一部の実施形態において、本開示の抗体は単一ドメイン抗体である。単一ドメイン抗体は、抗体の重鎖可変ドメインの全てもしくは一部分または軽鎖可変ドメインの全てもしくは一部分を含む、単一のポリペプチド鎖である。ある特定の実施形態において、単一ドメイン抗体は、ヒト単一ドメイン抗体である(Domantis,Inc.,Waltham,Mass.、例えば、米国特許第6,248,516 B1号を参照されたい)。一実施形態において、単一ドメイン抗体は、抗体の重鎖可変ドメインの全てまたは一部分からなる。
一部の実施形態において、本明細書に記載される抗体のアミノ酸配列修飾(複数可)が企図される。例えば、抗体の結合親和性及び/または他の生物学的特性を向上させることが望ましい場合がある。抗体のアミノ酸配列変異形は、抗体をコードするヌクレオチド配列中に適切な変化を導入することによって、またはペプチド合成によって、調製されても良い。かかる修飾には、例えば、抗体のアミノ酸配列からの残基の欠失、及び/またはそこへの残基の挿入、及び/またはその中での残基の置換が含まれる。最終構築物に到達するように、欠失、挿入、及び置換を任意に組み合わせることができるが、但し、その最終構築物が、所望の特性を保有することを条件とする。アミノ酸の改変は、配列が作製されるときに対象の抗体アミノ酸配列中に導入され得る。
ある特定の実施形態において、1つ以上のアミノ酸置換を有する抗体変異形が提供される。置換型突然変異生成に対する目的の部位には、HVR及びFRが含まれる。保存的置換が、「保存的置換」の見出しの下に表1において示される。より実質的な変化は、表1において、「例示的な置換」の見出しの下に提供され、またアミノ酸側鎖クラスを参照して以下に更に記載される。アミノ酸置換が目的の抗体中に導入され、その産物が、所望の活性、例えば、抗原け都合の保持/向上、免疫原性の減少、またはADCCもしくはCDCの向上についてスクリーニングされても良い。

ある特定の実施形態において、1つ以上のアミノ酸修飾が、本明細書に提供される抗体のFc領域に導入され、それによってFc領域変異形を産生しても良い。Fc領域変異形は、1つ以上のアミノ酸位置にアミノ酸修飾(例えば、置換)を含む、ヒトFc領域配列(例えば、ヒトIgG1、IgG2、IgG3、またはIgG4 Fc領域)を含んでも良い。
本開示の抗体は、当該技術分野で既知であり、容易に入手可能な、追加の非タンパク質性部分を含有するように、更に修飾され得る。ある特定の実施形態において、抗体の誘導体化に好適な部分は、水溶性ポリマーである。水溶性ポリマーの非限定的な例としては、ポリエチレングリコール(PEG)、エチレングリコール/プロピレングリコールのコポリマー、カルボキシメチルセルロース、デキストラン、ポリビニルアルコール、ポリビニルピロリドン、ポリ−1,3−ジオキソラン、ポリ−1,3,6−トリオキサン、エチレン/無水マレイン酸コポリマー、ポリアミノ酸(ホモポリマーまたはランダムコポリマーのいずれか)、及びデキストランまたはポリ(n−ビニルピロリドン)ポリエチレングリコール、プロプロピレン(propropylene)グリコールホモポリマー、プロリプロピレン(prolypropylene)オキシド/エチレンオキシドコポリマー、ポリオキシエチル化ポリオール(例えば、グリセロール)、ポリビニルアルコール、ならびにそれらの混合物が挙げられるが、これらに限定されない。ポリエチレングリコールプロピオンアルデヒドは、水中でのその安定性に起因して、製造における利点を有し得る。ポリマーは、任意の分子量のものであっても良く、分岐していても非分岐であっても良い。抗体に結合したポリマーの数は様々であり得、1つを超えるポリマーが結合される場合、それらは、同じ分子または異なる分子であり得る。一般に、誘導体化に使用されるポリマーの数及び/または種類は、向上させるべき抗体の特定の特性または機能、抗体誘導体が規定の条件下で療法において使用されることになるかどうか等を含むがこれらに限定されない考察に基づいて決定することができる。
抗体はまた、組み換え法を使用して生成されても良い。抗抗原抗体の組み換え生成では、抗体をコードする核酸が単離され、更なるクローニング(DNAの増幅)または発現のために、複製可能なベクター中に挿入される。抗体をコードするDNAは、従来の手順を使用して(例えば、抗体の重鎖及び軽鎖をコードする遺伝子に特異的に結合可能である、オリゴヌクレオチドプローブを使用することによって)、容易に単離されて配列決定され得る。多くのベクターが利用可能である。ベクター構成要素としては、概して、シグナル配列、複製源、1つ以上のマーカ遺伝子、エンハンサー要素、プロモータ、及び転写終結配列のうちの1つ以上が挙げられるが、これらに限定されない。
本開示の抗体は、直接的にのみではなく、異種ポリペプチドとの融合ポリペプチドとしても組み換え的に生成され得る。この異種ポリペプチドは、好ましくは、成熟タンパク質またはポリペプチドのN末端にて特異的切断部位を有するシグナル配列または他のポリペプチドである。選択される異種シグナル配列は、好ましくは、宿主細胞によって認識され処理されるもの(例えば、シグナルペプチダーゼによって切断される)ものである。天然の抗体シグナル配列を認識し処理しない原核宿主細胞については、シグナル配列は、例えば、アルカリホスファターゼ、ペニシリナーゼ、lpp、または耐熱性エンテロトキシンIIリーダーの群から選択される原核性シグナル配列によって置換される。
発現及びクローニングベクターの両方は、ベクターが1つ以上の選択される宿主細胞において複製できるようにする核酸配列を含有する。一般に、クローニングベクターにおいて、この配列は、ベクターに宿主染色体DNAとは無関係に複製させるものであり、かつ複製または自己複製配列の起点を含むものである。かかる配列は、多様な原核宿主細胞についてよく知られている。例えば、プラスミドpBR322からの複製起点が大部分のグラム陰性細菌に好適である。
発現及びクローニングベクターは、選択可能なマーカとも称される選択遺伝子を含有しても良い。典型的な選択遺伝子は、(a)抗生物質または他の毒素、例えば、アンピリシン、ネオマイシン、メトトレキサート、またはテトラサイクリンに対する耐性を与える、(b)栄養要求性欠乏を補充する、あるいは(c)複合培地から得ることができない必須の栄養素、例えば、桿菌のためのDアラニンラセマーゼをコードする遺伝子を供給する、タンパク質をコードする。
発現及びクローニングベクターは、概して、宿主生物によって認識されるプロモータを含有し、抗体をコードする核酸に動作可能に連結する。原核宿主との使用に好適なプロモータは、phoAプロモータ、β−ラクタマーゼ及びラクトースプロモータ系、アルカリホスファターゼプロモータ、トリプトファン(trp)プロモータ系、及びプロモータ等のハイブリッドプロモータが挙げられる。しかしながら、他の既知である細菌性プロモータも好適である。細菌系における使用のためのプロモータはまた、抗体をコードしているDNAに動作可能に連結するシャインダルガーノ(S.D.)配列を含有することになる。
上記のように、翻訳開始領域(TIR)は、原核宿主細胞における組換えタンパク質の翻訳に重要であることが知られている(例えば、Simmons LC and Yansura DG 1996 Nat.Biotechnol.14:629、及びVimberg V et al.2007 BMC Mol.Biol.8:100を参照されたい)。TIRは、翻訳単位の翻訳の効率を決定し得る。TIRは、典型的には、開始コドン、シャインダルガーノ(SD)配列、及び翻訳エンハンサー等の翻訳単位特性を含む。TIRは、シグナルペプチドをコードする分泌シグナル配列を更に含んでも良い。配列及びTIRの特性間の離間により翻訳開始効率が調節され得る。タンパク質生成におけるTIRの使用の更なる説明については、例えば、発現及び分泌を最適化するための翻訳開始領域(TIR)及びシグナル配列を説明している、米国特許第5,840,523号(Simmonsら)を参照されたい。
原核宿主細胞において使用される発現ベクターはまた、転写の終結及びmRNAの安定化に必要な配列を含有しても良い。原核細胞において、終結因子は、Rho依存性またはRho独立終結因子を含んでも良い。原核宿主細胞において有用な終結因子の一例としては、λt0終結因子が挙げられるが、これに限定されない(Scholtissek and Grosse,Nucleic Acids Res.15:3185,1987)。
宿主細胞を培養してポリペプチドの2つの鎖を発現させることによって原核宿主細胞に2つの鎖を含有するポリペプチドを生成する方法が本明細書で提供され、この場合、発現時に、2つの鎖が折り畳まれ組み立てられて宿主細胞中に生物学的に活性のポリペプチドを形成し、宿主細胞は、成長温度及び成長撹拌速度を含む成長相、ならびに生成温度及び生成撹拌速度を含む生成相を含む、条件下で、培養培地において培養され、成長温度は、生成温度を2〜10℃上回り、成長撹拌速度は、生成撹拌速度を50〜250rpm上回る。ある特定の生成プロセス最適化により、本明細書に記載のデータが示すように2鎖タンパク質の収率が劇的に増加することは、本開示の驚くべき発見である。
本開示のある特定の態様は、原核宿主細胞に関する。本明細書のベクターにおけるDNAのクローニングまたは発現に好適な原核細胞としては、グラム陰性またはグラム陽性生物等の真正細菌、例えば、エシェリキア属等の腸内細菌科、例えば、大腸菌、エンテロバクター属、クレブシエラ属、プロテウス属、サルモネラ属、例えば、サルモネラ・チフィムリウム、セラチア属、例えば、セラチア・マルセッセンス、及び赤痢菌属、ならびにバシラス・サブチリス及びバシラス・リケニフォルミス等のバシラス(例えば、1989年4月12日出版のDD 266,710に開示されているバシラス・リケニフォルミス41P)、1989)、緑膿菌及びストレプトミセス等のシュードモナス属が挙げられる。1つの好ましい大腸菌クローニング宿主は大腸菌294(ATCC 31,446)であるが、大腸菌B、大腸菌X1776(ATCC 31,537)、及び大腸菌W3110(ATCC 27,325)等の他の株が好適である。これらの例は、限定的なものではなく例示的なものである。
本開示のある特定の態様は、成長相及び生成相を含む条件下での培養培地中の宿主細胞の培養に関する。これらの相の各々は、更に、宿主細胞が特定の相において成長する条件を指し得る。例えば、本明細書で使用される場合、成長相は、成長温度及び成長撹拌速度を含んでも良く、生成相は、生成温度及び生成撹拌速度を含んでも良い。
本開示のある特定の態様は、宿主細胞から生物学的に活性のポリペプチドを回収することに関する。典型的には、本開示の生物学的に活性のポリペプチドを回収すること(「精製すること」または「精製」という用語が本明細書で互換的に使用されても良い)は、ポリペプチドを宿主細胞(ポリペプチドが培地中に排出される場合には細胞培養培地)から単離して、ポリペプチドを、他の関連する高分子、例えば、細胞残屑及び他のポリペプチドから精製することを伴う。多様なタンパク質を多様な宿主細胞内コンパートメントから精製するための多数の技法が当該技術分野で既知である(例えば、Evans,Jr.,TC and Xu MQ(eds.)Heterologous Gene Expression in E.coli(2011)Methods in Molecular Biology Vol 705,Humana Pressを参照されたい)。例示的な技法が以下に記載されるが、これらは、当業者の理解を助けるための例示のみを目的に含まれるものであり、限定するものでは全くない。
組み換え技法を使用する異種分泌タンパク質の生成は、多くの治療用分子にとって重要である。多特異性抗体(例えば、二重特異性抗体)は、多くの重要な治療的使用、例えば、癌における免疫療法及び他の用途を持つ異種分泌タンパク質の1つの非限定的な例である。治療的使用のための多特異性抗体の生成は、半抗体(hAb)等のこれらの抗体の構成単位を産業規模で生成する能力を必要とする。この要求を満たすために、標準的な方法と比べて顕著に増加した生成をもたらす最適化された発現ベクター及び処理ステップが本明細書に記載される。重要なことに、シャペロンタンパク質FkpA、DsbA、及びDsbCと組み合わせたhAbの重鎖(HC)及び軽鎖(LC)を共発現させるための単一プラスミドまたは適合性プラスミド系が、組み立てられたhAbの生成を顕著に向上させることが分かった。この系を試験して、複数のhAbの生成を改善することが分かったが、これは、多くの分泌タンパク質の生成に対するその幅広い有用性を示す。続く処理ステップの最適化(例えば、培養の異なる相における、撹拌速度、pH、FkpAプロモータ、及び培養温度を含む)により、産物収率がなお更に顕著に増加した。
半抗体(hAb)ベクター構築
ベクターを、EP1356052に記載されているものに類似した様式で構築した。具体的には、種々の発現ベクターをhAbの発現のために作製した。各ベクターでは、発現カセットをEcoRI部位にて大腸菌プラスミドpBR322のフレームワークにクローニングした(Sutcliffe,Cold Spring Harbor Symp.Quant.Biol.43:77−90,1978)。各発現カセットは、少なくとも次の構成要素を含有した。(1)転写の制御のためのphoAプロモータ(Kikuchi et al.,Nucleic Acids Res.9:5671−5678,1981)、(2)翻訳開始のための大腸菌trp由来のシャインダルガーノ配列、耐熱性エンテロトキシンII(STII)シグナル配列、またはこれら両方の組み合わせ(Chang et al.,Gene 55:189−196,1987)、及び(3)転写を終了するためのλt0終結因子(Scholtissek and Grosse,Nucleic Acids Res.15:3185,1987)。追加的に、STIIシグナル配列またはそのシグナル配列のサイレントコドン変異形は、軽鎖または重鎖をコードする配列に先行した。この配列は、ポリペプチドのペリプラズムへの分泌を導く(Picken et al.,Infect.Immun.42:269−275,1983、及びSimmons and Yansura,Nature Biotechnology 14:629−634,1996)。
xIL13、xIL4、xIL17、及びxIL33 hAbの最高力価を得るための上記のベクターと組み合わせるシャペロン発現を決定するために、プラスミドを共発現に対して提供した。シャペロン活性を持つペプチジルプロイルシス−トランスイソメラーゼであるFkpAタンパク質を含む、ある数の既知のシャペロンを試験した。そのために、適合性プラスミド(pACYC、Novagen,Madison,WI)を、EP1356 052に記載されているように、FkpAのためのORFを含有するように構築した(例えば、実施例9〜10、特に段落[0207]を参照されたい)。
FkpAの共発現について記載した適合性プラスミド系と同様に、適合性プラスミドを利用して、既に記載したFkpA TIR変異形のうちの1つを組み込むhAb発現プラスミドと組み合わせた種々の既知のオキシドレダクターゼをスクリーニングした。この作業は、pxIL13.2.2.FkpAc13として特定されたTIR2,2プラスミドを用いて行った。加えて、適合性プラスミドを利用して、xIL4及び他のIgG1 hAbと組み合わせたFkpA及びオキシドレダクターゼをスクリーニングした。
大規模生成は、本質的にはEP1356052に記載されている通りのものであった(例えば、実施例4及び段落[0159]〜[160]を参照されたい)。10リットルの発酵の各々では、0.5mLの凍結保存培養(10〜15%のDMSOを含む)を解凍して使用し、0.5mlのテトラサイクリン溶液(5mg/ml)ならびに/または2mLのカナマイシン溶液(5mg/mL)及び2.5mlの1Mリン酸ナトリウム溶液を補充した500mlのSoy LB培地を含む2Lの振盪フラスコに播種した。この種培養物を振盪しながら30℃で約16時間成長させた後、これを使用して10リットルの発酵器に播種した。
非還元性可溶性試料調製は、EP1356052に記載されているものと同様である(例えば、実施例4、特に段落[0162]を参照されたい)。具体的には、非還元性可溶性試料を次のように調製した。発酵過程で採取した1mLの凍結した全培養液試料を室温で解凍した。100μLの解凍した全培養液を500μLの抽出緩衝液に添加した。(抽出緩衝液:10mMのトリス、pH6.8、5mMのEDTA、新たに添加した0.2mg/mLの鶏卵リゾチーム、及び新たに調製したヨード酢酸、最終濃度5〜10mM)。抽出緩衝液を加えた全培養液試料を、氷上で5〜10分間インキュベートした後、2×10のパルスで超音波処理し、続いて4℃及び14,000rpmで15〜20分間遠心分離した。上清を可溶性画分として除去した。SDS−PAGE及び免疫ブロットによる分析では、可溶性画分を、還元剤なしで、2倍のNovex Tricine試料緩衝液中に1:10で希釈した。10μLのこの調製物を、10ウェルのNovex 10% Bis−Tris NuPageゲルに充填し、MES緩衝液を用いて150Vで電気泳動させた。その後、このゲルを使用して、免疫ブロットまたはクマシーブルーによる染色を行った。
親和性精製した抗IL13及び抗IL17半抗体を1:1の質量比で組み合わせた後、pH8.5に滴定した。組み立てプロセスから予測した二重特異性抗体の理論最大量に対して還元した100倍の過剰モル比のL−グルタチオン(GSH)を混合物に添加し、温度を室温から35℃に調整した。2つの半抗体を酸化還元反応においてインビトロで組み立てて、抗IL13/抗IL17二重特異性抗体を形成した。酸化還元反応を35℃で12〜24時間継続した。組み立て反応を、pHを6.5に調整することによってクエンチし、プールを水で希釈した後、二重特異性抗体に対して、イオン交換クロマトグラフィーまたは疎水性カラムクロマトグラフィー単独による精製、またはそれらの組み合わせによる精製のいずれかを行った。
hAb力価に与えるFkpA発現の影響
図1Aに示すように、xIL13 hAb TIR1,1ベクターから軽鎖及び重鎖サブユニットを生成すると、総量2.9g/L及び1.2g/Lの軽鎖及び重鎖がそれぞれ生成された。TIR2,2については、生成された軽鎖及び重鎖の総量は、それぞれ、6.4g/L及び4.1g/Lであった。
xIL13 hAb生成を、オキシドレダクターゼDsbA及びDsbCの発現のために上記のpxIL13.2.2.FkpAc13(単一プラスミド)を適合性プラスミドと組み合わせることによって更に最適化した(図9)。pxIL13.2.2.FkpAc13を伴うDsbAとDsbCとの両方を発現しているプラスミド(JJ247)は、hAb力価を増加させた。図10は、DsbA及びDsbCのIPTG誘導の存在下及び不在下での、経時的に生成されたxIL13 hAbの力価を示す。JJ247を伴うpxIL13.2.2.FkpAc13についての最高力価は、発酵開始から52時間で達成された。この時点では、IPTGのない条件(非誘導条件)で、xIL13力価は1.2±0.2g/Lであり、IPTGのある条件(誘導条件)では、xIL13力価は1.0±0.2g/Lであった(図10)。試験した両条件では、52時間〜72時間における力価の低下が顕著であったが、これは実施例2に記載する発酵プロセス中の酸素取り込み速度(OUR)の低下及び浸透圧の上昇に起因するものであった。
上記の結果は、hAb HC及びLCと、FkpA、DsbA、及びDsbCとの共発現によって達成されるhAb生成の改善を示す。しかしながら、このように生成が向上しても、力価は、プラトーであるか、または更には生成から52時間後の時点では減少したことが分かった。したがって、hAb生成を更に最適化するための追加の実験を開始した。
これまでの実施例はhAb生成の顕著な増大を示すが、生成プロセスを更により最適化するための追加の試験を開始した。このため、xIL13 hAb生成では、実施例1で説明した高性能の単一プラスミド系及び実施例2で説明した約1.75mmol/L/分のOUR設定値を開始点として使用した。このプロセスを、以下に記載のように、顕著に優れた生成収率を得るために更により最適化した。
これまでの実施例はhAb生成の顕著な増大を示すが、宿主株66F8と67A6との間のhAb生成における潜在的な差異を特性評価するための追加の試験を開始した。このため、hAb生成では、実施例1で説明した高性能の単一プラスミド系、実施例2で説明したOUR移行、及び実施例3で記載した温度移行を、66F8及び67A6の2つの大腸菌宿主株において評価した。66F8株の遺伝子型は、W3110 ΔfhuA ΔphoA ilvG+ Δprc spr43H1 ΔdegP ΔmanA lacIQ ΔompT ΔmenE742である。67A6株の遺伝子型は、W3110 ΔfhuA ΔphoA ilvG+ Δprc spr43H1 ΔdegP ΔmanA lacIQ ΔompT ΔmenE742 degPS210Aである。
xIL33 hAb(MD501プラスミド)を、xIL13 hAb単一プラスミド(MD157)のLC及びHCのためのORFをxIL33のLC及びHCのORFで置き換えることによって構築した。抗体断片及びシャペロンの発現に使用したプロモータは、xIL13(MD157)プラスミドと同一であった(図39)。単一発酵を、xIL33のLC及びHC(MD481)のORFのみを含有するxIL33 hAb発現ベクターを用いて行った。MD481プラスミドは、分子シャペロンDsbA、DsbC、またはFkpAのORFを含有しなかった。発酵を、30℃の一定温度、7.0のpH、及び650rpmの撹拌速度で行った(図40の規範事例)。次に、単一発酵を、xIL33のLC及びHCのORF、ならびに分子シャペロンDsbA、DsbC、またはFkpAのORFを含有するxIL33 hAb発現ベクター(MD501)を用いて行った。MD481の発酵に対する条件と同じ動作条件をMD501の発酵に使用した。単一プラスミド(MD501)の使用により、規範事例と比較してxIL−33 hAb力価が約10倍増加した。
FkpAの発現レベルを最適化するために、2つの追加のFkpA TIR変異形を、xIL13、xIL17、及びxIL33 hAbのための単一プラスミド(MD157、MD341、及びMD501プラスミド)において試験した。FkpA TIR変異形を、内因性FkpAシグナル配列との比較において説明したように特性評価した(図42)。MD157、MD341、及びMD501プラスミドは、3つのTIR強度と相関したFkpAc13のORFを組み込んだ(図43)。MD157、MD341、及びMD501において、単一プラスミドFkpAc13 ORFをFkpA TIR1またはTIR2 ORFで置き換え、各hAbについて、既に特定した最適化した発酵条件において試験した。
実施例3で特定したxIL13 hAbの最良の条件を第2のOTR方針を用いてまた試験した。実験(N=3)において、容器背圧(BP)及び散布速度を、それぞれ、1.0から0.3バールに、また20から13SLPMに減少させた。背圧及び散布速度の減少に起因するOTRの損失を回復するために、成長相及び生成相の撹拌速度を、650から880rpmに、また475から650rpmに増加させた。容器背圧、散布速度、及び撹拌速度の異なる組み合わせを使用して、前述の実施例で述べたものと同様のOTR条件を達成することができる。

Claims (69)
- 原核宿主細胞における2つの鎖を含むポリペプチドの生成方法であって、
(a)
成長温度及び成長撹拌速度を含む成長相、ならびに
生成温度及び生成撹拌速度を含む生成相、を含む、条件下で、培養培地において、前記ポリペプチドの前記2つの鎖を発現させるように、前記宿主細胞を培養することであって、それによって、発現時に、前記2つの鎖が折り畳まれ、かつ組み立てられて、前記宿主細胞において生物学的に活性のポリペプチドを形成し、
前記宿主細胞が、
