Abstract
Description
本出願は、2015年7月25日に出願された「Reticulation and Surface Modification of Polymer Foams Using Plasma」と題する米国仮特許出願第62/196,966号に対する優先権を主張し、その内容は参照により本明細書に組み込まれる。
プラズマガスの組成
表I.プラズマガスの組成および流量
シリコンウェハ(University Wafer,Inc.)を約1.25cm×1.7cmの長方形に切断し、水およびイソプロパノール中で各々を15分間超音波洗浄し、1Torrで一晩乾燥させ、乾燥下で保存した。
プラズマ膜は、Aurora 0350 Plasma Surface Treatment System(Plasma Technology Systems)を用いて堆積した。反応チャンバは、反応チャンバの底部から第3のラックスペースに据え付けられた単一のガラス棚を保持した。プラズマフィールドの空間的反応性をマッピングするために、ガラス棚上の6×6グリッドに配置された4.5cmの高さのガラスキュベットに、シリコンウェハを固定した。他のすべての実験試料は、棚の後部3分の1を中心として4.5cmの高さのガラスキュベットの頂部に固定した。チャンバを10mTorrのベース圧力まで排気し、続いて、プラズマ点火に先立って、指定されたプロセス流量でチャンバにプロセスガスを1分間導入した。連続した13.56MHzのRF電源を用いてプラズマフィールドを発生させ、プラズマ処理後にプロセスガスを、チャンバにわたり1分間流し続けた。次いで、チャンバを大気で10分間パージし、大気圧に戻した。すべての試料をポリプロピレン製バッグまたはトレイに乾燥下で保存した。
エリプソメトリーでの厚さは、380〜890nmおよび入射角70°で動作するAlpha−SE Spectroscopic Ellipsometer(J.A.Woollam Co.)を使用して測定した。データ分析は、シリコン基板上の薄い透明な膜を仮定してJ.A.Woollam光学モデルを採用した。
水の静的接触角の測定は、CAM 200 Goniometer(KSV Instruments)により、5マイクロリットルの液滴の体積を用いて行った。ヤング/ラプラスフィッティングモデルを用いて、報告された各プロセスについて6つの接触角を測定した。
XPS分析のため、シリコンウェハをLPおよびLPAプロセスで処理した。プラズマ処理時間は、各プロセスに対して30nmの規準化した膜厚を堆積するように選択された。MgのX線源およびCN10除電器を含むOmicron ESCA+システムを用いてXPSスペクトルを取得した。XPS動力、ステップ、エネルギーを渡す。原子比は、ソフトウェアCasa XPSを用いて計算した。
ポリマーのガラス転移温度(Tg)は、Q200 DSC(TA Instruments)を用いて測定した。乾燥Tgについては、1〜3mgの試料を、孔を設けたアルミニウム鍋に室温で装填し、−40℃に冷却し、−40℃〜120℃の加熱/冷却/加熱サイクルを、温度速度10℃/minで実行した。第2の加熱サイクルでの転移変曲点をTgの推定値として使用した。湿潤Tgの分析については、ワイヤ上発泡体試料を52℃の水浴に5分間浸漬し、その後、#3925 Hydraulic press(Carver,Inc)を使用して、1メートルトンで30秒間、実験室グレードのキムワイプシート(Kimberly−Clark Inc.)の間で圧縮した。次いで、発泡体を排気したアルミニウム鍋に入れ、−40℃に冷却し、10℃/minで80℃に加熱した。湿潤Tgは加熱サイクルの転移変曲点として推定した。それぞれの報告されたプラズマ組成物の5つの試料について、湿潤および乾燥Tgを測定した。
立体顕微鏡(Leica KL 2500 LCD)を用いて、圧縮したワイヤ上発泡体の直径を測定した。ワイヤ上発泡体試料は、カスタムのアルミニウム固定具を使用して、12mmの幾何学的基準と同じ平面に保持した。固定具を37℃の水浴に浸し、上から30秒間隔で30分間撮像した。ImageJを使用して、取り込んだ画像内の各発泡体試料の長さに沿って、5つの直径の測定値を取得した。各プラズマ組成物の3つの試料を測定した。
プラズマフィールド内の試料の位置、RFパワー、ガスの組成、圧力、および処理時間を含むプロセスパラメータを、膜堆積速度および疎水性への影響を判定するために系統的に評価した。次いで、選択されたプラズマプロセスを、臨床的に関連のある塞栓装置に適用して、水分可塑化およびその後のSMP発泡体の作動速度に対する影響を判定した。
図1に見られるように、プラズマ堆積速度は、棚の周辺部の周りにある反応性の低い縁を除き、プラズマチャンバの背面に向かって増加する。この反応性の勾配は、チャンバを通るプロセスガスの流れと平行になっている。したがって、ガス類がRFフィールドにより長い時間曝されると、堆積特性が高まり、その反応性が増加する。チャンバ内に試料を配置したことに起因する変動を軽減するために、その後のすべての試料を図1に示す100〜120nmの堆積領域内に固定した。
各プラズマプロセスの堆積速度を判定するために、膜堆積特性を測定した。図2(A)および図2(B)のグラフは、高流量エチレン(HE)および高流量エチレン/アセチレン(HEA)プロセスの典型的な堆積特性を示す。すべてのプロセスは、線形時間依存性堆積特性を示す。全32の線形回帰の平均R2値は0.992である。堆積速度は、動力が増加すると、また同様のプロセス状態でアセチレン(すなわち、HEA)を導入すると、増加した。
プロセスガスおよび動力の各組み合わせに対する水の静的接触角を図4(A)および(B)にまとめている。接触角は、プロセスの動力を増加させると、またアセチレンをプロセスに導入すると、減少した。プロピレンプロセスは一貫して、エチレンプロセスよりも疎水性があり、それはおそらく膜内に組み込まれたメチル基からであった。概して、膜の堆積速度を増加させるプロセスパラメータは、表面の疎水性を低下させる傾向があった。すべてのプラズマ堆積膜は、37±1℃の平均接触角を有する未処理のシリコンウェハよりも有意に疎水性であった。
様々なプロセスの動力、ガスの組成、圧力、および表面の疎水性の範囲を包含するために、4つのプラズマプロセスを選択した。これらのプロセスは、20nmの規準化された厚さで100のHDI発泡体に適用した。これらの発泡体の乾燥および湿潤飽和でのガラス転移温度を表IIにまとめている。

図5(A)および(B)は、未処理の発泡体と比較した場合における2つの異なるプラズマ処理のインビトロでの発泡体の膨張をまとめたものである。作動時間は点線の標準偏差を表す切片であると仮定され、それは神経血管コイルを送達するために使用される従来の0.021インチのマイクロカテーテルの内腔を表す。
Singhalら(Controlling the Actuation Rate of Low−Density Shape−Memory Polymer Foams in Water;Macromol.Chem.Phys.2013,214,1204〜1214)によって記載されたプロトコルに従って、SMP発泡体を調製した。トリメチルヘキサメチレンジイソシアネート(TM)100モル%およびN,N,N’,N’−テトラキス(2−ヒドロキシプロピル)エチレンジアミン(HPED)に対するトリエタノールアミン(TEA)の33:67のヒドロキシル当量モル比のイソシアネート組成物の状態である。硬化した発泡体を抵抗性のワイヤカッターで50mm×50mm×25mmのブロックに切断し、洗浄して、残留界面活性剤および触媒を除去した。以前に報告された洗浄プロトコルは、逆浸透(RO)水中で、15分間の超音波処理間隔2回、RO水中の20vol%Contrad(登録商標)70溶液(Decon Laboratories、キングオブプラシャ、ペンシルベニア州)中で1回、RO水での15分間隔4回に関与した。洗浄した試料をアルミニウムトレイで凍結し、3日間凍結乾燥した。乾燥した発泡体をポリプロピレン製バッグに乾燥下で保存した。
プラズマ処理は、Aurora 0350 Plasma Surface Treatment System(Plasma Technology Systems)を用いて行った。ポリマー発泡体ブロックを、反応チャンバの後部3分の1を中心とする12.5cmの高さのアルミニウムのメッシュ固定具に固定した。処理中、ポリマー膜を4.5cmの高さのガラスキュベットに固定した。発泡体のドッグボーンを、処理中に2つの4.5cmの高さのガラスキュベットの橋渡しをするよう固定した。チャンバを10mTorrのベース圧力まで排気し、次いでO2およびCF4プロセスガスをそれぞれ200sccmおよび800sccmでチャンバに導入してチャンバを385mTorrの圧力にした。ブロックの発泡体試料は、部分網状化または完全網状化用に、300ワットでそれぞれ8分または15分間プラズマ処理した。発泡体のドッグボーンを300ワットで1分、2分および3分間プラズマ処理して網状化の程度を高め、3分間で完全網状化を達成した。他に記載がない限り、すべての膜試料を15分間のプロセスでプラズマ処理した。すべてのプラズマ処理した試料をポリエチレン製バッグに乾燥下で保存した。
抵抗性のワイヤカッターを用いて、発泡体試料を発泡軸に対して平行(軸方向)および直交(横断)させて1mmのスライスに切断した。各スライスを、カーボンブラックテープを備えたステージに据え付け、Cressington Sputter Coater(Ted Pella,Inc.、レディング、カリフォルニア州)を用いて20mAで90秒間スパッターコーティングした。次いで、試料を、Joel NeoScope JCM−5000走査型電子顕微鏡(SEM)(Nikon Instruments、Inc.、メルビル、ニューヨーク州)を用いて視覚化した。
プラズマ網状化発泡体および未処理発泡体の多孔質媒体特性を以前に報告されたような透過率システムを用いて測定し、Forchheimer−Hazen−Dupuit−Darcy(FHDD)の式(1)を用いて計算した:

式中、δP/δxは、流れる方向における試料じゅうの圧力勾配(Pa/m)であり、μは作動流体の動的粘性(Pa/s)であり、Kは試料の透過率(m2)であり、V_0はダルシー流速(流量を試料の断面積で除算したもの)(m/s)であり、pは流体の密度(kg/m3)であり、Cは試料の発泡体ファクタ(m-1)である。透過率、K、および発泡体ファクタCは、発泡体の幾何学的パラメータである。
一軸引張試験は、50mm/minの一定のひずみ速度でInsight 30 Material Tester(MTS Systems Corporation、エデン・プレイリー、ミネソタ州)を用いて室温で行った。それぞれの試料の応力−ひずみ曲線から、極限引張強度(kPa)、破断時のひずみ(%)および弾性率(kPa)を算出した。
生検用パンチとレザーブレードを用いて、網状化の状態ごとに、6mm×5mm(OD×長さ)の3つの円筒形試料を調製した。発泡体試料が直径200ミクロンのニチノールワイヤ(NDC、フリーモント、カリフォルニア州)上に軸方向に通され、100℃に予熱したSC150ステントクリンパー(Machine Solutions、フラッグスタッフ、アリゾナ州)に充填した。試料を径方向に圧縮する前に15分間平衡化した。試料は、冷却して室温に戻すまで、動かないようにした。クリンプしたワイヤ上発泡体試料を、膨張試験の前に、最低24時間乾燥下で保存した。試料を、12mmの測定基準と同じ撮像面に試料を保持するカスタムのアルミニウム固定具内に据え付けた。固定具を37℃の水浴に浸し、1分間間隔で15分間撮像した。ImageJ分析ソフトウェア(National Institute of Health、メリーランド州、米国)を用いて、各時点で各試料の長さに沿って5つの直径測定を行った。
CAM 200ゴニオメーター(KSV Instruments)および、5μlの液滴の体積を用いて、未処理の膜およびプラズマ処理膜(300ワットで15分間)で、水の静的接触角の測定を行った。網状化後0日、7日、14日、21日および28日目に接触角を測定して、試料の疎水性の緩和を調べた。ヤング/ラプラスフィッティングモデルを用いて4つの別個の膜で3つの接触角を測定し、各プロセスおよび時点について計12の測定値を求めた。
ウシの血液を、動物の屠殺直後に屠殺場で採取した。凝固を防止するために、血液と、リン酸緩衝生理食塩水(PBS、pH7.4)中の3.2%クエン酸ナトリウム溶液との体積比が9:1の状態で、1Lのガラスジャーにおいて、血液にクエン酸塩加した。すべての血液調査は、動物の屠殺の8時間以内に完了した。
細胞と材料の相互作用を、プラズマ処理(処理後0日と28日目)および未処理の膜で評価した。緑色蛍光タンパク質(GFP)発現3T3線維芽細胞(NIH3T3/GFP、Cell Biolabs Inc.、米国)を200cell/mm2で表面に播種する前に、膜(処理状態ごとにn=3)をUV光下で3時間滅菌した。設定した時点(3時間、1日、3日および7日目)で、蛍光顕微鏡(励起:488nm、Nikon FN1 Upright Microscope、NY、米国)を用いて細胞を撮像した。画像当たり5つの代表的な細胞を測定して、各々の膜の状態について、3つの画像にわたり、ImageJソフトウェアを使用して、細胞の面積を定量した。手動の検証によるImageJの粒子分析を用いて、各々の膜の状態について、3つの画像にわたって細胞の増殖を定量化した。
発泡体の形態が流体透過率へ与える影響
発泡体の形態におけるプラズマ誘起の変化は、SEMの撮像により明瞭に視覚化されている(図6A)。未処理の発泡体は、依然として大部分変化していない孔間膜を有する。部分網状化発泡体は、膜の大部分のピンホールに関して中程度の膜の分解があり、血液−材料の相互作用用の膜の大きな表面積を維持しながら、ある程度の孔の相互連結性をもたらす。発泡体ストラットは完全なままであるが、わずかな割合の膜しか完全網状化発泡体には残らない。完全網状化は、流体の透過率および細胞の浸潤を改善するための多孔質マトリックスを提供する。
力学的完全性への影響

図7に見られるように、非網状化発泡体試料は37℃でゆっくりとした膨張速度を有し、浸漬の15分後にわずか2mmの膨張を示した。比較すると、部分網状化発泡体は、15分以内に直径が約5mmに拡張した。完全網状化試料は、わずか7分で完全な形状回復(直径6mm)を果たした。
接触角の測定が、プラズマ処理をした膜にて行われ、表面の親水性の増加が膨張速度の増加の要因であるという仮説を確認した(図8)。プラズマ処理の日、膜は、接触角96±2°の未処理の膜と比較して、接触角が45±3°のものの方が親水性であった。しかし、プラズマ処理された表面は、乾燥空気中で4週間保存した後、75±4°の増加した接触角で疎水性の回復を示した。
細胞が拡散する形態に基づいて、プラズマ処理された表面に対する細胞の親和性は、播種後わずか3時間で実証されている。インビトロで1週間後に見られるように、この最初の接着により、後の時点でプラズマ処理された基板にて細胞の拡散が有意に増すことが可能になる。図9Aに示すように、対照膜上の細胞は培養約1日目で細胞の面積の閾値に達するが、プラズマ処理された表面上の細胞は、高次に伸長した仮足を示し、1週間の培養にわたって細胞の拡散が一貫して増加している。類似した線維芽細胞の拡散挙動が、匹敵する範囲の表面接触角63〜68°の組織培養ポリスチレンについて報告されている。網状化後4週間後の細胞拡散の観察された減少は、表面の一時的な疎水性回復に起因する(図8)。
Claims (24)
- 第1および第2の状態を有するモノリシック形状記憶ポリマー(SMP)発泡体
を含むシステムであって、前記SMP発泡体は、(a)ポリウレタン、(b)内側ストラットによって画定された内側網状化セルを有する内側半分部分、および(c)前記SMP発泡体の断面を備える平面において前記内側部分を囲む、外側ストラットによって画定された外側網状化セルを有する外側半分部分を備えるシステム。 - 前記外側網状化セルが前記内側網状化セルより網状化しているような網状化勾配を含む、請求項1に記載のシステム。
- 前記SMP発泡体が、遠位部分および近位部分を含み、
前記遠位部分が、前記内側半分部分および前記外側半分部分を含み、
前記遠位部分および近位部分のうちの1つは、前記遠位部分および近位部分の別のものよりも大いに網状化したセルを含む、請求項1に記載のシステム。 - 前記SMP発泡体が、経時的に、疎水性の増加に向かい回復するように構成された表面エネルギーを有する、請求項1に記載のシステム。
- 前記内側および外側ストラットは両方がN,N,N’,N’−テトラキス(2−ヒドロキシプロピル)エチレンジアミン(HPED)、トリエタノールアミン(TEA)およびヘキサメチレンジイソシアネート(HDI)を含み、TEAはHPEDよりも、高いヒドロキシル基のモル比に寄与する、請求項1に記載のシステム。
- 前記SMP発泡体が0.06g/cm3未満の密度を有する、請求項5に記載のシステム。

前記炭化水素膜が前記内側および外側ストラットの両方を覆うシステム。 - 前記炭化水素膜が500nmほど厚くなく、前記炭化水素膜が疎水性である、請求項16に記載のシステム。
- プラズマチャンバ内に第1および第2の状態を有するモノリシック形状記憶ポリマー(SMP)発泡体を配置すること、
前記チャンバを第1の圧力レベル範囲まで排気すること、
前記第1の圧力レベル範囲にある間、第1のガス混合物を前記チャンバに供給すること、
前記第1のガス混合物を第1のプラズマにイオン化すること、
第1の期間にわたって前記SMP発泡体を前記第1のプラズマに曝して前記SMP発泡体のセルを網状化し、前記網状化したセルの内側および外側ストラットの表面を酸化させること、および
前記SMP発泡体を大気圧に曝すこと
を含む方法であって、
前記SMP発泡体が、(a)ポリウレタンと、(b)前記内側ストラットを含む内側半分部分と、(c)前記SMP発泡体の断面を備える平面において前記内側部分を囲む、前記外側ストラットを含む外側半分部分とを含む方法。 - 前記SMP発泡体を前記第1のプラズマに曝した後に、続いて、
前記SMP発泡体を含む前記チャンバおよび別のチャンバの少なくとも1つを第2の圧力レベル範囲まで排気すること、
前記第2の圧力レベル範囲にある間に第2のガス混合物を前記チャンバおよび別のチャンバの前記少なくとも1つに供給すること、
前記第2のガス混合物を第2のプラズマにイオン化すること、および
第2の期間にわたって前記SMP発泡体を前記第2のプラズマに曝して、前記内側および外側ストラットの両方の前記酸化した表面上に炭化水素膜を形成すること
を含む方法であって、
前記炭化水素膜が500nmほど厚くない、請求項18に記載の方法。 - 前記第1のガス混合物が酸素およびテトラフルオロメタンを含み、前記第2のガス混合物がアセチレン、エチレン、プロピレン、メタン、イソブチレン、エタン、プロパン、ブタンおよびアルゴンからなる群から選択される少なくとも1つのメンバーを含む、請求項19に記載の方法。

- 前記第1の期間、前記SMP発泡体を前記第1のプラズマに曝す前に前記SMP発泡体の一部をマスキングすることを含む、請求項18に記載の方法。
- 前記内側および外側ストラットは両方がN,N,N’,N’−テトラキス(2−ヒドロキシプロピル)エチレンジアミン(HPED)、トリエタノールアミン(TEA)およびヘキサメチレンジイソシアネート(HDI)を含み、TEAはHPEDよりも、高いヒドロキシル基のモル比に寄与する、請求項8に記載のシステム。
