Abstract
Description
実施形態Aは、

上記水性組成物を1〜25℃の温度で少なくとも7日の期間保管する工程と、を含み、
上記水性組成物は、第1の時点で、初期ATP−ジホスホヒドロラーゼ活性を有し、
上記pHと有効量のポリオール、緩衝試薬、タンパク質、及びATP−ジホスホヒドロラーゼとの組み合わせは、組成物を第1の時点から第1の時点の少なくとも7日後である第2の時点まで、約25℃で保持した後に、初期ATP−ジホスホヒドロラーゼ活性の90%以上を保持することを可能にし、
上記ATP−ジホスホヒドロラーゼ活性は、非律速濃度のATP、ルシフェリン、及びルシフェラーゼを用いる、pH7.75での生物発光カップルアッセイにおいて測定される、方法である。
約6.0〜7.2のpHを有する水性組成物を含むキットであって、当該組成物は、有効量のソルビトール、HEPES、Trisスクシネート、血清アルブミン、及びATP−ジホスホヒドロラーゼ酵素活性成分を含む、キットである。
試料及び水性組成物を含む第1の混合物を形成する工程であって、上記水性組成物が約6.8〜約7.2のpHを有し、有効量のポリオール、緩衝試薬、タンパク質、及びATP−ジホスホヒドロラーゼを含む、工程と、
上記第1の混合物を、第1の期間にわたって所定の温度に保持する工程と、
第1の混合物を、細菌細胞抽出剤、ルシフェラーゼ、ルシフェリン、及び緩衝試薬と組み合わせて、pHが約7.75の第2の混合物を形成する工程と、ルシフェラーゼに触媒された生物発光反応を測定する工程と、を含む方法である。
25℃で様々な期間保管した後の凍結乾燥ATP−ジホスホヒドロラーゼ酵素活性の安定性

Claims (20)
- 約6.0〜約7.2のpHを有する水性組成物を含み、前記組成物が、有効量のポリオール、緩衝試薬、タンパク質、及びATP−ジホスホヒドロラーゼを含む、キット。
- 体細胞抽出剤を更に含む、請求項1に記載のキット。
- 前記組成物が、有効量の殺微生物化合物を含まない、請求項1又は2に記載のキット。
- 前記組成物が、有効量のジチオトレイトール、ジチオエリスリトール、又はβ−メルカプトエタノールを含まない、請求項1〜3のいずれか一項に記載のキット。
- ルシフェラーゼ酵素活性成分を更に含む、請求項1〜4のいずれか一項に記載のキット。
- 前記ルシフェラーゼ酵素活性成分が、前記水性組成物から分離されている、請求項5に記載のキット。
- 前記水性組成物のpHが約6.4〜約7.0である、請求項1〜6のいずれか一項に記載のキット。
- 前記ポリオールが、ソルビトール、キシリトール、グリセロール、及びこれらの混合物からなる群から選択される、請求項1〜7のいずれか一項に記載のキット。
- 前記ポリオールがソルビトールを含み、前記ソルビトールが、前記水性組成物中に約1モル/リットル〜約1.6モル/リットルの濃度で存在する、請求項8に記載のキット。
- 前記水性組成物が、有効量の前記体細胞抽出剤を含む、請求項1〜9のいずれか一項に記載のキット。
- 前記体細胞抽出剤が、非イオン性界面活性剤を含む、請求項1〜10のいずれか一項に記載のキット。
- 前記体細胞抽出剤が、ポリエチレングリコールp−(1,1,3,3−テトラメチルブチル)−フェニルエーテル及びポリエチレングリコールモノアルキルエーテルからなる群から選択される、請求項11に記載のキット。
- 前記緩衝試薬が、HEPESとTrisスクシネートとの混合物を含む、請求項1〜12のいずれか一項に記載のキット。
- 前記緩衝試薬が、前記水性組成物中に約0.5mM〜約200mMの濃度で存在する、請求項1〜13のいずれか一項に記載のキット。
- 前記タンパク質が、血清アルブミン及び精製コラーゲンからなる群から選択される、請求項1〜14のいずれか一項に記載のキット。
- 前記ATP−ジホスホヒドロラーゼが、前記水性組成物中に約250ユニット/リットル〜約2500ユニット/リットルの濃度で存在する、請求項1〜15のいずれか一項に記載のキット。
- 細菌細胞抽出剤を更に含む、請求項1〜16のいずれか一項に記載のキット。
- 0℃超で少なくとも7日間保管された水性組成物において初期ATP−ジホスホヒドロラーゼ酵素活性の90%以上を保持する方法であって、
約6.0〜7.2のpHを有し、有効量のポリオール、緩衝試薬、タンパク質、及びATP−ジホスホヒドロラーゼを含む、水性組成物を形成する工程と、
前記水性組成物を1〜25℃の温度で少なくとも7日の期間保管する工程と、を含み、
前記水性組成物は、第1の時点で、初期ATP−ジホスホヒドロラーゼ活性を有し、
前記pHと前記有効量の前記ポリオール、前記緩衝試薬、前記タンパク質、及び前記ATP−ジホスホヒドロラーゼとの組み合わせは、前記組成物を前記第1の時点から前記第1の時点の少なくとも7日後である第2の時点まで約25℃で保持した後に、前記初期ATP−ジホスホヒドロラーゼ活性の90%以上を保持することを可能にし、

試料及び水性組成物を含む第1の混合物を形成する工程であって、前記水性組成物が、約6.8〜約7.2のpHを有し、有効量のポリオール、緩衝試薬、タンパク質、及びATP−ジホスホヒドロラーゼを含む、工程と、
前記第1の混合物を、第1の期間にわたって所定の温度に保持する工程と、
前記第1の混合物を、細菌細胞抽出剤、ルシフェラーゼ、ルシフェリン、及び緩衝試薬と組み合わせて、pHが約7.75の第2の混合物を形成する工程と、
