Abstract
本发明公开了一株唾液乳杆菌ZLp4b及其在防治幼畜腹泻病中的应用。该菌株具有广谱的抑菌作用,可抑制大肠杆菌、沙门氏菌、金黄色葡萄球菌及巴氏杆菌等多种致病菌的生长。此外,该菌株具有很好的体外益生特性,包括较高的胃肠道耐受性、高温耐受性和黏附性等。将本发明唾液乳杆菌应用于饲喂小鼠和腹泻仔猪、羔羊、犊牛中,可促进小鼠血清中免疫球蛋白及细胞因子的分泌,治愈或明显减缓幼畜腹泻症状。

技术领域
本发明涉及一株细菌及其应用。
背景技术
在畜牧养殖中,益生菌制剂作为新型饲料添加剂有着绿色健康、无毒、无残留且不易产生耐药 性等特性,可作为饲料添加剂长期使用。随着对益生菌的不断研究,我们认识到益生菌不仅是存在 于体内的正常微生物,更是调节人体和动物体各项机能的重要物质,益生菌在医疗、食品、养殖中 的应用越来越广泛。益生菌是指活的微生物,当摄入足够的数量时,能给予宿主健康作用。益生菌 大多是来自于人或者动物的胃肠道,对机体发挥着重要的生理作用。目前商业化产品中应用最广泛 且公认安全的益生菌为乳酸菌。乳酸菌进入肠道后通过以下途径拮抗病原菌的生长:(1)产生细菌 素和有机酸等抑菌物质;(2)通过与病原菌竞争肠道上皮细胞的黏附位点和营养素来抑制致病菌在 肠道的定植和生长;(3)增强肠道上皮细胞屏障功能,防治病原菌对肠道上皮细胞的损伤;(4) 通过竞争、置换和排斥的方式抑制病原菌对肠道上皮细胞的黏附;(5)调控致病菌引起的免疫炎症 反应。此外,乳酸菌可通过调节胃肠道平衡、促进营养物质吸收、提高机体免疫力等方式发挥益生 作用。大量研究表明,动物源益生菌具有耐胃肠道性能好、定植率高、抗病能力强、饲喂后机体的肠道菌群稳定等特点,并且添加到动物饲料中后可预防并缓解动物腹泻、规避畜禽养殖的风险,降 低饲养成本并提高收益。
在养猪业中,仔猪腹泻是规模化养殖场面临的最严重的问题,已成为影响养猪业健康稳定发展 和经济收益的主要问题之一。许多病原微生物可导致仔猪、羔羊和犊牛等幼畜的腹泻,其中以大肠 杆菌、沙门氏菌和链球菌等为主。在大规模养殖场中,解决此类问题的方法是使用抗生素。抗生素 虽然在防病抗病等方面起到一定的积极作用,但其长期使用给人类健康和环境带来的危害不言而 喻。随着多数国家对抗生素的限用和禁用,寻找作为新型抗生素替代品的工作已得到了极大的关 注。有研究证实,在畜禽饲料中补充一定的益生菌对预防以及治愈家畜胃肠道疾病、促进家畜的营 养吸收等有良好效果。
因此,为满足我国养殖业需求,促进畜禽健康生长,开发具有益生菌特性并能防治或减缓幼畜 腹泻的益生菌菌株具有重要意义。
发明内容
本发明提供一株唾液乳杆菌ZLp4b,该菌株于 2019 年 9 月 6 日保藏于中国北京的中国微生物菌种保藏管理委员会普通微生物中心,分类名称为唾液乳杆菌(lactobacillus salivarius),保藏编号为CGMCC NO:18471。
本发明的唾液乳杆菌ZLp4b在制备防治仔猪、羔羊、犊牛腹泻药物中的应用。

本发明所述唾液乳杆菌ZLp4b具有良好的抗逆性,包括对低pH、高胆盐及模拟胃肠道溶液的耐 受性。菌株ZLp4b在不同pH条件下都能生长,在pH=3.0及以上环境中可至少耐受4h,分别在含有 0.0%、0.1%、0.2%、0.3%猪胆盐的条件下耐受4h后活菌数基本不变,均高于1.0×107CFU/mL,在人 工模拟胃液中耐受3h后存活率为88.72%,在模拟肠液中耐受6h后存活率为79.07%。
本发明提供的唾液乳杆菌体外益生特性良好,具有广谱抑菌作用、良好的表面性质和较高的黏 附性,且能抑制大肠杆菌黏附Caco-2细胞。ZLp4b对大肠杆菌、副猪嗜血杆菌、沙门氏菌、金黄色葡 萄球菌、巴氏杆菌及支原体等18株致病菌有抑制作用,其中对副猪嗜血杆菌CYF-5的抑菌圈达23.04 mm;ZLp4b对Caco-2细胞的黏附性与参考菌株鼠李糖乳杆菌GG(Lactobacillus rhamnosus GG)的黏 附性相当,无显著差异,且可通过竞争和置换的方式抑制大肠杆菌的黏附细胞,是一种益生菌。
本发明所述唾液乳杆菌ZLp4b可明显减缓并治愈幼畜腹泻。将本发明唾液乳杆菌ZLp4b与其他 益生菌菌株配伍成复合微生态制剂后共计饲喂五个养殖场的腹泻仔猪1511头,其中一个养殖场治愈 率为100%,无病畜死亡,其余四个养殖场的治愈率分别为94.96%、93.23%、92.63%及87.60%。
本发明唾液乳杆菌ZLp4b与其他益生菌菌株配伍成复合微生态制剂初步应用于腹泻羔羊、犊牛 中,均有缓解并治愈腹泻病的效果。
本发明提供的菌种益生性好,对腹泻病畜的治愈率高,可作为微生态制剂应用于仔猪、羔羊、 犊牛腹泻中。

具体实施方式
以下是结合附图详细阐述本发明的方案。
实施例一菌株的分离、纯化
1.1样品采集
采集甘肃省平凉市多个养殖场中健康猪、牛、羊的新鲜粪便120份于50mL的无菌离心管中保 存,加入冰袋后低温运输至实验室。
1.2菌株的分离及纯化
称取粪便样品10g加入到90mL无菌PBS(磷酸盐缓冲溶液)中混匀并过滤,滤液依次稀释至10-4、10-5、10-6倍后各取0.1mL涂布于含钙的MRS平板上,37℃厌氧培养24h;挑取平板上产生溶钙圈 的乳酸菌疑似菌落并进行革兰氏染色,挑取革兰氏阳性且菌体形态呈杆状的菌株的单菌落于MRS固 体平板上划线纯化培养,并依此编号;共分得1126株不同来的乳酸菌疑似株,其中来自于猪粪便649 株,牛粪便273株,羊粪便204株;待菌株纯化3次后,挑取单菌落接种于10mL MRS液体培养基中 37℃静置培养20h。
本发明上述使用的无菌PBS配方如下:
称取NaCl 8.0g,KCl 0.2g,Na2HPO4 1.42g,KH2PO4 0.27g加入到800mL去离子水中,充分 搅拌溶解后,滴加浓盐酸调节pH至7.4,加去离子水定容至1L后,121℃高压灭菌20min,室温保 存备用。
本发明上述使用的MRS培养基配方如下:
蛋白胨10.0g,牛肉浸粉10.0g,酵母浸粉5.0g,葡萄糖20.0g,吐温801.0mL,K2HPO4·3H2O 2.0g,无水乙酸钠5.0g,柠檬酸三铵2.0g,MgSO4·7H2O 0.29g,MnSO4·H2O0.058 g,将上述成分加入蒸馏水中,定容至1000mL,调节pH为6.3,于121℃高压灭菌20min。
实施例二益生菌侯选珠的筛选及鉴定
2.1高抗逆性乳酸菌的筛选
耐酸能力测定:分别取1mL上述1126株新分离乳酸菌的新鲜培养液接种至pH=2.5的无菌PBS 中,于37℃条件下耐受3h后,取样并连续稀释至10-4、10-5,吸取0.1mL稀释液涂布于MRS平板上, 37℃厌氧培养24h后计数测定活菌数,以0h活菌数为对照,计算存活率。
耐胆盐能力测定:分别取1mL上述1126株分离菌的新鲜培养液接种至含0.3%胆盐的无菌PBS 中,于37℃条件下耐受3h后计数测定活菌数,以0h活菌数为对照,计算存活率。
耐受结果表明,在1126株受试菌中,344株乳酸菌疑似株同时对pH=2.5和0.3%胆盐条件的耐受性 高于80%,其余菌株对酸和胆盐表现出不同程度的敏感性。
耐高温能力测定:取1mL上述344株细菌的新鲜培养液分别于50℃、60℃和70℃条件下加热10 min,高温处理后取样测定OD(600nm),以加热前各菌株的OD(600nm)值为初始值,计算各菌株对高温 的耐受性。结果表明,65%的菌株在50℃条件下处理10min后活菌数减半,加热温度在60℃及以上 时,所有菌株存活率普遍较低,加热温度为70℃时,仅有26株细菌存活率高于10%。
2.2高黏附性乳酸菌的筛选
FITC(异硫氰酸荧光素)标记上述26株高抗逆性菌株:将活化后的上述各菌株的菌液于4℃,6000r/min离心收集菌体,用无菌PBS洗涤3次后加入到工作浓度为500μg/mL的FITC标记液中, 37℃暗处理2h;孵育后的菌液6000r/min离心,用PBS洗涤3次去除未结合的FITC,并将菌体重 悬于RPMI-1640细胞培养液中,调整菌体浓度为2×108CFU/mL,吸取菌液于96孔板中,并用酶标 仪测定其在吸收光波长为485nm及发射光波长为530nm时的相对荧光强度,记录为N0。
黏附试验向培养至单层细胞的12孔细胞培养板中加入0.5mL FITC标记的各菌株,于37℃、 5%CO2培养箱中孵育2h后,用无菌PBS漂洗3-5次去除未黏附的菌株。鼠李糖乳杆菌 (Lactobacillus rhamnosus GG)作为阳性对照。漂洗后的12孔细胞培养板每孔加入300μL胰酶消化细 胞5min后加入RPMI-1640细胞培养液终止反应并混匀每孔液体。吸取细胞悬液于96孔板中,测定 其在吸收光波长为485nm及发射光波长为530nm时的相对荧光强度,并记录为N1。
菌株黏附率计算公式为:黏附率=N1/N0×100%
式中:N1为黏附Caco-2细胞后菌株的相对荧光强度;
N0为黏附Caco-2细胞前菌株的相对荧光强度。
黏附实验表明,26株试验菌株表现出对Caco-2细胞不同的黏附率,其中编号为ZLp4b的菌株黏附 率最高(表1),为12.15%,与鼠李糖乳杆菌GG对Caco-2细胞的黏附率相当,无显著差异。

基于上述试验结果,最终筛选出菌株ZLp4b作为益生菌候选株。
2.3菌株ZLp4b的形态学鉴定
用无菌接种环挑取一环菌株ZLp4b的菌液于MRS平板上划线,于37℃、厌氧条件下培养24h后观 察单菌落形态,菌株ZLp4b株在MRS平板上呈乳白色,凸起,圆形,边缘整齐,表面光滑(图1)。 革兰氏染色后油镜下观察,菌株ZLp4b呈蓝紫色且菌体形态呈杆状,两端钝圆,单个、成双或聚集排 列(图2)。
2.4菌株ZLp4b的分子生物学鉴定
提取菌株ZLp4b的基因组DNA将ZLp4b的菌液以5%接种量接种至10mL液体MRS培养基中,于 37℃静置培养20h后,取1mL新鲜菌液离心收集菌体并用PBS漂洗;用购自大连宝生物工程有限公司 的细菌基因组DNA提取试剂盒提取菌株ZLp4b的基因组DNA,提取后冻存于-20℃,备用。
16S rDNA序列的PCR扩增采用细菌鉴定16S rDNA通用引物27F及1492R进行PCR扩增,扩增体 系为:2×Premix Taq 25μL,上、下游引物各1μL,基因组DNA 1μL,ddH2O 22μL。扩增程序为: 95℃预变性7min;95℃变性45s;55℃变性45s;72℃延伸70s,共35个循环;72℃延伸10min,4℃ 保存PCR产物。
琼脂糖凝胶电泳鉴定PCR扩增产物经0.1%琼脂糖凝胶电泳鉴定,得到目的片段长约为1500bp (图3),其中第一泳道为DNA分子质量为2000的DNA Marker。1、2泳道为菌株ZLP4b的片段长度。
ZLp4b的16S rDNA测序PCR产物送西安擎科泽西生物有限公司测序,测序结果提交NCBI进行 Blast比对,序列与Lactobacillus salivarius的序列同源性为99.7%,结合ZLp4b的单菌落形态及革兰氏 染色结果,鉴定该菌株为唾液乳杆菌。该菌株序列于NCBI申请获得登录号为:MK208492。
本发明的广谱抑菌作用的唾液乳杆菌ZLp4b于2019年9月6日保藏于北京的中国微生物菌种保 藏管理委员会普通微生物中心,保藏编号为CGMCC NO:18471。
实施例三唾液乳杆菌ZLp4b的生物学特性研究
3.1唾液乳杆菌ZLp4b的酸耐受性研究
取ZLp4b新鲜菌液以5%接种量接种于不同pH值(pH梯度为2.0、2.5、3.0、3.5、4.0)的无菌 PBS中,置于37℃条件下静置培养0、2、4h,每一个处理3个重复。取1mL菌液于9mLPBS中 混匀连续稀释,制成10-4、10-5、10-6稀释度溶液,取0.1mL稀释液于MRS固体平板上涂布计数, 37℃培养24h后记录平板上的菌落数。菌株ZLp4b对酸性环境的耐受性结果见表2,ZLp4b在不同 pH条件下都能生长,在3.0及以上时,培养2h及4h后活菌数目均有高于1.0×108CFU/mL且基本 不变,说明ZLp4b可在低pH环境中至少耐受4h,也能确保摄入的活菌顺利通过胃液;在pH为2.0 和2.5时,培养4h后,菌落数低于1.0×108CFU/mL,存活率较低,表明ZLp4b在pH为2.0的环境 中不能长时间存活。该菌具有很强的耐酸能力,在2.0-3.0条件下培养4h的菌落少于2h,说明在这 个pH范围内,随着处理时间的延长,菌的生长受到抑制。
3.2唾液乳杆菌ZLp4b的胆盐耐受性研究
取ZLp4b新鲜菌液以5%接种量接种于含有不同猪胆盐(含量梯度为0.0%、0.1%、0.2%、0.3%、 0.6%)的无菌PBS中,置于37℃条件下下分别培养0、2、4h,每个处理3个重复。各取1mL培养物 于9mL PBS中混匀并连续稀释,制备成10-4、10-5、10-6稀释度溶液,取0.1mL稀释液于MRS固体平板 上涂布计数,于37℃厌氧培养24h后用平板进行菌落计数。唾液乳杆菌ZLp4b在含有不同浓度猪胆盐 的PBS中耐受结果如表3所示,菌株在含有0.0%、0.1%、0.2%、0.3%猪胆盐的条件下耐受4h后活菌 数基本不变,均高于1.0×107CFU/mL。ZLp4b在含有0.6%猪胆盐的条件下耐受4h后,活菌数下降为8.8×106CFU/mL,与0h相比存活率仅为43.3%。由此可知,菌株ZLp4b对胆盐具有很好的耐受性。
3.3唾液乳杆菌ZLp4b对人工模拟胃肠道的耐受性研究
配制人工模拟胃液:称取胃蛋白酶(1:15000)溶于灭菌的生理盐水(0.9%w/v,调节pH至 3.0)中,配制成浓度为3g/L的溶液。0.22μm无菌滤膜过滤后4℃保存。
配制人工模拟肠液:称取胰蛋白酶(1:250)溶于灭菌的生理盐水(0.9%w/v,调节pH至8.0) 中,配制成浓度为1g/L的溶液,加入0.3%胆盐,0.22μm无菌滤膜过滤后,4℃保存。
将ZLp4b离心并收集菌体,用无菌的PBS重悬后调节其浓度为1×109CFU/mL,取1mL菌液加入 到9mL模拟胃液中37℃静置培养1h、2h、3h,分别取样。3h后取1mL上述培养液加入到9mL模拟 肠液中,37℃培养2h,4h,6h后,分别取样并采用MRS平板计数测定活菌总数。菌株ZLp4b在人工 模拟的胃肠道内存活情况如表4所示,可以看出ZLp4b在人工模拟的胃液和肠液中存活率较高,在模 拟胃液中耐受3h后存活率为88.72%,在模拟肠液中耐受6h后存活率为79.07%,ZLp4b经过人工模拟 的胃液和胰液处理后活菌数目均高于106CFU/mL,高于其发挥作用的最低活菌数,说明其耐受性较 好。
实施例四唾液乳杆菌ZLp4b的抑菌性能研究
选取金黄色葡萄球菌、大肠杆菌、支原体等致病菌作为指示菌。菌株ZLp4b以2%种液的接种量 接种于MRS培养基37℃培养18h后离心,上清液经0.22μm微孔滤膜过滤后,备用。将活化后的不 同致病菌用无菌PBS调节浓度约为1×106CFU/mL,取100μL致病菌均匀涂布于其对应的平板上, 室温条件下静置10min;待平板表面菌液略干后,用孔径为7mm的打孔器均匀打孔并用加热后的 琼脂封底,在孔中加入150μL无菌上清液后,于室温下预扩散2h,置于培养箱中37℃培养18h。 ZLp4b的抑菌效果如表5所示,唾液乳杆菌ZLp4b抑制多种致病菌的生长,其中对副猪嗜血杆菌 CYF-5抑制能力最强,抑菌圈直径为23.04mm;其次对沙门氏菌、大肠杆菌、牛支原体等有一定的 抑制作用。

实施例五唾液乳杆菌ZLp4b的表面性质研究
5.1唾液乳杆菌ZLp4b的自凝集能力测定
唾液乳杆菌ZLp4b以5%的接种量接种于MRS培养基中,37℃静置培养20h后于5000r/min、4℃ 条件下离心10min收集菌体,用无菌PBS洗涤3次后调节其OD600为0.5±0.02,并记录吸光值为A0,静 置24h后测定菌液吸光值A24,PBS作为空白对照。
ZLp4b的自凝集率(%)公式为:A%=(A0-A24)/A0×100。
唾液乳杆菌ZLp4b的自凝集率如表6所示。
5.2唾液乳杆菌ZLp4b的共聚集能力测定
唾液乳杆菌ZLp4b以5%的接种量接种于MRS培养基中,37℃静置培养20h;大肠杆菌ATCC 43888接种于LB培养基中,于37℃、200rpm/min条件下振荡培养18h;上述菌液于5000r/min、4℃ 条件下离心10min收集菌体,用无菌PBS分别洗涤3次后调节唾液乳杆菌ZLp4b和大肠杆菌ATCC 43888的混合悬菌液OD600为0.5±0.02,并记录吸光值为A0。静置24h后测定吸光值A24,PBS作为空白 对照。
本发明上述使用的LB培养基配方如下:
称取胰蛋白胨10.0g,酵母浸粉5.0g,氯化钠10.0g,将上述成分加入950mL蒸馏水中,调节 pH为7.0,用蒸馏水定容至1L,于121℃高压灭菌20min。
ZLp4b的它凝集率(%)公式为:A%=(A0-A24)/A0×100。
唾液乳杆菌ZLp4b的它凝集率如表6所示。
5.3唾液乳杆菌ZLp4b的疏水性测定
采用碳氢化合物黏着法测定唾液乳杆菌ZLp4b的疏水性。过夜培养的菌株ZLp4b在5000r/min、 4℃条件下离心10min收集菌体,用无菌PBS洗涤3次后调节其OD600为0.5±0.02,并记录吸光值为 A0。将二甲苯加入到ZLp4b菌悬液中,37℃温育10min后将两相体系涡旋振荡120s,于37℃条件下静 置培养1h待两相分离后去除二甲苯相,于600nm处测定水相的吸光值A1,PBS作为空白对照。
ZLp4b的疏水性(%)公式为:A%=(A0-A1)/A0×100。
唾液乳杆菌ZLp4b的疏水性如表6所示。
由表6可知唾液乳杆菌ZLp4b的自凝集率为52.17%,与大肠杆菌的共凝集率为69.95%,疏水性为 46.39%,说明唾液乳杆菌ZLp4b能够很好的与大肠杆菌聚集,且具有良好的疏水性能,表明ZLp4b黏 附性较好。

实施例六唾液乳杆菌ZLp4b抑制病原菌黏附Caco-2细胞性能的研究
FITC标记致病菌将活化后的大肠杆菌ATCC438886000r/min离心收集菌体,用无菌PBS洗涤 3次后加入到工作浓度为500μg/mL的FITC溶液中,37℃暗处理2h后6000r/min离心菌体,PBS洗 涤3次去除未结合的FITC;将菌体重悬于RPMI-1640细胞培养液中,调整菌株浓度为2×108 CFU/mL。
唾液乳杆菌ZLp4b的制备活化后的唾液乳杆菌ZLp4b于6000r/min离心收集菌体,用无菌PBS 洗涤3次后用1640细胞培养液调整菌株浓度为2×108CFU/mL。
竞争试验实验组向培养至单层细胞的12孔细胞培养板中分别加入唾液乳杆菌ZLp4b和已标记 的致病菌各0.25mL并混合均匀,对照组加入0.5mL FITC标记的大肠杆菌,37℃、5%CO2培养箱 中孵育2h,用无菌PBS洗涤3~5次。
排斥实验实验组向培养至单层细胞的12孔培养板中加入0.5mL唾液乳杆菌ZLp4b,对照组加 入0.5mL细胞培养液,于37℃、5%CO2培养箱中孵育1h后弃去孔内液体,用无菌PBS漂洗3次 后每孔加入0.5mL FITC标记的大肠杆菌,37℃封闭孵育1h后,用无菌PBS漂洗3~5次。
置换试验向培养至单层细胞的12孔培养板中加入0.5mL FITC标记的大肠杆菌,于37℃、5% CO2培养箱中封闭孵育1h后弃去孔内液体,用无菌PBS漂洗3次,实验组加入0.5mL唾液乳杆菌 ZLp4b,对照组加入0.5mL细胞培养液,37℃封闭孵育1h后,用无菌PBS漂洗3~5次。
上述各试验结束后,向孔中加入300μL胰酶,消化细胞5min后加入0.5mL细胞培养液终止反 应。混匀细胞液体后,吸取细胞悬液0.1mL于96孔板中,并用酶标仪测定其在吸收光波长为485 nm以及发射光波长为530nm时的荧光强度。计算唾液乳杆菌ZLp4b对大肠杆菌ATCC43888黏附 Caco-2细胞的抑制率,计算公式如下:
唾液乳杆菌ZLp4b的黏附抑制率(%)=1-(实验组细胞悬液相对荧光强度/对照组细胞悬液相对荧 光强度)×100%
菌株ZLp4b对大肠杆菌黏附Caco-2细胞的影响如表7所示,菌株ZLp4b可通过竞争和置换的方式 抑制大肠杆菌的黏附,且竞争的方式抑制率更高;排斥试验结果显示,菌株ZLp4b不能通过排斥的方 式抑制大肠杆菌对Caco-2细胞的黏附。

实施例七唾液乳杆菌ZLp4b在提高小鼠机体免疫方面的研究
选取体重约为8g的昆明系小鼠30只,将小鼠随机分为三组(各10只),实验分组如下:Ⅰ组 (对照组),基础日粮+无菌MRS培养基;Ⅱ组,基础日粮+2×106CFU/只/天菌株唾液乳杆菌 ZLp4b;Ⅲ组,基础日粮+2×107CFU/只/天菌株唾液乳杆菌ZLp4b。试验期为30d,试验期间小鼠自 由采食和饮水。试验结束后,小鼠摘眼球采血,颈椎脱臼法处死小鼠。小鼠血液于室温放置过夜后 10000r/min离心,分离血清。参照试剂盒操作说明采用酶联免疫法检测小鼠血清中抗体IgA、IgM、 IgG及细胞因子IFN-γ、IL-2的含量。结果显示(表8),两种剂量的试验组与对照组小鼠血清中 IgA、IgM、IgG和IFN-γ4种检测指标的含量均高于对照组,第Ⅱ组的小鼠血清中IL-2含量略低于 对照组,第Ⅲ组的小鼠血清中IL-2含量最低。

实施例八唾液乳杆菌ZLp4b在在幼畜腹泻病中的应用研究
将本发明唾液乳杆菌ZLp4b与实验室自分离约氏乳杆菌GHZ10a及枯草芽孢杆菌BSC16a按一定比 例配伍成复合微生态制剂,初步应用于仔猪腹泻病的防治中。结果如表9所示,分别饲喂甘肃永靖、 平凉、民乐、陕西杨凌和河南明权5个养殖场共计1511头腹泻仔猪,饲喂第二天,腹泻症状明显缓 解,饲喂3天后,80%的病畜不再发生腹泻症状,持续饲喂3-5天后,病畜基本痊愈,饲喂期间甘肃平 凉养殖场无动物死亡发生,治愈率达100%,其余养殖场中个别病畜因腹泻严重及其他因素导致死 亡,其中甘肃民乐养殖场中有5头猪因脱水严重而死亡。除陕西杨凌养殖场中仔猪腹泻治愈率为87.60%外,其余养殖场中腹泻病例治愈率均高于90.00%。上述饲喂试验中,益生菌制剂对仔猪腹泻 治愈效果显著,且未出现病情反复等现象。此外,将上述复合微生态制剂初步应用于腹泻羔羊和犊 牛中,均有缓解并治愈腹泻病的效果。因此,本发明唾液乳杆菌ZLp4b可作为复合微生态制剂复配菌 株应用于幼畜腹泻病防治中。

1.一株具有广谱抑菌作用的唾液乳杆菌(Lactobacillus salivarius)ZLp4b,于2019年 9 月 6 日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCCNO:18471。
2.权利要求1所述的唾液乳杆菌ZLp4b在制备防治仔猪、羔羊、犊牛腹泻药物中的应用。
3.权利要求1所述的唾液乳杆菌ZLp4b在制备防治仔猪、羔羊、犊牛腹泻饲料中的应用。
