Description
본 발명은 면역관문을 제어할 수 있는 인자를 발현하는 면역세포, 이의 제조를 위한 발현 벡터, 이를 포함하는 조성물 및 이의 용도에 관한 것이다.
암을 치료하기 위한 방법은 꾸준한 발전과 변화 과정을 거쳐 왔으며, 외과적 수술, 화학요법, 방사선 요법 등의 방법이 현재까지 계속 이용되고 있다. 그러나 이러한 기존의 암 치료 방법들은 암이 전이되지 않은 초기 상태에만 효과가 있는 경우가 대부분이며, 이미 전이가 진행된 상태에서는 예컨대 외과적 수술을 하더라도 추후 재발의 가능성이 높다는 문제점이 있다. 이에, 암을 치료하기 위해 면역 반응을 이용하는 방법에 대한 연구가 계속되고 있다.
최근 혈액암 치료에서 키메라 항원 수용체(chimeric antigen receptor, CAR)를 발현하는 T세포(CAR-T)의 치료적 가능성이 입증되었으며, CD19 또는 BCMA (B-cell maturation antigen)를 타깃으로 하는 CAR-T 치료제가 시판된 바 있다. 하지만 혈액암과는 다른 고형암의 특성, 그 중에서도 면역 반응을 억제하는 종양 미세환경(tumor microenvironment, TME) 때문에 고형암 CAR-T 치료제 이외에도 효율적으로 TME를 공략하여 암을 치료할 수 있는 있는 전략이 추가적으로 필요하다.
한편, 인간백혈구 항원 G (HLA-G, human leukocyte antigen G)는 비전형적인(non-classical) HLA 클래스 I 분자로 태반기저부의 융모외영양막(extravillous cytotrophoblast)에서만 특이적으로 발현한다고 알려져 있다. 그리고 이러한 발현을 통해 태아와 모체 사이의 면역관용(immune tolerance)을 유지하는데 중요한 역할을 한다고 생각되고 있다.
HLA-G의 면역세포 수용체는 ILT2 (Ig-like transcript 2), ILT4, KIR2DL4 (Killer cell immunoglobulin like receptor, two Ig domains and long cytoplasmic tail 4) 등으로 알려져 있으며, 이 수용체들은 모두 ITIM 모티프 (immunoreceptor tyrosine-based inhibitory motif)를 가지고 있다. HLA-G가 수용체에 결합하면 이 모티프로 인해 면역세포를 억제하는 신호전달체계가 활성화된다.
HLA-G의 발현은 일반세포에서 제한적이나, 여러 종류의 암종에서는 과발현되며(Lin A, Yan WH. Human Leukocyte Antigen-G (HLA-G) Expression in Cancers: Roles in Immune Evasion, Metastasis and Target for Therapy. Mol. Med. 21(1), 782-791 (2015)), 이는 면역 억제 미세환경(immune suppressive microenvironments)이나 진행암의 단계, 불충분한 치료 반응 또는 예후와 연관되어 있다.
또한, HLA-G는 다양한 암종에서 과발현하는 특성으로 인해 내인성(endogenous) 면역세포뿐만 아니라 특정 항원을 타깃으로 하는 면역세포치료제들의 효능을 저해시킬 수 있다. 즉, HLA-G는 면역을 억제하는 면역관문(immune checkpoint)의 역할을 수행하기 때문에 내인성 및 외인성 면역세포의 효능을 저하시킬 수 있다. 따라서, 면역반응을 이용하여 암을 효과적으로 치료하기 위해서는 HLA-G의 면역관문 기능을 극복할 수 있는 방법이 필요하다.
상기와 같은 상황에서 본 발명자들은 인간 백혈구 항원 G (HLA-G) 과발현 암종, 특히, 고형암의 종양 미세환경을 극복할 수 있는 면역세포를 개발하고자 예의 연구 노력하였다. 그 결과 HLA-G에 특이적으로 결합하는 항-HLA-G 항체 유래의 가용성 scFv를 발현 및 분비하는 면역세포를 제조하고, 제조한 면역세포가 HLA-G를 발현하는 세포주에 대해 일반 면역세포보다 높은 항암 활성을 나타내는 것을 규명함으로써, 본 발명을 완성하게 되었다.
본 발명에 따르면, 본 발명의 면역세포는 HLA-G와 그 수용체의 상호작용을 저해할 수 있는 가용성 단일쇄 Fv(scFv)를 분비하므로, 종양 미세환경이 개선되어 종양침윤림프구와 같은 면역세포들을 활성화시킬 수 있고, 내인성 면역세포의 활성도 추가적으로 유도할 수 있다. 따라서 본 발명의 면역세포는 더 효율적으로 암세포를 제거할 수 있고, HLA-G의 고발현과 관련된 암뿐만 아니라 다양한 암의 치료에 사용될 수 있다.
이에 본 발명의 목적은 면역억제 활성을 갖는 HLA-G 및 그의 수용체로 이루어진 군에서 선택되는 표적에 특이적으로 결합하는 가용성 scFv를 발현하여 항종양 활성이 높은 면역세포를 제공하는 것이다.
또한, 본 발명의 다른 목적은 상기 면역세포에 항원 인식 수용체를 추가로 발현시킨 면역세포를 제공하는 것이다.
본 발명의 또 다른 목적은 상기 면역세포를 제조하기 위한 발현 벡터를 제공하는 것이다.
본 발명의 또 다른 목적은 상기 면역세포 및 약제학적으로 허용되는 담체를 포함하는 암 치료용 약제학적 조성물을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명의 일 양태는
항원 인식 수용체; 및
인간 백혈구 항원 G (human leukocyte antigen G, HLA-G) 및 그의 수용체로 이루어진 군에서 선택되는 표적에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편;을 발현 및 분비하는, 면역세포를 제공한다.
또한, 본 발명의 다른 양태는 인간 백혈구 항원 G(HLA-G) 및 그의 수용체로 이루어진 군에서 선택되는 표적에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 발현 및 분비하는 면역세포를 제공한다.
본 발명에서, 상기 면역세포는 자연 살해 세포, T 세포, B 세포, 대식세포, 수지상 세포, 자연 살해 수지상 세포, 비만세포 및 이들의 전구세포로 이루어진 군으로부터 선택될 수 있다. 보다 구체적으로는 T 세포, B 세포와 같은 효과기 세포(effector cell)일 수 있다.
본 발명에서, 상기 면역세포는 자가 세포의 집단 또는 동종 세포의 집단을 포함한다. 즉, 상기 면역세포는 인간 백혈구 항원 G (HLA-G) 및 그의 수용체로 이루어진 군에서 선택되는 표적에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 발현 및 분비하는 자가 세포의 집단 또는 동종 세포의 집단을 포함한다.
본 명세서에서, 용어 "자가"는 개체에게 재도입 될 예정인, 동일한 개체로부터 유래된 임의의 물질을 나타낸다. 본 명세서에서, 용어 "동종"은 물질이 도입되는 개체와 동일한 종의 상이한 동물로부터 유래된 임의의 물질을 나타낸다.
본 발명에서, 상기 두 면역세포의 차이는 항원 인식 수용체의 존재 유무로 본 발명에서, 상기 항원 인식 수용체는 표현 그대로 세포에 존재하는 특정 항원을 인식하여 결합하는 수용체로 항원과 결합하면 면역세포를 활성화시킨다.

상기 외인성 항원 인식 수용체란 해당 면역세포에서 자연적으로는 발현되지 않으나, 키메라 항원 수용체(chimeric antigen receptor, CAR)와 같이 인공적으로 수용체 컨스트럭트를 제작한 다음 면역세포에 전달하여 발현시킨 수용체이다.
상기 내인성 항원 인식 수용체란 해당 면역세포에서 자연적으로 발현되는 수용체를 말하며, 예로는 T 세포 수용체(T cell receptor, TCR), 또는 NK 세포가 가진 비정상세포를 인지하는 NKG2D 등과 같은 활성화 수용체가 있다.
상기 외인성 또는 내인성 항원 수용체의 구분은 상대적인 것으로 T 세포에서 TCR은 내인성 항원 수용체에 해당하지만 다른 면역세포에서 발현되는 TCR은 외인성 항원 수용체에 해당한다.
상기 항원 인식 수용체는 일반적으로 항원 결합 도메인, 신호 서열(leader sequence, LS), 힌지(hinge), 막관통 도메인(transmembrane domain, TM), 및 세포 내 신호전달 도메인(intracellular signaling domain, ICD)으로 구성될 수 있으나, 그 구조나 구성이 상기에 언급한 범위로 제한되는 것은 아니다.
상기 항원 결합 도메인은 타깃 세포의 항원을 인식하며, 항체 또는 항체 단편일 수 있다. 상기 신호 서열은 항원 인식 수용체를 세포막으로 이동시켜 세포 표면에 발현 및 노출될 수 있도록 하는 서열을 말하며, 막관통 도메인은 항원 결합 도메인과 세포내 신호전달 도메인을 세포막 사이로 연결하는 도메인이다.
본 발명에서, 항원 인식 수용체에 사용되는 신호 서열, 힌지, 막관통 도메인 및 세포내 전달 도메인은 본 발명이 속하는 기술 분야에서 통상적으로 사용되는 것일 수 있다.

보다 구체적으로 상기 인간 백혈구 항원 G (HLA-G)에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편은 서열번호 1 내지 3으로 이루어진 군에서 선택되는 아미노산 서열을 포함할 수 있으며, 서열번호 4 내지 6으로 이루어진 군에서 선택되는 아미노산 서열을 포함할 수 있다.

본 발명에서 용어, "항체"는 면역학적으로 특정 항원과 반응성을 갖는 면역글로불린 분자를 포함하는, 항원을 특이적으로 인식하는 수용체 역할을 하는 단백질 분자를 의미한다. 상기 항체에는 단일 클론 항체, 다클론 항체, 단일클론 및/또는 다중클론 항체의 혼합물, 전장 항체(full-length antibody) 및 항체 단편이 모두 포함된다. 전장 항체 또는 온전한 항체(intact antibody)는 천연 항체 구조와 실질적으로 유사한 구조를 가지거나, 본원에서 정의된 바와 같은 Fc 영역을 함유하는 중쇄를 갖는 항체를 언급하기 위해 상호 교환적으로 사용된다.
또한, 항체는 이가 (bivalent) 또는 이중 특이성 분자(예컨대, 이중특이성 항체)를 포함할 수 있다. 아울러, 항체는 서열의 유래에 따라 인간 항체, 인간화 항체, 또는 키메라 항체일 수 있다.

본 발명에서, 용어 "경쇄"는, 항원에 특이성을 부여하기 위해 충분한 가변영역의 아미노산 서열을 포함하는 가변 영역 도메인 VL 및 불변 영역 도메인 CL을 포함하는 전체 길이 경쇄 및 이의 단편을 모두 일컫는다.
본 발명에서 용어, "단편", "항체 단편", "항원 결합 단편" 또는 "항원 결합 도메인"은 항체의 항원결합 기능을 보유하는 항체의 임의의 단편을 지칭하는 것으로 호환적으로 사용된다. 예시적인 항원 결합 단편은 Fab, Fab', scFV, F(ab')2 및 Fv 등을 포함하나, 이에 제한되지 않는다.

단일쇄 Fv는 항체 중쇄 및 경쇄의 가변 영역을 10개 내지 25개 아미노산의 짧은 링커로 연결시킨 항체 단편의 일종이다.
본 발명의 일 구체예에 따르면, 상기 인간 백혈구 항원 G (HLA-G)에 특이적으로 결합하는 scFv는 세포 외부에서 도입된 상기 scFv를 코딩하는 뉴클레오티드 서열로 이루어진 핵산에 의해 면역세포에서 발현 및 분비될 수 있다.
따라서, 상기 면역세포는 인간 백혈구 항원 G (HLA-G)에 특이적으로 결합하는 scFv를 코딩하는 뉴클레오티드 서열로 이루어진 핵산을 포함할 수 있으며, 해당 뉴클레오티드 서열은 하기에서 선택될 수 있다:

한편, 본 발명자들은 면역세포에서 인간 백혈구 항원 G (HLA-G)에 특이적으로 결합하는 가용성 scFv 또는 상기 가용성 scFv와 항원 인식 수용체를 발현시키기 위해 발현 벡터를 제작하였다.
따라서, 본 발명의 다른 양태는 상기 면역세포를 제조하기 위한 하기의 발현 벡터를 제공한다:

상기 (a), (b) 및 (c)의 발현 벡터에서, 인간 백혈구 항원 G에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 코딩하는 뉴클레오티드 서열은 면역세포에서 설명한 뉴클레오티드 서열 서열과 동일하다.
본 발명의 일 구체예에 따르면, 상기 (a)의 발현 벡터는, 항원 인식 수용체를 코딩하는 뉴클레오티드 서열 및 인간 백혈구 항원 G 및 그의 수용체로 이루어진 군에서 선택되는 표적에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 코딩하는 뉴클레오티드 서열이 자기 절단형 펩티드를 코딩하는 뉴클레오티드 서열을 통해 연결된 것일 수 있다 (도 1a 내지 1c). 자기 절단형 펩티드를 코딩하는 뉴클레오티드 서열 서열은 서열번호 7의 아미노산 서열을 코딩하는 서열이다.
본 발명의 일 구체예에 따르면, 상기 (a), (b) 및 (c)의 발현 벡터에서, 인간 백혈구 항원 G에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 코딩하는 뉴클레오티드 서열은 구체적으로는 신호 서열을 코딩하는 뉴클레오티드 서열, 항-HLA-G 결합 scFv를 코딩하는 뉴클레오티드 서열 및 인간 면역글로불린(human immunoglobulin)의 단편 결정화 부위(Fc)를 코딩하는 뉴클레오티드 서열이 순차적으로 연결된 것일 수 있다 (도 1a 내지 1c).
또한, 상기 항-HLA-G 결합 scFv를 코딩하는 뉴클레오티드 서열은 중쇄 및 경쇄 가변영역을 코딩하는 뉴클레오티드 서열 사이에 링커 서열을 코딩하는 뉴클레오티드 서열이 포함된 것으로 링커 서열은 서열번호 13의 서열일 수 있다.
본 발명의 일 구체예에 따르면, 상기 신호 서열은 세포 내에서 발현된 항-HLA-G 결합 scFv를 세포 외부로 이동시켜 분비하는 역할을 한다. 펨브롤리주맙 (Pembrolizumab; 상품명 키트루다(Keytruda))과 같은 기존의 면역관문 억제 항체는 항체 자체를 체내에 투여하나, 본 발명은 면역세포를 투여해도 항-HLA-G 결합 scFv가 세포 외부로 분비되므로 면역세포와 scFv의 작용 효과를 모두 기대할 수 있다. 또한, 전신이 아닌 면역세포가 존재하는 암세포 부근에서만 항-HLA-G 결합 scFv가 발현 및 분비되므로 전신 부작용을 감소시킬 수 있다.
신호 서열을 코딩하는 뉴클레오티드 서열은 서열번호 8 내지 12로 이루어진 군에서 선택되는 아미노산 서열 또는 서열번호 8 내지 12로 이루어진 군에서 선택되는 아미노산 서열과 80% 이상, 바람직하게는 90% 이상, 더욱 바람직하게는 95% 이상, 가장 바람직하게는 99% 이상의 서열동일성을 가지는 아미노산 서열을 코딩하는 뉴클레오티드 서열일 수 있다. 본 발명의 실시예에서는 서열번호 8의 신호 서열을 사용하였다.
Fc 부위는 항-HLA-G 결합 scFv의 안정성 및 생물학적 활성을 유지시키며, Fc 부위를 코딩하는 뉴클레오티드 서열은 본원발명이 속하는 기술 분야에서 통상적으로 사용되는 서열일 수 있으며, 본 발명의 실시예에서는 서열번호 14의 서열을 사용하였다.
본 발명에서, 상기 항-HLA-G 결합 scFv는 항-HLA-G scFv, 항체단편 scFv 등으로도 지칭될 수 있다.
본 발명에서, 인간 백혈구 항원 G에 특이적으로 결합하는 가용성 scFv를 코딩하는 핵산을 함유하는 발현 벡터는 가용성 scFv 발현벡터로도 지칭될 수 있다.
본 발명에서 사용되는 벡터는 당 분야에 공지된 벡터를 다양하게 사용할 수 있고, 항원 인식 수용체 및/또는 가용성 scFv를 발현시키고자 하는 세포의 종류에 따라 프로모터 (promoter), 종결자 (terminator), 인핸서 (enhancer) 등과 같은 발현조절 서열, 막 표적화 또는 분비를 위한 서열 등을 적절히 선택하고 목적에 따라 다양하게 조합할 수 있다.
본 발명의 벡터를 세포 내로 도입하고 발현시키는 방법은 관련 기술분야에 잘 알려져 있다. 벡터는 당업계에 공지된 방법에 의해 숙주 세포, 예를 들어 포유동물, 박테리아, 효모, 또는 곤충 세포 내로 쉽게 도입될 수 있다. 예를 들면, 벡터는 물리적, 화학적, 또는 생물학적 수단에 의해 숙주 세포 내로 전달될 수 있다. 상기 물리적 수단은 인산칼슘 침전, 리포펙션, 입자 폭격(particle bombardment), 미세주입, 전기천공(electroporation) 등을 포함한다. 상기 화학적 수단은 콜로이드 분산액 시스템, 예컨대 거대분자 복합체, 나노 캡슐, 마이크로스피어, 비드, 및 수중유 에멀젼, 마이셀(micelle), 혼합된 마이셀, 및 리포좀을 포함하는 지질-기반 시스템을 포함한다. 또한, 상기 생물학적 수단은 상술한 렌티바이러스, 레트로바이러스 등, DNA 또는 RNA 벡터의 사용을 포함한다.
본 발명의 벡터는 플라스미드 벡터, 코즈미드 벡터, 박테리오 파아지 벡터 및 바이러스 벡터 등을 포함하나 이에 제한되지 않는다. 적합한 벡터는 프로모터, 오퍼레이터, 개시코돈, 종결코돈, 폴리아데닐화 시그널 및 인핸서 같은 발현 조절 요소 외에도 막 표적화 또는 분비를 위한 시그널 서열 또는 리더 서열을 포함하며 목적에 따라 다양하게 제조될 수 있다.
본 발명의 일 구체예에서는 렌티-바이러스 벡터를 사용하였다. 본 발명의 구체적인 일 구현예에 있어서, 상기 벡터는 프로모터를 추가로 포함한다. 상기 프로모터는 예컨대 RSV 프로모터일 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 또 다른 양태는 상기 항-HLA-G 결합 가용성 scFv를 코딩하는 뉴클레오티드 서열을 포함하는 단리된 핵산이다.
본 명세서에서 사용되는 용어, "핵산"은 DNA 및 RNA 분자를 포괄적으로 포함하는 의미를 가지며, 핵산 분자에서 기본 구성단위인 뉴클레오티드는 자연의 뉴클레오티드뿐만 아니라, 당 또는 염기 부위가 변형된 유사체(analogue)도 포함한다 (Scheit, Nucleotide Analogs, John Wiley, New York(1980); Uhlman 및 Peyman, Chemical Reviews(1990) 90:543-584).
본 발명의 항-HLA-G 결합 scFv를 코딩하는 뉴클레오티드 서열은 변형될 수 있으며, 상기 변형은 뉴클레오티드의 추가, 결실, 또는 비보존적 치환 또는 보존적 치환을 포함한다.
본 발명에 따른 가용성 scFv를 코딩하는 뉴클레오티드 서열을 포함하는 핵산(폴리뉴클레오티드)은 코돈 최적화에 의해 변형될 수 있으며, 이는 코돈의 축퇴성(degeneracy)에서 기인한 것으로, 폴리펩타이드 또는 이의 변이체 단편을 코딩하는 많은 뉴클레오티드 서열이 존재한다는 것은 통상의 기술자가 잘 이해할 수 있을 것이다. 이들 핵산의 일부는 임의의 자연 발생형 유전자의 뉴클레오티드 서열과 최소 상동성을 보유한다.
특히 코돈 활용법의 차이로 인해 가변적인 핵산, 예를 들어 인간, 영장류 및/또는 포유동물의 코돈 선택에 최적화된 핵산이 바람직하다.
본 발명에서, 상기 단리된 핵산은 하기 서열을 포함할 수 있다:
(a) 서열번호 21의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR1을 코딩하는 뉴클레오티드 서열, 서열번호 22의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR2를 코딩하는 뉴클레오티드 서열 및 서열번호 23의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR3을 코딩하는 뉴클레오티드 서열; 및

보다 구체적으로, 상기 단리된 핵산은 하기 서열을 포함할 수 있다:
(a) 서열번호 15의 아미노산 서열을 갖는 중쇄 가변 영역을 코딩하는 뉴클레오티드 서열 및 서열번호 16의 아미노산 서열을 갖는 경쇄 가변 영역을 코딩하는 뉴클레오티드 서열;
(b) 서열번호 17의 아미노산 서열을 갖는 중쇄 가변 영역을 코딩하는 뉴클레오티드 서열 및 서열번호 18의 아미노산 서열을 갖는 경쇄 가변 영역을 코딩하는 뉴클레오티드 서열; 또는
(c) 서열번호 19의 아미노산 서열을 갖는 중쇄 가변 영역을 코딩하는 뉴클레오티드 서열 및 서열번호 20의 아미노산 서열을 갖는 경쇄 가변 영역을 코딩하는 뉴클레오티드 서열.
또한, 본 발명의 일 구체예에 따르면, 상기 단리된 핵산은 중쇄 가변 영역을 코딩하는 뉴클레오티드 서열과 경쇄 가변 영역을 코딩하는 뉴클레오티드 서열 사이에 서열번호 13의 링커 서열을 코딩하는 뉴클레오티드 서열을 추가로 포함한다.
한편, 본 발명자들은 항-HLA-G 가용성 scFv 발현 면역세포에서 가용성 scFv가 세포 밖으로 잘 분비되고 (도 5 및 도 11), 가용성 scFv를 발현하는 HER2 CAR-T 세포는 HLA-G 발현 암세포주에 의해 면역세포의 기능 저하가 일어나지 않는 것을 확인하였다 (도 10).
즉, 본 발명의 면역세포는 HLA-G와 그 수용체의 상호작용을 저해할 수 있는 가용성 scFv를 항원 인식 수용체와 함께 동시 발현하여 항암 효과를 나타낼 수 있을 뿐만 아니라, 가용성 scFv를 단독 발현할 때에는 타겟(암세포)을 내인성 항원 인식 수용체로 인지하여 제거하는 과정에서 가용성 scFv가 항암 효능 증대에 도움을 줄 수 있다. 이러한 항암 효능 증대 효과는 종양침윤림프구와 같은 내인성 면역세포에도 작용할 수 있기 때문에 내인성 면역세포의 활성화도 추가적으로 유도할 수 있다.
따라서, 본 발명은 또한 전술한 본 발명의 면역세포를 포함하는 약제학적 조성물을 제공한다.
본 발명의 일 구체예에 따르면, 상기 약제학적 조성물은 암의 예방 또는 치료 용도로 사용될 수 있다.
본 발명에 있어서, "암"과 "종양"은 동일한 의미로 사용되며, 전형적으로 조절되지 않은 세포 성장/증식을 특징으로 하는 포유동물의 생리학적 상태를 지칭하거나 의미한다.
본 명세서에서 사용되는 용어, "예방"은 본 발명의 조성물의 투여로 질환을 억제시키거나 진행을 지연시키는 모든 행위를 의미하며, "치료"는 질환의 발전의 억제, 경감 또는 제거를 의미한다.
본 발명에서, 상기 암(또는 종양)의 예는 암종, 림프종(예를 들어, 호지킨 및 비-호지킨 림프종), 모세포종, 육종 및 백혈병을 포함하나, 이에 한정되지는 않는다. 보다 바람직한 암의 예는 췌장암, 유방암, 난소암, 신경교종, 자궁경부암, 자궁내막암, 식도암, 위암, 간암, 폐암, 대장암, 비인두암, 구강암, 갑상선암, 전립선암, 신장암, 흑색종, 담낭암, 담관암, 흑색종, 편평세포암, 소세포 폐암, 비소세포 폐암, 폐의 선암종, 폐의 편평세포 암종, 복막암, 간세포성암, 방광암, 간세포암, 결장암, 결장직장암, 타액선 암종, 외음부암, 중피종, 백혈병 및 다른 림프구증식성 장애, 및 다양한 유형의 두경부암을 포함한다.
보다 구체적으로 상기 약제학적 조성물은 HLA-G 발현 양성인 암의 치료에 사용될 수 있으며, 상기 암은 췌장암, 유방암, 난소암, 신경교종, 자궁경부암, 자궁내막암, 식도암, 위암, 간암, 폐암, 대장암, 비인두암, 구강암, 갑상선암, 전립선암, 신장암, 담낭암, 담관암 및 흑색종으로 이루어진 군으로부터 선택될 수 있다.
본 발명의 약제학적 조성물은 약제학적으로 허용되는 담체를 추가로 포함할 수 있으며, 상기 약제학적으로 허용되는 담체는 제제시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산칼슘, 알기네이트, 젤라틴, 규산칼슘, 미세결정성 셀룰로오스, 폴리비닐피롤리돈, 셀룰로오스, 물, 시럽, 메틸셀룰로오스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 암 예방 또는 치료용 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약제학적으로 허용되는 담체 및 제제는 문헌[Remington's Pharmaceutical Sciences(19th ed., 1995)]에 상세히 기재되어 있다.
본 발명의 약제학적 조성물은 경구 또는 비경구로 투여할 수 있고, 비경구 투여인 경우에는 정맥내 주입, 피하주입, 근육 주입, 복강 주입, 내피 투여, 국소 투여, 비내 투여, 폐내 투여 및 직장 내 투여 등으로 투여할 수 있다. 경구 투여시, 단백질 또는 펩타이드는 소화가 되기 때문에 경구용 조성물은 활성 약제를 코팅하거나 위에서의 분해로부터 보호되도록 제형화될 수 있으며, 본 발명의 조성물은 활성 물질이 표적 세포로 이동할 수 있는 임의의 장치에 의해 투여될 수 있다.
본 발명의 약제학적 조성물의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성별, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하며, 보통으로 숙련된 의사는 소망하는 치료 또는 예방에 효과적인 약제학적 유효량을 용이하게 결정 및 처방할 수 있다. 본 명세서에서 용어 "약제학적 유효량"은 질환의 치료, 예방 및 진단하는 데 충분한 양을 의미한다.
본 발명의 약제학적 조성물은 1회 투여량 내에 상기 면역세포(예컨대, T 세포)의 수가 치료 대상 내의 상기 종양세포, 예를 들어 췌장암 세포, 유방암 세포, 난소암 세포 또는 신경교종 세포 수의 1배 내지 10배로 포함된 것일 수 있다.
본 발명의 약제학적 조성물은 당해 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화하여 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질중의 용액, 현탁액 또는 유화액 형태이거나 엑스제, 산제, 좌제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.
본 발명의 약제학적 조성물은 개별 치료제로 투여되거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와 순차적 또는 동시에 투여될 수 있다.
본 발명의 또 다른 양태는 암의 예방 또는 치료용 약제학적 조성물의 제조를 위한 제1항 내지 제10항 중 어느 한 항에 따른 면역세포의 용도, 암의 예방 또는 치료를 제1항 내지 제10항 중 어느 한 항에 따른 면역세포의 용도를 제공한다.
본 발명은 또한 전술한 면역세포 또는 약제학적 조성물을 이를 필요로 하는 대상체에 투여하는 단계를 포함하는 암의 예방 또는 치료 방법을 제공한다.
상기 대상체는 암이 발병하였거나 발병할 수 있는 인간을 포함한 원숭이, 소, 말, 양, 돼지, 닭, 칠면조, 메추라기, 고양이, 개, 마우스, 쥐, 토끼 또는 기니아 피그를 포함한 모든 동물을 의미한다.
상기 암은 HLA-G가 과발현된 암일 수 있으나 이에 제한되지는 아니한다.
본 발명은 면역관문을 극복한 면역세포와 이를 이용한 약제학적 조성물을 제공한다. HLA-G는 다양한 고형암에서 면역관문으로 작용하여 종양침윤림프구의 정상적인 기능을 억제하는데, 본 발명에 따른 면역세포는 항-HLA-G 항체 유래의 가용성 scFv를 발현하여 분비하므로 면역 억제 반응을 극복하여 더 효율적으로 암세포를 제거할 수 있고, 내인성 면역세포의 활성화도 추가적으로 유도할 수 있기 때문에 HLA-G의 고발현과 관련된 다양한 암 등의 치료에 사용될 수 있다.

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예 1. 세포주 및 배양
인간 난소암 세포주 SK-OV-3와 인간 급성 백혈병 세포주 Jurkat은 한국세포주은행(Korea)으로부터 공급받아 사용하였다. HLA-G를 안정적으로 과발현하는 SK-OV-3-HLA-G 세포주와 HLA-G 수용체인 ILT2(Ig-like transcript 2)를 안정적으로 과발현하는 Jurkat-ILT2 세포주는 렌티바이러스 벡터(lentivral vector)를 이용하여 각각 SK-OV-3 또는 Jurkat 세포주로부터 제작하였다. SK-OV-3-HLA-G와 Jurkat-ILT2 세포주는 10% FBS(Gibco, USA)와 1% 항생제 (Antibiotic-Antimycotic;Gibco, USA), 및 0.5 ug/mL 퓨로마이신(puromycin; Sigma-aldrich, USA)을 포함하는 RPMI 1640(Gibco, USA) 배지에서 배양하였다.
실시예 2. 항-HLA-G 가용성 scFv 컨스트럭트 구축
본 발명의 항원 인식 수용체와 항-HLA-G 가용성 scFv 발현 컨스트럭트는 인공적으로 합성되었다. 좀 더 상세하게는, 항원 인식 수용체와 가용성 scFv는 자기 절단형 펩티드로 연결된 상태이며, 가용성 scFv는 인간 면역글로불린 중쇄 신호 서열, HLA-G 항체단편 scFv, 인간 면역글로불린 Fc 부위가 연결된 상태로 SOE-PCR(Splicing by overlap extension PCR)을 통해 인공적으로 합성되었다(도 1a 내지 1c).
PCR 산물을 MluI 및 XbaI으로 절단한 후 3세대 자가-불활성화 렌티바이러스 발현 벡터인 pLV-EF1A-MCS 벡터의 MluI 및 XbaI 사이트에 삽입하였다(도 2a 내지 2c).
내인성 항원 인식 수용체를 사용하는 면역세포를 위하여 항원 인식 수용체 없이 가용성 scFv만 발현하는 컨스트럭트 또한 제작하였다. 가용성 scFv는 인간 면역글로불린 중쇄 신호 서열, HLA-G 항체단편 scFv, 인간 면역글로불린 Fc 부위가 연결된 상태로 SOE-PCR을 통해 인공적으로 합성되었으며, PCR 산물을 MluI 및 XbaI으로 절단한 후 3세대 자가-불활성화 렌티바이러스 발현 벡터인 pLV-EF1A-MCS 벡터의 MluI 및 XbaI 사이트에 삽입하였다(도 3a 내지 3c).
본 발명의 실시예에 따른 항-HLA-G 가용성 scFv 및 그의 제조에 사용된 도메인들의 서열들의 정보를 표 1에 정리하였다. 가용성 scFv를 구성하는 각 부위는 서로 직렬로(in tandem) 연결되며, 또한 인프레임(in frame)으로 연결된다. 상세하게는, 신호 서열, 항체단편 scFv, Fc 부위와 종결 코돈(stop codon)인 TAA가 연결된 것이다.

실시예 3. 항-HLA-G 가용성 scFv를 발현하는 CAR-Jurkat 세포주의 제작
3-1. 항-HLA-G 가용성 scFv 유전자 발현 렌티바이러스의 제조
항원 인식 수용체의 예시로 HER2 CAR를 선정하였다. HER2 CAR와 항-HLA-G 가용성 scFv가 자기 절단형 펩티드를 통해 동시에 발현될 수 있도록 하였으며, 이들 유전자의 전달을 위한 렌티바이러스는 플라스미드 DNA 형질전환법으로 제작하였다. TransIT-293 Transfection Reagent(Mirus Bio LLC, USA)을 이용하였고, 제조사의 프로토콜에 따라 수행하였다. 실험 전날 100 mm 접시에 Lenti-XTM 293T 세포주(Clontech, USA)를 5x106개로 파종하고, 다음날 pLV-EF1A-항원 인식 수용체-가용성 scFv 렌티바이러스 벡터, gag-pol 발현 벡터, 및 VSV-G 외피 발현 벡터를 형질전환시켰다. 항원 인식 수용체-가용성 scFv 렌티바이러스 벡터는 다음과 같다; HER2 CAR, HER2 CAR-가용성 scFv 2, 또는 HER CAR-가용성 scFv 3. 형질전환 후 세포를 약 72시간 배양하고 배양 완료 후 세포 상층액을 모두 수확하였다. 상층액을 0.45 um 필터(Millipore, USA)로 여과하여 세포 조각을 제거하였다. Lenti-XTM Concentrator(Clontech, USA)를 이용하여 제조사의 프로토콜에 따라 렌티바이러스를 약 100배 농축하고 사용 전까지 -80℃에 냉동 보관하였다.
3-2. 항-HLA-G 가용성 scFv 발현 CAR-Jurkat 세포주 제작 및 배양
MOI(Multiplicity of infection, 감염 배수) 5 정도의 렌티바이러스와 함께 Jurkat-ILT2 세포주를 6 웰 플레이트에 웰 당 1x106개를 넣어주고 배양액이 2 mL이 되도록 하였다. 추가로 배지에 폴리브렌(polybrene)을 8 ug/mL 농도로 첨가하였다. 그리고 24시간 동안 정치 배양한 후 새로운 배지로 교체하였다. 형질도입된 Jurkat-ILT2 세포주는 2~3일 간격으로 mL당 2x105개로 계대 배양하여 유지하였다.
3-3. 항-HLA-G 가용성 scFv 발현 CAR-Jurkat 세포주에서 가용성 scFv의 분비 확인
앞서 제작한 항-HLA-G 가용성 scFv 발현 CAR-Jurkat 세포주에서 세포 표면에 HER CAR가 발현하는지 확인하였다. HER2 CAR와 가용성 scFv는 자기 절단형 펩티드로 연결되어 있기 때문에, CAR가 정상적으로 발현한다면 가용성 scFv 또한 정상적으로 발현함을 예측할 수 있다.
1x105개의 세포를 수확한 후 100 uL D-PBS에 부유시켜 세포 부유액을 준비하였다. 상기 세포 샘플에 Recombinant Protein L(PE conjugated)(Sino Biological, China)과 Human ILT2 APC-conjugated Antibody(R&D systems, USA)를 첨가하여 4℃에서 30분 동안 반응시켰다. 반응 종료 후 세포를 D-PBS로 2회 세척하고, 유세포분석법으로 HER2 CAR와 ILT2의 발현 수준을 확인하였다.
그 결과, 가용성 scFv를 발현하는 CAR-Jurkat 세포주에서 거의 모든 세포가 HER2 CAR를 발현하고 있었고, 그 중 절반 이상은 ILT2를 동시에 발현하고 있었다(도 4).
가용성 scFv는 세포 밖으로 분비되어 작용해야 하기 때문에, 가용성 scFv가 정상적으로 발현하여 세포 밖으로 분비되는지 확인하였다. 24 웰 플레이트에 8x105개의 세포를 파종한 다음, 다음날 배지를 모두 수확하여 MinuteTM High-Efficiency Protein Precipitation Kit(Invent Biotechnologies, USA)로 배지 내 단백질을 획득하였다. Western blotting을 통해 획득한 단백질을 Goat Anti-Human IgG Antibody, Fc, HRP conjugate(Sigma-aldrich, USA)로 검출하였다.
그 결과, 가용성 scFv를 발현하는 CAR-Jurkat 세포주에서 가용성 scFv가 잘 분비되고, 안정적으로 존재하고 있음을 확인하였다(도 5).
실시예 4. 생체 외(
in vitro
)에서 항-HLA-G 가용성 scFv 발현 CAR-Jurkat 세포주의 항암 효능 확인
항-HLA-G 가용성 scFv 발현 CAR-Jurkat 세포주의 항암 효능을 확인하기 위하여 SK-OV-3-HLA-G 세포주를 이용하였다. SK-OV-3-HLA-G 세포주는 HER2와 HLA-G가 모두 양성인 세포주로, HER2 CAR에 의해서 면역세포가 타깃 세포주에 대해 항암 효능을 나타낼 수 있으며, 반대로 HLA-G에 의해서는 면역세포가 타깃 세포주로 인해 그 효능이 억제될 수 있다. 항-HLA-G 가용성 scFv 발현 CAR-Jurkat 또는 대조군 Jurkat 세포주를 48 웰 플레이트에 웰 당 2x105 세포/150 uL로 첨가한 후, SK-OV-3-HLA-G 세포주를 2x105 세포/150 uL 첨가하여 48시간 반응시켰다.
세포 상층액을 수확하여, 수확한 상층액에서 Human IL-2 Quantikine ELISA Kit(R&D systems, USA)로 IL-2의 농도를 측정하였다. 그 결과, HER2 CAR만 발현하는 Jurkat 세포주 대비 항-HLA-G 가용성 scFv를 발현하는 HER2 CAR-Jurkat 세포주와 타깃 세포주를 공배양한 경우에 IL-2의 분비가 유의적으로 증가함을 확인하였다(도 6).
이와 동시에 공배양한 세포를 수확한 다음, Jurkat 세포주의 활성화 마커인 CD25를 CD25 Antibody, anti-human, PE-Vio® 770, REAfinityTM(Miltenyi Biotec, Germany)를 이용하여 유세포분석법으로 확인한 결과, HER2 CAR만 발현하는 Jurkat 세포주 대비 항-HLA-G 가용성 scFv를 발현하는 HER2 CAR-Jurkat 세포주와 타깃 세포주를 공배양한 경우에만 CD25의 세포 표면 발현율이 상당히 증가함을 확인하였다(도 7).
상기 결과들을 통해, 항-HLA-G 가용성 scFv를 발현하여 분비시키는 경우에 타깃 세포주가 발현하는 HLA-G로 인한 면역세포의 기능 저하가 다시 일정 부분 회복되는 것을 알 수 있었다.
실시예 5. 항-HLA-G 가용성 scFv를 발현하는 CAR-T 세포의 제작
5-1. ILT2 유전자 발현 렌티바이러스와 항-HLA-G 가용성 scFv 유전자 발현 렌티바이러스의 제조
ILT2 유전자와 항-HLA-G 가용성 scFv 유전자의 전달을 위한 렌티바이러스는 플라스미드 DNA 형질전환법으로 제작하였다. TransIT-293 Transfection Reagent(Mirus Bio LLC, USA)을 이용하였고, 제조사의 프로토콜에 따라 수행하였다. 실험 전날 100 mm 접시에 Lenti-XTM 293T 세포주(Clontech, USA)를 5x106개로 파종하고, 다음날 pLVX-puro-ILT2 렌티바이러스 벡터 또는 pLV-EF1A-항원 인식 수용체-가용성 scFv 렌티바이러스 벡터, gag-pol 발현 벡터, 및 VSV-G 외피 발현 벡터를 형질전환시켰다. 항원 인식 수용체-가용성 scFv 렌티바이러스 벡터는 다음과 같다; HER2 CAR, HER2 CAR-가용성 scFv 2, 또는 HER CAR-가용성 scFv 3. 형질전환 후 세포를 약 72시간 배양하고 배양 완료 후 세포 상층액을 모두 수확하였다. 상층액을 0.45 um 필터(Millipore, USA)로 여과하여 세포 조각을 제거하였다. 5-2. 항-HLA-G 가용성 scFv 발현 CAR-T 세포 제작 및 배양
공여자를 모집하고 백혈구성분채혈(leukapheresis)을 통해 백혈구를 얻었다. 상기 백혈구에서 SepMateTM-50(STEMCELL technology, Canada)과 Ficoll-Paque PLUS(GE healthcare, Sweden)를 이용하여 말초혈 단핵세포(peripheral blood mononuclear cell, PBMC)를 획득하였다. 그 후 QuadroMACSTM Separator(Miltenyi Biotec, Germany)와 LS 컬럼(Miltenyi Biotec, Germany), 인간 CD4 MicroBeads(Miltenyi Biotec, Germany) 및 인간 CD8 MicroBeads(Miltenyi Biotec, Germany)을 이용하여 양성 선택(positive selection) 방법으로 인간 T 세포를 분리하였다. 인간 T 세포는 TexMACSTM 배지(Miltenyi Biotec, Germany)를 배양액으로 사용하고, T Cell TransActTM, human(Miltenyi Biotec, Germany)을 첨가하여 활성화시켰다. T 세포의 성장을 위해 배양액에 인간 IL-2(R&D systems, USA)를 100 U/mL로 첨가하여 배양하였다. 24시간 배양 후 활성화된 T 세포를 수확하여 렌티바이러스 전달에 사용하였다.
ILT2 유전자와 항-HLA-G 가용성 scFv 유전자를 모두 전달하기 위해 하루의 간격을 두고 유전자 도입을 진행하였으며, 도입 효율을 증가시키기 위해 RetroNectin®(Takara, Japan)을 사용하였다. MOI 10 정도의 ILT2 유전자 발현 렌티바이러스를 RetroNectin이 코팅된 24 웰 플레이트에 넣고, 제조사의 프로토콜에 따라 렌티바이러스를 흡착시켰다. 이후 렌티바이러스가 흡착된 플레이트에 활성화된 인간 T 세포를 웰 당 5x105개를 넣어주고 배양액이 1.25 mL이 되도록 하였다. 그리고 1,000 xg로 15분간 원심분리하여 T 세포에 ILT2 유전자 발현 렌티바이러스를 전달하였다. 24시간 후 역시 같은 방식으로 항-HLA-G 가용성 scFv 유전자 발현 렌티바이러스를 T 세포에 전달하였다. 그 다음 T 세포를 48시간 동안 정치 배양한 후 새로운 배지로 교체하였다. 형질도입된 인간 T 세포는 2~3일 간격으로 mL당 5x105개로 계대 배양하고, 세포 수가 mL당 2x106개를 넘지 않도록 유지하였다. 특별한 언급이 없는 한, 인간 T 세포는 배양액에 IL-2(R&D systems, USA)를 100 U/mL로 첨가하여 배양하였다.
5-3. 항-HLA-G 가용성 scFv 발현 CAR-T 세포에서 가용성 scFv의 분비 확인
앞서 제작한 항-HLA-G 가용성 scFv 발현 CAR-T 세포에서 세포 표면에 HER CAR가 발현하는지 확인하였다. HER2 CAR와 가용성 scFv는 자기 절단형 펩티드로 연결되어 있기 때문에, CAR가 정상적으로 발현한다면 가용성 scFv 또한 정상적으로 발현함을 예측할 수 있다.
1x105개의 세포를 수확한 후 100 uL D-PBS에 부유시켜 세포 부유액을 준비하였다. 상기 세포 샘플에 Recombinant Protein L(PE conjugated)(Sino Biological, China)과 Human ILT2 APC-conjugated Antibody(R&D systems, USA)를 첨가하여 4℃에서 30분 동안 반응시켰다. 반응 종료 후 세포를 D-PBS로 2회 세척하고, 유세포분석법으로 HER2 CAR와 ILT2의 발현 수준을 확인하였다.
그 결과, HER2 CAR를 도입한 세포에서는 CAR 발현율이 35~45% 수준이었고, ILT2를 도입한 세포에서는 ILT2 발현율이 20~30% 수준이었다(도 8).
가용성 scFv는 세포 밖으로 분비되어 작용해야 하기 때문에, 가용성 scFv가 정상적으로 발현하여 세포 밖으로 분비되는지 확인하였다. 24 웰 플레이트에 8x105개의 세포를 파종한 다음, 다음날 배지를 모두 수확하여 MinuteTM High-Efficiency Protein Precipitation Kit(Invent Biotechnologies, USA)로 배지 내 단백질을 획득하였다. 실시예 6. 생체 외(
in vitro
)에서 항-HLA-G 가용성 scFv 발현 CAR-T 세포의 항암 효능 확인
항-HLA-G 가용성 scFv 발현 CAR-T 세포의 항암 효능을 확인하기 위하여 SK-OV-3-HLA-G 세포주를 이용하였다. SK-OV-3-HLA-G 세포주는 HER2와 HLA-G가 모두 양성인 세포주로, HER2 CAR에 의해서 면역세포가 타깃 세포주에 대해 항암 효능을 나타낼 수 있으며, 반대로 HLA-G에 의해서는 면역세포가 타깃 세포주로 인해 그 효능이 억제될 수 있다. SK-OV-3-HLA-G 세포주(타겟 세포=T)를 96 웰 플레이트에 웰 당 1x104 세포/150 uL 첨가한 다음, 24시간 후에 항-HLA-G 가용성 scFv 발현 CAR-T 또는 대조군 T 세포(효과기 세포=E)를 웰 당 5x104 세포/70 uL 또는 3x104 세포/70 uL(E:T 비율=5:1 또는 3:1)로 첨가하여 48시간 반응시켰다.
세포 상층액을 수확하여, 수확한 상층액에서 Human IL-2 Quantikine ELISA Kit(R&D systems, USA)로 IL-2의 농도를 측정하였다. 그 결과, E:T 비율 5와 3 모두의 경우에서 HER2 CAR만 발현하는 CAR-T 세포 대비 ILT2를 추가로 발현하는 HER2 CAR-T-ILT2 세포의 IL-2의 분비가 감소하였으며, 이러한 감소는 항-HLA-G 가용성 scFv를 발현하는 HER2 CAR-T 세포에서 다시 유의적으로 회복됨을 확인하였다(도 10; 도 10a-E:T=5:1, 도 10b-E:T=3:1).
상기 결과들을 통해, 항-HLA-G 가용성 scFv를 발현하여 분비시키는 경우에 타깃 세포주가 발현하는 HLA-G로 인한 면역세포의 기능 저하가 다시 일정 부분 회복되는 것을 알 수 있었다.
실시예 7. 항-HLA-G 가용성 scFv를 발현하는 Jurkat 세포주의 제작
7-1. 항-HLA-G 가용성 scFv 유전자 발현 렌티바이러스의 제조
CAR와 같은 외인성 항원 인식 수용체 없이 내인성 항원 인식 수용체를 사용하는 면역세포를 위하여 제작한 항-HLA-G 가용성 scFv 단독 발현 컨스트럭트를 사용하여, 이들 유전자의 전달을 위한 렌티바이러스는 플라스미드 DNA 형질전환법으로 제작하였다. TransIT-293 Transfection Reagent(Mirus Bio LLC, USA)을 이용하였고, 제조사의 프로토콜에 따라 수행하였다. 실험 전날 100 mm 접시에 Lenti-XTM 293T 세포주(Clontech, USA)를 5x106개로 파종하고, 다음날 pLV-EF1A-가용성 scFv 렌티바이러스 벡터, gag-pol 발현 벡터, 및 VSV-G 외피 발현 벡터를 형질전환시켰다. 가용성 scFv 렌티바이러스 벡터는 다음과 같다; 가용성 scFv 2, 또는 가용성 scFv 3. 형질전환 후 세포를 약 72시간 배양하고 배양 완료 후 세포 상층액을 모두 수확하였다. 상층액을 0.45 um 필터(Millipore, USA)로 여과하여 세포 조각을 제거하였다. Lenti-XTM Concentrator(Clontech, USA)를 이용하여 제조사의 프로토콜에 따라 렌티바이러스를 약 100배 농축하고 사용 전까지 -80℃에 냉동 보관하였다.
7-2. 항-HLA-G 가용성 scFv 발현 Jurkat 세포주 제작 및 배양
MOI(Multiplicity of infection, 감염 배수) 5 정도의 렌티바이러스와 함께 Jurkat-ILT2 세포주를 6 웰 플레이트에 웰 당 1x106개를 넣어주고 배양액이 2 mL이 되도록 하였다. 추가로 배지에 폴리브렌(polybrene)을 8 ug/mL 농도로 첨가하였다. 그리고 24시간 동안 정치 배양한 후 새로운 배지로 교체하였다. 형질도입된 Jurkat-ILT2 세포주는 2~3일 간격으로 mL당 2x105개로 계대 배양하여 유지하였다.
7-3. 항-HLA-G 가용성 scFv 발현 Jurkat 세포주에서 가용성 scFv의 분비 확인
가용성 scFv는 세포 밖으로 분비되어 작용해야 하기 때문에, 가용성 scFv가 정상적으로 발현하여 세포 밖으로 분비되는지 확인하였다. 24 웰 플레이트에 8x105개의 세포를 파종한 다음, 다음날 배지를 모두 수확하여 MinuteTM High-Efficiency Protein Precipitation Kit(Invent Biotechnologies, USA)로 배지 내 단백질을 획득하였다. Western blotting을 통해 획득한 단백질을 Goat Anti-Human IgG Antibody, Fc, HRP conjugate(Sigma-aldrich, USA)로 검출하였다.
그 결과, 가용성 scFv를 발현하는 Jurkat 세포주에서 가용성 scFv가 잘 분비되고, 안정적으로 존재하고 있음을 확인하였다(도 11).
Claims (22)
- 항원 인식 수용체; 및인간 백혈구 항원 G (human leukocyte antigen G, HLA-G) 및 그의 수용체로 이루어진 군에서 선택되는 표적에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편;을 발현 및 분비하는, 면역세포.
- 인간 백혈구 항원 G(HLA-G) 및 그의 수용체로 이루어진 군에서 선택되는 표적에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 발현 및 분비하는 면역세포.
- 제1항 또는 제2항에 있어서, 상기 인간 백혈구 항원 G에 특이적으로 결합하는 항체 또는 항원 결합 단편은 하기에서 선택되는 서열을 포함하는, 면역세포:(a) 서열번호 21의 아미노산 서열을 갖는 중쇄 상보성 결정 부위 1 (complementarity determining region 1, CDR1), 서열번호 22의 아미노산 서열을 갖는 중쇄 CDR2 및 서열번호 23의 아미노산 서열을 갖는 중쇄 CDR3를 포함하는 중쇄 가변 영역; 및서열번호 24의 아미노산 서열을 갖는 경쇄 CDR1, 서열번호 25의 아미노산 서열을 갖는 경쇄 CDR2 및 서열번호 26의 아미노산 서열을 갖는 경쇄 CDR3를 포함하는 경쇄 가변 영역;(b) 서열번호 27의 아미노산 서열을 갖는 중쇄 CDR1, 서열번호 28의 아미노산 서열을 갖는 중쇄 CDR2 및 서열번호 29의 아미노산 서열을 갖는 중쇄 CDR3를 포함하는 중쇄 가변 영역; 및서열번호 30의 아미노산 서열을 갖는 경쇄 CDR1, 서열번호 31의 아미노산 서열을 갖는 경쇄 CDR2 및 서열번호 32의 아미노산 서열을 갖는 경쇄 CDR3를 포함하는 경쇄 가변 영역; 또는(c) 서열번호 33의 아미노산 서열을 갖는 중쇄 CDR1, 서열번호 34의 아미노산 서열을 갖는 중쇄 CDR2 및 서열번호 35의 아미노산 서열을 갖는 중쇄 CDR3를 포함하는 중쇄 가변 영역; 및서열번호 36의 아미노산 서열을 갖는 경쇄 CDR1, 서열번호 37의 아미노산 서열을 갖는 경쇄 CDR2 및 서열번호 38의 아미노산 서열을 갖는 경쇄 CDR3를 포함하는 경쇄 가변 영역.
- 제1항 또는 제2항에 있어서, 상기 인간 백혈구 항원 G에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편은 단일쇄 Fv(single chain Fv, scFv)인, 면역세포.
- 제4항에 있어서, 상기 단일쇄 Fv는 면역세포로부터 분비되는 것인, 면역세포.
- 제4항에 있어서, 상기 단일쇄 Fv는 상기 면역세포 및 종양침윤림프구의 면역 반응을 향상시키는, 면역세포.
- 제1항 또는 제2항에 있어서, 상기 면역세포는 자연 살해 세포, T 림프구, B 림프구, 대식세포, 수지상 세포, 자연 살해 수지상 세포, 비만세포 및 이들의 전구세포로 이루어진 군으로부터 선택되는, 면역세포.
- 제1항에 있어서, 상기 항원 인식 수용체는 외인성 또는 내인성 항원 인식 수용체인, 면역세포.
- 제1항에 있어서, 상기 항원 인식 수용체는 키메라 항원 수용체(chimeric antigen receptor, CAR) 또는 T 세포 수용체(T-cell receptor, TCR)인, 면역세포.
- 제1항에 있어서, 상기 항원 인식 수용체는 CD19, CD20, CD22, CD7, CD10, CD30, CD33, CD34, CD38, CD41, CD44, CD49f, CD56, CD74, CD123, CD133, CD138, CD171, EGFR (epidermal growth factor receptor), PSMA (prostate specific membrane antigen), GD2, EGFR 변이체, ROR1(Tyrosine-protein kinase transmembrane receptor 1), c-Met, HER2 (human epidermal growth factor receptor-2), CEA(Carcino embryonic antigen), 메소텔린(mesothelin), GM2, MUC16, MUC1, CS1(CD319), 인터루킨 13 수용체 알파2 (IL-13R a2), BCMA(B-cell maturation antigen), LewisY, IgG 카파 사슬, 엽산 수용체-알파(Folate receptor-alpha), PSCA(Prostate stem cell antigen), EpCAM(Epithelial cell adhesion molecule), NY-ES0-1(New York esophageal squamous cell carcinoma 1), WT-1(Wilms' tumor gene1), MAGE-A1 (MAGE Family Member A1), TAG-72(Tumor associated glycoprotein-72), NKG2D(KLRK1 (killer cell lectin like receptor K1)), 클라우딘 18.2(Claudin 18.2), 클라우딘 3, 클라우딘 4 및 클라우딘 6으로 이루어진 군으로부터 선택되는 항원을 인식하는 것인, 면역세포.
- 제1항 또는 제2항에 따른 면역세포를 제조하기 위한 이하의 (a)~(c) 중 어느 하나의 발현 벡터:(a) 항원 인식 수용체를 코딩하는 뉴클레오티드 서열 및 인간백혈구 항원 G 및 그의 수용체로 이루어진 군에서 선택되는 표적에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 코딩하는 뉴클레오티드 서열을 함유하는 발현 벡터;(b) 인간백혈구 항원 G 및 그의 수용체로 이루어진 군에서 선택되는 표적에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 코딩하는 뉴클레오티드 서열을 함유하는 발현 벡터; 및(c) 이하의 (c-1) 및 (c-2)의 2개의 발현 벡터:(c-1) 항원 인식 수용체를 코딩하는 뉴클레오티드 서열을 함유하는 발현 벡터; 및(c-2) 인간백혈구 항원 G 및 그의 수용체로 이루어진 군에서 선택되는 표적에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 코딩하는 뉴클레오티드 서열을 함유하는 발현 벡터.
- 제11항에 있어서, 상기 (a)의 발현 벡터는, 항원 인식 수용체를 코딩하는 뉴클레오티드 서열 및 인간 백혈구 항원 G 및 그의 수용체로 이루어진 군에서 선택되는 표적에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 코딩하는 뉴클레오티드 서열이 자기 절단형 펩티드 코딩 뉴클레오티드 서열을 통해 연결되어 있는 것인, 발현 벡터.
- 제11항 또는 제12항에 있어서, 상기 인간 백혈구 항원 G에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 코딩하는 뉴클레오티드 서열은 신호 서열을 코딩하는 뉴클레오티드 서열, 항-HLA-G 결합 scFv를 코딩하는 뉴클레오티드 서열 및 Fc 부위를 코딩하는 뉴클레오티드 서열이 연결되어 있는 것인, 발현 벡터.
- 제11항에 있어서, 상기 인간 백혈구 항원 G에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 코딩하는 뉴클레오티드 서열은,(a) 서열번호 21, 27 또는 33의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR1을 코딩하는 뉴클레오티드 서열; 서열번호 22, 28 또는 34의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR2를 코딩하는 뉴클레오티드 서열; 및 서열번호 23, 29 또는 35의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR3을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 가변영역 코딩 뉴클레오티드 서열; 및(b) 서열번호 24, 30 또는 36의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR1을 코딩하는 뉴클레오티드 서열; 서열번호 25, 31 또는 37의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR2를 코딩하는 뉴클레오티드 서열; 및 서열번호 26, 32 또는 38의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR3을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 가변영역 코딩 뉴클레오티드 서열;을 포함하는 것인, 발현 벡터.
- 제14항에 있어서, 상기 인간 백혈구 항원 G에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 코딩하는 뉴클레오티드 서열은(a) 서열번호 21의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR1을 코딩하는 뉴클레오티드 서열, 서열번호 22의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR2를 코딩하는 뉴클레오티드 서열 및 서열번호 23의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR3을 코딩하는 뉴클레오티드 서열; 및서열번호 24의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR1을 코딩하는 뉴클레오티드 서열, 서열번호 25의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR2를 코딩하는 뉴클레오티드 서열 및 서열번호 26의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR3을 코딩하는 뉴클레오티드 서열;(b) 서열번호 27의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR1을 코딩하는 뉴클레오티드 서열, 서열번호 28의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR2를 코딩하는 뉴클레오티드 서열 및 서열번호 29의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR3을 코딩하는 뉴클레오티드 서열; 및서열번호 30의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR1을 코딩하는 뉴클레오티드 서열, 서열번호 31의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR2를 코딩하는 뉴클레오티드 서열 및 서열번호 32의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR3을 코딩하는 뉴클레오티드 서열; 또는(c) 서열번호 33의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR1을 코딩하는 뉴클레오티드 서열, 서열번호 34의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR2를 코딩하는 뉴클레오티드 서열 및 서열번호 35의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 중쇄 CDR3을 코딩하는 뉴클레오티드 서열; 및서열번호 36의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR1을 코딩하는 뉴클레오티드 서열, 서열번호 37의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR2를 코딩하는 뉴클레오티드 서열 및 서열번호 38의 아미노산 서열을 코딩하는 뉴클레오티드 서열을 포함하는 경쇄 CDR3을 코딩하는 뉴클레오티드 서열;을 포함하는 것인, 발현 벡터.
- 제1항 내지 제10항 중 어느 한 항에 따른 면역세포를 포함하는 약제학적 조성물.
- 제16항에 있어서, 상기 약제학적 조성물은 암의 예방 또는 치료를 위한 것인, 약제학적 조성물.
- 제17항에 있어서, 상기 암은 췌장암, 유방암, 난소암, 신경교종, 자궁경부암, 자궁내막암, 식도암, 위암, 간암, 폐암, 대장암, 비인두암, 구강암, 갑상선암, 전립선암, 신장암, 담낭암, 담관암, 흑색종으로 이루어진 군으로부터 선택되는, 약제학적 조성물.
- 제16항 내지 제18항 중 어느 한 항에 있어서, 약제학적으로 허용되는 담체를 추가로 포함하는, 약제학적 조성물.
