Claims (14)
- 生体試料中のドナー特異的抗体の存在または不存在を判定する方法であって、
レシピエント免疫抗体が存在するならば、これがドナー細胞表面抗原(Ag)と結合して免疫抗体−Ag複合体を形成するのに十分な条件下で、ドナーからの細胞試料をレシピエントからの生体試料と混合することによって混合物を形成する工程、
前記混合物を、前記免疫抗体−Ag複合体と特異的に結合する抗体をその表面に有するビーズと接触させる工程、
前記ビーズに結合した前記免疫抗体−Ag複合体と特異的に結合する検出可能標識抗体を溶解条件下で添加する工程、および
前記免疫抗体−Ag複合体と結合した前記検出可能標識抗体の存在または不存在を検知して前記レシピエントからの生体試料中のドナー特異的抗体の存在または不存在を判定する工程を含む前記方法。 - 前記検知が半定量的である、請求項1に記載の方法。
- 前記免疫抗体が同種抗体、自己抗体、または補体結合抗体(CFAb)である、請求項1または2に記載の方法。
- 前記検知が、フローサイトメーター中に前記複合体を流すこと、または酵素結合免疫吸着検査法(ELISA)によって前記複合体を検出することを含む、請求項1から3のいずれか一項に記載の方法。
- 前記検出可能な標識が蛍光色素、発色団、酵素、リンカー分子、ビオチン分子、電子供与体、電子受容体、染料、金属、または放射性核種を含む、請求項1から4のいずれか一項に記載の方法。
- 前記ドナー特異的抗体が実際のドナー特異的抗体である、請求項1から5のいずれか一項に記載の方法。
- 前記AgがHLA−A、HLA−B、HLA−C、HLA−DRB1、HLA−DRB2、HLA−DRB3、HLA−DRB4、HLA−DRB5、HLA−DQ、およびHLA−DPから成る群から選択される、請求項1から6のいずれか一項に記載の方法。
- 前記生体試料が血清、血液、唾液、または血漿を含む、請求項1から7のいずれか一項に記載の方法。
- 前記ドナーからの細胞試料が0.001×106〜2.0×106個の細胞を含む、請求項1から8のいずれか一項に記載の方法。
- 前記ドナーからの細胞試料が0.2×106個未満の細胞を含む、請求項1から9のいずれか一項に記載の方法。
- 前記ビーズがアガロースビーズ、ラテックスビーズ、磁気ビーズ、またはポリスチレンビーズである、請求項1から10のいずれか一項に記載の方法。
- 前記方法が12時間以下で実施される、請求項1から11のいずれか一項に記載の方法。
- 前記溶解条件がトレーサ、洗浄剤、およびDNアーゼを含む溶解緩衝液を与えることを含む、請求項1から12のいずれか一項に記載の方法。
- ドナー特異的抗体が前記レシピエントからの前記生体試料に存在するか示す報告を作成することを含む、請求項1から13のいずれか一項に記載の方法。
