- 前記改変クロストリジウム毒素は、pIが少なくとも6である、先行請求項の何れかに記載の改変クロストリジウム毒素。
- 前記改変クロストリジウム毒素は、pIが6乃至10である、先行請求項の何れかに記載の改変クロストリジウム毒素。
- 前記改変クロストリジウム毒素は、pIが7乃至9、好ましくは8乃至9である、請求項7記載の改変クロストリジウム毒素。
- 前記少なくとも1つのアミノ酸修飾は、アミノ酸置換、アミノ酸挿入、及びアミノ酸欠失から選択される、先行請求項の何れかに記載の改変クロストリジウム毒素。
- 前記少なくとも1つのアミノ酸置換は、酸性アミノ酸残基の塩基性アミノ酸残基による置換、酸性アミノ酸残基の非荷電アミノ酸残基による置換、及び非荷電アミノ酸残基の塩基性アミノ酸残基による置換から選択される、請求項9記載の改変クロストリジウム毒素。
- 前記改変クロストリジウム毒素は、1乃至80個のアミノ酸修飾を含む、先行請求項の何れかに記載の改変クロストリジウム毒素。
- 前記改変クロストリジウム毒素は、4乃至40個のアミノ酸修飾を含む、請求項11記載の改変クロストリジウム毒素。
- 前記少なくとも1つのアミノ酸修飾は、クロストリジウム毒素の受容体結合ドメイン(HCドメイン)に位置する、先行請求項の何れかに記載の改変クロストリジウム毒素。
- 前記少なくとも1つのアミノ酸修飾は、クロストリジウム毒素のHCNドメインに位置する、請求項13記載の改変クロストリジウム毒素
- 前記少なくとも1つのアミノ酸修飾は、表面露出アミノ酸残基の修飾である、先行請求項の何れかに記載の改変クロストリジウム毒素。
- 前記少なくとも1つのアミノ酸修飾は、アスパラギン酸残基、グルタミン酸残基、ヒスチジン残基、セリン残基、スレオニン残基、アスパラギン残基、グルタミン残基、システイン残基、又はチロシン残基から選択されるアミノ酸残基の修飾を含む、先行請求項の何れかに記載の改変クロストリジウム毒素。
- 前記アミノ酸残基は、リジン残基又はアルギニン残基により置換される、請求項16記載の改変クロストリジウム毒素。
- 請求項1乃至17の何れかに記載の改変クロストリジウム毒素をコードする核酸配列を含む核酸。
- 軽鎖及び重鎖を有する単鎖改変クロストリジウム毒素タンパク質を生産する方法であって、請求項18記載の核酸を適切な宿主細胞内で発現させることと、前記単鎖改変クロストリジウム毒素タンパク質を含有する宿主細胞のホモジネートを提供するために前記宿主細胞を溶解することと、前記単鎖改変クロストリジウム毒素タンパク質を単離することと、を含む方法。
- 改変クロストリジウム毒素を活性化する方法であって、請求項19記載の方法により取得可能な単鎖改変クロストリジウム毒素タンパク質を提供することと、前記軽鎖と重鎖との間に位置する認識部位(切断部位)においてポリペプチドを切断するプロテアーゼにポリペプチドを接触させることと、前記ポリペプチドを、前記軽鎖と重鎖とがジスルフィド結合により互いに連結された二本鎖ポリペプチドに転換させることと、を含む方法。
- 医薬において用いる、請求項1乃至17の何れかに記載の改変クロストリジウム毒素。
- 斜視、眼瞼痙攣、斜視(squint)、ジストニア(例えば、痙性ジストニア、顎口腔ジストニア、限局性ジストニア、遅発性ジストニア、喉頭ジストニア、四肢ジストニア、頸部ジストニア)、斜頚(例えば、痙性斜頚)、細胞/筋肉の(SNAREの下方制御又は不活性化による)無能力化が有効となる美的療法(美容)用途、眼球運動の神経筋障害又は状態(例えば、共同斜視、上下斜視、外側直筋麻痺、眼振、甲状腺機能低下ミオパチー)、書痙、眼瞼痙攣、歯ぎしり、ウィルソン病、振戦、チック、分節性ミオクローヌス、攣縮、慢性多発性硬化症による痙縮、異常な膀胱の制御をもたらす痙縮、アニムス、背部痙攣、筋肉硬直、緊張性頭痛、骨盤挙筋症候群、二分脊椎、遅発性ジスキネジア、パーキンソン病、吃音症、片側顔面痙攣、眼瞼障害、脳性麻痺、限局性痙縮、痙性大腸炎、神経因性膀胱、アニスムス、四肢痙縮、チック、振戦、歯ぎしり、裂肛、アカラシア、嚥下障害、流涙、多汗症、過剰唾液分泌、過剰消化管分泌、筋肉痛(例えば、筋痙攣による痛み)、頭痛(例えば、緊張性頭痛)、額の皺、皮膚の皺、癌、子宮障害、泌尿生殖器障害、泌尿生殖器神経障害、慢性神経性炎症、及び平滑筋障害から選択される疾患又は状態の予防又は治療に用いる、請求項1乃至17の何れかに記載の改変クロストリジウム毒素。
- 実質的に本明細書において添付図面を参照して説明された改変クロストリジウム毒素。
