(技術分野)
本発明は、ヒトB細胞を含む、B細胞を産生及びサポートする方法に関連し、抗原特異的抗体、より具体的には、モノクロナール抗体を産生するために用いることができる。より具体的には本発明は、ハイブリドーマ技術又はEBV−形質転換B細胞を用いることなくモノクロナール抗体を産生するためB細胞を培養することに関連する。
モノクロナール抗体は、例えば、抗体ベースの生物学的又は薬学的発展を通じて、特に医学において有用性を見出されてきた。モノクロナール抗体を産生する現在の方法は、マウスハイブリドーマ法(すなわち、抗体産生細胞を骨髄腫細胞と融合させること)を含む。その他の方法としては、EBV−形質転換B細胞系及びファージディスプレイが挙げられる。ヒトモノクロナール抗体を産生するために用いられるそれぞれの異なる方法は、それを使いにくくしている技術的な限界に苦しんでいる。例えば、ハイブリドーマ技術については、モノクロナール抗体の非ヒト部分に対する免疫反応を促進する頻度を下げるため、抗体を「ヒト化」する必要がある。加えて、特異の抗原又はエピトープに対する免疫反応は、種に特異的であり得る。その他の方法には、ヒトの治療及び診断に用いるためのモノクロナール抗体の開発に用いるためそれらを広く適用することに制約を与える欠点がある。
本発明は、培養中のB細胞を拡張及び分化するための高度に効率的な方法の発見にも基づいており、B細胞の単一細胞クローニングを可能とする。ある態様において、本発見は、培養中のB細胞を拡張及び分化するための高度に効率的な方法に関連し、ヒトB細胞の単一細胞クローニング及び抗体の産生を可能とする。
本発明は、培養中のB細胞を拡張及び分化するための高度に効率的な方法の発見に基づいており、それは、B細胞の単一細胞クローニング(単一B細胞を播種し、そのB細胞をB細胞のクローンに拡張すること)を一定量及びモノクロナール抗体を一定量生成するのに十分な時間であって、産生されたモノクロナール抗体の特徴づけを円滑化する時間内で可能とする。本方法のある態様において、ヒトB細胞は、単一細胞クローンとして拡張及び分化され、ヒトモノクロナール抗体を産生する。これらの方法の別の態様においては、本方法は培養を拡張するために抗原を必要としない及び/又は添加しない。また別の態様においては、B細胞の抗原特異性は知られていない。また別の態様において、B細胞はインビトロで抗原に曝されない。
本発明の別の態様において、フィーダー細胞系が提供され、それは改変された間葉系間質細胞又は改変された胸腺上皮細胞を含み、それらは効率的に培養中の多くの数のB細胞を促進及びサポートすることができ、それによって、改変マウス3T3細胞とともにB細胞を培養する過去の試みにおいて観察されたよりも高いレベルでのモノクロナール抗体産生を可能とする。この態様に関連して、フィーダー細胞系は、間葉系間質細胞又は胸腺上皮細胞を含み、それらは、CD154(また、CD40L又はCD40リガンドとしても知られている)及びBLyS(Bリンパ球刺激因子、またBAFF(B細胞活性化因子)としても知られている)を含む組合せ、又はCD154、BLyS、及びIL−21を含む組合せを発現するよう改変される。本発明のフィーダー細胞は、ヒトB細胞及びマウスB細胞を含む哺乳類のB細胞の著しい繁殖及び分化をサポートし、B細胞が改変されたフィーダー細胞の存在下でインビトロで培養される場合、外因性の抗原又は外因性のIL−4を加える必要がない(すなわち、無しでよい)。
本発明は、培養中のB細胞を拡張及び分化するための効率的な方法の発見に基づいており、単細胞クローンから一定量のB細胞へ、最初のクローン及び免疫グロブリン遺伝子の発現に頼ることなく更に特徴づけることを可能とするだけの十分な量のモノクロナール抗体を産生する。更なる特徴づけは、当該技術分野で公知の、1つ以上のハイスループットスクリーニングアッセイを用いた、1つ以上の抗原特異性、機能、結合及び/又は中和の特徴づけを含んでもよい。本発明は、例えば、NIH−3T3細胞系(マウス繊維芽細胞系)のような、以前マウスB細胞を拡張及び分化するため用いられたフィーダー細胞系の、培養中でヒトB細胞を拡張する際の、特にモノクロナール抗体の産生に有用な量及び分化の状態における、欠点を克服している。
「B細胞」という用語は、本明細書において用いられる場合、ヒトB細胞及びマウスB細胞などの哺乳類のBリンパ球を意味するが、これらに限定されるものではない。任意のB細胞を本発明の方法により拡張することができ、本発明の方法は、例えば、(a)B細胞の発達中にDNAの再配置によって生成する様々な抗体レパートリーを含むナイーブB細胞、及び(b)特異の抗原に対して曝されている、又は曝された記憶のあるB細胞のような抗体産生を含む。B細胞は、B細胞個体群、サブ個体群、又はその部分集合を含み、ナイーブB細胞、メモリーB細胞、活性化B細胞、B1細胞、胚中心B細胞、辺縁帯B細胞、制御性B細胞及び濾胞B細胞が挙げられるが、これらに限定されるものではないが、。
「モノクロナール抗体」という用語は、本明細書で用いられる場合、抗原のエピトープへの単一結合特異性及び親和性を示す抗体を意味する。「組換えモノクロナール抗体」は、モノクロナール抗体であって、組換え手法、例えば免疫グロブリン遺伝子配列を含有する組換え発現ベクター(例えば、VH及びVL cDNA)を用い、ホスト細胞に形質移入することによって産生されるものを指す。そのような組換え発現ベクターは、ヒト免疫グロブリン遺伝子配列を含んでもよく(例えば、ヒトVH及びヒトVL cDNA)、それらは次いでホスト細胞に形質移入される。組換えモノクロナール抗体はまた、マウスVH及びVL cDNAを含み、「ヒト化」抗体を産生するために操作され、それは、ヒトにおける治療、診断、又はセラノスティックスへの適用に用いることができる。
「抗原」という用語は、本明細書で用いられる場合、抗体の産生を刺激し、物質に特異的な抗体に結合することができる物質を意味する(すなわち、抗体は、結合特異性を有する抗原に対して特異的に結合することができる)。抗原は、タンパク質、ペプチド、脂質、炭水化物、核酸及び小分子(無機又は有機)の1つ以上を含む物質を含んでよい。抗原は、ヒトの体にとって異質な物質、ウィルス抗原、バクテリア抗原、寄生虫抗原、腫瘍抗原、毒素抗原、真菌性抗原、自己抗原、変容自己抗原(病気状態の結果として変容又は改変された自己抗原)、改変された抗原(病気状態の結果、健康な又は非病気状態の抗原に比較して、ミスフォールド、又は酸化、又は変容グリコシル化を伴う、又は過剰発現、又は変異したもの)を含んでもよい。
CD154ポリペプチドは任意のCD40アゴニストで置き換えられてよく、CD40抗体及びその断片、CD40リガンド、CD154ポリペプチド及びそのポリペプチド断片、小分子、合成薬、ペプチド(環状ペプチドを含む)、ポリペプチド、タンパク質、核酸、アプタマー、合成又は天然無機分子、模倣剤、及び合成又は天然有機分子であって、CD40を活性化することができるものが挙げられるがこれらに限定されるものではないが、。ある実施形態では、CD40アゴニストはCD40抗体である。CD40抗体は任意の形式であることが可能である。CD40に対する抗体は当該技術分野で公知である(例えば、Buhtoiarov et al., 2005, J. Immunol. 174:6013-22; Francisco et al., 2000, Cancer Res. 60:3225-31; Schwulst et al., 2006, 177:557-65を参照。これらは参照によりその全体が本明細書に組み込まれる)。CD40アゴニストは、CD154ポリペプチドであってよく、フィーダー細胞の表面で発現しても、溶解性の形態で発現してもよい。ヒトCD154ポリペプチドが実施例において用いられ、261アミノ膜結合タンパク質(配列番号:68)または149アミノ酸溶解性タンパク質として認められ、配列番号:02又は配列番号:03を含むヌクレオチド配列、又は類似のヌクレオチド配列であって、同じアミノ酸をコードしているものを含むヌクレオチド配列によってコードされることができる。この観点から、CD154は、様々な長さの膜結合タイプ又は溶解性形式として産生することができる。CD40LとCD40の相互作用のために重要なアミノ酸は、アミノ酸128から258を含むことが示されている。
別途特定又は分脈上示されない限り、「1つの(a)」、「1つの(an)」及び「その(the)」という用語は、「1つ以上」を意味する。例えば、「タンパク質( a protein)」、又は「RNA(an RNA)」は「1つ以上のタンパク質」又は「1つ以上のRNA」をそれぞれ意味すると理解されるべきである。
以下の実施例は、例示とすることのみを意図するものであり、本発明または付属する特許請求の範囲のスコープを制限することを意図するものではない。
本実施例において、本発明によるヒトB細胞のインビトロ拡張のための方法が提供される。ヘパリン処置された血液を健康な大人のヒトドナーから収集した。血液単核細胞は、Sepmate50チューブ中のフィコール−メトリゾ酸ナトリウムの層上で300xgで10分間遠心分離により単離された。末梢血単核球(PBMCs)を緩衝液(PBS)で2倍に希釈し、続いて遠心分離し(300×g、10分)、MACS緩衝液(0.5%ウシ血清アルブミン(BSA、w/v)、2.5mM PBS中ETDA)の中で〜3×107細胞/mlに再懸濁した。CD19+B細胞が、製造者の指示に従いCD19磁気ビーズを用いた陽性選択によって単離された。簡潔に述べると、単離された単核細胞は、CD19モノクロナール抗体で覆われたマイクロビーズ(60μLマイクロビーズ/mLのPBMC)と共に4℃で15分間培養され、10倍に希釈され、遠心分離により再ペレット化され、1mLのMACS緩衝液中で再懸濁された。細胞はあらかじめ湿らせたLSカラム(速やかで優しく細胞を分離することを可能とするコーティングで覆われた強磁性の球体で構成されるマトリックスを含むカラム)に磁場の下で搭載された。続いてカラムは、追加的な6mLのMACS緩衝液と共に洗浄された。CD19+細胞は、カラムを磁場から外し、10%FCS(ウシ胎児血清)を含む3mLのRPMI1640培地をカラムに添加し、別のコニカルチューブに溶出液を収集することにより溶出された。生存細胞の濃度を、顕微鏡下で死細胞のトリパン青染色とともに血球計算盤を用いて決定した。B細胞純度は、免疫蛍光染色及びフローサイトメトリー分析により、98%以上のCD19+細胞頻度とともに決定された。追加的な選択の繰り返しを、純度を高めるために用いることができる。
本実施例で例示されるのは、本発明の改変されたフィーダー細胞の、インビトロで拡張されたヒトB細胞の分化を誘発する能力である。インビトロ、MS5Duo又はMS5Trio培養システム中で拡張されたヒトB細胞を、分化の表示であるマーカーについて分析した。ヒトB細胞表現型は、PerCP−、FITC−、PE/Cy7、FITC、及びPE/Cy7−接合CD19(HIB19)、IgM(MHM−88)、IgG(HP6017)、CD38(HIT2)、及びCD138(MI15)モノクロナール抗体を用いて評価された。生存細胞はフローサイトメトリーによって分析された。MS5Duo又はMS5Trio培養システム中で拡張されたヒトB細胞は、CD19+及びCD20+のままであり、それらがB細胞起源であることが確認された(図2)。概ね80%の新たに単離されたヒト血液B細胞は、改変されたフィーダー細胞とともに培養される前に、IgMを発現し、〜4%がIgGをともに発現した(図2A)。改変されたフィーダー細胞単分子膜とともに培養して第7日に、B細胞の半分がIgMを発現し、〜16%がIgGを発現した(図2B)。培養の終わりまでに、MS5Trio培養システム中で拡張されたほとんどのB細胞は、細胞表面IgMを発現したが、IgM-IgG-IgA+CD19+B細胞の頻度は拡張した(図2B−C)。10%未満の新たに分離されたB細胞がCD38又はCD138を、改変されたフィーダー細胞と共に培養される前に発現し(図2A)、これは活性化B細胞上に発現した活性化マーカーであり、プラズマ細胞上に高い密度で発現した。培養第6−7日までには、16%のB細胞がMS5Trio培養システム中で拡張し、CD38活性化マーカーを発現した(図2B)。培養第14日までに、56%までのこれらB細胞がCD38を発現し、23%のB細胞がCD38とCD138の両方を発現した(図2C)。全く同じではないにしても類似の結果が、MS5Duo培養システム中で拡張されたB細胞について得られた。本発明の改変されたフィーダー細胞とともに培養されたヒトB細胞のいくつかは、それらの抗体分泌プラズマブラストへの活性化及び分化と整合的な表現型を獲得した。
本実施例で例示されるのは、本発明の改変されたフィーダー細胞の、インビトロで拡張されたヒトB細胞の抗体産生を誘発する能力である。インビトロ、MS5Duo又はMS5Trio培養システム中で拡張されたヒトB細胞を、抗体産生について分析した。ヒトIgG、IgM及びIgA抗体レベルは、酵素結合免疫アッセイ(ELISA)によって決定された。プレートは、細胞培養上清液(バルクB細胞培養由来であれば1:10に希釈されたもの、又は単一B細胞培養由来であれば希釈されていないもの)がプレートに添加される前に、1%のBSAを含むトリス−緩衝生理食塩水でブロックした。抗IgG1抗体と接合したアルカリ性ホスファターゼが、結合した抗体を検出するために用いられ、1Mジエタノールアミン/0.5M MgCl2が4−ニトロフェニルホスファート二ナトリウム塩六水和物とともに検出試薬として用いられた。405nmでの吸収を読み取った。対照プレートは、リン酸緩衝食塩水(PBS)中1%BSAを用いてコートし、上述のとおり、培養上清液及び検出試薬を添加する前にブロックした。抗体濃度は、商業的に入手できるヒトIgM、IgG及びIgA標準を用いて各ELISAから得られた検量線に基づいて定量した。
本発明の方法の更なる工程は、各B細胞クローンから産生したモノクロナール抗体の特徴づけ、例えば、当該技術分野で公知の方法を用いて、モノクロナール抗体の抗原に対する特異性を決定することを含んでもよい。本発明の方法に従って産生したモノクロナール抗体の特異性を決定するため、当該技術分野で公知のいくつかの方法のうち任意の1つを用いてよい。例えば、抗原アレイが今では入手可能であり、これは少量の希釈されていないB細胞クローンからの細胞培養上清液とともに100までの異なる抗原のスクリーニングを可能とし、モノクロナール抗体が特異性を有する抗原を確認する効率を、抗体産生数と並行的に著しく増加させる。簡潔に述べると、精製されたビオチン化した抗原は、ストレプトアビジンで覆われた96ウェルマイクロタイタープレートに、0.2又は0.4mmのベタ印刷ピンを用いて直接接触印刷によって滴下される。印刷されたプレートは、洗浄せずに置かれ、プレートは使用準備ができるまで4℃で保管される。スクリーニングのため、B細胞クローン上清液が直接ウェルに添加され、もし望むのであれば、場合によってはブロッキング工程を行う(例えばBSA)。印刷されたアレイ中で抗原に特異的なビオチン化されたヒト抗体が、陽性対照及び位置決めマーカーとして用いられる。B細胞クローン上清液は抗原アレイに添加され、一晩4℃で培養された。緩衝液(PBS+0.1%ツイン)で洗浄した後、アレイは2時間、抗ヒト抗体(ヒト抗体を検出するために)又は抗マウス抗体(マウス抗体を検出するために)とともに培養され、蛍光検出のため蛍光体、又は比色検出のため酵素(例えば、アルカリホスファターゼ)及び非色基質でラベルされた。抗原アレイは、最適化及び定量検出のためイメージキャプチャ及び画像化ソフトウェアを使用してハイスループット顕微鏡で分析することが可能である。
本実施例では、特異の抗原、典型的には公知の抗原、に結合するヒトモノクロナール抗体を産生するための方法が例示され、そこでは、抗原への曝露がインビボで起こり、インビボでのこの抗原への曝露の結果、ヒト個体がこの抗原に対する結合特異性を有する抗体を産生することができるB細胞を有する。ある例示されたこの方法の適用においては、個体は病態、不調、又は病気の過程を有していてよく、その結果、その個体のB細胞が病態、不調、又は病気の過程と関連又はこれらによって引き起こされる1つ以上の抗原に対し曝露されていてもよい。そのような曝露によって産生される抗体は、状態、不調、又は病気の過程の発達又は進行のいずれかに貢献してもよく(「病理的抗体」)、又はその逆に、状態又は病気の発達を阻害又は妨害する試みにおいて産生されていてもよい(「治療的抗体」)。該方法は、B細胞をそのようなヒト個体から単離し、単離されたB細胞を単一細胞に分離し、インビトロで単離されたB細胞(単一細胞)を、本発明の改変されたフィーダー細胞の存在下で、培養中で2週間未満の間に少なくとも数において104倍の拡張を達成するための条件及び十分な時間、培養する工程を含み、培養中の拡張されたB細胞クローンから産生されたものはモノクロナール抗体である。この共培養から産生した各モノクロナール抗体は、次に抗原特異性を評価してよい。
Claims (35)
- インビトロでB細胞を拡張する方法であって、
(a)少なくとも1つのB細胞を単離するステップと、
(b)ステップ(a)からの少なくとも1つのB細胞を、CD154ポリペプチド(又はCD40アゴニスト)及びBLySポリペプチド(又はB細胞生存因子)を発現するよう改変された間質細胞系を含むフィーダー細胞系とともに、IL−21と組合せて、かつ外因的なIL−4を添加することなしに培養するステップであって、該B細胞をフィーダー細胞系と、ヒトB細胞を数において拡張させるのに十分な条件の下、かつ十分な時間、培養するステップを含む、方法。 - 前記B細胞がヒトB細胞である、請求項1に記載の方法。
- 前記IL−21が外因的に培養物に添加される、請求項1又は2に記載の方法。
- 前記フィーダー細胞がIL−21ポリペプチドを発現するように改変される、請求項1又は2に記載の方法。
- 前記B細胞が、数において少なくとも平均104倍に拡張される、請求項1から4のいずれか1項に記載の方法。
- ステップ(b)における少なくとも1つのB細胞のフィーダー細胞系との培養が、2週間未満行われる、請求項1から5のいずれか1項に記載の方法。
- 1つのB細胞が単離される、請求項1から6のいずれか1項に記載の方法。
- 少なくとも1つのB細胞がステップ(a)の単離の前に抗原にさらされる、請求項1から7のいずれか1項に記載の方法。
- ステップ(b)における少なくとも1つのB細胞のフィーダー細胞系との培養が、抗原の存在下で行われる、請求項1から8のいずれか1項に記載の方法。
- ステップ(b)の培養が、抗原の添加を行うことなく行われる、請求項1から9のいずれか1項に記載の方法。
- 少なくとも1つのB細胞が、抗原に結合することによって単離される、請求項1から10のいずれか1項に記載の方法。
- ステップ(b)の後に、少なくとも10%の拡張されたB細胞がIgGを発現する、請求項1から11のいずれか1項に記載の方法。
- ステップ(b)の後に、少なくとも2.5%の拡張されたB細胞がIgAを発現する、請求項1から12のいずれか1項に記載の方法。
- 前記フィーダー細胞が請求項28から35のいずれか1項に記載のフィーダー細胞である、請求項1から13のいずれか1項に記載の方法。
- モノクロナール抗体を産生する方法であって、
(a)B細胞を単離するステップと、
(b)ステップ(a)からのB細胞を単一のB細胞に分離するステップと、
(c)複数のB細胞クローンを生成するため、単一のB細胞を、CD154ポリペプチド(又はCD40アゴニスト)及びBLySポリペプチド(又はB細胞生存因子)を発現するよう改変される間質細胞系を含むフィーダー細胞系とともに、IL−21と組合せて、かつ外因的なIL−4を添加することなしに培養するステップであって、該B細胞をフィーダー細胞系と、ヒトB細胞を数において拡張させ、モノクロナール抗体を産生するB細胞クローンに分化するのに十分な条件の下かつ十分な時間、培養するステップを含む、方法。 - さらに(d)複数のB細胞クローンにより産生された少なくとも1つのモノクロナール抗体の抗原特異性を評価するステップを含む、請求項15に記載の方法。
- さらに(e)複数のB細胞クローンにより産生された少なくとも1つのモノクロナール抗体を精製するステップを含む、請求項15又は16に記載の方法。
- 前記B細胞がステップ(a)の単離の前に抗原にさらされる、請求項15から17のいずれか1項に記載の方法。
- 前記B細胞がヒトB細胞である、請求項15から18のいずれか1項に記載の方法。
- 前記B細胞がマウスB細胞又はマウスB細胞の部分集合である、請求項15から18のいずれか1項に記載の方法。
- 抗原がステップ(c)に含まれていない、請求項15から20のいずれか1項に記載の方法。
- ステップ(c)の後に、少なくとも10%の拡張されたB細胞がIgGを発現する、請求項15から21のいずれか1項に記載の方法。
- ステップ(c)の後に、少なくとも2.5%の拡張されたB細胞がIgAを発現する、請求項15から21のいずれか1項に記載の方法。
- 前記フィーダー細胞が請求項28から35のいずれか1項に記載のフィーダー細胞である、請求項15から23のいずれか1項に記載の方法。
- 哺乳類B細胞をインビトロで拡張及び分化するためのキットであって、フィーダー細胞、生体試料からB細胞を単離するための少なくとも1つの抗体、及びB細胞を末梢血試料から収集するための試薬を含み、該フィーダー細胞が請求項28から35のいずれか1項に記載のフィーダー細胞であり、該抗体がCD19抗体であってよい、キット。
- 改変されたフィーダー細胞と単離されたB細胞を培養するために必要な、1つ以上の試薬、場合によりIL−21、を更に含む、請求項25に記載のキット。
- B細胞から産生されるモノクロナール抗体を特徴づけるためであって、1つ以上の試薬、場合によりIgM、IgG、IgA又はIgEに特異的な抗体、を更に含む、請求項25又は26に記載のキット。
- フィーダー細胞系が、CD154ポリペプチド、BLySポリペプチド、及び場合によっては、IL−21ポリペプチドを発現するよう改変される間質細胞系を含む、細胞系。
- 前記改変される間質細胞系が間葉系間質細胞系を含む、請求項28に記載のフィーダー細胞系。
- 前記改変される間質細胞系が線維芽細胞系を含む、請求項28に記載のフィーダー細胞系。
- フィーダー細胞系が、培養物中で2週間未満の間に、ヒトB細胞を数において少なくとも平均104倍に拡張することができる、請求項28から30のいずれか1項に記載のフィーダー細胞系。
- 間質細胞系がMS−5細胞系を含む、請求項28から31のいずれか1項に記載のフィーダー細胞系。
- 前記CD154ポリペプチドが、配列番号68又はその機能的断片若しくは変種を含む、請求項28から31のいずれか1項に記載のフィーダー細胞系。
