Abstract
【解決手段】特定抗原に特異的なIgG陽性B細胞を含む抗原特異的B細胞集団の製造方法であって、IgG陽性B細胞を、IL−21の存在下で、CD40、BAFF受容体、Fasを介した刺激を付与しながら、前記特定抗原と共に培養して前記特定抗原に特異的な抗原特異的B細胞を選別し、前記特定抗原に特異的なIgG陽性B細胞を含む抗原特異的B細胞集団を得ることを含む当該製造方法。

Description
しかしながら、動物個体を用いることや、高い親和性を示す抗体を産生するハイブリドーマを選択することには時間がかかり、組換え技術により得られた抗体の活性を確認する必要がある。このため、この方法では、目的とする抗体の作製に時間がかかる上に、その効果はヒトに投与するまで確定できない。
また、免疫抗原が個体毒性を示す場合には個体への免疫は困難である。さらに、動物種間で保存性の高いタンパク抗原を抗原とする場合、免疫学的寛容のために抗体が産生されにくい。
従って、本発明の目的は、特定抗原に対して特異的なIgG陽性B細胞を含む抗原特異的B細胞集団を簡便に製造する方法を提供することである。
[1] 特定抗原に特異的なIgG陽性B細胞を含む抗原特異的B細胞集団の製造方法であって、IgG陽性B細胞を、IL−21の存在下で、CD40、BAFF受容体及びFasを介した刺激を付与しながら、前記特定抗原と共に培養して前記特定抗原に特異的な抗原特異的B細胞を選別し、前記特定抗原に特異的なIgG陽性B細胞を含む抗原特異的B細胞集団を得ることを含む当該製造方法。
[2] 前記CD40、BAFF受容体及びFasを介した刺激は、CD40L、BAFF及びFasLにより付与される[1]に記載の製造方法。
[3] 前記IgG陽性B細胞は、B細胞を含む細胞集団を、CD40及びBAFF受容体を介した刺激を付与しながら、IL−4の存在下で培養する一次培養及びIL−21の存在下で培養する二次培養により培養することによって得られる[1]又は[2]に記載の製造方法。
[4] 前記CD40L、BAFF、FasL及び特定抗原を提示した担体が使用される[1]〜[3]のいずれかにに記載の製造方法。
[5] 前記CD40L、BAFF、FasL及び抗原を提示したフィーダー細胞が使用される[1]〜[4]のいずれかに記載の製造方法。
[6] [1]〜[5]のいずれかに記載の製造方法により得られた抗原特異的B細胞集団を用いてモノクローナル抗体を製造するモノクローナル抗体の製造方法。
[7] CD40L、BAFF、FasL及び特定抗原を表面に有する抗原特異的B細胞選別用フィーダー細胞。
従って、複数回の免疫を経ることなく、特定抗原に対して特異性を有するIgG陽性B細胞を含む抗原特異的B細胞集団が効率よく得られる。
以下、本発明について説明する。
また、本発明に用いられる細胞集団は、IL−21受容体(IL−21R)、CD40、BAFF受容体(BAFF−R)及びFasを有し且つ抗原を認識可能なIgG陽性B細胞を含むものであればよく、本発明の目的を妨げない範囲で分化段階における他のステージのB細胞や多種の細胞が含まれていてもよい。培養による選択の効率の観点から、IgG陽性B細胞以外のB細胞、例えば、IgE陽性細胞、CD138陽性(プラズマ)細胞や、B細胞以外の細胞、例えば、T細胞、単球、NK細胞を除去することが好ましい。
培養温度などの培養条件は、一般的なB細胞に対して用いられる培養条件をそのまま適用することができ、例えば、37℃5%CO2の条件が挙げられる。
細胞集団の培地への播種密度は、細胞集団の由来や組織から調製した細胞の状態、また同一培養系内で行う培養日数によって異なるが、一般に、1×102個〜1×107個/cm2、好ましくは1×103個〜1×107個/cm2とすればよく、特にヒトB細胞は高密度で培養を開始した方が増殖率が良いことから、好ましくは1×104個〜1×107個/cm2とすればよい。この範囲内であれば、4日間程度の培養後に、過増殖状態になることを予防できる。
遊離型CD40Lは、IgG陽性B細胞を含む細胞集団を形成するためには、B細胞が増殖を維持可能な濃度で培養系に存在すればよく、例えば、10ng/ml〜10μg/mlだが、B細胞の増殖に伴う相対的な減少を考慮して、好ましくは100ng/ml〜10μg/mlとすることができる。
FasLは、一般的にB細胞に対してアポトーシスを誘導可能な濃度で培養系に存在していればよく、例えば、10ng/ml〜10μg/ml、また抗原刺激による細胞死の抑制を誘導する観点から、好ましくは10ng/ml〜1μg/mlの濃度とすることができる。
これらの抗体又は抗体断片は、担体の表面に提示された形態であっても、抗体表面に固定化されていない遊離型又は可溶化形態であってもよい。
各分子を表面に提示するために用いられる担体としては、細胞、人工脂質二重膜、ポリスチレン若しくはポリエチレンテレフタレートなどのプラスチック、コラーゲン、ナイロン、ニトロセルロース、寒天、アガロース、セファロースなどの多糖、紙、ガラスなどを、上記分子を表面に提示するために通常用いられるものであれば、特に制限されることなく挙げることができる。担体の形状には特に限定はなく、シート状、平板状、球状、スポンジ状、繊維状などのいずれの形状としてもよい。担体としては、細胞の確実な選択性の観点から、細胞であることが好ましい。担体として使用可能な細胞としては、線維芽細胞、上皮細胞(例えばHeLa細胞)、胎児腎臓細胞(例えば、HEK293など)、濾胞樹状細胞などを挙げることができる。なかでも、増殖速度が速く、細胞表面積が大きい、フィーダー細胞の除去が簡便であるとの観点から、線維芽細胞が好ましい。
フィーダー細胞の由来としては、上述した細胞集団と同様に、哺乳動物の霊長類、有蹄類、小型哺乳類の齧歯類、鳥類などが挙げられ、好ましくは、齧歯類及び哺乳類由来のものであり、マウス、ヒトなどを例示することができる。また、フィーダー細胞は、提示対象となる上記細胞集団と同一の種に由来する細胞であってもよく、異なる種に由来する細胞であってもよい。
B細胞の抗原に対する親和性を向上させるために、工程を繰り返す毎に抗原の濃度や価数を変化(減少)させてもよい。さらに、B細胞の抗原に対する親和性を向上させるために、直前の工程の培養上澄、または培養上澄中に産生された抗体を次の工程を行う際に培養系へ加えてもよい。これにより、B細胞受容体と抗体との競合が生じて、より高い親和性のB細胞受容体をもつB細胞が選別され得る。
このような異なるサイトカインを用いた二段階の培養を行うことによって、IgG陽性B細胞が大量に増殖する。培養工程の開始時の播種密度及び細胞種によって異なるが、本培養工程を行うことにより、IgG陽性B細胞を、培養開始時の103倍〜105倍まで増やすことが可能となる。
また一次培養は、播種密度によって異なる場合もあるが、B細胞集団の増殖速度の観点から一般に培養開始後2日〜8日としてもよく、細胞集団中のIgG陽性B細胞の密度の観点から3日〜6日とすることが好ましく、3日〜5日とすることがより好ましい。
二次培養は、播種密度によって異なる場合もあるが、B細胞集団の増殖速度の観点から一般に二次培養開始後2日〜10日としてもよく、細胞集団中でのプラズマ芽細胞の数の抑制とIgG陽性B細胞の数の増加の観点から2日〜7日とすることが好ましく、2日〜5日とすることがより好ましいが、細胞の生存能が維持されれば、より長期であってもよい。
増殖工程で好ましく用いられるIL−21については、前述した二次培養又は選別工程で適用した条件をそのまま適用することができる。また、増殖工程に好ましく用いられるCD40L及びBAFFについても、前述した事項をそのまま適用することができる。
従って、本抗原特異的IgG陽性B細胞集団であれば、特定抗原に対して親和性を有するIgG陽性B細胞を大量に必要とするモノクローナル抗体の製造や、細胞治療等に好適に用いることができる。
V領域に対して変異を導入する方法としては、公知の方法を適用することができるが、変異導入の確実性の観点から、抗体遺伝子の体細胞突然変異(SHM)と抗体定常部領域遺伝子のクラススイッチ組み換え(CSR)の双方に寄与するAID(activation induced cytidine deaminase)の利用が挙げられる。あるいは、AIDの発現を増強するサイトカインなどを用いてもよい。
これにより、特定抗原に対するモノクローナル抗体を簡便に且つ迅速に得ることができる。
本フィーダー細胞を用いることによって、本発明の抗原特異的B細胞集団を効率よく得ることができる。
[A]IgG陽性B細胞の増殖
(1)細胞の培養
B細胞調製物及び他の細胞の培養は、特に断らない限り、RPMI−1640(10%(v/v) FCS、ペニシリン/ストレプトマイシン、2mM L−グルタミン、55nM 2−ME、10mM HEPES及び1mM ピルビン酸ナトリウム添加)をB細胞培養培地として用いて、5%(v/v)CO2、37℃の条件下で行った。
マウス(C57BL/6、メス8週齢)から脾臓細胞を得て、常法により、ビオチン−抗マウスCD43抗体(BD Pharmingen社製)、ビオチン−抗マウスCD4抗体(Biolegend社製)、ビオチン−抗マウスTer−119抗体(eBioscience社製)、ビオチン−抗マウスCD11c抗体(eBioscience社製)を反応させ、CD43、CD4、Ter−119及びCD11c陰性のB細胞を、Streptavidin-Particle Plus-DM、BD IMagnet(BD Bioscience Pharmingen社製)を用いて回収し、上記培養培地に懸濁してB細胞調製物を得た。
実験には、直径10cm細胞培養プレート(BD Falcon社製)に、40LB細胞を3×106個播種し、24時間培養して単一層を形成させた後、120Gyのγ線を照射してから使用した。
40LB上に、上記で得られたマウスB細胞を、マウスIL−4(10ng/ml、PEPRO TECH社製)を含有するB細胞培養培地に1.25×104個/mlの細胞密度で懸濁して、CO2インキュベーターにて培養を行った(一次培養)。
培養4日目に全細胞を、2mMのEDTAと0.5%のBSAを含むPBSを用いてフィーダーごと剥がし、ピペットでフラッシュして回収した。新たに準備された40LB細胞が播種されたプレートに、IL−21(10ng/ml、PEPRO TECH社製)を含有するB細胞培養培地に、回収された細胞集団を、2.5×103個/ml以下の細胞密度で播種して、培養を行った(二次培養)。
図3に示されるように、培養4日目ではIgG1及びIgE型抗原受容体を発現したB細胞が存在しているが、培養10日目にはほぼ全てのB細胞はIgG1型のみとなったことが確認できた。また、一部がCD138陽性(形質細胞系)となることを除けば、ほぼ全ての細胞が胚中心B細胞の表現型を示していた(GL7+、Fas+、CD38low、PNA結合性+)(データは示さず)。
[B]B1−H8ノックインマウス由来B細胞集団の抗体産生能
(1)細胞の調製
NP抗原特異的IgG陽性B細胞集団を調製するために、B1−8重鎖ノックインマウスのB細胞集団を用いた。
B1−8重鎖ノックインマウス(Lam KP et al., 1997: Cell 90:1073)の脾臓より、実施例1(1)と同様にして、ナイーブB細胞を調製した。B1−8重鎖ノックインマウスのB細胞は、4−ヒドロキシ−3−ニトロフェニルアセチル(NP)抗原に対して親和性を示すB細胞(λL鎖陽性)が約5%存在していることが知られている。
B1−8重鎖ノックインマウス由来脾臓B細胞を、実施例1と同様にして、回収した後、κ軽鎖陰性且つNP結合性B細胞を、FITC−抗マウスκ軽鎖抗体(BD Pharmingen社製)、抗FITC-microbeads(Miltenyi Biotec社製)、NP−BSA−ビオチン(Biosearch Technologies社製)、及びアビジン-microbeads(Miltenyi Biotec社製)を使用し、さらにMACS Separation Columns (Miltenyi Biotec社製)を用いて回収した。このNP特異的B細胞を、実施例1と同様にして培養開始から4日目に回収して、二次培養を開始した。二次培養開始から2日目と4日目における全B細胞中の抗NPIgG1産生細胞数を、ELISPOT法により測定した。また、実施例1と同様にしてフローサイトメトリーを用いて、IgG1陽性細胞の数を算出した。フローサイトメトリーの結果から算定したIgG1陽性細胞数を基に、抗NP−IgG1抗体産生細胞の割合(%)を算出した。具体的には、NP−CGGをコーティングしたMultiScreen(MILLIPORE社製)に、回収された細胞を200個/wellで播種し、5時間培養後、細胞を洗浄除去し、抗マウスIgG1−HRP抗体(SouthernBiotech社製)を反応後、AEC substrate(DAKO社製)を用いて発色させ、NP特異的IgG1産生細胞数を測定した。結果を図4示す。
[C]B1−H8ノックインマウス由来のIgG陽性B細胞集団の調製
(1)細胞の調製
抗原特異的IgG陽性B細胞集団が選択できることを確認するため、B1−8重鎖ノックインマウスのB細胞集団及び40LB−FcY−FL細胞を用いた。
B1−8重鎖ノックインマウス(Lam KP et al., 1997: Cell 90:1073)の脾臓より、実施例1(1)と同様にして、ナイーブB細胞を調製した。
40LB−FcY−FL細胞は、40LB細胞にマウスFas−ligand(FL)(Cell, Vol.75, pp.1169-1178 (1993))とニワトリIgY(IgG)受容体(Proc Natl Acad Sci U S A, Vol.104, pp.11718-11723 (2007))を、常法に従ってレトロウィルスベクターにより導入し(Int J Hematol, Vo.67, pp.351-359 (1998))、恒常的に発現させてクローン作製した。
40LB−FcY−FL細胞に提示する抗原としては、NP42−チキン−グロブリン(CGG)抗原(定法によりNPとCGGを共役させた)を用いた。実施例1と同様に、10cmプレートに40LB−FcY−FL細胞を播種した後、NP42−CGGを10ug/mlで1時間、室温で反応させ結合させた。
B1−8重鎖ノックインマウス由来脾臓B細胞を、実施例1と同様にして、3日間一次培養した後、2日間二次培養した。
次にNP抗原特異的B細胞の選択培養を行うため、回収した培養B細胞から、ビオチン−抗マウスIgE抗体(BD Pharmingen社製)及びビオチン−抗マウスCD138抗体(BD Pharmingen社製)とStreptavidin-Particle Plus-DM(BD Pharmingen社製)とを用いてIgE陽性細胞及び抗体産生細胞を除いた後、NP42−CGGを結合させた40LB−FcY−FL細胞でIL−21(10ng/ml、PEPRO TECH社製)と共に培養した。36時間後、全細胞を回収し、ここからビオチン−抗マウスH−2Kd抗体とStreptavidin-Particle Plus-DM(BD Pharmingen社製)を用いて40LB−FcYR−FL細胞を除き、得られたB細胞を新たな40LB細胞上でIL−21(10ng/ml、PEPRO TECH社製)を加えて3日間、培養した。
[D]AID強制発現によるGCL−B細胞における体細胞超突然変異の誘導
B1−8重鎖ノックインマウス由来脾臓B細胞を用いて、実施例1と同様に一次培養を開始し、培養2日目にAIDを、レトロウィルスベクターを用いて強制発現させた。具体的には、pMX-IRES-GFPレトロウィルスベクター(Proc Natl Acad Sci U S A, Vol.97, pp.3062-3066 (2000))のGFP遺伝子をヒトCD8(hCD8)に組み換えたpMX-IRES-hCD8レトロウィルスベクターを作成し、これにマウスAID(J Biol Chem, Vol.274, pp.18470-18476 (1999))を常法に従ってサブクローニングして作成したpMX-AID-IRES-hCD8レトロウィルスベクターを用いて、培養2日目のB細胞に感染させて培養4日目で2次培養に移行した。培養開始後2日ごとに抗軽鎖抗体を用いて抗原受容体刺激を行い、培養9日目に、ビオチン−抗ヒトCD8抗体(Biolegend社製)とアビジン-APC (eBioscience社製)を用いて染色し、FACS Vantage(BectonDickinson社製)を用いてhCD8陽性細胞をソーティングした。この細胞のゲノムに対し、鋳型として以下のプライマー(5’−GGCCGTCGACTGAGCACACAGGACCTCAC−3’:配列番号1)及び(5’−CCGGGAATTCTTCTGACTCCCAAGGTGTCC−3’:配列番号2)を用いてノックイン重鎖をPCR法により増幅し、増幅した断片をプラスミドベクターにクローニングした後、シークエンス解析を行った。
従って、実施例1及び実施例2で示される一次培養及び二次培養では、AIDを発現させることによって、重鎖V領域に高頻度の変異を導入可能であることが示された。この重鎖V領域に高頻度に変異が導入されたIgG陽性B細胞を、実施例3と同様に抗原を用いて選別することによって、この抗原に特異的なIgG陽性B細胞を含む抗原特異的B細胞集団が得られることが示唆される。
[E]ヒトB由来細胞を用いたIgG陽性B細胞集団の調製
健常人のヒト末梢血からFicoll(GE Healthcare Bio-Sciences社製)を用いて単核球を分離し、さらにCD2陰性且つCD19陽性のB細胞を、ビオチン−抗ヒトCD2抗体、FITC−抗ヒトCD19抗体、Streptavidin-Particle Plus-DM(BD Pharmingen社製)及び抗FITC microbeads(Miltenyi Biotec社製)を使用し、更にMACS Separation Columns (Miltenyi Biotec社製)を用いて回収した。回収されたB細胞を、実施例1と同様に準備した40LB上で、ヒトIL−4(50ng/ml、PEPRO TECH社製)、ヒトIL−2(25unit/ml、PEPRO TECH社製)を加えて4日間一次培養した。一次培養後の全細胞を実施例1と同様に回収し、新たに準備した40LB上に一次培養B細胞を撒き、ヒトIL−2(25unit/ml、PEPRO TECH社製)とヒトIL−21(10ng/ml、PEPRO TECH社製)を加え二次培養した。3日後に全細胞を回収し、二次培養と同条件でさらに3日間培養を続けた。
Claims (7)
- 特定抗原に特異的なIgG陽性B細胞を含む抗原特異的B細胞集団の製造方法であって、
IgG陽性B細胞を、IL−21の存在下で、CD40、BAFF受容体、Fasを介した刺激を付与しながら、前記特定抗原と共に培養して前記特定抗原に特異的な抗原特異的B細胞を選別し、前記特定抗原に特異的なIgG陽性B細胞を含む抗原特異的B細胞集団を得ること
を含む当該製造方法。 - 前記CD40、BAFF受容体及びFasを介した刺激は、CD40L、BAFF及びFasLにより付与される請求項1記載の製造方法。
- 前記IgG陽性B細胞は、B細胞を含む細胞集団を、CD40及びBAFF受容体を介した刺激を付与しながら、IL−4の存在下で培養する一次培養及びIL−21の存在下で培養する二次培養により培養することによって得られる請求項1又は請求項2記載の製造方法。
- CD40L、BAFF、FasL及び特定抗原を提示した担体が使用される請求項1〜請求項3のいずれか1項記載の製造方法。
- 前記CD40L、BAFF、FasL及び特定抗原を提示したフィーダー細胞が使用される請求項1〜請求項4のいずれか1項記載の製造方法。
- 請求項1〜請求項5のいずれか1項記載の製造方法により得られた抗原特異的B細胞集団を用いてモノクローナル抗体を製造するモノクローナル抗体の製造方法。
- CD40L、BAFF、FasL及び特定抗原を表面に有する抗原特異的B細胞選別用フィーダー細胞。
