Description
発色性アッセイ(chromogenic assay)または1段階アッセイ(one stage assay)を用いて第VIII因子活性を測定し、第VIII因子の単位を国際単位(IU)で表した。
1段階凝固アッセイおよび/または発色性アッセイを用いて第IX因子の生物学的活性を測定し、現在のWHOの第IX因子濃縮物標準によって定義される国際単位(IU)で第IX因子の単位を表した。
HES−G−CSFのResource S HPLC分析
溶離液Aでサンプルを0.1mg/mLに希釈する。20μgをResource S 1mLカラム(GEヘルスケア社、ドイツ、ミュンヘン)に注入する。

組換え第VIII因子
実験に用いる第VIII因子は、欧州特許出願公開第1739179A号(Schroder et al)に記載のプロセスに従ってヒト細胞系HEK293F中で産生された組換えヒトBドメイン欠失第VIII因子タンパク質である。精製プロセスは、5つのクロマトグラフィーステップからなり、ヒトグリコシル化様パターン(Sandberg et al、国際出願第PCT/EP2009/060829号)を示す高純度の第VIII因子タンパク質標品が得られた(Winge et al、国際公開第2009/156430A号)。
これらの実験に用いる材料は、SD処理およびナノ濾過された高純度の第IX因子濃縮物である市販の製品Nanotiv(登録商標)に由来する。これらの実験に使用する前に、ゲル濾過カラムを用いて材料を更に精製し、第IX因子のモノマーピークをその後の実験に用いた。
用いる細胞系は、無血清増殖に適応させたヒト胚腎臓細胞293(HEK293)に由来する細胞系である。この宿主、HEK293Fを、G−CSFのcDNAコード配列を有する発現カセットで安定的にトランスフェクトした。カセットに強力なプロモーターを用いた。一般的なプロセスは欧州特許第1739179(Schroder et al)にも記載されている。
調製
上記実験セクションの記載に従って組換え第VIII因子(rFVIII)を調製した。この実験では、溶液中のrFVIIIに対するメレジトースの安定化効果を、一般的に用いられている安定化剤であるトレハロースと比較した。rFVIII濃度は100IU/mlとした。本実験で調べた製剤の組成を表1に示す。
調製
上記実験セクションの記載に従って組換え第VIII因子(rFVIII)を調製した。本実験では、凍結乾燥プロセス中および凍結乾燥製剤中でのメレジトースの安定化効果を、一般的に用いられている安定化剤であるトレハロースと比較した。rFVIII濃度は100IU/mlとした。本実験で調べた製剤の組成を表3に示す。
調製
上記実験セクションの記載に従って組換え第VIII因子(rFVIII)を調製した。本実験では、凍結乾燥プロセスおよび凍結乾燥製剤中における種々の濃度のメレジトースの安定化効果を比較し、更に安定化効果を四糖であるスタキオースと比較した。rFVIII濃度は170IU/mlでとした。本実施例で調べた製剤の組成を表5に示す。
調製
上記実験セクションの記載に従って血漿由来第IX因子(pFIX)を調製した。本実験は、pFIXに対するメレジトースの安定化効果を調べる。pFIX濃度は100IU/mlとした。本実験で調べた製剤の組成を表7に示す。
凍結乾燥ステップのタンパク質回収率は約100%であった。安定性試験の結果を表8に示す。
調製
上記実験セクションの記載に従って組換えHES化G−CSF(rHES−G−CSF)を調製した。本実験では、凍結乾燥形態のrHES−G−CSFに対するメレジトースの安定化効果を、一般的に用いられている安定化剤トレハロースと比較した。rHES−G−CSF濃度は0.3mg/mlとし、調べた製剤の組成を表9に示す。
調製
上記実験セクションの記載に従って、血漿由来第IX因子(pFIX)を調製した。本実験では、四糖であるスタキオースと比べた凍結乾燥ステップでのpFIXに対するメレジトースの安定化効果を調べた。pFIX濃度は100IU/mlとした。本実験で調べた製剤の組成を表11に示す。
凍結乾燥ステップのタンパク質回収率は、製剤6Aでは約100%であったが、製剤6Bでは84%であった。
Claims (23)
- 分子量が10KDaを超えるヒト血漿タンパク質を含む溶液に、前記ヒト血漿タンパク質を安定化する量のメレジトースを添加することにより、前記分子量が10KDaを超えるヒト血漿タンパク質を安定化する方法であって、前記ヒト血漿タンパク質が、第VII因子、第VIIa因子、第IX因子、第VIII因子、およびHES化G−CSF、ならびにその誘導体および変異タンパク質からなる群から選択される、薬学的または生物学的意義のあるヒト血漿タンパク質または組換えにより生成されたヒト血漿タンパク質である、方法。
- 前記溶液を固体状態に転移させる、請求項1に記載の方法。
- 凍結乾燥により前記溶液を固体状態に転移させる、請求項2に記載の方法。
