本発明は、以下の3つの鍵となるタンパク質要素を含む新規治療用融合タンパク質である:(1)Treg細胞に対し選択性が高くなるように改変されている改変型IL-2サイトカイン、(2)該タンパク質の循環半減期を増加するエフェクター機能欠損型Fcタンパク質、および(3)融合タンパク質の高い生物学的活性に必要である、2つの部分の間のペプチドリンカー。該融合タンパク質は、IL-2ドメインがIL-2ドメインのC末端によりリンカーペプチドのN末端に結合されるように構成される。FcドメインはそのN末端によりリンカーのC末端に結合される。短いペプチドリンカーを用いるおよび用いない従来研究では、n-IL-2:c-Fc融合タンパク質は著しい生物活性を有しないことが報告された。このことによって、Fc領域がリンカーのN末端に結合され、IL-2ドメインが融合タンパク質のカルボキシ末端を形成する、逆の構成であるn-Fc:c-IL-2が注目されることとなった。
本明細書で使用される「配列番号1に対する少なくとも○パーセント(例えば、90または95または97%)の配列同一性」とは、2つ以上の核酸またはポリペプチドの配列が同一である程度を指す。評価窓にわたる、例えば目的の配列の長さにわたる、目的の配列と第2の配列の間のパーセント同一性は、配列を整列し、同一性を最大化するためにギャップの導入を許容しながら、同一残基の反対側ににある、評価の窓の中にある残基(ヌクレオチドまたはアミノ酸)の数を決定し、窓の中に位置する目的の配列または第2の配列の残基の合計数(いずれかより大きな方)で割り、さらに100を掛けることによって計算することができる。特定のパーセント同一性を達成するのに必要な同一な残基の数を計算する際には、割合を最も近い全体数に丸めることになる。パーセント同一性は、様々なコンピュータープログラムを使用して計算することができる。例えば、BLAST2、BLASTN、BLASTP、Gapped BLAST等のコンピュータープログラムは、アラインメントを生成し、目的の配列間のパーセント同一性を提供する。Karlin and Altschul, Proc. Natl. Acad. ScL USA 90:5873-5877, 1993において改変されたKarlin and Altschulのアルゴリズム(Karlin and Altschul, Proc. Natl. Acad. ScL USA 87:22264-2268, 1990)はAltschul et al.(Altschul, et al., J. Mol. Biol. 215:403-410, 1990)のNBLASTおよびXBLASTプログラムに組み込まれている。比較目的のためのギャップ有アラインメントを取得するために、Altschul et al.(Altschul, et al. Nucleic Acids Res. 25: 3389-3402, 1997)に記載されるようにGapped BLASTが使用される。BLASTおよびGapped BLASTプログラムを使用する際には、各プログラムの初期設定パラメーターを使用してもよい。PAM250またはBLOSUM62行列を使用してもよい。BLAST解析を行うためのソフトウェアはNational Center for Biotechnology Information(NCBI)を通じて公共に利用できる。これらのプログラムについては「ncbi.nlm.nih.gov」のURLワールドワイドウェブアドレスを有するウェブサイトを参照されたい。具体的な実施形態では、パーセント同一性はNCBIによって提供されるような初期設定パラメーターを用いてBLAST2を使用して計算される。
本発明のIL-2バリアントタンパク質は、IL-2αβγ選択的アゴニストである。機能的には、本発明のIL-2バリアントタンパク質は、IL2Rβγ受容体複合体に比べてIL2Rαβγ受容体複合体を選択的に活性化する。本発明のIL-2バリアントタンパク質は、野生型IL-2に由来し、配列番号1の野生型IL-2に対して少なくとも95%の配列同一性を有するものとして構造的に規定され、Treg細胞を優先的に活性化する能力によって機能的に規定される。該タンパク質はまた、CD4+ CD25-/low T細胞またはNK細胞に比べてTreg細胞におけるリン酸化STAT5タンパク質のレベルによって測定される、TregにおいてIL-2受容体シグナリングを選択的に活性化する能力によって、あるいはNK細胞に対するフィトヘマグルチニン刺激T細胞の選択的活性化によって機能的に規定することができる。
「Treg」または「Treg細胞」は、制御性T細胞を指す。制御性T細胞は、他の免疫細胞の活性を抑制するT細胞のクラスであり、細胞マーカー表現型CD4+CD25+FOXP3+によってフローサイトメトリーを使用して規定される。FOXP3は細胞内タンパク質であり、染色のためには細胞固定と透過処理を必要とするため、細胞表面表現型CD4+CD25+CD127-を、生きたTregを規定するために使用することができる。Tregはまた、様々なTregサブクラス、例えばtTreg(胸腺由来)およびpTreg(末梢由来で、末梢のナイーブT細胞から分化したもの)を含む。すべてのTregはIL-2Rαβγ受容体を発現し、それ自身のIL-2を産生せず、増殖はIL-2に依存していて、当業者であれば、両方のクラスがIL-2Rαβγ選択的アゴニストによって選択的に活性化されることを認識する。
「ペプチドリンカー」は、融合タンパク質を構成する2つのタンパク質の間に位置するアミノ酸配列であって、リンカーペプチド配列がいずれのパートナータンパク質にも由来しないものとして定義される。ペプチドリンカーは、正しいタンパク質フォールディングと構成要素タンパク質部分の安定性を促進し、タンパク質発現を改善し、2つの融合パートナーの生物活性をより良くすることを可能にするために、スペーサーとして融合タンパク質に組み入れられる(Chen, et al., 2013, Adv Drug Deliv Rev. 65(10):1357-69)。ペプチドリンカーは、非構造化可撓性ペプチドまたは硬い構造化ペプチドのカテゴリーに分けることができる。
「Fc融合タンパク質」は、新規の単一の組換えポリペプチドを生成するために、哺乳類IgGタンパク質のFcドメインの翻訳読み枠が別のタンパク質(「Fc融合パートナー」)の翻訳読み枠と融合した、組換えDNA技術によって作製されるタンパク質である。Fc融合タンパク質は、通常、ヒンジ領域に位置するジスルフィド結合によって一つに連結された、ジスルフィド連結二量体として生成される。
「生物活性」は、細胞に基づく定量的インビトロアッセイにおける生物学的活性の測定を指す。
Fc融合タンパク質の設計と構築には複数の選択肢があり、所望の生物学的活性と薬学的特性を有する分子の生成が可能になるように、これらの設計の選択肢からの選択が以下に提示される。鍵となる設計の選択肢は、以下の通りである:(1)IL2選択的アゴニストの性質、(2)Fcタンパク質部分の選択、(3)融合タンパク質における融合パートナーの構成、および(4)Fcと融合パートナータンパク質の間の接合部のアミノ酸配列。
一般的に、本発明の融合タンパク質の調製は、本明細書に開示される方法によって、および例えばポリメラーゼ連鎖増幅反応(PCR)、プラスミドDNAの調製、制限酵素を用いたDNAの切断、オリゴヌクレオチドの調製、DNAのライゲーション、mRNAの単離、適切な細胞へのDNAの導入、宿主の形質転換または遺伝子導入、宿主の培養等を含む認知された組換えDNA技術によって達成することができる。さらに、該融合分子は、カオトロピック試薬と周知の電気泳動の、遠心分離の、およびクロマトグラフィーの方法を使用して単離し精製することができる。これらの方法に関する開示については、Sambrook et al., Molecular Cloning: A Laboratory Manual 2nd ed.(1989);およびAusubel et al., Current Protocols in Molecular Biology, John Wiley & Sons, New York (1989)を一般的には参照されたい。
置換N88Rを有するIL-2は、IL2Rαβγ受容体に対するIL2選択的アゴニストの例示的なケースである(Shanafelt, A. B., 2000, Nat Biotechnol.18:1197-202)。IL2/N88Rは、IL2Rβ受容体サブユニットおよびIL2Rβγ受容体複合体への結合が欠損しているものの、野生型IL-2(wt IL-2)と同程度に効果的に、IL2Rαβγ受容体複合体に結合し、IL2Rαβγ発現PHA活性化T細胞の増殖を刺激することができるが、一方でIL2Rβγ発現NK細胞の増殖を刺激する3,000倍低下した能力を示す。同様の活性プロファイルを有する他のIL2Rαβγ選択的アゴニストは、置換N88GおよびD20Hを有するIL-2を含み、置換Q126LおよびQ126F(IL2RGサブユニットとの接触残基)を有する他のIL2バリアントもまたIL2Rαβγ選択的アゴニスト活性を有する(Cassell, D. J., et al., 2002, Curr Pharm Des., 8:2171-83)。当業者は、Fc融合タンパク質が同様の活性を有することを予想して、これらのIL2選択的アゴニスト分子のいずれについてもIL2/N88R部分に置換することができることを認識するだろう。前述した変異のすべてを、wt IL-2、または置換C125S(対のないシステイン残基を除去することによってIL-2安定性を促進する置換である)を有するwt IL-2の背景に導入することができる。本発明はまた、IL-2受容体活性化活性に著しく影響することなく、産生または安定性を改善する他の変異またはトランケートとともに使用することができる。

鍵となる設計上の選択肢は、Fcタンパク質部分の性質である。Fc融合タンパク質の主要な治療用途は、(1)融合パートナータンパク質に免疫グロブリンFcエフェクター機能を与えること;または(2)融合パートナータンパク質の循環半減期を増加させることである(Czajkowsky, et al., 2012, EMBO Mol Med. 4:1015-28)。IgGタンパク質の主要エフェクター機能は、補体依存性細胞傷害(CDC)および抗体依存性細胞傷害(ADCC)、すなわち補体タンパク質C1qへのおよびIgG-Fc受容体(FcγR)へのそれぞれのFc結合によって媒介される機能である。これらのエフェクター機能は、治療用タンパク質が特定の抗原標的または細胞に対する免疫反応を指向するまたは増強するために使用される場合に、重要である。本発明の融合タンパク質は、IL2選択的アゴニスト部分の循環半減期を増大させるためだけに設計され、エフェクター機能は必要なく、毒性を有することさえあり得るため、はっきり言えば望ましくない。例えば、エフェクター機能適格性Fcを有するIL2選択的アゴニスト-Fc融合タンパク質は、本発明の融合タンパク質が活性化し拡大しようとするTreg細胞を可能性として殺傷することができ、これは自己免疫疾患のための治療目標の真逆である。エフェクター機能(CDC、ADCC)、循環半減期、および安定性の点で異なる4つのヒトIgGサブクラスがある(Salfeld, J. G., 2007, Nature Biotechnology 25:1369-72)。IgG1は、Fcエフェクター機能を有し、最も多いIgGサブクラスであり、米国FDA承認治療用タンパク質の中で最も一般的に使用されている。IgG2は、Fcエフェクター機能を欠損しているが、他のIgG2分子との二量体化を受けやすく、またヒンジ領域のジスルフィド結合の組み換えのために不安定化しやすい。IgG3はFcエフェクター機能を有し、極めて長く硬いヒンジ領域を有する。IgG4は、Fcエフェクター機能を欠損しており、他のサブクラスよりも短い循環半減期を有しており、IgG4二量体は、異なるIgG4分子との間でH鎖の交換につながるヒンジ領域中の1つのみのジスルフィド結合により、生化学的に不安定である。当業者であれば、IgG2およびIgG4に由来するFcタンパク質部分はエフェクター機能を有しておらず、本発明に使用することができることを認識するだろう。当業者であればまた、IgG2 Fcのヒンジ領域は凝集を防ぐために改変することができ、またはIgG4 Fcのヒンジ領域は二量体を安定化させるために改変することができるように、Fc配列改変が当技術分野において記述されていることを認識するだろう。あるいは、IgG1のエフェクター機能欠損型バリアントが生成されている。一つのそのようなバリアントは、N結合型糖鎖付加部位の場所である、位置N297にアミノ酸置換を有する。このアスパラギン残基の置換は、糖鎖付加部位を除去し、ADCCおよびCDC活性を著しく低下させる(Tao, M. H., et al., 1989, J Immunol. 143:2595-2601)。このバリアントは本明細書における本発明の例示的なケースとして使用される。別のエフェクター機能欠損型IgG1バリアントはIgG1(L234F/L235E/P331S/)であり(Oganesyan, et al., 2008, Acta Crystallogr D Biol Crystallogr. 64:700-4)、これはC1qおよびFcγR結合部位におけるアミノ酸を変異するものであり、当業者であれば、エフェクター欠損型で安定なIL2SA-Fc融合タンパク質を生成するためにこれらのまたは類似したFcバリアントの使用を考慮するだろう。当業者であれば、二量体よりも安定な単量体となるように改変されたFcタンパク質部分の形態(Dumont, J. A., et al., 2006, BioDrugs 20:151-60;Liu Z, et al., J Biol Chem. 2015 20;290:7535-62)もまた本発明のIL-2選択的アゴニストと組み合わせられることを認識するだろう。さらに、当業者であれば、Fc H鎖ポリペプチドと組み合わせたIL-2-Fc H鎖ポリペプチドから構成され、二重特異性抗体技術を使用して構築される、機能的に単量体のヘテロ二量体(Zhu Z, et al., 1997 Protein Sci. 6:781-8)もまた、本発明のIL-2選択的アゴニストと組み合わせられることを認識するだろう。IgG部分における抗原特異性を有する(Penichet, M. L., et al.,1997, Hum Antibodies. 8:106-18)または有しない(Bell, et al., 2015, J Autoimmun. 56:66-80)いくつかのIL-2 Fc融合タンパク質が、完全IgG抗体分子を用いて作製された。その上、当業者であれば、ヒンジ領域の一部または全部を有しないFcバリアントを本発明と共に使用することができることを認識する。
Fcと融合パートナータンパク質との間の接合部のアミノ酸配列は、(1)2つのタンパク質配列の直接的な融合物であるか、(2)介在するリンカーペプチドを有する融合物のいずれであってもよい。米国FDAによって臨床使用のために現在承認されている10個のFc融合タンパク質(表1)のうち、8個は融合パートナータンパク質とFcとの直接融合物であり、一方2個はリンカーペプチドを有し、したがって多数のFc融合タンパク質がリンカーペプチドなしに機能的であることが可能である。リンカーペプチドは、2つのタンパク質部分の間のスペーサーとして含まれる。リンカーペプチドは、適切なタンパク質フォールディングと構成要素タンパク質部分の安定性を促進することができ、タンパク質の発現を改善し、構成要素タンパク質部分のより良好な生物活性を可能にする(Chen, et al., 2013, Adv Drug Deliv Rev. 65:1357-69)。多数の融合タンパク質に使用されるペプチドリンカーは、非構造化可撓性ペプチドとなるように設計される。天然タンパク質における独立した構造ドメインの間のリンカーペプチドの長さ、配列、および立体構造の研究により、可撓性ペプチドリンカーの設計のための理論的基礎が提供されている(Argos, 1990, J Mol Biol. 211:943-58)。Argosは、長い可撓性リンカーペプチドはグリシンのような小さい無極性残基、ならびにセリンおよびスレオニンのような小さい極性残基から構成され、複数のグリシン残基は可撓性の高い立体構造を可能にし、セリンまたはスレオニンは極性を有する表面領域を提供しペプチド内のまたは構成要素融合タンパク質部分との疎水性相互作用を制限するという指針を提供した。文献に記載される多くのペプチドリンカーでは、配列GGGGSの繰り返し等、グリシンとセリンが豊富であるが、当業者であれば、Argos(Argos, 1990, J Mol Biol. 20;211(4):943-58)の一般的な推奨に従う他の配列もまた使用することができることを認識する。例えば、本明細書に記載されるタンパク質のうちの1つは、グリシンおよびアラニンから構成されるリンカーペプチド(配列番号15)を含有する。完全に伸展したベータストランド立体構造を有する可撓性リンカーペプチドは、残基当たりの端から端までの長さが約3.5Åである。したがって、5、10、15、20、25または30残基のリンカーペプチドは、最大の完全に伸展した長さがそれぞれ17.5Å、35Å、52.5Å、70Å、87.5Åまたは105Åである。ペプチドリンカーの最大の端から端までの長さはまた、本発明におけるペプチドリンカーの性質を規定するための指針となり得る。
質は、ペプチドリンカーがなくても、または5残基のペプチドリンカーを有していても、生物活性が低下するかまたは生物活性が全くないことを報告した(Gavin, et al.,米国特許出願公開第20140286898号)。この特許中で報告されたこの仕事は、少なくとも6および好ましくは少なくとも9アミノ酸のペプチドリンカーが、Tregに対する頑健なIL-2生物活性に必要であることを示し、生物活性の改善は15アミノ酸でプラトーに達し、リンカーの長さが30アミノ酸になるまで維持されることをさらに示す。
候補タンパク質の生物学的活性の特性決定には、頑健な定量的なバイオアッセイが必要である。これらのアッセイは、IL2受容体の活性化を測定し、Tregにおける活性化の下流の機能的な結果を測定し、さらに活性化したTregの治療上関連する成績と機能を測定すべきである。これらのアッセイは、IL2選択的アゴニスト分子の治療活性と能力を測定するために使用することができ、また動物またはヒトにおけるIL2選択的アゴニストの薬物動態の測定のために使用することもできる。一つのアッセイは、シグナル伝達タンパク質STAT5のリン酸化を測定し、リン酸化タンパク質(pSTAT5)に特異的な抗体を用いてフローサイトメトリーを測定した。STAT5のリン酸化は、IL-2シグナル伝達経路における必須のステップである。STAT5はTreg発生に必須であり、CD4+CD25+細胞において発現されたSTAT5の構成的活性化形態は、IL-2の不在下でのTreg細胞の産生に十分である(Mahmud, S. A., et al., 2013, JAKSTAT 2:e23154)。したがって、Treg細胞におけるリン酸化STAT5(pSTAT5)の測定は、これらの細胞におけるIL-2活性化を反映するものとして当業者には認識され、適切な暴露時間と条件があれば、IL-2処理の他の生物学的結果を予測する。機能的活性化の別のアッセイは、Treg細胞のIL-2刺激性増殖を測定する。当業者であれば、Treg増殖は、精製されたTreg細胞へのトリチウム標識チミジンの取り込みによって;フローサイトメトリーにより測定される細胞の混合集団中のTreg細胞数、およびCD4+CD25+FOXP3+またはCD4+CD25+CD127-マーカー表現型の頻度の増加によって;Ki-67等の増殖関連細胞周期タンパク質のTreg細胞における発現の増加によって;またはTreg細胞におけるフローサイトメトリーによる、カルボキシフルオレセインスクシンイミジルエステル(CFSE)等の生体染色色素の細胞分裂関連希釈の測定によって、測定できることを認識する。IL-2によるTregの機能的活性化の別のアッセイは、Tregの増大した安定性である。pTreg細胞は不安定であると一部で考えられており、Th1およびTh17エフェクターT細胞に分化する能力を有する。TregのIL-2活性化は、Tregを安定化させ、この分化を防止することができる(Chen, Q., et al., 2011, J Immunol. 186:6329-37)。TregのIL-2刺激の別の結果は、Tregの免疫抑制活性に寄与する、CTLA4、GITR、LAG3、TIGIT、IL-10、CD39およびCD73等のTreg機能的エフェクター分子のレベルの刺激である。
本発明の融合タンパク質の医薬組成物は、従来方法に従う、非経口(特に静脈内または皮下)投与のために製剤化されるものとして規定される。一般的に、医薬製剤は、生理食塩水、緩衝食塩水、5%デキストロース水溶液等の薬学的に許容可能なビヒクルと組み合わせた、本発明の融合タンパク質を含む。製剤は、1つ以上の賦形剤、保存剤、可溶化剤、緩衝剤、バイアル表面でのタンパク質損失を防ぐためのアルブミン等をさらに含んでもよい。製剤方法は当技術分野において周知であり、例えば、Remington: The Science and Practice of Pharmacy, Gennaro, ed., Mack Publishing Co., Easton, Pa., 19.sup.th ed., 1995に開示されている。
本発明の融合タンパク質は、好ましくは非経口経路によって投与される。皮下経路は好ましい経路であるが、静脈内、筋内、および皮内投与もまた使用することができる。皮下または筋内経路、デポおよびデポ製剤を使用することができる。特定の疾患には、特別な投与経路を使用することができる。例えば、炎症性眼疾患には、眼球内注射を使用することができる。融合タンパク質は、0.01mcg/ml~100mcg/mlの範囲の濃度を使用してもよいが、全容量の約0.1~10mcg/mlの濃度で使用することができる。
本発明の治療法から利益を得ることができる疾患の一部は言及されている。しかし、自己免疫疾患におけるTreg細胞の役割は、非常に活発な研究分野であり、追加の疾患が本発明によって治療可能なものとして同定される可能性が高い。自己免疫疾患は、免疫系が自身のタンパク質、細胞および組織を攻撃するヒトの疾患として規定される。自己免疫疾患の網羅的列挙および概説は、The Autoimmune Diseases(Rose and Mackay, 2014, Academic Press)に見出すことができる。Treg増強から利益を受けることの証拠がある疾患は、移植片対宿主病、尋常性天疱瘡、全身性エリテマトーデス、強皮症、潰瘍性大腸炎、クローン病、乾癬、1型糖尿病、多発性硬化症、筋萎縮性側索硬化症、円形脱毛症、ブドウ膜炎、視神経脊髄炎およびデュシェンヌ型筋ジストロフィーを含む。
本発明におけるFcタンパク質部分の目的は循環半減期を増加させることのみであるため、当業者であれば、分子サイズを増加させ腎クリアランスの速度を減少させる同一の目標を達成するために、本発明で見出された構造-活性関係を使用して、IL-2選択的アゴニスト部分を他のタンパク質のN末端に融合することができることを認識する。IL2選択的アゴニストは血清アルブミンのN末端に融合することができ(Sleep, D., et al., 2013, Biochim Biophys Acta.1830:5526-34)、ここで血清アルブミンはIL-2部分に比べて融合タンパク質の流体力学半径を増加させ、且つFcRNによって再利用される。当業者であればまた、本発明のIL2選択的アゴニスト部分はまた組換え非免疫原性アミノ酸ポリマーのN末端に融合することができることを認識するだろう。この目的のために開発された非免疫原性アミノ酸ポリマーの2つの例は、A、E、G、P、SおよびTアミノ酸の鎖であるXTENポリマー(Schellenberger, V., et al., 2009, Nat Biotechnol. 27:1186-90)、およびP、AおよびSアミノ酸残基の鎖であるPASポリマー(Schlapschy, M., et al., 2007, Protein Eng Des Sel. 20:273-84)である。
N88RL9AG1(配列番号4)をコードするcDNAをDNA合成とPCR構築によって構築した。N88RL9AG1構築物は、マウスIgG1シグナル配列、置換N88RおよびC125Sを有する成熟ヒトIL-2(配列番号1)配列、9アミノ酸リンカーペプチド配列(配列番号15)、ならびに置換N297Aを含有するヒトIgG1のFc領域(配列番号2)から構成された。N88R/IL2は、IL2RBへの結合が低減していて、IL2Rαβγ受容体発現細胞に対する選択的アゴニスト活性を有するIL2選択的アゴニストである(Shanafelt, A. B., et al., 2000, Nat Biotechnol.18:1197-202)。IgG1 Fc上のN297におけるN結合型糖鎖付加部位の除去によって、Fcエフェクター機能は減少する(Tao, M. H., et al., 1989, J Immunol. 143:2595-2601)。D20HL0G2は、マウスIgG1シグナル配列、置換D20HおよびC125Sを有するIL-2(配列番号1)、ならびにヒトIgG2(配列番号3)に由来するFcタンパク質部分から構成された。D20H IL-2バリアントは、N88Rと同様の選択的アゴニスト活性を有することが報告されている(Cassell, D. J., et al., 2002, Curr Pharm Des., 8:2171-83)。
N88RL9AG1およびD20HL0G2が正しくフォールディングしたかどうかを決定するために、IL-2受容体サブユニットIL2RAおよびIL2RBへのその親和性をBiacore T-200装置(GE Healthcare)を使用して表面プラスモン共鳴(SPR)によって決定した。IL2RAおよびIL2RB細胞外ドメインタンパク質およびIL-2タンパク質(R&D Systems, Minneapolis, MN)を、それぞれ30および484の最終RU(共鳴単位)値まで、NHS/EDCカップリングによってCM-5 Biacoreチップ上に固定した。IL2RAへの結合キネティクスは、50μl/分の流速で、0.6nM~45nMの範囲の5種類の濃度のIL2およびN88RL9AG1で測定された。IL2RBへの結合キネティクスは、IL2については16.7nM~450nMの、Fc融合タンパク質については14nM~372nMの範囲の5種類の濃度で、10μl/分の流速で測定された。解離定数(Kd)は、IL-2については1:1フィット、N88RL9AG1およびD20HL0G2については2価フィットを仮定して、Biacore評価ソフトウェア、バージョン2.0を使用して、該速度定数から計算された。平衡Kd値は定常状態結合値を使用してBiacore評価ソフトウェアによって計算された。
T細胞に対するN88RL9AG1およびD20HL0G2の生物活性を、特定のT細胞サブセットにおけるリン酸化STAT5(pSTAT5)レベルを測定することによって決定した。pSTAT5のレベルは、リン酸化STAT5ペプチドに対する抗体を使用し、固定し透過処理した細胞においてフローサイトメトリーによって測定した。Treg細胞は恒常的にCD25を発現し、CD25発現レベルの上位1%の細胞はTreg細胞について高度に濃縮されている(Jailwala, P., et al., 2009, PLoS One. 2009; 4:e6527;Long, S. A., et al., 2010, Diabetes 59:407-15)。したがって、フローサイトメトリーのデータは、TregおよびCD4エフェクターT細胞サブセットについて、それぞれCD25high(CD25発現細胞の上位1~2%)およびCD25-/low群にゲートされた。
実施例3に記載された予想外の結果は、N88RL9AG1で検出され、D20HL0G2で検出されないIL2生物活性が、リンカーペプチドの存在によるものであったことを示唆した。この知見を検証し、他の変数、例えばFc部分のアイソタイプやIL-2部分における選択性変異等の寄与を取り除くために、一群の類似体(すべてIgG1 N297A Fcを使用するもの)を設計し生成した(表III)。
より幅広い生物学的文脈においてN88R/IL2-Fc融合タンパク質の選択性を決定するために、粗非分画ヒトPBMC中のすべての鍵となる免疫細胞種についてSTAT5活性化を測定するアッセイが開発された。ヒトPBMCは、正常なボランティアからフィコールハイパーク遠心によって単離された。106個のPBMCをグルコース(Lonza)および10%FBS(Omega)を有するX-VIVO15培地に懸濁し、37℃で20分間10-8Mの試験タンパク質で処理した。次に製造者の指示書に従ってFoxp3/Transcription Factor Staining Buffer Set(EBIO)で細胞を処理した。次に、実施例3に記載されるようにCytofix緩衝液で細胞を固定し、Perm Buffer IIIで透過処理した。固定し透過処理した細胞を次に1%FBS/PBSで洗浄し、暗所にて室温で60分間抗体混合物で染色した。染色された細胞を次に1%FBS/PBSで洗浄し、PBSに再懸濁し、Fortessaフローサイトメーター(BD Biosciences)で解析した。抗体混合物は、抗CD4-PerCP-Cy5.5(BD、#560650)、抗pSTAT5-AF-488(BD、#612598)、抗CD25-PE(BD、#560989)、抗CD56-PE-CF594(BD、#562328)、抗FOXP3-AF647(BD、#560889)、抗CD3-V450(BD、560366)、および 抗CD8-BV650(Biolegend、#301041)からなるものだった。この染色方法によって、7種類の鍵となる免疫細胞種におけるpSTAT5レベルの監視が可能になった。
実施例5に提示された結果は、N88R/IL-2-Fc融合タンパク質の頑健な生物活性のための、長さが6~20アミノ酸のリンカーペプチドの強い必要性を示し、リンカーペプチドの長さが増加するほど生物活性が増加した。さらにより長いリンカーペプチドが生物活性の増加を促進するかどうかを決定するために、ペプチドリンカーの長さが25および30アミノ酸の追加のタンパク質構築物を実施例1に記載されるように調製し、N88RL15AG1およびN88RL20AG1の独立な調製と併せて実施例3に記載されるようにT細胞pSTAT5生物活性アッセイにおいて試験した。この実験の結果は、ペプチドリンカーを25(N88RL25AG1)または30(N88RL30AG1)アミノ酸に増加させても、20アミノ酸リンカーを有するタンパク質よりも、CD25hi細胞に対してより大きな生物活性がもたらされなかったことを示した(表VII)。これらの結果は、実施例4に提示される結果と共に、IL-2生物活性を促進するペプチドリンカーの能力は15~20アミノ酸の長さでプラトーに達すること、およびより長いリンカーペプチドは生物活性のさらなる増加を促進しないことを示す。
本発明は以下の態様も提供する。

ここで前記IL-2融合タンパク質は高親和性IL-2受容体を選択的に活性化し、それによってヒト制御性T細胞を選択的に活性化する能力を有する、前記融合タンパク質。
[2] IL-2バリアントタンパク質が、ヒトIL2タンパク質(配列番号1)に対してN88R、N88G、D20H、Q126LおよびQ126Fからなる群より選択される置換を有するヒトIL-2を含む、[1]に記載の融合タンパク質。
[3] IL-2バリアントタンパク質が置換C125Sを有するヒトIL-2を含む、[1]に記載の融合タンパク質。
[4] 両方のIL-2バリアントタンパク質が配列番号1に提供されるN88Rである、[1]に記載の融合タンパク質。
[5] リンカーがグリシンおよびセリン残基を含み、リンカーが10~30アミノ酸である、[1]に記載の融合タンパク質。
[6] IgG Fcタンパク質が、該融合タンパク質のFc部分のエフェクター機能を低減する1つ以上のアミノ酸置換を含有する、[1]に記載の融合タンパク質。
[7] a. ヒトIL-2(配列番号1)に対してアミノ酸置換N88RおよびC125Sを有するIL-2バリアントタンパク質、
b. 配列番号15に記載のリンカーペプチド、ならびに
c. 配列番号2に記載のヒトIgG1 Fcバリアントタンパク質
を含む融合タンパク質であって、高親和性IL-2受容体を選択的に活性化し、それによってヒト制御性T細胞を選択的に活性化する能力を有する、前記融合タンパク質。
[8] a. ヒトIL-2(配列番号1)に対してアミノ酸置換N88RおよびC125Sを有するIL-2バリアントタンパク質、
b. 配列番号15に記載のリンカーペプチド、ならびに
c. 配列番号3に記載のヒトIgG2 Fcタンパク質
を含む、融合タンパク質。
[9] [1]の融合タンパク質および薬学的に許容可能な担体を含む、医薬組成物。
[10] ヒト制御性T細胞を選択的に活性化する方法であって、該方法は、ヒトIL-2(配列番号1)に対してアミノ酸置換N88RおよびC125Sを有するIL-2バリアントタンパク質、配列番号15に記載のリンカーペプチド、ならびに配列番号2に記載のヒトIgG1 Fcタンパク質を含む医薬組成物を投与することを含み、ここで前記医薬組成物は、ヒト制御性T細胞の濃度が所望のレベルに到達するまで治療上有効量で投与される、前記方法。
[11] ヒト制御性T細胞を選択的に活性化する方法であって、該方法は、[2]のIL-2バリアントタンパク質、ならびに
a. 配列番号2に記載のヒトIgG1 Fcタンパク質、
b. 配列番号3に記載のヒトIgG2 Fcタンパク質、および
c. 配列番号24に記載のヒトIgG4 Fcタンパク質ドメイン
からなる群より選択されるヒトIgG Fcタンパク質
を含む医薬組成物を投与することを含み、ここで前記医薬組成物は、ヒト制御性T細胞の濃度が所望のレベルに到達するまで治療上有効量で投与される、前記方法。
[12] ヒト血液細胞を0.01nM~1nMの濃度の[1]記載の融合タンパク質と接触させ、次にフローサイトメトリーによって該タンパク質に結合する細胞を検出することによって、ヒト血液試料中のTreg細胞の数を測定する方法。
[13] 2つの同一の鎖を含む2量体タンパク質であって、ここで各鎖はN末端ヒトIL-2バリアントタンパク質部分およびC末端IgG Fcタンパク質部分を含み、ここで
N末端ヒトIL-2バリアントタンパク質部分は、
a. N末端およびC末端を有し;
b. N88R、N88G、D20H、Q126LおよびQ126Fからなる群より選択される少なくとも1つの置換によって配列番号1のヒトIL-2野生型と相違し;
c. 配列番号1に対して少なくとも97%の配列同一性を有し;ならびに
d. Treg細胞上のIL2Rαβγに結合することによってTreg細胞を活性化する能力を有し;
N末端ヒトIL-2バリアントタンパク質はそのC末端で6~30アミノ酸残基のアミノ酸リンカーのN末端と連結し、ここで該リンカーはまたC末端を有し;ならびに
該アミノ酸リンカーのC末端は、配列番号2に対して95%の配列同一性を有しシステイン残基を含む、IgG Fcタンパク質部分のN末端に連結し;ここで2つの鎖はIgG Fcタンパク質部分のシステイン残基を通して互いに連結されている、前記2量体タンパク質。
[14] IL-2バリアントタンパク質が置換C125Sを有するヒトIL-2をさらに含む、[13]に記載の2量体タンパク質。
[15] アミノ酸リンカーが、グリシン残基、セリン残基、ならびにグリシンおよびセリン残基の混合物の群から選択されるリンカーからなる、[13]に記載のタンパク質。

Claims (19)
- ヒトIL-2バリアントタンパク質ドメイン、6~35アミノ酸のペプチドリンカーセグメントドメイン、およびIgG Fcドメインを含む融合タンパク質であって、各ドメインはアミノ末端(N末端)およびカルボキシ末端(C末端)を有し;該融合タンパク質は、ヒトIL-2バリアントタンパク質ドメインのC末端がペプチド結合によりペプチドリンカーのN末端と融合し、IgG Fcタンパク質部分のN末端がペプチド結合によりペプチドリンカーのC末端と融合するように構成され;
ここで前記IL-2融合タンパク質は高親和性IL-2受容体を選択的に活性化し、それによってヒト制御性T細胞を選択的に活性化する能力を有する、前記融合タンパク質。 - IL-2バリアントタンパク質が、ヒトIL2タンパク質(配列番号1)に対してN88R、N88G、D20H、Q126LおよびQ126Fからなる群より選択される置換を有するヒトIL-2を含む、請求項1に記載の融合タンパク質。

- 両方のIL-2バリアントタンパク質が配列番号1に提供されるN88Rである、請求項1に記載の融合タンパク質。
- リンカーがグリシンおよびセリン残基を含み、リンカーが10~30アミノ酸である、請求項1に記載の融合タンパク質。
