Abstract
Description
特に、医薬、化粧料、化学物質などの安全性や効能の評価には、動物の生体を使わずに、in vitroで実施する試験法(動物実験代替法)の開発が大きな課題となっている。しかしながら、生体における細胞の機能や薬剤への反応の過程を、in vitroで完全に再現することは難しい。ここで、一般的な二次元培養(単層培養)を用いるよりも、三次元培養を用いる方が生理的条件を模倣するために有用であることが、例えば、皮膚、肝臓、がん腫瘍などの研究において、既に知られている。
また、少なくとも2つの異なる表現型の哺乳類細胞の三次元凝集体及び液体増殖培地を含有する哺乳類組織のin vitroモデルが提案されている(例えば、特許文献2参照)。また、特許文献2には、少なくとも1つの表現型の細胞は三次元凝集体を形成する前に蛍光染色で標識されており、哺乳類組織のin vitroモデルを用いて、前記細胞の増殖を阻害する抗がん剤などの物質をスクリーニングする方法が開示されている。
前記第一の細胞は、外部からの刺激に応答して、化学発光、生物発光又は蛍光によって光を発する細胞である。
本発明の三次元組織体は、第一の細胞を含む第一の細胞領域と、前記第一の細胞とは異なる第二の細胞を含む第二の細胞領域とが存在し、前記第一の細胞は、外部からの刺激に応答して、化学発光、生物発光又は蛍光によって光を発する細胞(以下、「発光細胞」と称することもある)である。
前記三次元組織体は、三次元の各方向において、少なくとも細胞2個分の厚みで配置されている。
前記三次元組織体は、細胞同士が直接結合していてもよく、例えば、コラーゲン、プロテオグリカン、ヒアルロン酸、フィブロネクチン、ラミニン、エラスチン、フィブリリン、キチン、セルロースなどの細胞外基質(ECM)や足場材料(Scaffold)や後述の固定剤が細胞の間に充填されていてもよい。
前記発光細胞とは異なる細胞(第二の細胞)を含む領域(第二の細胞領域)とは、前記発光細胞は存在していないことで特定される領域である。
前記発光細胞を含む領域と、前記発光細胞とは異なる細胞を含む領域とは、隣接していることが好ましい。ただし、2つの領域の境の間に、少量(体積で50%未満)の前記発光細胞が散在している領域があってもよい。
前記外部からの刺激に対し、前記三次元組織体内の特定な位置にある細胞は、例えば、周辺組織の構造の違い又は刺激源からの距離により、他の領域に位置する細胞とは異なる反応を示す可能性がある。例えば、前記三次元組織体に、前記外部からの刺激を一定の方向から与えると、刺激を与える界面に垂直な軸に対する深さに応じて、刺激の度合い、すなわち細胞の反応が変化する可能性が推測される。よって前記発光細胞を含む領域は、その変化の評価を可能にするための適切な深さに配置することが好ましい。
前記三次元組織体は、例えば、刺激物質の安全性評価もしくは効能評価又は薬剤のスクリーニングに適用することができる。また、薬物動態研究や再生医療の開発などを目的に、三次元組織体を生きた動物に移植した後に、組織・臓器再生の経過を観察することもできる。
三次元組織体10は、細胞Aを含む領域11、細胞Bを含む領域12及び培地13が、培養容器14内に順次積層されている。
第二の細胞としての細胞Aは、細胞Bとは異なる発光細胞であってもよいし、発光細胞でなくてもよい。
細胞Bを含む領域12の表面は、培養容器の存在による人為的結果を避けるために、培養容器14の周壁に接触していないことが好ましい。具体的には、細胞Bを含む領域12は、三次元組織体10の中心部に配置されていることが好ましい。これにより、細胞Bを含む領域12と培養容器14の界面の影響を排除することができる。
細胞Bを含む領域12は、刺激を与える界面に垂直な軸に対して、一定の深さに存在することが好ましい。これにより、刺激源からの距離又は刺激物質の浸透性の影響を評価することができる。
なお、三次元組織体は、細胞Aを含む領域の内部に、細胞Bを含む領域が存在していればよく、細胞Bを含む領域は、層状でなくてもよい。
また、三次元組織体は、細胞A及び細胞Bとは異なる細胞を含む領域が更に存在していてもよい。この場合、細胞A及び細胞Bとは異なる細胞は、発光細胞であってもよいし、発光細胞でなくてもよい。
細胞の配置方法としては、特に制限はなく、公知の方法を適宜使用することができ、例えば、ピペット分注、マイクロマニピュレーター、インクジェット法、ゲル押し出し法、スクリーン印刷等の転写法、光ピンセットなどが挙げられる。これらの中でも、細胞を効率よく数十μm2〜数mm2以下の狭い範囲の領域に薄く、均一に配置する点から、インクジェット法が好ましい。前記インクジェット法によれば、数pLの液滴を吐出するため、細胞1〜10個を精密に配置することができる。
前記細胞A及びBとしては、三次元組織体を形成することが可能であれば、特に限定されず、分類学的に、例えば、真核細胞、原核細胞、多細胞生物細胞、単細胞生物細胞を問わず、すべての細胞を使用することができる。これらの中でも、細胞と細胞とが互いに接着し、物理化学的に処理しなければ、単離しない程度の細胞接着性を有する哺乳類又はヒト由来の接着性細胞が好ましい。
前記発光細胞は、化学発光、生物発光又は蛍光によって光を発する。このため、外部からの刺激に応答して、細胞内に起きる様々な分子メカニズムを解析することができる。
化学発光は、化学反応に伴って発光する現象であり、例えば、ペルオキシダーゼが過酸化水素を分解すると起こるルミノール反応などが挙げられる。
生物発光は、生物体の行う化学発光であり、発光酵素などの働きによってエネルギーが光として放出される現象をいい、例えば、ホタルが持つルシフェラーゼ(酵素)がルシフェリン(基質)を酸化すると光が放出されることなどが挙げられる。
蛍光は、励起状態にある物質が基底状態へもどる際に,電子遷移に伴って放出される光であり、生物においては、例えば、オワンクラゲが持つGFP(緑色蛍光タンパク質)に励起光を当てると緑色の光が放出されることなどが挙げられる。
この場合、レポーター遺伝子は、発現レベルを測定する標的遺伝子のプロモーターの下流に連結されている。
前記レポーター遺伝子としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、ルシフェラーゼ遺伝子、GFP(緑色蛍光蛋白質)遺伝子、又はこれらの派生物などが挙げられる。
その他の細胞を発光させる方法としては、特に限定されないが、細胞内の異なる分子の相互作用を評価する蛍光/発光共鳴エネルギー移動(FRET/BRET)、酵素や結合因子などの活性や遊走の指標となる蛍光/発光基質の取り込み(蛍光カルシウムインジケーター、Aldefluorアッセイ、Glucose取込みイメージング、蛍光ナノ粒子プローブなど)、などの公知の方法を用いることができる。
なお、前記細胞A及び細胞Bが発光細胞である場合は、異なる条件やタイミングで発光する発光細胞、異なる波長の光を発光する発光細胞などを用いて区別することができる。例えば、三次元組織体の深さに応じて、異なる波長の光を発光する発光細胞を配置することができる。また、異なる条件で発光する疾患細胞と正常細胞を共培養として配置し、お互いの反応を比較したりすることができる。
前記培養容器は、三次元組織体の形成と維持に必要な土台や支持体となる基材からなる。
前記基材を構成する材料としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、樹脂、ガラス、金属などが挙げられる。
前記基材の形状としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、平面状のみならず、穴が開いた形状、メッシュ状、凹凸のある形状、ハニカム状などが挙げられる。
前記培養容器としては、細胞の接着と増殖に適した基材からなり、光透過性が高く、自己発光しないマルチウェルタイプのカルチャープレートやインサートなどを用いることが好ましい。
なお、三次元組織体10では、細胞Aを含む領域11が培養容器14に接触しているが、細胞Aを含む領域11が培養容器14に常時接触していなくてもよい。また、三次元組織体10は、培地13や安全性評価又は効能評価する物質やスクリーニングする薬剤を出し入れするために、開放系となっているが、閉鎖系で環流培養が可能なものやマイクロ流路を持つチップ型などであってもよい。
前記培地は、三次元組織体の形成と維持に必要な成分を含み、乾燥を防ぎ、浸透圧などの外部環境を整える溶液である。
前記培地としては、特に制限はなく、公知の培地の中から、用途に応じて、適宜選択すればよい。
なお、表面が空気に暴露されている皮膚のように、常時培地内に浸しておく必要のない三次元組織体においては、培地を適宜除去してもよい。
細胞Bを特定の位置に固定し、三次元組織体の構造を保持するために、固定剤が必要な場合がある。細胞Bを固定する時間は、一時的であってもよいし、永続的であってもよいが、少なくとも三次元組織体を形成した後、細胞培養時及び使用時に形状が崩れない程度の時間を必要とする。また、固定剤は、細胞に悪影響を与えないような生体適合性があるものが望ましい。前記固定剤は、細胞と混合して配置してもよいし、細胞とは別に配置してもよい。
前記固定剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、生体由来高分子(コラーゲン、エラスチン、ゼラチン、フィブロインなど)、凝固因子(フィブリノーゲン/トロンビンなど)、接着因子(フィブロネクチン、ラミニン、リコンビナントペプチドなど)、ポリ乳酸等の合成高分子、アルギン酸、ジェランガム等の多糖類/多価金属塩などが挙げられる。
細胞の配置方法によっては、固定剤は、2液以上の混合により増粘する相変化材料であることが好ましい。
前記固定剤として、2液以上の混合により増粘する相変化材料を使う際には、そのうちの1液を事前に細胞懸濁液に混合してもよいし、別途液滴を適切な位置に配置してもよい。なお、固定剤を用いる場合、前記固定剤は、三次元組織体中に残留していてもよいし、残留していなくてもよい。
−細胞の準備−
細胞Aとして、正常ヒト皮膚繊維芽細胞(Normal Human Dermal Fibroblast(Lonza社製)(以下、「NHDF細胞」という)を用いた。
細胞Bとして、NF−κB転写因子の応答配列のプロモーターの下流にルシフェラーゼ配列が連結されている遺伝子を含む市販のマウス繊維芽細胞NFkB−luciferase NIH3T3 Reporter Stable Cell Line(Panomics社製)(以下、「NFkB−luc細胞」という)を用いた。
まず、NHDF細胞及びNFkB−luc細胞を、メーカー推奨の手順に従い、拡大培養した。
次に、トリプシン処理により細胞を単離した後、発光反応とは独立的に共焦点蛍光顕微鏡観察で識別可能にするために、NHDF細胞及びNFkB−luc細胞を、それぞれCellTracker Green及びCellTracker Orange(以上、Thermo Fisher社製)で染色した。
図2A〜図2Eに示すとおり、染色したNFkB−luc細胞及びNHDF細胞を用いて、三次元組織体を作製した。
まず、培養容器14としての96ウェルプレートサイズのウェル(0.3cm2)に、0.2質量%コラーゲン液に1×107細胞/mLの密度で懸濁したNHDF細胞を、マイクロピペットMで20μL分注し、細胞層が固化するまで37℃のCO2インキュベーターに静置した(図2A参照)。
図3に、三次元組織体10をZ−Stack撮影した画像を示す。図3から、厚みが約180μmの細胞A(NHDF細胞)を含む領域11の中心部に異なる色の細胞B(NFkB−luc細胞)を含む領域12が細胞B(NFkB−luc細胞)の1個分の厚みで配置されていることが確認された。
実施例1において、厚みが1.6mmの細胞A(NHDF細胞)を含む領域11の底部に細胞B(NFkB−luc細胞)を含む領域12を配置した以外は、実施例1と同様にして、三次元組織体を得た(図4参照)。
実施例1において、厚みが1.6mmの細胞A(NHDF細胞)を含む領域11の頂部に細胞B(NFkB−luc細胞)を含む領域12を配置した以外は、実施例1と同様にして、三次元組織体を得た(図5参照)。
実施例2において、細胞A(NHDF細胞)を含む領域11の代わりに、DMEM培地を配置した以外は、実施例2と同様にして、三次元組織体を得た(図6参照)。
図7に、比較例1の三次元組織体を示す。
特開2012−179020号公報の実施例1及び2で使用されている「3D培養皮膚モデルEpiDermTM(MatTek corporationより入手)」のように、観察する領域を特定する発光細胞が含まれていない場合、又は図7に示すように、含まれていても三次元組織体全体に無作為に分散している場合には、三次元組織体全体をバルクとした平均的な反応しか計測できず、三次元組織体の中のある特定な領域の反応がその他の領域とは異なるかを非破壊的に評価するには、発光細胞を対照となる領域に配置する技術を用いる必要がある。
図8に、国際公開第2000/75286号パンフレットの実施例2(図3)を再現した比較例2の三次元組織体を示す。
図8に示すように、発光細胞とその他の細胞が全体的に混合されている場合には、三次元組織体全体をバルクとした平均的な反応しか計測できず、三次元組織体の中のある特定な領域の反応がその他の領域とは異なるかを非破壊的に評価するには、発光細胞を興味の対照となる領域に配置する技術を用いる必要がある。
NFkB−luc細胞の発光を誘導するために、刺激物質71としての、50ng/mLのTNF−αの水溶液と、発光基質としての、200μMのD−ルシフェリンの水溶液を三次元組織体に加え、37℃のCO2インキュベーターに4時間静置して反応させた。
次に、プレートリーダーCytation5(BioTek社製)を用いて、ゲイン175、積分時間3秒間の設定で、三次元組織体(各8個)の発光強度を測定した。
このとき、ブランクとして、DMEM培地を用いた。表1に、三次元組織体の発光強度の測定結果を示す。
図10に、実施例2、実施例3、及び対照例1の三次元組織体の平均発光強度と標準偏差を示す。
図9A及び図10は、対照例1である。
図9C及び図10から、実施例3の三次元組織体は、細胞A(NHDF細胞)を含む領域11の頂部に細胞B(NFkB−luc細胞)を含む領域12が配置されているため、細胞B(NFkB−luc細胞)が、頂部から添加した刺激物質(TNF−α)71に応答し、比較例1と同等の発光強度が検出されることがわかった。
一方、図9B及び図10から、実施例2の三次元組織体は、細胞A(NHDF細胞)を含む領域11の底部に細胞B(NFkB−luc細胞)を含む領域12が配置されているため、発光強度が低下した。この結果から、実施例2の三次元組織体は、細胞A(NHDF細胞)を含む領域11の厚みの影響により、刺激物質(TNF−α)71が、細胞B(NFkB−luc細胞)を含む領域12まで浸透していないことが示唆された。
発光強度の低下が単に細胞A(NHDF細胞)を含む領域11に光吸収されていることによる可能性は、細胞B(NFkB−luc細胞)の代わりに、刺激物質とは非依存的に発光する細胞を用いることで除外できた(例えば、恒常的に発現するプロモーターの下流にルシフェラーゼ配列が連結されている遺伝子を含むTK−Luc細胞)。
これらの結果から、実施例2及び実施例3の三次元組織体は、特定の領域における発光強度、即ち、刺激物質に対する応答を非破壊的に評価できることが示された。また、実施例2及び実施例3を比較することにより、刺激物質への応答が三次元組織体内の領域の位置によって変化するかを評価できた。
細胞Bを含む領域12は層状でなくともよい。図11A及び図11Bに示す例は、予め低吸着処理プレート(住友ベークライト株式会社製)を用いて、細胞B(NFkB−luc細胞)の約300μm直径の球体の塊(スフェロイド)を作製した。
次に、実施例1と同様にして、0.2質量%コラーゲン液に懸濁した細胞A(NHDF細胞)を、培養容器14に配置した。この時、培養容器14としては、頂部からのみ刺激物質が添加可能な培養プレートではなく、Falconカルチャーインサート0.4μmを用いた(Corning社製)。この培養容器を用いると、多孔膜で作られた底部81(図11B参照)からも三次元組織体全体を培養液及び刺激物質に水浸できた。
最後に、マイクロマニピュレーターを用いて、細胞B(NFkB−luc細胞)のスフェロイドを、細胞A(NHDF細胞)を含む領域11の中心部に配置し、更にスフェロイドを完全に包埋するように0.2質量%コラーゲン液に懸濁した細胞Aを追加する。実施例4を用いた場合、三次元組織体の外部から内部への刺激を受けて、中心部の発光強度、即ち刺激物質への応答が評価できた。
また、このような形状の容器を用いた場合、頂部の培地13と底部の培養液41は異なる刺激物質を含んでもよく、例えば、2種類の刺激物質が同時に中心の細胞Bを含む領域12に到達した場合のみの反応を評価できた。
細胞Bを含む領域12は一体化していなくともよく、図12A及び図12Bに示すように、同一の三次元組織体において複数の細胞Bを含む領域が、X、Y及びZの特定な座標に配置されていてもよい。実施例5を作製するためには、培養容器14としての3.5mmサイズのカルチャーディッシュに、実施例1と同じインクジェット方式のバイオプリンターPを用いて、予め設定した座標に細胞B(NFkB−luc細胞)を含む領域12を9個のエリアに分けて配置した以外は、実施例1と同様にして、三次元組織体を構築した。
実施例5を用いた場合、刺激物質を添加した時の深さにおける浸透だけでなく、広範囲における反応の伝達速度などの評価も可能となる。
細胞Bを含む領域12は、図13に示すように、細胞Aを含む領域11を包み込む形でもよい。実施例1と同じインクジェット方式のバイオプリンターPを用いて予め細胞Aを含む細胞シートと、細胞Bを含む細胞シートが2枚重ねに積層されるように構築し、筒状に丸めて内部を培養液で環流させた。この場合、筒の内側から放射状に外側に向かって刺激反応が変化するかを評価することができる。
