Abstract
Description
本発明の組成物は、黒生姜と、生姜、桂皮、薬用人参及びビタミンBからなる群から選ばれる少なくとも1種類の成分とを含有する。本発明の組成物は、その態様によって、PPARγ発現促進組成物、皮膚外用組成物、抗肥満組成物、褐色脂肪活性化組成物、褐色脂肪増殖組成物、冷え症改善組成物、熱産生促進組成物、代謝促進組成物、エネルギー消費促進組成物、及び抗インスリン抵抗性組成物に分けられる。上記代謝促進組成物は特に限定されないが、例えば、脂質代謝促進組成物である。
黒生姜の根茎を洗浄後、1〜10mm程度にスライスし、1日天日干しにした。その後、40〜100℃に設定したオーブン乾燥機で4〜6時間乾燥し、粗粉砕後、130〜200℃で5〜20秒間殺菌を行った。殺菌した粗粉砕物を粉砕機によって粉砕し、黒生姜の根茎粉砕物を得た。同様の方法により、黒生姜の茎粉砕物、葉粉砕物及び花粉砕物を得た。これらを黒生姜粉砕物とした。
上記した方法により得た黒生姜粉砕物 300gを秤量し、60%(V/V)エタノール 3Lと共に三角フラスコに入れた。途中で何回か攪拌しながら室温で24時間静置して1回目の抽出を行った。これを減圧ろ過して、1回目の抽出液を得た。減圧ろ過後の残渣を60%(V/V)エタノール 3Lに浸漬して、室温で24時間静置して2回目の抽出を行った。これを減圧ろ過して、2回目の抽出液を得た。これら1回目及び2回目の抽出液を併せた抽出混合液を減圧濃縮して黒生姜抽出物を得た。得られた黒生姜抽出物は、57DMF含量が9.27%(W/W)であった。
得られた黒生姜抽出物または発酵黒生姜抽出物、生姜抽出物、桂皮粉末、薬用人参抽出物、ビタミンB1粉末を被験物質として用いた。
コラゲナーゼが0.2%(W/V)、BSAが1%(W/V)となるように調製したDMEM培地を、室温で1時間振盪することによって、コラゲナーゼ溶液を調製した。
Sea:ICRマウスをジエチルエーテルで安楽殺した後、背部から褐色脂肪組織を摘出した。得られた褐色脂肪組織から、ピンセットを用いて、褐色脂肪の周辺に付着している脂肪組織、血管、筋肉などを取り除いた後、褐色脂肪をDMEM中で3回洗浄した。次いで、コラゲナーゼ溶液に褐色脂肪を浸し、溶液中で褐色脂肪をハサミで細かく切り刻む細断処理に供した。細断処理した褐色脂肪をコラゲナーゼ溶液中で、37℃で1時間インキュベートすることにより細胞分散液を得た。得られた細胞分散液を、100μmの孔径のセルストレイナーでろ過し、次いで遠心処理(800rpm、5min、20℃)に供した。次いで、沈殿した細胞をDMEMで懸濁して、細胞懸濁液を得た。次いで、オートクレーブで滅菌処理した25μmフィルターを用いて、得られた細胞懸濁液をろ過した。ろ過上清を遠心(800rpm、5min、20℃)した後、沈殿した細胞を10%(V/V)FBS含有DMEMで懸濁し、褐色脂肪細胞液とした。
増殖培地として、10%(V/V)FBS含有DMEMを用いた。
75cm2フラスコにコラーゲンコート溶液を5ml入れ、室温で約1時間静置後、コラーゲンコート溶液を吸引除去し、5mlのPBSで2回洗浄を行うことによって、コラーゲンコートフラスコを得た。
全体を100質量部として、黒生姜抽出物 0.01質量部、生姜抽出物 0.01質量部、グリセリン 10質量部、ジグリセリン 3質量部、1,3−ブチレングリコール 12質量部、ペンチレングリコール 3質量部、ヒアルロン酸ナトリウム 0.1質量部、クエン酸 0.01質量部、クエン酸ナトリウム 0.02質量部、キサンタンガム 0.1質量部、メチルパラベン 0.15質量部、カルボマー 0.2質量部、水酸化ナトリウム 0.03質量部及び水 残部を混合して、化粧水の態様で本発明の組成物を調製した。
全体を100質量部として、黒生姜抽出物 0.01質量部、桂皮抽出物 0.02質量部、ラウレス硫酸ナトリウム 7.5質量部、コカミドプロピルベタイン 4.2質量部、コカミドDEA 3質量部、1,3−ブチレングリコール 0.1質量部、ポリクオタニウム−10 0.225質量部、クエン酸 0.15質量部、クエン酸ナトリウム 0.05質量部、フェノキシエタノール 0.9質量部及び水 残部を混合して、シャンプーの態様で本発明の組成物を調製した。
全体を100質量部として、黒生姜粉砕物 0.5質量部、薬用人参抽出物 0.2質量部、グリセリン 2質量部、オリーブ油 1質量部、EDTA−4ナトリウム 0.1質量部、エチドロン酸4ナトリウム 0.2質量部及び石ケン素地 残部を混合及び固化することにより、石鹸の態様で本発明の組成物を調製した。
全体を100質量部として、黒生姜抽出物 0.1質量部、チアミン塩酸塩 0.1質量部、ショ糖脂肪酸エステル 3質量部、グリセリン 12質量部、スクアラン 6質量部、ジメチルシリコーンオイル 24質量部、ポリプロピレングリコール 1質量部、増粘剤 0.06質量部、フェノキシエタノール 0.2質量部、エタノール 5質量部、水酸化ナトリウム 0.01質量部及び精製水 残部を混合して、乳液の態様で本発明の組成物を調製した。
全体を100質量部として、黒生姜抽出物 0.1質量部、オタネニンジンエキス 0.1質量部、スクワラン 15.0質量部、ミリスチン酸オクチルドデシル 4.0質量部、水素添加大豆リン脂質 0.2質量部、ブチルアルコール 2.4質量部、硬化油 1.5質量部、ステアリン酸 1.5質量部、親油型モノステアリン酸グリセリン 1.5質量部、モノステアリン酸ポリグリセリル 0.5質量部、ベヘニルアルコール 0.8質量部、モノミリスチン酸ポリグリセリル 0.7質量部、サラシミツロウ 0.3質量部、d−δ−トコフェロール 0.1質量部、メチルパラベン 0.3質量部、C10〜30アルキル変性カルボキシビニルポリマー 0.2質量部、カルボキシビニルポリマー 0.1質量部、1,3−ブタンジオール 18.0質量部、水酸化ナトリウム 0.1質量部及び精製水 残部を混合して、化粧用クリームの態様で本発明の組成物を調製した。
全体を100質量部として、黒生姜抽出物 0.1質量部、ケイヒエキス 0.01質量部、ポリビニルアルコール 20.0質量部、グリセリン 5.0質量部、エタノール 20.0質量部、カオリン 6.0質量部、防腐剤 0.2質量部、香料 0.1質量部及び精製水 残部を混合して、パック剤の態様で本発明の組成物を調製した。
全体を100質量部として、黒生姜粉末 10質量部、ケイヒ末 8質量部、ビタミンB1 5質量部、結晶性セルロース 20質量部、乳糖 50質量部、ステアリン酸マグネシウム 4質量部及びコーンスターチ 残部を混合及び打錠することにより、錠剤の態様で本発明の組成物を調製した。
全体を100質量部として、黒生姜粉末 10質量部、薬用ニンジン末 15質量部、乳糖 10質量部、ステアリン酸カルシウム 1質量部及び結晶セルロース 残部を混合及び顆粒化することにより、顆粒剤の態様で本発明の組成物を調製した。
全体を100質量部として、黒生姜抽出物 10質量部、生姜抽出物 20質量部、レシチン 8質量部及びオリーブ油 残部を混合して調製したものを内容液として、これをカプセル殻に内包することにより、カプセル剤の態様で本発明の組成物を調製した。
全体を100質量部として、黒生姜エキス粉末 0.84質量部、ビタミンB1 1質量部、果糖ブドウ糖液糖 10質量部、クエン酸 1質量部、安息香酸ナトリウム 0.02質量部、香料製剤 2質量部、スクラロース 0.05質量部、アセスルファムカリウム 0.03質量部、及び精製水 残部を混合して、液剤の態様で本発明の組成物を調製した。

- 黒生姜のエタノール抽出物又は含水エタノール抽出物と、薬用人参又はビタミンB1とを含有する冷え改善組成物。
