Claims (14)
- N476、S564、N578、E599、L647、D650、D651、V675、I685、N687、T755、E757、N761、N763、I831、T847、及びI849からなる群から選択される少なくとも1つのアミノ酸のリジン又はアルギニンでの置換を含み、かつ、少なくとも6.6の等電点(pI)を有する改変ボツリヌス毒素血清型A(BoNT/A);
ここで、
前記少なくとも1つのアミノ酸置換は、当該改変BoNT/AのpIを、前記少なくとも1つのアミノ酸置換を欠く、その他の点においては同一のBoNT/AのpIよりも、少なくとも0.2pI単位高い値に増大し;
当該改変BoNT/Aは、前記少なくとも1つのアミノ酸置換を欠く、その他の点においては同一のBoNT/Aの標的細胞結合、転位置及び標的SNAREタンパク質(単数又は複数)の切断の能力を保持し; かつ
当該改変BoNT/Aは、
a. 配列番号2、4、及び6から選択される核酸配列に対して少なくとも90%の配列同一性を有する核酸配列によってコードされる; 及び/又は
b. 配列番号1、3、及び5から選択されるアミノ酸配列に対して少なくとも90%の配列同一性を有するアミノ酸配列を含む。 - 前記改変BoNT/Aが:
a. 配列番号2、4、及び6から選択される核酸配列に対して少なくとも95%の配列同一性を有する核酸配列によってコードされる; 及び/又は
b. 配列番号1、3、及び5から選択されるアミノ酸配列に対して少なくとも95%の配列同一性を有するアミノ酸配列を含む;
請求項1に記載の改変BoNT/A。 - 前記改変BoNT/Aが:
a. 配列番号2、4、及び6から選択される核酸配列に対して少なくとも99%の配列同一性を有する核酸配列によってコードされる; 及び/又は
b. 配列番号1、3、及び5から選択されるアミノ酸配列に対して少なくとも99%の配列同一性を有するアミノ酸配列を含む;
請求項1又は2に記載の改変BoNT/A。 - 前記置換は、改変BoNT/AのpIを、前記置換を欠く、その他の点においては同一のBoNT/AのpIよりも、少なくとも0.5pI単位高い値に増大する、請求項1から3のいずれか一項に記載の改変BoNT/A。
- 前記置換は、改変BoNT/AのpIを、前記置換を欠く、その他の点においては同一のBoNT/AのpIよりも、少なくとも1pI単位高い値に増大する、請求項1から4のいずれか一項に記載の改変BoNT/A。
- 前記置換は、改変BoNT/AのpIを、前記置換を欠く、その他の点においては同一のBoNT/AのpIよりも、少なくとも2pI単位高い値に増大する、請求項1から5のいずれか一項に記載の改変BoNT/A。
- 前記置換は、改変BoNT/AのpIを、前記置換を欠く、その他の点においては同一のBoNT/AのpIよりも、2から5の間のpI単位高い値に増大する、請求項1から6のいずれか一項に記載の改変BoNT/A。
- 少なくとも3つのアミノ酸置換を含む、請求項1から7のいずれか一項に記載の改変BoNT/A。
- 4箇所から40箇所の間のアミノ酸置換を含む、請求項1から8のいずれか一項に記載の改変BoNT/A。
- 請求項1から9のいずれか一項に記載の改変BoNT/Aをコードする核酸配列を含む、核酸。
- 軽鎖及び重鎖を有する単鎖改変BoNT/Aタンパク質を生産する方法であって、請求項10に記載の核酸を、適した宿主細胞中で発現させることと、宿主細胞を溶解して、単鎖改変BoNT/Aタンパク質を含有する宿主細胞ホモジネートを提供することと、単鎖改変BoNT/Aタンパク質を単離することとを含む、方法。
- 改変BoNT/Aを生産する方法であって、請求項11に記載の方法によって得られる単鎖改変BoNT/Aタンパク質を提供することと、当該単鎖改変BoNT/Aタンパク質を、当該単鎖改変BoNT/Aタンパク質を軽鎖と重鎖の間に位置する認識部位(切断部位)で切断するプロテアーゼと接触させることと、当該単鎖改変BoNT/Aタンパク質を、軽鎖及び重鎖がジスルフィド結合によって一緒に連結している活性化二本鎖ポリペプチドに変換することとを含む、方法。
- 請求項1から9のいずれか一項に記載の改変BoNT/A又は請求項12に記載の方法によって得られる改変BoNT/Aを含む医薬。
