Description
本出願は、2017年7月15日に出願された米国仮特許出願第62/532,998号の優先権の利益を主張し、その内容は参照によりその全体が本明細書に組み込まれる。

拡張培地配合物には、以下を含む:(1)対照−成長培地(GF添加なし);(2)ウシ線維芽細胞成長因子(bFGF;10ng/ml);(3)上皮成長因子(EGF;62ng/ml);(3)インスリン様成長因子1(IGF−1;100ng/ml);(4)Pro−LIF;bFGF 10ng/ml、EGF62ng/ml、IGF−1 100ng/ml、およびZnCl2 50μM);(6)GFウィーニング:Pro−LIF培地で3日間、その後成長培地で3回洗浄(5分ごと)し、その後成長培地で1日。(7)細胞は、DiIで染色し、成長培地で成長させ、細胞を追跡するために追加された蛍光色であるDiIが細胞分化に影響を及ぼすかを確認した。

Claims (30)
- 食用組成物を製造するための方法であって、
a.三次元多孔性足場ならびに(i)筋芽細胞またはそれらの前駆細胞;および(ii)少なくとも1種の細胞外マトリックス(ECM)分泌細胞、または(iii)内皮細胞またはそれらの前駆細胞のうちの少なくとも1つを含む複数の細胞型をインキュベートすることであって、前記筋芽細胞またはそれらの前駆細胞および前記内皮細胞またはそれらの前駆細胞は、10:1〜1:10の範囲の比でインキュベートされる、インキュベートすることと、
b.筋芽細胞またはそれらの前駆細胞を筋管への分化に誘導することであって、
それによって、前記食用組成物が産生される、誘導することと、を含む、方法。 - 前記複数の細胞型が、筋芽細胞またはそれらの前駆細胞、少なくとも1種の細胞外マトリックス(ECM)分泌細胞、および内皮細胞またはそれらの前駆細胞を含む、請求項1に記載の方法。
- 前記ECM分泌細胞が、間質細胞、線維芽細胞、周皮細胞、平滑筋細胞およびそれらの前駆細胞からなる群から選択される、請求項1または2に記載の方法。
- 前記複数の細胞型が、筋芽細胞、ECM分泌細胞および内皮細胞を含む、請求項1〜3のいずれか一項に記載の方法。
- 前記筋芽細胞の前駆細胞が衛星細胞である、請求項1〜4のいずれか一項に記載の方法。
- 前記内皮細胞が、骨格微小血管内皮細胞、大動脈平滑筋細胞、またはそれらの組み合わせから選択される、請求項1〜5のいずれか一項に記載の方法。
- 前記複数の細胞型が、衛星細胞、ECM分泌細胞、および内皮細胞を含む、請求項1〜6のいずれか一項に記載の方法。
- 前記筋芽細胞またはそれらの前駆細胞およびECM分泌細胞が、10:1〜1:1の範囲の比でインキュベートされる、請求項7に記載の方法。
- 前記ECM分泌細胞および内皮細胞が、1:10〜1:1の範囲の比でインキュベートされる、請求項7に記載の方法。
- 前記衛星細胞、前記ECM分泌細胞、および前記内皮細胞が、10:1:1〜2:1:10の範囲の比でインキュベートされる、請求項7〜9のいずれか一項に記載の方法。
- 前記ECM分泌細胞が、線維芽細胞、その前駆細胞、またはその組み合わせである、請求項7〜10のいずれか一項に記載の方法。
- 前記三次元多孔性足場が、テクスチャ加工タンパク質、非テクスチャ加工タンパク質、および多糖類からなる群から選択される、請求項1に記載の方法。
- 前記テクスチャ加工タンパク質が、テクスチャ加工大豆タンパク質である、請求項12に記載の方法。
- 前記三次元多孔性足場が、20〜1,000マイクロメートルの範囲の平均直径を有する細孔を含む、請求項12または13に記載の方法。
- 前記複数の細胞型が非ヒト細胞である、請求項1〜14のいずれか一項に記載の方法。
- 前記複数の細胞が家畜哺乳類に由来する、請求項1〜15のいずれか一項に記載の方法。
- 前記筋芽細胞またはそれらの前駆細胞および前記三次元多孔性足場は、前記筋芽細胞またはそれらの前駆細胞が、前記三次元多孔性足場10mgに103〜107の範囲の比でインキュベートされる、請求項1〜16のいずれか一項に記載の方法。
- 前記三次元多孔性足場が細胞外マトリックスをさらに含む、請求項1〜17のいずれか一項に記載の方法。
- a.三次元多孔性足場と、
b.前記三次元多孔性足場1mm3あたり10,000〜250,000個の筋管核を含む筋管と、
c.複数の細胞型であって、(i)筋芽細胞またはそれらの前駆細胞;および(ii)少なくとも1種のECM分泌細胞;または(iii)前記複数の細胞の15%未満を構成する、内皮細胞またはそれらの前駆細胞のうちの少なくとも1つからなる群から選択される複数の細胞型と、を含む、組成物。 - 前記複数の細胞型が、筋芽細胞またはそれらの前駆細胞、少なくとも1種の細胞外マトリックス(ECM)分泌細胞、および内皮細胞またはそれらの前駆細胞を含む、請求項19に記載の組成物。
- 前記筋芽細胞の前駆細胞が衛星細胞である、請求項19または20に記載の組成物。
- 前記複数の細胞型が、衛星細胞、ECM分泌細胞、および内皮細胞を含む、請求項19〜21のいずれか一項に記載の組成物。
- 前記ECM分泌細胞が、線維芽細胞、その前駆細胞、またはその組み合わせである、請求項19〜22のいずれか一項に記載の組成物。
- 前記三次元多孔性足場が、テクスチャ加工タンパク質、非テクスチャ加工タンパク質、および多糖類からなる群から選択される、請求項19〜23のいずれか一項に記載の組成物。
