ペプチドは、当該分野において周知のいずれの方法でも合成し得る。タンパク質の化学的な合成に適した方法には、例えば、Solid Phase Peptide Synthesis,Second Edition,Pierce Chemical Company(1984)及びMethods Enzymol.289,Academic Press,Inc,New York(1997)にStuart及びYoungが記載したものが含まれる。
本明細書で使用の芳香族カチオン性ペプチドは、疾患の予防又は処置に有用である。具体的には、本開示では、本明細書に記載の芳香族カチオン性ペプチドを投与することによる、疾患のリスクにさらされている(すなわち罹患しやすい)被験体に処置をする予防的方法及び治療的方法の両方を提供する。したがって、この方法では、有効量の芳香族カチオン性ペプチドをそれを必要とする被験体に投与することにより被験体において疾患を予防及び/又は処置する。
上記のペプチドは、それを必要とする哺乳動物において酸化的損傷を減少させるのに有用である。酸化的損傷を減少させる必要がある哺乳動物は、酸化的損傷に関係した疾患、状態又は処置を被っている哺乳動物である。典型的には、酸化的損傷はフリーラジカル、例えば活性酸素種(ROS)及び/又は活性窒素種(RNS)によって引き起こされる。ROS及びRNSの例には、ヒドロキシルラジカル、スーパーオキシドアニオンラジカル、一酸化窒素、水素、次亜塩素酸(HOCl)及びペルオキシニトライトアニオンが含まれる。有効量の上記の芳香族カチオン性ペプチドの投与後に哺乳動物、取り出された臓器又は細胞における酸化的損傷の量が低下したならば、酸化的損傷は「減少した」とみなされる。典型的には、酸化的損傷がペプチドで処置していないコントロールの被験体と比較して少なくとも約10%、少なくとも約25%、少なくとも約50%、少なくとも約75%又は少なくとも約90%低下したら、酸化的損傷が減少したとみなされる。
上記のペプチドは、ミトコンドリア膜透過性遷移(MPT)に関係した疾患又は状態の処置において有用である。このような疾患又は状態には、以下に限定するものではないが、組織又は臓器の虚血及び/又は再灌流、低酸素症及び多数の神経変性疾患が含まれる。MPTの阻害又は防止が必要な哺乳動物は、これらの疾患又は状態を患っている哺乳動物である。
様々な実施形態において、適切なインビトロ又はインビボのアッセイを行うことによって特定の芳香族カチオン性ペプチド系治療薬の効果及びその投与が処置に適応があるか否かを判定する。様々な実施形態において、インビトロのアッセイを代表的な動物モデルで実施して、所定の芳香族カチオン性ペプチド系治療薬が疾患の予防又は処置において所望の効果を発揮するか否かを判定することができる。治療に使用する化合物は、ヒト被験体での試験に先立って適切な動物モデル系で試験することができ、この適切な動物モデル系には、以下に限定するものではないが、ラット、マウス、ニワトリ、ブタ、ウシ、サル、ウサギ等が含まれる。同様に、インビボの試験でも、当該分野で公知の動物モデル系のいずれをも、ヒト被験体への投与に先立って使用することができる。
一態様において、本発明は、被験体において、被験体に、状態の惹起又は進行を防止する芳香族カチオン性ペプチドを投与することでその疾患を予防する方法を提供する。予防を目的とした用途においては、芳香族カチオン性ペプチドの医薬組成物又は薬物を、ある疾患又は状態を発症しやすい又はそのリスクがある被験体に、そのリスクを取り除く若しくは低下させる、重症度を低下させる又はその疾患の発症を遅らせるのに十分な量で投与する(疾患の生化学的、組織学的及び/又は行動上の症状、疾患の発症中に認められる合併症及び中間的な病理学的表現型を含む)。予防用の芳香族カチオン性ペプチドを、異常の特徴である症状が顕在化する前に投与することができ、これによって疾患又は障害が予防される、あるいはその進行が遅延する。適切な化合物は、上記のスクリーニングアッセイに基づいて決定することができる。
本テクノロジーの別の態様には、治療を目的として、被験体において疾患を処置する方法が含まれる。治療を目的とした用途においては、組成物又は薬物を、このような疾患を疑われる又は既に患っている被験体に、その疾患の症状を治癒する又は少なくとも部分的にその進行を阻止するのに十分な量で投与する(疾患の発症中に認められる合併症及び中間的な病理学的表現型を含む)。
細胞、臓器又は組織をペプチドと接触させるための当該分野で公知のいずれの方法も利用し得る。適切な方法には、インビトロ、エクスビボ又はインビボの方法が含まれる。インビボの方法には、典型的には、芳香族カチオン性ペプチド(上記のもの等)の哺乳動物、適切にはヒトへの投与が含まれる。治療を目的としてインビボで使用する場合は、芳香族カチオン性ペプチドを、被験体に有効量(すなわち、所望の治療効果を有する量)で投与する。用量及び投与レジメンは、被験体の損傷の程度、使用する特定の芳香族カチオン性ペプチドの特徴(例えば、その治療係数)、被験体及び被験体の既往歴に左右される。
ミトコンドリアによるATP合成は、ミトコンドリア内膜(IMM)の電子伝達系(ETC)を介した電子流によって推進される。この電子伝達系を流れる電子流は、一連の酸化/還元過程として表すことができる。電子は、電子供与体(NADH又はQH2)から一連の電子受容体(複合体I−IV)を経て最終的には末端電子受容体である分子酸素に受け渡される。IMMと緩く会合しているチトクロムc(cytochrome c)は、複合体IIIとIVとの間で電子を伝達する。
実施例で示すように、試料において、アミノ酸配列:Tyr−D−Arg−Phe−Lys−NH2(SS−01)、2’,6’−Dmt−D−Arg−Phe−Lys−NH2(SS−02)、Phe−D−Arg−Phe−Lys−NH2(SS−20)又はD−Arg−2’,6’−Dmt−Lys−Phe−NH2(SS−31)を含むペプチドを含めた本明細書で開示の芳香族カチオン性ペプチドの濃度を変化させることで、チトクロムcの電気及びフォトルミネッセンス特性が変化する。具体的には、チトクロムcに対する芳香族カチオン性ペプチド濃度を上昇させると、チトクロムcの導電率及びフォトルミネッセンス効率が上昇する。芳香族カチオン性ペプチドの適切な濃度範囲には、以下に限定するものではないが、0−500mM、0−100mM、0−500μm、0−250μm及び0−100μmが含まれる。
図8は、本明細書で開示のペプチド、例えばTyr−D−Arg−Phe−Lys−NH2(SS−01)、2’,6’−Dmt−D−Arg−Phe−Lys−NH2(SS−02)、Phe−D−Arg−Phe−Lys−NH2(SS−20)又はD−Arg−2’,6’−Dmt−Lys−Phe−NH2(SS−31)のいずれかをドープしたチトクロムcの層110の導電率(抵抗)における変化を測定することによって試験基板130のpH及び/又は温度における変化を検出するセンサ例100を示す。基板130の温度及び/又はpHが変化するにつれて、芳香族カチオン性ペプチドがペプチドドープチトクロムc層110内外に拡散し、これがペプチドドープチトクロムc層110の導電率の変化を引き起こす。計測器120は、アノード122及びカソード124を介してチトクロムc層110に電位(電圧)を印加することによって導電率における変動を測定する。導電率が上昇すると、アノード122とカソード124との間を流れる電流が増加する。導電率が低下すると、アノード122とカソード124との間を流れる電流が減少する。代替案のセンサは、より高感度で抵抗を測定するために追加の電気端子(すなわち、アノード及びカソード)を含み得る。例えば、代替案のセンサは、抵抗をケルビン検出により測定するために4個の電気端子を含み得る。
加えて、ペプチドドープチトクロムcセンサを、マイクロフルイディック及びオプトフルイディックデバイスで使用して例えば圧力、温度、pH、印加電場等における変動を、ハイブリッド生物学的/化学的/電子的プロセッサで使用するために、電流及び/又は電圧に変換することができる。また、米国特許出願公開第2009/0201497号、米国特許出願公開第2010/0060875号及び米国特許出願公開第2011/0039730号(各文献は参照により本明細書に全て組み込まれる)の明細書に記載されるもの等のマイクロフルイディック及びオプトフルイディックデバイスにおいて使用することができる。
芳香族カチオン性ペプチドをドープしたチトクロムcを電気又は光スイッチ、例えば図10に図示のスイッチ201として又はそれにおいて使用することもできる。このスイッチ201は、芳香族カチオン性ペプチド200、例えばTyr−D−Arg−Phe−Lys−NH2(SS−01)、2’,6’−Dmt−D−Arg−Phe−Lys−NH2(SS−02)、Phe−D−Arg−Phe−Lys−NH2(SS−20)又はD−Arg−2’,6’−Dmt−Lys−Phe−NH2(SS−31)の入った、チトクロムc又はペプチドドープチトクロムc110と導管221及びチャネル210を介して流体連通している貯留部220を含む。運転時、導管221を開放するとペプチド200が方向212に向かってチャネル210へと流れる。チャネル210とチトクロムc130との間の境界を隔てて温度及び/又はpH勾配を作り出すことによってスイッチ201を作動させる。勾配の方向に応じて、ペプチド200はチトクロムc130内外へと拡散し、これによって上述したように導電率及びフォトルミネッセンス特性が変化する。ペプチド濃度の変動による導電率の変化を用いて、アノード222とカソード224との間の電流を調節することができる。
チトクロムc及び/又は本明細書で開示するような芳香族カチオン性ペプチド、例えばTyr−D−Arg−Phe−Lys−NH2(SS−01)、2’,6’−Dmt−D−Arg−Phe−Lys−NH2(SS−02)、Phe−D−Arg−Phe−Lys−NH2(SS−20)又はD−Arg−2’,6’−Dmt−Lys−Phe−NH2(SS−31)をドープしたチトクロムcをベースとしたトランジスタは、デジタルコンピューティングで使用するための情報を保存するスタティック又はダイナミックランダムアクセスメモリ(RAM)等のメモリの実装に使用することもできる。当然のことながら、6個のトランジスタを連結して、定期的なリフレッシュを必要とすることなく1ビットの情報を保存するスタティックRAM(SRAM)セルを形成することができる。チトクロムc及び/又は芳香族カチオン性ペプチドをドープしたチトクロムcをベースとしたトランジスタは他のタイプのメモリの実装にも使用することができ、デジタルコンピューティング用のダイナミックランダムアクセスメモリ(DRAM)が含まれる。当然のことながら、RAMを使用して、上述したもの等の用途向けのデジタルコンピューティングを行うことができる。
チトクロムc及び/又は本明細書で開示の芳香族カチオン性ペプチド、例えばTyr−D−Arg−Phe−Lys−NH2(SS−01)、2’,6’−Dmt−D−Arg−Phe−Lys−NH2(SS−02)、Phe−D−Arg−Phe−Lys−NH2(SS−20)又はD−Arg−2’,6’−Dmt−Lys−Phe−NH2(SS−31)をドープしたチトクロムcを有機発光トランジスタ(OLET)の作製に使用することもでき、これはより安価なデジタルディスプレイ及びコンピュータチップ上の高速スイッチング光源につながり得る。OLETをベースとした光源は、ダイオードよりはるかに速く切り替えを行い、またその平面的な設計により、コンピュータチップ上により容易に集積させ得て、銅線より速くデータをチップで伝送することができる。より高い効率の鍵となるのは三層構造であり、薄膜が互いに積層されている。電流は上層及び下層を水平に流れ(一方は電子を輸送し、他方は正孔を輸送する)、中央の層に入り込むキャリアは再結合し、光子を放出する。チャネルにおける接合領域の位置はゲート及びドレインの電圧に左右されるため、発光領域を調節することができる。
チトクロムc及び/又は本明細書で開示されるような芳香族カチオン性ペプチド、例えばTyr−D−Arg−Phe−Lys−NH2(SS−01)、2’,6’−Dmt−D−Arg−Phe−Lys−NH2(SS−02)、Phe−D−Arg−Phe−Lys−NH2(SS−20)又はD−Arg−2’,6’−Dmt−Lys−Phe−NH2(SS−31)をドープしたチトクロムcを、有機発光ダイオード(OLED)及びエレクトロルミネッセントディスプレイで使用することができる。OLEDは多様な消費者製品、例えば時計、電話、ノートパソコン、ポケットベル、携帯電話、デジタルビデオカメラ、DVDプレーヤー及び電卓において有用である。OLEDを備えたディスプレイには、慣用の液晶ディスプレイ(LCD)に勝る数多くの利点がある。OLED系のディスプレイはバックライトを必要としないため濃黒色レベルを表示し、また視野角が広くても比較的高いコントラスト比を達成することができる。また、重量があって電力消費量が多いバックライトを必要とするLCDより薄く、より効率よく明るくなり得る。これらの特徴が組み合わさる結果、OLEDディスプレイは、LCDディスプレイより軽量且つコンパクトである。
チトクロムc活性層における芳香族カチオン性ペプチドの濃度レベルもまた、ヘテロ接合を得るための空間及び/又は時間の関数として変動し得て、ヘテロ接合とは、米国特許第7897429号明細書(参照により全て本明細書に組み込まれる)に記載されるような、異なるエネルギーギャップの2種の半導体材料間の界面であり、図18、19の光電池に図示される。芳香族カチオン性ペプチド濃度の適切な範囲には、以下に限定するものではないが、0−500mM、0−100mM、0−500μM、0−250μM及び0−100μMが含まれる。例えば、ヘテロ接合を用いて、OLED及び他のデバイスにおけるより強い発光のための多重量子井戸構造を作り出すことができる。p型及びn型の結晶性の薄膜の活性層で構成された有機ヘテロ接合トランジスタが高導電率であると発見されてから、有機ヘテロ接合はますます注目を浴びている。無機ヘテロ接合において形成される空乏層とは対照的に、電子及び正孔蓄積層は、有機ヘテロ接合界面の両側で観察することができる。高導電率のヘテロ接合膜を、電荷注入バッファ層及びタンデムダイオード用の接続ユニットとして使用することができる。両極性トランジスタ及び発光トランジスタ(上述のもの)は、有機ヘテロ接合膜を活性層として使用して実現することができる。
チトクロムc及び/又は芳香族カチオン性ペプチド、例えばTyr−D−Arg−Phe−Lys−NH2(SS−01)、2’,6’−Dmt−D−Arg−Phe−Lys−NH2(SS−02)、Phe−D−Arg−Phe−Lys−NH2(SS−20)又はD−Arg−2’,6’−Dmt−Lys−Phe−NH2(SS−31)等をドープしたチトクロムcを使用してバッテリの内部抵抗を低下させることもでき、これにより放電中、バッテリをほぼ定電圧に維持することが可能になる。当該分野では理解されるように、バッテリは、化学エネルギーを直接電気エネルギーに変換するデバイスである。これには多数のボルタ電池が含まれ、それぞれアニオン及びカチオンを含有する伝導性電解質によって直列に接続された2個の半電池を含む。一方の半電池は電解質及びアニオン(負に帯電したイオン)が移動する電極、すなわちアノード又は負電極を含み、もう一方の半電池は電解質及びカチオン(正に帯電したイオン)が移動する電極、すなわちカソード又は正電極を含む。バッテリに電力を供給する酸化還元反応において、カチオンはカソードで還元され(電子が付加される)、アニオンはアノードで酸化される(電子が除去される)。電極は互いには接触しないが、電解質によって電気的に接続されている。電解質が異なる2個の半電池を使用する電池もある。半電池を隔てるセパレータはイオンが流れるのは許すが、電解質が混ざるのは防止する。
チトクロムcの単一分子を分子バッテリとして使用することもでき、分子バッテリの充電及び/又は放電時間を1種以上の芳香族カチオン性ペプチド、例えばTyr−D−Arg−Phe−Lys−NH2(SS−01)、2’,6’−Dmt−D−Arg−Phe−Lys−NH2(SS−02)、Phe−D−Arg−Phe−Lys−NH2、(SS−20)又はD−Arg−2’,6’−Dmt−Lys−Phe−NH2(SS−31)で調節することができる。本明細書に記載するように、チトクロムcは、荷電酸素及び窒素原子とは膜の反対側に炭素及び硫黄を有する膜タンパク質である。荷電酸素及び窒素で覆われた領域は水系環境を好み、膜の両面から飛び出している。この配列はチトクロムcによって果たされる役割にとって完璧であり、酸素から水への反応を分子ポンプの動力源として使用する。酸素が消費されるにつれて、水素イオンを膜の片側からもう一方側に送り出すことでエネルギーが蓄積される。後に、このエネルギーを使用してATPを作り上げたり、あるいは水素イオンに膜を浸透させ戻すことでモータに動力を供給することができる。
有機光電池(OPV)は、価格設定における大きな崩壊、芸術的外観、また低光量条件下でのめざましい効率を約束する。OPV材料はまたフレキシブル且つ形によくフィットする。OPVは潜在的に様々な材料の周囲に巻き付けることができ、あるいはペイントすることさえできる。現在のOPV効率は5%−6.25%である。これらの効率は慣用の発電形態に置き換わるに十分なものではないものの、OPVは、半導体太陽電池の高コストを考えると、著しく高い効率を必要とはしない用途に適している。例えば、オフィス、自宅又は会議室という環境のような低光量条件下、連続的なトリクル充電設定で携帯電話を充電するのに光電池を使用することができる。
チトクロムc、芳香族カチオン性ペプチド又は芳香族カチオン性ペプチド、例えばTyr−D−Arg−Phe−Lys−NH2(SS−01)、2’,6’−Dmt−D−Arg−Phe−Lys−NH2(SS−02)、Phe−D−Arg−Phe−Lys−NH2(SS−20)若しくはD−Arg−2’,6’−Dmt−Lys−Phe−NH2(SS−31)をドープしたチトクロムcを含む光電池及び他の用途向けの有機膜は、スピンコーティング、気相堆積及び米国特許第6734038号、米国特許第7662427号及び米国特許第7799377号明細書に記載の方法で堆積させ得て、各文献は参照により全て本明細書に組み込まれる。スピンコーティング技法を用いて高速で広い表面積をコーティングすることができるが、ある層に溶媒を使用すると既に存在しているポリマー層が劣化する場合がある。スピンコーティングした材料を別のパターニングステップでパターニングしなくてはならない。
上述したように、例示的な芳香族カチオン性ペプチド、例えばTyr−D−Arg−Phe−Lys−NH2(SS−01)、2’,6’−Dmt−D−Arg−Phe−Lys−NH2(SS−02)、Phe−D−Arg−Phe−Lys−NH2(SS−20)又はD−Arg−2’,6’−Dmt−Lys−Phe−NH2(SS−31)を使用して導電率を上昇させることができる。その結果として、例示的な芳香族カチオン性ペプチドを使用して、(廃)熱エネルギー生産時の損失をより少なくして電流を伝導することができる。この効果を利用して、バッテリを動力とするデバイス(家庭用電化製品等)及びより大型の電力系統(電力伝送用途におけるもの等)において動作寿命を延ばすことができる。廃熱の発生を減少させることで冷却の必要性も低下し、更に効率が上昇し、また本発明の芳香族カチオン性ペプチドをドープした導電性材料(チトクロムc等)を動力源とする電子機器の寿命が延びる。
本明細書で開示の芳香族カチオン性ペプチドを使用してチトクロムcバイオセンサにおける電子流を強化し、またその感度レベルを上昇させ得る。実施例で示すように、本明細書で開示のペプチド、例えばD−Arg−2’,6’−Dmt−Lys−Phe−NH2はチトクロムcの還元を増進させ(図1)、またチトクロムcを流れる電子流を増加させる(図2)。
本明細書で開示の芳香族カチオン性ペプチドは、環境汚染物質の生物的環境浄化に有用である。特に、このペプチドは、生物的環境浄化反応の速度及び/又は効率を上昇させるのに有用であり、ここで細菌cチトクロムは電子の環境汚染物質への伝達を仲介し、これによってその物質の原子価を変化させ、その相対毒性を低下させる。本明細書で開示の方法において、芳香族カチオン性ペプチドは細菌cチトクロムと相互作用し、また電子伝達を促進する。一態様において、芳香族カチオン性ペプチドは細菌cチトクロムの還元を促進する。別の態様において、ペプチドは、細菌cチトクロムを通る電子拡散を強化する。別の態様において、ペプチドは、細菌cチトクロムにおいて電子容量を強化する。別の態様において、ペプチドは、細菌チトクロムのヘム基周囲で電子拡散に有利に働く新規なπ−π相互作用を引き起こす。最終的には、芳香族カチオン性ペプチドの細菌cチトクロムとの相互作用が、環境汚染物質の異化的還元を増進及び/又は強化する。
本明細書で開示の芳香族カチオン性ペプチド、チトクロムc及び/又はペプチドドープチトクロムcは、ナノワイヤ用途において有用である。典型的には、ナノワイヤは、直径がナノメートル(10-9メートル)オーダーのナノ構造体である。あるいは、ナノワイヤを、数十ナノメートル以下に制約された厚さ又は直径と制約のない長さを有する構造体と定義することができる。このスケールだと、量子力学的効果が働き始める。多くの異なるタイプのナノワイヤが存在し、金属(例えば、Ni、Pt、Au)、半導体(例えば、Si、InP、GaN等)及び絶縁(例えば、SiO2、TiO2)ナノワイヤが含まれる。分子ナノワイヤは、有機(例えば、DNA、本明細書で開示の芳香族カチオン性ペプチド、チトクロムc及び/又はペプチドドープチトクロムc等)又は無機(例えば、Mo6S9−xIx)の繰り返し分子単位から構成される。本明細書で開示のナノワイヤは、例えば部品を極めて小さな回路に連結するのに有用である。ナノテクノロジーを用いて、部品を化合物から作り出す。
ナノワイヤを合成するには2つの基本的アプローチ、すなわちトップダウン方式のアプローチとボトムアップ方式のアプローチがある。トップダウン方式のアプローチでは、大きな材料片を、リソグラフィ、電気泳動等の様々な手段で小さくカットする。これに対してボトムアップ方式のアプローチでは、ナノワイヤを、構成吸着分子の組み合わせにより合成する。合成技法の殆どはボトムアップ方式のアプローチをベースとしている。
チトクロムcの還元
増量しながら芳香族カチオン性ペプチドを酸化チトクロムcの溶液に添加した。還元チトクロムcの生成を、500nmでの吸光度でモニタした。チトクロムcの還元速度を、非線形解析(Prizmソフトウェア)により求めた。
新鮮なミトコンドリアを、以前に記載されたようにラットの腎臓から単離した。電子束を、Cl(グルタメート/マレート)、C2(スクシネート)及びC3(TMPD/アスコルベート)について異なる基質を使用し、以前に記載されたようにO2消費(Oxygraphクラーク電極)により測定した。アッセイを、酵素反応の飽和を回避するために、低基質条件下で行った。単離したミトコンドリアでのATP産生を、96ウェル発光プレートリーダー(Molecular Devices社)においてルシフェラーゼ法(Biotherma社)により動力学的に測定した。ATP合成の初期最高速度は、最初の1分で求められた。
サイクリックボルタンメトリを、Bioanalytical System社のCV−50Wボルタンメトリックアナライザで、電位+0.237V(対NHE)のAg/AgCl/1M KCl基準電極(Biometra社、ゲッチンゲン、ドイツ)及び白金対電極を使用して行った。金線電極を、確立されたプロトコルに従って清浄にした。溶液状のチトクロムcの電気化学的研究を、メルカプトプロパノール修飾電極(20mMのメルカプトプロパノール中で24時間にわたってインキュベートした)を使用して行った。サイクリックボルタモグラム(1MのKCl及び10mMのリン酸ナトリウムバッファ中の20μMのチトクロムc、pH7.4/7.8)を記録した。見掛電位を、異なるスキャン速度(100−400mV/秒)でのアノード及びカソードピーク電位間の中点値として計算し、拡散係数をRandles−Sevcik式に従って異なるスキャン速度でのピーク電流から計算した。
固相ペプチド合成法を用い、また全てのアミノ酸誘導体は市販されている。ペプチド会合の完了後、ペプチドを通常のやり方で樹脂から切断した。未精製のペプチドを、分取逆相クロマトグラフィで精製する。ペプチドの構造同一性をFAB質量分析により確認し、その純度を分析逆相HPLC及び薄層クロマトグラフィにより3つの異なる系で評価した。>98%の純度が達成される。典型的には、5gの樹脂を使用した合成により約2.0−2.3gの純粋なペプチドが得られる。
吸収分光法(UltroSpec 3300 Pro、220−1100nm)を用いて、SS−31がチトクロムc還元を変化させるのか否かを判定した(図1)。グルタチオンでのチトクロムcの還元は、Qバンド(450−650nm)での複数のシフトと関係していて、550nmでのシフトが顕著である。SS−31を添加すると、550nmで著しいスペクトル重みシフト(spectral weight shift)が発生した(図1A)。時間依存分光は、SS−31がチトクロムc還元の速度を上昇させたことを示す(図1B)。これらのデータは、SS−31がチトクロムcの電子構造を変化させ、またFe3+からFe2+ヘムへの還元を強化したことを示す。
サイクリックボルタンメトリ(CV)を行って、SS−31が電子流及び/又はチトクロムcの還元/酸化電位を変化させたか否かを判定した(図2、上パネル)。CVを、Au作用電極、Ag/AgCl基準電極及びPt補助電極を使用して行った。SS−31は、チトクロムcの還元及び酸化過程の両方について電流を増加させた(図2、上パネル)。SS−31は還元/酸化電位を変化させないものの(図2、上パネル)、チトクロムcを通る電子流を増加させ、これはSS−31が複合体III−IV間の抵抗を低下させることを実証している。図2(下パネル)の場合、全てのボルタンメトリ測定を、BASi C3セルスタンドに連結されたBASi−50Wボルタンメトリックアナライザを使用して行った。Ag/AgCl電極を基準として使用し、ガラス状炭素及び白金電極を標準測定に使用した。各測定に先立って、電極の汚損を回避するために、溶液を窒素で完全に脱気した。図2(下パネル)に示すように、サイクリックボルタモグラムを、トリス−ホウ酸塩−EDTA(TBE)バッファ、バッファ+チトクロムc及びバッファ+チトクロムc+2つの異なるSS−31用量について記録した。電流(電子拡散速度)は、SS−31の用量がチトクロムcの2倍になるとほぼ200%増加する(チトクロムc:SS−31=1:2)。この結果は、SS−31がチトクロムcにおける電子拡散を増進させ、より感度の高いバイオ検出器の設計にとってこのペプチドがより有用なものになることを示す。
フォトルミネッセンス(PL)を行うことで、SS−31がチトクロムcのヘムの伝導帯(電子輸送に関与するエネルギー状態)の電子構造に与える影響を調べた(図3)。Nd:YDO4レーザー(532.8nm)を使用して、チトクロムcにおいて電子を励起させた(図2A)。チトクロムcにおける強いPL放出状態をはっきりと650nmで確認することができる(図2B)。PL強度は、SS−31を添加すると用量依存的に上昇し、これはチトクロムcの伝導帯における利用可能な電子状態の増加を実証している(図2B)。これは、SS−31がチトクロムcの伝導帯の電子容量を上昇させることを示し、これはSS−31が仲介するチトクロムcを流れる電流の増加と一致している。
円偏光二色性分析(Olis社の分光偏光計、DSM20)を行うことで、ソーレー帯(415nmで負ピーク)を、チトクロムcにおけるπ−π*ヘム環境についてのプローブとしてモニタした(図4)。SS−31はこのピークの「赤色」シフトを440nmに推進し、これはSS−31が変性を伴うことなくチトクロムc内で新規なヘム−チロシンπ−π*遷移を引き起こしたことを実証している(図4)。これらの結果は、SS−31がヘムの現在の環境を、ヘムへの電子トンネル効果のための追加のTyrを供給すること又は内在性Tyr残基とヘムとの間の距離を減少させることで変化させることを示す。ヘム周囲でのπ−π*相互作用における上昇は、電子拡散に好適となる電子トンネル効果を強化し得る。
単離したラットの腎臓のミトコンドリアの酸素消費を、Oxygraphを使用して求めた(図5)。呼吸速度を、異なる濃度のSS−31の存在下、状態2(400μMのADPのみ)、状態3(400μMのADP及び500μMの基質)及び状態4(基質のみ)で測定した。全ての実験を、n=4−7で3重に行った。結果は、SS−31が、ミトコンドリアを脱共役することなく酸素への電子伝達を増進させたことを示す(図5)。
ミトコンドリアによるATP合成速度を、400mMのADPを添加してから1分後に単離したミトコンドリアから回収した呼吸緩衝液におけるAPTを測定することで求めた(図6)。ATPをHPLCで分析した。全ての実験をn=3で3重に行った。単離したミトコンドリアにSS−31を添加すると、ATP合成速度が用量依存的に上昇した(図6)。これらの結果は、SS−31による電子伝達の強化がATP合成に結びついていることを示す。
ミトコンドリア呼吸に対するSS−31の作用におけるチトクロムcの役割を実証するために、ミトコンドリアでのO2消費に対するSS−31の効果を、一旦凍結させたラットの腎臓のミトコンドリアから作成したチトクロムc欠乏マイトプラストにおいて判定した(図7)。呼吸速度を、100μMのSS−31を伴って又は伴うことなく、500μMのスクシネートの存在下で測定した。実験を、n=3で3重に行った。これらのデータは、(1)SS−31がIMMに緊密に結合したチトクロムcを介して働くこと、(2)SS−31が機能性チトクロムcにおける減退を救うことができると示している。
SS−31及びSS−20は、還元剤としてのグルタチオン(GSH)によって引き起こされるチトクロムcの還元の動力学を加速させることができる(図13)。チトクロムcの還元を、550nmでの吸光度における上昇でモニタした。GSHの添加は、550nmでの吸光度における時間依存性の上昇をもたらした(図13)。同様の結果が、還元剤としてN−アセチルシステイン(NAC)を使用して得られた(図示せず)。SS−31だけを濃度100μMで添加してもチトクロムcは還元されなかったが、SS−31は用量依存的にNAC誘発性のチトクロムc還元の速度を上昇させ、これはSS−31が電子を供与しないが電子の伝達速度を加速可能なことを実証している。
SS−20及びSS−31は共に、単離されたラットの腎臓のミトコンドリアにおけるO2消費で測定される電子束を増進させることができる(図14)。SS−20又はSS−31を濃度100μMで、0.5mMのスクシネート(複合体II基質)及び400μMのADPを含有する呼吸緩衝液中の単離されたミトコンドリアに添加した。O2消費における同様の増加が、低濃度の複合体I基質(グルタメート/マレート)を使用した場合に観察された(データは示さず)。電子束における増加は、低濃度のスクシネートでエネルギー供給した単離されたミトコンドリアにおけるATP産生速度における著しい上昇と相関関係にあった(図15)。これらのデータは、SS−20及びSS−31の標的をIMMにすることで、電子伝達系における電子束を促進し、またATP合成を特には基質の供給が低下した条件下で改善することができることを示す。
チトクロムcを単離及び精製する方法は当該分野で公知である。1つの例示的で非限定的な方法を提供する。チトクロムcは幾つかの正に帯電した基を有し、これがチトクロムcのpIを約10している。このため、チトクロムcは通常、膜上のリン脂質の負電荷へのイオン性引力によりミトコンドリアの膜に結合されている。硫酸アルミニウム溶液における低pHでのブレンダでの均質化により、まず組織及びミトコンドリアが分解される。正に帯電したアルミニウムイオンは、負に帯電したリン脂質に結合することによって膜からチトクロムcを外してタンパク質を溶液中に遊離させることができる。過剰な硫酸アルミニウムを、pHを8.0にまで上昇させることで除去し、アルミニウムは水酸化アルミニウムの形態で沈殿する。
本明細書に記載の生物的環境浄化組成物及び方法を、以下の実施例で更に説明する。この実施例は説明だけを目的としたものであり、限定することを意図してはいない。化学物質及び他の成分は典型例として提示される。本明細書に記載の方法及び組成物の範囲内で、上記の開示を鑑みて修正を加え得る。
芳香族カチオン性ペプチドをコードしているオリゴヌクレオチドを化学的に合成する。このオリゴヌクレオチドを、マルチプルクローニングサイトの上流に構成的プロモータを担持する細菌プラスミドへのディレクショナルクローニングを可能にする独特の制限酵素部位が両端に含まれるように設計する。プラスミドを、オリゴヌクレオチド端部上の制限酵素部位に対応する酵素での制限酵素消化により調製する。分子生物学の慣用の技法を用いて、オリゴヌクレオチドをアニールし、調製したプラスミドにライゲーションする。ライゲーション産物を、選択培地上で成長させたE.コリで形質転換させる。当該分野で公知の方法を使用したDNAシーケンシングにより幾つかの陽性クローンをcDNAインサートについてスクリーニングする。陽性クローンを増幅し、発現構築物のストック溶液を調製する。
D.デスルフリカンスの100mlの一夜培養物(OD600=0.6)を遠心分離し、ペレットを滅菌水で3回洗浄し、最終体積200μlの滅菌水に再懸濁させる。30μlのアリコートを4μlのプラスミド調製物(1μg)と混合し、エレクトロパルセータ装置による5000V/cn電気パルスに6ミリ秒にわたって供する。組み換え細菌を、組み換えプラスミドによって付与された抗生物質耐性をベースとして選択する。
野生型及び組み換えD.デスルフリカンス株を、可溶性サルフェートを還元する容量について試験する。細菌を、Deutsche Sammlung von Mikroorganismen und Zellkulturen GmbH社(ドイツ微生物・細胞培養物コレクション社)が推奨する培地において30℃、嫌気性条件下で培養する。1280ppmのサルフェート水溶液を野生型及び組み換えD.デスルフリカンスと共に播種し、12時間にわたって培養する。
サルフェート濃度を、比濁技法(Icgen et al.,2006)を用いて測定する。サルフェートを、塩酸媒体中、塩化バリウムで沈殿させて不溶性の硫酸バリウム結晶を形成させる。グリセロール(104.16mL)、濃塩酸(60.25mL)及び95%イソプロピルアルコール(208.33mL)を含有する改良調整混合物を新たに調製する。各反応につき、2mLの細胞非含有上清を、250mLの三角フラスコ内のミリポア水で1:50に希釈し、5mLの調整混合物を添加する。懸濁液全体を撹拌してよく混合する。約1グラムの塩化バリウム結晶を、撹拌を1分間にわたって継続しながら添加する。この混合物を2分間にわたって静的条件下で静置してから、濁度を420nmで分光測光法により測定する。サルフェートイオンの濃度を、0−40ppmのNa2SO4の標準物質を使用して作成した曲線から求める。
芳香族カチオン性ペプチドを発現する組み換え細菌が、これらの条件下でより高い異化的サルフェート還元速度を示すことが予測される。
本発明が、本発明の個々の態様の単独の実例としての本願に記載の特定の実施形態によって限定されることはない。本発明の趣旨及び範囲から逸脱することなく数多くの本発明の改良形及び変化形を形成可能であることは当業者に明らかである。本発明の範囲内の機能的に同等の方法及び装置についても、本明細書で列挙したものに加えて、上記の説明から当業者には明らかである。このような改良形及び変化形は、付随する請求項の範囲内に含まれるものとする。本発明は、付随する請求項が権利を有する均等物の全範囲を含め、付随する請求項の用語によってのみ限定される。本発明は、特定の方法、試薬、化合物、組成物又は生物学的な系に限定されず、当然のことながら変化し得ることを理解されたい。また、本明細書で使用の用語が特定の実施形態だけを説明することを目的とし、限定を意図していないことを理解されたい。
本発明のまた別の態様は、以下のとおりであってもよい。
〔1〕チトクロムc含有試料においてチトクロムc還元を増加させる方法であって、
試料を有効量のペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2と接触させることを含む
ことを特徴とする方法。
〔2〕チトクロムc含有試料においてチトクロムcを通る電子拡散を強化する方法であって、
試料を有効量のペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2と接触させることを含む
ことを特徴とする方法。
〔3〕チトクロムc含有試料においてチトクロムc周囲にπ−π相互作用を引き起こす方法であって、
試料を有効量のペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2と接触させることを含む
ことを特徴とする方法。
〔4〕センサであって、
あるレベルのペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2をドープしたチトクロムcと、前記ペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2のレベルの変化によって引き起こされるチトクロムcの性質における変化を測定するための計測器とを含む
ことを特徴とするセンサ。
〔5〕前記ペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2のレベルが、前記チトクロムcの温度及び前記チトクロムcのpHの少なくとも一方における変動に応答して変化する、前記〔4〕に記載のセンサ。
〔6〕前記性質が導電率であり、前記計測器が、前記チトクロムcと電気的に連通するアノード及びカソードを含む、前記〔4〕に記載のセンサ。
〔7〕前記性質がフォトルミネッセンスであり、前記計測器が、前記チトクロムcが放出する光の強度及び前記チトクロムcが放出する光の波長の少なくとも一方における変化を測定するための光検出器を含む、前記〔4〕に記載のセンサ。
〔8〕検出方法であって、
ペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2のレベルにおける変化によって引き起こされる、あるレベルのペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2をドープしたチトクロムcの性質における変化を測定することを含む
ことを特徴とする方法。
〔9〕前記ペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2のレベルが、前記チトクロムcの温度及び前記チトクロムcのpHの少なくとも一方における変動に応答して変化する、前記〔8〕に記載の方法。
〔11〕前記性質が、導電率、フォトルミネッセンス強度及びフォトルミネッセンス波長の少なくとも1つである、前記〔8〕に記載の方法。
〔12〕スイッチであって、
チトクロムcと、前記チトクロムcと通じているペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2のソースと、前記チトクロムcと通じている前記ペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2の量を制御するためのアクチュエータとを含む
ことを特徴とするスイッチ。
〔13〕前記アクチュエータが、前記チトクロムcの温度及び前記チトクロムcのpHの少なくとも一方を制御する、前記〔11〕に記載のスイッチ。
〔14〕スイッチング方法であって、
チトクロムcと通じているペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2のレベルを変化させることを含む
ことを特徴とする方法。

〔24〕試料における基質の検出方法であって、
(a)試料を、
(i)基質特異性の酸化還元活性酵素と、
(ii)ペプチドD−Arg−2’,6’−Dmt−Lys−Phe−NH2をドープしたチトクロムcと、
(iii)電極
とを含むバイオセンサと接触させ、
(b)前記バイオセンサ内で電子の流れを検出することを含む
ことを特徴とする方法。

Claims (10)
- 環境汚染物質の生物的環境浄化方法であって、
環境汚染物質含有材料を、1種以上の芳香族カチオン性ペプチドを発現している組み換え細菌を含む生物的環境浄化組成物と接触させることを含む
ことを特徴とする方法。 - 前記環境汚染物質が、金属、非金属、パークロレート、ニトレート、放射性核種、メチル−tert−ブチル−エーテル(MTBE)、塩化ビニル又はジクロロエチレンを含む、請求項1に記載の方法。
- (a)前記金属が、Sc、Ti、V、Cr、Mn、Fe、Co、Ni、Cu、Zn、Y、Zr、Nb、Mo、Tc、Ru、Pd、Ag、Cd、Hf、Ta、W、Re、Os、Ir、Pt、Au、Hg、Rf、Db、Sg、Bh、Hs、Cn、Al、Ga、In、Sn、Ti、Pb若しくはBiを含む、
(b)前記非金属がサルフェートを含む、
(c)前記パークロレートが、NH4ClO4、CsClO4、LiClO4、Mg(ClO4)2、HClO4、KClO4、RbClO4、AgClO4若しくはNaClO4を含む、
(d)前記ニトレートが、HNO3、LiNO3、NaNO3、KNO3、RbNO3、CsNO3、Be(NO3)2、Mg(NO3)2、Ca(NO3)2、Sr(NO3)2、Ba(NO3)2、Sc(NO3)3、Cr(NO3)3、Mn(NO3)2、Fe(NO3)3、Co(NO3)2、Ni(NO3)2、Cu(NO3)2、Zn(NO3)2、Pd(NO3)2、Cd(NO3)2、Hg(NO3)2、Pb(NO3)2若しくはAl(NO3)3を含む、又は
(e)前記放射性核種がアクチニド若しくはウラニウムを含む、請求項2に記載の方法。 - 前記生物的環境浄化をインシチュ又はエクスシチュで行う、請求項1に記載の方法。
- 前記細菌が、前記1種以上の芳香族カチオン性ペプチドをコードする核酸配列を含む、請求項1に記載の方法。
- 前記核酸配列が、誘導プロモータ又は構成的プロモータの制御下で発現する、請求項5に記載の方法。
- 前記核酸配列がプラスミドDNAを含む、請求項5に記載の方法。
- 前記核酸配列がゲノムインサートを含む、請求項5に記載の方法。
- 前記組み換え細菌が、表7に挙げた細菌種由来である、請求項1に記載の方法。
- 前記芳香族カチオン性ペプチドが、D−Arg−2’,6’−Dmt−Lys−Phe−NH2を含む、請求項1〜9のいずれか1項に記載の方法。
