Abstract

a presente invenção refere-se a um método para padronizar a sensibilidade de neurônios derivados de células-tronco pluripotentes induzidas (ips) a um polipeptídeo de neurotoxina que compreende as etapas: a) cultivar diferentes lotes de neurônios derivados de células-tronco pluripotentes induzidas em um meio de cultura celular que compreende gt1b por pelo menos 3 horas; b) colocar os diferentes lotes de neurônios derivados de células-tronco pluripotentes induzidas da etapa a) em contato com um polipeptídeo de neurotoxina; c) cultivar os diferentes lotes de neurônios derivados de células-tronco pluripotentes induzidas da etapa b) por pelo menos 24 horas na presença de gt1b sob condições que permitem que o polipeptídeo de neurotoxina exerça sua atividade biológica e, assim, padronizar a sensibilidade dos neurônios derivados de células-tronco pluripotentes induzidas a um polipeptídeo de neurotoxina. a invenção refere-se, ainda, a um método para a geração de neurônios derivados de células-tronco pluripotentes induzidas que têm sensibilidade padronizada a um polipeptídeo de neurotoxina, que compreende as etapas: a) fornecer diferentes lotes de neurônios derivados de células-tronco pluripotentes induzidas; b) cultivar diferentes lotes de neurônios derivados de células-tronco pluripotentes induzidas da etapa a) em um meio de cultura celular que compreende gt1b por pelo menos 3 horas e, assim, padronizar a sensibilidade dos neurônios derivados de células-tronco pluripotentes induzidas a um polipeptídeo de neurotoxina. adicionalmente, é abrangido pela presente invenção um método para determinar a atividade biológica de um polipeptídeo de uma neurotoxina que compreende as etapas: a) cultivar neurônios derivados de células-tronco pluripotentes induzidas em um meio de cultura celular que compreende gt1b por pelo menos 3 horas; b) colocar os neurônios derivados de células-tronco pluripotentes induzidas da etapa a) em contato com um polipeptídeo de neurotoxina; c) cultivar os neurônios derivados de células-tronco pluripotentes induzidas da etapa b) por pelo menos 24 horas na presença de gt1b sob condições que permitem que o polipeptídeo de neurotoxina exerça sua atividade biológica; e d) determinar a atividade biológica do polipeptídeo de neurotoxina nas ditas células. finalmente, a invenção refere-se ao uso de gt1b para a) padronizar a sensibilidade de diferentes lotes de neurônios derivados de células-tronco pluripotentes induzidas a um polipeptídeo de neurotoxina; ou b) reduzir a variabilidade da sensibilidade de diferentes lotes de neurônios derivados de células-tronco pluripotentes induzidas a um polipeptídeo de neurotoxina.
