Abstract
Description
しかしながら、現在、本分野では、満足できるPD-L1に対するナノ抗体がまだ欠けている。そのため、本分野では、新しいPD-L1に対して有効な特異性ナノ抗体の開発が切望されている。
(a)配列番号1で示されるFR1、配列番号2で示されるFR2、配列番号3で示されるFR3、配列番号4で示されるFR4からなるか、あるいは
(b)配列番号10で示されるFR1、配列番号11で示されるFR2、配列番号12で示されるFR3、配列番号13で示されるFR4からなる、
VHH鎖を提供する。
本発明の第五の側面では、本発明の第四の側面に記載のポリヌクレオチドを含有する発現ベクターを提供する。
もう一つの好適な例において、前記の宿主細胞は原核細胞または真核細胞を含む。
もう一つの好適な例において、前記の宿主細胞は大腸菌、酵母細胞からなる群から選ばれる。
(a) ナノ抗体の生成に適する条件において、本発明の第六の側面に記載の宿主細胞を培養することによって、前記抗PD-L1ナノ抗体を含む培養物を得る工程と、
(b) 前記培養物から前記の抗PD-L1ナノ抗体を単離または回収する工程と、
を含む方法を提供する。
(a) 本発明の第二の側面に記載の抗PD-L1ナノ抗体のVHH鎖、または本発明の第三の側面に記載の抗PD-L1ナノ抗体と、

(i) 本発明の第一の側面に記載の抗PD-L1ナノ抗体のVHH鎖の相補性決定領域CDR、本発明の第二の側面に記載の抗PD-L1ナノ抗体のVHH鎖、または本発明の第三の側面に記載の抗PD-L1ナノ抗体、または本発明の第八の側面に記載の免疫抱合体と、
(ii) 薬学的に許容される担体と、
を含有する薬物組成物を提供する。
(i)ヒトPD-L1分子の検出、
(ii)フローサイトメトリーによる検出、
(iii)細胞免疫蛍光による検出、
(iv)腫瘍の治療、
(v)腫瘍の診断、
のうちの一つまたは複数における使用を提供する。
(i) 本発明の第二の側面に記載の重鎖可変領域VHHの配列または本発明の第三の側面に記載のナノ抗体の配列と、
(ii) 任意の発現および/または精製を補助するタグ配列と、
を有する組換えタンパク質を提供する。
もう一つの好適な例において、前記のタグ配列は6HisタグおよびHAタグを含む。
もう一つの好適な例において、前記の組換えタンパク質は特異的にPD-L1タンパク質と結合するものである。
ここで、前記試薬、カセットまたはキットはサンプルにおけるPD-L1タンパク質の検出に使用され、
ここで、前記薬剤はPD-L1タンパク質を発現する(すなわちPD-L1陽性の)腫瘍の治療または予防に使用される、
使用を提供する。
(1) サンプルを本発明の第三の側面に記載のナノ抗体と接触させる工程と、
(2) 抗原-抗体複合体が形成したか検出し、ここで、複合体が形成したというのはサンプルにPD-L1タンパク質が存在することを意味する工程と、
を含む方法を提供する。
もう一つの好適な例において、前記の対象は、哺乳動物、たとえばヒトを含む。
本発明者は幅広く深く研究し、大量の選別によって、1種類の抗PD-L1ナノ抗体を得ることに成功し、実験結果から、本発明で得られたPD-L1ナノ抗体が有効にPD-L1とPD-1の間の相互作用を遮断することができ、意外なことに、本発明のヒト化後のPD-L1ナノ抗体がより有効にPD-L1とPD-1の結合を遮断することができ、かつBiaCore T200によって同定されたところ、ヒト化後のPD-L1ナノ抗体の親和力が高く、安定性が良く、かつ腫瘍抑制作用が顕著である。これに基づき、本発明を完成させた。
診断の目的に使用される検出可能なマーカーは、蛍光または発光マーカー、放射性マーカー、MRI(磁気共鳴画像)またはCT(コンピューターX線断層撮影技術)造影剤、または検出可能な生成物を生成させる酵素を含むが、これらに限定されない。
さらに、本発明は組成物を提供する。好ましくは、前記の組成物は薬物組成物で、上記の抗体またはその活性断片あるいはその融合タンパク質と、薬学的に許容される担体とを含有する。通常、これらの物質を無毒で不活性で薬学的に許容される水系担体で配合し、ここで、pH値は配合される物質の性質および治療しようとする疾患にもよるが、pH値は通常5〜8程度、好ましくは6〜8程度である。配合された薬物組成物は通常の経路で給与することができ、腫瘍内、腹膜内、静脈内、あるいは局部給与が含まれるが、これらに限定されない。
本発明の一つの好適な例において、前記ナノ抗体は検出可能なマーカーを持つものである。より好ましくは、前記のマーカーは、同位元素、コロイド金マーカー、有色マーカーまたは蛍光マーカーからなる群から選ばれる。
本発明の抗PD-L1ナノ抗体は良い特異性を有し、高い力価を持つ。
また、本発明は、PD-L1タンパク質を検出する方法にも関する。当該方法は、基本的に、細胞および/または組織のサンプルを得る工程と、サンプルを媒体に溶解させる工程と、前記溶解されたサンプルにおけるPD-L1タンパク質のレベルを検出する工程とを含む。
また、本発明は、本発明の抗体(またはその断片)あるいはカセットを含有するキットを提供するが、本発明の一つの好適な例において、前記のキットはさらに容器、使用説明書、緩衝液などを含む。
上記のように、本発明のナノ抗体は幅広い生物応用価値および臨床応用価値を有し、その応用はPD-L1に関連する疾患の診断と治療、基礎医療研究、生物学研究などの多くの分野にわたる。一つの好適な応用はPD-L1に対する臨床診断および標的治療に使用されることである。
(a) 本発明のナノ抗体の高特異性はヒトの正確な空間構造を有するPD-L1タンパク質に対するものである。
(b) 本発明のナノ抗体は親和力が強い。
(c) 本発明のナノ抗体は生産が簡単である。
(1)ヒトPD-L1のヌクレオチド配列をpCDNA3.1(-)ベクター(Invivogenから購入)に合成した後、その細胞外領域の配列をpFUSE-IgG1ベクター(Invitrogenから購入)にサブクローンし、ここで、Fcタグ無しのhPD-L1(ECD)タンパク質が製造しやすいように、hPD-L1(ECD)のC末端に1つのTEV酵素切断部位を導入した。
(2)Omegaプラスミドマキシキットで構築できたpFΜSE-IgG1-hPD-L1(ECD)プラスミドを抽出した。
(3)ODが2.0×106個/mLになるまでHEK293F細胞を培養した。
(4)プラスミドを形質移入試薬PEIと1:3で均一に混合した後20min静置し、さらにHEK293F細胞に入れ、37℃、6% CO2のシェーカーインキュベーターで5〜6日培養した。
(5)細胞上清を収集し、Protein Aビーズと室温で1時間結合させた。
(6)リン酸塩緩衝液pH7.0でビーズを洗浄した後、さらに0.1M pH3.0 グリシンでタンパク質を溶離させた。
(7)溶離されたタンパク質をPBSに限外ろ過し、収量を測定した後サンプリングしてSDS-PAGE検出を行い(検出結果を図1Bに示す)、残ったタンパク質を-80℃の冷蔵庫に保存した。
(8)発現されたhPD-L1(ECD)-Fcタンパク質を酵素切断し、0.1mgのTEV酵素で1mgのhPD-L1(ECD)-Fcタンパク質を切断し、4℃で16時間酵素切断し、タンパク質液にニッケルカラムを通させ、さらにProtein Aカラムを通させ、流出液を収集し、サンプリングしてSDS-PAGE検出を行った(検出結果を図1Cに示す)。
(1)1mgのhPD-L1(ECD)-Fc抗原をフロイントアジュバントと等体積で混合させ、1頭の新疆フタコブラクダを、毎週1回で、合計7回免疫させ、B細胞を刺激して抗原特異的ナノ抗体を発現させた。
(2)7回の免疫が終了した後、100 mLのラクダの末梢血のリンパ球を取って全RNAを抽出した。
(3)cDNAを合成してネステッドPCRによってVHHを増幅した。
(4)制限酵素PstIおよびNotIで20 μgのpMECSファージディスプレイベクター(Biovectorから購入)および10 μgのVHHを酵素切断して2つの断片を連結した。
(5)連結産物を電気的形質転換感受性細胞TG1に形質転換させ、PD-L1ナノ抗体ライブラリーを構築してライブラリーサイズを測定したところ、ライブラリーサイズが1.3×109CFΜであった(結果を図2に示す)。
それと同時に、ランダムに24個のクローンを取って集落PCR検出を行い、結果から構築されたライブラリーの挿入率が100%であったことが示され、集落PCRの結果を図3に示す。
抗体の選別:
(1)100 mM NaHCO3、pH 8.2に溶解させた10 μgのhPD-L1(ECD)抗原をNUNCマイクロプレートにカップリングさせ、4℃で一晩置いた。
(2)2日目に、100 μLの0.1%BSAを入れ、室温で2 hブロッキングした。
(3)2 h後、100 μLのファージ(2×1011 CFΜ免疫ラクダナノ抗体ファージディスプレイ遺伝子ライブラリー)を入れ、室温で1 h作用させた。
(4)0.05% PBS+Tween 20で5回洗浄し、非特異的なファージを除去した。
(5)100 mMのトリエタノールアミンでPD-L1と特異的に結合したファージを解離させ、そして生長対数期にある大腸菌TG1細胞に感染させ、37℃で1 h培養し、ファージを生成させて精製して次の選別に使用し、同じ選別過程を3回繰り返し、その濃縮結果を図4に示す。
ファージの酵素結合免疫法(ELISA)によって特異的な単一の陽性クローンを選別した。
(1)2〜3回の上記選別後のファージを含有する細胞シャーレから、96個の単一集落を取って100 μg/mLのアンピシリンを含有するTB培地(1LのTB培地に2.3g KH2PO4、12.52g K2HPO4、12g ペプトン、24g酵母エキス、4mLグリセリンを含有する)に接種し、対数期に生長したら、最終濃度1mMのIPTGを入れ、28℃で一晩培養した。
(2)浸透法によって粗抽出抗体を得、そして抗体を抗原で被覆されたELISAプレートに移し、室温で1時間置いた。
(3)PBSTで結合しなかった抗体を洗浄し、抗ネズミ抗HA抗体(北京康為世紀生物科技有限公司から購入)を入れ、室温で1時間置いた。
(4)PBSTで結合しなかった抗体を洗浄し、ヤギ抗マウスアルカリホスファターゼ標識抗体を入れ、室温で1時間置いた。
(5)PBSTで結合しなかった抗体を洗浄し、アルカリホスファターゼ呈色液を入れ、ELISA装置で、405 nmの波長で、吸収値を読み取った。
(6)サンプルウェルのOD値が対照ウェルのOD値の3倍以上よりも大きい場合(Ratio+/->3)、陽性クローンウェルと判定した。
(7)陽性クローンウェルの菌を100 μg/mLのLB含有液に移してプラスミドを抽出して配列決定を行った。
(1)上記配列決定で得られたクローン株のプラスミドを大腸菌WK6に電気的形質転換し、そしてそれをLA+Glucose、すなわちアンピシリンとブドウ糖を含有する培養プレートに塗布し、37℃で一晩培養した。
(2)単一集落を取って5 mLのアンピシリン含有LB培養液に接種し、37℃のシェーカーで一晩培養した。
(3)1 mLの一晩培養された菌種を330 mLのTB培養液に接種し、37℃のシェーカーでOD値が0.6〜1に達するまで培養し、IPTGを入れ、28℃のシェーカーで一晩培養した。
(4)遠心し、菌を収集した。
(5)浸透法によって抗体粗抽出液を得た。
(6)ニッケルカラムイオンアフィニティークロマトグラフィーによって純度が90%以上に達したナノ抗体を製造し、精製結果を図5に示す。
(1)hPD-1-Fc-ビオチンタンパク質(hPD-1-Fcの製造方法は実施例1と同様で、SDS-PAGE検出結果を図1Eに示す)を製造し、タンパク質のビオチン化の方法はビオチン試薬の説明書を参照した。
(2)各サンプルは1×106個のヒトPD-L1全長タンパク質を一過性発現するHEK293F細胞を取って0.5% BSA-PBS緩衝液に再懸濁させ、上記精製されたPD-L1ナノ抗体を10μg入れ、同時に陰性対照(hIgG1)およびブランク群(PBS)を設け、すべてのサンプルに5μgのhPD-1-Fc-ビオチンを入れ、4℃で20minインキュベートした。
(3)PBSで2回細胞を洗浄し、eBioscienceのSA-PEを入れ、4℃で20minインキュベートし、PBSで細胞を2回洗浄した後、フローサイトメーター流式細胞計(BD FACS Calibur)によって検出し、検出結果を図6に示す。
PD-L1ナノ抗体のヒト化改造
(1)まず配列番号8で示されるPD-L1ナノ抗体の配列を鋳型として構造データベースで相同構造の検索を行ったところ、1306個の構造が見つかり、中からE value=0.0で配列相同性≧70%の34個の構造を選択した。
(2)この34個の構造を比較し、結晶構造の解像力の大きさおよび構築された系統樹から、最終的に3dwtを含む9つのタンパク質を選び、配列番号8で示されるPD-L1ナノ抗体の配列のマルチテンプレートのホモロジーモデリングを行い、最終的に10個の構造を得、さらに採点関数の点数ランキングから、molpdfが最低の構造を選び、以下の操作を続けた。
(3)モデリングによる最適な構造に対し、ProtSAサーバーで残基の溶媒接触性を計算し、すなわち残基のアンフォールディング状態に対するフォールディング状態の溶媒に接触可能な面積の比に準じて判断し、40%超の残基を溶媒に露出する残基とした。
(4)モデリングによる最適な構造およびDP-47に対して配列アラインメントを行い、相応する溶媒に露出する残基を入れ替えた。最終的に配列番号14で示されるアミノ酸配列によってコードされるヒト化PD-L1ナノ抗体を決めた。ヒト化前後の抗体の配列は下記表2に示す。
本実験は既に2種類の市販の抗PD-L1抗体(アテゾリズマブ(Atezolizumab),ATEおよびデュルバルマブ(Durvalumab),DUR)を陽性対照抗体として使用し、MOA検出細胞株(Promega)を使用し、蛍光レポーター遺伝子の発現を検出することによってNFATシグナルの活性化状況を反映することで、抗体(配列は実施例5を参照する)のPD-1/PD-L1結合に対する抑制作用を検出した。工程は以下の通りである。
(1)活性検出の1日前のCHOK1-PDL1細胞の用意:CHOK1-PDL1は1〜2日前に継代した。培養上清を捨て、PBSで洗浄した。適量のトリプシンを入れて37℃/5% CO2で3〜5 min消化した。4倍のトリプシン体積の培地を入れ、細胞を50 ml遠心管に移して計数した。必要な体積の細胞を取って、230 gで10 min遠心した。培地を入れ、細胞を4×105個細胞/mLになるように再懸濁させた。細胞を96ウェル白色細胞培養プレートに100 μl/ウェルで入れた。サイドウェルにPBSを200 μl/ウェルで入れた。細胞を37℃、5%CO2のインキュベーターで一晩培養した。
(2)Jurkat-PD1細胞の処理:活性検出の2日前に細胞の継代を行った。計数後、必要な体積の細胞を取って、170 gで5 min遠心した。アッセイ緩衝液で細胞を1.25×106 個細胞/mlになるように再懸濁させた。
(3)サンプルおよびJurkat-PD1細胞の検出プレートへの仕込み:95 μl/ウェルのCHOK1-PDL1細胞上清を捨てた。40 μlのサンプル(ハイブリドーマ上清または勾配希釈されたハイブリドーマ上清精製抗体)および陽性対照、陰性対照を入れた。40 μlのJurkat-PD1細胞を入れた。37℃/5%CO2のインキュベーターで6時間培養した。
(4)検出:事前にBio-GloTM緩衝液を融解させ、Bio-GloTM基質を入れ、均一に混合した。6時間後、Bio-GloTM試薬を80 μl/ウェルで入れた。室温で5〜10 min置いた。数値を読み取った。
実験結果は、表4および図11に示すように、各濃度において、本発明のヒト化前のナノ抗体は基本的にいずれも陽性対照群の抗体よりも活性が強く、ヒト化後の本発明のナノ抗体は活性が陽性対照群に相当する。そのため、Nb-Fcおよびヒト化Nb-Fc抗体がいずれも有効にPD1/PD-L1の相互作用を遮断することができる。
(1)ヒト化前およびヒト化後のPD-L1 Nbの配列をpFUSE-IgG1ベクター(Invivogenから購入)に合成し、OmegaプラスミドマキシキットでpFUSE-IgG1-Nb(ヒト化)プラスミドを抽出した。
(2)ODが2.0×106個/mLになるまでHEK293F細胞を培養した。
(3)プラスミドを形質移入試薬PEIと1:3で均一に混合した後20min静置し、さらにHEK293F細胞に入れ、37℃、6% CO2のシェーカーインキュベーターで5〜6日培養した。
(4)細胞上清を収集し、Protein Aビーズと室温で1時間結合させた。
(5)リン酸塩緩衝液pH7.0でビーズを洗浄した後、さらに0.1M pH3.0 グリシンでタンパク質を溶離させた。
(6)溶離されたタンパク質をPBSに限外ろ過し、収量を測定した後サンプリングしてSDS-PAGE検出を行い(検出結果を図1Dおよび図7に示す)、残ったタンパク質を-80℃の冷蔵庫に保存した。図7から精製後のヒト化ナノ抗体の純度が90%以上に達したことがわかる。
方法は実施例5と同様である。
(1)各サンプルは2×105個の天然にPD-L1を発現するヒト肺癌細胞系EBC-1を取って0.5% BSA-PBS緩衝液に再懸濁させ、10μgの精製されたヒト化PD-L1ナノ抗体を入れ、同時に陰性対照(hIgG1)およびブランク群(PBS)を設け、すべてのサンプルに5μgのhPD-1-Fc-ビオチンを入れ、4℃で20minインキュベートした。
(2)PBSで細胞を2回洗浄し、eBioscienceのSA-PEを入れ、4℃で20minインキュベートし、PBSで細胞を2回洗浄した後、セットして検出したが、検出結果は図8に示すように、ブランク群および陰性対照群ではPD-1-Fc-ビオチンとEBC-1細胞の結合率が90%以上であったが、PD-L1ナノ抗体およびヒト化ナノ抗体を入れた後、PD-1-Fc-ビオチンとEBC-1細胞の結合率が10%以下で、これは入れられたナノ抗体が顕著にPD-1とPD-L1の相互作用を遮断することができることを示す。
BiaCore T200によって検出した。(1)固定化:カルボキシ基とアミノ基の反応を利用して固定相の抗原をCM-5センサーチップの表面に固定化した。
(2)結合:抗体をHBS緩衝液で適当の濃度(5つの濃度勾配)に希釈し、抗原と抗体の結合過程を観察した。
(3)チップの再生:次の抗体の測定時10mMのグリシンで洗浄して再生させた。
(4)実験結果の分析:測定結果は図9に示すように、ヒト化前のナノ抗体の親和力が2.34×10-9Mで、ヒト化後のナノ抗体の親和力が2.26×10-9Mであった。 ヒト化の改造は抗体の親和力に影響しなかった。
(1)通常の方法によってヒト化前およびヒト化後のナノ抗体をビオチン化させた。
(2)抗原タンパク質のPD-L1(ヒト)、PD-L1(ネズミ)、PD-L1(サル)、PD-L2(ヒト)、B7H4(ヒト)、B7H3(ヒト)の被覆:各ウェルに0.5 μg(5μg/mL、100μL)、IgG1を対照として、4℃で一晩被覆させた。
(3)2日目にPBSTで3回洗浄し、200 μLの1%BSAを入れて室温で2時間ブロッキングした。
(4)各株のビオチン化されたナノ抗体を10 μg/mLに希釈し、それぞれ各ウェルに100μL取ってインキュベートし、室温で1時間反応させた。
(5)PBSTで結合しなかった抗体を洗浄し、100 μLのストレプトアビジン-HRP(1:1000で希釈)を入れ、室温で1時間置いた。
(6)呈色液を入れ、ELISA装置で、450 nmの波長で、吸収値を読み取った。吸収値からナノ抗体の特異性を判断し、検出結果は図10に示すように、ヒト化前後のナノ抗体はいずれもヒト由来およびサル由来のPD-L1と相互作用することができたが、ネズミ由来のPD-L1と相互作用せず、2株の抗体は優れた種属特異性を有し、かつヒト化前後のナノ抗体はいずれもPD-L1ファミリーのメンバーと相互作用せず、優れたファミリー特異性を有する。
本実験は、抗体を体外で培養された、異なるドナー由来の成熟DC細胞およびCD4+ T細胞とともにインキュベートし、培養系におけるIL2およびIFN-γの相対発現量を検出することによって、異なる抗体のT細胞に対する活性化作用を反映した。工程は以下の通りである。
(1)PBMCの単離:ドナーから新鮮な血液を50 ml取り、2.5倍のPBSを添加し、軽くFiColl(Thermo) 12.5 mlに入れて4管に分け、400 gで30 min遠心し、0減速度で停止させた。そのうちの白色バンドを吸ってPBS(Gibco)に入れ、PBSで2回洗浄した。
(2)DC細胞の単離:単離されたPBMC細胞を取って5 mlのT細胞培地に入れ、37 ℃、6% CO2で2 h付着培養し、懸濁細胞液を吸い取ってCD4+細胞を単離し、残った細胞を3 mlのDC培地に添加し、2日培養した後3 mlのDC培地を添加し、さらに5日培養した後、rTNFa (R&D Systems)(1000 U/ml)、IL-1b(R&D Systems) (5 ng/ml)、IL-6 (R&D Systems)(10 ng/ml) および1 μM PGE2(Tocris)を入れて2日培養し、リンパ球混合反応(MLR)のDC細胞とした。
(3)CD4+細胞の単離:PBMCを2 h静置培養し、懸濁細胞液を15 ml遠心管に吸い取り、200 gで10 min遠心し、沈殿に加入500 μlの単離液、100 μl のAB型血清、100 μlの精製抗体を入れて再懸濁させ、4℃で20 minインキュベートし、単離液で1回洗浄し、さらに500 μlのビーズ緩衝液を入れて15 minインキュベートし、磁場でビーズを除去し、T細胞培地で1回洗浄し、8 mlの培地で再懸濁させ、37℃、6% CO2で培養した。「ヒトCD4+ T細胞濃縮キット(Human CD4+ T cell Enrichment Kit)」(19052、Stemcell)の説明書に従って操作した。
(3)MLR実験:成熟したDC細胞をCD4+細胞と混合し、各ウェルに体積200 μl、DC細胞10000個、CD4+細胞100000個で、抗体を入れ、DC、T細胞、MLRを陰性対照とし、DC+T細胞+anti-CD3/CD28ビーズを陽性対照とし、混合して5日培養し、cisbioキット(ヒトIL2キット1000 Test、ヒトIFN-γ1000test)によってIL2、IFN-γの濃度を検出した。
実験結果は図12に示すように、本発明のナノ抗体(配列は実施例5を参照する)がドナーを刺激して生成したサイトカインはいずれも陽性対照よりも高く、ヒト化後、生成したサイトカインは陽性対照に相当するため、本発明のナノ抗体およびヒト化改造後の抗体はいずれも有効にT細胞を活性化させることができ、かつ活性化効果が陽性対照群の抗体に相当することを示す。
本実験は、ヒトPD-L1を発現するMC38細胞(MC38-PDL1)(南京銀河公司)でPD-L1遺伝子組換えマウスにおいてヒト化Nb-Fcの抗腫瘍作用を測定した。まず、皮下接種の手段によってMC38-PDL1腫瘍担持マウスモデルを構築し、腫瘍形成後群分けを行い、異なる抗体(配列は実施例5を参照する)で異なる投与量の治療を与え、投与期間における各群のマウスの腫瘍体積および体重の変化をモニタリングし、投与頻度は2回/週で、モニタリングの頻度はいずれも2回/週で、連続5週でモニタリングし、投与量および投与形態は表5および図13に示す。
1) MC38/PD-L1細胞懸濁液の調製:PBS(1×)でMC38細胞を分散させ、細胞密度が1×107個/mlで、MC38細胞懸濁液を調製した。
2) 接種:25匹のC57Bl/6背景のPD-L1マウスの右側の背部の毛を剃り、MC38/PD-L1細胞を1×106個/0.1 ml/匹で皮下注射した。腫瘍細胞の接種の6日後、各マウスの腫瘍体積を検出し、腫瘍体積が87.4 mm3〜228.4 mm3の範囲内にある25匹のマウスを選んで腫瘍体積で平均に群分けを行った。
3) 投与:図13を参照する。
4) 検出:毎回の投与前に、体重と腫瘍体積を測定し、重量を記録した。電子天秤で体重を週に2回測定した。
5) 腫瘍体積の測定:ノギスで腫瘍の最大長さ(L)および最大幅(W)を測定し、腫瘍体積はV=L×W2/2という公式で計算した。
実験結果は図14に示すように、時間の経過につれ、ヒト化Nb-Fcを接種されたマウスを示し、その腫瘍体積は対照群に対して良く抑えられ、かつ顕著に増加した状況が現れず、ヒト化Nb-Fcが顕著な腫瘍抑制作用を有することが説明された。
本実験は、PEG沈殿の方法によって、候補の抗体(配列は実施例5を参照)の異なる濃度のPEGにおける溶解状況を検出することによって、抗体の可溶性を反映した。工程は以下の通りである。
1) 抗体のサンプルを5 mg/mlに濃縮させた。
2) 96ウェル細胞培養プレートにサンプルを仕込み、各ウェルに40 μlずつ抗体のサンプルを入れ、最終濃度が1 mg/mlであった。1〜12列目にそれぞれ30% PEG を26.7 μl、40 μl、46.7 μl、53.3 μl、60μl、66.7 μl、73.3 μl、80μl、86.7 μl、93.3 μl、100 μl、106.7 μlで入れ、IBI301緩衝液で合計体積が200 μlになるまで補充した。PEGの濃度勾配はそれぞれ4% 、6%、 7% 、8%、 9%、 10%、 11% 、12%、 13% 、14% 、15%、16%であった。
3) 室温で1 h置いて、OD500 nmを測定した。
研究結果は表6および図15に示すように、ヒト化Nb-Fcはいずれも8%(w/v)PEGで混濁したが、対照抗体は(すでに市販の薬物ヒュミラ(Humira))6%(w/v)PEGで混濁したことで、本発明のヒト化Nb-Fcが対照抗体よりも優れた可溶性を有することが示された。
本実験は、抗体(配列は実施例5を参照する)の40℃で30日置いた後の純度および生物学的活性の変化を検出することによって、当該抗体の長期間熱安定性を評価した。SECの方法によって目的抗体の40℃で0、14および30日置いた後の純度を測定したが、実験結果は表7および図16に示すように、ヒト化Nb-Fcの純度が顕著に変化しなかった。本実験は、FACS方法によって加速安定性実験のサンプルとCHO-PDL1細胞の結合状況を検出したが、工程は以下の通りである。
1) 細胞の準備:CHO-PDL1細胞を計数し、そして2×106個細胞/mlに希釈し、U底96ウェルプレートに100 μl/ウェルで入れ、1列目のウェルに50 μl追加した。
2) 検出工程:1つ目のウェルに抗体を最終濃度が200 nMになるように入れ、均一に混合し、50 μlを2つ目のウェルに吸い取り、同様に順に操作し、陰性対照はIgG対照であった。氷浴で20 min処理した。PBSを100 μl/ウェル入れ、400 gで5 min遠心して上清を除去し、PBSで細胞を1回洗浄した。1:100で希釈されたヤギ抗ヒトIgG-PE(eBioscience)を100 μl /ウェル入れ、氷浴で20 min処理し、400 gで5 min遠心して上清を除去し、PBSを100 μl/ウェル入れて1回洗浄し、100 μl PBSで再懸濁させ、FACSによって検出した。
実験結果は図17に示すように、ヒト化Nb-FcとCHO-PDL1細胞の結合に顕著な変化がなかった。研究結果から、ヒト化Nb-Fcが優れた熱安定性を有することがわかる。
Claims (10)
- 抗PD-L1ナノ抗体のVHH鎖の相補性決定領域CDR領域であって、前記VHH鎖の相補性決定領域CDRは配列番号5で示されるCDR1、配列番号6で示されるCDR2、配列番号7で示されるCDR3からなることを特徴とするCDR領域。
- 抗PD-L1ナノ抗体のVHH鎖であって、前記VHH鎖はフレームワーク領域FRおよび請求項1に記載の相補性決定領域CDRを含み、前記のフレームワーク領域FRは、
(a)配列番号1で示されるFR1、配列番号2で示されるFR2、配列番号3で示されるFR3、配列番号4で示されるFR4からなるか、あるいは
(b)配列番号10で示されるFR1、配列番号11で示されるFR2、配列番号12で示されるFR3、配列番号13で示されるFR4からなる、
ことを特徴とするVHH鎖。 - PD-L1エピトープに対するナノ抗体で、配列番号8または配列番号14で示されるアミノ酸配列のVHH鎖を有することを特徴とする抗PD-L1ナノ抗体。
- 請求項1に記載のCDR領域、請求項2に記載の抗PD-L1ナノ抗体のVHH鎖、または請求項3に記載の抗PD-L1ナノ抗体からなる群から選ばれるタンパク質をコードすることを特徴とするポリヌクレオチド。
- 配列番号9または15で示されるヌクレオチド配列を有することを特徴とする請求項4に記載のポリヌクレオチド。
- 請求項4に記載のポリヌクレオチドを含有することを特徴とする発現ベクター。
- 請求項6に記載の発現ベクターを含有するか、あるいはゲノムに請求項4に記載のポリヌクレオチドが組み込まれていることを特徴とする宿主細胞。
- 抗PD-L1ナノ抗体を生成する方法であって、
(a) ナノ抗体の生成に適する条件において、請求項7に記載の宿主細胞を培養することによって、前記抗PD-L1ナノ抗体を含む培養物を得る工程と、
(b) 前記培養物から前記の抗PD-L1ナノ抗体を単離または回収する工程と、
を含むことを特徴とする方法。 - (a) 請求項2に記載の抗PD-L1ナノ抗体のVHH鎖、または請求項3に記載の抗PD-L1ナノ抗体と、
(b) 検出可能なマーカー、薬物、毒素、サイトカイン、放射性核種、または酵素からなる群から選ばれる抱合部分と、
を含有することを特徴とする免疫抱合体。 - 請求項3に記載の抗PD-L1ナノ抗体の使用であって、(a)PD-L1分子を検出する試薬製造、または(b)腫瘍を治療する薬物の製造に用いられることを特徴とする使用。
