Abstract
Description
本発明の他の様態は、ラクトバチルスサリバリウスCJLS1511(KCCM11829P)またはその死菌体を含む動物飼料添加剤組成物に係わる。
前記培養液を70℃ないし160℃の温度で加熱し、
前記加熱された培養液を10℃ないし60℃の範囲の温度に冷却し、
前記冷却された培養液で、ラクトバチルスサリバリウスCJLS1511(KCCM11829P)死菌体を分離することを含むラクトバチルスサリバリウスCJLS1511(KCCM11829P)死菌体の製造方法に係わる。
公知の菌株であるラクトバチルスサリバリウスKCCM40210を比較菌株にし、本発明のラクトバチルスサリバリウスCJLS1511(KCCM11829P)菌株の安全性、耐酸性、耐胆汁性、抗菌活性、消化酵素活性及び中性脂肪分解活性を下記のように評価した。
ラクトバチルスサリバリウスCJLS1511(KCCM11829P)菌株の安全性を評価するために、韓国バイオベンチャー協会団体標準で提示した安全性評価試験法により、溶血現象検査、ゼラチン液化反応検査、有害代謝産物(アンモニア)生成確認及びフェニルアラニン脱アミン検査を行った。その結果を下記表3に示した。
MRS培地で事前培養したラクトバチルスサリバリウスCJLS1511菌株と、比較菌株であるラクトバチルスサリバリウスKCCM40210標準菌株とに対して、pH2,3,7にそれぞれ調整したMRS溶液で、10−6倍希釈した。希釈した菌液を37℃で培養しながら、一定時間帯別に、MRS agar培地に塗抹し、48時間嫌気的に培養した後、コロニー数を測定した。該耐酸性実験の結果は、表4に示した。
MRS培地で事前培養したラクトバチルスサリバリウスCJLS1511菌株と、比較菌株であるラクトバチルスサリバリウスKCCM40210標準菌株とを、pH4にそれぞれ調整後、胆汁酸液(Oxgall)を、それぞれ0%、0.3%、1%になるように濃度を合わせた後、MRS溶液で10−6倍希釈した。希釈した菌液を37℃で培養しながら、一定時間帯別に、MRS agar培地に塗抹し、48時間嫌気的に培養した後、コロニー数を測定した。菌数を測定した結果は、表5に要約した。
TSB(Tryptic soy broth)培地(BD、米国)に、24時間液体培養された3種の病源菌(E.coli K88、E.coli ATCC 25922、Salmonella typhimurium KCCM 25922、Salmonella cholerasuis KCCM 10709)を、滅菌された綿棒で均一に105〜6cfu/mlで塗抹した。
消化酵素活性実験は、炭水化物、タンパク質、リンを分解することができる酵素を有している乳酸菌であるか否かということを確認するために行われた。
ラクトバチルスサリバリウスCJLS1511のBSH(Bile salt hydrolase)活性を調査した結果、2mMのTDCA(taurodeoxycholate hydrate、Sigma、米国)を添加したMRS固体培地で白沈澱を生成し、酵素活性があることが確認され、TDCA陽性対照群菌株であるラクトバチルスサリバリウスKCCM 40210標準菌株の沈澱と類似した沈澱様相を示した。しかし、2mMのGDCA(sodium glycodeoxycholate、Sigma、米国)を添加した培地においては、ラクトバチルスサリバリウスCJLS1511菌株の場合、菌が良好に育ったが、ラクトバチルスサリバリウスKCCM 40210標準菌株の場合、沈澱を示さず、菌が良好に育たなかった。該結果を下記表8に記載した。
ラクトバチルスサリバリウスCJLS1511とラクトバチルスサリバリウスKCCM 40210標準菌株とを、それぞれMRS寒天培地にループで塗抹し、37℃で48時間培養し、培養液を製造した。

ラクトバチルスサリバリウスCJLS1511の生菌と死菌との疎水性と凝集性との差を確認するために、自家凝集反応と同時凝集反応とを評価した。
生菌と死菌との自家凝集反応と同時凝集反応との結果は、下記表9に記載した。
EU肉鶏の基礎飼料に、ラクトバチルスサリバリウスCJLS1511死菌体0.2%(w/w)を投与した飼料と、それを投与していない基礎飼料(対照群)とに対して、通常の飼料給餌のように、29日間給餌し、開始平均体重(g)、終了平均体重(g)、1日当たり平均増体量(g/d)、平均1日当たり飼料摂取量(g/d)及び飼料要求率をそれぞれ測定した。その結果を下記表10に示した。

- ラクトバチルスサリバリウスCJLS1511(KCCM 11829P)またはその死菌体。
- ラクトバチルスサリバリウスCJLS1511(KCCM 11829P)またはその死菌体を含む動物飼料添加剤組成物。
- 前記動物飼料添加剤組成物が前記死菌体を含むことを特徴とする請求項2に記載の動物飼料添加剤組成物。
- 前記ラクトバチルスサリバリウスCJLS1511(KCCM 11829P)またはその死菌体が、動物飼料添加剤組成物1g当たり1.0x108cfuないし1.0x1010cfuで含まれることを特徴とする請求項2に記載の動物飼料添加剤組成物。
- ラクトバチルスサリバリウスCJLS1511(KCCM 11829P)またはその死菌体の保護剤を追加して含むことを特徴とする請求項2ないし4のうちいずれか1項に記載の動物飼料添加剤組成物。
- 前記保護剤が、酵母、酵母抽出物、単糖類、多糖類、澱粉類、砂糖類のうち1種以上であることを特徴とする請求項5に記載の動物飼料添加添加剤組成物。
- 前記動物が肉鶏であることを特徴とする請求項2ないし4のうちいずれか1項に記載の動物飼料添加剤組成物。
- 前記ラクトバチルスサリバリウスCJLS1511(KCCM 11829P)またはその死菌体が、動物飼料の重量基準で、0.1重量%ないし1重量%であることを特徴とする請求項2ないし4のうちいずれか1項に記載の動物飼料添加剤組成物。
- 請求項8に記載の動物飼料添加剤組成物を含む動物飼料。
- ラクトバチルスサリバリウスCJLS1511(KCCM 11829P)を培養し、培養液を製造し、
前記培養液を70℃ないし160℃の温度で加熱し、
前記加熱された培養液を10℃ないし60℃の範囲の温度に冷却し、
前記冷却された培養液において、ラクトバチルスサリバリウスCJLS1511(KCCM 11829P)死菌体を分離することを含むラクトバチルスサリバリウスCJLS1511(KCCM 11829P)死菌体の製造方法。 - 前記加熱が、熱交換機を介して、間接加熱されることを特徴とする請求項10に記載のラクトバチルスサリバリウスCJLS1511(KCCM 11829P)死菌体の製造方法。
- 前記冷却が、10℃/min〜60℃/minの速度で冷却されることを特徴とする請求項10に記載のラクトバチルスサリバリウスCJLS1511(KCCM 11829P)死菌体の製造方法。
- 前記分離された死菌体に保護剤を混合することを追加して含むことを特徴とする請求項10ないし12のうちいずれか1項に記載のラクトバチルスサリバリウスCJLS1511(KCCM 11829P)死菌体の製造方法。
- 前記混合された死菌体を粉末化することを、追加して含むことを特徴とする請求項13に記載のラクトバチルスサリバリウスCJLS1511(KCCM 11829P)死菌体の製造方法。
- 前記保護剤が、酵母、酵母抽出物、単糖類、多糖類、澱粉類、砂糖類のうち1種以上であることを特徴とする請求項13に記載のラクトバチルスサリバリウスCJLS1511(KCCM 11829P)死菌体の製造方法。
