タンデムダイアボディ
Reusch et al.,2014,mAbs 6:3,728−739に記載されているようにタンデムダイアボディ(図3)を構築する。タンデムダイアボディを構築するために、抗EGFR Fvドメイン(配列番号1、2)を抗CD16A Fvドメイン(配列番号12、13)と組み合わせる。抗EGFRドメインおよび抗CD16AドメインがVH_EGFR−L1−VL_CD16A−L2−VH_CD16A−L3−VL_EGFRの順に配置されるように、タンデムダイアボディ用の発現カセットをクローニングする。リンカーL1およびL3には9アミノ酸リンカー(G2S)3(配列番号36)を使用し、リンカーL2には6アミノ酸リンカー(G2S)2(配列番号35)を使用する。得られたEGFR/CD16Aタンデムダイアボディは、配列番号27に示されるアミノ酸配列を有する2つのポリペプチドからなる。
国際公開第2017/064221号パンフレットに記載されているようにaTriFlex(図4)を構築する。aTriFlexを構築するために、抗EGFR Fvドメイン(配列番号1、2)を抗CD16A Fvドメイン(配列番号12、13)および抗HSA Fvドメイン(配列番号31、32)と組み合わせる。抗EGFRドメインおよび抗CD16Aドメインが、第1のポリペプチドではVH_EGFR−L1−VL_EGFR−L2−VL_CD16A−L2−VL_CD16A−L2−VH_HSA−L1−VL_HSA、および第2のポリペプチドではVH(CD16A)−L2−VH(CD16A)順序の順に配置されるように、aTriFlex用の発現カセットをクローニングし、リンカーL1には18アミノ酸リンカー(G2S)6(配列番号18)を使用し、リンカーL2には9アミノ酸リンカー(G2S)3(配列番号36)を使用する。
CHO細胞内のBi−scFv−Fc抗原結合タンパク質(図2)の発現のために、哺乳動物発現ベクター系に分子のコード配列をクローニングした。要約すると、Thermo Fisher Scientific GeneArt(Regensburg,Germany)により、ペプチドリンカーによって接続された抗EGFR Fvドメイン(配列番号1、2)および抗CD16A Fvドメイン(配列番号12、13)をコードする遺伝子配列を合成した。対応するプライマーを用いて、様々な可変ドメインのPCRアンプリコンと、サイレンシング点変異を含むFc部分(配列番号20)のPCRアンプリコンとを生成した。その後、様々なオーバーラップしたDNA断片と線形化されたバックボーンベクターとを1つの等温反応で組み合わせる。それぞれ抗体の分泌および精製を容易にするために、N末端シグナルペプチドおよびFc部分のコード配列を含むようにBi−scFv−Fc発現構築物を設計した。プライマー対5´−TAATACGACTCACTATAGGG−3´(配列番号33)および5´−TAGAAGGCACAGTCGAGG−3´(配列番号34)を使用して、GATC(Koln,Germany)でのDNA配列決定により構築物の配列を確認した。抗EGFRドメインおよび抗CD16AドメインがVL_CD16A−L1−VH_CD16A−ヒンジ−CH2−CH3−L2−VH_EGFR−L3−VL_EGFRの順に配置されるように、Bi−scFv−Fc用の発現カセットをクローニングし、リンカーL1には(G2S)7を使用し、リンカーL2には(G4S)2を使用し、リンカーL3には(G2S)6を使用する。得られたBi−scFv−Fc−_02は、配列番号30に示されるアミノ酸配列を有する2つのポリペプチドからなる。
同じベクターからの共発現のために、Fcサイレンシング点変異(配列番号15、16)を有する重鎖定常ドメインおよび軽鎖定常ドメインを含むCMV制御発現カセットを含む改変された哺乳動物発現ベクターに、抗CD16A Fvドメインのコード配列(配列番号12、13)をクローニングすることによって、scFv−IgAbをコードするDNA発現構築物を生成する。その後、対応するプライマーを用いて、配列番号18(VH−(G2S)6−VL)に示されるアミノ酸配列を有するペプチドリンカーにより分離された抗EGFR Fvドメイン(配列番号1、2)をコードする遺伝子配列からPCRアンプリコンを生成する。結果として生じるオーバーラップしたDNA断片を、関連する位置で共発現ベクターに挿入する。Fcサイレンシング点変異を含む可変ドメインおよび定常ドメインをコードする必要な遺伝子配列はいずれも、Thermo Fisher Scientific GeneArt(Regensburg,Germany)によって合成された。それぞれ抗体の分泌および精製を容易にするために、N末端シグナルペプチドおよびFc部分のコード配列を含むようにscFv−IgAb発現構築物を設計した。カスタムメイドプライマーを使用して、GATC(Koln,Germany)でのDNA配列決定により全構築物の配列を確認した。抗EGFRドメイン、抗CD16Aドメインおよび定常ドメインが、第1のポリペプチドではVH_CD16A−CH1−ヒンジ−CH2−CH3−L1−VH_EGFR−L2−VL_EGFR、および第2のポリペプチドではVL_CD16A−CLambdaの順に配置されるように、scFv−IgAb用の発現カセットをクローニングする。リンカーL1には(G4S)2(配列番号35)を使用し、リンカーL2には(G2S)6(配列番号18)を使用する。得られたscFv−IgAb_02は、配列番号29に示されるアミノ酸配列を有する軽鎖と会合した、配列番号28に示されるアミノ酸配列を有する重鎖からなる。
L−グルタミン、10%FCSおよび100μg/mlゼオシンを補充したHam’s F−12 Nutrient Mix中で、Flp−In CHO細胞(Life Technologies)、CHO−K1チャイニーズハムスター卵巣細胞(ATCC、CCL−61)の派生物(Kao and Puck,1968)を培養した。0.25%トリプシン−EDTAを用いて付着細胞を分離し、Life Technologiesによって提供される標準的な細胞培養プロトコルに従って継代培養した。
懸濁液に適応させた宿主細胞のトランスフェクションによって、分泌された組換え抗体、Fc融合構築物またはコンパレーター抗体および膜係留抗原を安定に発現する組換えFlp−In CHO細胞株を生成した。このために、ポリエチレンイミン(PEI)を使用して、目的のタンパク質(pcDNA5−FRT)をコードする発現プラスミド(2.5μg)と、Flpリコンビナーゼ(pOG44、Life Technologies)とを共トランスフェクションする1日前に、ゼオシン不含標準培地に細胞を入れた。要約すると、DNA:PEI比1:3(μg/μg)で合計100μLのOptiMEM I培地中でベクターDNAとトランスフェクション試薬とを混合し、10分間インキュベートしてから、1mlのCHO−S−SFMII培地(Life Technologies)に懸濁した2E+6 Flp−In CHO細胞に加えた。24〜48時間のインキュベーション後、CHO−S−SFMII培地中で培養物を0.1E+6生細胞/mLの密度に希釈した後、6〜7μg/mLピューロマイシン二塩酸塩を加えて、安定にトランスフェクトされた細胞の選択を開始した。Flpリコンビナーゼは、部位特異的DNA組換えを介して、組み込まれたFRT部位でのゲノムへのFlp−In発現構築物の挿入を媒介する(O’ Gorman et al 1991)。選択中に、生細胞密度を週に2回測定し、細胞を遠心分離し、最大密度0.1E+6生細胞/mLで、新鮮な選択培地に再懸濁した。選択の2〜3週間後に、組換えタンパク質産物を安定に発現する細胞プールを回収し、その時点で、振盪フラスコ内の標準的な培養培地に細胞を移した。Criterion Stain−Free(Bio−Rad)技術(以下を参照)またはフローサイトメトリーをそれぞれ使用した細胞培養上清のタンパク質ゲル電気泳動によって、組換え分泌タンパク質または膜係留タンパク質の発現を確認した。7.5%DMSOを含有する培地中で、安定な細胞プールを凍結保存した。
細胞培養上清への分泌により、安定にトランスフェクトされたCHO細胞の10日間または11日間の流加培養で、組換えタンパク質を作製した。このために、ガス透過性キャップを備えたポリカーボネートErlenmeyerフラスコ(Corning)内の標準的な培養培地に、6E+5細胞/mLの開始密度で組換え抗体、Fc融合抗原またはコンパレーター抗体を安定に発現する細胞を播種し、140rpmで撹拌しながら37℃および5%CO2でインキュベートした。流加培養中、培地には、40mL/L ActiCHO Feed A(GE Healthcare)および4mL/L ActiCHO Feed B(GE Healthcare)を0日目(開始日)に、これらを2倍の量で3日目、5日目および7日目に補充した。典型的に>75%の培養生存率で10日後または11日後に細胞培養上清を収集した。フィーディング前に1日おきに生成培養物から試料を収集し、細胞密度および生存率を評価した。収集日に、遠心分離と、Millipore Express PLUSメンブレンフィルター(Millipore)を使用した真空濾過(0.22μm)とにより細胞培養上清を除去し、その後使用した。
生成培養物の5日目、7日目および10日目または11日目のSDS−PAGEにより、細胞培養上清(CSS)のタンパク質発現力価および生成物完全性を分析する。4〜20%Criterion TGX Precast SDS PAGEゲル(Biorad)にロードする前に、試料とSDS PAGE試料緩衝液とを混合する。Criterion Stain−free Molecularイメージングシステム(Biorad)を使用して、ゲル中の総タンパク質を可視化する。既知の濃度の参照抗体と比較することにより、生成物の力価を半定量的に決定する。
プロテインAおよび分取SECを含む2段階手順で、清澄化したCHO細胞培養上清から抗EGFR抗原結合タンパク質を精製した。プロテインAの場合、清澄化した上清をHiTrap MabSelectSuReカラムにロードした。リン酸緩衝食塩水pH7.4および10mMリン酸ナトリウムpH7.0で洗浄した後、50mM酢酸ナトリウムpH3.5および10mMグリシン/HCL pH2.0を用いた2段階勾配でタンパク質を溶出した。SE−HPLCおよびSDS−PAGEを使用して画分の純度を分析した。許容可能な純度を示す画分をプールし、Superdex 200 prep gradeカラムを使用した分取ゲル濾過に供した。精製した抗EGFR抗原結合タンパク質を含有する溶出画分をプールし、10mM酢酸ナトリウム、4.5%ソルビトールpH5.0に対してSephadex G−25カラムを使用した緩衝液交換に供し、限外濾過により約1mg/mLの典型的な濃度に濃縮した。還元および非還元条件下でのSDS−PAGEによって、最終試料の均一性(scFv−IgAb_2約79%およびBi−scFv−Fc_2約85%)を評価した(図5を参照)。非還元2x SDS−PAGE試料緩衝液、または還元剤としてジチオスレイトール(DTT)を含有する還元2x SDS−PAGE試料緩衝液と試料を混合した。4〜20%のCriterion TGX Precast SDS Pageゲルにロードする前に、全試料を95℃で5分間加熱した。1レーン当たり2μgの精製タンパク質試料を使用した。ゲル中のタンパク質を分離するために、SDS−PAGEを1x Tris/グリシン/SDS緩衝液で300Vで約22分間実行した。Criterion Stain−free Molecularイメージングシステム(BioradBio−Rad)を使用して、ゲル中の総タンパク質を可視化した。分子量マーカーとしてPage Ruler Unstained Protein ladderを使用した。Superdex 200 Increase 10/300GLカラムを使用した分析用SE−HPLCにより、純度(scFv−IgAb_2約99%およびBi−scFv−Fc_2約97%)を評価した。精製タンパク質は、その後使用するまでアリコートとして−80℃で保存した。
ELISAにおける結合の分析
100mMカーボナート−バイカーボナート緩衝液中の組換え抗原または抗体により、96ウェルELISAプレート(Immuno MaxiSorp;Nunc)を4℃で一晩コーティングした。EGFR−mFc抗原は2.5μg/mL、EGFR−Fcは3μg/mL、CD16A−Fcは1.5μg/ml、EGFR/CD16A scFv−IgAbは3.8μg/ml、またはEGFR/CD16A Bi−scFv−Fcは3μg/mLの濃度でコーティングした。PBSに溶解した3%(w/v)脱脂粉乳(Merck)を用いたブロッキング工程の後、0.3%(w/v)脱脂粉乳を含有するPBS中の様々な抗体または可溶性抗原の連続希釈液をプレート上で室温で1.5時間インキュベートした。0.1%(v/v)Tween 20を含有するPBSを1ウェル当たり300μL用いて3回洗浄した後、プレートを検出抗体と室温で1時間インキュベートした。Hisタグ付き検体の検出には、Penta−HIS−HRP(Qiagen)を1:3000希釈で使用した。EGFR/CD16A scFv−IgAbまたはEGFR/CD16A Bi−scFv−FcがEGFR−mFc抗原に結合していることを検出するために、ビオチン化CD16−Fcをプレート上で1μg/mLで1時間、室温でインキュベートした後、洗浄し、ストレプトアビジン−HRPコンジュゲート(Roche)と1:10000希釈で、室温で1時間インキュベートした。0.1%(v/v)Tween 20を含有するPBSを1ウェル当たり300μL用いて3回洗浄した後、発色がはっきりと見えるまで、プレートをテトラメチルベンジジン(TMB)基質(Seramun)とインキュベートした。1ウェル当たり100μLの0.5M H2SO4を加えて、反応を停止した。マルチラベルプレートリーダー(Victor、Perkin Elmer)を使用して、450nmで吸光度を測定した。吸光度値をプロットし、非線形回帰、シグモイド型用量反応(可変勾配)、GraphPad Prismバージョン6.07による最小二乗(通常)適合(GraphPad Software,La Jolla California USA)を使用して分析した。
EGFR/CD16A抗原結合タンパク質中のサイレンシングされたFcの特性評価のために、表面プラズモン共鳴分光法(SPR)を使用して、様々なFcγ受容体およびFcRn(pH6.0)に対する結合を測定した。
リガンド分子
モノFc(mFc)およびAvi−Tagに融合した様々なFcγ受容体(ヒト、イヌ、カニクイザル、マウス)をCHOに発現させ、プロテインAおよびサイズ排除クロマトグラフィー(SEC)により精製した。ビオチン−タンパク質リガーゼ/BirAキット(GeneCopoeia)を使用して、Avi−Tagで分子をビオチン化した。

HBS−P+を使用して、様々なFcγ受容体に対するEGFR/CD16A抗原結合タンパク質の結合をBiacore T200機器上で25℃で測定した。
ランニング緩衝液として、および希釈のために(1×Gibco PBS、0.005% Tween20、4M HClを使用してpH6.0まで滴定)、pH6.0のPBS−T緩衝液を使用して、25℃で、Biacore T200機器上で、FcRn(ヒト、カニクイザル、マウス)に対するEGFR/CD16A抗原結合タンパク質の結合を測定した。この目的のために、Biotin CAPtureキット(GE Healthcare)を使用してMulti Cycle Kinetic実験を行った。センサー表面を活性化するために、フローセルFc 1〜4にBiotin CAPture試薬(GE Healthcare)を注入し(100秒、5μL/分)、2100RU〜2800RUの反応をもたらした。フローセルFc 2に様々な種のビオチン化FcRnを注入し(5μL/分)、約8〜約15RUの反応をもたらした。ランニング緩衝液として、および希釈のために、EGFR/CD16A抗原結合タンパク質の希釈系列を(pH7.4)に入れた。この目的のために、Biotin CAPtureキット(GE Healthcare)を使用して、Single Cycle Kinetic実験を行った。センサー表面を活性化するために、フローセルFc 1〜4にBiotin CAPture試薬(GE Healthcare)を注入し(100秒、5μL/分)、2100RU〜2800RUの反応をもたらした。フローセルFc 2、Fc 3、Fc 4)内にビオチン化Fcγ受容体を捕捉した(35RU〜55RU)。Single Cycle Kineticモード(30μL/分、会合180秒、解離240秒、6000nM〜1.47nM希釈1:4)で、フローセルFc 1〜4に抗EGFR抗体構築物の希釈系列を注入した。再生溶液(GE Healthcare)を使用してチップを再生した(10μL/分、フローセル1〜4、120秒)。ゼロ濃度サイクルの減算、および参照チャネルFc 1(Fc 2−1、Fc 3−1、Fc 4−1)内のシグナルの減算によってセンサーグラムを参照する。フローセルFc 1、2(30μL/分、会合240秒、解離100秒、3000nM〜12.5nM希釈1:3)に導入したBiacore T200評価ソフトウェアを使用して、1:1結合モデル(0に対するRI定数)にデータを適合させることによって結合速度を評価した。再生溶液(GE Healthcare)を使用してチップを再生した(10μL/分、フローセル1〜4、120秒)。ゼロ濃度サイクルの減算、および参照チャネルFc 1(Fc 2−1)内のシグナルの減算によってセンサーグラムを参照する。Biacore T200評価ソフトウェアを使用して、1:1結合モデルにデータを適合させることによって(RmaxおよびRIをローカルに適合)、結合速度を評価した。
Fcγ受容体結合アッセイでは、scFv−IgAb_02は、ヒトCD16A FcγRIIIa(48R−158V)−mFc−Avi(KD 12.5nM)およびカニクイザルCD16 FcγRIII−mFc−Avi(KD 19.9nM)に対する結合を示したが、他の試験したFcγ受容体についてはいずれも、結合相互作用は検出されなかった(表1)。Bi−scFv−Fc_02は、CD16A FcγRIIIa(48R−158V)−mFc−Avi(KD 12.2nM)およびカニクイザルCD16 FcγRIII−mFc−Avi(KD 25.2nM)のみに対する結合を示した。対照分子EGFR/CD16Aタンデムダイアボディは、ヒトCD16A FcγRIIIa(48R−158V)−mFc−Avi(KD 4.4nM)およびカニクイザルCD16 FcγRIII−mFc−Avi(KD 8.9nM)に対する結合を示した。様々なIgG1対照分子によって、試験した全Fcγ受容体の機能性を示した(データは示さず)。
10mg/mLポリクローナルヒトIgGの存在下または非存在下での初代ヒトNK細胞に対する二重特異性EGFR/CD16A抗原結合タンパク質の結合
方法
バフィーコートからのPBMCの単離およびヒトNK細胞の濃縮
密度勾配遠心分離によって、バフィーコート(German Red Cross,Mannheim,Germany)からPBMCを単離した。2〜3倍容量のPBS(Invitrogen、カタログ番号:14190−169)を用いてバフィーコート試料を希釈し、Lymphoprep(Stem Cell Technologies、カタログ番号:07861)のクッション上に重層し、ブレーキをかけずに室温で800×gで25分間遠心分離した。界面に位置するPBMCを収集し、PBSで3回洗浄した後、10%FCSを補充した完全RPMI 1640培地中で一晩刺激することなく培養した。NK細胞を濃縮するために、PBMCを一晩培養物から収集し、製造業者の指示に従って未接触ヒトNK細胞の免疫磁気単離のためのEasySep(商標)ヒトNK細胞濃縮キット(Stem Cell Technologies、カタログ番号:19955)と、Big Easy EasySep(商標)Magnet(Stem Cell Technologies、カタログ番号:18001)とを使用する1ラウンドの陰性選択のために使用した。
2%熱不活性化FCS(Invitrogen、カタログ番号:10270−106)、0.1%アジ化ナトリウム(Roth,Karlsruhe,Germany、カタログ番号:A1430.0100)を含有するFACS緩衝液(PBS、Invitrogen、カタログ番号:14190−169)中の10mg/mLポリクローナルヒトIgG(Gammanorm,Octapharma)の存在下または非存在下で、様々な二重特異性EGFR/CD16A抗原結合タンパク質の連続希釈液100μLとともに、示された細胞型のアリコートを37℃で45分間インキュベートした。FACS緩衝液で繰り返し洗浄した後、10mg/mL抗EGFR mAb(クローン62−1−1 Biogenes)、続いて15μg/mL FITCコンジュゲートヤギ抗マウスIgG(Dianova、カタログ番号:115−095−062)を用いて、細胞結合抗体を検出した。AlexaFluor 488コンジュゲートストレプトアビジン(Dianova 016−540−084)によって、ビオチン化セツキシマブおよびビオチン化抗EGFR IgG抗体(IgAb_wtFc、IgAb_enhFc)を検出した。最後の染色工程の後、細胞を再度洗浄し、2μg/mLヨウ化プロピジウム(PI)(Sigma、カタログ番号:P4170)を含有するFACS緩衝液0.2mLに再懸濁して、死細胞を除外した。Millipore Guava EasyCyteフローサイトメーター(Merck Millipore,Schwalbach,Germany)を使用して、2〜5x103の生細胞の蛍光を測定した。Incyteソフトウェア(Merck Millipore,Schwalbach,Germany)を使用して、細胞試料の平均蛍光強度を計算した。二次および三次試薬のみを用いて染色した細胞の蛍光強度値を差し引いた後、それらの値をGraphPad Prismソフトウェア(Windows用GraphPad Prismバージョン6.00、GraphPad Software,La Jolla California USA)を使用した非線形回帰分析に使用した。KDの計算には、1部位結合(双曲線)の方程式を使用した。
二重特異性EGFR/CD16A抗原結合タンパク質の結合能に対するヒトIgGの生理学的濃度の影響を評価するために、図10に例示された10mg/mLポリクローナルヒトIgGの存在下または非存在下で、初代ヒトNK細胞に対していくつかの二重特異性EGFR/CD16A抗原結合タンパク質を用いた結合アッセイを行った。表3は、両条件下での示された二重特異性抗原結合タンパク質の結合親和性を要約している。初代ヒトNK細胞に対するEGFR/CD16Aタンデムダイアボディの見かけの親和性は、10mg/mLポリクローナルヒトIgGの存在下では実質的に変化しない。しかし、IgGを加えると、Bi−scFv−Fc_02の親和性が5nMから20nMに約4分の1低下する。
組換えヒトEGFRまたはEGFRvIIIを発現するCHO細胞に対するEGFR/CD16Aタンデムダイアボディの結合
細胞結合アッセイおよびフローサイトメトリー分析

EGFR + 腫瘍細胞株に対する二重特異性EGFR/CD16A抗原結合タンパク質の細胞傷害活性
方法
細胞株の培養
10%熱不活性化FCS、2mM L−グルタミンおよび100IU/mLペニシリンGナトリウムおよび100μg/mLストレプトマイシン硫酸塩(成分はいずれもInvitrogen製)を補充したDMEM培地中で、標準条件下で、A−431(ATCC、カタログ番号:CRL−1555)を培養した。10%熱不活性化FCS、2mM L−グルタミンおよび100IU/mLペニシリンGナトリウムおよび100μg/mLストレプトマイシン硫酸塩を補充したRPMI 1640培地(成分はいずれもInvitrogen製、本明細書では完全RPMI 1640培地と呼ぶ)中で、標準条件下で、HCT−116(ATCC、カタログ番号:CCL−247)を培養した。細胞株はいずれも、5%CO2を含む加湿雰囲気中で37℃で培養した。
密度勾配遠心分離によって、バフィーコート(German Red Cross,Mannheim,Germany)からPBMCを単離した。2〜3倍容量のPBS(Invitrogen、カタログ番号:14190−169)を用いてバフィーコート試料を希釈し、Lymphoprep(Stem Cell Technologies、カタログ番号:07861)のクッション上に重層し、ブレーキをかけずに室温で800×gで25分間遠心分離した。界面に位置するPBMCを収集し、PBSで3回洗浄した後、10%FCSの代わりに10%ヒトプール血清(Sigma、カタログ番号:H4522)を補充した完全RPMI 1640培地中で一晩刺激することなく培養した。NK細胞を濃縮するために、PBMCを一晩培養物から収集し、製造業者の指示に従って未接触ヒトNK細胞の免疫磁気単離ためのEasySep(商標)ヒトNK細胞濃縮キット(Stem Cell Technologies、カタログ番号:19055)と、Big Easy EasySep(商標)Magnet(Stem Cell Technologies、カタログ番号:18001)とを使用する1ラウンドの陰性選択のために使用した。
カルセイン放出細胞傷害性アッセイのために、示された標的細胞を培養物から収集し、FCS不含RPMI 1640培地で洗浄し、37℃でFCS不含RPMI培地中で30分間かけて10μMカルセインAM(Invitrogen/Molecular Probes、カタログ番号:C3100MP)を用いて標識した。標識細胞を穏やかに洗浄した後、完全RPMI培地(10%熱不活性化FCS、4mM L−グルタミン、100U/mLペニシリンGナトリウム、100μg/mLストレプトマイシン硫酸塩を補充したRPMI 1640培地)に1x105/mLの密度まで再懸濁した。次いで、丸底96ウェルマイクロプレートの個々のウェルに、総容量200μL/ウェルで2連で、12段階希釈で、E:T比5:1の濃縮初代ヒトNK細胞、および示された抗体とともに、1×104の標的細胞を播種した。抗体の非存在下でのエフェクターによる標的の自発的放出、最大放出および殺傷を、各プレートに対して4連で決定した。
EGFR+A−431およびHCT−116標的細胞に対する4時間カルセイン放出細胞傷害性アッセイでは、対照構築物とともに、様々な二重特異性EGFR/CD16A抗原結合タンパク質を試験した。抗原結合タンパク質はオフターゲットの細胞傷害性を示さなかった。図12は、1つの例示的な実験の結果を示している。表6(A−431標的細胞)および表7(HCT−116標的細胞)に、3つの独立した実験から得られた結果を要約する。
EGFRリン酸化の阻害
EGF誘発性EGFRシグナル伝達に対する様々なEGFR/CD16A抗原結合タンパク質の阻害作用を比較するために、A−431細胞を用いたリン酸化アッセイを行った。
細胞株の培養
10%熱不活性化FCS、2mM L−グルタミンおよび100IU/mLペニシリンGナトリウムおよび100μg/mLストレプトマイシン硫酸塩(成分はいずれもInvitrogen製)を補充したDMEM培地中で、5%CO2を含む加湿雰囲気中、37℃で、標準条件下でA−431(ATCC、カタログ番号:CRL−1555)を培養した。
要約すると、10%熱不活性化FCS、2mM L−グルタミンおよび100IU/mLペニシリンGナトリウムおよび100μg/mLストレプトマイシン硫酸塩(成分はいずれもInvitrogen製)を補充したDMEM培地中で、5%CO2を含む加湿雰囲気中、37℃で20時間かけて、96ウェルプレートの個々のウェルに、5x104のA−431細胞(ATCC、カタログ番号:CRL−1555)のアリコートを播種した。次いで、血清不含培地中で4時間かけて細胞を飢餓状態にしてから、示された抗体構築物の連続希釈液を加えた。37℃で30分間インキュベートした後、EGF(Sigma、カタログ番号:10605−HNAE−250)を100ng/mLの最終濃度まで加え、培養物を37℃で10分間さらにインキュベートした後、氷冷PBS(Invitrogen、カタログ番号:14190−169)で細胞を洗浄し、溶解させ、製造業者の指示に従ってPhospho−EGFR ELISAキット(RayBiotech、カタログ番号:PEL−EGFR−Y)を使用したリン酸化EGFRの相対的定量のために使用した。マルチプレートリーダー(Victor 3、Perkin Elmer)を用いて、450nmで吸光度を測定した。GraphPad Prismソフトウェア(Windows用GraphPad Prismバージョン6.00、GraphPad Software,La Jolla California USA)を使用して、吸光度値を分析しプロットした。
2%熱不活性化FCS(Invitrogen、カタログ番号:10270−106)、0.1%アジ化ナトリウム(Roth,Karlsruhe,Germany、カタログ番号:A1430.0100)を含有するFACS緩衝液(PBS、Invitrogen、カタログ番号:14190−169)中の示された抗体の連続希釈液100μLとともに、示された細胞のアリコートを37℃で45分間インキュベートした。FACS緩衝液で繰り返し洗浄した後、10μg/mL抗His mAb 13/45/31−2(Dianova,Hamburg,Germany、カタログ番号:DIA910−1MG)、続いて15μg/mL FITCコンジュゲートヤギ抗マウスIgG(Dianova、カタログ番号:115−095−062)を用いて、または抗CD16A Fvドメイン((配列番号12、13)に対して生成されたmAb 4−1−1、続いて15μg/mL FITCコンジュゲートヤギ抗マウスIgGを用いて、細胞結合タンデムダイアボディを検出した。FITCコンジュゲートヤギ抗ヒトIgG(Dianova、カタログ番号:109−095−088)によって、またはmAb 4−1−1、続いて15μg/mL FITCコンジュゲートヤギ抗マウスIgGを用いて、細胞表面に結合したBi−scFv−Fc_02(配列番号30)、scFv−IgAb_01およびscFv−IgAb_02(配列番号28、29)を検出した。FITCコンジュゲートヤギ抗ヒトIgG(Dianova、カタログ番号:109−095−088)によって、細胞表面に結合したセツキシマブおよびIgAb_wtFc(IgAb_049)を検出した。最後の染色工程の後、細胞を再度洗浄し、2μg/mLヨウ化プロピジウム(PI)(Sigma、カタログ番号:P4170)を含有するFACS緩衝液0.2mLに再懸濁して、死細胞を除外した。MXPソフトウェア(Beckman−Coulter,Krefeld,Germany)を使用するBeckman−Coulter FC500 MPLフローサイトメーター、またはMillipore Guava EasyCyteフローサイトメーター(Merck Millipore,Schwalbach,Germany)を使用して、2〜5x103の生細胞の蛍光を測定した。CXPソフトウェア(Beckman−Coulter)またはIncyteソフトウェア(Merck Millipore,Schwalbach,Germany)を使用して、細胞試料の平均蛍光強度を計算した。二次および三次試薬のみを用いて染色した細胞の蛍光強度値を差し引いた後、それらの値をGraphPad Prismソフトウェア(Windows用GraphPad Prismバージョン6.00、GraphPad Software,La Jolla California USA)を使用した非線形回帰分析に使用した。KDの計算には、1部位結合(双曲線)の方程式を使用した。
陽性対照として使用された抗EGFR IgGセツキシマブ、およびイムガツズマブ(imgatuzumab)(IgAb_065)由来の抗EGFR Fabドメインを有するヒトIgG1は、EGF誘発性EGFRリン酸化を用量依存的に阻害し、EC50値は7μg/mL〜9μg/mLの範囲であった。
薬物動態(PK)特性の評価
それぞれの抗体の単回静脈内注射後のCD1マウスを対象としたscFv−IgAb_02およびBi−scFv−Fc_02の血清中濃度の決定
CD1マウスを対象とした単回投与PK試験では、EGFR/CD16A抗原結合タンパク質のPK評価を行った。CD1マウスでは名目上の標的に対する結合はなく、このモデルでは標的を介した作用を検討することはできないことに留意しなければならない。しかし、被験物質はマウスのFcRn受容体と完全に交差反応性であり、半減期に対するFcRnの作用は完全に反映されるはずである。マウス血清に対するPK分析では、2つの試験系を実施して、EGFR/CD16A抗原結合タンパク質を評価した。最初の試験系をELISAフォーマットで設定し、その後このプラットフォームをMSDリーダープラットフォームに移行させた。2つのアッセイから、一貫した血清中濃度およびPKデータが明らかにされた。

A−431細胞およびA−549細胞におけるEGF刺激性EGFRリン酸化の阻害
EGF誘発性EGFRシグナル伝達に対する様々なEGFR/CD16A抗原結合タンパク質の阻害作用を比較するために、A−431細胞およびA−549細胞を用いたリン酸化アッセイを行った。
細胞株の培養
10%熱不活性化FCS、2mM L−グルタミンおよび100IU/mLペニシリンGナトリウムおよび100μg/mLストレプトマイシン硫酸塩(成分はいずれもInvitrogen製)を補充したDMEM培地中で、5%CO2を含む加湿雰囲気中、37℃で、標準条件下でA−431(ATCC、カタログ番号:CRL−1555)およびA−549(DSMZ、カタログ番号:ACC 107)を培養した。
要約すると、10%熱不活性化FCS、2mM L−グルタミンおよび100IU/mLペニシリンGナトリウムおよび100μg/mLストレプトマイシン硫酸塩(成分はいずれもInvitrogen製)を補充したDMEM培地中で、5%CO2を含む加湿雰囲気中、37℃で20〜22時間かけて、96ウェルプレートの個々のウェルに、5x104のA−431細胞またはA−549細胞のアリコートを播種した。次いで、血清不含培地中で4時間かけて細胞を飢餓状態にしてから、示された抗体構築物の連続希釈液を加えた。37℃で30分間インキュベートした後、EGF(Sigma、カタログ番号:10605−HNAE−250)を100ng/mLの最終濃度まで加え、培養物を37℃で10分間さらにインキュベートした後、氷冷PBS(Invitrogen、カタログ番号:14190−169)で細胞を洗浄し、溶解させ、製造業者の指示に従ってPhospho−EGFR ELISAキット(RayBiotech、カタログ番号:PEL−EGFR−Y)を使用したリン酸化EGFRの相対的定量のために使用した。マルチプレートリーダー(Victor 3、Perkin Elmer)を用いて、450nmで吸光度を測定した。GraphPad Prismソフトウェア(Windows用GraphPad Prismバージョン6.00、GraphPad Software,La Jolla California USA)を使用して、吸光度値を分析しプロットした。
A−431細胞(図15a)およびA−549細胞(図15b)を使用して、EGFによる刺激時のEGFRのリン酸化に対するEGFR/CD16A scFv−IgAb(scFv−IgAb_02)、参照抗EGFR IgGセツキシマブ、およびパニツムマブならびに様々な対照抗体の阻害作用を評価した。抗体の概要、およびEGFRリン酸化の用量依存的阻害について決定されたEC50値の要約を表10に示す。

方法
細胞株の培養
10%熱不活性化FCS、2mM L−グルタミンおよび100IU/mLペニシリンGナトリウムおよび100μg/mLストレプトマイシン硫酸塩(成分はいずれもInvitrogen製)を補充したDMEM培地中で、標準条件下で、A−431(ATCC、カタログ番号:CRL−1555)を培養した。実験に使用する前に、飢餓培地(1%FCSを含むRPMI 1640培地(Invitrogen))中で細胞を1時間培養した。
示されていれば、20μg/mLのそれぞれの抗体とともに、加湿雰囲気中、37℃で3x106の細胞を1時間インキュベートした。続いて、加湿雰囲気中、37℃で5分間または15分間、100ng/mLの濃度で組換えヒトEGF(ThermoFisher、#10605HNAE250)を加えることにより、細胞を刺激した。次いで、細胞を洗浄し、150mM NaCl(AppliChem、#131659.1211)、1%Triton X 100(Roth、#30512)、0.05%デオキシコール酸ナトリウム(Sigma、#D6750)、0.1%SDS(Roth、#CN30.1)、50mM Tris(Biomol、#08003.1)、プロテアーゼ阻害剤(Roche、#11697498001)およびホスファターゼ阻害剤(Roche、#4906845001)を含有する放射免疫沈降アッセイ緩衝液(RIPA)を用いて、氷上で45分間溶解させた。300xg、4℃で15分間遠心分離した後、上清を、62.5mM Tris−HCl pH6.8、2%SDS、5%グリセリン(Applichem、#A2926,055)、200mMブロモフェノールブルー(Roth、#A512.1)、0.1M DTT(Roth、#6908.2)を含有する還元試料緩衝液と1:1で混合し、10分間かけて95℃に加熱し、続いて、1×Tris/グリシン/SDS緩衝液(Bio−Rad、#1610732)中の4〜20%Criterion TGX Precast SDS−PAGEゲル(Bio−Rad、#5678095)上で300Vで22分間SDS−PAGEに供した。イムノブロッティングでは、製造業者の指示に従ってTrans−Blot Turbo Transfer System(Bio−Rad)を使用して、タンパク質をPVDFメンブレン(BioRad、#1704157)に転写した。次いで、メンブレンをTBS中の5%脱脂乳(Sigma、#70166)中で室温で1時間ブロッキングし、TBSで3回洗浄し、供給業者によって推奨されるように希釈した、5%BSA(Sigma、#A3059)中の一次抗体、TBS中の0.05%NaN3(Roth、#K305.1)とともに、室温で1時間または4℃で一晩インキュベートした。続いて、メンブレンをTBSで3回洗浄し、TBSおよび5%脱脂乳中のHRPコンジュゲート二次抗体と室温で1時間インキュベートした。TBSで洗浄した後、ChemiDoc MPイメージングシステム(Bio−Rad)を使用して、ECL溶液(ThermoFisher、#32209)を加えた後の化学発光を測定し、Image Labソフトウェア(BioRad)を使用して分析した。試験した抗体の一覧を表11に示し、タンパク質の検出に使用した抗体の一覧を表12に示す。
EGF誘発性EGFRシグナル伝達の阻害に対するEGFR/CD16A scFv_IgAb_02の作用を評価するために、scFv−IgAb_02とともに、および対照としてEGFR/RSV scFv−IgAb_45、または抗EGFR IgG1セツキシマブとともにA−431細胞をインキュベートした。刺激はそれぞれ5分および15分行い、ウエスタンブロットを介してリン酸化の誘発を評価した。
scFv−IgAb_02によって誘発される、PBMCからのサイトカイン放出
バフィーコートからのPBMCの単離
密度勾配遠心分離によって、バフィーコート(German Red Cross,Mannheim,Germany)からPBMCを単離した。2〜3倍容量のPBS(Invitrogen、カタログ番号:14190−169)を用いてバフィーコート試料を希釈し、Lymphoprep(Stem Cell Technologies、カタログ番号:07861)のクッション上に重層し、ブレーキをかけずに室温で800×gで25分間遠心分離した。界面に位置するPBMCを収集し、PBSで3回洗浄した後、10%FCSを補充した完全RPMI 1640培地中で一晩刺激することなく培養した。
10%熱不活性化FCS、2mM L−グルタミンおよび100IU/mLペニシリンGナトリウムおよび100μg/mLストレプトマイシン硫酸塩(成分はいずれもInvitrogen製)を補充したDMEM培地中で、5%CO2を含む加湿雰囲気中、37℃で、標準条件下でA−431(ATCC、カタログ番号:CRL−1555)を培養した。
エフェクター対標的比50:1で、EGFR+A−431標的細胞と、5x105の初代ヒトPBMCとを共培養した。総容量200μLで漸増濃度のscFv−IgAb_02の存在下または非存在下で、10%FCSを補充した完全RPMI 1640培地中で、共培養物をインキュベートした。scFv−IgAb_02の存在下または非存在下で、PBMCまたはA−431細胞のみの培養を含めることによって、培養物中のバックグラウンドサイトカインレベルを評価した。陽性対照として、共培養物をDynaBeads Human T−Activator CD3/CD28(Gibco、カタログ番号11132D)とインキュベートし、PBMC集団内のT細胞からの試験した全サイトカインの放出を刺激した。加湿インキュベーター内で全培養物を37℃、5%CO2で4時間、24時間または48時間インキュベートしてから、70xgで2分間、室温で遠心分離した。各ウェルから細胞培養上清(70μL)を収集し、BD(商標)Cytometric Bead Array(CBA)Human Th1/Th2 Cytokine Kit II(BD Bioscience)を使用してBioassay GmbH(Heidelberg,Germany)でビーズベースのマルチプレックス法によりサイトカインを定量するまで、−80℃で保存するために丸底96ウェルマイクロプレートに移した。Windows用GraphPad Prism(V6.00/7.03、GraphPad Software,La Jolla,California,USA)を使用して、結果を分析およびプロットした。
サイトカイン定量のために上清を収集した後、細胞ペレットのフローサイトメトリーによって、NK細胞の活性化を評価した。このために、細胞を洗浄し、100μLのFACS緩衝液(2%熱不活性化FCSおよび0.1%アジ化ナトリウムを含有するPBS)の総染色容量で、CD56−PC7(5μL/試験;Beckman Coulter、A21692)、CD25−PE(10μL/試験;Beckman Coulter、A07774)およびCD69−PC5(5μL/試験;Beckman Coulter、IM2656)に再懸濁した。暗所で氷上で15分間インキュベートした後、細胞を洗浄し、FACS緩衝液に再懸濁し、フローサイトメトリーによって分析した。
漸増濃度のscFv−IgAb_02の存在下または非存在下で、PBMCとA−431とを4時間、24時間および48時間共培養した後の細胞培養上清中の6つのサイトカイン、すなわち、インターロイキン−2(IL−2)、インターロイキン−4(IL−4)、インターロイキン−6(IL−6)、インターロイキン−10(IL−10)、腫瘍壊死因子(TNF)およびインターフェロンγ(IFN−γ)の放出を評価した。
薬力学的インビボ試験(POC)
異種移植されたヒトEGFR+腫瘍を担持するヒト化マウスモデルを対象に予防的および治療的投薬レジメンを用いて、scFv−IgAb_02のいくつかのインビボPOC試験を行った。モデルは、臍帯血由来ヒトCD34+造血幹細胞をあらかじめ移植した、流体力学的にIL−15によりブーストしたNOD/Shi−scid/IL−2Rγnullマウス(NOG)からなった。0日目(D0)に、1x106のA−431細胞の皮下接種により、腫瘍を移植した。モデルは、TransCure BioServices SAS,Franceにより提供された。前試験では、ヒト免疫エフェクター細胞(ヒトNK細胞を含む)を用いた確実な再構成、ならびに一貫したA−431腫瘍の取り込みおよび増殖が実証された。このモデルは現在、ヒトNK細胞再構成(末梢血1mL当たり約1〜2x104のヒトNK細胞で生じる)に関して最良のヒト化マウスモデルであるように思われる。
A)CD16A+NK細胞および/またはマクロファージとの相互作用を強制することによる、EGFR+腫瘍細胞に対するADCCおよび/またはADCPの誘発
B)EGFR受容体を遮断することによる、EGFR+腫瘍細胞に対する直接の増殖阻害効果、からなる二重の抗腫瘍作用機序を有すると結論付けられ、
ADCC(および/またはADCP)の誘発は、scFv−IgAB_02によって発揮される主要な抗腫瘍効果である(EGFRリン酸化を遮断することによる直接的な増殖阻害が主要であるセツキシマブとは対照的)。
D17にA−431腫瘍が50〜100mm3のサイズに達した際に、動物に、5、15および45mg/kgの用量(q7d×4)で、scFv−IgAB_02を毎週静脈内投与した。
動物に、D1から45mg/kgの用量(q7d×4)でscFv−IgAB_02またはRSV−EGFRを毎週静脈内投与した。
予防群(1群当たりマウス6匹)の右側腹部に、1×106のA431細胞を移植した。腫瘍細胞接種の1日後、マウスを処置した(4週間にわたり週1回静脈内注射、および殺処分の3日前に最後の注射)。0日目は腫瘍細胞接種日と規定される。以下の処置を行った:

・第7群:IL15−huNOG+A431+RSV/EGFR(2.5mg/kg、毎週、IV)
・第8群:IL15−huNOG+A431+RSV/EGFR(5mg/kg、毎週、IV)

処置群の右側腹部に、1x106のA431細胞を移植した。腫瘍体積が50〜100mm3に達した際に、腫瘍体積、ヒト化率およびNK細胞数に基づいてマウスを無作為化し、処置を開始した。0日目は、最初の処置日として規定される。第10群の動物、ならびに第11群および第12群の追加の動物6匹は、フローサイトメトリー分析のためのサテライト動物として機能させた。

薬物動態
scFv−IgAB_02の薬物動態プログラムには以下が含まれる
カニクイザル血清マトリックスにおける高感度分析アッセイの開発
カニクイザルを対象とした静脈内単回投与PK試験(3つの用量レベル)

カニクイザルを対象としたピボタル28日毒性試験からのPK/TK評価(データは未だ保留中)
カニクイザル由来の薬物動態試料の生物分析のために、MSD(登録商標)プラットフォームに基づく電気化学発光免疫アッセイを行った。MSD(登録商標)プラットフォームは、ELISAのような同様の設定を利用するが、主な相違は、読み出しが従来のELISAのような酵素的基質変換に基づくのではなく、電気化学発光反応に基づくことである。そのため、捕獲抗体を吸着する電極表面とともに専用のマイクロプレートが使用される。さらに、SULFO−TAG(商標)(NHSエステルとしてコンジュゲートしたルテニウム(II)トリス−ビピリジン)と呼ばれる電気化学発光標識によって標識された検出器が必要である。MSD読み取り緩衝液(トリプロピルアミン、TPAを含有する)の存在下で、電気化学発光に適した化学環境が提供される。MSD(登録商標)イメージャーは、プレート電極に電圧を印加し、プレートの底部に近接したSULFO−Tagが一連の還元反応および酸化反応を通じて光を放出するようにする。放出された光の強度が検出される。各標識の複数の励起サイクルによってシグナルを増幅して、光レベルを高め、感度を向上させることができる。刺激機構(電気)はシグナル(光)から分離されているため、最小のバックグラウンドシグナルおよび高いシグナル対バックグラウンド比が可能である。アッセイは5ng/mlのLLOQを有し、LLOQで優れた選択性を示す。14/14の個体がRE%<20を示す。
Citoxlab試験では、合計9匹の雄カニクイザルを登録した。以下の表に従って、8mg/kg(雄3匹)、25mg/kg(雄3匹)および75mg/kg(雄3匹)の用量レベルでscFv−IgAB_02を投与される3群に動物を割り当てた。投与は2時間の注入により5mL/kg/hの速度で行った。
・全動物に対して、被験物質に対する全身曝露を達成した、
・予測通り、scFv−IgAB_02注入の終了時にTmaxに達した、
・終末半減期は33.4〜154時間の範囲であった、
・用量正規化AUC0−t値に基づいて、投与された用量レベルの範囲にわたって血清被験物質曝露の用量比例的な増加を超える増加が認められたが、用量正規化Cmaxを考慮すると、ほぼ用量比例的な増加が観察された。
・用量レベルの増加に伴う終末半減期値のわずかな増加が観察されたことから、scFv−IgAB_02の観察された全身CLの用量関連変化が確認された。
・終末分布容積(terminal volume of distribution)VZはサルにおける全血漿容積に近似し、scFv−IgAB_02が血漿容積内に主に位置することを示す
・用量の増加に伴って終末分布容積値の減少が観察され、被験物質の組織分布/内在化の飽和機構の可能性が示唆された。
EGFR + 腫瘍担持マウスにおけるscFv−IgAB_02の組織分布
A431細胞を用いたマウス異種移植モデルへの125I−scFv−IgAB_02の静脈内投与によって、scFv−IgAB_02の組織分布の評価を行った。
毒性学
関連種としてカニクイザルを用いて、後期腫瘍患者を対象としたFIM試験を裏付ける、二重特異性EGFR/CD16抗原結合タンパク質の毒性学試験を行った。他の種を対象とした毒性学試験は行わなかった。
これまでに行ったscFv−IgAB_02の毒性学プログラムには、以下が含まれる
カニクイザルを対象とした28日間の静脈内反復投与範囲決定試験(intravenous repeated dose range finder study)(非GLP)
カニクイザルを対象とした28日間のピボタル静脈内反復投与毒性試験(GLP)
この試験の目的は、カニクイザルへの4週間(合計5回の注入)にわたる毎週の反復IV注入(2時間の椅子注入(chair infusion))、および5週間の回復期に続いて、被験物質の最大耐量を決定することであった。モーリシャス原産のカニクイザル10匹(雄5匹および雌5匹)を以下のように用量群に割り当てた。
カニクイザルを対象とした4週間の期間のピボタルGLP準拠反復投与毒性試験を行った(CRO:Covance)。この文献の執筆時点で、試験の存続期間は完了しており、初期または部分的な結果が得られた。ただし、この試験の特定の最終分析(例えば、組織病理学、毒物動態学)は未だ保留中であった。
血液学的検査に関して
・全群の動物が、投与期の8日目から網赤血球数の増加を示した。これは、頻繁な採血の生理的代償作用と考えられる。
・投与期の1日目、投与後4時間の時点で、第2群〜第4群の動物ではWBC数が増加し、好中球数が確実に増加した。差は用量依存的でなかった。
中用量群(24mg/kg)の2匹では、増加は投与後24時間の時点で完全に可逆的であった。他の動物および群については、24時間の時点で試料を収集しなかった。他の全群では8日目に回復が示された。ただし、24時間の時点での全群の完全な回復が予測される。

- 抗原結合タンパク質が、2つのポリペプチド鎖からなり、各ポリペプチド鎖が、第1の抗原結合部位のVHにペプチドリンカーによって連結されたVLを含む第1の一本鎖Fvユニット(scFv1)であって、CH2−CH3ポリペプチドのCH2ドメインにヒンジ領域によってN末端で融合される第1の一本鎖Fvユニット(scFv1)と、第2の抗原結合部位のVHにペプチドリンカーによって連結されたVLを含む第2の一本鎖Fvユニット(scFv2)であって、CH2−CH3ポリペプチドのCH3ドメインにペプチドリンカーによってC末端で融合される第2の一本鎖Fvユニット(scFv2)と、を含む、請求項3に記載の多特異性抗原結合タンパク質。
- scFv1が、CD16Aに対する抗原結合部位であり、scFv2が、EGFRに対する抗原結合部位である、請求項4に記載の多特異性抗原結合タンパク質。
- 抗体が、F(ab´)2断片およびFc部分を含み、
(i)F(ab´)2断片が、CD16Aに対する2つのFv抗原結合部位を含み、EGFRに対する抗原結合部位を含む2つの一本鎖Fv(scFv)が、Fc部分に融合され、scFvのそれぞれが、Fc部分のCH3ドメインにC末端で融合され、または
(ii)F(ab´)2断片が、EGFRに対する2つのFv抗原結合部位を含み、CD16Aに対する抗原結合部位を含む2つの一本鎖Fv(scFv)が、Fc部分に融合され、scFvのそれぞれが、Fc部分のCH3ドメインにC末端で融合される、請求項3に記載の多特異性抗原結合タンパク質。 - 重鎖および軽鎖を含む、請求項6に記載の多特異性抗原結合タンパク質であって、
(i)重鎖が、構造VH(CD16A)−CH1−CH2−CH3−VH(EGFR)−VL(EGFR)を有し、軽鎖が、構造VL(CD16A)−CLを有し、または
(ii)重鎖が、構造VH(EGFR)−CH1−CH2−CH3−VL(CD16A)−VH(CD16A)を有し、軽鎖が、構造VL(EGFR)−CLを有する、多特異性抗原結合タンパク質。 - 抗原結合タンパク質が、Fcガンマ受容体に結合しないが、新生児Fc受容体に結合する、請求項1から7のいずれか一項に記載の多特異性抗原結合タンパク質。
- タンパク質が、配列番号28に記載のアミノ酸配列を有する重鎖および配列番号29に記載のアミノ酸配列を有する軽鎖、配列番号43に記載のアミノ酸配列を有する重鎖および配列番号44に記載のアミノ酸配列を有する軽鎖、または配列番号45に記載のアミノ酸配列を有する重鎖および配列番号44に記載のアミノ酸配列を有する軽鎖を含む、請求項1から8のいずれか一項に記載の多特異性抗原結合タンパク質。
- (i)血清アルブミンに対する抗原結合部位、または
(ii)抗原結合タンパク質に融合した血清アルブミン
をさらに含む、請求項1または9のいずれか一項に記載の多特異性抗原結合タンパク質。 - 治療用化合物として使用するための、請求項1から10のいずれか一項に記載の多特異性抗原結合タンパク質。
- EGFR陽性細胞またはEGFRvIII陽性細胞を特徴とするがんの治療に使用するための、請求項11に記載の多特異性抗原結合タンパク質。
- がんが、結腸直腸がん、頭頸部がん、肺がんおよび神経膠芽腫からなる群から選択される、請求項12に記載の使用のための多特異性抗原結合タンパク質。
- 増殖性疾患または腫瘍性疾患を治療または改善する方法であって、それを必要とする対象に、請求項1から10のいずれか一項に記載の多特異性抗原結合タンパク質を投与する工程を含む、上記方法。
