Abstract
Description
〔1〕GSK3阻害剤と、肝細胞増殖因子受容体の活性化剤、副腎皮質ホルモン、カルシトリオール、及びジメチルスルホキシドからなる群から選ばれる少なくとも1種とを含むことを特徴とする腸細胞分化用培地。
(1)多能性幹細胞を、アクチビン受容体様キナーゼ-4,7の活性化剤を含む培地中で培養する工程、
(2)工程(1)で得られた細胞を、GSK3阻害剤及びγ-セクレターゼ阻害剤を含む培地中で培養する工程、
(3)工程(2)で得られた細胞を、〔1〕乃至〔4〕のいずれかに記載の腸細胞分化用培地中、高密度コラーゲンゲル膜上で培養し、腸細胞を得る工程。
(A)腸細胞分化用培地
本発明の腸細胞分化用培地は、GSK3阻害剤と、肝細胞増殖因子受容体の活性化剤、副腎皮質ホルモン、カルシトリオール、及びジメチルスルホキシドからなる群から選ばれる少なくとも1種とを含むことを特徴とするものである。
本発明の腸細胞の作製方法は、下記の工程(1)〜(3)を含むことを特徴とするものである。
工程(1)では、多能性幹細胞を、アクチビン受容体様キナーゼ-4,7の活性化剤を含む培地中で培養する。
a)細胞接合部にタイトジャンクション関連タンパク質であるZO1を発現する。
b)単層膜を形成する。TEER値の測定により、この単層膜は十分なバリア機能を有することが確認されている。
c)取り込みトランスポーター遺伝子(例えば、SLC15A1)及び排出トランスポーター遺伝子(例えば、ABCB1及びABCG2)が成体小腸と同等なレベルで発現している。
d)ジゴキシン及びプラゾシンの基底側から頂端側への経細胞方向性輸送が観察される。
e)CYP酵素(例えば、CYP2C9、CYP2C19、CYP3A4)遺伝子発現量が正常成体レベルの発現量に近いレベルである。
f)VILLINを発現し、その発現は頂端側に局在する。
(A)結果
(1)コラーゲンビトリゲルは、ヒトiPSCが、膜形成及び腸のマーカー発現を特徴とする腸細胞へ分化することをサポートする。
ヒトiPS細胞の成熟腸細胞への分化を誘導する試みにおいて、本発明者は最初にM15細胞上でヒトiPSCから胚体内胚葉(DE)細胞を誘導した。3日目(D3)のDE細胞を解離させ、CVMインサート上に置き換え、15日目までM2培地で培養した。その後、培地を成熟培地(M3、M3-1、又はM3-2)に交換し、40日目まで培養した(図1A、図6A)。あるいは、3日目DE細胞をiMatrix511プレコートプレート上に再プレーティングし、M2培地中で10日目まで培養し、次いで10日目の腸前駆細胞ストックとして凍結保存した。使用時に、凍結保存した10日目細胞ストックを凍結融解し、CVMインサート上に播種し、15日目までM2中で培養し、次いでM3-1又はM3-2培地に交換した。3日目DEは凍結細胞ストックとして保存することもできた。本発明者の実験では、3日目DE細胞ストック又は10日目腸前駆細胞ストックのいずれかを使用した。免疫細胞化学分析の結果、CVM上にプレーティングした4日目のDE細胞はSOX17発現を示し、その後10日目にCDX2発現が上昇することが明らかになった(図1B)。
次に、定量的PCR分析により、腸のマーカー、トランスポーター及び代謝酵素の発現を分析した。この一連の実験では、凍結保存された3日目のDE細胞を使用した。凍結保存された3日目のDE細胞を、CVMインサート上にプレーティングし、最初にM2培地で培養し、次いでM3培地に交換し、分化の40日目まで培養することによって、腸分化を誘導した(図2A)。
次に、hiPSC由来腸前駆細胞の分化能を試験した。この実験では、凍結保存された3日目DE細胞、10日目及び15日目の hiPSC由来腸前駆細胞を使用した。これらの細胞をCVMインサート上に播種し、35日又は40日まで培養し、その後回収し、腸マーカー、トランスポーター及びCYP酵素をリアルタイムPCRによって分析した(図3A)。hiPSC由来10日目及び15日目の腸前駆細胞は分化し、低レベルのLGR5発現を示した。3日目DE細胞及び10日目腸前駆細胞は、35日目でCDX2及びVILLIN上を同様のレベルで発現する腸細胞を生じる。しかし、15日目の腸前駆細胞由来の40日目の腸細胞は、3日目のDE細胞由来のものより高いレベルでCDX2又はVILLINを発現した(図3B)。この結果は、10日目及び15日目のhiPSC由来腸前駆細胞の凍結保存された細胞ストックが、CVM上で腸細胞に分化できたことを示唆している。
結果は、hiPSC由来の腸細胞が、腸細胞と類似する腸マーカー、トランスポーター及びCYP酵素を発現することを示している。次に、hiPSC由来腸細胞のマーカーの発現レベルを、Caco-2細胞の発現レベルと比較した。Caco-2細胞は、薬剤開発プロセスにおける薬物の腸内吸収特性を評価するための最も一般的に使用されるヒト腸細胞モデルである。Caco-2細胞をCVM 24ウェルインサート上にプレーティングし、21日間培養した。2つの異なるhiPSC細胞株(RPChiPS771及びChiPS18)由来の3日目のDE細胞をCVM上にプレーティングし、それぞれ18日間培養し(分化日数は21日)、分子マーカーの発現を調べた(図4A)。
ABCG2及びABCB1 mRNAはhiPSC由来の腸細胞で高度に発現するので、細胞培養インサート上の腸細胞における排出トランスポーター(P-gp、BCRP)の輸送活性を調べるために双方向における経細胞輸送アッセイを行った(図5A、右)。P-gpの典型的な基質であるジゴキシンの基底側から頂端側への方向性の輸送(BからAへの輸送)は、時間依存的に、頂端側から基底側への輸送(AからBへの輸送)より高いレベルで観察された(図5A、左パネル)。一方、P-gpの典型的な阻害剤であるベラパミルの存在下では、ジゴキシンのBからAへの輸送は、AからBへの輸送とほぼ同じであった。同様に、BCRP及びP-gpの基質であるプラゾシンのBからAへの輸送は、AからBへの輸送よりも高いことが判明した。BCRP及びP-gp両トランスポーターの阻害剤であるエラクリダーの存在下では、プラゾシンのBからAへの輸送は減少したが、AからBへの輸送は増加した。したがって、これらの結果は、hiPSC由来の腸細胞が、腸管排出トランスポーターであるP-gp及びBCRPの輸送活性を示すことを示唆した(図5A、中央パネル)。
上記の分化法において、本発明者は、hiPS由来腸細胞が15日目に一定レベルでトランスポーターABCB1を発現し(図2C)、15日目以降ABCG2及びSLC15A1の発現を増加させることを見出した。これらのhiPS由来腸細胞はM3中で培養した後、15日目から薬物代謝酵素も発現し始める(図2D)。前駆細胞の成熟をトランスポーター及び代謝酵素を発現する成熟腸細胞に導くために重要である分子を明らかにするために、本発明者は、M3培地をM3-1又はM3-2培地で置き換える効果を調べた(図6-10)。その結果、BIO、DMSO、デキサメタゾン、カルシトリオール(M3-2)を細胞に添加した場合、hiPS由来腸細胞は、従来のM3培地での培養下で生成したhiPS由来腸細胞と同等のレベルでトランスポーター及び代謝酵素CYP3A4を発現し、また、そのレベルはM3-1培地によって得られた結果よりも高いことが明らかとなった(図8、9)。
M3-2培地の代わりに、M3-2培地からカルシトリオールを除いた培地を用いて、CYP3A4の発現を調べた。カルシトリオールを除いた培地中で培養されたhiPSC由来腸細胞は、分化21日目において、ほとんどCYP3A4を発現しなかった(図12)。
ここでは、hiPSC由来の内胚葉又は腸前駆細胞をCVM上で培養することにより、hiPSCから機能性腸細胞を生成することを確立した。本発明者は、CVMが腸の成熟細胞の誘導及び維持のための良好な基材であることを見出した。本発明者は以前に、M15細胞上でhiPSCを培養し、Wntシグナル活性化剤であるBIO及びNotchシグナル阻害剤であるDAPTを添加することによって、CDX2陽性腸細胞を得ることができることを報告した。CVM上で培養されたhiPSC由来腸前駆細胞は、頂端膜に局在するVILLINを発現する単層を形成することができる。分化した細胞はZO1陽性であり、TEER測定によって明らかにされたバリア機能を示す。これらの特徴は、機能性腸細胞が生成されることを示している。
(1)ヒトiPS細胞株
ChiPS18(Asplund et al., 2015)(TakaraBio)及びRPChiPS771(ReproCell)の二つのヒトiPS細胞株を使用した。未分化の細胞を、Synthemax II(Corning Cat No.3535)で予めコーティングした細胞培養皿(Invitrogen)上のAK02 StemFit培地(Ajinomoto)で維持した。メチオニン除去のために、ChiPS18細胞及びRPChiPS771細胞を、StemFit培地(Ajinomoto)完全又はMet除去KA01培地(Ajinomoto)で培養した。
コラーゲンキセロゲル膜は、関東化学株式会社(Tokyo, Japan)によって製造されている。簡単に説明すると、CVMは、以前に報告されているように(Oshikata-Miyazaki and Takezawa, 2016, Cytotechnology 68, 1801-1811;Yamaguchi et al., 2013, Toxicol. Sci. 135, 347-355;Yamaguchi et al., 2016, J. Appl. Toxicol. 36, 1025-1037)以下の3工程によって調製された:1)0.2mLの0.25%I型コラーゲンゾルから、直径35mmの培養皿に不透明な軟質ゲルを形成させたゲル化工程;2)十分な乾燥によってゲルが硬質材料となったガラス化工程;3)水分を補給することによってガラス化材料をゲル強度が増強された薄い透明なゲル膜に変換する再水和工程。続いて、分離可能なシート上にコラーゲンビトリゲル膜を再ガラス化することによって、遊離水を含まない乾燥CVMとして定義されるコラーゲンキセロゲル膜を調製した。コラーゲンキセロゲル膜を、内外径11〜13mm、長さ15mmのプラスチックシリンダーの底端に貼り付け、シリンダーの上端に2本のハンガーを接続した。これにより、以前報告したように(Oshikata-Miyazaki and Takezawa, 2016, Cytotechnology 68, 1801-1811;Yamaguchi et al., 2013, Toxicol. Sci. 135, 347-355;Yamaguchi et al., 2016, J. Appl. Toxicol. 36, 1025-1037)、培養培地で再水和することによって容易にCVMチャンバーに変換可能なコラーゲンキセロゲル膜チャンバーを作製できた。
未分化ChiPS18又はRPChiPS771細胞を、まずM15細胞を用いて内胚葉に分化させた。簡潔には、直径100mmのプレートに、マイトマイシンで処理した凍結M15フィーダー細胞を、5×106細胞/皿の密度でプレコートした。内胚葉分化のために、未分化ChiPS18又はRPChiPS771細胞をM15細胞被覆100mm直径プレート上に5×105細胞/皿の密度でプレーティングし、3μM CHIR99021(Wako, Tokyo)を補充した内胚葉分化培地であるM1培地で1日間培養し、次にM1培地(CHIR99021を含まない)に変更し、さらに2日間培養した。M1培地は、DMEM(Thermo Fisher)、4,500mg/Lグルコース、NEAA(ThermoFisher)、L-Gln(Nacalai Tesque)、PS(Nacalai Tesque)、0.1mM β-ME(Sigma)、無血清B27サプリメント(#12587001, ThermoFisher)、100ng/mL組換えヒトアクチビンA(Cell Guidance Systems, Cambridge, UK)からなる。3日目(D3)に、ChiPS18又はRPChiPS771由来内胚葉細胞を集め、Bambanker hRM(NIPPON Genetics Co Ltd, CS-07-001)で2.0×106細胞/mLで凍結し、その後の使用まで液体窒素中に貯蔵した。
10日目の腸前駆凍結保存細胞を調製するために、3日目のDE細胞を、4×106細胞/皿の密度で、iMatrix-551(FUJIFILM WAKO, 380-13041)でプレコートされた直径100mmの正常組織培養プレート(又はCellSeed, CS3005)上にプレーティングし、10日目(D10)までM2培地で培養した。培地は、2日ごとに新鮮なM2培地と交換した。その後、細胞を収集し、10日目の腸前駆凍結保存細胞ストックとして凍結した。
腸管分化のために、10日目の腸前駆細胞ストックを凍結融解し、M2培地中の再水和ビトリゲル膜(CVM)24ウェルインサート(ad-MED Vitrigel TM 2, KANTO KAGAKU, 培養面積:0.33cm2/insert)上に、1.6×105細胞/ウェルの濃度でプレーティングした。最初の2日間のプレーティングの際に、Y-27632(WAKO:251-00514)を培地に添加した。細胞をM2中で15日目まで培養した。培地の量は、セルカルチャーインサートの上層については200μL、下層については700μLとした。培地を15日目から21日目の間、又は30日目まで、M3-1又はM3-2に変更した。培地は2日ごとに新しい培地と交換した。
M3-2培地:HGFの代わりに0.5%のジメチルスルホキシド(DMSO;Sigma-Aldrich, D2650)を使用すること以外はM3-1培地と同じ成分。
Caco-2細胞を、直径10cmの組織培養皿中の10%ウシ胎仔血清を補充したDMEM培地(low glucose)中で5.0×105細胞/ mlで培養し、およそ80%の集密度で3〜4日ごとに継代した。Caco-2細胞を新たに凍結融解し、使用前に一度継代した。膜の完全性の測定のために、Caco-2細胞を1.0×104細胞/ウェルでCVM24ウェルインサート上において継代し、培養を続けた。Caco-2細胞は、典型的には、およそ7日間培養した後に単層を形成した。
CVM上で培養したヒトiPSC由来の腸細胞とCaco-2細胞の膜の完全性を、TEER Measuring System(Kanto kagaku, Inc Tokyo)を用いて測定した。
細胞を、4%パラホルムアルデヒド(Nacalai Tesque)を含むPBS中で固定し、0.1%Triton X-100(Nacalai Tesque)で透過処理し、次いで20%Blocking One(Nacalai Tesque, Japan)を含むPBST(0.1%Tween-20 in PBS)でブロッキングした。抗体を、20%Blocking One(Nacalai Tesque, Japan)を含むPBST(0.1%Tween-20 in PBS)で希釈した。細胞を6-ジアミジノ-2-フェニルインドール(DAPI)(Roche Diagnostics, Switzerland)で対比染色した。
RNeasy micro-kit又はQiaxol(Qiagen, Germany)を用いてiPS細胞からRNAを抽出し、次いでDNase(Qiagen)で処理した。逆転写反応のために、PrimeScript TM RT Master Mix(Takara, Japan)を用いて2.5μgのRNAを逆転写した。リアルタイムPCR分析のために、mRNA発現をStepOne Plus(Applied Biosystems, Foster City, CA)を用い、SyberGreenで定量した。PCR条件は以下の通りであった:95℃で30秒間の初期変性、続いて95℃で5秒間の変性、60℃で30秒間のアニーリング及び伸長を1サイクルとし、40サイクル繰り返した。標的mRNAレベルを任意単位として表し、ΔΔCT法を用いて決定した。プライマーの詳細は下表に示す。また、プライマーの配列を配列番号1〜18に示す。
P-gpのトランスポーター活性を選択的に評価するために、細胞単層を通り抜ける[3 H]-ジゴキシン(0.1μCi/3mL)の時間依存的方向性輸送(BからAへの輸送及びAからBへの輸送)を、100μMベラパミルの非存在下又は存在下で測定した。BCRP及びP-gp活性を評価するために、20μMエラクリダーの非存在下又は存在下で、[3 H]-プラゾシン(0.1μCi/3mL)の時間依存的方向性輸送を測定した。細胞単層を通り抜ける受動的膜透過及び傍細胞輸送によって媒介される経細胞輸送を評価するために、[3 H]-プロプラノロール(0.1μCi/3mL)及び[3 H]-マンニトール(0.1μCi/3mL)をそれぞれ試験した。
dQ/dtが単位時間当たりに透過した化合物の量である場合、Aはセルカルチャーインサート膜の表面積(0.33cm2)であり、そしてC0はドナーチャンバー中の初期化合物濃度である。ローダミン123の流出比は以下のように計算した。
流束比= Papp, basolateral to apical/Papp, apical to basolateral
分化21日目のhiPS由来腸細胞におけるCYP3A4活性を評価した。hiPS由来腸細胞培養培地(M3)を除去し、37℃でトランスポートバッファー(TB)に置き換え、10分間プレインキュベートした。ミダゾラムを含むTBを上部(200μl)及び下部(500μl)区画の両方に添加することによってアッセイを開始した。120分のインキュベーション後、すべてのインキュベーション培地を上部及び下部区画の両方から集め、ミダゾラムの代謝産物である1'-OHミダゾラムのLC-MS / MS分析を行うまでサンプルを-80℃で保存した。 Pierce BCAタンパク質アッセイキット(ThermoFisher, Rockford, IL)を製造元の指示に従って使用して、1ウェルあたりのタンパク質量を定量した。代謝クリアランスは、細胞タンパク質量で標準化した値として求めた。
データは、平均値±S.D.(n = 3)として表した。 グループ間の差は、スチューデントのt検定又はポストホックダネットの検定を用いた一元配置ANOVAによって分析した。各図の説明には、それぞれの統計分析とP値が記載されている。スチューデントのt検定による*p<0.05又は**p<0.01;ポストホックダネットの検定を用いた一元配置ANOVAによる§P<0.05又は§§P<0.01が有意であるとみなされる。

Claims (10)
- GSK3阻害剤と、肝細胞増殖因子受容体の活性化剤、副腎皮質ホルモン、カルシトリオール、及びジメチルスルホキシドからなる群から選ばれる少なくとも1種とを含むことを特徴とする腸細胞分化用培地。
- GSK3阻害剤が、BIOであることを特徴とする請求項1に記載の腸細胞分化用培地。
- 肝細胞増殖因子受容体の活性化剤が、HGFであることを特徴とする請求項1又は2に記載の腸細胞分化用培地。
- 副腎皮質ホルモンが、デキサメタゾンであることを特徴とする請求項1乃至3のいずれか一項に記載の腸細胞分化用培地。
- 以下の工程(1)〜(3)を含むことを特徴とする腸細胞の作製方法、
(1)多能性幹細胞を、アクチビン受容体様キナーゼ-4,7の活性化剤を含む培地中で培養する工程、
(2)工程(1)で得られた細胞を、GSK3阻害剤及びγ-セクレターゼ阻害剤を含む培地中で培養する工程、
(3)工程(2)で得られた細胞を、請求項1乃至4のいずれか一項に記載の腸細胞分化用培地中、高密度コラーゲンゲル膜上で培養し、腸細胞を得る工程。 - 工程(2)において、工程(1)で得られた細胞を高密度コラーゲンゲル膜上で培養することを特徴とする請求項5に記載の腸細胞の作製方法。
- 工程(1)において、多能性幹細胞をM15細胞上で培養することを特徴とする請求項5又は6に記載の腸細胞の作製方法。
- 工程(1)において、アクチビン受容体様キナーゼ-4,7の活性化剤が、アクチビンAであることを特徴とする請求項5乃至7のいずれか一項に記載の腸細胞の作製方法。
- 工程(2)において、GSK3阻害剤がBIOであり、γ-セクレターゼ阻害剤がDAPTであることを特徴とする請求項5乃至8のいずれか一項に記載の腸細胞の作製方法。
- 請求項5乃至9のいずれか一項に記載の方法によって作製された腸細胞。
