Abstract
Description
PD−1(Programmed Cell Death Protein 1、プログラム細胞死タンパク質1)は、細胞表面受容体であり、T細胞とプレB細胞の表面に発現され、活性化されたT細胞には発現を向上させ、PD−L1とPD−L2の二つのリガンドと結合することをできる。PD−1は、負の調節機能を有する免疫チェックポイントタンパク質であり、T細胞の活性化を防止でき、自身免疫の低下と免疫耐受の促進には重要な役割を果たす。PD−1の機能が、二つの経路で達成し、一つは、リンパ節における抗原特異性T細胞のアポトーシスを促進すること、もう一つは、調節性T細胞(Treg、制御性T細胞ともいう)のアポトーシスを低下することである。
CD47(白血球分化抗原47)はインテグリン関連タンパク質(インテグリン関連タンパク質、IAP)とも呼ばれ、膜インテグリンを伴う膜貫通タンパク質であり、トロンボスポンジン−1(トロンボスポンジン−1、TSP−1)およびシグナル調節タンパク質α(シグナル調節タンパク質α、SIRPα)にも結合する。CD47は、ヒト細胞で広く発現し、かつさまざまな腫瘍細胞で過剰発現し、細胞のアポトーシス、増殖、接着、遊走などのさまざまな細胞活動に関与している。
PD−1/PD−L1シグナル伝達経路はT細胞のエフェクター段階にのみ作用するため、抗PD−1/PD−L1薬物効果には腫瘍抗原の提示が必要で、臨床的には、この治療法は低免疫原性腫瘍の約70%には効果がないことが多く、化学療法、放射線療法、モノクローナル抗体薬と組み合わせる必要がある;さらに、がん細胞は、異なる経路または複数の経路を介して免疫回避を達成することがあるが、PD−1/PD−L1経路を単に遮断するだけでは、一部のがん細胞の免疫回避に効果がない場合がある。
CD47分子に結合しつつ、PD−L1分子に結合できるCD47とPD−L1標的にする二重機能融合タンパク質。
抗体(antibody)とは、身体の免疫系が、抗原の刺激下でBリンパ球から分化した形質細胞によって産生され、対応する抗原に特異的に結合することができる免疫グロブリンであり、また、インビトロで、ハイブリドーマや遺伝子工学および他の方法によって調製することができる。物理化学性質と生物学的な機能によって、抗体は、IgM、IgG、IgA、IgE、IgDの五種類に分けられる。IgG(Immunoglobulin G)タイプの抗体は、血清の主要な抗体成分であり、血清免疫グロブリンの約75%を占め、国内外の既存の抗体医薬品の主なタイプである。
SEQ ID NO:4(SIRPα変異体と抗体Fc区域が連結してなる融合ペプチドフラグメントをコードし、Y349C、T366S、L368A、Y407Vが突然変異して形成したHole構造を有するもの)、SEQ ID NO:5(抗PD−L1抗体の軽鎖をコードするもの)、SEQ ID NO:6(抗PD−L1抗体の重鎖をコードし、T366W、S354Cが突然変異して形成したKnob構造を有するもの)のヌクレオチド配列から、それぞれに相応するDNAフラグメントを合成し、遺伝子工学技術でpcDNA3.1(+)ベクターに挿入し、発現ベクターとするpcDNA3.1−SIRPα−m−Fc、pcDNA3.1−antiPDL1−LとpcDNA3.1−antiPDL1−Hを構築し、リポソーム法で、3個の発現ベクターをCHO−K1にコトランスフェクトし、ホスト細胞に発現し、トランスフェクトされた細胞を、20%ウシ胎児血清FBSを含むDMEM完全培地(Invitrogen社)に24時間培養し、その後、1mg/ml G418抗生素を含む選択的培地に圧力スクリーニングし、続いて細胞を2週間培養した。限界希釈法でサブクローンスクリーニングを行った;Dot−Blot法で、細胞培養上清液における目標タンパク質の含有量(上清をニトロセルロースメンブレンにスポットし、ブロッキング後、西洋ワサビペルオキシダーゼHRPで標識されたマウス抗ヒトIg抗体を加え、ジアミノベンジジンDABで発色させた)を比較した後に、目標タンパク質とするIABの発現量が一番高いクローンをスクリーニングし、発現細胞株とした。
Protein Aアフィニティークロマトグラフィーカラムで二重機能融合タンパク質IABを精製し、10倍カラム体積のバランス液(150mM NaClを含む20mMリン酸塩緩衝液、pH7.0)でカラムをバランスした後に、培養上清液をロードし、ロードしたから5〜10倍カラム体積のバランス液で、導電率がバランスになるまで洗浄した。その後、溶離液(50mMクエン酸緩衝液、pH3.5)で溶離し、OD280が200mAUより大きくなったから、溶離ピークを收集した。1M Trisで、目標タンパク質を含む溶離液のpHを6.5〜7.0に調整した後に、ゲル脱塩カラムで緩衝液(pH7.4リン酸塩緩衝液)を交換し、それから滅菌し、ろ過し、UV法でタンパク質濃度を検測した後に、将来の使用のために4℃に保存した。
精製された抗体サンプルを、それぞれに非還元(8%の分離ゲル)と還元(12%の分離ゲル)の条件において、ポリアクリルアミドゲル電気泳動法(SDS−PAGE)で完全なタンパク質分子量と還元分子量を検測し、試薬を調製・操作する方法について、《分子クローン実験指南》を参照し、電気泳動の結果を明細書図面の図3(非還元SDS−PAGE)と図4(還元SDS−PAGE)に示す。
発現・精製されたIAB融合タンパク質を50mMのNH4HCO3溶液へ脱塩し、UV法で脱塩されたタンパク質溶液を定量化した(50mM NH4HCO3をブランクコントロールとして、消散係数は1.60である)。脱塩されたタンパク質溶液を、50mMのNH4HCO3の添加で1mg/mLまで希釈し、ロードした。
SEQ ID NO:9(SIRPαに高い親和力を持つ変異体SIRPα−mとIgG1抗体Fc区域が連結してなる融合ペプチドフラグメントをコードするが、「Hole」構造を有しない)、SEQ ID NO:10(抗PD−L1抗体の重鎖をコードするが、「Knob」構造を有しなく、かつN297Aの突然変異でFc区域のグリコシル化修飾を除去した)のヌクレオチドから、相応するDNAフラグメントを合成し、それぞれにpcDNA3.1ベクターに挿入し、発現ベクターとするpcDNA3.1−SIRPα−m−Fc'とpcDNA3.1−antiPDL1−H'を構築した。
(1)競争ELISA法でIABとヒトPD−L1の結合力を測定する
anti−PDL1'はビオチン(Biotin)で標識され、使用に備えた。組換え発現されたヒトPD−L1−Fc融合タンパク質を、0.1M NaHCO3溶液(pH9.6)で5μg/mlまで希釈し、100μL/ウェルの量でマイクロプレートをコーティングし、洗浄し、ブロッキングした後に、ウェルごとにビオチンで標識されたanti−PDL1'と勾配希釈された競争物を加え、競争物は、標識されないanti−PDL1'或いは実施例2に製造されたIABタンパク質であり、競争物の濃度は、1μg/mlから、二倍に希釈し、濃度ごとに三つのウェルを使用し、37℃で1時間インキュベート後に、反応液を捨て、プレートを洗浄しアビジンで標識された西洋ワサビペルオキシダーゼ(Avidine−HRP)を加え、基質TMBと発色反応を行い、OD 450nmにおける吸光値を測定し、結果をプロットし、図6を参照する;図において、横軸は、競争物濃度(ng/mL)で、縦軸は、OD値で、中空三角形は、anti−PDL1'の競争データで、黒丸は、IABの競争データである。
同じ方法でIABタンパク質とCD47の結合力を測定した;マイクロプレートをコーティングした抗原は、組換え・発現されたヒトCD47タンパク質であり、ビオチンで標識されたSIRPα−m−Fc'と勾配希釈された競争物を加え、競争物は、標識されないSIRPα−m−Fc'或いは実施例2に製造されたIABタンパク質であり、競争物の濃度は、1μg/mlから、二倍に希釈し、濃度ごとに三つのウェルを使用し、インキュベート後に西洋ワサビペルオキシダーゼとTMBで発色し、OD 450nmにおける吸光値を測定し、結果をプロットし、図7を参照する;図において、横軸は、競争物濃度(ng/mL)で、縦軸は、OD値で、中空三角形は、anti−PDL1'の競争データで、黒丸は、IABの競争データである。
フローサイトメトリーでそれぞれにIABタンパク質とMC38細胞(マウス結腸がん細胞)、HL60細胞(人白血病細胞)の結合状況を検測し、かつSIRPα−m−Fc'、anti−PDL1'をコントロールとし、検測結果について、図19、図20(二つの図には、点線はブランクコントロール、実線はIABタンパク質、短い破線はanti−PDL1'、長い破線はSIRPα−m−Fc'を表示する)を参照する。検測結果より、IABタンパク質がMC38細胞およびHL60細胞の表面に結合できることを示している。
健康な人から20mlの静脈血を採取し、密度勾配遠心分離により末梢血単核細胞(PBMC)を分離し、10%FBSを含むRPMI1640培地に再懸濁し、96ウェル細胞培養プレートを1×10^5細胞/ウェルで広げ、異なる濃度(0.01ng/ml、0.1ng/ml、1ng/ml、10ng/ml、100ng/ml)のブドウ球菌エンテロトキシンB(SEB)と20μg/mL関連タンパク質(IAB、anti−PDL1'或いはSIRPα−m−Fc')を含むRPMI1640培地を各ウェルに添加し、37℃、5%CO2のインキュベーターで72時間インキュベートし、毎日顕微鏡を倒して細胞の状態を観察し、72時間後、培養上清を採取し、ELISAでIL−2分泌量を測定した。実験結果を図8に示し、SEBがPBMC細胞を活性化した後、IL−2の分泌量を増加し、特定の濃度範囲(0−100ng/mL)のSEBで用量依存曲線を示した。異なるSEB濃度で、IABグループとanti−PDL1'グループのIL−2の量は、SIRPα−m−Fc'よりも有意に高かった。IABが、インビトロでT細胞を活性化する機能を持っていることを証明した。
2×106のマウス結腸がん細胞株MC38を2000μLの5μM CSFE(2−カルボキシフルオレセインジアセテートスクシンイミドエステル、リビング蛍光染料)溶液で37°Cの暗所で20分間インキュベートした後、10mLの完全培地を加えて5分間インキュベートし、溶液から未結合のCSFEを除去した。標識された腫瘍細胞を遠心分離によって収集し、完全培地に再懸濁し、濃度が1×106/mLの腫瘍細胞懸濁液を作成した。
ルシフェラーゼレポーター遺伝子を有するFcγRIIIaを発現するJurkat細胞(エフェクター細胞)およびMC38腫瘍細胞(標的細胞)とさまざまな濃度の試験抗体(IAB融合タンパク質、陽性コントロールSIRPα−m−Fc'、陰性コントロールanti−PDL1')を、8%CO2を含む37℃のインキュベーターで4時間インキュベートし、蛍光基質を添加し、蛍光測定値をGlomaxRプレート蛍光アッセイマイクロプレートリーダーで測定し、結果を図11に示した。
15匹の健康なBalb/cマウス(6〜8週間)を、体重(平均約20g)に応じてランダムに3グループ(グループあたり5匹)に分けた。マウスの各グループは、100mg/kgの用量で腹腔内薬物投与された(IAB、SIRPα−m−Fc'または静脈内ヒトIgG)、投与の24時間後、約500μLの血液を眼窩からEPチューブに採取し、血液凝固を防ぐために10μLの1%ヘパリンナトリウムをすぐに添加した。サンプルは、マウスの血液分析のために「上海南部モデル動物研究センター」に送られた。プロトタイプ抗体とIAB融合タンパク質の血液毒性は、投与後のマウスの血液中の赤血球、ヘモグロビン、ヘマトクリット、血小板などの指標の変化をモニタリングすることで評価し、結果を図12、図13、および図14に示した;結果より、IABタンパク質を注射した実験グループの赤血球含有量(9.1×10^12/L)、ヘマトクリット(43.0%)、およびヘモグロビン(137.8g/L)は、ヒトIgGを注射したコントロールグループの赤血球含有量(9.4×10^12/L)、ヘマトクリット(42.56%)、およびヘモグロビン(142.4g/L)と有意差がなかったが、SIRPα−m−Fc注射グループには、血液毒性に影響された関連データは、赤血球含有量(6.8×10^12/L)、ヘマトクリット(35.07%)、およびヘモグロビン(121.6g/L)で、他の2つの群とは有意に異なっていた。
(1)IABでBalb/cマウスMC38腫瘍モデルを治療する:
合計40匹のBalb/c雌マウス(6−8週間)に、MC38腫瘍細胞を2×10^5で皮下接種した。6日目に腫瘍が大きすぎるか小さすぎるマウスを除外した後、腫瘍の体積(約100mm3)に応じて、4つのグループ(各グループに8匹のマウス)に分け、7日目と10日目に、腫瘍にIAB融合タンパク質、無関係なコントロール(ヒトIgG1)またはプロトタイプ抗体(SIRPα−m−Fc'、anti−PDL1')を10mg/kgの用量で注射した。週に2回マウスの腫瘍形成を観察し、ノギスを使用し腫瘍の最大直径(長さ)と垂直距離(幅)を測定し、腫瘍の体積を計算する式は、体積=0.5×長さ×幅2である。マウスの腫瘍体積が2000mm3を超える場合、または腫瘍部位に広い範囲の潰瘍が出現した場合、マウスは頸椎脱臼法で殺され、通常21日間観察された。試験結果は、図15を参照。
IAB融合タンパク質治療グループの腫瘍が完全に消失したマウス(接種部位に触れると、腫瘍には触れられない)と、健康なBalb/cマウスをMC38二次接種実験に使用した。新たに2×10^5の接種量に従って、マウスの元の接種部位の反対側にMC38腫瘍細胞を皮下に再接種した。週に2回マウスの腫瘍形成を観察し、ノギスを使用し腫瘍の最大直径(長さ)と垂直距離(幅)を測定し、腫瘍の体積を計算する式は、体積=0.5×長さ×幅2である。マウスの腫瘍体積が2,000mm3を超える場合、または腫瘍部位に広い範囲の潰瘍が出現した場合、マウスは頸椎脱臼法で殺され、通常21日間観察された。試験結果は、図16を参照。
(1)MC38担癌マウス(マクロファージ欠損タイプ)のIAB治療:
15匹の6週齢Balb/c健康な雌マウスに、MC38腫瘍細胞を2×10^5細胞の数に従って皮下接種し、6日目に、腫瘍の体積に従って平均に3つのグループに分け、各グループに5匹、すなわち実験グループ、コントロールグループ1、およびコントロールグループ2を配置した。実験グループには、200μL/マウスのクロフゾン(英語名Clophosome、マクロファージ除去剤、上海邦律生物テクノロジー有限会社)を注射した後、100μL/マウスのクロフルクソンを1日おきに注射し、IAB融合タンパク質を10mg/kgの用量で7日目と10日目に腫瘍に注射した;コントロールグループ1では、クロフルクソンの代わりにブラックリポソームをマウスに注射し、IAB融合タンパク質を7および10日目に10mg/kgの用量で腫瘍に注射した;コントロールグループ2では、クロフゾンの代わりにブラックリポソームをマウスに注射し、無関係なヒトIgG抗体を7および10日目に10mg/kgの用量で腫瘍に注射した。週に2回各グループのマウスの腫瘍形成を観察し、ノギスを使用し腫瘍の最大直径(長さ)と垂直距離(幅)を測定し、腫瘍の体積を計算する式は、体積=0.5×長さ×幅2である。マウスの腫瘍体積が2,000mm3を超える場合、または腫瘍部位に広い範囲の潰瘍が出現した場合、マウスは頸椎脱臼法で殺され、通常21日間観察された。試験結果は、図17を参照。
各匹のBalb/cマウスに抗CD8抗体または無関係のIgG抗体(200μg/マウス/回)を週2回注射して、マウスのCD8+陽性T細胞をクリアした。
Claims (10)
- 二重機能融合タンパク質が、CD47とPD−L1を標的にし、CD47とPD−L1の両方の分子に結合できることを特徴とする二重機能融合タンパク質。
- 上記の二重機能融合タンパク質が、CD47分子に結合する部分は、SIRPα変異体と抗体Fcセグメントを連結してなるSIRPα−m−Fcペプチドセグメントである;PD−L1分子に結合する部分は、抗PD−L1抗体的軽鎖anti−PD−L1−Lと重鎖anti−PD−L1−Hからなるものであることを特徴とする請求項1に記載の二重機能融合タンパク質。
- 上記のCD47に結合する部分の抗体Fc区域及PD−L1に結合する部分の抗体は、いずれもIgG1サブタイプであることを特徴とする請求項2に記載の二重機能融合タンパク質。
- 上記の二重機能融合タンパク質は、Knob−in−Hole構造を有することを特徴とする請求項2又は3に記載の二重機能融合タンパク質。
- 上記のCD47に結合する部分のSIRPα−m−Fcは、Knob−in−Hole構造におけるHole構造を有し、上記のPD−L1に結合する部分の重鎖anti−PD−L1−Hは、Holeと合わせるKnob構造を有することを特徴とする請求項4に記載の二重機能融合タンパク質。
- 上記のCD47に結合する部分のアミノ酸配列は、SEQ ID NO:1である;上記のPD−L1に結合する部分の軽鎖anti−PD−L1−Lのアミノ酸配列は、SEQ ID NO:2であり、重鎖anti−PD−L1−Hのアミノ酸配列は、SEQ ID NO:3であることを特徴とする請求項5に記載の二重機能融合タンパク質。
- 上記のCD47に結合する部分のヌクレオチド配列は、SEQ ID NO:4である;上記のPD−L1に結合する部分の軽鎖anti−PD−L1−Lのヌクレオチド配列は、SEQ ID NO:5であり、重鎖anti−PD−L1−Hのヌクレオチド配列は、SEQ ID NO:6であることを特徴とする請求項5に記載の二重機能融合タンパク質。
- 請求項1〜7のいずれか一つに記載の二重機能融合タンパク質の、腫瘍を予防・治療する医薬品の調製における応用。
- 請求項1〜7のいずれか一つに記載の二重機能融合タンパク質の、免疫を調節する医薬品の調製における応用。
- 請求項1〜7のいずれか一つに記載の二重機能融合タンパク質の、CD47を標的にする医薬品の血液毒性を低下する医薬品の調製における応用。
