Abstract

本出願は、2009年2月20日に出願された米国仮特許出願番号61/154,210に対する優先権を主張し、上記米国仮特許出願は、参照によって本明細書に援用される。
本発明は全般的に分子生物学および医学の分野に関する。より詳細には、本明細書は、胚性幹細胞から造血前駆細胞および内皮前駆細胞などの前駆細胞を作製するための方法および組成物に関する。本発明はさらに、前駆細胞を作製するためのキット、および多能性幹細胞の分化を促進する物質をスクリーニングする方法に関する。
ヒト胚性幹細胞は、インビトロで無限に培養増殖することができるため、少なくとも原理的には、問題または障害のあるヒト組織に代わる細胞および組織を供給することができる。造血幹細胞および前駆細胞は医療において大きな可能性があることから、胚性幹細胞からの造血前駆細胞の分化の方法を改良しようとする多くの研究がなされてきた。ヒト成人の場合、造血幹細胞の少数は主に骨髄に存在し、活発に分裂する造血前駆細胞(CD34+)の不均一な集団を形成しており、これが血液系のすべての細胞に分化する。CD34は他の細胞型、特に内皮細胞(血管)にも発現するため、CD34+マーカーは造血細胞の十分なマーカーとは言えない。このため、造血前駆細胞を同定しやすくするため、CD43マーカーなど他のマーカーも使用してもよい(たとえば、非特許文献1;非特許文献2)。成人ヒトの造血前駆体は増殖および分化して1日に数千億個の成熟血液細胞を産生する。造血前駆細胞は臍帯血にも存在する。
本発明は、間質フィーダー細胞を使用したり、または胚様体を形成したりせずに多能性幹細胞から造血細胞および内皮細胞を作製し、再現性よく分化する方法および組成物を提供することにより従来技術の限界を克服するものである。さらに、本発明の方法は、たとえば規定された分化培地および特定の大気条件を使用することにより多能性幹細胞の分化を改善する。本明細書で使用する場合、「規定された条件」、「規定された培地」および「規定された分化」という用語は、培地の全成分が既知量であり、かつ明らかでない成分、血清またはフィーダー細胞(たとえば、マウス胎仔線維芽細胞)を使用しない培養条件をいう。「明らかでない成分」とは、未知の構成要素を含むか、あるいは既知の構成要素を未知の量で含む成分である。規定された条件は、たとえば、分化させた細胞をヒト患者などの被験体に治療投与する場合がある用途で特に有用であり得る。「血清」という用語は、本明細書で使用する場合、増殖細胞に栄養素を与えるために培地に加えてもよいヒト以外の動物の生成物をいう。
より分化しているとはいえ、依然として2種以上の細胞に分化する能力を持つ。たとえば、造血前駆細胞は多能性幹細胞より分化しているが、造血前駆細胞はなお、たとえば赤血球(erythrocyte)、マクロファージ、顆粒球、巨核球、樹状細胞またはマスト細胞に分化する能力を持つ。本発明のいくつかの実施形態では、前駆細胞は造血前駆細胞である。他の実施形態では、前駆細胞は内皮前駆細胞である。なお他の実施形態では、前駆細胞は、造血細胞または内皮細胞に分化できる血液内皮(hematoendothelial)(また
は血管芽細胞)前駆細胞である。

本発明は、間質細胞または胚様体を必要とすることなくヒト多能性幹細胞から造血細胞または内皮細胞を作製する方法および組成物を提供する。いくつかの方法は規定された分化培地を使用するものであり、低酸素雰囲気条件を含んでもよい。こうした方法を使用してヒト造血前駆細胞を作製し、これをさらに赤血球(erythrocyte)、マクロファージ、顆粒球および/または巨核球などの細胞系統へ分化させてもよい。さらに、本方法を使用してヒト内皮前駆細胞を作製し、これをさらに内皮細胞に分化させてもよい。本発明の分化培地は、増殖因子(たとえば、BMP−4、VEGF、bFGF)を含んでもよく、フィブロネクチンなどのマトリックス成分と一緒に使用してもよい。
従来の多能性幹細胞の培養方法は、血清製品およびマウスフィーダー層に依存して多能性幹細胞を様々な細胞型に分化させてきた。これらの手順は、分化を行える規模を制限し、生物学的ばらつきおよびコンタミネーションの可能性を増大させ、そうでなければ有用であるはずのトランスレーショナル治療において多能性幹細胞の使用を著しく妨げている。
当該技術分野においては様々な増殖因子が公知であり、本発明と共に使用してもよい。ある種の実施形態では、本発明の分化培地は、たとえば、BMP−4、VEGFおよびbFGFなど1種、2種またはそれ以上の増殖因子を含んでもよい。マウス胎仔線維芽細胞培養系を使用してこれらの増殖因子を用いれば、ヒト胚性幹細胞を造血細胞および内皮細胞に分化させることができる(Wang et al.,2007)。
骨形成タンパク質−4(BMP−4)は骨形態形成タンパク質群のメンバーで、腹側中胚葉誘導因子である。BMPは成人ヒト骨髄(BM)に発現し、骨のリモデリングおよび成長に重要である。ある種の実施形態では、BMP4を加える必要があるのは、培養の最初の2〜3日間だけであり、その後は分化に有害な影響を与えずに系から除去し得る。
al.,2000)。たとえば、BMP−4は、成人および新生児由来の高度に精製された原始ヒト造血細胞の増殖および分化を調節することができ(Bhatia et al.,1999)、BMP−4はヒト胚性幹細胞の造血分化を促進することもできる(Chadwick,2003)。BMP−4はさらに、内皮前駆細胞からの内皮細胞の分化を促進することもできる(Wang et al.,2007)。
内皮細胞増殖因子(VEGF)は、胎児の循環系の形成および血管形成に関与する重要なシグナル伝達タンパク質である。VEGFは、内皮および他の細胞型(たとえば、ニューロン、がん細胞、腎上皮細胞)など様々な細胞型に影響を与え得る。VEGFはインビトロで内皮細胞の分裂および遊走を刺激し得る。またVEGFの機能は、がん、糖尿病、自己免疫疾患および眼血管疾患など様々な病状において重要であることも明らかになっている。
塩基性線維芽細胞増殖因子(bFGF、FGF−2とも呼ばれる)は、四肢および神経系の発生、創傷治癒および腫瘍増殖など多様な生物学的プロセスに関係していると考えられている増殖因子である。これまでの研究から、bFGFは、ヒト胚性幹細胞のフィーダー非依存性増殖を支持するために使用されてきたが(Ludwig et al.,2006a)、造血細胞の発生または生存に影響を与えないと考えられることが明らかになっている(Ratajczak et al.,1996)。ある種の実施形態では、分化を誘導するのにbFGFを必要としない。このため、種々の実施形態では、bFGFを本発明の培地に加えても、あるいは加えてなくてもよい。
SCF(SCF、kitリガンド、KLまたはスティール因子とも呼ばれる)は造血、精子形成およびメラニン形成に関与するサイトカインである。本発明の方法では、SCFを培地に約2.5ng/mL、5ng/mL、10ng/mL、15ng/mL、20ng/mL、25ng/mL、30ng/mL、35ng/mL、40ng/mL、45ng/mL、50ng/mL、55ng/mL、60ng/mL、65ng/mL、70ng/mL、75ng/mL、80ng/mL、85ng/mL、90ng/mL、95ng/mLまたは約100ng/mLの濃度で加えてもよい。好ましい実施形態では、SCFを培地に約25ng/mLの濃度で加える。
TPOも、造血前駆細胞を、たとえば、巨核球に分化させるのに関係している。本発明の方法では、TPOを培地に約2.5ng/mL、5ng/mL、10ng/mL、15ng/mL、20ng/mL、25ng/mL、30ng/mL、35ng/mL、40ng/mL、45ng/mL、50ng/mL、55ng/mL、60ng/mL、65ng/mL、70ng/mL、75ng/mL、80ng/mL、85ng/mL、90ng/mL、95ng/mLまたは約100ng/mLの濃度で加えてもよい。好ましい実施形態では、TPOを培地に約25ng/mLの濃度で加える。
一部の実施形態では、本発明の分化培地を使用して多能性幹細胞を播種しても、培養しても、維持しても、または分化させてもよく、本発明の分化培地は生存因子を含んでもよい。生存因子は、本発明の方法を使用して個別化した多能性幹細胞の生存および分化の効率を高めるために使用してもよい。いくつかの実施形態では、使用してもよい生存因子として、ミオシンIIのインヒビター、Rho非依存性キナーゼ(Rho−independent kinase)(ROCK)のインヒビターおよびプロテインキナーゼC(PKC)のインヒビターがあるが、これに限定されるものではない。ある種の実施形態では、TeSR1培地、TeSR2培地またはmTeSR培地を含む培養用培地に生存因子を加えてもよい。
本発明の分化培地は、栄養素、アミノ酸、抗生物質、緩衝剤および同種のものなど追加の構成要素をさらに含んでもよい。ある種の実施形態では、本発明の分化培地は、非必須アミノ酸、L−グルタミン、ペニシリン、ストレプトマイシンおよびモノチオグリセロールを含んでもよい。
Replacement 3(Sigma、カタログ番号S2640)を使用してもよい。他の実施形態では、規定された培地を必要としない実施形態では、BIT9500の代わりにKOSRを使用してもよい。KOSRは、市販されている(たとえばGibco/Invitrogen、カタログ番号10828)規定されていない培地であり、国際公開第98/30679号に以前記載された。
本発明の分化培地は、好ましくはフィブロネクチン、コラーゲンまたはRGDペプチドなど1種または複数種のマトリックス成分を用いて本明細書に記載の方法で使用する。どのような理論にも拘泥するわけではないが、マトリックス成分は、多能性幹細胞の増殖の固体支持体となり得る。好ましい実施形態では、マトリックス成分を培養面に適用し、培養培地および細胞と接触させる。
本発明の規定された細胞培養培地のマトリックス成分として、フィブロネクチンを使用してもよい。どのような理論にも拘泥するわけではないが、フィブロネクチンは、フィーダー細胞または胚様体を使用せずに増殖および分化させるための、ヒト胚性幹細胞のマトリックスとなり得る。
本発明の細胞培養培地のマトリックス成分としてコラーゲンを使用してもよい。コラーゲンは、結合組織の主要なタンパク質構成要素であり、組織および細胞を支持する細胞外マトリックスの主な構成要素である。どのような理論にも拘泥するわけではないが、コラーゲンは、フィブロネクチンの場合と同様、フィーダー細胞または胚様体を使用せずに増殖および分化させるための、ヒト胚性幹細胞のマトリックスとなり得る。コラーゲンは、本発明の分化培地に、たとえば、約0.5μg/cm2〜5μg/cm2または約1.5μg/cm2の濃度で加えてもよい。ある種の実施形態では、細胞を培養する表面をコーティングするためにコラーゲンを使用してもよい。ある態様では、開示された方法に有用なコラーゲンはコラーゲンIVである。
本発明の規定された細胞培養培地のマトリックス成分として、RGDペプチドを使用してもよい。RGDペプチドは、Arg−Gly−Asp(RGD)配列を含む接着タンパク質であり、ある種のRGDペプチドは、細胞の接着、遊走および増殖に重要な役割を果たし得る。どのような理論にも拘泥するわけではないが、RGDペプチドは、フィブロネクチンと同様、胚性幹細胞の分化および増殖を可能にする胚性幹細胞のマトリックス物質になり得る。ある種の実施形態では、合成RGDペプチドを本発明に使用してもよい。
本明細書では当該技術分野の慣習に従い(Ezashi et al.,2005)、周囲の酸素濃度を通常酸素濃度という。本明細書で使用する場合、「低酸素雰囲気」とは、約15〜25%酸素を含む周囲空気より酸素が少ない雰囲気をいう。好ましくは、低酸素雰囲気は、約5.5%未満酸素を含む。
「多能性」という用語は、細胞が、胚形成中に発生する3つの胚葉、外胚葉、内胚葉および中胚葉から生じる細胞型のいずれかに分化する能力という意味で細胞生物学の技術分野において一般に使用される。本明細書の「多能性細胞」および「多能性幹細胞」という用語は、本質的に任意のヒト胎児または成人細胞型に分化する能力を持つ細胞を説明するために使用される。例示的な多能性幹細胞型として、胚性幹細胞および誘導多能性幹細胞(すなわちiPS細胞)を挙げることができるが、これに限定されるものではない。本明細書で使用する場合、「胚性幹細胞」または「多能性幹細胞」という用語は、胚盤胞に自然発生する細胞、または胚盤胞に由来する細胞だけでなく、多能性になるように、または幹細胞様の状態に戻るように誘導された細胞をいう場合がある(たとえば、Nakagawa et al.,2007;Yu et al.,2007を参照されたい)。
et al.,2007;Takahashi et al.,2007;Nakagawa et al.,2007)。iPS細胞の作製方法は、当該技術分野において公知であり、本質的に体細胞を再プログラムする任意の適切な方法を使用し、本明細書に開示された方法に使用できる多能性幹細胞を作製すればよい。iPS細胞を作製する例示的な方法として、たとえば、どちらも参照によってその全体を本明細書に援用するThomson(米国特許出願公開第2008/0233610号)およびDaleyおよび共同研究者(米国特許出願公開第2009/0004163号)により開示された方法が挙げられる。
TeSR培地は、未分化ヒト多能性幹細胞の培養に使用してもよい規定された培地である。TeSRは、bFGF、LiCl、γ−アミノ酪酸(GABA)、ピペコリン酸およびTGFβを含むもので、TeSRを使用した様々な方法については、たとえば、参照によってその全体を本明細書に援用する米国特許出願公開第2006/0084168号およびLudwig et al.(2006a;2006b)などに以前に記載されている。「TeSR培地」という用語は、本明細書で使用する場合、TeSR1培地、TeSR2培地またはmTeSR培地を包含する。TeSR2培地(Stem Cell Technologies,Vancouver,BC,Canada)は、TeSR1培地と本質的に同一であり、TeSR1と同様、TeSR2培地もヒト化されている。本明細書に開示された方法にはTeSR1培地、TeSR2培地またはmTeSR培地を使用してもよい。
ヒト多能性幹細胞の培養および維持には様々なマトリックス成分を使用してもよい。参照によってその全体を援用するLudwig et al.(2006)に記載されているように、多能性細胞増殖の固体支持体を与える手段として培養面をコーティングするには、たとえば、コラーゲン、フィブロネクチン、ラミニンまたはビトロネクチンの1種または複数種を使用してもよい。一実施形態では、コラーゲンはコラーゲンIVである。
本発明に使用してもよい多能性幹細胞は、細胞培養の技術分野において公知の任意の方法を用いて播種培地に播種してもよい。たとえば、多能性幹細胞は、播種培地に単一のコロニーとして播種しても、またはクローン群として播種してもよいし、あるいは多能性幹細胞は、本質的に個別の細胞として播種してもよい。いくつかの実施形態では、当該技術分野において公知の機械的または酵素的方法を用いて、多能性幹細胞を本質的に個別の細胞に分離する。非限定的な例として、多能性幹細胞は、細胞間、および細胞と培養面との間の結合を破壊するタンパク質分解酵素と接触させてもよい。分化のため多能性幹細胞を個別化するのに使用してもよい酵素として、様々な市販処方物としてのトリプシン、trypLE(Invitrogen,Carlsbad,CAから市販されている安定なトリプシン様酵素)またはAccutase(登録商標)など酵素の混合物を挙げることができるが、これに限定されるものではない。
胚性幹細胞からの造血幹細胞および内皮前駆細胞の調製後、造血前駆細胞または内皮前駆細胞を精製すると望ましい場合がある。不均一な細胞集団から造血前駆細胞または内皮前駆細胞などの細胞のサブセットを分離または実質的に精製するには、FACSまたはMACSなどのフローサイトメトリーを使用した細胞の分離方法を用いてもよい。
造血細胞を分離するには、分化させたヒト胚性幹細胞(hESC:human embryonic stem cell)から磁気活性化細胞ソーター(MACS)を用いてCD34+またはCD43+細胞を分離すればよい。MACSは典型的には、カラムにおいて細胞を分離する磁性ビーズと組み合わせて抗CD34抗体などの抗体を使用する。ある種の実施形態では、MACSは、FACSに比べて細胞に対して穏やかであり、細胞の生存率および完全性に好ましい影響を与え得る。
CD34+造血細胞またはCD31+内皮細胞の分離には、蛍光活性化細胞ソーティング(FACS)を使用してもよい。当該技術分野において周知のように、FACSは、たとえば、細胞を分離するために結合された蛍光タグを含む抗CD34抗体により細胞が示す程度または蛍光を利用する。こうして、FACSを使用して不均一な細胞集団からCD34+造血細胞またはCD31+内皮細胞を分離することができる。
本発明では、造血前駆細胞を赤血球(erythrocyte)、顆粒球、マクロファージ、巨核球、樹状細胞およびマスト細胞などの細胞系統にさらに分化させるため、様々なアプローチを使用してもよい。これらのアプローチは、赤血球分化培地、メチルセルロース、および巨核球分化培地の使用を含んでもよい。ある種の実施形態では、造血前駆細胞を内皮細胞に分化させてもよいし、あるいは血管を作製するために使用してもよい。

situで光学顕微鏡法により行ってもよいし、またはコロニーを個別に採取してから細胞化学的方法および免疫細胞化学的方法により細胞を染色して行ってもよい。CFCアッセイには寒天、アガロース、メチルセルロース、コラーゲンおよびフィブリンの塊など様々なゲル化剤が使用されている。
造血前駆細胞は、たとえば、赤血球分化培地を用いて赤血球細胞に分化させてもよい。赤血球分化培地は、無血清培地でも、または規定された培地でもよく、培地はSCF、EPO、インスリン、デキサメタゾンおよび/またはトランスフェリンを含んでもよい(Slukvin et al.,2007)。
造血前駆細胞から赤血球(erythrocytes)、マクロファージおよび/または顆粒球を分化誘導するのにメチルセルロースを使用してもよい。メチルセルロースは、光学的透明度に優れた安定なゲルを形成する比較的不活性なポリマーである。メチルセルロースは一般に、ウシ胎仔血清(FBS:fetal bovine serum)、ウシ血清アルブミン(BSA)、2−メルカプトエタノール、インスリン、トランスフェリン、組換えサイトカインなどの化合物を補充した培養培地、またはコロニー刺激因子の供給源であるコンディション培地において約0.9〜1.2%の最終濃度で使用される。メチルセルロースによる細胞分化に関する方法については、たとえば、Kaufman et al.(2001)に記載されている。
巨核球の生成を誘導するには、巨核球分化培地を使用してもよい。巨核球の生成に関する様々な製品およびアプローチについては、たとえば国際公開第2006/050330号に記載されており、これを本発明に使用してもよい。さらに、Stem Cell Technologies(Vancouver,BC,Canada)からMegacultTMが市販されており、巨核球の生成/分化に使用してもよい。種々の実施形態では、巨核球分化培地にトロンボポエチン(thrombopoeitin)(TPO)、インターロイキン3(IL−3)、インターロイキン6(IL−6)、Flt−3リガンドおよび/または幹細胞因子を加えてもよい。細胞を巨核球分化する方法については、たとえば、Kaufman et al.(2001)に記載されている。
本明細書に記載の方法により得られたCD34+集団は、血液内皮(または血管芽細胞)および内皮前駆体をさらに含んでもよい。内皮細胞は、たとえば、以下のプロトコルにより作製することができ、動物またはヒト被験体への移植に使用してもよい。ヒトES細胞由来のCD34+細胞は、EGMTM−2培地(Lonza,Walkersville,MD)、あるいは50ng/mLのrhVEGFおよび5ng/mLのrhFGF−2を含む分化培地で7〜10日間培養すればよい。内皮細胞は、4℃、25mMのHepes(Sigma)緩衝EGMTM培地中のラットテイルコラーゲン、タイプ1(BD Biosciences,Bedford,MA)などのコラーゲン(1.5mg/mL)およびヒト血漿フィブロネクチン(90mg/mL)(Sigma)の約1mLの溶液に懸濁してもよい。pHは、1N NaOH(Fisher Science,NJ)を使用して7.4に調整すればよい。次いでこの細胞懸濁物を、12ウェルプレート(Falcon)にピペットで移し、30分間37℃に加温し、コラーゲンを重合させてもよい。凝固させた各ゲル構造物は、加温した1mLのEGM培地でカバーしてもよい。細胞は、5%CO2で約1日間培養してもよい。ある種の実施形態では、細胞を厚いMatrigelTM層内で増殖させ、内皮表現型のマーカーとなる管状構造の形成を調査して細胞が本当に内皮細胞であることを確認してもよい。
本発明の方法は、胚性幹細胞から内皮細胞を分化させるために使用してもよい。いくつかの実施形態では、本方法は、胚性幹細胞を内皮前駆細胞に分化させる最初のステップ、続いて内皮前駆細胞を分取し、さらに内皮細胞に分化させる追加のステップを含む。たとえば、幹細胞(hESCまたはiPS細胞など)は、本明細書に記載するように播種し、増殖させてもよい。いくつかの実施形態では、フィブロネクチンまたはコラーゲンコートプレートなどのマトリックス成分を使用してhESCまたはiPS細胞を播種する。ある種の実施形態では、少なくとも一部がマトリックス成分でコーティングされた固体支持層に細胞を播種する。細胞は、培養面1平方センチメートル当たり約10,000幹細胞から培養面1平方センチメートル当たり約80,000幹細胞で播種してもよい。特定の実施形態では、培養面1平方センチメートル当たり約20,000幹細胞から培養面1平方センチメートル当たり約70,000幹細胞の密度で細胞を播種する。細胞は、ブレビスタチンなどのミオシンIIインヒビターまたはH1152などのROCKインヒビターを含んでもよいTeSR培地で培養してもよい。
幹細胞の培養および/または多能性幹細胞からの造血前駆細胞および内皮前駆細胞の分化を行う1つまたは複数のステップは、自動化してもよい。ロボットまたは他の自動化によりプロセスを自動化すれば、細胞の作製、培養および分化の方法をより効率的かつ経済的なものにすることができる。たとえば、ロボットによる自動化は、その全体を参照によって本明細書に援用する米国特許出願公開第2009/0029462号に記載されているように利用してもよい。
本発明はさらに、本発明に従い使用されるキットも意図している。たとえば、キットは、1つまたは複数の密封バイアルに本明細書に記載の分化培地を含んでもよい。キットは、多能性幹細胞、前駆細胞、造血前駆細胞または内皮前駆細胞などの細胞を含んでもよい。
本発明は、多能性幹細胞の前駆細胞への分化を促進する能力について候補物質を見分けるのに有用なスクリーニングアッセイなどスクリーニングアッセイを意図している。
本発明はさらに、本明細書に開示された方法により得られた薬学的有効量の前駆細胞、造血細胞または内皮細胞を被験体に投与することにより疾患、障害または傷害を処置する方法も意図している。本発明によるこれらの組成物の投与は、その経路により標的組織に利用可能になるのであれば、一般に使用されるどのような経路により行ってもよい。これには、静脈内注射、脊髄内注射または脳内、皮内、皮下、筋肉内または腹腔内の方法など全身性または非経口の方法による投与が含まれる。投与は、治療の性質に応じ経口手段、経鼻手段、頬粘膜手段、直腸手段、経膣手段または局所手段により行ってもよい。
以下の例は、本発明の好ましい実施形態を説明するために含まれる。以下の実施例に開示された技法は、本発明者により発見された技法であり、本発明の実施の際に非常に役立つものであり、したがって本発明を実施するための好ましい態様になると考えられることを当業者は理解されたい。しかしながら、本開示に照らして、開示される個々の実施形態において多くの変更が可能であり、依然として本発明の精神および範囲を逸脱せずに同様または類似の結果が得られることを当業者は理解されたい。
(ヒト胚性幹細胞のCD34+造血細胞への規定された分化)
継代41代目のヒト胚性幹細胞をフィブロネクチンコートプレートに蒔き、TeSR培地で7日間培養した。7日目に、培地をTeSR培地からCD34分化培地に交換した。CD34分化培地は、表1にIMDMとして上述したものであり、BIT9500、BMP4、VEGF、bFGF、非必須アミノ酸、L−グルタミン、Pen−strepおよびモノチオグリセロールを含む。
(個別化された胚細胞のCD34+造血細胞への規定された分化)
どのような分化方法を実施しても、投入されるESのコロニーサイズおよびコロニー密度が変われば、成績に著しいばらつきが生じる可能性がある。したがって、分化を惹起する前にESコロニーの個別の細胞への分散について調査した。培養したES細胞の集団を分散させるか、あるいはトリプシンまたはTrypLEで個別化した。次いでROCKインヒビター(H1152、1μM)を使用して、あるいは使用せずに、分散させたES細胞をTeSR培地に播種した。個別化した培地を用いて本明細書に記載の分化手順を実施した。
TeSR培地のフィブロネクチンコート表面に蒔いた、個別化したES細胞は、ROCKインヒビター(たとえば、1uMのH1152)などの生存因子を添加しなければ、接着せず、生存できない。さらに、播種密度が低すぎると(約1×104細胞/cm2未満)、細胞はROCKインヒビターの存在下でも剥離し、生存能を失う。高すぎる細胞密度(5×104細胞/cm2超)で播種すると、細胞は接着した状態が続くが、血液内皮前駆体に分化できない。これらの上限と下限との間の最適な播種密度は、細胞増殖に使用する方法、継代数およびES細胞の全体的な状態によって異なる。

この分化方法の再現性および効率の改善を目的として、血液内皮前駆細胞の生成における低酸素状態を検討した。胚の増殖の非常に早い段階で低酸素がインビボで重要な役割を果たしていることは明らかになっている。心血管系が確立される前の哺乳動物の発生は、3%酸素環境で起こる。諸研究により、生理的低酸素状態が胚の血管形成および造血の重要な制御因子であり得ることが明らかになっている(Forsythe et al.,1996;Ramirez−Bergeron et al.,2004;Harrison et al.,2002;Cipolleschi et al.,1993)。

(材料および方法)
hESCまたはiPS細胞を20,000〜70,000細胞/cm2の密度でフィブロネクチン(3〜5μg/cm2)またはコラーゲンコートプレートに播種した。この細胞を、ROCKインヒビターH1152を含むTeSR1培地で増殖させた。細胞は、低酸素(low oxygen)インキュベーター(5%酸素)に一晩入れた。
この方法により得られた細胞は、内皮細胞の分子的および機能的特徴を示した。たとえば、この細胞はCD31を発現し、一生を通じてCD31を発現し続けた。また、細胞は、CD105(エンドグリン)およびvon Willebrand因子(第VIII因子とも呼ばれる)も発現した。さらに、細胞は、アセチル化LDLを取り込むこともできた。この細胞は機能的には、マトリゲルの厚い層に血管様チューブ構造を形成することができた。これらの結果から、使用した方法により内皮細胞が作製されることが示される。
