序論
本発明は、抗HLA−G特異的抗体の抗原結合ドメイン、任意に(i)ヒトIgG4ヒンジドメイン、(ii)ヒトIgG4ヒンジドメイン及びCH3ヒトIgG4ドメイン、若しくは(iii)変異CH2ヒトIgG4ドメイン、ヒトIgG4ヒンジドメイン及びCH3ヒトIgG4ヒンジドメインを含むか又はそれらからなるヒンジドメインにより大部分が構成される細胞外ドメインと、膜貫通ドメインと、CAR設計の生成に応じて1、2又は3つの共刺激構造、任意に切断可能リンカー及び任意にレポーターを含む細胞内ドメインとを含むキメラ抗原レセプター(CAR)に関する。
本発明の理解を容易にするために、いくつかの用語を以下に定義する。
「HLA−G」は、少なくとも7つのアイソフォームを含むヒト白血球抗原Gを示す。その発現は主に、絨毛外栄養膜細胞層上の胎児−母体インターフェース;胎盤、羊膜;胸腺、角膜、気管支上皮細胞、及び膵臓などの少数の健康な成人組織;並びに間葉系幹細胞、少数の活性化単球;並びに赤血球及び内皮前駆体などの異なるタイプの細胞に限定される。可溶性HLA−Gは、血漿、脳脊髄液、悪性腹水、胸水、精子などの体液中にも見いだされる。HLA−G遺伝子は一部の組織では活性でないが、プロゲステロンや抗癌剤などのある種の分子によってその発現を誘導することができる。さらに、この分子は、癌、多発性硬化症、炎症性疾患及びウイルス感染などの病態でも、又は同種異系移植後にも新規に発現する(neo-expressed)。可溶性HLA−G(sHLA−G)は個体の血清/血漿中に検出できる。
本発明は、7つのHLA−Gアイソフォームのうちの1つ〜6つ、好ましくは2つ〜5つのHLA−Gアイソフォームに特異的に結合するが、全てのHLA−Gアイソフォームには特異的に結合又は認識しない抗体を提供する。例えば、抗体は、1つ、2つ、3つ、4つ、5つ、又は6つのHLA−Gアイソフォームに特異的に結合することができる。HLA−GアイソフォームはHLA−G1、HLA−G2、HLA−G3、HLA−G4、HLA−G5、HLA−G6及びHLA−G7からなる群から選択することができ、好ましくはHLA−G1、HLA−G2、HLA−G5及びHLA−G6からである。

CARは、典型的には、エクトドメイン(細胞外ドメイン)及びエンドドメイン(細胞質ドメイン)を含み、好ましくは、それらはヒンジドメイン及び膜貫通ドメインによって結合される。特に、本発明のCARは、任意に、切断可能なリンカー及びレポーターを含む。細胞の表面に発現されるエクトドメインは、抗原結合ドメインすなわちレセプタードメインを含み、任意に、プロセシングのために抗原結合ドメインを小胞体に向けるシグナルペプチドを含む。細胞外ドメインは、標的抗原を特異的に認識する抗原結合ドメインを含む。非限定的な例として、抗原結合ドメインは、抗体、好ましくはscFvなどの一本鎖抗体であり得る。スペーサー領域は、抗原結合ドメインを膜貫通ドメインに連結し、抗原結合ドメインが抗原を認識できるように配向できるよう十分に柔軟に設計される。膜貫通ドメインは典型的には疎水性アルファヘリックスであり、典型的には、細胞膜の脂質二重層を横切って広がる。CARのエンドドメインは、活性化シグナルを細胞内部的に細胞質に広げ、そのことによりCARを発現する細胞を刺激する、シグナル伝達ペプチドからなる。エンドドメインは、下に説明するように、いくつかのシグナル伝達ドメインを含み得る。
本発明のCARの外部ドメインは、抗原結合ドメインである。特に、この抗原結合ドメインは、上の節で定義した抗体に由来する。

ヒンジドメイン
T細胞が抗原提示細胞と相互作用すると、約15nmの膜間距離を有する免疫シナプスが形成される。この距離はTCRとペプチド−MHC複合体の構造によって決まる。この空間的分離は、リン酸化カスケード及びT細胞活性化の効果的なトリガーに重要である。人為的に短縮すると、このタンパク質はシナプス内に留まる機会を得て、活性化シグナルの抑制をもたらす。
膜貫通モジュールは、レセプターを細胞表面に固定するように機能する。膜貫通ドメインに関して、CARは、CARの抗原結合ドメインに融合される膜貫通ドメインを含むように設計することができる。特に、CARは、CARの抗原結合ドメイン及びエンドドメインの両方に融合される膜貫通ドメインを含むように設計することができる。
ある実施形態では、T細胞レセプターζ鎖に由来するものなどの細胞質又は細胞内シグナル伝達ドメインは、標的抗原とキメラレセプターとの結合に続いて、Tリンパ球増殖及びエフェクター機能に対する刺激シグナルを産生するために、キメラレセプターの少なくとも一部として使用される。
抗原標的化ドメイン、ヒンジドメイン、膜貫通ドメイン、及びシグナル伝達エンドドメインに加えて、本発明のCARは、CARのN末端に連結されたシグナルペプチド配列をさらに含むことができる。シグナルペプチド配列は、多くの分泌タンパク質及び膜タンパク質のN末端に存在し、典型的には15〜30アミノ酸長を有する。上記のタンパク質分子の多くは、シグナルペプチド配列を有するので、これらのシグナルペプチドは、本発明のCARのためのシグナルペプチドとして使用することができる。

本発明の範囲には、本明細書に記載の本発明のCARの機能的部分が含まれる。用語「機能的部分」は、CARについて使用される場合、本発明のCARの任意の部分又はフラグメントを指し、その部分又はフラグメントは、それがその一部であるCAR(親CAR)の生物学的活性を保持する。機能的部分は、例えば、抗原若しくは標的細胞を認識する能力を保持するCARの部分、又は親CARと類似の程度、同程度、若しくはより高い程度で疾患を検出、治療、若しくは予防するCARの部分を包含する。親CARについては、機能性部分は例えば、親CARの約10%、25%、30%、50%、68%、80%、90%、95%、又はそれより多くを含むことができる。

本発明は、本明細書に記載のCARを発現する細胞に関する。この細胞は、CARを発現することができる免疫細胞、又はCARをコードする発現ベクターを保有する細菌細胞などの細胞を含む、あらゆる種類のものであってよい。
遺伝子を細胞に導入して細胞内で遺伝子を発現させる方法は当技術分野で公知である。
本明細書で開示される細胞の増殖及び遺伝子改変の前に、細胞は、対象から−例えば、自己療法を含む実施形態では−又は商業的に入手可能な培養物から得られ得る。
当業者に知られているように、各種の方法は、対象から免疫細胞を単離するために容易に利用可能であるか、又は例えば、Life TechnologiesのDynabeads(登録商標)システム;STEMcell TechnologiesのEasySep(商標)、RoboSep(商標)、RosetteSep(商標)、SepMate(商標);Miltenyi BiotecのMACS(商標)細胞分離キット、細胞表面マーカー発現並びに他の市販の細胞分離及び単離キット(例えば、Pierce,Rockford,ILのISOCELL)を使用して、本出願に適合させることができる。免疫細胞の特定のサブ集団は、独特の細胞表面マーカーに特異的なそうしたキットにおいて利用可能なビーズ又は他の結合剤の使用によって単離され得る。例えば、MACS(商標)CD4+及びCD8+マイクロビーズを用いて、CD4+及びCD8+T細胞を単離し得る。ヒト疾患の治療的介入としての、抗原特異的T細胞を単離し、拡大する戦略もまた、臨床試験において検証されている(Riddell et al., 1992; Walter et al., 1995; Heslop et al., 1996)。
所望のCARを発現するための免疫細胞の遺伝子改変の前又は後にかかわらず、一般に知られている方法、又は米国特許第6,352,694号;同第6,534,055号;同第6,905,680号;同第6,692,964号;同第5,858,358号;同第6,887,466号;同第6,905,681号;同第7,144,575号;同第7,067,318号;同第7,172,869号;同第7,232,566号;同第7,175,843号;同第5,883,223号;同第6,905,874号;同第6,797,514号;同第6,867,041号及び米国特許出願公開第2006/0121005号、Life Technologies Dynabeads(登録商標)システム活性化及び増殖キット;BD Biosciences Phosflow(商標)活性化キット、Miltenyi Biotec MACS(商標)活性化/増殖キット、並びに関連する細胞の活性化部分に特異的な他の市販の細胞キットに記載があるものなどの、本発明の用途に容易に適合する方法で、細胞を活性化及び増殖させることができる。HLA−G抗原によりエクスビボで刺激すると、選択されたCAR発現細胞サブ集団を活性化し、増殖させることができる。或いは、細胞は、HLA−G抗原との相互作用により、インビボで活性化され得る。
本発明の核酸コンストラクトを免疫細胞に形質導入して、本発明の抗HLA−G CARを発現する免疫細胞を作製することができる。ある実施形態では、細胞は、少なくとも1つの本発明のCARを含むよう形質導入される。
本発明はまた、上記の抗HLA−G抗体又は抗体フラグメント、CAR、核酸コンストラクト、ベクター及び/又は細胞を含む医薬組成物又は獣医学組成物に関する。
本発明は、医薬として使用するための、又は対象の疾患又は障害の治療に使用するための、本発明の医薬組成物又は本発明のCAR発現細胞に関する。本発明はまた、対象の疾患又は障害を治療するための医薬の製造における、本発明の医薬組成物又は本発明のCAR発現細胞の使用に関する。最後に、本発明の医薬組成物又は本発明のCAR発現細胞の治療有効量を対象に投与することを含む、対象の疾患又は障害を治療するための方法に関する。
疾患又は障害の治療及び併用療法における使用
一態様では、治療されるべき疾患又は障害は、増殖性の疾患又は障害、好ましくは癌;感染性の疾患又は障害、好ましくはウイルス感染;炎症性の疾患又は障害;及び免疫性の疾患又は障害、好ましくは自己免疫又は自己免疫疾患、アレルギー及び移植片対宿主拒絶から選択される病態である。いくつかの実施形態では、病態は癌であり得る。
HLA−Gは、その場で、多くのヒト固形悪性腫瘍及び悪性造血疾患(乳癌、卵巣癌、腎明細胞癌、結腸直腸癌、胃癌、食道癌、肺癌、肝細胞癌、並びに急性骨髄性白血病及び慢性リンパ性白血病(B−CLL)を含む)で異常発現する。悪性腫瘍におけるHLA−Gの異種発現は、結腸直腸癌(CRC)、胃癌(GC)、非小細胞肺癌(NSCLC)、食道扁平上皮癌(ESCC)、乳癌、肝細胞癌、及びB−CLLの患者の芳しくない臨床転帰と有意に相関する。さらに、血清可溶性HLA−Gは、正常な健常対照又は良性疾患症例と比較すると、様々なタイプの癌患者(メラノーマ、急性白血病、多発性骨髄腫、神経芽細胞腫、リンパ増殖性疾患、乳癌又は卵巣癌、非小細胞肺癌、食道癌、結腸直腸癌;胃癌及び肝細胞癌の患者を含む)で増加する。
本明細書で開示される抗HLA−Gモノクローナル抗体又はscFvはまた、診断方法及び予後方法に有用である。したがって、本発明は、対象のHLA−G関連医学的病態の診断における、本明細書で開示される抗体の使用に関する。
いくつかの実施形態では、本方法は、HLA−G CAR療法に応答する可能性が高いと判定された患者に有効量のHLA−G CAR療法薬を投与する追加の工程を含む。
本明細書に記載の組成物のいずれもが、本発明によって提供されるキットに含まれ得る。したがって、キットは、適切な容器手段中に、本発明の組換え/操作された細胞、及び/又は本発明の核酸コンストラクトをコードするベクター、及び/又は本発明の核酸コンストラクト若しくは関連試薬を含む。いくつかの実施形態では、キットは、癌又は感染症を治療するための追加の薬剤をさらに含み、追加の薬剤は、核酸コンストラクト若しくは細胞又は本発明のキットの他の要素と組み合わせてもよく、或いはキット中で別々に提供されてもよい。いくつかの実施形態では、個体から試料を採取する、及び/又は試料を分析する手段は、キット中に提供され得る。ある実施形態では、キットは例えば、細胞、緩衝液、細胞培地、ベクター、プライマー、制限酵素、塩などを含む。キットはまた、滅菌された、薬学的に許容される緩衝液及び/又は他の希釈剤を含有するための手段を含み得る。
結果
抗HLA−G抗体パラトープの特異性
HLA−Gの異なるアイソフォームは、β2M及びペプチドに会合しているか否かにかかわらず、1〜3個の球状ドメインを含有して発現され得ることは以前に記載した。異なる特異的抗HLA−G抗体は、HLA−Gの異なる領域又は「エピトープ」に対して以前に作製されていた。抗HLA−Gモノクローナル抗体の特異性は、以前にHLA−Gの異なるアイソフォームに対して決定されていた。簡潔に言えば、図2Aに示すように、15E7抗体はHLA−G1/5 β2Mフリー及びHLA−G6に対して高い親和性を提示し、このMabがβ2Mに会合しないHLA−Gアイソフォームに特異的であることを示している。図2Bに示すように、K562 HLA−G1pは、HLA−G β2M−会合アイソフォームに特異的なMEM−G/09モノクローナル抗体と比較して、15E7モノクローナル抗体での染色後に、K562 HLA−G1Lvよりも高い割合のHLA−G β2M−フリーを提示する(データは示さず)。同様に、15E7の特異性をJurkat wt、Jurkat HLA−G1Lv及びJeg−3細胞株に対して評価したところ、図2Bに示すように、15E7モノクローナル抗体は、HLA−G β2Mフリーアイソフォームに対して高い親和性を提示することが示された。
15E7及びLFTT−1モノクローナル抗体パラトープに基づいて、HLA−Gに対する2つの異なるキメラ抗原レセプターを作製した。既に報告されている(Pule et al. 2005 Molecular Therapy)第3世代のCARに対応したCARの設計は、シグナルペプチド、15E7若しくはLFTT−1のいずれかのscFv、以下の(i)〜(iv)を含むか、若しくはそれらからなるスペーサードメイン;(i)CD8a若しくはCD28ヒンジ、(ii)ヒトIgG4ヒンジドメイン、(iii)ヒトIgG4ヒンジドメイン及びCH3ヒトIgG4ドメイン、又は(iv)変異CH2ヒトIgG4ドメイン、ヒトIgG4ヒンジドメイン及びCH3ヒトIgG4ヒンジドメイン;、CD28膜貫通領域、並びに4−1BBとCD3ζの2つの共活性化セグメントから構成される。図3はCD8a又はCD28ヒンジドメインを有する15E7及びLFTT−1に基づくCAR構造の概略設計の要約であり、図4は、切断可能リンカーP2A及びレポーターとしての切断型hCD19を有する15E7及びLFTT−1に基づくCAR構造の概略設計の要約である。
免疫細胞は、APC又は腫瘍細胞抗原−提示細胞などの標的細胞から細胞表面膜パッチを獲得することが以前に実証された。エフェクター細胞による標的細胞からの膜獲得は、(i)細胞間接触を必要とし、(ii)迅速であり、且つ(iii)獲得側細胞の活性化状態に依存する能動的プロセスであり、獲得側細胞の活性化状態は、(iv)エフェクター細胞の抗原特異的活性化を反映する。従って、膜転移は、機能的に成熟した免疫細胞の活性化に関連している。
エフェクター細胞を発現する15E7及びLFTT−1 CARの細胞毒性機能を、JEG−3、K562及びK562−HLA−G1トランスフェクト細胞株に対して評価した(図8及び9)。そうするために、標的細胞をCFSEで標識した後、示されたエフェクター:標的比(E:T)でCAR−T細胞と24時間、共培養した。共インキュベーションの24時間後、培地を回収してIFN−γ分泌を測定し、細胞を回収してフローサイトメトリーにより標的細胞溶解を調べた。
HLA−G CAR−T細胞のインビボでの機能を調べるために、ルシフェラーゼ(K562−HLA−G1P−luc)を発現するK562−HLA−G1P腫瘍細胞をNGSマウスにインプラントした後、HLA−G CAR−T細胞を接種し、HLA−G腫瘍細胞に対するHLA−G CAR−T細胞の細胞毒性をモニターした(図10)。
細胞株
Jurkat細胞株は、ATCC(アメリカン・タイプ・カルチャー・コレクションTIB−152)から購入したヒトCD4+T細胞である。Jurkat細胞株に、HLA−G1、HLA−G−LFTT−1、又はHLA−G−15E7 CARレンチウイルスそれぞれを形質導入した。Jurkat wt及びJurkat形質導入細胞株を、2mM L−グルタミン、1mg/mlのペニシリン及びストレプトマイシン(Gibco)、並びに10%熱不活化FCS(Invitrogen)を添加したRPMI1640(Invitrogen)で培養した。
HLA−G−LFTT−1及びHLA−G−15E7 CARコンストラクトを、既に説明したように作製した[7]。簡単に説明すると、抗HLA−G認識ドメインは、HLA−G1/β2M会合特異的抗体LFTT−1又はHLA−G1/β2Mを含まない特異的抗体15E7に由来する一本鎖可変断片(scFv)である。IgG1ヒンジ領域に由来する短いスペーサーを使用して、このscFvを膜貫通ドメインに連結させた。HLA−G CAR内部ドメインは、CD28、OX40、及びCD3ζ活性化分子の融合によって構成されていた。CARコンストラクトを、CMV前初期プロモーター下で、特定のプライマーを用いたPCRによる抽出後、消化/ライゲーションによってpTripプラスミドベクターにクローン化した。MabのscFvの軽鎖及び重鎖をコードするアミノ酸配列は、LFTT−1については配列番号1(VH)、2(VL)及び31(scFV)に、15E7については配列番号3(VH)、4(VL)及び30(scFV)に詳述されている。CAR配列は、15E7 CARについては、配列番号83及び84(それぞれアミノ酸配列及び核酸コード配列)に開示され、LFTT−1 CARについては、配列番号85及び86(それぞれアミノ酸配列及び核酸コード配列)に開示される。
HLA−Gを発現する安定したJurkat細胞株を、形質導入によって作製し、レンチウイルス粒子を次のように作製した:Zhao et al.[38]に従ってネイティブHLA−G1 cDNA(NM_002127.5)修飾K334A及びK335Aに対応する特定の配列を、CMV前初期プロモーター下で、特定のプライマーを用いたPCRによる抽出後、消化/ライゲーションによってpTripプラスミドベクターに個別にクローン化した。
HIV−1由来のベクター粒子を、HEK−293T細胞(ATCC)と組換えプラスミドpTRIP、VSV由来の糖タンパク質をコードするエンベロープ発現プラスミド、Indiana血清型糖タンパク質、及びp8.74キャプシド形成プラスミドのリン酸カルシウム共トランスフェクションによって作製した。ウイルスストックを、形質導入HEK−293T細胞からの細胞溶解物のリアルタイムPCRによって漸増し、1ml当たりの形質導入単位(TU)として表した。Jurkat HLA−G−LFTT−1、HLA−G−15E7 CAR細胞、及びJurkat HLA−Gを作製するために、1×105のJurkat細胞を、500μlのcRPMI培地及び106TU(293T)のTrip CMV−HLA−G−LFTT−1−CAR、Trip CMV−HLA−G−15E7−CAR、又はTrip CMV−HLA−Gベクターそれぞれを含む12ウェルプレートに播種した。細胞を37℃で1時間培養し、次いで、37℃、1200gで1時間遠心分離した。その後、1mlのcRPMI培地を添加し、37℃で培養した。2週間後、陽性細胞を抗HLA−G抗体を使用したフローサイトメトリーで選別した。HLA−Gの発現を、Jurkat HLA−G CAR活性化アッセイの前にフローサイトメトリーによって評価した。同じ手順を行い、K562 HLA−G1Lv及びNKT1.2 HLA−G−LFTT−1 CAR又はHLA−G−15E7 CAR細胞を得た。
eBiosciences(Paris; France)からのPE結合マウスIgG1(クローンP.3.6.8.2.1. 12-4714)、BD BiosciencesからのFITC結合ラットIgG2a(クローン:R35-95 553929, Le Pont de Claix; France)及びMEM−G/09(Exbio, Praha)。フローサイトメトリーは、対応する細胞株をモノクローナル抗体と共にRTで1時間インキュベートし、2回洗浄し、PE結合ヤギ抗マウスIgG抗体(405307、Biolegend, USA)と共にRTで30分間インキュベートした。フローサイトメトリー分析は、LSR FORTESSA(Beckton Dickinson, Le Pont-de-Claix, France)を用いて実施し;データはFlowJo Xソフトウェア(Tree star, Ashland, USA)を用いて分析した。ポジティブ標識集団の%は、アイソタイプ対照の99%によって示されるものよりも高い染色強度を有するものとして定義した。
Jurkat HLA−G CARエフェクター細胞及びJurkat又はJurkat HLA−G1腫瘍細胞のいずれかを、製造業者の仕様に従って、PKH26及びPKH67蛍光色素(Sigma)でそれぞれ標識した。
ベクタープラスミドpTRIP(血清型インディアナ(IND)であるVSV由来の糖タンパク質をコードする)及び組み込みレンチウイルスベクター粒子(ILV)の作製のためのp8.74カプシド化プラスミドと、HEK293T細胞(ATCC)との一過性リン酸カルシウム同時遺伝子導入によって、HIV−1由来のベクター粒子を作製した。ベクター遺伝子導入能力は、以前に報告されているように(Coutant, F., et al., PLoS One, 2008. 3(12): p. e3973)、293T細胞の形質導入後の定量的PCRによって決定し、ベクターの形質導入単位(TU)/mLで表した。
フィコールの単離後、3人の異なる健康なドナー(EFS、Rungis)の血液試料からPBMCを抽出した。T細胞を、カラム精製(Miltenyi)によって選別し、CD3+CD28+被覆マイクロビーズ(Miltenyi)で活性化し、次いで、10%FCS 1%ペニシリン−ストレプトマイシン(Gibco)、50μMベータ−メルカプトエタノール(Gibco)、非必須アミノ酸、10mM Hepes(Gibco)及び1mMピルビン酸ナトリウム(Gibco)を補充したRPMI 1640 Glutamax(Gibco)中、37℃、5%CO2で48時間培養した。次に、細胞を洗浄し、ゆっくりと振盪しながら4時間、1M/mlの200μl中、MOI20のレンチウイルスベクターで形質導入した。次いで、細胞をU底96ウェルプレートに移した。24時間後、細胞を1.106細胞/mlに調整し、50U/mlのヒトIL2を添加した(Preprotech)。2〜3日毎に、細胞を計数し、IL2を含む完全培地中で1.106細胞/mlに調整した。8日後、細胞を機能分析に使用した。
JEG−3、K562及びK562−HLA−G1標的細胞について細胞毒性分析を行った。
IFNγの定量は、製造業者の説明書に従って、CBAキット(BD Biosciences)を用いて共培養培地上で直接行った。
共培養細胞を、以前の記載のように調製し(E:T比10)、抗CD107a(クローンH4A3 PE、Biolegend)を、共培養物に直接添加した。共インキュベーションの1時間後、モネンシン(GolgiStop, BD Bioscience)を添加した。共インキュベーション実験の5時間後に細胞を回収し、CD4、CD8、CD19及びLive/dead抗体で標識した。Attuneサイトメーターを用いて獲得し、FlowJoソフトウェアにより結果を分析した。
NOD/SCID/IL2Rγc欠損(NSG)マウス(6〜8週齢、The Jackson laboratory, Sacramento, CA, USA)を照射(2.5Gy)し、K562−HLA−G1P(1.106個/マウス)の腫瘍細胞を発現する適切な数のルシフェラーゼを静注した。K562−HLA−G1P−Lucモデルには、3日目にT細胞(形質導入されていないもの、されたもの)を尾静脈に注射した。移植後17日目まで3〜4日毎にインプラントをモニターし、次いでBLI測定によって週1回モニターした:マウスは、画像化(IVIS Lumina Series III、Perkin Elmer、Massachusetts、USA)の10分前以内に3mgのルシフェリン(VivoGlo Luciferin、#P1043、Promega、Wisconsin、USA)の腹腔内投与を受けた。
データは平均値+/−標準偏差(SD)として表す。スチューデントのt検定を使用して、0.05未満のP値が有意であると見なした。代表的な実験を示す図では、エラーバーは3連のSDを表す。
CAR免疫療法の基本的な開発基準は、適正な腫瘍関連抗原の特定、遺伝子導入エフェクター細胞の接近性、免疫抑制性腫瘍微小環境の可逆性であることを考えると、HLA−Gは、この目的のための非常に興味深い候補であることは明らかであるように思われる。HLA−Gは、免疫調節に重要な役割を果たすことからICPと考えられ、この分子が多様な起源の多数の腫瘍滲出液中に発現していることも広く報告されていて、それに対する細胞性又は体液性応答はこれまで報告されていない。さらに、HLA−Gは特異な分子であり、その機能は特徴づけられているが、そのタンパク質構造は非常に複雑で不均一であることが判明している。したがって、HLA−G機能を阻害するには、単に1つのアプローチだけでは十分ではないだろう。
Claims (23)
- 抗HLA−G抗体の抗原結合ドメイン、膜貫通ドメイン、及び細胞内ドメインを含むキメラ抗原レセプター(CAR)であって、1〜6個のHLA−Gアイソフォーム、好ましくは2〜5個のHLA−Gアイソフォームに特異的に結合する、CAR。
- β2MフリーHLA−Gアイソフォーム又はβ2M会合HLA−Gアイソフォームに特異的に結合する、請求項1に記載のCAR。
- HLA−Gのアルファ1ドメインに結合しない、請求項1又は2に記載のCAR。
- HLA−G1、HLA−G2、HLA−G5及びHLA−G6からなる群から選択されるHLA−Gアイソフォームと特異的に結合するか、或いはHLA−G1及びHLA−G5の両方、若しくはHLA−G2及びHLA−G6の両方、及び/又はHLA−G1/β2Mフリー及びHLA−G5/β2Mフリーの両方のアイソフォームに特異的に結合する、請求項1〜3のいずれか一項に記載のCAR。

- 前記膜貫通ドメインは、CD28、CD3及びCD8膜貫通ドメインから選択され、好ましくは、前記膜貫通ドメインはCD28膜貫通ドメインである、請求項1〜7のいずれか一項に記載のCAR。
- N末端からC末端に向かって連続的に、ペプチドシグナル配列、抗HLA−G抗体又はその抗原結合フラグメント、スペーサードメイン、膜貫通ドメイン、細胞内ドメイン、切断可能リンカー、及び切断型ヒトCD19ドメインを含む、請求項1〜8のいずれか一項に記載のCAR。
- 前記抗原結合ドメインを前記膜貫通ドメインに連結するヒンジ領域、好ましくは(i)ヒトIgG4ヒンジドメイン、(ii)ヒトIgG4ヒンジドメイン及びCH3ヒトIgG4ドメイン、(iii)変異CH2ヒトIgG4ドメイン、ヒトIgG4ヒンジドメイン及びCH3ヒトIgG4ヒンジドメイン、(iv)CD28ヒンジドメイン、並びに(v)CD8aヒンジドメインからなる群から選択されるヒンジ領域をさらに含む、請求項9に記載のCAR。
- 前記ヒンジドメインは、(i)配列番号25、(ii)配列番号25及び配列番号27、(iii)配列番号25、配列番号26及び配列番号27、(iv)配列番号18若しくは(v)配列番号19の配列、又はそれと80%超、好ましくは90%超、さらに好ましくは95%超の同一性を示す相同配列を含むか、或いはそれからなる、請求項10に記載のCAR。
- 前記シグナルペプチドは、CD8aシグナルペプチド、マウスIgカッパシグナルペプチド、ヒトIgG4シグナルペプチド、IL2シグナルペプチド、ヒトIgG2シグナルペプチド、及びガウシアlucシグナルペプチドからなる群から選択される、請求項9〜11のいずれか一項に記載のCAR。
- 前記切断可能リンカーは、P2A、T2A、E2A、B2A及びF2Aからなる群から選択される、請求項9〜12のいずれか一項に記載のCAR。
- 前記切断型ヒトCD19ドメインは、配列番号29の配列からなる、請求項9〜13のいずれか一項に記載のCAR。
- 少なくとも2つの異なる抗原結合ドメイン、膜貫通ドメイン及び細胞内ドメインを含む多重特異性CARコンストラクトであって、前記抗原結合ドメインの少なくとも1つは、HLA−Gのβ2Mドメインに会合したHLA−Gアイソフォームと特異的に結合し、前記抗原結合ドメインの少なくとも別のものは、HLA−Gのβ2Mドメインに会合しないHLA−Gアイソフォームと特異的に結合し、好ましくは、前記多重特異性CARコンストラクトは、(i)配列番号11、12及び13の配列をそれぞれ含むCDR1、2及び3(HCDR1、HCDR2、HCDR3)を含むドメイン、(ii)配列番号14、15及び16の配列をそれぞれ含むCDR1、2及び3(LCDR1、LCDR2、LCDR3)を含むドメイン、(iii)配列番号5、6及び7の配列をそれぞれ含むCDR1、2及び3(HCDR1、HCDR2、HCDR3)を含むドメイン、(iv)配列番号8、9及び10の配列をそれぞれ含むCDR1、2及び3(LCDR1、LCDR2、LCDR3)を含むドメインを含む二重特異性CARコンストラクトであり、任意に、各CDRは任意に1、2、3又は4個のアミノ酸置換、欠失又は挿入を含み得る、多重特異性CARコンストラクト。
- 請求項1〜15のいずれか一項に記載の前記CARをコードする、核酸分子。
- 請求項16に記載の前記核酸分子を含む、発現ベクター。
- 請求項1〜15のいずれか一項に記載のCAR、又は請求項16に記載の核酸分子、又は請求項17に記載の発現ベクターを含む細胞であって、好ましくは、T細胞、CD4+T細胞、CD8+T細胞、B細胞、NK細胞、NKT細胞、単球及び樹状細胞からなる群から選択され、好ましくは、T細胞、B細胞又はNK細胞である、細胞。
- 請求項16に記載の核酸分子、請求項17に記載の発現ベクター、又は請求項18に記載の細胞を含み、かつ任意に薬学的に許容される担体を含む、医薬組成物。
- 請求項19に記載の医薬組成物であって、
(i)β2M会合HLA−G、好ましくはHLA−G1及びHLA−G5の両方に、特異的に結合する細胞であって、好ましくは請求項5に記載のCARを含む細胞、並びにβ2MフリーHLA−G、好ましくはHLA−G2及びHLA−G6の両方、及び/又はHLA−G1/β2Mフリー及びHLA−G5/β2Mフリーの両方のアイソフォームに、特異的に結合する細胞であって、好ましくは請求項5に記載のCARを含む細胞、を含む医薬組成物、或いは
(ii)HLA−G1及びHLA−G5の両方に若しくはβ2MフリーHLA−Gアイソフォームに特異的に結合するCAR、好ましくは請求項5に記載のCAR、を含む細胞、並びにHLA−G2及びHLA−G6の両方に、若しくはβ2M会合HLA−Gアイソフォームに、特異的に結合するCAR、好ましくは請求項5に記載のCARを含む細胞、を含む医薬組成物。 - 癌の治療に使用するため、若しくはウイルス感染の治療に使用するための、請求項18に記載の細胞又は請求項19〜20のいずれか一項に記載の医薬組成物であって、好ましくは、HLA−Gを標的としないCAR療法と組み合わせて投与される、細胞又は医薬組成物。
- HLA−G β2M会合アイソフォームに、好ましくはHLA−G1及びHLA−G5の両方に、特異的に結合する抗HLA−G抗体であって、好ましくは、
(a)(i)重鎖可変領域は、配列番号1、若しくは配列番号1と80%超、好ましくは90%超、さらに好ましくは95%超の同一性を示す相同配列を含み、かつ
(ii)軽鎖可変領域は、配列番号2、若しくは配列番号2と80%超、好ましくは90%超、さらに好ましくは95%超の同一性を示す相同配列を含む、又は
(b)(i)重鎖可変領域のCDR(相補性決定領域)1、CDR2及びCDR3は配列番号1を含み、軽鎖可変領域のCDR1、CDR2及びCDR3は配列番号2を含む、
抗HLA−G抗体。 - 前記抗HLA−G抗体若しくはその抗原結合フラグメント、好ましくはscFvは、β2MフリーHLA−G若しくはβ2M会合HLA−Gアイソフォームに選択的に結合するが、HLA−Gアイソフォームの全てには結合しない、請求項1〜15のいずれか一項に記載のCAR又は請求項22に記載の抗HLA−G抗体。
