本発明は、腫瘍細胞に対する老化および/または細胞共食いおよび/または免疫応答を誘導するための、特定のナノ粒子と電離放射線との組み合わせ効果の、本発明者らによって実証された驚くべき利点に由来する。
ナノ粒子は、放射線増感剤として作用する高Z元素、例えば、40より大きい、例えば50より大きい原子番号Zを有する元素を含有する。特定の実施形態では前記高Z元素が重金属の中から選択され、より好ましくはAu、Ag、Pt、Pd、Sn、Ta、Zr、Tb、Tm、Ce、Dy、Er、Eu、La、Nd、Pr、Lu、Yb、Bi、Hf、Ho、Pm、Sm、In、およびGd、ならびにそれらの混合物の中から選択される。
−PETまたはSPECTシンチグラフィー、

細胞、試薬およびガドリニウム系ナノ粒子
大腸癌(p53+/+、p53R248W/+およびp53R248/−)HCT116細胞を、10%熱不活化ウシ胎児血清(Hycultec GmbH)、2mMのL−グルタミンおよび100IU/mLペニシリン−ストレプトマイシン(Life technology)を添加したMcCoyの5A培地(Life technology)で維持した。ヒト大腸癌変異体HCT116p53R248W/−、p53R248W/+およびHCT116p53+/+を、Dr.Christophe Bourdonから入手した。ベンジルオキシカルボキシル−Val−Ala−Asp(OMe)フルオロメチルケトン(Z−VAD−fmk)は、Bachemから入手した。ROCK1阻害剤(Y27632)およびN−アセチルシステイン(NAC)はSigma製であり、マンガン(III)−テトラキス(4−安息香酸)ポルフィリン(MnTBAP)はMerck chemicals製であった。ガドリニウム系ナノ粒子(GdBN)は、NH TherAguixから入手した。
ROCK1に特異的な低分子干渉RNA(siRNA)(siRNA−1 ROCK1:5’ GCC GCC GGG ACC CAA CUA U3’; siRNA−2 ROCK1:5’ GGA AUC CAG UUG AAU ACA 3’)およびコントロールsiRNA(siRNA−1 Co.:5’ GCC GGU AUG CCG GUUU AAG U 3’)をSigmaから入手した。SMARTpool siGENOME NOX5 siRNA(siRNA NOX5)(D−010195−05)は、4個のsiRNA(siRNA−1:5’ GGA GCA AGG UGU UCC AGA 3’; siRNA−2:5’ CUA UAG ACC UGG UGA CUA 3’; siRNA−3:5’ GCU AU U GGG CUA GGU A 3’およびsiRNA−4:5’ CCU U CUUG CAG AGC GAU 3’)を含む。コントロールsiGENOME非標的siRNA(siRNA−2 Co.として示される)は、4つのオン標的プラス非標的siRNAのプールである(D-0012206-13-05)。SMARTpool siGENOME NOX5 siRNAおよびsiGENOME非標的siRNA Pool#1は、Dharmaconから購入した。RNA干渉のために、HCT116細胞を、siRNA形質導入の48時間前に播種した(6ウェルプレート中で5.0×105細胞/2mL/ウェル)。次いで、細胞を、Lipofectamine RNAi max(#13778150、Life technologies)を製造者の指示に従って使用して10nMのsiRNAで形質導入し、その後の実験の前に37℃で24時間インキュベートした。
HCT116細胞を6ウェルプレートに播種し、37℃で1時間、示された濃度のGdBNと共にインキュベートした。次に、X線照射器(1Gy/min,200keV,15mA,2mm銅厚、X-RAD 320、Precision X-Ray)またはγamma線照射器(IBL-637,Cs137,1Gy/min,gamma CIS-BioInternational、IBA、Saclay、France)で細胞を照射した。その後の実験のために、照射後の指示された時点で細胞を回収した。
細胞共食いは既報13の通り決定した。簡単に述べると、処理細胞を10μMの5−(および6)−(((4−クロロメチル)ベンゾイル)アミノ)テトラメチルローダミン(Cell Tracker Orange CMTMR、Invitrogen)で染色し、未処理細胞を10μMの5−クロロメチルフルオレセインジアセテート(Cell tracker Green CMFDA、Invitrogen)で染色した。次いで、細胞を、ROCKの薬理学的阻害剤Y27632(30μM、TOCRIS)またはpanカスパーゼ阻害剤zVAD−fmk(100μM、Calbiochem)の存在下で、示された時間共培養する。単一および共食い細胞上でのサイクリン依存性キナーゼ阻害剤p21の特異的細胞下染色のために、細胞を4%パラホルムアルデヒド/リン酸緩衝生理食塩水(PBS)中で10分間固定し、PBS中の0.3% Triton X−100(Sigma)中で透過性にし、5% FCS−PBSと共に1時間インキュベートした。ヒトp21に対するウサギ抗体(form Cell Signaling Technology、#2947)を、1mg/mlのBSAを含有するPBS中での免疫検出に使用し、Invitrogenのアレクサ488蛍光色素に結合したヤギ抗ウサギIgGで明らかにした。細胞をHoechst 33342(Invitrogen)で対比染色し、Zeiss LSM510での蛍光共焦点顕微鏡により、または63×対物レンズを用いたLEICA DMi8での蛍光顕微鏡により分析した。フローサイトメトリーによる細胞死の誘導を検出するために、細胞を37℃および5%のCO2で30分間、40nMの3,3’−ジヘキシルオキサカルボシアニン、DIOC6(3)、および2μg/mLのヨウ化プロピジウム(Invitrogen、分子プローブ、OR、USA)で染色した。サイトフルオロメトリー測定をGuava EasyCyte(Millipore EMD)で行い、Incyteソフトウェア(Millipore)により分析した。細胞周期分布は既報14に従い、10μg/mlのHoechst 33342(Invitrogen)で評価した。LSR II(商標)フローサイトメーター(Becton-Dickinson)を用いて、細胞蛍光を定量した。蛍光ヒストグラムをKaluzaソフトウェアバージョン1.5で分析した。
所定の時間に、細胞を固定し、既報13の通り、老化β−ガラクトシダーゼ染色キット(Cell signaling technologies)を用いて染色した。
全細胞溶解物を、プロテアーゼおよびホスファターゼ阻害剤カクテル(EDTAフリー阻害剤、Roche−diagnostics、Meylan、France)を補充した溶解緩衝液(0.1%のNP40;200mMのHEPES;100mMのKCl;1mMのEDTA;1%グリセロール最終濃度)中で調製した。タンパク質抽出物を、Bradfordアッセイ(Bio−Rad Laboratories、Hercules、CA、USA)を用いて定量した。タンパク質抽出物(20〜40μg)を4〜12%のNuPAGE Bis−trisゲル(Invitrogen)上に流し、4℃でニトロセルロース膜上に移した。ブロッキング後、膜を、製造業者の説明書に従って、一次抗体と共に4℃で一晩インキュベートした:p21WAF1/CIP1(Cell signaling #2947);NOX−5(Abcam#191010);切断カスパーゼ−3(Cell signaling #9664);MLC2S19*(Cell signaling #3671)またはMLC2(Cell signaling #8505)。次いで、西洋ワサビペルオキシダーゼ結合ヤギ抗マウスまたは抗ウサギ(Southern Biotechnology)抗体を1時間インキュベートし、直接化学発光画像スキャニング(G:BOX Syngene)を用いて増強化学発光プライム検出(Amersham、GE−Healthcare)で明らかにした。GAPDH(#MAB374、EMD Millipore)をローディングコントロールとして使用した。
細胞をカバーガラス皿(World Precision Instruments)に播種した。細胞内ROSレベルを、蛍光プローブ(2,7−ジクロロヒドロ−フルオレセインジアセテート(H2DCFDA))を用いて、製造者の指示書(Invitrogen)に従って評価した。細胞をHoechst 33342(Invitrogen)で対比染色し、蛍光顕微鏡(Leica DMi8、63x対物レンズを使用)によって分析した。
NOX5サイレンスHCT116クローンを、GIPZレンチウイルスベクター中で発現されたNOX5遺伝子に対する3つの特定小ヘアピンRNA(shRNA)のプールを用いて行った。NOX5に特異的なshRNA(sh1NOX5(クローンID、V3LHS 353964:5’ CTTGGACCCTTCGATCCA3’;sh2NOX5(クローンID、V2LHS 136069):5’ TAGAACCTCAAAAGTGGC3’;sh3NOX5(クローンID、V2LHS 136068):5’ ACAAAGTCAGTGTGAG3’)およびコントロールshRNA(shControl:5’ GCCGGGUAGCCGUGUUAAGU3’)は、Dharmaconから購入した。コントロールクローンは、非サイレンシングGIPZコントロールレンチウイルスベクター(クローンID:RHS4346、Dharmacon)を用いて実施した。
NOX5 mRNAの検出には、NOX5遺伝子(Hs00225846_m1)およびGAPDH(Hs02758991_g1)の予め設計されたApplied Biosystemsプローブ(正規化のため)を使用した。これらのプローブは、Applied Biosystemsから入手したpremade TaqMan Gene発現ミックスに含まれた。結果を周期しきい値法(CT)で分析し、それぞれの試料を内因性GAPDH mRNAの量に対して正規化した。
NOX5を枯渇させた(shNOX5)またはさせなかった(shControl)ヒト結腸直腸HCT116細胞を、37℃で1時間、1.2mMのGdBNで前処理し、単一線量が6GyのX線(XR)で1回照射した。次に、2.5 106 shControlまたはshNOX5 HCT116細胞を6週齢のスイスヌードマウス(Charles River Laboratories)に移植し、腫瘍増殖を経時的にモニターした。実験は、無作為化されず、EU Directive 63/2010に従って実施され、Gustave Roussy Cancer Campus(CEEA IRCIV/IGR n°26)のEthical Committeeによって承認された。
サンプルサイズを予め決定するための統計的方法は使用されなかった。統計的有意性は、一方向ANOVA試験を用いて決定した。統計的に有意な値は、図の説明で報告される。全ての実験は、独立して少なくとも3回行った。データは、平均±SEMとして表す。GraphPad Prismバージョン6.0b(GraphPad Software)を用いて統計解析を行った。
ガドリニウム系ナノ粒子と電離放射線との組み合わせは、細胞老化を誘導する
我々が以前に報告したように13,14、癌細胞は、細胞老化と同時に誘導される可能性のある別個の致死モダリティ(NCADまたはCAD死を含む)を介して電離放射線後に排除される。ガドリニウム系ナノ粒子の存在下での癌細胞の照射の生物学的効果を決定するために、本発明者らは最初に、癌細胞の老化を誘導するこの治療の能力を分析した。従って、ヒト結腸直腸HCT116細胞を、1.2mMのガドリニウム系ナノ粒子(GdBN)で1時間前処理し、次いで、単一線量が6GyのX線(XR)で1回照射した。2日後、細胞質老化関連β−ガラクトシダーゼ(senescence-associated β−galactosidase:SA−β−Gal)活性、サイクリン依存性キナーゼ阻害蛋白質p21の発現、および処理HCT116細胞の細胞周期進行を分析した。我々は、HCT116細胞をXRで処理した後、コントロールと比較してSA−β−Gal+細胞の頻度の有意な増加を検出し、また、GdBN+XRの組合せがXRで処理した後に検出されるSA−β−Gal+細胞の数を有意に増加させることに特に注意を払った(図1aおよび1b)。これらの結果は蛍光顕微鏡(図1cおよび1d)およびウェスタンブロット(図1e)によってp21の発現の増加を検出することによって確認され、ガドリニウム含有粒子と電離放射線との組み合わせが、XR後の老化の誘導に対して癌細胞を感作することを実証する。並行して、本発明者らはまた、フローサイトメトリーを使用して、6Gy単独で、または1.2mMのGdBNと組み合わせて照射され、そして24時間および48時間培養されたHCT116細胞の細胞周期分布を分析した。コントロールHCT116細胞と比較して、XR単独またはGdBN+XRで処理したHCT116細胞のG2/M期における有意な蓄積が、24時間のインキュベーション後に検出されたが、48時間のインキュベーション後には検出されなかった(図1f〜1h)。これらの結果は、本発明者らがXRとGdBNとを結び付けた後に検出したp21発現の有意な増加と一致する(図1dおよび1e)。転写因子p53が、p21発現の制御を介して、細胞老化15,16の誘導中の長期G2停止を引き起こす可能性を考慮して、野生型(p53+/+)、p53に対して変異または/および転写不活性化された(p53R248W/+またはp53R248W/−)HCT116細胞を、6Gy単独または1.2mMのGdBNとの併用で照射し、p21発現およびSA−β−Gal活性について48時間後に分析した。予測されたように、本発明者らは、p21発現(図1f)およびSA−β−Gal活性(図1g)がXRおよびGdBN+XR治療後に有意に減少することを観察し、p53の転写活動がXR−またはXR+GdBN−誘導細胞老化の両方に必要であることを示した。まとめると、これらの結果は、ガドリニウム含有ナノ粒子の存在下での癌細胞の放射線治療が細胞老化の誘導を通して照射された癌細胞の排除に有利であることを実証する。
GdBNおよびXRを用いた癌細胞の組み合わせ治療によって誘導される致死プロセスをさらに特定するために、本発明者らは、細胞自律および非細胞自律死を誘導するこの治療の能力を決定した。この文脈において、既報13,14の通り、我々は最初に、5−(および−6)−(((4−クロロメチル)ベンゾイル)アミノ)テトラメチルローダミン(CMTMR)、赤)蛍光バイタルプローブで標識化された未処理HCT116細胞と、5−クロロメチルフルオレセインジアセテート(CMFDA、緑)蛍光バイタルプローブで標識化された等原性HCT116細胞との間で、共培養を実施し、示された濃度のGdBNの存在下(または非存在下)で、異なる線量で照射した。24時間の共培養後、CMFDA+細胞およびCMTMR+細胞の細胞運命を、細胞飲み込みについて分析した。共焦点解析の結果、照射(XR)またはGdBN+XR処理CMTMR+HCT116細胞と未処理CMFDA+HCT116細胞との共培養中(図2b)、細胞飲み込みが検出された。このプロセスは線量依存的に起こり(図2c)、癌細胞をγ線照射した場合にも観察される(図2d)。細胞の飲み込みに関与する生物学的機構をさらに特徴付けるために、本発明者らは、細胞侵入または細胞共食い作用機構のいずれかを誘導するこれらの治療の能力を試験した。蛍光顕微鏡を用いて、我々は、照射(XR)およびGdBN+XR治療CMTMR+HCT116細胞が、隣接(CMFDA+またはCMTMR+)HCT116細胞が受けた治療とは無関係に、隣接(CMFDA+またはCMTMR+)HCT116細胞を内在化することを観察した(図2e)。本発明者らは、照射された細胞と比較して、GdBN+XR治療細胞の共食い活性の有意な増加に注目し、癌細胞のGdBNおよびXRの組み合わせ治療が、照射細胞の共食い活性を示す能力を増加させることを明らかにした。これらの実験と並行して、本発明者らはまた、照射された細胞が他のCADモダリティ(例えば、アポトーシスおよび壊死)を同時に受ける能力を評価した。処理細胞のミトコンドリア膜および細胞膜の透過性を(フローサイトメトリーによる3,3′−ジヘキシルオキサカルボシアニンヨージドおよびヨウ化プロピジウム(DiOC6(3)/IP)染色の同時検出を介して)分析することにより、XRまたはGdBN+XR処理に反応して検出した細胞共食いがアポトーシスまたは壊死の有意な誘導なしに誘導されることを観察した(図2f−2j)。まとめると、これらの結果はガドリニウム含有ナノ粒子で癌細胞を治療した後に、照射された細胞の細胞共食いを増強することができるという事実を強調している。
上記の細胞共食いに関与する分子メカニズムを正確にするために、本発明者らは、次いで、隣接癌細胞の生きた細胞飲み込みに関連するシグナル伝達経路を研究した。本発明者らは最初に、XRまたはGdBN+XR治療後に観察された細胞飲み込みが、(XRまたはGdBN+XR治療後のカスパーゼ−3のプロアポトーシス切断の非存在によって明らかにされるように(図3a))細胞死誘導の非存在下で生じ、そしてpanカスパーゼ阻害剤である、Z−VAD−fmkによって阻害されなかったことを明らかにし(図3c)、このプロセスが、死んだ細胞のアポトーシス取り込みに依存せず、そして生きた癌細胞の内在化を可能にすることを示した。さらに、本発明者らは、HCT116細胞のGdBN、XRでの治療、またはGdBNおよびXRでの併用治療が(セリン19(MLC2S19*)上のミオシン軽鎖2のリン酸化によって明らかにされるように)キナーゼROCK1を活性化することを示した(図3b)。本発明者らは、GdBNの存在下または非存在下での癌細胞の照射後にMLC2S19*の有意な増加したリン酸化を観察し(図3c)、ROCK1の活性化がGdBN+XR治療後に検出された隣接癌細胞の取り込みに必要であり得ることを示した。XRまたはGdBN+XR媒介細胞飲み込み中のROCK−1依存性シグナル伝達経路の寄与を決定するために、1.2mMのGdBNの存在下(または非存在下)で6Gyを照射したHCT116細胞の共培養にROCK1の薬理学的阻害剤(Y27632)を添加し、24時間の共培養後の細胞共食いの頻度を決定した。本発明者らは、Y27632によるROCK1の薬理学的阻害(図3bで明らかにされるように)が6Gy単独照射後、および1.2mMのGdBNの存在下でのHCT116細胞の照射後に検出される細胞共食いを阻害し(図3c)、XRまたはGdBN+XR治療後に検出される細胞共食いにROCK1の生物学的活性が必要であることを明らかにした。次いで、我々は、飲み込まれた(標的)細胞または飲み込んだ(共食い)細胞のいずれかで特異的な2つの低分子干渉RNA(siRNA)でROCK1を枯渇させることによって、GdBN+XR誘導細胞共食いに対するキナーゼROCK1の寄与をより良く定義した。我々は、双方の相互作用細胞上のROCK1の枯渇がXRまたはGdBN+XR治療後に検出される細胞共食いの頻度を減少させたことを観察し(図3dおよび3e)、キナーゼROCK1がGdBN+XR媒介細胞共食いの誘導の間、飲み込まれた標的細胞および飲み込んだ共食い細胞の両方において中心的な役割を果たすことを示した。
XRまたはGdBN+XR治療後に検出された飲み込まれた細胞および共食い細胞の両方の運命を決定するために、蛍光顕微鏡により、1.2mMのGdBNの存在下(または非存在下)で6Gyで照射されたHCT116細胞の24時間共培養後に観察された、飲み込まれたHCT116細胞の核におけるクロマチン凝縮および核断片化の存在を決定した。本発明者らは、照射のみの後、またはGdBNの存在下での癌細胞の照射後に検出された全ての飲み込まれた細胞が標的細胞分解の徴候を示したことを観察した(図4aおよび4d)。興味深いことに、本発明者らはpanカスパーゼ阻害剤であるZ−VAD−fmkが標的細胞分解を抑制できなかったことを実証し(図4d)、カスパーゼの活性化はX線照射またはX線照射とGdBNとの組み合わせによって誘導される細胞共食いの実行に必要ではないことを示した。さらに、XR−またはGdBN+XR誘導共食い細胞はp21発現(図4bおよび4e)およびSA−β−Gal+活性(図4cおよび4f)の増大を示すことも明らかにし、XR−またはGdBN+XR治療後、共食い細胞がエンテセンス(entescence)を受け得るか、あるいは単一老化細胞が隣接細胞を内在化し得ることを示した。エンテセンスの誘導または老化細胞の共食い活性の活性化がXRまたはGdBN+XR治療後に検出された共食い活性と関連するかどうかを決定するために、XRまたはGdBN+XR治療後にSA−β−Gal+活性を示した単細胞および共食い細胞の頻度を測定した。SA−β−Gal+細胞の大部分は単細胞であることが観察され(図4g)、細胞共食いは、XRまたはGdBN+XRで治療された癌細胞の細胞自律的老化の誘導後に起こるようことを示唆している。
活性酸素種(ROS)産生が電離放射線後の老化誘導に関与する主要な細胞ストレスの1つであることを考慮して17、XRおよびGdBN+XR治療後のROS産生を分析した。蛍光顕微鏡を用いて非蛍光色素2,7−ジクロロヒドロフルオレセインジアセテート(H2DCFDA)の蛍光2,7−ジクロロヒドロフルオレセイン(DCF)への転換を検出することにより、XRまたはGdBN+XR治療後に得られた単細胞および共食い細胞のROS生成能を測定した。本発明者らは、これらの治療が単一細胞および共食い細胞の両方においてROSの産生を誘導したことを観察した(図5aおよび5b)。さらに、ROS産生を鈍らせることが知られている抗酸化剤N−アセチルシステイン(NAC)およびスーパーオキシドジスムターゼ(SOD)模倣Mn(III)テトラキス(4−安息香酸)(MnTBAP)は18、XR単独またはΓδBN+XR併用によるHCT116細胞の治療後に検出される細胞老化(p21発現の検出によって明らかにされるように(図5c))を低下させることを明らかにした。これらの結果は、ROS産生がXRおよびGdBN+XR治療に関連する老化および細胞共食いの誘導に重要な役割を果たすことを実証した。
腫瘍増殖に対するNOX5不活性化の影響を特徴付けるために、短いヘアピンRNAを用いたRNA干渉により枯渇した(shNOX5)または枯渇していない(shControl)ヒト結腸直腸HCT116細胞(図6a)を、37℃で1時間、1.2mMのGdBNで前処理し、次いで、単一線量が6GyのX線(XR)で1回照射した。腫瘍細胞をBalb/c Nudeマウスに皮下注射し、腫瘍増殖を評価した。我々は、GdBN治療マウスまたはコントロールマウスと比較して、電離放射線が癌細胞移植後の腫瘍の増殖を損なうことを観察し、ガドリニウム系粒子と電離放射線(GdBN+XR)との組み合わせが、腫瘍増殖を停止させるXRの能力を増強することを確認した(図6b)。我々はまた、NOX5の不活性化が、NOX5を枯渇させたコントロール、GdBN治療、XR治療またはGdBN+XR治療HCT116細胞の移植後に得られた腫瘍の増殖を強く損なうことを観察した(図6c)。これらの結果は、NOX5が腫瘍移植またはGdBN単独での治療によって、およびXR単独またはGdBN+XRの腫瘍抑制活性によって誘導される細胞ストレスに対して癌細胞を保護することを示す。全体として、これらの結果は、GdBN+XRとNOX5不活性化との組み合わせが放射線治療の効率を強く改善することを実証する。

Claims (17)
- 治療を必要とする対象における腫瘍を治療する方法において使用するためのナノ粒子であって、該方法は、
a.有効量のナノ粒子の懸濁液を、治療を必要とする対象の腫瘍に投与するステップであって、前記ナノ粒子は40を超える、好ましくは50を超える原子番号Zの元素を含み、10nm未満、好ましくは5nm未満、例えば1〜5nmの平均流体力学的直径を有する、投与するステップと、
b.前記ナノ粒子を含む前記腫瘍を、効率的な線量の電離放射線に曝露するステップと、を含み、
前記電離放射線と前記ナノ粒子との組み合わせ効果は、照射された腫瘍細胞に老化および/または細胞共食いを誘導する、ナノ粒子。 - 前記腫瘍が少なくとも3Gy、好ましくは3Gy〜9Gy、より好ましくは5〜7Gyの分割当たりの電離放射線の線量に曝露され、総線量が好ましくは最大10分割で投与される、請求項1に記載のナノ粒子。
- 前記方法が前記ステップaおよびbの前に、
i.対象の腫瘍におけるNOX5および/またはROCK1発現レベルまたは活性を決定するステップと、
ii.得られた発現値または活性を対応するコントロール値と比較するステップと、
iii.NOX5および/またはROCK1発現レベルまたは活性がコントロール値と実質的に同一またはそれより高い腫瘍を有する対象の中から対象を選択するステップと、をさらに含む、請求項1または2に記載のナノ粒子。 - 前記方法がNOX5および/またはROCK1活性のエンハンサーまたはモジュレーター剤を、電離放射線への曝露ステップの前に、またはそれに付随して、またはその後に投与するステップをさらに含む、請求項1または2に記載のナノ粒子。
- 治療ステップの前に、腫瘍におけるNOX5および/またはROCK1発現レベルまたは活性を決定するステップをさらに含む、請求項1、2または4に記載のナノ粒子。
- 前記電離放射線と前記ナノ粒子との組み合わせ効果が、前記腫瘍細胞に対する、NOX5活性によって媒介される免疫応答を誘導する、請求項1〜5のいずれか1項に記載のナノ粒子。
- 電離放射線への暴露ステップの前、またはそれに付随して、またはその後に、免疫治療剤を投与するステップをさらに含む、請求項1〜6のいずれか1項に記載のナノ粒子。
- 前記免疫治療薬が、PD1/PDL1阻害剤およびCTLA4阻害剤などの免疫チェックポイント阻害剤の中から選択される、請求項7に記載のナノ粒子。
- 治療される腫瘍が、アポトーシスを誘導する化学療法治療に抵抗性であることが示されている腫瘍の中から選択される、請求項1〜8のいずれか1項に記載のナノ粒子。
- 非照射細胞が、隣接する照射された細胞の細胞共食いによってさらに殺される、請求項1〜9のいずれか1項に記載のナノ粒子。
- 腫瘍細胞中の老化細胞の数が、ナノ粒子の存在なしに、同じ電離放射線の曝露によって誘導された老化細胞の数と比較して、少なくとも10%、20%、30%、40%、または少なくとも50%の倍率で治療後に増加する、請求項1〜10のいずれか1項に記載のナノ粒子。
- 細胞共食いが、ナノ粒子の存在なしに、同じ電離放射線への曝露によって観察される細胞共食いと比較して、少なくとも10%、20%、30%、40%、または少なくとも50%の倍率で増強される、請求項1〜11のいずれか1項に記載のナノ粒子。
- 前記電離放射線に曝露される腫瘍の体積が、治療される腫瘍の総体積よりも小さく、例えば、少なくとも(体積で)10%小さく、または少なくとも(体積で)20%小さい、請求項1〜12のいずれか1項に記載のナノ粒子。
- 前記方法が、前記電離放射線に曝露された領域の外側に位置する腫瘍の治療をさらに可能にする、請求項1〜13のいずれか1項に記載のナノ粒子。
- 前記ナノ粒子が、高Z元素として希土類金属、好ましくはガドリニウムを含む、請求項1〜14のいずれか1項に記載のナノ粒子。
- 前記ナノ粒子が、高Z元素のキレート、例えば希土類元素のキレートを含む、請求項1〜15のいずれか1項に記載のナノ粒子。
