Abstract
Description
本明細書の文脈において、ポリアルキルオキサゾリン(POx)という用語は、アルキル誘導体(特にメチル−、エチル−、又はプロピル−置換された)オキサゾリン又はオキサジンのポリマーを指す。
本発明の第1の側面によれば、以下の工程を含むマクロ多孔質ゲルを提供するための方法を提供する。
a)第1及び第2の溶質を含む水溶液を提供する工程であって、ここで第1の溶質は、ポリエチレングリコール(PEG)の架橋可能な誘導体及びポリオキサゾリンの架橋可能な誘導体から選択される架橋可能なポリマーであり;及び第2の溶質はコスモストロピック試薬であり、
b)該混合物へ架橋可能なポリマーを架橋することができる架橋試薬を添加し、それにより架橋可能なポリマーのゲル化及び相分離を同時に開始する工程。

PEGは、その水組織化特性によって、デキストラン及びいくつかの塩と水性二相系を形成することは古くから知られいている。この物理的効果は、タンパク質抽出のために広く使われてきており、ゲルミクロスフィア形成に有用であることが示されているが、マクロ多孔質足場のインサイチュ自己会合についての可能性はまだ示されていない。これはおそらく、相分離は典型的には高速ミクロスフィア形成、それに続く融合及びデカンテーションを通じて進み、それは細孔及びゲルの浸透したネットワークを提供しないからである。相互接続されたネットワークは、PEG/デキストラン混合物の高割合のデキストランをタンブリングしながら一晩光架橋することによって得ることができた。その利点にもかかわらず、この方法はインサイチュゲル化及び細胞封入を可能とせず、そして得られる足場の透明性は低い。本発明者らは、細胞封入及び注入に適合する方法でマクロ多孔質ハイドロゲルを作製するための鍵が、低いポリマー含有量及び高い粘度を組み合わせることであることを見出した。低いポリマー含有量は、PEG及び多糖類が最初は混和性であることを保証する。次に、水系において分子量に大きく依存する相分離は、架橋試薬存在下で起こるPEG鎖伸長によって誘発される(図7)。
粘度の増加はゆるやかな分布を与え、よって動態を与える。しかし、この効果は、容積排他を通した濃縮による相殺以上であり、そして結果として亢進された動態で多孔質PEGゲルが形成される。特に興味深いことに、これが生理的なpHで最適な動態のゲルの形成を可能にし、一方、同様に良好なゲル化時間は一般的にはpH8.0でのみ達成され(図2a/c)、それは一般的に細胞にストレスを与え、神経細胞のような感受性細胞の損傷又は死をもたらす。高粘度はまたゲル化が起こる前に細胞が沈降することを防止する。
DRGは、完全な移植片として、末梢軸索再生の最も一般的なインビトロモデルであり、化学的及び生物学的ストレスに対する耐性のため、培養が容易である。それらは、移植片の周囲のプロテアーゼの濃度が高くなるために数日以内に分解する傾向があるにもかかわらず、それらは、通常様々な基質の生理活性を評価するために2Dで、及びフィブリンまたはコラーゲン中の3Dで培養する。本発明者らは非分解性多孔質PEGゲルが末梢軸索浸潤に許容的であり、そして長時間安定であることを見出した(図3)。このため、E9.5ニワトリ胚由来のDRG移植片を、HA 0.525%(w/v)及びデキストラン 1.4%(w/v)存在下で、PEGジチオールの化学量数で架橋した4アームPEG−VS 0.8%(w/v)から形成されるPEGゲルのディスク(4 mm×1.5 mm)に封入した。30日後の生存細胞プロセス(カルセインAMで染色)の全ゲル画像化により、ゲル全体で軸索の密集した網が侵入していることが明らかとなり、許容性および長期安定性が示された。ヨウ化プロピジウム(PI、死細胞)/カルセインAM(生細胞)染色パターンから観測されるように、細胞死は移植片の表面では明白であったが、内部ではみられなかった。これは、外側の細胞層が封入に関連するストレスから内部細胞を保護するのに役立つことを示唆している。
初代ラット皮質神経細胞をHAの量を増加させながら形成した多孔質ゲル中に封入した(図4)。神経細胞生存率、神経突起伸長割合及びゲル分解は、3D神経ネットワーク形成について最適化するために最も重要なパラメーターであるため、定量化した。市販のQ−ゲルは、現在の最良の標準を代表するものであり、多くの異なる細胞の封入に成功しているため、非多孔質な古典的なPEGゲルの対照として使用した。
初代ラット神経細胞の培養は、長年、細胞生物学研究のための対照モデルとされてきたが、ヒトの神経学的条件のより関連性の高いモデルとしてhiPSC由来神経細胞が出現してきている。それらは、特定の突然変異又は薬剤の効果を研究することができる、規定された細胞の再現性ある供給を提供し、そしてそれらは治療可能性を有すると考えられている。それらは、典型的には2Dで、規定された無血清培地中で増殖するが、しかし3D培養には一般的にマトリゲルが使用される。ここで、本願発明者らは、臨床応用への転換を妨げる免疫原性材料に依存しないhiPSC由来神経細胞について効果的に規定された3D培養系を確立する。hiPSC由来神経細胞を高い生存率(最大85%)を有する0.525%(w/v)HAで微細構造化されたPEGゲル中に封入し、そして初代神経細胞と同じ高速度で神経突起を伸長した(図6)。それらは剛性及び細胞密度に非常に敏感であり、ゲルを著しく速く分解した。結果として、ゲルは接続したネットワークを形成するために必要な5日間は安定であったが、残念ながらスパイク活性を得るために必要な2〜3週間は安定ではなかった。速い神経突起伸長及び高い生存率を有して1週間以上ゲルを安定させる最良の妥協点は、これらの微細構造化条件において2%(w/v)PEG及び5M cell/mlであった。細胞は、神経細胞マーカーであるニューロフィラメント、βIII−チューブリン及びMAP2に対して陽性であり、星状細胞マーカーGFAPに対して陰性であり、完全に分化していた。
典型的には塩析出、凍結乾燥又はエレクトロスピニングによって巨視的又は階層的な多孔性を有する足場を製造するための多くの方法が開発されてきた。しかしながら、それらの方法は、細胞を封入又はインサイチュ形成が可能ではない。ここで本願発明者らは、PEGが段階的な成長重合を起こす際に、PEGと高粘度の多糖類との間の相分離により形成される多孔質構造に細胞を直接封入することが可能な独創的な方法を提示する。速い神経突起伸長及びシナプス形成を可能にする規定された3D培養は、軸索誘導、成長円錐機構、シナプス会合及び再生のような神経細胞−マトリックス相互作用に依存する研究について主要な関心事である。空白のバックグランドにおいて細胞外合図の生物学的活性を研究するための可能性は重要なツールであり、特に神経系疾患の再現性ある重要なモデルを提供する、様々な患者特異的及びトランスジェニック細胞系由来の市販されているhiPSC由来神経細胞と組み合わせた重要なツールである。それらの完全な透明性により記載した微細構造化ゲルは特に画像化研究に適している。
特に明記しないかぎり、全ての化学物質はシグマ−アルドリッチ製であり、細胞培養試薬はインビトロジェン製であり、実験は3回行っており、画像は最小限に改変している(神経突起と細胞本体の両方が鮮明に区別できるように、全ての図において輝度/コントラスト及び神経細胞の図のみガンマを調節した)。
20 kDaの4アーム−PEG−チオール(Laysan Bio)1 g(50 μmol)を300mM、pH8.0のトリエタノールアミン(TEOA)緩衝液4mlに溶解し、そしてこれを直ちに、1004 μl(10 mmol)のビニルスルホンを溶解した同緩衝液4mlへ激しく撹拌しながら滴下して添加した。該反応は数分以内に実質的に完了し、そして2h進行させた。生成物をmQ−水に対して透析し(各少なくとも6時間に対して少なくとも6回水の交換)、凍結乾燥した。チオールはプロトン−NMR(400 Mhz装置で重クロロホルム中)の検出限界内において完全に置換されていることが確認された。生成物をmQ−水中に4%(w/v)で再懸濁し、ろ過滅菌し、200 μl(8 mg)に等分し、凍結乾燥し、使用するまで−20℃で保存した。
Ac−GCRD−GPQGIWGQ−DRCG−NH2(1746 Da)配列を有するMMP切断可能なペプチド(ナガセ H., Biopolymers 40, 339−416, 1996; ルトルフ, B. M. P., Adv. Mater. 15, 888−892, 2003)はAnawa(スイス)製で、純度95%以上であった。HPLC精製されたペプチドは、酸性TFA塩として生じ、それは中和する必要がある。この目的のため、〜100 mgのペプチド粉末を最小量の水(〜1 ml)に分散させ、pH指示薬として最小量のフェノールレッドを補充し、そして300 mMのNaOH水溶液で中和した(黄色の溶液をオレンジに変化させた)。プロトン化された形態のペプチドは難溶性であるが、最初のスラリーは工程中に透明になる。チオールがpH7.0より大きい(指示薬は赤みがかった橙色に変わる)と速やかに酸化するので過剰のNaOHを添加することは避けることが重要である。次いで、中和したペプチドをmQ−水で1%(w/v)に希釈し、ろ過滅菌し、400 μl(4 mg)に等分し、凍結乾燥し、密封して使用するまで−20℃で保存した。この形態で少なくとも6ヶ月は安定であることがわかった。ペプチド粉末は典型的には有意な量の対イオン及び少量の酸化ペプチドを含むため、各バッチについて、再懸濁した分割量中の遊離チオールの正確な濃度をβ−メルカプトエタノール較正曲線を用いたエルマンアッセイ(エルマン, G. L., Arch. Biochem. Biophys. 70−77, 1959)で決定した。pHバランスのために使用する少量のフェノールレッドは、このアッセイの文脈においては無視できるほどの吸光度を有するように制御した。
非常にpH感受性であるマイケル付加(ルトルフ, M.P., Biomacromolecules 4, 713−722, 2003)によるゲル形成で使用するために、特別に注意を払ってHEPES緩衝液(200 mMのHEPES及び100 mMのNaOH mQ−水の水溶液、pH7.4に調整)を調製した。558/433 nmでのpH指示薬フェノールレッドの吸光度測定は、より漂う傾向にある較正溶液及び電極に頼らず再現性のある実験を保証するために、最も信頼できるpH値を提供すると判明した。558/433 nmでの吸光度の比は2に調整し、これは24℃、pH7.40及び300 mMイオン強度に相当する(ヤオ, W., Environ. Sci. Technol. 35, 1197−201, 2001)(両性イオンが2つの電荷を有していると仮定している。代わりにそれらが電荷を持たない分子のように扱われる場合は、これは100 mMイオン強度及びpH7.5に相当する)。
高粘度(図S1参照)バクテリアMA(エルテバーグ, H., Methods Biotechnol. 10, 71−78, 1999)は、スクジャック−ブラエク教授(ノルウェー科学技術大学バイオポリマー研究所)から寄贈されたものである。HAとは異なり、アルギン酸塩は哺乳動物には存在しないため、それ故にいずれの神経細胞受容体によっても特異的に認識されないと予想される。さらに、アルギン酸塩のこの形態は純粋な粘性があり、グルロン酸塩(野生型アルギン酸塩のカルシウム架橋を可能にする)が欠損しているためである。
支持体は事前に準備した。1 mmの層のPDMS(Sylgard(登録商標)184 10%(w/v)架橋剤)を50℃で2時間、ペトリ皿上で加熱した。4及び6 mmの連続的な同心の打ち抜き(カイメディカル製真皮パンチ)でリング状の型を得た。ウェル当たり1つの滅菌カバースリップを用い、その上に1つのPDMSモールドを置いた24ウェルプレートを準備した。PDMSはガラスに貼り付いており、そしてゲルはこの支持体中に直接作製することができ、これにより、神経細胞を機械的に妨害することなく全て容易な移動、画像化又は免疫染色が可能となる。他の細胞型において通例であるゲルを自由に浮遊させるという試みは、はるかに悪い結果であった:神経細胞は典型的には、培地交換中又はゲル移動中に機械的に破損を被る。PDMSモールドは、洗浄剤及びUV滅菌で洗浄した後、再利用することができる。エタノールはPDMS中に内包され、培地中に溶出するため避けるべきである。ゲル作製の直前に、4アーム−PEG−VSの分割量をpH7.4のHEPESへ20%(w/v)となるように再懸濁した(ビニルスルホン40 mMとなる)。MMP切断可能ペプチドはpH6.0のPBS中に8%(w/v)となるように再懸濁した(エルマンテストで一般的にチオールが50 mMとなる)。ペプチド溶液のpHがわずかに酸性であるのは、チオールの酸化を避けるのに重要であり、これはpH7.4で数時間で顕著になる。ヒアルロナン(HA、シグマ53747)を0.75%(w/v)添加したろ過滅菌済みの完全培地の分割量を4℃で保存した。HAを0.5%(w/v)より多く含む溶液は粘度が高いため、正確に溶液をピペットするために、Gilson Microman M50Eのようなポジティブディスプレイスメントピペットが必要である。PEG 2%(w/v)、HA 0.525%(w/v)及び1e7 cells/mlを含む典型的なゲル100 μlを調製するために、ストック溶液を以下の様に組み合わせた:4アーム−PEG−VSのストック溶液 10 μl、pH7.4のHEPES緩衝液 12 μl、HAを添加した培地中に細胞が1.43e7 cells/mlの細胞懸濁液 70 μl、及び最後にMMP切断可能ペプチドのストック 8μlをチューブの側面に水滴として置いた。素早くボルテックス後、ポジティブディスプレイメントピペットで〜10秒激しく混合し、液体前駆体をPDMSモールド当たり15μl分注した。ゲル化はこの濃度で3〜4分以内で起こるが、柔らかいゲルについては15分までかかるので全てのゲルを細胞培養インキュベーター内(37℃、5%CO2)で1時間進行させてから1mlの増殖培地を添加した。ゲル化混合物は、生理学的なpH及び浸透圧だけでなく、完全培地が大半である:結果として、細胞が受けるストレスは最小限であり、1時間でゲル化を停止した時には、神経突起伸長が既に位相差で観察される。強く緩衝されたpH7.4のHEPESの〜20%の溶液によってpHを安定に保つ。各実験において示されているように、4アーム−PEG−VS及びMMPペプチドストックを対応する混合物へ添加する量を変更し、緩衝液をpH8.0のTEOAに置換え、HA及びストック内の細胞濃度を変更し、培地を各細胞型に合わせたものに変更し、又はHAを別の高粘度の添加物で置換えることによって、他の条件は得られる。同一の緩衝液及びチオール濃度において、MMP切断可能ペプチドをPEG−ジチオール(Laysan Bio, 3.4 kDa)によって置換することによって、非分解性ゲルが形成された。例示した場合では、CSRARKQAASIKVAVSADR−NH2の配列を有するラミニンα1(2110−2127)ペプチド(単にIKVAVと示す、ANAWA)を添加した。それを実施するために、IKVAVペプチドは、最初に4 mg/mlストックから4アーム−PEG−VS溶液に添加し、結合するために30分維持し、残りのプロトコルを実施した(この場合、終容量が同じとなるように、添加するHEPES緩衝液の量を減少させた)。クマリン標識IKVAVペプチドを用いてFRAP試験を用いて、ゲル中へのペプチドの固定が成功していることを確認した。IKVAV生物活性は、公表されたプロトコル(タシロ,K.,J. Biol.Chem.264、16174−82,1989)に従って2Dでアッセイした。
動的せん断測定は、Anton Paar MCR 301レオメータで行った。ゲルの液体前駆体は前述のように調製し、完全増殖培地は有しているが細胞は有しておらず、それを金属床プレートと直径20 mmの平行なプレートからなる形状に挿入した。貯蔵弾性率G’及び損失弾性率G’’の増加を水飽和雰囲気下、ペルチェ制御により37℃の温度、0.25 mmギャップ及び5%ひずみで、1Hz振動で観測した。このひずみは、ゲルの線形粘弾性領域内に良好に入るように選択された(1Hzで少なくとも1〜400%ひずみが及ぶ)。剛性は、60分での貯蔵弾性率(全てのサンプルがプラトーに達し、細胞培養培地の添加によりゲル化が停止したポイント)として定義され、及びゲル化時間は、補足図1において典型的な実験曲線で示すように、Logスケールでプラトーな剛性の半分に達する時間として定義した。増殖培地中の0.5%(w/v)HA及び2%(w/v)MAの複合粘度は、水飽和雰囲気下、37℃、0.2 mmギャップ及び2%ひずみ、及び0.01〜10 Hzの間で測定した。
20 kDaの4アームPEG−チオール 100 mg(5 μmol)を2 mlのmQ−水に溶解した。次いでテトラメチルローダミン−5−マレイミド(シグマ 94506) 0.12 mg(0.25 μmol)のpH7.4PBS溶液 2 mLを撹拌しながら滴下した。結合は数秒内に起こるが、さらに10分間進行させた。得られた混合物を、300 mM、pH8.0のTEOA緩衝液4 ml中に220 μl(2.2 mmol)のジビニルスルホンを溶解させた溶液に滴下し、30分間反応させ、透析し、凍結乾燥した。全ての操作は暗闇中で行われた。
ゲルはPEG 1.9%(w/v)、細胞なし、及びTMR標識4アーム−PEG−VSを用いて上記のとおり作製した。異なる細孔サイズを作製するために、培地(混合物が70%)にHAを0又は0.5%又は2%添加した。各ゲルの微細構造は、pH7.4のPBS中で3日間培養後、63×/1.4油浸対物レンズを有するライカSP8共焦点顕微鏡上で及び520 nm励起で、50×50×200 nm3ピクセルサイズの25 μmの積層立方体として画像化された。検出のために、非常に低ノイズで光子計測を行い、そしてそれゆえに「純粋」な黒レベルを提供する利点があるライカHyDsを使用した。漂白補正、2pxガウス平滑化、閾値処理、Matlab等値面による表面三角測量、及びパッチ機能削減及びブレンダーレンダリングをした細孔の結合性の3Dビューが得られた。細孔サイズの定量化について、条件あたりの3つの積層それぞれを、漂白補正(フィジー、単純比、バックグラウンドレベル0)及び1pxガウス平滑化でフィルタリングした。次いで積層は、閾値処理し、反転処理して、1の値をつけた細孔と0とした蛍光PEGとの2進画像を得、そして2次相関関数S2(x,y)をカスタムMatlabスクリプトで計算した。等方性が確認されたので、rについて50 nm binを使用してデータを
E17ウィスターラットの胚由来の皮質を以前の記述のとおり解剖し、及び解離した(ブリュリ, T., Nat. Protoc. 2, 3090−101, 2007)。簡単に述べると、細かく刻んだ皮質を、1 mg/mlBSA、10 mMグルコース、0.5 μg/ml DNAse(シグマ D−5020)及び0.5 mg/mlパパイン(シグマ P−4762)を添加したPBS中で、37℃で15分間インキュベートし、DMEMと10%FBSからなるブロッキング培地で洗浄した。次いで、それらを2 mlのブロッキング培地に再懸濁し、火仕上げしたパスツールピペットを用いた倍散によって解離させた。解離した細胞を、1×グルタマックス及びペニシリン/ストレプトマイシンを添加したNeurobasal+B27(ギブコ21103−049及び17504−044)からなる無血清増殖培地に再懸濁し、そしてPLLで被覆したフラスコ(50 μg/ml PLLホウ酸溶液pH8.4を37℃で1時間インキュベートしたもの)上で一晩平面培養した。翌日、まず平面培養した神経細胞をTrypLE expressで5分間洗浄し、弱く接着している非生存細胞及び細胞の残骸を剥離した。その後、残りの健常細胞をトリプシン/EDTA(ギブコ12605−010及び25200−072)で剥離させた。2倍容量のブロッキング培地を添加後、細胞を遠心分離し、無血清増殖培地へ再懸濁した。この後の2D又は3Dにおける細胞培養は全て、無血清増殖培地で行い、週に1回その半分を交換した。
DRGは封入直前に、E10ニワトリ胚から単離した。それらを、1 mg/mlアルブミン及び50 ng/ml NGFを添加した無血清増殖培地で培養した。封入は、ゲル形成セクションに記載されているとおり、4アーム−PEG−VS 0.8%(w/v)を、HA 0.525%(w/v)及びデキストラン1.4%(w/v)存在下で、化学量論量のPEGチオールで架橋した。
hiPSC由来神経細胞前駆細胞(Axol ax0016)を増殖させ、オンライン供給者プロトコルに従って分化させた。次いで、得られた大脳皮質神経細胞を、初代神経細胞と同様のプロトコルで、微細構造化のためのHA0.525%(w/v)存在下で、増殖培地をAxol維持培地(また無血清)と置換えて、ゲル内へ封入した。
カルシウム画像化のためにゲルの培地を除去し、1 mMのオレゴングリーンBAPTA−1(ライフテクノロジーズ)乾燥DMSO溶液を6 μMとなるように添加した新鮮な完全培地と交換した。1時間インキュベーション後、ゲルを元の培地で1時間洗浄し、8kHzの共鳴スキャナー及び488 nmの照射を伴い、20×/0.95水浸対物レンズを用いてライカSP8共焦点顕微鏡で画像化した。得られた画像率は7.44Hzであった。
生存率アッセイは、ゲルの培地に、カルセインAM(緑色の細胞質、生細胞、4 mMのストックDMSO溶液から2μM)、ヘキストの核染色(青色の核、全ての細胞、10 mg/mlのストックmQ−水溶液から10 μg/ml)、エチジウムホモダイマー−1(赤色の核、死細胞、2mMストックDMSO/水1:4溶液から2 μM)、及びヨウ化プロピジウム(赤色の核、死細胞、0.33mg/mlのmQ−水溶液から6.6 μg/ml)を添加することにより行った。ゲルを、添加した培地で1時間インキュベートし、次いで、1時間新鮮な培地で洗浄し、そしてライカSP8多光子顕微鏡で、解離した細胞については200 μM深さに対する20×/0.95水浸対物レンズで、又はDRG外植片については、単一スライスとして10×/0.3空気対物レンズで、画像化した。生細胞及び死細胞を手動で数えた。
Zeiss Observer 2顕微鏡上で5×空気対物レンズで明視野において、全ゲルをカバーするタイルスキャンを行うことにより、直径4 mmのゲル層上において2日後に(培養1ヶ月以内に溶解する条件では、既にこの時点で可視的に分解された)ゲル安定性を定量した。タイルはMicrosoft Image Composite Editorで組み立てた。分解した領域は、それらは懸濁した細胞体がなく、分解した部分の側面や底に凝集した暗い細胞凝集体で輪郭が縁取られているため、分解した領域ははっきりと目に見える。それらは、Fijiを用いて手動でセグメント化し、及び分解のパーセンテージを、ゲル全体に対する分解領域として報告した(6サンプルからの平均及び標準偏差)。
粘性成分として、10%(w/v)の高分子量PEG(シグマ 200 kPa)を用いて、神経細胞を封入する際に、混合直後に細胞クラスター化が観測された。画像化は20倍空気対物レンズでPlasDICを用いて画像化を行った。最良の可視条件においてPEGの取り込みを研究するために、神経細胞はPLLを被覆した組織培養プラスチック上で6日間、2Dで培養し、組織剥離から完全に回復させた。次いでそれらの培地に1%(w/v)TMR標識4アーム−PEG−VSを添加し、封入中のPEGへの暴露を模倣した。培養物を新しい増殖培地で10分間5回注意深く洗浄し、そして直ちにin PlasDICにおいて、及び20×空気対物レンズにおける蛍光を画像化した。さらに培養12日後に同一の観測を行いPEGの保持を観測した。蛍光チャネルは、バックグランドの差分及び3px中央値フィルタリングで光の不均一性及びノイズについて補正した。
神経突起の長さを、D2では、最大の正確性と信頼性をあたえることから手動追跡で定量化したが、D5では、この時点での長さが手動追跡では困難になるため、自動追跡で定量した。自動追跡から得られる全てのイメージは、適切な追跡について視覚的にチェックした。手動追跡は単純な神経突起トレーサー(ロンゲア, M. H., Bioinformatics 27, 2453−2454, 2011)を用いて行い、及びNeuriteQuantを用いた自動追跡(ディーメルト, L., BMC Neurosci. 12, 100, 2011)を行った。追跡は200 μmの深さ及び620 μmの幅の2光子画像化3D積層で行った。蛍光染色はデフォルトでβIII−チューブリン(固定した試料の全ての神経突起を染色)であり、及び、免疫染色に耐えるには弱すぎるものである部分的に分解したゲルを含む一連についてはカルセインAMを用いた(図4及び6に記載のHA応答曲線)。DAPIで染色された核の数を計測し、細胞数の標準化に用いた。
1次抗体は、βIII−チューブリン(シグマ T5076、1:500)、ニューロフィラメント(Neurofilament)(シグマ N4142、1:150)、シナプトタグミン(UCSF からライハルト教授によって開発されたDSHB mAB 30、8 μg/ml)、MAP−2(シグマ M3696、1:200)、GFAP(R&D システムズ AF2594、10 μg/ml)に対する抗体を用いた。Alexa488及び594を結合した2次抗体(インビトロジェン A11005、A11008、A10680、A11015)を1:200で使用した。本願発明者らは標準的な免疫細胞化学のプロトコルを適用して、ゲル全体を直接染色し、画像化した。試料を、0.1%トリトンX−100を含む10%ホルマリン中、4℃で1時間、固定及び浸透処理し、5%BSAのPBS溶液で一晩ブロッキングし、PBSで洗浄した(1時間を2回、一晩を1回)。次いで、それらを1次抗体でインキュベートし(3%BSAのPBS溶液で希釈し、室温で一晩穏やかに振盪)、PBSで洗浄し、2次抗体中で一晩インキュベートし、PBSで洗浄し、0.3 μM DAPIのPBS溶液で1時間染色し、最後にPBSで洗浄した。免疫染色した試料の画像化は、620μm幅及び200μm深さにわたり20×/0.95水浸対物レンズを備えたLeica SP8多光子顕微鏡上で、1 μmステップ、600Hz走査速度、1024×1024px、及びMaiTai Deepsee及びInsight Deepseeレーザーをそれぞれ使用した710 nm及び1100 nmの同時励起(Spectra Physics)で行った。2〜3色を同時に収集した。結果として得られた積層に最大強度投影(MIP)を適用し、フルオレセイン参照画像によって割ることにより光の不均一性を補正し、それらの色の表示を調整した(輝度/コントラスト/ガンマ)。
一元配置分散分析(one−way ANOVA)及びテューキーポストホックテスト(Tukey’s post hoc test)を用いて系列内の差異を定量化し、p<0.01を有意であるとみなした。
Claims (24)
- マクロ多孔質ハイドロゲルを提供するための方法であって、以下の工程、
a)第1及び第2の溶質を含む水溶液を提供する工程であって、ここで
i)該第1の溶質が、ポリエチレングリコール(PEG)の架橋可能な誘導体及びポリオキサゾリン(POx)の架橋可能な誘導体から選択される架橋可能なポリマーであり;及び
ii)該第2の溶質がコスモトロピック試薬であり;及び
b)前記架橋可能なポリマーを架橋することができる架橋試薬を前記混合物へ添加し、それにより前記架橋可能なポリマーのゲル化及び相分離を同時に開始する工程
を含む、マクロ多孔質ハイドロゲルを提供するための方法。 - 前記架橋可能なポリマーが、
