et al.,Nature Immunology 6,1245);しかしながら、後の報告では、TIM−3およびGal9の相互作用は本来非特異的であると示された(Leitner et al.PLoS Pathog 9(3):e1003253)。CEACAM1は、T細胞耐容能および消耗を調節するTIM−3リガンドとして報告された(Huang et al.Nature 517,386)。しかしながら、Huangの結果は、TIM−3がCEACAM1と結合しないと示す本発明者ら自身のデータと一致しない(下記参照)。

現在あるGIAに対して保存的修飾の導入によってGIAを製造してもよい。例えば、修飾GIAは、上記製造された抗体の相当品と相同性である重鎖および軽鎖可変領域ならびに/またはFc領域を含む。本明細書に開示されている方法に使用することができる修飾GIAは、Gal3:TIM−3シグナル伝達経路を遮断することができる所望の機能特性を保持しなければならない。

いくつかの周知のアッセイを使用して、候補、例えば、Gal3タンパク質またはコンビナトリアルライブラリーからの試験化合物を含む抗原で動物を免疫することにより生成された抗体がGal3とTIM−3との相互作用を遮断することができるかどうかを評価することができる。通常、これは、次の種類のアッセイのうち1つ以上を使用して候補の評価を含む:i)候補が標的タンパク質、すなわち、Gal3またはTIM−3と結合するかどうかを試験する結合アッセイ;ii)候補がGal3とTIM−3との相互作用を遮断することができるかどうかを試験する遮断アッセイ;iii)Gal3とTIM−3との相互作用の遮断によって、候補がT細胞を活性化することができるかどうかを試験する細胞ベース機能アッセイ;ならびにiv)候補が腫瘍量を低減することができるかどうかを試験するインビボ有効性アッセイ。
2つの分子の相互作用を評価するために使用されるいずれものアッセイを使用して、候補が標的タンパク質と結合することができるかどうかを決定することができる。非限定的例示的アッセイとしては、−酵素結合免疫吸着アッセイ(ELISA)、ラジオイムノアッセイ(RIA)などの−結合アッセイ、蛍光標識細胞分取(FACS)分析が挙げられる。場合によっては、標的タンパク質および候補の結合を複合体中の標識化された標的タンパク質の検出によって決定することができるように、標的タンパク質、すなわち、Gal3またはTIM−3タンパク質を放射性同位元素または酵素標識と結合することができる。例えば、標的タンパク質を直接的にまたは間接的に125I、35S、14C、または3Hで標識化し、放射性同位元素を電波放射の直接的計数もしくはシンチレーション測定によって検出することができる。あるいは、標的タンパク質分子を、例えば、ホースラディッシュペルオキシダーゼ、アルカリホスファターゼ、またはルシフェラーゼで酵素的に標識化することができ、候補と標的タンパク質との結合を適切な基質を生成物に変換することによって決定する。
て推奨されている適切な濃度、流速において緩衝液(Biacore ABにより提供されている)中に候補を流すことによって、結合を測定する。会合速度および解離速度を記録し、会合速度および解離曲線を、BIA評価ソフトウェア(Biacore AB)を使用して結合モデルにフィットする。相互作用のKD値、Kon値およびKoff値を測定することができる。好ましいGal3:TIM−3阻害剤は、1×10−7M以下、例えば、5×10−7M以下または1×10−8M以下でその標的タンパク質と結合することができる。
それから、標的タンパク質の結合能を示した候補を、遮断アッセイにおいてGal3とTIM−3との相互作用を遮断するその能力について評価する。いくつかの実施形態では、遮断アッセイは、免疫アッセイ、例えば、ELISAである。1つの実施形態では、候補がTIM−3とGal3との相互作用を遮断するかどうかの決定方法は、プレートを標的タンパク質、TIM−3またはGal3のうち1つで被覆すること、ならびに候補および他の標的タンパク質、すなわち、Gal3またはTIM−3の混合物を被覆されたプレートに添加して、TIM−3とGal3との結合に対応するシグナルを検出することを含む。候補を添加しないコントロール反応と比較したシグナルの減少は、候補がGal3とTIM−3との相互作用を遮断することができることを示す。
場合によっては、標的タンパク質との結合を示した候補を、混合リンパ球培養反応(MLR)アッセイを使用して、T細胞を活性化するその能力についてさらに評価する。1つの例示的アッセイは、米国特許第8,008,449号に記載されており、関連する開示はその全文を参照することにより本明細書に組み入れられる。MLRアッセイを使用して、T細胞増殖、IL−2および/またはIFN−γの産生を測定することができる。1つの例示的アッセイでは、候補を、種々の濃度において抗原提示細胞(APC)と共に培養されたいくつかの精製T細胞に添加する。それから、37℃において4〜7日の期間、候補の存在下で細胞を培養する。それから、特定の体積の培養培地を、サイトカイン測定のために採取する。IFN−γおよび他のサイトカインのレベルを測定することができる。サイトカイン産生の測定方法は周知であり、市販キットを容易に入手することができ、例えば、OptEIA ELISAキット(BD Biosciences社)がある。いくつかの実施形態では、12〜24時間、例えば、18時間の期間、3H−チミジンの存在下細胞を培養し、細胞増殖と正の相関性がある細胞内の3H−チミジンの取込み量について分析する。コントロールと比較して、候補を含む培養液はT細胞増殖の増加、IL−2および/またはIFN−γの産生増加を示すことを示している結果は、該候補はTIM−3およびGal3の相互作用の遮断によってT細胞の活性化に有効であることを示す。T細胞の活性化における候補の能力を評価するために使用することができるMLRの1つの例示的アッセイは、実施例11に開示されている。
別の実施形態では、インビボアッセイを使用して、候補ががん治療に有効であるかどう
かを評価する。インビボアッセイを確立された既知の手順に従って、マウス腫瘍モデルなどの腫瘍モデルで行うことができる。手短に言えば、動物、例えば、マウスにヒト腫瘍細胞株を皮下に移植する。腫瘍が増殖し、特定のサイズ、例えば、100〜300mm3に達した場合、適切な投与量で、所定の頻度でマウスに候補を投与する。腹腔内注射または静脈内注射などのいくつかの経路によって候補を投与することができる。通常、4〜8週間継続し、腫瘍増殖について、毎週1回または2回動物をモニターする。腫瘍を、三次元的に(高さ×幅×長さ)測定し、腫瘍体積を算出する。マウスが腫瘍エンドポイント、例えば、1500mm3に達した、またはマウスが著しく体重減少、例えば、15%超、20%超、もしくは25%超の体重減少を示した場合、実験終了時に、マウスを通常安楽死させる。コントロールと比較して、候補治療群において腫瘍増殖がより遅い、または腫瘍エンドポイント体積に達する平均時間がより長いことを示す結果は、候補ががん増殖を阻害する作用を有することを表している。腫瘍治療における候補の能力を評価するために使用することができるインビボ有効性アッセイの1つの例示的アッセイは、実施例4に開示されている。
本明細書に開示されているGal3:TIM−3阻害剤療法は腫瘍量を低減し、有益な臨床転帰をがん患者、特にGal3を過剰発現するがんに罹患しているものに与えることができる。これらの応答の測定方法は、例えば、ctep.cancer.gov/protocolDevelopment/docs/recist_guideline.pdfで入手することができる固形がんの治療効果判定のためのガイドライン(「RECIST)に記載されているように、がん治療分野において当業者に周知である。

アメリカ培養細胞系統保存機関(ATCC、米国バージニア州マナサス)から得られたA20、マウスBリンパ腫細胞株を、FlagタグヒトGal3タンパク質またはFlagタグヒトPDL1タンパク質をコードする核酸構築物でトランスフェクトした。加えて、構築物は、抗生物質耐性マーカーを含む。形質転換された細胞を抗生物質耐性に基づいて選択し、FlagタグヒトGal3タンパク質を安定して発現するA20細胞(A20
Gal3細胞)またはFlagタグヒトPDL1タンパク質を安定して発現するA20細胞(A20 hPDL1細胞)を作製した。
本実施例は、Gal3とTIM−3との相互作用の評価を行った様々なアッセイを説明する。
TIM−3がGal3と特異的に相互作用するかどうかを試験するために、共免疫沈降実験を行った。293T細胞を、HAタグTIM−3をコードするプラスミドおよびFlagタグGal3、FlagタグGal9、またはFlagタグCEACAM1をコードするプラスミドでコトランスフェクトした。製造者プロトコールに従って、リポフェクタミン3000(米国マサチューセッツ州ウォルサム)を使用して、トランスフェクションを行った。トランスフェクトした細胞を一夜増殖させてから、洗浄し、1ml溶解バッファー中に溶解した。溶解細胞を遠心分離し、上澄み(ライセート)を回収した。ライセートを調製し、SDS PAGEで分離し、それぞれ、抗HA(図1A)および抗Flag抗体(図1B)でプローブした。抗Flagおよび抗HA抗体の両方ともSigma社から購入した。図1Aおよび1Bの矢印は、様々なタンパク質の存在を示す。
EACAM1をコードするプラスミドでコトランスフェクトされた細胞から得られたライセートからの結果を示す。
次に、細胞接着アッセイを行い、Gal3およびTIM−3間の結合を確認した。この実験では、96ウェルプレートを4℃一夜でヒトFc、ヒトPD1−Fc、ヒトVISTA−Fc、ヒトTIM−3−Fcで被覆し、それから、37℃で2時間PBS中の2%BSAで遮断した。A20、ヒトGal3を過剰発現するA20細胞(A20 Gal3)、またはヒトPDL1を過剰発現するA20細胞(A20 PDL1)を上記様々なFcタンパク質で被覆されたウェル中に播種した。それから、プレートを720rpmで遠心分離してから、止めた。プレートを37℃で30分間インキュベートし、それから、PBS中に浸した。プレートをゆっくりと180度ひっくり返し、30分間ひっくり返した位置で保持した。プレートを反転から戻した後、PBSから取り出し、各ウェルから200μlの溶液を取り出して捨て、約100μlの残った溶液を96ウェルプレート中に移動した。細胞をフローサイトメトリー分析によってカウントした。
フローサイトメトリー分析を行い、A20細胞を使用してTIM−3およびGal3間
の結合を評価した。A20 Gal3細胞を、氷上20分間、マウスTIM−3 Fcを含むかまたは含まない10%FBS HBSS溶液と共にインキュベートした。5つの実験群があった:群1では、A20 Gal3細胞をコントロールとしてmTIM−3 Fcタンパク質なしでインキュベートし;群2では、A20 Gal3細胞をmTIM−3
Fcタンパク質と共にインキュベートし;群3、4、5では、mTIM−3 Fcタンパク質に加えて、抗マウスTIM−3ポリクローナル抗体(R&D System社、米国ミネソタ州ミネアポリス)(群3)、モノクローナル抗体RMT3−23(Bio X
cell社、米国ニューハンプシャー州ウェストレバノン)(群4)、モノクローナル抗体215015(R&D System社)(群5)も加えてこれらの抗体がGal3およびTim3結合を遮断することができるかどうかを試験した。遮断のため、細胞を指定された抗体を含む10%FBS HBSSと共にインキュベートし、それから、20分間、mTIM−3 Fcを含む10%FBS HBSSと共に添加した。サンプルを遠心分離し、20分間、APC結合抗hFc抗体(Jackson ImmunoResearch社、米国ペンシルベニア州ウェストグローブ)を含む10%FBS HBSSにペレットを添加した。遠心した後、生/死細胞をバイオレット死細胞染色キット(Life
Technologies社)で染色した。染色細胞をフロー分析に付した。
ELISAも行い、Gal3とTIM−3との相互作用を試験した。96ウェルELISAプレート(ThermoFisher Scientific社)をPBS中のマウスGal3タンパク質(BioLegend社、米国カリフォルニア州サンディエゴ)またはPBS中のヒトGal9タンパク質(R&D system社)またはエタノール中のホスファチジルセリン(PS)(Sigma社)で被覆し、4℃で一夜インキュベートした。プレートをTBSTで3回洗浄し、それから、室温で1時間2%BSAを含有するPBSバッファーで遮断した。図5Aでは、種々の抗Gal3抗体、すなわち、mGal3ポリクローナル抗体(R&D system社)、mAb IMT001、mAb M3/38(Thermofisher Scientific社)(図5A)を、Gal3で被覆されたウェルに添加した。それから、抗体を10分間インキュベートしてから、TIM−3 Fcをプレートに添加し、更に1時間インキュベートした。それから、プレートを3回洗浄し、次いで、室温で1時間、抗ヒトIgG−HRP(Jackson ImmunoResearch社)と共にインキュベートした。TBSTで3回洗浄後、TMB基質(GeneTex社、米国カリフォルニア州アーバイン)で発色し、反応を1N
HClで停止した。光学密度(OD)を450nmにおいて読み取った。結果を、二重反復試験の平均OD±SDとして表した。図5Aでは、結果は、試験された全抗体の中で、マウスGal3ポリクローナル抗体およびモノクローナル抗体IMT001がGal3とTIM−3との相互作用を遮断したことを示した(図5A)。
よび2)またはエタノール中のPS(Sigma−Aldrich社、米国ミズーリ州セントルイス)(群3および4)をプレート上に被覆し、4℃で一夜インキュベートした。抗mTIM−3マウス抗体、mAb RMT3−23(Bio X Cell社)を、群2および4の被覆プレートのみに添加した。二次抗ヒトIgG−HRP抗体および基質を上記のように添加し、mTIM−3のmGal3またはPSとの結合を検出した。結果は、群3と比較して群4におけるシグナルの著しい低下を示し、RMT3−23はPSがTIM−3と結合するのを遮断したことを示し;一方、結果は、群1と比較して群2における有意な低下は示されず、RMT3−23はGal3がTIM−3と結合するのを遮断しなかったことを示す。TIM−3は死細胞表面上で外面化され暴露されたそのPSとの相互作用により死細胞と結合するので、これらの実験は、図4A〜4Cにおいて、Gal3およびPSがTIM−3の異なるエピトープと結合するという観察を実証した。
この実施例は、A20細胞内のGal3の過剰発現の機能特性を評価するために行われた実験を説明する。
インビボでのGal3:TIM−3阻害剤の抗腫瘍効率を評価するために、この実施例における実験を行った。分子医学研究所動物実験委員会によって承認されたプロトコールに従って、動物実験を行った。C57BL/6マウスを、到着次第、実験動物管理公認協会公認の施設に入れた。36匹の7週齢雌マウスを無作為に3つの群(n=12)に割り当てた。0日目、B16F10細胞(0.1mLのPBS中、2×105)を洗浄し、PBS中に再懸濁した後、27ゲージ針の注射器を使用してマウスの尾静脈内に注射した。
B16F10細胞の注射後、動物に、0、3、7および10日目に10mg/KgのマウスIgG2b(Bio X Cell社、米国ニューハンプシャー州ウェストレバノン)を腹腔内投与し、0、3および7日目にmPD1抗体(Bio X Cell社、米国ニューハンプシャー州ウェストレバノン)または0、3、7、10および15日目にGal3抗体IMT001を腹腔内投与した。この実験で使用されたGal3抗体クローンIMT001は、Gal3上のエピトープ(配列番号5)を認識する。21日目、動物を人道的に犠牲にし、肺組織を取り出し、10%緩衝ホルムアルデヒド溶液中に固定した。肺の左葉の1つの表面上の黒色転移コロニー数をカウントした(図7B)。結果は、平均±SEMとして表した。1元配置分散分析(one−way ANOVA)を使用してIgGコントロール群と比較して統計解析を行った。
HBSS溶液と共にインキュベートした。遠心した後、生/死細胞をバイオレット死細胞染色キット(Thermo Fisher Scientific社、米国マサチューセッツ州ウォルサム)で染色した。染色細胞をフロー分析に付した。図7Bは、3処置群由来の肺全体の代表的画像を示す。図7Cは、左肺葉表面上の多数の転移コロニーを示す(平均値±SEM)。図7Dおよび図7Eは、種々の処置群の肺重量および体重を示す(平均値±SEM)。アイソタイプコントロール群と比較して、Gal3抗体処置群は、黒色転移コロニー数によって示される腫瘍数の有意な(約46%)減少(p<0.01)を示した。しかしながら、アイソタイプコントロール群と比較して、抗マウスPD1抗体29Fは、この肺転移モデルにおいて有意な抗腫瘍作用を示さなかった(p>0.05)。
分子医学研究所動物実験委員会によって承認されたプロトコールに従った動物実験。7週齢雌Balb/cマウスを、到着次第、実験動物管理公認協会公認の施設に入れた。腫瘍移植の日に、4T1細胞を集め、洗浄し、PBS中に再懸濁した。吸入麻酔薬(医療用グレードの空気中3〜5%イソフルラン)によってマウスを麻酔した。0.1mL PBS中の2×105細胞を、25ゲージ針の注射器により乳腺中に皮下注射した。マウスを無作為に2つの群(n=10)に割り当てた。4T1細胞の注射後、マウスに、0、3および7日目に10mg/KgのマウスIgG2b(Bio X Cell社)または0、3、7、10および14日目にGal3抗体IMT001を腹腔内投与した。腫瘍体積および体重を毎週2回モニターした。30日目、マウスを人道的に犠牲にし、肺組織を30%ショ糖で膨張させ、取り出して、ボーインズ溶液(Sigma−Aldrich社)中に固定した。肺の左葉の1つの表面上の転移コロニー数をカウントした。結果は、平均±SEMとして表した。対応のないt検定を使用してIgGコントロール群と比較して統計解析を行った。
原発性腫瘍モデルにおいてGal3:TIM−3阻害剤の抗腫瘍効率を評価するために
実験を行った(図9)。分子医学研究所動物実験委員会によって承認されたプロトコールに従って、動物実験を行った。Balb/cマウスを、到着次第、実験動物管理公認協会公認の施設に入れた。7週齢雌マウスを無作為に3つの群(n=15)に割り当てた。腫瘍移植の日に、吸入麻酔薬(医療用グレードの空気中3〜5%イソフルラン)によってマウスを麻酔し、Renca細胞を洗浄し、PBS中に再懸濁した後、25ゲージ針の注射器を使用して0.1mLのPBS中の2×105細胞を皮下注射した。Renca細胞の注射後、マウスに、0、3および7日目に10mg/KgのマウスIgG2b(Bio X Cell社)または0、3および7日目にmPD1抗体(Bio X Cell社)または0、3、7、10および14日目にGal3抗体IMT001抗体を腹腔内投与した。コントロール群における腫瘍体積が2000〜2500mm3に達した場合、動物を人道的に犠牲にした。結果は、平均±SEMとして表した。対応のないt検定を使用してIgG2bコントロール群と比較して統計解析を行った。
分子医学研究所動物実験委員会によって承認されたプロトコールに従った動物実験。7週齢雌C57BL/6マウスを、到着次第、実験動物管理公認協会公認の施設に入れた。腫瘍移植の日に、MC38マウス大腸腺がん細胞を集め、洗浄し、PBS中に再懸濁した。吸入麻酔薬(医療用グレードの空気中3〜5%イソフルラン)によってマウスを麻酔した。0.1mLのPBS中の5×105細胞を、25ゲージ針の注射器によりマウスの右側に皮下注射した。7日目、腫瘍体積を測定し、マウスを無作為に2つの群(n=8)に割り当てた。マウスに、7、10、14、17および22日目に、10mg/KgのマウスIgG2b(BioXCell社)またはGal3抗体IMT001を腹腔内投与した。腫瘍体積および体重を毎週2回モニターした。腫瘍体積が3000mm3に達した場合、動物を人道的に犠牲にした。結果は、平均±SEMとして表した。対応のないt検定を使用してIgGコントロール群と比較して統計解析を行った。
10アミノ酸により重複し、全ヒトGal3タンパク質配列を包含する24の20アミノ酸ペプチドを含むペプチドアレイを合成した(Genscript社、米国ニュージャージー州ピスカタウェイ)(図11A)。各ペプチド20μgを、膜上にドットブロットした。PBS中の5%ミルクで遮断後、4℃で一夜、膜を、1μg/mlのIMT001抗体と共にインキュベートした。3回の洗浄後、膜を、1時間、1:8000希釈抗mIgG HRP抗体(Southern Biotech社、 米国アラバマ州バーミンガム)と共にインキュベートした。3回の洗浄後、膜を、Western ECLブロッティング基質(Bio−Rad社、米国カリフォルニア州ハーキュリーズ)と共にインキュベートし、反応させた(図11B)。ペプチド5および6は良好なシグナルを示し、IMT001が結合するhGal3上のエピトープはPGAYPGQAPPGAYPGQAPPGAYPGAPGAYP(配列番号7)であることを示した。
マウスに、百万個のB16F10細胞を静脈内に移植した。それから、マウスを、0、1、3および7日目にIMT001またはアイソタイプコントロール(10mg/kg、腹腔内)で処置し、8日目に肺免疫細胞を単離するために犠牲にして、表現型を決定した。肺から細胞を単離し、それから、リンパ球マーカーCD3、CD4、CD8、CD19、DX5に対する蛍光標識抗体で染色して、フローサイトメトリーによって分析した。図12の結果は、アイソタイプコントロール抗体処置と比較して、抗Gal3抗体IMT001処置は、腫瘍の宿主である肺におけるCD3 Tリンパ球、CD4 Tヘルパー、CD8細胞傷害性T細胞、CD19 B細胞およびDX5ナチュラルキラー細胞を含む様々な免疫エフェクター細胞数を増加させたことを示す。これは、抗Gal3抗体は免疫細胞を活性化することができたことを示す。
ヒト肺がんにおけるGal3発現を検出するために免疫組織化学(IHC)実験を行った。ヒト肺がんの凍結組織スライド(US Biomax Inc.)を、室温で10分間、10%中性緩衝ホルマリン(Fisher Scientific社)中に固定し、PBS中で5分間2回洗浄した。内在過酸化酵素を、室温で10分間、3%H2O2中にスライドを浸すことによって遮断した。PBS中で5分間2回洗浄後、スライドを、室温で15分間、ストレプトアビジン試薬(Molecukar Probes社)中でインキュベートし、次いで、PBSで徹底的に洗浄し、15分間ビオチン試薬(Molecukar Probes社)中でインキュベートし、PBS中で更に洗い流して、内在ビオチンバックグラウンドを遮断した。スライドを10%FBS、200μg/mLのmIgGおよび200μg/mLのhIgGで1時間遮断し、4℃で一夜、一次抗体IMT001−ビオチン(5μg/mL)と共にインキュベートし、3回洗浄してから、1時間1:100で二次抗体HRPアビジン(BioLegend社)と共にインキュベートし、3回洗浄した。DAB基質(Vector Laboratories社)と共にインキュベートすることによって染色を行い、蒸留水中にスライドを浸すことにより停止した。そして、ヒト肺がんスライドをヘマトキシリンQS(Vector Laboratories社)中で対比染色し、蒸留水中で洗浄し、段階的の濃度を変えた一連のエタノールおよびキシレン溶液中で脱水し、VectaMount(商標)封入剤(Vector Laboratories社)で封入した。
まずヒトCD14単球を、CD14細胞ポジティブセレクションキット(Miltenyi社、米国カリフォルニア州オーバーン)を使用して末梢血液単核細胞(PBMC)から単離し、それぞれ、GM−CSF+IL−4またはGM−CSFまたはM−CSF(Rocky Hill、米国ニュージャージー州)の存在下、樹状細胞(DC)、またはM1マクロファージ、またはM2マクロファージに分化した。それから、ヒト樹状細胞(DC)、M1およびM2マクロファージ細胞上のGal3発現を検出するために、フローサイトメトリー分析を行った。詳細には、100,000のDC、M1またはM2細胞を、氷上で20分間、10μg/mlで、コントロールmIgG−ビオチン(BioLegend社)またはIMT001−ビオチンを含む100μlの10%FBS HBSS溶液と共にインキュベートした。それから、細胞を洗浄し、氷上で20分間、1:1000でPE−ストレプトアビジン(BioLegend社)と共にインキュベートした。遠心した後、生/死細胞をバイオレット死細胞染色キット(Life Technologies社)で染色した。染色細胞をフロー分析に付した。図14の結果は、IMT001で染色されたM2細胞の平均蛍光強度(MFI)が、アイソタイプコントロール抗体で染色された細胞の平均蛍光強度よりずっと高いことを示し、IMT001がM2細胞と特異的結合するが、樹状細胞(図14A)およびM1マクロファージ(図14B)を染色することができなかったことを示す。
マウスマクロファージ上のGal3の発現を、IHCおよびフローサイトメトリー分析によって検出した。IHCの詳細では、ウェル当たり100,000細胞を一夜播種した。2日目、細胞を、PBSで1回洗浄し、室温で10分間、3%ホルムアルデヒドで固定し、それから、PBSで2回洗浄し、室温で1時間、10%FBSおよび200μg/mLを含有するPBS中で遮断した。遮断後、細胞を、40℃で一夜、10μg/mLの一次抗体mIgG−ビオチン(BioLegend社)またはIMT001−ビオチンと共にインキュベートし、PBSTで3回洗浄し、室温で1時間アビジン−HRP(1:1000)で染色し、それから、PBSTで再度3回洗浄した。過酸化酵素基質を使用して染色を発色し、ヘマトキシリンQS(Vector Laboratories社)で対比染色した。結果は、mIgGコントロール(図15A)と比較して、IMT001はマクロファージ上のGal3発現を明白に検出したことを示す(図15B)。

Claims (39)
- Gal3とTIM−3との相互作用を妨げるGal3:TIM−3阻害剤を、患者に投与することを含む、前記患者の免疫応答の活性化方法であって、前記阻害剤を、免疫応答を活性化するのに充分な量で投与する、方法。
- 前記TIM−3は免疫細胞上で発現される、請求項1に記載の方法。
- 前記患者はがんの宿主であり、前記Gal3とTIM−3との相互作用は腫瘍微小環境中で起こり、前記免疫応答の活性化は前記患者の前記がん量を低減する、請求項1または2に記載の方法。
- 前記がんは腫瘍微小環境中の細胞を含み、前記細胞は細胞表面上でGal3を過剰発現する、請求項3に記載の方法。
- 腫瘍微小環境中の細胞を含むがんの宿主である患者における免疫応答の活性化方法であって、前記細胞はその表面上でGal3を過剰発現し、前記方法は前記腫瘍微小環境中の前記免疫細胞上の前記Gal3とTIM−3との相互作用を妨げるGal3:TIM−3阻害剤を前記患者に投与することを含み、前記阻害剤を前記免疫応答の活性化によって前記患者の前記がん量を低減するのに充分な量で投与する、方法。
- 免疫細胞はT細胞であり、前記免疫応答の活性化はT細胞の活性による、請求項2または5に記載の方法。
- 前記がんは、転移がんまたは原発がんである、請求項3〜5のいずれか一項に記載の方法。
- 前記阻害剤は、TIM−3と結合する、請求項1〜7のいずれか一項に記載の方法。
- 前記阻害剤は、Gal3と結合する、請求項1〜8のいずれか一項に記載の方法。
- 前記Gal3:TIM−3阻害剤は抗体である、請求項1〜9のいずれか一項に記載の方法。
- 前記抗体は、配列番号5〜8から成る群から選択される配列を含むペプチドを認識する、請求項5に記載の方法
- 前記抗体は、一本鎖抗体またはFabである、請求項5に記載の方法。
- 前記抗体は、ヒト化抗体またはヒト抗体である、請求項5に記載の方法。
- 前記Gal3:TIM−3阻害剤の前記投与は、点滴静注による、請求項1〜13のいずれか一項に記載の方法。
- 前記Gal3:TIM−3阻害剤を、1つ以上の他の治療と併用して投与する、請求項1〜14のいずれか一項に記載の方法。
- 前記1つ以上の他の治療は、化学療法、放射線療法、チェックポイント阻害剤療法から成る群から選択される、請求項15に記載の方法。
- 前記チェックポイント阻害剤療法は、抗PD−1療法および抗CTLA4療法から成る群から選択される、請求項15または16に記載の方法。
- 前記阻害剤の前記投与を、隔週に10μg/体重kg〜100mg/体重kgの投与量で投与する、請求項1〜17のいずれか一項に記載の方法。
- 患者のがんがGal3:TIM−3阻害剤を用いた治療に適しているかどうかの決定方法であって、前記方法は:
患者の公知の型のがんの腫瘍微小環境から得られた細胞を、Gal3に対して特異的な抗体と組み合わせること;
前記細胞上のGal3レベルを決定すること;
前記細胞表面上の前記Gal3レベルを、Gal3の第一活性閾値と比較すること;及び
前記細胞表面上の前記Gal3レベルが前記第一活性閾値より高い場合に、前記患者のがんは、Gal3:TIM−3阻害剤を用いた治療に適していると判定すること、
を含む、方法。 - Gal3の前記第一活性閾値は、前記患者と同型のがんに罹患している少なくとも100試験個体のコホートから誘導される、請求項19に記載の方法。
- 前記患者のがんが治療ステップに適していると判定することは、腫瘍微小環境から得られる細胞表面上のGal3レベルがGal3の第二活性閾値と比較して25%以上であるかどうかの判定をさらに含み、前記第二活性閾値は健常な患者由来の対応する細胞を含むサンプルから誘導される、請求項20に記載の方法。
- 前記腫瘍微小環境から得られる前記細胞は、少なくともがん細胞および/または腫瘍関連マクロファージを含む、請求項19〜21のいずれか一項に記載の方法。
- 前記患者のがんが治療ステップに適していると判定することは、腫瘍微小環境から得られる細胞表面上のGal3レベルが前記第二活性閾値と比較して75%以上であるかどうかの判定をさらに含む、請求項21に記載の方法。
- がん患者のT細胞上のGal3とTIM−3との相互作用を妨げることができる滅菌液剤であって、前記溶液は1〜4時間にわたる静脈内送達に適した100mlの溶液中の患者体重キログラム当たり10μg〜100mgの抗体を含み、前記抗体はT細胞上のGal3とTIM−3との相互作用を妨げることができる、滅菌液剤。
- 前記滅菌液剤は、1つ以上の他のチェックポイント阻害抗体をさらに含む、請求項24に記載の滅菌液剤。
- 1つ以上の他のチェックポイント阻害抗体は、抗PD−1および抗CTLA−4抗体から成る群から選択される、請求項24に記載の滅菌液剤。
- 前記滅菌液剤は、10〜100nmの径を有する1つ以上のナノ粒子をさらに含む、請求項24〜26のいずれか一項に記載の滅菌液剤。
- 前記抗体は、抗Gal3抗体である、請求項24に記載の滅菌液剤。
- 前記抗体は、抗TIM−3抗体である、請求項24に記載の滅菌液剤。
- Gal3とTIM−3との相互作用を妨げることができる抗Gal3抗体の製造方法であって、前記方法は:配列番号5〜8から成る群から選択される配列を含むペプチドを動物に導入することを含み、前記動物は抗Gal3抗体を産生する、方法。
- ヒト化抗Gal3抗体であって、前記抗体は、
(1)相補性決定領域(CDR)L1、CDR L2、およびCDR L3を含む軽鎖可変領域ならびに(2)CDR H1、CDR H2、およびCDR H3を含む重鎖可変領域を含み、
前記CDR L1は、配列番号17のアミノ酸配列を含み、
前記CDR L2は、配列番号18のアミノ酸配列を含み、
前記CDR L3は、配列番号19のアミノ酸配列を含み、
前記CDR H1は、配列番号9のアミノ酸配列を含み、
前記CDR H2は、配列番号10のアミノ酸配列を含み、および
前記CDR H3は、配列番号11のアミノ酸配列を含む、
ヒト化抗Gal3抗体。 - 前記重鎖可変領域は、配列番号25のアミノ酸配列と少なくとも90%同一性を有する配列を有する、請求項31に記載のヒト化抗Gal3抗体。
- 前記軽鎖可変領域は、配列番号26のアミノ酸配列と少なくとも90%同一性を有する配列を有する、請求項31または32に記載のヒト化抗Gal3抗体。
- Gal3とTIM−3との相互作用を遮断し、患者の免疫応答の活性化および/またはがん治療をすることができる化合物の選択方法であって、前記方法は:
(a)化合物のライブラリーを、Gal3およびTIM−3と接触させること、ならびに
(b)前記Gal3とTIM−3との相互作用を遮断することができる前記ライブラリーから1つ以上の候補化合物を選択すること、
を含む、方法。 - (c)前記工程(b)から選択された1つ以上の候補化合物を、T細胞を含む混合物、および同種抗原提示細胞と接触させて、T細胞を刺激することができる1つ以上の化合物を特定すること、ならびに/または
(d)腫瘍の宿主である哺乳類に、前記(b)から選択された1つ以上の候補化合物を投与し、前記哺乳類の腫瘍量を低減することができる1つ以上の化合物を特定すること、ならびに必要に応じて、
(e)前記T細胞を刺激することができ、および/または前記哺乳類の腫瘍量を減らすことができる化合物の有効量を前記患者に投与し、それにより、前記患者の免疫応答を活性化および/またはがんを治療すること、
をさらに含む、請求項34に記載の方法。 - 前記化合物は、抗体である、請求項35に記載の方法。

- 抗体を患者に投与することを含む前記患者の免疫応答の活性化方法であって、前記抗体はGal3とTIM−3との相互作用を阻害するための手段を含む、方法。
- 前記抗体は、Gal3またはTIM−3と結合するための手段をさらに含む、請求項38に記載の方法。
