Abstract
Description
本実施形態に係るバリア機能改善剤は、アスチルビンを有効成分として含有するものである。
乾燥した黄杞葉部100gに10倍量の50容量%エタノールを加え、90℃で2時間還流抽出し、ろ過してろ液を得た。得られた抽出ろ液を減圧濃縮し、黄杞葉部抽出物(20.8g)を得た。得られた黄杞葉部抽出物20.0gに水を加え懸濁し、多孔性樹脂(ダイヤイオンHP−20,三菱化学社製)に付し、水200g、50容量%メタノール200g、メタノール200gの順で溶出させた。次いで、50容量%メタノール200gで溶出させた画分について溶媒留去および凍結乾燥を行い、黄杞葉部抽出物50%メタノール溶出画分(11.3g)を得た。

製造例で得られたアスチルビン(試料1)について、以下のようにして経上皮電気抵抗(transepithelial electrical resistance,TER)の値を測定した。
製造例で得られたアスチルビン(試料1)について、タイトジャンクション(TJ)構成因子のmRNA発現に対する影響を、以下のようにして評価した。
製造例で得られたアスチルビン(試料1)について、TJ構成因子のタンパク質発現に対する影響を以下のようにして評価した。
腫瘍壊死因子−α(TNF−α)やインターロイキン−6(IL−6)等の炎症性サイトカインは、消化管でのバリア機能に損傷を与えることが知られている。そこで、製造例で得られたアスチルビン(試料1)について、炎症性サイトカインによる消化管バリア機能の損傷に対し、どのように影響するのかを検討した。
また、IL−6(100ng/mL)についても、TNF−αと同様に試験を行い、TERを測定した。
結果を図3に示す。
Claims (2)
- アスチルビンを有効成分として含有することを特徴とするバリア機能改善剤。
- 前記アスチルビンが、タイトジャンクション機能強化作用、クローディン−1発現促進作用、オクルディン発現促進作用、およびZO−2発現促進作用からなる群より選択される1または2以上の作用を有することを特徴とする請求項1に記載のバリア機能改善剤。
