也可以存在于所有基底表面上。涂层可以部分覆盖或完全覆盖基底的特定的表面。涂层的覆盖程度将取决于所要求保护的产品的预期用途。
设想基底可以是基于聚合物的基底。聚合物是由较小的重复亚单元组成的大分子。优选地,用于本发明的基底可以包括聚氨酯、聚丙烯和/或基于胶原的聚合物。根据本公开,该基材可以包括基于聚合物的水凝胶或基于聚合物的水胶体。热固性和热塑性聚氨酯均可适用于本发明。但是,可以设想,适合于稳定地保持金属组分和/或橡胶的任何材料都可以单独使用或组合使用以作为本发明的基底。例如,如果合适,如羊毛、棉、皮革、亚麻、苎麻、丝绸、大麻、竹子、黄麻、人造丝、氯丁橡胶、弹性纤维、橡胶、聚酯这些材料都可以适合用作基底。在某些情况下,应当理解,基底可以是不同类型的聚合物的组合。这样的组合可以在基底上赋予其他的有利的性质以实现所期望的目的或便于制造和存储。具体地,设想可以将海藻酸盐和纤维素掺入基底中以增强吸收性、柔韧性和舒适性。技术人员会认识到,基于聚合物的水凝胶由于存在亲水性官能团而特别有利地用于伤口敷料中。该功能可以控制特定表面的水分。
优选地,制造后的基底可以包含以下成分:
a)羧甲基纤维素钠
b)表面活性剂
c)甘油
d)柠檬酸
e)选定的聚合物,以及
f)选定形式的橡胶(优选硅酮橡胶)
合适的表面活性剂的实例包括硬脂酸钠、磺基琥珀酸二辛酯钠盐和全氟辛烷磺酸盐。合适的表面活性剂可以属于以下任何组:阴离子、阳离子、非离子或两性离子表面活性剂。如果需要,柠檬酸部分可以被其他弱酸例如乙酸、乳酸和磷酸代替。以上列表的部分e)可以被上述任何聚合物替代。优选地,所选择的聚合物被单独使用;然而,如果认为最终的抗微生物试剂是更有效的并且仍占基底组合物的68.5%,则可以组合使用不同的聚合物。
优选地,3-15重量%的基底由金属组分组成。例如3-5%、3-6%、3-7%、3-8%、3-9%、3-10%、3-11%、3-12%、3-13%、3-14%、5-6%、5-7%、5-8%、5-9%、5-10%、5-11%、5-12%、5-13%、5-14%、10-11%、10-12%、10-13%、10-14%。
设想如果需要,在材料中包含其他添加物以改善抗微生物性质。这些添加物可以包括螯合剂、硫酸镁和/或铜肽。这些添加物可以以0.1至1重量%,例如约0.5重量%的量掺入到基底中。术语“螯合剂”用于描述可以与单个金属离子形成多个键从而形成更稳定的络合物的物质。技术人员会认识到此类物质的作用可以增强抗微生物性质。
当与任何被污染的表面接触时,本发明可为有效的。在优选的实施方案中,本发明可以掺入到医疗设备中。在另一个优选的实施方案中,本发明可以掺入到导液管、鼻胃管、套管或腹腔镜管中。
通过术语“导管”、“鼻胃管”、“套管”和“腹腔镜管”,我们意指可插入受试者(人或动物)以治疗疾病、进食、进行外科手术的管,例如,腹腔镜手术,或用于执行受试者目前无法独立管理的生理功能,例如,排空膀胱中的尿液。实例包括但不限于导尿管、积液引流(脓肿)、从肺部周围引流空气/液体、静脉输液给药(血液、药物、营养)、体液收集(例如,血液样本、脑脊液样本)、用于血管成形术和血管造影、在硬膜外或蛛网膜下腔给药(例如麻醉剂)、气管导管和脐带。
本领域技术人员将理解管的特性(例如柔韧性程度)将取决于预期的应用。
本发明提供了高水平的抗微生物活性,因此具有广泛的应用。本发明包括包含本发明的抗微生物材料的感染控制产品。这种产品可能在医疗保健环境中具有实用性,最常见的是作为医疗材料。“感染控制产品”是指任何治疗、预防或减轻感染发展和/或传播的产品。此类产品的实例包括伤口敷料、绷带、医疗设备、药物容器和用于感染保护的个人防护服。
在第二方面,本发明提供了一种制造包含基底和金属组分的抗微生物材料的方法,其中所述金属组分包含化学键合的铜和锌且基底包含橡胶。该方法包含以下步骤:a)将铜和锌结合以制备所述金属组分;b)将金属组分加热到熔融状态;c)用高速气体碎裂所述熔融状态,和d)将已被碎裂的金属组分与基底结合,其中所述基底包含如本文所述的橡胶组分。优选地,橡胶组分是硅酮橡胶。
因此,一种生产本发明的金属组分的方法可以包括等离子体或气体雾化过程。可以设想,金属的粉末形式可以用在本发明的方法中,按照本领域技术人员所理解的其他的形式也可能是适合的。
可以设想,等离子体或气体雾化过程会产生粉末形式的金属组分,其可以在适当时与基底结合,如本领域技术人员所理解的那样。
在优选的实施方案中,在等离子或气体雾化过程开始之前,可任选地通过使用机械破碎过程来减小金属组分的尺寸。使用“机械破碎”,我们意指采用能够将金属组分逐渐分成更小的个体的任何过程。该过程可以通过使用许多的破碎设备来实现,这些设备包括但不限于:磨碎机、卧式磨、一维振动磨、3D振动磨和行星式磨。所有上述装置都会使尺寸减小,由于在研磨介质的撞击期间赋予了样品的能量。因此,在本发明的方法中,可以将金属形式的铜和锌研磨成我们所需要的合适形式。
一旦铜和锌结合在一起,就可以进行雾化过程。如本领域技术人员将会理解的,结合铜和锌的方式可以根据要使用的雾化过程而不同。
等离子体雾化要求金属组分为线状以用作原料。如本领域技术人员将会理解的,这通常是金属组分的合金线。与常规的气体雾化相反,等离子体雾化使用等离子炬在单个步骤中瞬间熔化和雾化线状物。然后使用冷却塔将形成的液滴转化为球形粉末。
或者,可以使用常规的气体雾化。这可以包括将铜-锌金属组分加热到约2000℃,以产生所述组分的熔融状态。使用“熔融状态”,我们意指所述金属组分在暴露于高温时的液体形式。如本领域技术人员将会理解的,高速气流可流过膨胀喷嘴,从输入室虹吸熔融金属组分。可用于此过程的气体的实例包括氮气、氩气、氦气或空气。技术人员将会认识到,可以在该过程中使用一种以上的气体,并且优选的气体或气体混合物将是惰性的/不反应的。所用气体的选择将取决于所需的最终碎裂的金属(粉末)的特性。为了提供合适的金属组分用于本发明的材料,可能需要高速的惰性气体。技术人员将会认识到,所需速度将根据所用的气体而有所不同,但是可能在100-2000m/s的范围内。该过程碎裂熔融金属的液流并且结果是产生细分散的颗粒,最终形成所需的金属组分的粉末形式。通过上述方法获得的粉末形式具有许多优点;高度球形颗粒的制备,低的氧含量以及对于生产铜和锌的适应性。技术人员还将会认识到,可以存在能够用于实现相同效果的生产金属粉末的替代性方法。
为了生产最终的抗微生物材料,将金属组分添加到基底中。具体地,金属粉末以少量的方式添加,直到将整个产物全部转移到基底。将所得的组合物在室温(20-22℃)下以350rpm混合2小时,随后使其固化。
本领域技术人员将了解可将橡胶组分引入本发明以制造符合所需规格的医疗设备的各种方法。例如,可以使用多种成型工艺。这包括液体注射成型、传递成型、压缩成型、微成型或二次成型。或者,可以采用挤压工艺。
本发明还提供了一种预防或治疗感染的方法,包括在医疗或兽医环境中使用本发明的抗微生物材料。
为了可以更清楚地理解本发明,现在将通过实施例的方式来描述本发明的实施方案。
实施例1
在两种不同的细菌菌株(金黄色葡萄球菌(Staphylococcus aureus)和肺炎克雷伯菌(Klebsiella pneumoniae))上进行抗微生物材料的测试
将每种测试生物体制备成约1x105菌落形成单位(CFU)/mL于0.85%NaCl中。对于每个样品,使用每种测试生物体进行5次平行接种。使用胰蛋白胨大豆琼脂(Tryptone SoyaAgar(TSA))的平板倾注法,在接种的点对接种物进行计数。将已经被接种的样品在24℃±1℃和>95%的湿度下保持24小时。在暴露时间之后,将已经被接种的试样在无菌下移至9ml稀释剂中。剧烈摇晃以确保任何残留的测试生物体完全地再悬浮。将得到的悬浮液置于TSALT(补充有0.3%大豆卵磷脂和3%Tween 80的TSA)中。将平板在31℃±1℃下培养至少5天。

对于样品“3%CuZn泡沫”和“15%CuZn泡沫”,可以看出在经历24小时的接触时间后这两种细菌菌株的数量均减少了>4log。将其与样品“0%CuZn泡沫”进行比较,后者对于这两种测试生物体均未显示出明显的抗微生物活性。
实施例2
在真菌白色念珠菌(Candida albicans)上进行抗微生物材料的测试
将测试生物体制备成约1x106 CFU/mL于0.85%NaCl中。对于每个样品,使用适当体积的测试生物体(表2)对于试样进行5次平行接种。使用沙氏葡萄糖琼脂(SabouraudDextrose Agar(SDA))的平板倾注法,在接种的点对接种物进行计数。然后将已经被接种的样品放置于24℃±1℃、>95%的湿度下的培养箱,时间持续1、8或24小时。在所需的暴露时间之后,将已经被接种的试样在无菌下移至9ml稀释剂中。剧烈摇晃以确保任何残留的测试生物体完全地再悬浮。将得到的悬浮液置于SDALT(补充有0.3%大豆卵磷脂和3%Tween 80的SDA)中。将平板在24℃±1℃下培养至少5天。对于阴性对照,样品用适当体积(表2)的无菌的0.85%NaCl接种,并以与测试样品相同的方式进行培育和分析。

对于样品“0%CuZn泡沫”,在24℃下1、8或24小时的接触时间之后,观察到白色念珠菌的数量没有明显减少。对于样品“2%CuZn泡沫”,在24℃下1小时的接触时间之后,观察到白色念珠菌的数量的减少大于3log;在24℃下8小时或24小时的接触时间之后,观察到白色念珠菌的数量的减少大于4log。对于样品“3%CuZn泡沫”,在24℃下1小时的接触时间之后,观察到白色念珠菌的数量没有明显减少;在24℃下8小时之后,观察到白色念珠菌的数量的减少大于3log;在24℃下24小时之后,观察到白色念珠菌的数量的减少大于5log。
实施例3:在牛冠状(bovine corona)病毒(BCV)毒株L9上进行抗微生物材料的测试
为了制备该材料,在无菌条件下切成1x1 cm的小块,并在折叠步骤后转移到Eppendorf杯中。为了制备测试病毒溶液,培养U373细胞。为了产生病毒,将BCV毒株L9加入已制备的单层中。在24-48小时的培育期后,通过快速的冷冻/融化循环来裂解细胞。去除细胞碎片,并将上清液直接用作测试病毒悬浮液。使用微量滴定法通过终点稀释滴定来测定感染性。通过计算滴定度的降低,与未经处理材料的杀病毒活性进行比较,以评估经过处理的材料的杀病毒活性。

在60分钟的接触时间后,在使用新的绿色/白色尼龙铜浸渍织物下,检测到仅一例材料中有残留病毒。相比之下,检测未经处理的材料,残留病毒可以在所有情况下被检测到。得出以下平均值:≤1.55±0.04(新的绿色/白色尼龙铜浸渍织物)和2.98±0.12(参考)。可见,在60分钟暴露时间后,基于10倍测定,两种材料之间的差异为1.43log10的差距。
Claims (24)
1.一种抗微生物材料,其包含基底和金属组分,其中金属组分包含化学键合的铜和锌,且其中基底包含橡胶组分。
2.根据权利要求1中所述的抗微生物材料,其中橡胶是硅酮橡胶。
3.根据权利要求1或2中所述的抗微生物材料,其中铜和锌形成合金。
4.根据权利要求1、2或3中所述的抗微生物材料,其中金属组分还包括选自以下的任何金属:锆、银、金、钯、铂、铱、铝、镍、钨、钼、钽、钛、碘和/或其任何合金。
5.根据权利要求1-4中所述的抗微生物材料,其中金属组分还包括锆、铜、锌、银、金、钯、铂、铱、铝、镍、钨、钼、钽、钛和/或碘中任一的一种或多种的盐。
6.根据权利要求1-5中任一项所述的抗微生物材料,其中金属组分包含尺寸为3-50μm,优选15-30μm的颗粒。
7.根据权利要求1-6中任一项所述的抗微生物材料,其中金属组分包含至少60%的铜。
8.根据权利要求1-7中任一项所述的抗微生物材料,其中金属组分包含75-80%的铜。
9.根据权利要求1-8中任一项所述的抗微生物材料,其中金属组分包含20-25%的锌。
10.根据权利要求1-9中任一项所述的抗微生物材料,其中金属组分散布在整个基底中,和/或其中金属组分在所述基底的表面上形成涂层。

16.一种医疗设备,其包含权利要求1-15中任一项所述的抗微生物材料或由其形成。
17.根据权利要求16中所述的医疗设备,其中所述医疗设备为导液管、鼻胃管、套管或腹腔镜管。
18.一种包含基底和金属组分的抗微生物材料的制备方法,其中金属组分包含化学键合的铜和锌,且其中基底包含橡胶组分,
该方法包含以下步骤:
a)将铜和锌结合以制备所述金属组分;
b)将金属组分加热到熔融状态;
c)用高速气体碎裂所述熔融状态,以及;
d)将被碎裂的金属组分与基底结合。
19.根据权利要求18中所述的方法,其中橡胶组分是硅酮橡胶。
20.根据权利要求18或19中所述的方法,其中在步骤(b)之前使用机械破碎方法来减小金属组分的尺寸。
21.根据权利要求18、19或20中所述的方法,其中步骤(c)产生所述金属组分的粉末形式。

24.一种预防或治疗伤口感染的方法,其包含在医疗或兽医环境中使用权利要求1-15中任一项所述的抗微生物材料或权利要求16或17中所述的医疗设备。
