本発明は、生物学的製剤の精製および/または濃縮プロセスのモニタリングおよび制御の分野ならびにこれらの方法で使用する装置に関する。具体的には、本発明は、タンパク質の濃度、純度および効力のオンラインモニタリングおよび制御ならびにパラメトリックまたはリアルタイムリリースの方法に関する。
下流工程中に、生物学的製剤は、それらの用途および必要性に従って精製および濃縮される。通例、オフラインで測定される製剤の所望の純度、濃度および効力を達成するために、一連の異なる精製プロセスステップが実施される。特に、微生物または哺乳類の細胞内での過剰発現は、例えばバイオ医薬品といった生物学的製剤の組み換え製造に使用される。そのような化合物は、例として、タンパク質(たとえば、抗体およびそのフラグメント)、核酸、炭水化物、脂質、有機小分子、非有機小分子、ウイルス、リポソームおよびそのような化合物のいずれかのハイブリッドまたは異型を含むことがある。
この理由のため、例えば組み換えによって製造されたバイオ医薬品といった生物学的製剤の下流工程をオンラインでモニターするためおよび/または制御するための改良された方法を提供することが望ましい。
目的は、請求項に係わる主題によって取り組まれる。
a) 少なくとも2つの独立したオンラインセンサーを動作ユニットに含め、
b) 前記センサーのそれぞれの出力に対応する多数のプロセスデータ値を得て;
c) 生物学的製剤の濃度、純度および/または効力の予測のためにオンラインおよびオフ ラインデータから得られた多変量統計解析を行うためのコンピューターのデータベー スにデータ値をインポートし、
d) 実際のプロセスデータ値を診断し;
e) 生物学的製剤の濃度、純度および/または効力をリアルタイムでモニターし、
f) 場合により、e)で得られた情報に基づいて、プロセス調節および/またはプロセス 最適化を行い、
g) 場合により、e)で得られた情報に基づいて、パラメトリックまたはリアルタイムで 生物学的製剤をリリースする、
というステップを含む。
a) クロマトグラフィーおよび/またはろ過ユニットを含み、
b) クロマトグラフィーまたはろ過ユニットが少なくとも1つのインラインおよび1つの 非侵襲の現場センサーを含み、
c) センサーがコンピューターに接続され、そこで収集されたデータ値をデータベースに 保管および/または診断することができる。

別段の言明がない限り、説明および特許請求の範囲を含むこの文書で使用される下記の用語は、下記の定義を有する。当業者は、せいぜい日常の実験を使用して、本明細書に記載する本発明の特定の態様と同等の数多くのものを認識するもしくは解明することができる。そのような同等のものは、本発明によって包含されることが意図される。
実施例7で詳述されているように、実施例で使用される予測モデルのために、UV−VIS、伝導率、圧力、pH、MALSおよびRI装置(p=計14個の変数)、ATR−FTIRスペクトル(分解能約 2 cm-1の結果としてp=1427個の予測因子が生じる)だけでなく、計14366個の蛍光変数を与える7つの励起波長での蛍光放出スペクトル(分解能およそ0.3 nm)からのオンラインデータが収集された。予測因子セットおよび応答に応じて、7〜14回のクロマトグラフィーランの変数データがモデル構築のために使用された。この後、時間整合ステップが行われた−いくつかの装置の間の既知の時間の遅延を考慮しながら、各オフライン画分の期間に対応する各オンライン変数の平均値が計算された。それから、変数選択手法としてのブースティング(Rパッケージ mboost)と組み合わせて、STAR(構造化加法回帰)モデルによって結果が得られた。相互検証(各ランからのデータを一旦除外し、いくつかの他のランのデータに基づいたモデルで予測)を介して、自動スケールされたデータ(すなわち、各予測因子から平均値が引かれ、その標準偏差で割られる)のパラメータ最適化およびモデル選択が行われた。相互検証された二乗平均平方根誤差(RMSE)で、モデルの品質が測定された。このようにして、所期の生物学的製剤の異なる特性のためのモデルを確立することができる。

一旦生物学的製剤のために予測モデルが確立されたら、本発明の方法を使用して生物学的製剤の精製および/または濃縮をモニターおよび制御することができる。したがって、本発明は、すぐにでも使用または商品化することができる対象の製剤を得るために、任意の従来の/当技術分野において公知の発現技術を介して前記製剤を発現させ、これもまた原理上は当技術分野において公知である方法によって発現産物を精製するが、加えて本発明の方法のいずれかを適用し、場合により精製された発現産物を仕上げる、たとえば調合する、というステップを含む、対象の生物学的製剤を製造するための改良された方法にも関する。

f) 実際のプロセスデータ値を診断し;
g) 場合によりプロセス制御および/またはプロセス最適化を行い、
h) 場合により工程内、中間体および/または最終製剤として生物学的製剤をパラメトリ
ックまたはリアルタイムリリースする、
というステップを含む、少なくとも1つの動作ユニットを含む、生物学的に製造された製剤の精製および/または濃縮プロセスまたはその一部をモニターおよび制御するためのコンピューターベースの方法が提供される。
本発明の方法で使用される多変量統計解析のために、所与の生物学的製剤のための個々の各ランに対してオンラインセンサーから得られたオンラインデータがコンピューターのデータベースにインポートされ、先に記載したように予測モデルによって評価され、生物学的製剤の濃度、純度および/または効力のリアルタイムモニタリングを可能にするために診断される。
生物学的製剤の精製および/または濃縮のための動作ユニットを含む装置もまた提供され、動作ユニットは、クロマトグラフィーおよび/またはろ過ユニットを含み、クロマトグラフィーまたはろ過ユニットは、少なくとも1つの非侵襲の現場センサーを含み、センサーは、コンピューターに接続され、そこで収集されたデータ値をデータベースに保管および/または診断することができる。
本発明は、下流プロセスの実質的な経済的改良を提供する。例として、発酵製剤の最後から例えばバイオ医薬品といった最終精製生物学的製剤までのバイオプロセスシステムの全てのステージにおける異なるバッチの比較を可能にする。
a) 少なくとも2つの独立したオンラインセンサーを前記精製ユニットに適用し、
b) プロセスデータをリアルタイムでモニターし、
c) 前記センサーのそれぞれの出力に対応する多数のプロセスデータ値を得て;
d) 生物学的製剤の濃度、純度および/または効力の予測のために、前記プロセスデータ
値に対してモデルベースの多変量統計解析および数理モデルを行い;
e) 実際のプロセスデータ値を診断し;
f) 場合によりプロセス制御および/またはプロセス最適化を行い、
g) 場合により生物学的製剤をパラメトリックまたはリアルタイムリリースする、
というステップを含む。
リアルタイムでモニターされた下流プロセスの一連の動作は、図1に見ることができる。
FGF−2濃度割り出し
逆相HPLCにTSK-GEL Superoctylカラムを使用して、この方法は、ヒト線維芽細胞増殖因子2(FGF2)の定量化を可能にする。それは、タンパク質とRP−HPLCカラムの固相との間の疎水性相互作用に基づいている。FGF2は、特定のアセトニトリル(ACN)濃度で溶出する。214および280 nmでの検出に二波長吸光度が使用される。
タンパク質Aをリガンドとするモノリスカラムを使用して、この方法は、抗体の高速定量化を可能にする。タンパク質Aは、抗体のFc領域に強く結合し、したがって全抗体(モノクローナルおよびポリクローナル両方)または抗体のFc部を含有する他の巨大分子の定量化に使用することができる。結合溶出モードで解析が行われ、塩酸溶液でpH値を下げることによって溶出が達成される。280 nmの波長で吸光度が検出される。
解析的サイズ排除クロマトグラフィー−単量体含有量ならびに高および低分子量不純物の割り出し
サイズ排除クロマトグラフィーは、サイズによる分子の分離を可能にする。この技術は、溶液の中に存在するかもしれない、活性単量体標的タンパク質とより高いまたはより低い分子量の不純物との間を区別するのに使用することができる。
Quant-iT(商標)PicoGreen(登録商標)dsDNA試薬は、溶液中の二本鎖DNA(dsDNA)を定量化するための超高感度な蛍光核酸染色である。PicoGreenアッセイの線形検出範囲は、一つだけの染料濃度で、1 ng/mLから1000 ng/mLまでのDNAである。
E.coli HCP ELISAキット
このキットは、組み換え発現によって製造された製剤の中のE.coli宿主細胞タンパク質汚染の存在を割り出すために、広く反応するポリクローナル抗体を使用して開発された。Cygnusのアッセイ検証は、このキットのLODが〜0.2 ng/mLであることを割り出している。

CHO ELISAは、CHOで発現されるさまざまな抗体および他の治療用タンパク質に対して、10億分の100の範囲でHCPsを検出することができる。

細胞培養アッセイを介したFGF−2抗原結合の割り出し
FGF−2の生物活性テストのための増殖アッセイ
FGF−2効力は、NIH−3T3細胞でのFGF−2誘導増殖の原理を使用してアッセイによって測定される。この線維芽細胞株は、FGF−2と組み合わせると増殖の増加を示す。細胞は、全ての推奨される成分(DMEM培地、10%仔ウシ血清、2 mMピルビン酸ナトリウム)と一緒に培地の中に播種される。24時間後、仔ウシ血清の濃度が10%から0.5%に下がる。それから、精製されたFGF−2(10〜100 ng/mL)の濃度を増加させながら、細胞が処理される。ポジティブコントロールとして市販のFGF−2が、ネガティブコントロールとして抗HER−2mAbが使用される。較正には、市販のFGF−2が使用される(Sinobiological)。
NIH−3T3細胞が25個のTフラスコの中でダルベッコ改変イーグル培地(DMEM)、10%仔ウシ血清、2 mMピルビン酸ナトリウムを含有する培地の中で37 ℃、5%CO2、95%空気および完全湿度で培養される。細胞は、一旦〜95%の密集度に達したら、0.05%トリプシン/EDTA を使用してはがされ、トリパンブルーを用いて血球計で数えられる。それから、これらの細胞は、5×105細胞/mLの濃度で再懸濁され、96ウェルプレートに(すなわち、200 μL/ウェルで、結果として1ウェル当たり細胞が1.0×105個になる)8チャンネルピペットで加えられる。
播種から36時間後に、20 μlまたは80 μlの培地が交換される。それから、培養された細胞は、選択された試料(1ウェル当たり20 μl試料)で処理される。DMEM培地で試料の事前希釈が行われる。
試料追加から24時間後に、MTTアッセイが行われる−先行実験で、24時間のインキュベーションが最も良い結果につながる。この目的のため、MTT溶液がPBSの中に1 mg/mLで調整され、0.2 μmフィルターでろ過される。それから、20 μLのMTTがウェルに加えられる。セルは、1時間の間、37 ℃、5%CO2、95%空気および完全湿度でインキュベートされる。インキュベーション後、MTT含有培地が取り除かれ、100 μLのDMSOに取り替えられる。プレートは、さらに30分間室温でインキュベートされ、ウェルの光学密度(OD)がプレートリーダーを使用してテスト波長570 nmおよびリファレンス波長690 nmで割り出される。
rhTNF−αを用いたWEHI−164細胞毒性アッセイ
抗TNF−α scFvおよびmAb効力は、WEHI−164細胞でのTNF−α誘導アポトーシスの原理を使用して、アッセイによって測定される。WEHI−164細胞でのアポトーシスを誘導するためのTNF−αのIC50値を割り出さなければならない。それから、細胞は、精製された抗TNF−α mAbの濃度を増加させながら、TNF−αのIC50濃度で処理される。市販の抗TNF−αモノクローナル抗体がポジティブコントロールとして、非特異的scFvがネガティブコントロールとして使用される。IC50を割り出すために、市販のTNF−αが使用される(Sinobiological)。
WEHI164細胞が25個のTフラスコの中でロズウェルパーク記念研究所培地(RPMI)、10%FBSを含有する培地の中で37 ℃、5%CO2、95%空気および完全湿度で培養された。細胞は、一旦〜95%の密集度に達したら、0.05%トリプシン/EDTA を使用してはがされ、トリパンブルーを用いて血球計の中で数えられた。それから、これらの細胞は、7.5×105細胞/mLの濃度で再懸濁され、96ウェルプレートに(すなわち、150 μL/ウェルで、結果として1ウェル当たり細胞が1.0×106個になる)8チャンネルピペットで加えられた。先行実験で、細胞密度が最適化された。

表面プラズモン共鳴(SPR)分光法は、リガンド結合相互作用を測定するために使用される方法である。それは、センサー表面に固定された相互作用パートナーを持つ異なる分子の結合アフィニティーおよび反応速度を研究するための無標識技術である。それは、光学測定方法に基づいており、センサー表面上の屈折率の小さい変化を検出する。
例えば下記のように、記載する方法でタンパク質の生物活性を割り出すことができる:
● FcyRIIIa(CD16) − 抗体
● TNF−アルファ − 抗TNFα抗体
● FGFR2 − FGF−2
全てのアッセイで、ランニングバッファーとしてM HEPES、0.15 M NaCl、3 mM EDTA、0.005% Tween20、pH7.4が使用される。それぞれのアッセイのための特定の方法パラメータを表1に示す。
下記の実施例は、本発明の理解を助けるために示されているが、いかなる形であっても本発明の範囲を限定することを意図するものでも解釈されるべきでもない。実施例は、従来の方法の詳細な説明を含まない:そのような方法は、当業者にとっては周知である。オフライン方法に関する一般的な設定および情報は、先にまとめられている。リアルタイムにモニターされた下流プロセスのためのツールおよび原理の概略的な概観を図1に示す。
1つは、トラスツズマブの重鎖遺伝子およびネオマイシン耐性遺伝子を運び、他方は、トラスツズマブの軽鎖および活性が低下したDHFR遺伝子を運ぶ、という2つのプラスミドの組み合わせをCHO−S細胞(Invitrogen)に形質導入した。細胞は、エレクトロポレーションによって形質導入され、ネオマイシン耐性のために選び出され、最も高い生産性のために選別された。その結果として生じたプールは、MTX選択に供された(400 nM)。細胞は、再び生産性のために選別され、それから3ヶ月間培養の中で維持され、安定したプロデューサーに選び出すために2回再選別された。最終選別中に、細胞は、1細胞/ウェルで選別することによってサブクローンされた。最終クローンは、〜20 pg/細胞/日の比生産性を有する。
標準培地は、8 mMグルタミンおよび400 nM MTXが含まれたCD−CHO(Invitrogen)だった。2×105細胞/mlを播種し、振とうフラスコの中で140 rpm、37 ℃および7%CO2で培養することによって、バッチ培養が行われた。培養は、生存率が70%未満に落ち込んだ時、通常10日目に、終了した。フィード時にバッチ量10%でCHO CD高効率フィードBを、それにプラスして3.3%でFunction Max Titer Enhancerを使用して、振とうフラスコの中で同じ条件下でフェッドバッチ培養が行われた。
内径16mmのXKカラム(GE Healthcare)を総量21.9mlまでタンパク質Aアフィニティー培地mAb Select SuRe(GE Healthcare)で充填した。プロセス全体が流速75 cm/時で行われた。カラムは、20 mM Na−リン酸、150 mM NaCl、pH7.4を含有するランニングバッファーで、5カラム容積(CV)に対して平衡化された。細胞培養上清は、10CVに対して、無菌の0.22 μmフィルターユニット(Nalgene)でろ過され、試料ポンプS9(GE Healthcare)を通ってカラムにロードされた。ロードの後には、5CVに対して20 mM Na−リン酸、2 M NaCl、pH7.4での洗浄ステップおよび5CVに対して20 mM Na−リン酸、150 mM NaCl、pH7.4でのもう一つの洗浄ステップが続く。標的タンパク質の溶出は、10CVに対して、平衡化バッファー中のpH3.5の100%glycine−HClでのステップグラジエントで達成された。溶出画分は、0.5 M Na−リン酸pH8.0(画分容積の10%)が含まれた中和バッファーが入っている容器に収集された。カラムは、0.1 M NaOHで30分間低流速で消毒された。
組換え大腸菌の原核細胞培養からの塩基性線維芽細胞増殖因子(FGF−2)のクロマトグラフィー精製のための捕捉ステップは、一連のオンラインセンサーによって記載およびモニターされる。
限外ろ過が使用されるのは、例としてクロマトグラフィーステップの前といった他のプロセスステップの前にタンパク質を濃縮するためか、精製の後で調合のために必要な濃度に達するためのどちらかである。さらなる用途は、バッファー交換または不純物の除去である。タンジェンシャルフローろ過は、膜の上のタンジェンシャルフローが連続的にろ過ケーキを取り除いて膜の目詰まりを防止することによって、膜を通った高流量に達する。溶液は、所望の濃度に達するまで膜モジュールを通って再循環するが、凝集による膜ファウリングまたは標的タンパク質の生物活性喪失のために膜の性能が低下するため、プロセスステップを設計する時には注意しなければならない。そのため、TFFユニット動作の膜(濃縮水流)の後に、一連のオンラインセンサー(多角度光散乱、屈折率、蛍光およびATR−FTIR装置)が具現化されている。
オンラインセンサーシステムおよびオフライン測定に由来する全ての入手可能なデータは、EVONデータベースに保管される。所与のタンパク質および株のための選択されたデータセットは、コンマ区切り値ファイルに抽出され、統計計算環境R(R Core Team、2015年)にインポートされ、そこで全てのデータ処理が行われる。初期のステップとして、荷重繰返しメジアンフィルター[18]を使用してオンライン信号の信号平滑化が実施され、スペクトルデータのバックグランド補正が行われる。最終的に、オンラインデータがオフライン時間軸に対して時間整合される。
組換え大腸菌の原核細胞培養からの塩基性線維芽細胞増殖因子(FGF−2)のクロマトグラフィー精製は、オンラインセンサー(静的光散乱、示差屈折率、UV280 nm吸光度、ATR−FTIRおよび蛍光分光)によって記載およびモニターされる。リアルタイムでモニターされたデータに基づいて、画分収集が行われる。そのため、オンライン信号は、多変量統計解析で評価される。独立したトレーニングデータセットに構築される統計モデルがオンライン信号に適用され、純度、効力および濃度を予測するのに使用される。システムは、EVONのソフトウェアによって制御される。
組換え大腸菌の原核細胞培養からの塩基性線維芽細胞増殖因子(FGF−2)のクロマトグラフィー精製は、オンラインセンサー(静的光散乱、示差屈折率、UV280 nm吸光度、ATR−FTIRおよび蛍光分光)によって記載およびモニターされる。リアルタイムでモニターされたデータに基づいて、カラムにロードするステップが行われる。オンライン信号は、多変量統計解析を用いて評価される。独立したトレーニングデータセットに構築される統計モデルがオンライン信号に適用され、標的タンパク質の漏出を予測するのに使用される。システムは、EVONのソフトウェアによって制御される。
モデル化のために入手可能なオンラインデータは、UV−VIS、伝導率、圧力、pH、MALSおよびRI装置(p=計14変数)、ATR−FTIRスペクトル(分解能約2 cm-1の結果としてp=1427予測因子が生じる)だけでなく、計14366蛍光変数を与える7つの励起波長での蛍光放出スペクトル(分解能約0.3 nm)からの信号を含む。予測因子セットおよび応答変数(濃度と同じことを意味する分量)、宿主細胞タンパク質(HCP)、二本鎖DNA(dsDNA)、単量体および高分子量HMW不純物濃度または受容体生物学的製剤相互作用のKD値として表される効力に応じて、7〜14回のクロマトグラフィーランからのデータがモデル構築のために入手可能である。
例示的なモデル化の結果およびその後の目標変数のための対応するオフライン値との比較が示されている:
1. タンパク質の分量(濃度)(単位:mg/ml)(図17)

対応する結果は、図17(分量)、18〜19(宿主細胞タンパク質、単位:それぞれng/mlおよびppm)、20〜21(dsDNA、単位:ng/mlおよびppm)、22〜23(単量体および高分子量不純物画分)および24(効力KD値)に示されている。これらの図では、実際に測定されたターゲットのオフライン値がバーで示され、1秒の時間グリッド上のオンラインデータに基づくモデル予測値が曲線として描写され、これらの予測値を各オフライン画分で平均化したものが横線として示されている。
結果は、一回のクロマトグラフィーランのものが示されており、予測誤差は、総合的な予測誤差(全てのランの平均)と類似している。横軸は、時間(単位:分)を表し、原点が最初のオフライン画分の開始に配置されている。

当たりのngの単位(ppm)で宿主細胞タンパク質を予測するのにも使用することが
できる。

E=0.97(図24に示すラン3のために割り出された予測性能は、RMSE=

S、RI、蛍光およびATR−FTIR)からの予測因子が使用されている。いく
つかの予測因子の線形関数の他に、UV−VIS、伝導率、圧力、pH、MALS
、RIおよび蛍光信号の平滑関数だけでなくUV−VIS、伝導率、圧力、pH、
MALSおよびRI信号間の製剤相互作用も予測モデルに組み込まれている。
Claims (18)
- 少なくとも1つの動作ユニットを使用することを含む、生物学的製剤の精製および/または濃縮プロセスをモニターおよび制御するためのコンピューターベースの方法であり、その方法は:
a) 少なくとも2つの独立したオンラインセンサーを動作ユニットに含める、
b) 前記センサーのそれぞれの出力に対応する多数のプロセスデータ値を得る;
c) 生物学的製剤の濃度、純度および/または効力の予測のためにオンラインおよびオフラインデータから得られた多変量統計解析を行うためのコンピューターのデータベースにデータ値をインポートする、
d) 実際のプロセスデータ値を診断する;
e) 生物学的製剤の濃度、純度および/または効力をリアルタイムでモニターする、
f) 場合により、e)で得られた情報に基づいて、プロセス調節および/またはプロセス最適化を行う、
g) 場合により、e)で得られた情報に基づいて、パラメトリックまたはリアルタイムで生物学的製剤をリリースする、
ステップを含む。 - オンラインセンサーのうち少なくとも1つが多角度光散乱センサー(MALS)、UV−VIS吸収センサー、蛍光センサー、赤外吸収センサー(IR)、減衰全反射フーリエ変換赤外分光法センサー(ATR−FTIR)、屈折率(RI)センサー、pHセンサー、温度センサー、伝導率センサー、圧力センサー、小角X線散乱(SAXS)センサーおよびレドックスセンサーからなる群より選択される請求項1記載の方法。
- オンラインセンサーのうち少なくとも1つがATR−FTIR、MALS、RIおよび蛍光センサーからなる群より選択される請求項1または2記載の方法。
- オンラインセンサーのうち少なくとも1つが非侵襲の現場センサーである請求項1〜3のいずれか一項記載の方法。
- 非侵襲の現場センサーがATR−FTIR、SAXS、温度、pH、伝導率およびレドックスセンサーからなる群より選択される請求項4記載の方法。
- 動作ユニットが少なくとも3つの、少なくとも4つの、少なくとも5つの、少なくとも6つの、少なくとも7つの、少なくとも8つの、少なくとも9つの、少なくとも10またはそれ以上の独立したオンラインセンサーを含む請求項1〜5のいずれか一項記載の方法。
- 動作ユニットが少なくとも1つのATR−FTIRセンサー、1つのMALSセンサー、1つのRIセンサーおよび1つの蛍光センサーを含む請求項1〜6のいずれか一項記載の方法。
- 動作ユニットが加えて少なくとも1つの温度センサー、少なくとも1つの伝導率センサー、少なくとも1つのpHセンサーおよび少なくとも1つの圧力センサーを含む請求項7記載の方法。
- センサーのうち少なくとも1つがATR−FTIRセンサー、温度センサー、SAXSセンサー、伝導率センサー、pHセンサーおよび圧力センサーからなる群より選択される非侵襲の現場センサーである請求項7または8記載の方法。
- 動作ユニットがクロマトグラフィーユニットおよび/またはろ過ユニットを含む請求項1〜9のいずれか一項記載の方法。
- クロマトグラフィーユニットがイオン交換クロマトグラフィー、アフィニティークロマトグラフィー、サイズ排除クロマトグラフィー、逆相クロマトグラフィー、疎水性相互作用クロマトグラフィー、マルチモーダル樹脂クロマトグラフィーからなる群より選択されて、結合/溶出またはフロースルーモードでアイソクラチック、リニア、セグメント化および/またはステップグラジエント溶出で動作され、ろ過ユニットが限外ろ過、精密ろ過、ナノろ過、深層ろ過からなる群より選択されて、タンジェンシャルフローろ過、デッドエンドろ過、絶対細孔サイズ膜でのろ過で動作される請求項10記載の方法。
- ステップf)で、ピーク収集、リフォールディング、ろ過または沈殿後の生物学的製剤の正しい収集、製剤の品質およびプロセス機器メンテナンスのいずれか1つまたは組み合わせに関してプロセスが調節される請求項1〜11のいずれか一項記載の方法。
- 生物学的製剤がバイオ医薬品、核酸分子または異種タンパク質であり、好ましくは、治療用タンパク質、酵素およびペプチド、タンパク質抗生物質、融合タンパク質、炭水化物−タンパク質複合体、構造タンパク質、調節タンパク質、ワクチンおよびワクチン様タンパク質または粒子、プロセス酵素、増殖因子、ホルモンおよびサイトカイン、抗体または前記生物学的製剤の代謝産物の中から選択される請求項1〜12のいずれか一項記載の方法。
- 生物学的製剤の濃度、純度および効力が方法のステップc)で予測され、ステップe)でモニターされる請求項1〜13のいずれか一項記載の方法。
- a)生物学的製剤を製造ことができる生物または細胞を培養するステップ;
b)ステップa)から結果として生じる製剤を精製するステップであって、精製が少なくとも1つの動作ユニットを使用してモニターおよび制御され、動作ユニットが少なくとも2つの独立したオンラインセンサーを含み、生物学的製剤の濃度、純度および/または効力の予測のために、オンラインおよびオフラインデータから得られた多変量統計解析を行うためのコンピューターのデータベースにオンラインセンサーからのデータ値がインポートされ、精製の制御がリアルタイムで実際のプロセスデータ値を診断することおよび生物学的製剤の濃度、純度および/または効力をモニターするステップを含む、生物学的製剤製造方法。 - 動作ユニットが
a) クロマトグラフィーおよび/またはろ過ユニットを含み、
b) クロマトグラフィーまたはろ過ユニットが少なくとも1つのインラインおよび/または1つの非侵襲の現場センサーを含み、
c) センサーがコンピューターに接続され、そこで収集されたデータ値をデータベースに保管および/または診断することができる、
生物学的製剤の精製および/または濃縮のための動作ユニットを含む装置。 - 非侵襲の現場センサーが温度センサー、小角X線散乱センサー(SAXS)、pHセンサー、伝導率センサー、ATR−FTIRセンサーおよびレドックスセンサーからなる群より選択される請求項16記載の装置。
- 多角度光散乱センサー(MALS)、UV−VIS吸収センサー、蛍光センサー、赤外吸収センサー(IR)、減衰全反射フーリエ変換赤外分光法センサー(ATR−FTIR)、光屈折率(RI)センサー、pHセンサー、温度センサー、伝導率センサー、圧力センサー、小角X線散乱(SAXS)センサーおよびレドックスセンサーからなる群より選択された、少なくとも1つのオンラインセンサーをさらに含む請求項16または17記載の装置。
