Abstract
Description
「インターロイキン−33」、「IL−33」、など、本明細書中で使用する場合、配列番号307番のアミノ酸配列を有するヒトIL−33タンパク質をいう。本明細書でタンパク質、ポリペプチド及びタンパク質断片という場合はいずれも、ヒト種でないことが明確に指示されていない限り(例えば、「マウスIL−33」、「サルIL−33」など)、それぞれヒトのタンパク質、ポリペプチド又はタンパク質断片を指すことを意図する。
−CH1−CH2;(xii)VL−CH1−CH2−CH3;(xiii)VL−CH2−CH3;及び(xiv)VL−CL。上記に挙げた例示的な立体配置を含む、可変及び定常ドメインの任意の配置では、可変及び定常ドメインは、互いに直接結合しあっても、又は、完全もしくは部分的ヒンジ領域もしくはリンカー領域により結合しても、どちらでもよい。ヒンジ領域は、少なくとも2個(例えば、5、10、15、20、40、60又はそれ以上)のアミノ酸で構成されてよく、隣接する可変領域及び/又は定常ドメイン間に単一ポリペプチド分子で柔軟又は半柔軟な結合をする。さらに、本発明の抗体の抗原結合性断片は、互いに及び/又は1つ以上の単量体のVHもしくはVLドメインと非共有的に会合した(例えば、ジスルフィド結合(複数可))、上記の可変及び定常ドメイン立体配置のいずれかのホモ二量体又はヘテロ二量体(又は他の多量体)を含んでよい。
本発明は、ヒトIL−33に結合し、IL−33媒介性シグナル伝達を抑制する又は減弱させる、抗IL−33抗体及びその抗原結合性断片を含む。抗IL−33抗体が「IL−33媒介性シグナル伝達を抑制する又は減弱させる」とみなされるのは、例えば、抗体が、(1)細胞ベースのバイオアッセイにおけるIL−33媒介性シグナル伝達の抑制;(2)IL−33誘発性のヒト好塩基球脱顆粒の抑制;(3)ヒトPBMCによるIL−33誘発性IFNγ産生の抑制;(4)哺乳類においてアレルゲン、例えば、IL−4又はIL−5に暴露されると上昇する、サイトカインレベルの低下;並びに(5)アレルゲン(例えば、チリダニ(HDM)への急性又は慢性的暴露で生じた肺炎症の抑制)からなる群から選択される1つ以上の性質を示す場合とする。
本発明のある実施形態によれば、FcRn受容体への抗体の結合性を、例えば、中性pHに比して酸性pHで高める又は減退させる、1つ以上の変異を含むFcドメインを含む、抗IL−33抗体を提供する。例えば、本発明は、FcドメインのCH2又はCH3領域における変異を含む抗IL−33抗体を含み、これにおいて、変異(複数可)により酸性環境(例えば、pH範囲約5.5〜約6.0のエンドソームで)におけるFcドメインのFcRnに対する親和性が高まる。こうした変異により、動物に投与した場合、抗体の血清中半減期が長くなり得る。このようなFc改変の非限定的な実施例には、例えば、250位(例えば、E又はQ);250及び428位(例えば、L又はF);252位(例えば、L/Y/F/W又はT)、254位(例えば、S又はT)、並びに256位(例えば、S/R/Q/E/D又はT)での改変;又は428位及び/又は433位(例えば、H/L/R/S/P/Q又はK)及び/又は434位(例えば、H/F又はY)での改変;又は250及び/又は428位での改変;又は307もしくは308位(例えば、308F、V308F)、及び434位での改変が含まれる。ある実施形態では、改変には、428L(例えば、M428L)及び434S(例えば、N434S)の改変;428L、259I(例えば、V259I)、及び308F(例えば、V308F)の改変;433K(例えば、H433K)及び434(例えば、434Y)の改変;252、254、及び256(例えば、252Y、254T、及び256E)の改変;250Q及び428Lの改変(例えば、T250Q及びM428L);並びに307及び/又は308の改変(例えば、308F又は308P)が含まれる。さらに別の実施形態では、改変には、265A(例えば、D265A)及び/又は297A(例えば、D297A)の改変が含まれる。
本発明は、IL−33の1個以上のアミノ酸と相互作用する抗IL−33抗体を含む。例えば、本発明は、IL−33のST2相互作用ドメイン内に位置する1個以上のアミノ酸と相互作用する抗IL−33抗体を含む。抗体が結合するエピトープは、IL−33の3つ以上の(例えば、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20以上の)アミノ酸の単一連続配列で構成されてよい。別の方法では、エピトープは、IL−33の複数の非連続アミノ酸(又はアミノ酸配列)で構成されてよい。
Methods Mol Biol 248:443−463)、及びペプチド開裂解析が含まれる。さらに、抗原のエピトープ切り出し(epitope excision)、エピトープ抽出及び化学的な修飾などの方法を使用することができる(Tomer, 2000, Protein Science 9:487−496)。抗体が相互作用するポリペプチド内のアミノ酸を同定するために使用できる別の方法は、水素/重水素交換を質量分析法で検出する方法である。一般的な用語で言うと、水素/重水素交換方法では、対象とするタンパク質を重水素で標識した後、その重水素標識タンパク質に抗体を結合させる。次に、タンパク質/抗体複合体を水に移し、抗体で保護された残基(重水素標識されたまま)を除くすべての残基で水素−重水素交換を起こさせる。抗体解離後、標的タンパク質をプロテアーゼで切断し質量分析を行うことで、重水素標識されている残基がわかれば、それが抗体が相互作用する特定のアミノ酸ということになる。例えば、Ehring(1999)Analytical Biochemistry 267(2):252−259; Engen and Smith(2001)Anal.Chem.73:256A−265Aを参照のこと。
完全ヒトモノクローナル抗体を含めたモノクローナル抗体の作製方法は当該技術分野で知られている。本発明の文脈においてヒトIL−33に特異的に結合するヒト抗体を作製するために、そのような既知のあらゆる方法を使用できる。
本発明の抗IL−33抗体及び抗体断片は、記載の抗体のアミノ酸配列とは異なるがヒトIL−33に結合する能力を保有するアミノ酸配列を有するタンパク質を包含する。このような改変抗体及び抗体断片は、親配列と比較した場合にアミノ酸の付加、欠失、又は置換を1つ以上含んでいるが、記載の抗体の生物学的活性と本質的に同等な生物学的活性を示す。同様に、抗IL−33抗体をコードする本発明のDNA配列は、開示の配列と比較した場合にヌクレオチドの付加、欠失、又は置換を1つ以上含むが、本質的に本発明の抗IL−33抗体又は抗体断片と生物学的に同等である抗IL−33抗体又は抗体断片をコードする配列を包含する。このような改変アミノ酸及びDNA配列の例については上述している。
ある実施形態によれば、本発明は、ヒトIL−33には結合するが他の種のIL−33には結合しない抗IL−33抗体を提供する。本発明はまた、ヒトIL−33及び1種以上の非ヒト種からのIL−33に結合する抗IL−33抗体を含む。例えば、本発明の抗IL−33抗体は、ヒトIL−33に結合してよく、また、マウス、ラット、モルモット、ハムスター、スナネズミ、ブタ、ネコ、イヌ、ウサギ、ヤギ、ヒツジ、ウシ、ウマ、ラクダ、カニクイザル、マーモセット、霊長類又はチンパンジーの1種以上のIL−33に、場合に応じて、結合しても結合しなくてもよい。本発明のある例示的な実施形態によれば、ヒトIL−33及びカニクイザル(例えば、Macaca fascicularis種)のIL−33に特異的に結合する抗IL−33抗体を提供する。
本発明は、細胞毒素、化学療法薬、免疫抑制剤又は放射性同位元素などの治療的部分(「免疫複合体」)に抱合させた抗IL−33モノクローナル抗体を包含する。細胞障害剤には細胞に有害な任意の薬剤が含まれる。免疫複合体を形成するための好適な細胞障害剤及び化学療法薬の例は、従来技術において既知である(例えば、国際公開第05/103081号を参照のこと)。
本発明の抗体は、単一特異性、二重特異的、又は多重特異性であってよい。多重特異性抗体は、1つの標的ポリペプチドの異なるエピトープに特異的であってよく、又は、2つ以上の標的ポリペプチドに特異的な抗原結合ドメインを含有してよい。例えば、Tutt et al., 1991, J.Immunol.147:60−69; Kufer et al., 2004, Trends Biotechnol.22:238−244を参照のこと。本発明の抗IL−33抗体を、別の機能性分子、例えば、別のペプチド又はタンパク質と結合又は共発現させてよい。例えば、抗体又はその断片を、別の抗体又は抗体断片など1つ以上の他の分子種(molecular entities)に機能的に結合させ(例えば、化学的共役、遺伝子融合、非共有性会合又はその他により)、第2の結合特異性を有する二重特異性又は多重特異性抗体を作製することができる。例えば、本発明は、免疫グロブリンの1本の腕はヒトIL−33又はその断片に対して特異的であり、免疫グロブリンのもう一方の腕は第2の治療標的に対して特異的である又は治療的部分に抱合される、二重特異性抗体を含む。
本発明は、IL−33にpH依存的に結合する抗体及びその抗原結合性断片を提供する。例えば、本発明の抗IL−33抗体は、中性pHに比して酸性pHでIL−33への低い結合性を示してよい。別の方法では、本発明の抗IL−33抗体は、中性pHに比して酸性pHで、その抗原への高い結合性を示してよい。
抗体のKD値の比(又はその逆)で表される。例えば、抗体又はその抗原結合性断片は、抗体又はその抗原結合性断片が約3.0以上の酸性/中性KD比を示す場合は、本発明の目的の「中性pHに比して酸性pHでのIL−33への低い結合性」を示すと考えてよい。ある例示的な実施形態では、本発明の抗体又は抗原結合性断片の酸性/中性KD比は、約3.0、3.5、4.0、4.5、5.0、5.5、6.0、6.5、7.0、7.5、8.0、8.5、9.0、9.5、10.0、10.5、11.0、11.5、12.0、12.5、13.0、13.5、14.0、14.5、15.0、20.0,25.0、30.0、40.0、50.0、60.0、70.0、100.0又はそれ以上であり得る。
本発明は、本発明の抗IL−33抗体又はその抗原結合性断片を含む医薬組成物を提供する。本発明の医薬組成物を、移行、送達、忍容性などで改善が得られる好適な担体、賦形剤などの薬剤とともに処方する。多数の適切な処方物を、すべての薬剤師に公知のRemington’s Pharmaceutical Sciences、Mack Publishing Company、Easton、PAに見出すことができる。これらの処方物には、例えば、粉末、ペースト、軟膏、ゼリー、ワックス、油、脂質、小胞含有脂質(陽イオン又は陰イオン)(LIPOFECTIN(商標)、Life Technologies、カリフォルニア州カールスバッド)、DNAコンジュゲート、無水吸収ペースト、水中油型及び油中水型エマルジョン、エマルジョンカーボワックス(さまざまな分子量のポリエチレングリコール)、半固体ゲル、及びカーボワックス含有半固体混合物が含まれる。Powell et al.”Compendium of excipients for parenteral formulations” PDA(1998)J Pharm Sci Technol 52:238−311も参照のこと。

本願発明者らが実施したマウスモデル系を使用する実験は、IL−33拮抗により治療、予防及び/又は改善が可能なさまざまな疾患及び病態の同定に寄与した。例えば、マウスIL−33DNAの流体力学的送達により、肺粘液蓄積を誘導しマウスでの血清総IgEを上昇させた。さらに、mIL−33DNA送達により、マイクロアレイ解析で測定したところ、ST2及び下流のさまざまなサイトカインが上方制御された。本願発明者らがIL−33ノックアウトマウスを使用して実施した実験でも、IL−33拮抗によりさまざまな治療上の利益が得られる可能性が高いことが明らかとなった。例えば、巨視的スコアリング及び皮膚浸潤は、IMQ誘導性乾癬モデルの野生型マウスとIL−33-/-マウスの間で比較できることがわかった。さらに、IL−33-/-マウスは、アレルゲン誘導性肺炎症モデルにおいて好酸球増加症及び残留粘液蓄積の低下を示した。
本発明は、本明細書に記載する抗IL−33抗体を1つ以上のさらなる治療的活性成分と組み合わせることを含む組成物及び治療的処方物、及びかかる組み合わせをそれを必要とする被検者に投与することを含む治療方法を含む。
本発明のある実施形態によれば、抗IL−33抗体(又は抗IL−33抗体と本明細書で言及する任意のさらなる治療的活性薬剤との組み合わせを含む医薬組成物)の複数用量をある一定期間被験者に投与してよい。本発明のこの態様による方法は、複数用量の本発明の抗IL−33抗体を被験者に順次投与することを含む。本明細書中で使用する場合、「順次投与する」とは、抗IL−33抗体の各用量を被検者に異なる時間ポイントで、例えば、予め設定された間隔(例えば、時間、日、週又は月)で区切った異なる日に投与することを意味する。本発明は、患者に単一の初回用量で抗IL−33抗体投与後、1つ以上の第2用量で抗IL−33抗体を投与し、必要に応じ、さらに1つ以上の第3用量で抗IL−33抗体を順次投与することを含む方法を含む。
本発明の抗IL−33抗体を、IL−33、又は試料中のIL−33発現細胞を検出及び/又は測定するため、例えば、診断するために使用することもできる。例えば、抗IL−33抗体、又はその断片を、IL−33の異常発現を特徴とする病態又は疾患を診断するため(例えば、過剰発現、低発現、発現の欠如など)に使用してよい。IL−33の例示的な診断アッセイは、例えば、患者から得た試料を本発明の抗IL−33抗体と接触させることを含んでよく、ここでは、抗IL−33抗体を検出可能標識又はレポーター分子で標識する。別の方法では、未標識抗IL−33抗体を、それ自体が検出可能に標識された第2抗体と組み合わせて診断用途に使用できる。検出可能標識又はレポーター分子は、3H、14C、32P、35S、又は125Iなどの放射性同位元素;フルオレセインイソチオシア
ネート、又はローダミンなどの蛍光又は化学発光部分;又はアルカリホスファターゼ、βガラクトシダーゼ、ホースラディッシュペルオキシダーゼ、又はルシフェラーゼなどの酵素であり得る。試料中のIL−33を検出又は測定するために使用可能な具体的試験法の例には、酵素結合免疫吸着法(ELISA)、ラジオイムノアッセイ(RIA)、及び蛍光活性化細胞分類(FACS)を含む。
以下の実施例は、本発明の方法及び組成物をいかに使用し作製するかについての開示及び説明を完全に当業者に提供するために提案するものであり、本発明者らが該発明者らの発明であるとみなす範囲を限定することは意図していない。使用する数字に関し、正確さを保証するべく試みがなされた(例えば、量、温度など)が、一部の実験上の誤差及び偏差は考慮されるべきである。特に指定しない限り、部は重量部、分子量は平均分子量、温度はセ氏、及び圧力は大気圧又はその近傍圧である。
ヒトIL−33に対するヒト抗体の作製
ヒトIL−33を含む免疫原を、免疫応答を刺激するためにアジュバントとともに、ヒト免疫グロブリンの重鎖及びκ軽鎖可変領域をコードするDNAを含む、VELOCIMMUNE(登録商標)マウスに直接投与した。抗体免疫応答をIL−33特異的イムノアッセイにより監視した。所望の免疫応答が達成されたら、脾細胞を回収してマウス骨髄腫細胞と融合させ、細胞の活動性を保存するとともにハイブリドーマ細胞株を形成した。IL−33特異性抗体を産生する細胞株を同定するために、ハイブリドーマ細胞株をスクリーニングし選択した。この手法を使用し、抗IL−33キメラ抗体(すなわち、ヒト可変ドメイン及びマウスの定常ドメインを有する抗体)を幾つか得た。この方法で作製した例示的抗体を、H1M9559N、H1M9566N、H1M9568N及びH1M9565Nという名称にした。キメラ抗体から得たヒト可変ドメインをその後ヒト定常ドメイン上にクローニングし、本明細書に記載する完全ヒト抗IL−33抗体を作製した。
重鎖及び軽鎖の可変領域アミノ酸配列
表1に、重鎖及び軽鎖の可変領域アミノ酸配列対、及びCDR配列、選択した抗IL−33抗体とその対応する抗体識別名称を記載する。
表面プラズモン共鳴で測定した抗体のヒトIL−33への結合性
精製抗IL−33モノクローナル抗体に結合するIL−33の平衡解離定数(KD値)を、Biacore4000instrumentを使用しリアルタイム表面プラズモン共鳴バイオセンサーで測定した。Biacoreセンサー表面を、抗マウスのウサギ・ポリクローナル抗体(GE、#BR−1008−38)又は抗ヒトFcマウス・モノクローナル抗体(GE、#BR−1008−39)いずれかとのアミンカップリングにより最初に誘導体化し、それぞれ、マウス又はヒトIgG4定常領域に発現した抗IL−33モノクローナル抗体を捕捉した。Biacore結合試験はすべて、0.01M ADA pH7.4、0.15M NaCl、3mM EDTA、及び0.05%v/v界面活性剤Tween−20(ABS−ETランニング緩衝液)で実施した。ABS−ETランニング緩衝液(100nM〜3.7nMの範囲で、3倍希釈)に調製した、各種濃度のヒトIL−33(hIL−33;R&D Systems、#3625−IL−010/CF)又はC末端ヘキサヒスチジンタグとともに発現されるカニクイザルIL−33(MfIL−33−6His;配列番号xx番)を、抗IL−33モノクローナル抗体捕捉表面上に流量30μL/分で注入した。hIL−33又はMfIL−33−6Hisと捕捉モノクローナル抗体との会合を4分間監視し、ABS−ETランニング緩衝液内での解離について10分間監視した。pHを低くしたことによる各抗IL−33抗体とhIL−33又はMfIL−33−6Hisとの結合への影響を、インラインpH追跡(in−ライン pH chase)試験フォーマットを用い、0.01M ADA pH6.0、0.15M NaCl、3mM EDTA、及び0.05%v/v界面活性剤Tween−20(ABS−ET pH6緩衝液)で試験した。これを達成するため、hIL−33又はMfIL−33−6Hisのいずれかと捕捉モノクローナル抗体との会合をABS−ETランニング緩衝液内で4分間監視した。hIL−33又はMfIL−33−6HisのいずれかをABS−ETランニング緩衝液中で30秒解離させた後、ABS−ET pH6緩衝液を3分間注入し、低pH条件下でのアナライトの解離を測定した。結合動態実験を25℃及び37℃の両温度で実施した。動態の会合(ka)及び解離(kd)定数は、Scrubber2.0c curve fittingソフトウェアを使用し、リアルタイム・センサーグラムを1:1結合モデルに当てはめて測定した。結合解離平衡定数(KD)及び解離半減期(t1/2)を以下の動態速度定数としての計算した:

抗IL−33抗体はヒトST2受容体へのIL−33の結合を遮断する
ヒトIL−33(hIL−33)又はヒトST2受容体に結合するカニクイザルIL−33 に対する抗IL−33抗体の遮断能力を、競合サンドイッチELISAを使用して測定した。C末端ヒトIgG1Fcタグ(hST2−hFc;配列番号306番)とともに発現した、ヒトST2タンパク質の細胞外ドメインの一部分を、PBS緩衝液中濃度1(g/mLで96ウェルマイクロタイタープレート上に4℃で一晩コーティングした。続いて、非特異的結合部位をBSAの0.5%(w/v)PBS溶液を用いてブロッキングした。一定濃度の30pMビオチン化hIL−33タンパク質(R&D Systems、Cat#3625−IL/CF)(ビオチン−hIL−33)又はヘキサヒスチジンタグとともに発現させた(MfIL−33−6His;配列番号305番)150pMカニクイザルIL−33いずれかを、抗体の最終濃度が0〜100nMの範囲になるよう抗体の系列希釈に別々に添加した。抗体/IL−33混合物は、室温にて1時間インキュベートしてから、hST2−hFcコーティングしたマイクロタイタープレートに移した。室温にて1時間インキュベートした後、ウェルを洗浄し、プレートに結合したビオチン−hIL−33を、ホースラディッシュペルオキシダーゼ(HRP)と抱合させたストレプトアビジン(Thermo Scientific, Cat#N200)で検出し、プレートに結合したMfIL−33−6Hisは、抗Hisモノクローナル抗体(Qiagen、#34460)と抱合させたHRPで検出した。全試料をTMB溶液(BD Biosciences、#51−2607KC)と反応させて呈色反応を起こさせ、次いで、1M硫酸による酸性化で反応を停止させてから、Victor X5マイクロプレートリーダーで450nmの吸光度を測定した。データ解析を、sigmoidal dose−response model within Prism(商標)ソフトウェアを使用して実施した。IC50値は、プレートにコーティングしたhST2−hFcに結合するビオチン−hIL−33又はMfIL−33−6Hisを、最高シグナルから50%まで低下させるのに必要とされる抗体濃度と定義され、この計算値を遮断力価の指標として用いた。遮断パーセントは、IL−33単独のシグナルとバックグラウンド(HRPで抱合させた第2抗体又はストレプトアビジン単独の時のシグナル)との差に対する、抗体存在下で観察されたシグナル低下の比率として計算した。一定濃度のビオチン−hIL−33又はMfIL−33−6His単独について測定した吸光度を、0%遮断と定義し、IL−33未添加について測定した吸光度を100%遮断と定義する。各抗体の最高濃度含有ウェルの吸光度値を、最大遮断パーセントを測定するために使用した。
Biacore解析により示される、抗IL−33モノクローナル抗体に結合するIL−33のST2による抑制
抗IL−33抗体の、予め形成したIL−33とST2との複合体への結合能力を、リアルタイム表面プラズモン共鳴バイオセンサーが付属したBiacore T−200 instrumentを使用して試験した。実験は、0.01M HEPES pH7.4、0.15M NaCl、3mM EDTA、及び0.05%v/v界面活性剤Tween−20(HBS−ET)で構成されるランニング緩衝液を用いて25℃にて実施した。Biacoreセンサー表面を、抗mycタグ特異的モノクローナル抗体とのアミンカップリング法により最初に誘導体化し(クローン#9E10)、この誘導体化されたセンサー上で、C末端myc−myc−ヘキサヒスタジンタグとともに発現させたヒトST2タンパク質(hST2−MMH;配列番号323番)をおよそ160応答単位(RU)捕捉した。次いで、捕捉したhST2−MMH表面を、ヒトIL−33(hIL−33;R&D Systems、#3625−IL−010/CF)100nMを3分間注入することにより飽和状態にし、その後、抗IL−33モノクローナル抗体溶液100nMを3分間注入した。リアルタイム結合応答を実験中の全期間監視し、抗IL−33抗体を予め形成したhIL−33と捕捉したhST2−MMHとの複合体に注入してから3分後に観察された結合応答を記録し、表にまとめ表7に示した。抗IL−33モノクローナル抗体の抗mycタグ捕捉表面への非特異的結合は観察されなかった。表7に示すように、17の被験抗体は、予めhST2−MMHと複合体を作製した後、hIL−33への測定可能な結合を示さなかったが、3抗体(H1M9565N、H1M9566N、及びH1M9568N)は予めhST2−MMHと複合体を作製した後でhIL−33に結合した。
抗IL−33抗体によるIL−33媒介性受容体シグナル伝達の抑制
インターロイキン−33(IL−33)は、付属タンパク質IL−1RAcPと会合する、toll様/インターロイキン−1受容体スーパーファミリー・メンバーであるST2のリガンドである(総説は、Kakkar and Lee, 2008を参照のこと)。IL−33によりST2/IL−1RAcPが活性化されると、MyD88(ミエロイド系分化因子88)及びTRAF6(TNF受容体関連因子6)などの下流分子を介してシグナル伝達カスケードが誘発され、特にNFκB(核因子−κB)の活性化を引き起こす。抗IL−33抗体を試験する、生物学的に関連するバイオ試験系を作成するため、ヒト胚性腎細胞(HEK293)を安定に移入させ、ヒトST2(NP_057316、accession番号1−556のアミノ酸)をルシフェラーゼ・レポーターとともに発現させた[NFκB応答配列(5x)−ルシフェラーゼ−IRES−GFP](HEK293/hST2/NFkB−ルシフェラーゼ細胞株)。HEK293細胞株はIL−1RAcPを内因的に発現し、HEK293細胞のIL−33によるNFκB活性化がこれまでに示されている(Schmitz et al., Immunity 23:479−490(2005))。安定な細胞株を単離し、10%FBS、DMEM、NEAA、ペニシリン/ストレプトマイシン、及びG418に維持した。
抗IL−33抗体によるIL−33誘発性ヒト好塩基球脱顆粒の抑制
本発明の選択抗IL−33抗体の特徴インビトロでさらに評価するために、IL−33誘発性好塩基球脱顆粒を遮断する能力を測定した。末梢血単核細胞(PBMC)を異なるヒトの提供者2人の新鮮全血から密度勾配遠心により精製した。K2 EDTA全血を、RPMI1640に1:1で希釈し、Ficoll−Paque(GE Healthcare、#17−1440−03)上に慎重に置き、遠心分離してPBMCを分離した。PBMCを含有する間期層を吸引し、新しい管に移し、MACS BSA溶液(Militenyi Biotec、#130−091−376)とMACSリンス溶液(Militenyi Biotec、#130−091−222)を1:20で希釈して構成したMACS緩衝液で2回洗浄した。その後、精製したPBMCを、MACS緩衝液100μL中約3.0x106細胞/mLの最終濃度でV字底96ウェルプレートに置いた。好塩基球をPBMC母集団に含有させるため、Ca++もMg++も含まないDulbeccoのリン酸緩衝生理食塩水(DPBS)50μLにIL−3(Sigma、#H7166−10UG)1ngを入れたものを細胞懸濁液に加えた後、37℃で10分間インキュベートした。
抗IL−33抗体による、IL−33誘発性のヒトPBMCからのIFN−γの抑制
本発明の抗IL−33抗体の特徴をさらに明らかにするため、末梢血単核細胞(PBMC)を使用する一次細胞ベースアッセイを用いた。本実施例で使用したアッセイは、Smithgall et al.in International Immunology, 2008, vol.20(8)pp.1019−1030により発表されている結果を基にした。このアッセイ用に、異なる提供者3人の新鮮全血から得たPBMCを密度勾配遠心により精製した。簡単に言うと、K2 EDTA全血をRPMI1640で2倍希釈し、Ficoll−Paque(GE Healthcare、#17−1440−03)上に慎重に置いて20分間遠心分離した。PBMCを含有する間期層を吸引し、新しい管に移してPBSで2回洗浄した。単離したPBMCを、10%FBS、L−グルタミン2mM、ペニシリン100U/mL、及びストレプトマイシン100μg/mLを添加したRPMI1640に最終濃度5x105細胞/mLで丸底96ウェルプレートに置いた。次いで、細胞を50g/mLのヒトIL−12(hIL−12;R&D Systems、#219−IL−025/CF)及び10nM〜0.64pMのヒトIL−33(hIL−33;R&D Systems、#3625−IL−010/CF)単独を系列希釈したものとともに、又は260pMのhIL−33と100nM〜6.4pMの各種抗体を系列希釈したものとの組み合わせとともにインキュベートした。最終容積はウェルあたり200μLであった。各条件を3連で試験した。抗体を存在させた場合は、それらを予めhIL−33とインキュベーションしてから30分後に細胞に加えた。
バイオレイヤー干渉法を使用したヒトIL−33交差競合
パネルの種々の抗IL−33モノクローナル抗体間の結合競合を、Octet(登録商標)HTXバイオセンサー(ForteBio、A Division of Pall Life Sciences)を使用してリアルタイム、非標識バイオレイヤー干渉法で測定した。実験は、0.01M HEPES pH7.4、0.15M NaCl、0.05%v/v界面活性剤Tween−20、及び0.1mg/ml BSA(HBS−ET kinetics buffer)緩衝液を使用し、プレートを速度1000rpmで撹拌しながら25℃にて実施した。2抗体がヒトIL−33への結合について互いに競合することができたかどうかを評価するため、予備混合アッセイフォーマットを使用し、これにおいては、100nMのヒトIL−33(R&D Systems;#3625−IL−010/CF)と500nMの異なる抗IL−33モノクローナル抗体(以後、mAb−2と呼ぶ)とを、結合競合アッセイに先立ち少なくとも2時間予備混合した。抗マウスFcポリクローナル抗体(Pall ForteBio Corp、#18−5088;以後、AMCと呼ぶ)又は抗ヒトFcポリクローナル抗体(Pall ForteBio Corp、#18−5060;以後、AHCと呼ぶ)いずれかでコーティングしたOctetバイオセンサーを、まず、個々の抗IL−33モノクローナル抗体20μg/mLを含有するウェルに3分間浸し、それぞれに、マウスFc又はヒトFcを発現している抗IL−33モノクローナル抗体(以後、mAb−1と呼ぶ)を捕捉した。捕捉ステップに続き、Octetバイオセンサー上の、何も結合していない抗マウスFcポリクローナル抗体及び抗ヒトFcポリクローナル抗体を、それぞれ、マウスFc又はヒトFcとともに非特異的モノクローナル抗体200μg/mLを含有するウェルに4分間浸して飽和させた。最後に、Octetバイオセンサーを、ヒトIL−33 100nMとmAb−2 500nMとの予備混合試料を含有するウェルに4分間浸漬した。各サイクルの終了時、非共有結合により捕捉した抗IL−33抗体と、結合した、ヒトIL−33とmAb−2とで予め作製した複合体とを、HCl 10mMとを20秒ずつ交互に3回浸漬してからHBS−ET kinetics bufferに浸す方法でバイオセンサーから除去した。バイオセンサーを、実験の各ステップごとにHBS−ET kinetics bufferで洗浄した。リアルタイム結合応答を結合イベントの間監視し、各ステップ終了時の結合応答(単位:nm)を記録した。解析中、与えられたmAb−2(mAb−1=mAb−2の場合、すなわち、マトリックスの対角線沿い)の自己−自己のバックグラウンド結合シグナルを、mAb−2の全結合イベントで観察されたシグナル(交差競合マトリックスの対応列)から減算し、バックグラウンドを補正した結果を図1に示している。ヒトIL−33と各種mAb−2試料それぞれとで予め作製した複合体へのmAb−1の結合応答を測定し、各種抗IL−33モノクローナル抗体のそれぞれに対する競合的/非競合的挙動を検討した。
バイオレイヤー干渉法を使用したサルIL−33交差競合
パネルの種々の抗IL−33モノクローナル抗体間の結合競合を、Octet(登録商標)HTXバイオセンサー(ForteBio、A Division of Pall Life Sciences)を使用してリアルタイム、非標識バイオレイヤー干渉法で測定した。実験は、0.01M HEPES pH7.4、0.15M NaCl、3mM EDTA、0.05%v/v界面活性剤Tween−20、及び0.1mg/ml BSA(HBS−ET kinetics buffer)緩衝液を使用し、プレートを速度1000rpmで撹拌しながら25℃にて実施した。C末端ヘキサヒスチジンタグとともに発現させた組換えサルIL−33(MfIL−33−6His;SEQ ID:xx)への結合について2抗体が互いに競合することができたかどうかを評価するために、2μg/mLのMfIL−33−6Hisを含有するウェルにバイオセンサーを85秒間浸すことにより、抗penta−His抗体でコーティングしたOctetバイオセンサー(Fortebio Inc、#18−5079)上に、およそ0.15nm結合単位のMfIL−33−6Hisを最初に捕捉した。その後、抗原捕捉バイオセンサーを、mAb−1の50μg/mL溶液含有ウェルに5分間浸漬させ、第1の抗IL−33モノクローナル抗体(以後、mAb−1と呼ぶ)で飽和させた。その後、バイオセンサーを、第2の抗IL−33モノクローナル抗体(以後、mAb−2と呼ぶ)の50μg/mL溶液含有ウェルに4分間浸した。バイオセンサーを、実験の各ステップごとにHBS−ET kinetics bufferで洗浄した。リアルタイム結合応答を実験の間監視し、各結合ステップの最大結合応答を記録した。予めmAb−1と作製した複合体MfIL−33−6HisへのmAb−2の結合応答を測定し、各種抗IL−33モノクローナル抗体のそれぞれに対する競合的/非競合的挙動を検討した。
インビボモデルでのmAb試験;急性HDM誘導性肺炎症モデルによる、肺炎症でIL−33が果たす役割の検討
関連インビボモデルにおける抗IL−33抗体H4H9675Pの影響を検討するため、急性HDM誘導性肺炎症試験を、マウスIL−33の代わりにヒトIL−33を発現するホモ接合体マウスにおいて実施した(IL−33 HumInマウス)。
放血後、各マウスから右肺の前葉及び中葉を取り出し、1X Haltプロテアーゼ阻害剤カクテル(Pierce、#78430)を添加した、組織タンパク質抽出試薬(1X T−PER試薬;Pierce、#78510)溶液を含有する管に入れた。以後の全ステップを氷上で実施した。T−PER試薬(プロテアーゼ阻害剤カクテル含有)の容積を、組織対T−PER比が1:8(w/v)に一致するよう各試料について調整した。肺試料を、使い捨て乳棒(Kimble Chase、#749625−0010)を使用して管内で手で均質化した。得られた溶解物を遠心分離し組織片をペレットにした。可溶性タンパク質抽出物を含有する上清を新たな管に移し、さらなる分析時まで4℃で保存した。
HDMを2週間投与されたIL−33 HumInマウスの肺で放出されたサイトカインIL−4及びIL−5のレベルは、生理食塩水緩衝液で惹起したIL−33 HumInマウスより有意に高かった。対照的に、急性HDM惹起期間中に抗IL−33抗体の処置を受けたIL−33 HumInマウスでは、未処理のHDM又はアイソタイプ・コントロールを投与されたIL−33 HumInマウスに比較して肺内IL−4及びIL−5レベルを低下させる傾向があった。
放血後、各マウスから右肺後葉を取り出し、およそ2〜3mmの角切りにした後、20μg/mLのDNAse(Roche、#10104159001)及び0.7U/mLのリベラーゼTH(Roche、#05401151001)をハンクス平衡塩類溶液(HBSS)(Gibco、#14025)に希釈した溶液を含有する管に入れ、それを37℃水浴で20分間でインキュベートし、5分おきにボルテックスによる撹拌を行った。エチレンジアミン四酢酸(Gibco、#15575)を最終濃度10mMで加えて反応を停止させた。続いて、各肺をgentleMACS dissociator(登録商標)(Miltenyi Biotec、#130−095−937)を使用して分離し、その後、70μmフィルターでろ過し、遠心分離した。得られた肺のペレットを1mLの1X赤血球溶解緩衝液(Sigma、#R7757)に再懸濁し赤血球を除去した。3分間室温にてインキュベーションした後、3mLの1X DMEMを加えて赤血球溶解緩衝液を不活化させた。その後、細胞懸濁液を遠心分離し、得られた細胞ペレットを、5mLのMACS緩衝液(autoMACS Running Buffer;Miltenyi Biotec、#130−091−221)に再懸濁した。再懸濁した試料を70μmフィルターでろ過し、ウェルあたり1x106細胞を96ウェルV字底プレートに置いた。その後、細胞を遠心分離し、ペレット1X PBSで洗浄した。第2遠心分離の後、細胞生存率を測定するため、細胞ペレットを、1X PBSで1:1000に希釈したLIVE/DEAD(登録商標)Fixable Aqua Dead Cell Stain(Life Technologies、#L34957)100μLに再懸濁し、遮光して室温で20分間インキュベートした。1X PBS洗浄を1回行った後、10μg/mLの精製ラット抗マウスCD16/CD32Fc Block(クローン:2.4G2;BD Biosciences、#553142)を含有するMACS緩衝液溶液で細胞を4℃で10分間インキュベートした。細胞を1回洗浄してから、MACS緩衝液に希釈した適切な抗体混合物(表16に記載の)内で遮光しながら4℃で30分間インキュベートした。抗体をインキュベーションした後、細胞をMACS緩衝液で2回洗浄し、BD cytofix(BD Biosciences、#554655)に再懸濁してから、遮光して4℃で15分間インキュベートした。細胞をその後洗浄してMACS緩衝液に再懸濁し、BD FACS管(BD Biosciences、#352235)に移して、好酸球、グループ2自然リンパ球(ILC2)及びリンパ球についてフローサイトメトリーで分析した。
表17に示すように、HDMを2週間投与されたIL−33 HumInマウスの肺における活性化CD4+T細胞、好酸球、及びILC2の頻度は、1X PBS対照で惹起したIL−33 HumInマウスより有意に高かった。対照的に、急性HDM惹起の期間中、抗IL−33抗体で処置したIL−33 HumInマウスでは、未処理のHDM又はアイソタイプ・コントロールを投与されたIL−33 HumInマウスに比較してこれらの浸潤頻度が低下する傾向が観察された。
抗IL−33抗体H4H9675Pのへの影響を関連インビボモデルで測定するため、慢性HDM誘導性線維症及び重度の肺炎症試験を、マウスIL−33の代わりにヒトIL−33を発現するホモ接合体マウスにおいて実施した(IL−33 HumInマウス)。
放血後、各マウスから右肺の前葉及び中葉を取り出し、1X Haltプロテアーゼ阻害剤カクテル(Pierce、#78430)を添加した、組織タンパク質抽出試薬(1X T−PER試薬;Pierce、#78510)溶液を含有する管に入れた。以後の全ステップを氷上で実施した。T−PER試薬(プロテアーゼ阻害剤カクテル含有)の容積を、組織対T−PER比が1:8(w/v)に一致するよう各試料について調整した。肺試料を、使い捨て乳棒(Kimble Chase、#749625−0010)を使用して管内で手で均質化した。得られた溶解物を遠心分離し組織片をペレットにした。可溶性タンパク質抽出物を含有する上清を新たな管に移し、さらなる分析時まで4℃で保存した。
HDMを4及び12週間投与されたIL−33 HumInマウスの肺で放出されたサイトカインIL−4及びIL−5のレベルは、1X PBSで惹起したIL−33 HumInマウスより有意に高かった。対照的に、慢性HDM惹起期間中に抗IL−33抗体の処置を受けたIL−33 HumInマウスでは、未処理のHDM又はアイソタイプ・コントロールを投与されたIL−33 HumInマウスに比較して肺内IL−4及びIL−5レベルを低下させる傾向があった。
Claims (39)
- IL−33媒介性シグナル伝達を抑制する又は減弱させる、ヒトインターロイキン33(IL−33)に結合する単離されたヒトのモノクローナル抗体又はその抗原結合性断片。
- 前記抗体又はその抗原結合性断片がIL−33及びST2の相互作用を遮断する、請求項1に記載の単離された抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、IL−33及びST2の相互作用をIC50値約10nM未満で遮断する、又は25℃でのインビトロ受容体/リガンド結合アッセイで測定したIL−33及びST2の相互作用を約50%を超えて遮断する、請求項2に記載の単離された抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、IL−33及びST2の相互作用を遮断しない、又は一部のみ遮断する、請求項1に記載の単離された抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、37℃での表面プラズモン共鳴アッセイで測定した場合に結合平衡解離定数(KD)約1nM未満でヒトIL−33に結合する、請求項1〜4のいずれか一項に記載の単離された抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、37℃での表面プラズモン共鳴アッセイで測定した場合に約8分を超える解離半減期(t1/2)でヒトIL−33に結合する、請求項1〜5のいずれか一項に記載の単離された抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、IL−33媒介性のヒト好塩基球脱顆粒を抑制する、請求項1〜6のいずれか一項に記載の抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、インビトロ好塩基球活性化試験(BAT)で測定した場合にIC50約600pM未満でIL−33媒介性のヒト好塩基球脱顆粒を抑制する、請求項7に記載の抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、ヒトPBMCのIL−33媒介性IFN−γ産生を抑制する、請求項1〜8のいずれか一項に記載の抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、インビトロでのPBMC IFN−γ産生アッセイで測定した場合にIC50約25nM未満でヒトPBMCのIL−33媒介性IFN−γ産生を抑制する、請求項9に記載の抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、インビトロでのPBMC IFN−γ産生アッセイで測定した場合にIC50約3nM未満でヒトPBMCのIL−33媒介性IFN−γ産生を抑制する、請求項10に記載の抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、インビトロでのPBMC IFN−γ産生アッセイで測定した場合にIC50約0.5nM未満でヒトPBMCのIL−33媒介性IFN−γ産生を抑制する、請求項11に記載の抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、アレルゲン誘導性肺炎症の動物モデルに投与した場合に肺におけるCD4陽性T細胞、好酸球及びILC2細胞の頻度を低下させる、請求項1〜12のいずれか一項に記載の抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、アレルゲン誘導性肺炎症の動物モデルに投与した場合に肺におけるIL−4及びIL−5のレベルを低下させる、請求項1〜13のいずれか一項に記載の抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、アレルゲン誘導性肺炎症の動物モデルに投与した場合、アイソタイプ・コントロール抗体を投与したアレルゲン誘発動物の場合に比して、IL−4レベルの少なくとも4倍低下及び/又はIL−5レベルの少なくとも5倍低下をもたらす、請求項14に記載の抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、配列番号2/10、18/26、34/42、50/58、66/74、82/90、98/106、114/122、130/138、146/154、162/170、178/186、194/202、210/218、226/234、242/250、258/266、274/282、290/298、及び308/316番からなる群から選択されるHCVR/LCVRの配列対を含む基準抗体と、IL−33への結合について競合する、請求項1〜15のいずれか一項に記載の抗体又は抗原結合性断片。
- 前記抗体又はその抗原結合性断片が、配列番号2/10、18/26、34/42、50/58、66/74、82/90、98/106、114/122、130/138、146/154、162/170、178/186、194/202、210/218、226/234、242/250、258/266、274/282、290/298、及び308/316番からなる群から選択されるHCVR/LCVRの配列対を含む基準抗体と同一のIL−33上のエピトープに結合する、請求項1〜16のいずれか一項に記載の抗体又は抗原結合性断片。
- ヒトのインターロイキン−33(IL−33)に特異的に結合する単離されたヒトのモノクローナル抗体又はその抗原結合性断片であって、前記抗体又は抗原結合性断片が(a)配列番号:2、18、34、50、66、82、98、114、130、146、162、178、194、210、226、242、258、274、290、及び308番からなる群から選択されるアミノ酸配列を有する重鎖可変領域(HCVR)の相補性決定領域(CDR);並びに(b)配列番号:10、26、42、58、74、90、106、122、138、154、170、186、202、218、234、250、266、282、298、及び316番からなる群から選択されるアミノ酸配列を有する軽鎖可変領域(LCVR)のCDR、を含む、単離されたヒトのモノクローナル抗体又はその抗原結合性断片。
- 前記抗体又は抗原結合性断片が、配列番号:2/10、18/26、34/42、50/58、66/74、82/90、98/106、114/122、130/138、146/154、162/170、178/186、194/202、210/218、226/234、242/250、258/266、274/282、290/298、及び308/316番からなる群から選択されるアミノ酸配列対のHCVR/LCVRの重鎖及び軽鎖CDRを含む、請求項18に記載の単離された抗体又は抗原結合性断片。

- ヒトのインターロイキン−33(IL−33)に特異的に結合する単離されたヒトのモノクローナル抗体又はその抗原結合性断片であって、前記抗体又は抗原結合性断片が(a)配列番号:2、18、34、50、66、82、98、114、130、146、162、178、194、210、226、242、258、274、290、及び308番からなる群から選択されるアミノ酸配列を有する重鎖可変領域(HCVR);並びに(b)配列番号:10、26、42、58、74、90、106、122、138、154、170、186、202、218、234、250、266、282、298、及び316番からなる群から選択されるアミノ酸配列を有する軽鎖可変領域(LCVR)、を含む、単離されたヒトのモノクローナル抗体又はその抗原結合性断片。
- 前記抗体又は抗原結合性断片が、配列番号:2/10、18/26、34/42、50/58、66/74、82/90、98/106、114/122、130/138、146/154、162/170、178/186、194/202、210/218、226/234、242/250、258/266、274/282、290/298、及び308/316番からなる群から選択されるHCVR/LCVRアミノ酸配列対を含む、請求項21に記載の単離された抗体又は抗原結合性断片。
- 請求項1〜22のいずれか一項に記載の抗体又は抗原結合性断片をコードする、単離された核酸分子。
- 請求項23に記載の核酸分子を含む、発現ベクター。
- 請求項24に記載の発現ベクターを含む、宿主細胞。
- 請求項1〜22のいずれか一項に記載の抗体又は抗原結合性断片を含む、医薬組成物、及び薬理学的に許容される担体又は溶剤。
- 炎症性の疾患もしくは障害、又はかかる炎症性の疾患又は障害に関連する少なくとも1つの症状を治療する方法であって、該方法は、請求項1から22のいずれか一項に記載の、IL−33に特異的に結合する抗体、又はその抗原結合性断片、又は請求項26に記載の医薬組成物を、それを必要とする患者に投与することを含み、これにおいて、前記炎症性の疾患又は障害が回復する、又は発症の重症度、持続時間もしくは頻度を低減する、又は前記炎症性の疾患又は障害に関連する少なくとも1つの症状が回復する、又は発症の重症度、持続時間もしくは頻度を低減する、治療方法。
- 前記炎症性の疾患又は障害が、喘息、アトピー性皮膚炎、慢性閉塞性肺疾患(COPD)、炎症性腸疾患、多発性硬化症、関節炎、アレルギー性鼻炎、好酸球性食道炎、及び乾癬からなる群から選択される、請求項27に記載の方法。
- 前記炎症性の疾患又は障害が喘息である、請求項28に記載の方法。
- 前記喘息が好酸球性又は非好酸球性喘息である、請求項29に記載の方法。
- 前記喘息がステロイド抵抗性又はステロイド感受性喘息である、請求項28〜30のいずれか一項に記載の方法。
- 前記炎症性の疾患又は障害がアトピー性皮膚炎である、請求項28に記載の方法。
- 前記炎症性の疾患又は障害が慢性閉塞性肺疾患(COPD)である、請求項28に記載の方法。
- 前記慢性閉塞性肺疾患が喫煙の結果である、又は原因の一部である、請求項33に記載の方法。
- アレルゲンに対する感受性を示す患者を治療する方法であって、該方法は、請求項1〜22のいずれか一項に記載の、IL−33に特異的に結合する有効量の抗体又はその抗原結合断片、又は請求項26に記載の医薬組成物をそれを必要とする患者にを投与することを含み、これにおいて、前記抗体又は抗体を含む組成物の投与後に、患者がアレルゲンに対する感受性低下もしくはアレルギー反応減退を示す、又はアレルゲンに対するいかなる感受性もしくはアレルギー反応、又はアナフィラキシー反応を経験しない、治療方法。
- 前記炎症性の疾患又は障害が、喘息、アレルギー、アナフィラキシー、アトピー性皮膚炎、慢性閉塞性肺疾患(COPD)、炎症性腸疾患、多発性硬化症、関節炎、アレルギー性鼻炎、好酸球性食道炎、及び乾癬からなる群から選択される、炎症性の疾患又は障害の治療で使用するための、請求項1〜22のいずれか一項に記載の抗体、又は請求項26に記載の医薬組成物。
- 前記炎症性の疾患又は障害が、喘息、アレルギー、アナフィラキシー、アトピー性皮膚炎、慢性閉塞性肺疾患(COPD)、炎症性腸疾患、多発性硬化症、関節炎、アレルギー性鼻炎、好酸球性食道炎、及び乾癬からなる群から選択される、炎症性の疾患又は障害の治療用薬物製造における、請求項1〜22のいずれか一項に記載の抗体、又は請求項26に記載の医薬組成物。
- 前記炎症性の疾患もしくは障害、又は前記炎症性の疾患もしくは障害の少なくとも1つの症状を緩和する、又はアレルゲンに対するアレルギー応答を減退させるために有用である、有効量の第2の治療剤を投与することをさらに含む、請求項27〜37のいずれか一項に記載の方法。
- 前記第2の治療剤が、非ステロイド性抗炎症剤(NSAID)、コルチコステロイド、副腎皮質ホルモン、気管支拡張剤、抗ヒスタミン薬、エピネフリン、うっ血除去薬、胸腺間質性リンパ球新生因子(TSLP)拮抗薬、IL−13拮抗薬、IL−4拮抗薬、IL−4/IL−13二重拮抗薬、IL−5拮抗薬、IL−6拮抗薬、IL−12/23拮抗薬、IL−22拮抗薬、IL−25拮抗薬、IL−17拮抗薬、IL−31拮抗薬、経口PDE4阻害剤及び別のIL−33拮抗薬又はIL−33に対する異なる抗体からなる群から選択される、請求項38に記載の方法。
