Abstract

a)該タンパク質、断片又はペプチドをコードするDNAを含むヘモフィルス・インフルエンザ又はE .コリを増殖させ、細菌を回収し、外膜又は封入体を単離し;
b)強い可溶化剤で封入体を可溶化し;
c)再生剤を添加し;
d)得られた懸濁液を8から10のpHの緩衝液に対して透析する。)。
本発明のある態様において、本明細書中で「タンパク質E」及び「タンパク質Eポリペプチド」と呼ばれるH .インフルエンザ(特に、型別不能H .インフルエンザ)のポリペプチドならびに生物学的、診断学的、予防的、臨床的又は治療的に有用なその変異体及びこれらを含む組成物が提供される。
(a)配列番号1−10の何れかの配列の配列と、少なくとも85%の同一性、好ましくは少なくとも90%の同一性、より好ましくは少なくとも95%の同一性、最も好ましくは少なくとも97−99%又は完全な同一性を有するアミノ酸配列を含む単離ポリペプチド;
(b)配列番号0の選択された配列の全長にわたり、配列番号11の何れかの配列に対して、少なくとも85%の同一性、好ましくは少なくとも90%の同一性、より好ましくは少なくとも95%の同一性、さらにより好ましくは少なくとも97−99%又は完全な同一性を有するポリヌクレオチド配列を含む単離ポリヌクレオチドによりコードされるポリペプチド;又は

本発明の目的は、タンパク質Eポリペプチドをコードするポリヌクレオチド、特に本明細書中でタンパク質Eと称するポリペプチドをコードするポリヌクレオチドを提供することである。

(b)配列番号1−10(又は断片)からのアミノ酸配列の全長にわたり、配列番号1−10(又はその断片)から選択された何れかのアミノ酸配列に対して、少なくとも85%の同一性、好ましくは少なくとも90%の同一性、より好ましくは少なくとも95%の同一性、さらにより好ましくは少なくとも97−99%又は100%完全な同一性を有するポリペプチドをコードするポリヌクレオチド配列、を含むか又はこれらからなる、単離ポリヌクレオチドを提供する。
本発明は、本発明のポリヌクレオチドを含むベクター、本発明のベクターにより遺伝子操作されている宿主細胞及び組み換え技術による本発明のポリペプチドの産生にも関する。本発明のDNAコンストラクト由来のRNAを用いてこのようなタンパク質を産生させるために無細胞翻訳系を使用することもできる。
本発明はまた、診断試薬としての使用のための、本発明のタンパク質Eポリヌクレオチド及びポリペプチドの使用にも関する。真核生物、特に哺乳動物、とりわけヒト中でのタンパク質Eポリヌクレオチド及び/又はポリペプチドの検出は、疾患の診断、疾患のステージ診断又は薬物に対する感染生物の反応に対する診断方法を提供する。真核生物、特に、哺乳動物及びとりわけヒト、とりわけ、タンパク質E遺伝子又はタンパク質を含む生物に感染している又は感染している疑いのあるものを、様々な周知の技術により、ならびに本明細書中で提供される方法により、核酸又はアミノ酸レベルで検出し得る。
(a)本発明のポリヌクレオチド、好ましくは配列番号11のヌクレオチド配列の何れかもしくはその断片;
(b)(a)の配列に相補的なヌクレオチド配列;
(c)本発明のポリペプチド、好ましくは配列番号1−10のポリペプチドの何れかもしくはその断片;又は
(d)本発明のポリペプチドに対する抗体、好ましくは配列番号1−10のポリペプチドの何れかに対するもの。
免疫原として、本発明のポリペプチド及びポリヌクレオチドもしくはその変異体又はこれらを発現する細胞を使用して、このようなポリペプチド及びポリヌクレオチドに対して免疫特異的な抗体をそれぞれ作製することができる。あるいは、本発明のポリペプチドに免疫特異的な抗体を作製するために、免疫原として、ポリペプチド配列内のエピトープの、ミモトープ、特にペプチドミモトープを使用することもできる。「免疫特異的」という用語は、先行技術におけるその他の関連ポリペプチドに対する親和性よりも、本発明のポリペプチドに対して、抗体が実質的により大きい親和性を有することを意味する。
例えば、細胞、無細胞標品、化学ライブラリ及び天然産物混合物中で小分子基質及びリガンドの結合を評価するために、本発明のポリペプチド及びポリヌクレオチドを使用することもできる。これらの基質及びリガンドは天然の基質及びリガンドであり得るか又は構造的もしくは機能的摸倣物であり得る。例えば、Coliganら、Current Protocols in Immunology 1(2):第5章(1991)を参照。
本発明の別の態様は、個体、特に哺乳動物、好ましくはヒトにおいて、免疫反応を誘発するための方法に関し、この方法は、感染、特に細菌感染及びとりわけ型別不能H .インフルエンザ感染から個体を保護するために抗体及び/又はT細胞免疫反応を生じさせるのに妥当なタンパク質Eポリヌクレオチド及び/又はポリペプチド又はその断片もしくは変異体を個体に接種することを含む。このような免疫反応が細菌複製を遅らせる方法も提供される。本発明のさらに別の態様は、個体において免疫反応を誘発する方法に関し、この方法は、抗体及び/又はT細胞免疫反応(例えばサイトカイン産生T細胞又は細胞毒性T細胞を含む。)を生じさせるため、個体、好ましくはヒトを疾患(その疾患が、既に個体中で確立されているものであれ、そうでないものであれ)から防御するためなど、免疫反応を誘発するために、インビボでタンパク質Eポリヌクレオチド及び/又はポリペプチド又はその断片もしくは変異体を発現させるために、タンパク質Eポリヌクレオチド及び/又はポリペプチド又はその断片もしくは変異体の発現を支配するための、核酸ベクター、配列又はリボザイムをこのような個体に送達することを含む。遺伝子を投与することの一例は、粒子その他に対するコーティングとして所望の細胞への遺伝子の投与を加速することである。このような核酸ベクターは、DNA、RNA、リボザイム、修飾された核酸、DNA/RNAハイブリッド、DNA−タンパク質複合体又はRNA−タンパク質複合体を含み得る。
本発明のさらなる態様において、細胞又は多細胞生物への投与のためのタンパク質Eポリヌクレオチド及び/又はタンパク質Eポリペプチドを含む組成物が提供される。
ポリヌクレオチド及びポリペプチド配列は、それらの2次元及び3次源構造を決定するため、ならびに同様のホモロジーのさらなる配列を同定するために、価値ある情報リソースを形成する。コンピュータ可読媒体に配列を保存し、次いで既知の巨大分子構造プログラムにおいて、又はGCGプログラムパッケージなどの周知の検索ツールを用いて配列データベースを検索するために保存データを使用することにより、これらのアプローチは、最も容易に促進される。
当技術分野で公知のように、「同一性」とは、場合によっては、配列を比較することにより決定される場合、2以上のポリペプチド配列又は2以上のポリヌクレオチド配列の間の関係である。当技術分野において、「同一性」はまた、場合によっては、このような配列の文字列間の一致によって決定される場合、ポリペプチド又はポリヌクレオチド配列間の配列関連性の程度も意味する。「同一性」は、以下に限定されないが、(Computational Molecular Biology、Lesk、A .M .編、Oxford University Press、New York、1988;Biocomputing:Informatics and Genome Projects、Smith、D .W .編、Academic Press、New York、1993;Computer Analysis of Sequence Data、Part I、Griffin、A .M .及びGriffin、H .G編、Humana Press、New Jersey、1994;Sequence Analysis in Molecular Biology、von Heine、G .、Academic Press、1987;及びSequence Analysis Primer、Gribskov、M .及びDevereux、J .編、M Stockton Press New York、1991;及びCarillo、H .及びLipman、D .、SIAM J、Applied Math .、48:1073(1988)に記載のものを含む既知の方法により容易に計算することができる。試験する配列間で最大の一致を与えるために、同一性を決定するための方法を設計する。さらに、公開されているコンピュータプログラムにおいて同一性を決定するための方法が体系化されている。2つの配列間の同一性を決定するためのコンピュータプログラム法には、以下に限定されないが、GCGプログラムパッケージにおけるGAPプログラム(Devereux、J .ら、Nucleic Acids Research 12(1):387(1984))、BLASTP、BLASTN(Altschul、S .F .ら、J .Molec .Biol .215:403−410(1990)及びFASTA(Pearson及びLipman Proc .Natl .Acad .Sci .USA 85;2444−2448(1988)が含まれる。プログラムのBLASTファミリーはNCBI及びその他のソースから公開されている(BLAST Manual、Altschul、S .ら、NCBI NLM NIH Bethesda、MD20894;Altschul、S .ら、J .Mol .Biol .215:403−410(1990))。同一性を決定するために、周知のSmith Watermanアルゴリズムも使用し得る。

試薬
b型H .インフルエンザ株MinnA及びNTHi3655は、Robert S .Munson Jr .(Washington University School of Medicine(St .Louis、Mo))の好意により入手した。型別不能H .インフルエンザ株NTHi772は、発明者らの部署での鼻咽頭スワブ培養物からの臨床分離株であった(2)。一連の様々なHaemophilus(ヘモフィルス)種も分析し、表1で述べる。ヒトIgD骨髄腫全血清IgD(λ)をThe Binding Site(Birmigham、England)より購入した。特異的な抗−pE抗血清を作製するために、完全フロイントアジュバント(Difco、Becton Dickinson、Heidelberg、Germany)中で乳化した組み換えpE22−160[pE(A)]又はキーホールリンペットへモシアニン(KLH)に共役されたpE41−68ペプチド200μgを用いて筋肉内投与でウサギに免疫付与し、不完全フロイントアジュバント中のタンパク質の同用量で第18日及び36日に免疫促進した。2から3週間後に採血した。CnBr−セファロースに共役されたpE(A)又は特異的pEペプチド(pE41−68)を用いて、得られたポリクローナル抗体をアフィニティークロマトグラフィーにより単離した(11)。ホースラディッシュペルオキシダーゼ(HRP)結合ヤギ抗−ヒトIgDはBiosource(Camarillo、CA)から入手した。ウサギ抗−ヒトIgD pAbはDakopatts(Gentofte、Denmark)から入手した。
NAD及びヘミン(Sigma、St .Louis、MO)を各10μg/mLずつ添加したブレインハートインフュージョン(BHI)培地(Difco Laboratories、Detroit、Mich .)中でH .インフルエンザb型(MinnA)を一晩増殖させた。2回洗浄した後、0 .5%Empigen(R)(Calbiochem Novabiochem、Bedford、MA)を含有する0 .05M Tris−HCl緩衝液(pH8 .8)中で細菌を抽出した。磁気撹拌装置により37℃で2時間、細菌懸濁液を混合した。4℃にて8000xgで20分間遠心した後、上清を滅菌フィルター(0 .45μm;Sterivex−HV、Millipore)でろ過した。0 .5%Empigen(R)中のH .インフルエンザ抽出物を6M尿素含有0 .05M Tris−HCl(pH8 .8)で平衡化したQ−セファロースカラム(Amersham Pharmacia Biotech)にかけた。同じ緩衝液中の0から1M NaClの直線的勾配を用いてカラムを溶出した。IgD(λ)骨髄腫血清により検出された分画を集め、0 .05M Tris−HCl(pH8 .8)に対してSpectraphorメンブレンチューブ(分子量カットオフ6−8,000;Spectrum、Laguna hills、CA)中で透析し、YM100ディスクメンブレン(分子量カットオフ10,000;Amicon 、Beverly、MA)で濃縮した。
ランニング(MES)、試料(LDS)及び転写緩衝液ならびにNovexからのブロッティング装置(San diego、CA)とともに10%Bis−Trisゲルを用いて150Vの一定電圧でSDS−PAGEを行った。試料を一様に100℃で10分間加熱した。クーマシーブリリアントブルーR−250(13;Bio−Rad、Sundbyberg、Sweden)によりゲルを染色した。ゲルからイモビロン−P膜(Millipore、Bedford、MA)へのタンパク質バンドの電気泳動的転写を30Vで2から3時間行った。転写後、5%粉乳を含有する0 .05%Tween20を添加したPBS(PBS−Tween)中でイモビロン−P膜をブロッキング処理した。PBS−Tween中で数回洗浄した後、2%粉乳を含有するPBS−Tween中の精製IgD骨髄腫タンパク質(0 .5μg/mL、hu IgD(λ)骨髄腫;The Bindingsite)とともにこの膜を室温で1時間温置した。PBS−Tween中で数回洗浄した後、1/1000希釈したHRP結合ヤギ抗−ヒトIgDを添加した。室温で40分間温置し、PBS−Tween中でさらに数回洗浄した後、ECLウエスタンブロッティング検出試薬(Amersham Pharmacia Biotech、Uppsala、Sweden)を用いて発色を行った。
イオン交換クロマトグラフィー後、IPGphor IEFシステム(Amersham Pharmacia Biotech)を用いて、H .インフルエンザ(MinnA)のEmpigen(R)抽出物を等電点電気泳動(IEF)に供した(5、12)。ゲルでの検定のために、標準物質を使用した(カタログ番号161−0320;Bio−Rad)。Immobilon−PVDFフィルター(0 .45mm;Millipore、Bedford、US)へと、2−Dポリアクリルアミドゲルを120mAで一晩電気的ブロッティングにより転写した。飽和後、温置、ブロッキング処理及び洗浄段階を上述のように行った。
Applied Biosystems(Foster City、CA)470ガス液体固体相シークエネーターを用いて自動化アミノ酸配列分析を行った。
Berns及びThomasの変法(2、13)を使用することにより、株772から染色体DNAを調製した。簡潔に述べると、Sau3Aで1時間、部分的に消化したDNA40μgからH .インフルエンザ772ゲノムライブラリを構築した。切断されたDNAをスクロース勾配上で分画した(14)。適切なサイズ(2から7kbp)のDNA断片を含有する分画を集め、DNAをBamHI消化したpUC18に連結し、次いでGene pulser(Bio−Rad、Richmond、CA)を用いてエレクトロポレーションにより大腸菌JM83に形質転換した。アンピシリン及びX−Gal(5−ブロモ−4−クロロ−3−インドリル−β−D−ガラクトピラノシド)を添加したLB寒天上に細菌を播種した。
寒天表面を乾いたフィルターで覆うことによって、LB寒天上で一晩培養したE .コリ形質転換体をニトロセルロースフィルター(Sartorius、Gottingen、Germany)に転写した。このプレートを15分間放置し、飽和クロロホルム蒸気に20分間曝露することにより、細胞を溶解した。オボアルブミンを含有するTris平衡塩類溶液(50mM Tris−塩酸塩、154mM NaCl、1 .5%オボアルブミン[pH7 .4])中で30分間フィルターを温置することにより、フィルター上の残りのタンパク質−結合部位をブロッキング処理した。ブロッキング処理後、ヒトIgD(λ)とともにこのフィルターを30分間温置した。洗浄後にHRP結合抗−ヒトIgDポリクローナル抗体を添加し、フィルターを30分間温置した。温置は全て室温で行った。最後に、4−クロロ−1−ナフトール及びH2O2を用いてフィルターを発色させた。陽性のクローンを拾い、NTHi772ゲノムDNAを含有するpUC18プラスミドDNAを精製した。隣接プライマーM13+及びM13−及びBigDye Terminator Cycle Sequencing v .2 .0 Ready Reaction配列決定キット(Perkin−Elmer、Foster City、Ca)を用いて、得られたNTHi772DNA挿入物の配列決定を行った。得られた挿入配列(3 .55kbp)は、タンパク質HI0175、HI0176、HI0177及び最後にHI0178(15)をコードするDNAを含有するストレッチに対応した。
PCR増幅断片を用いて、全コンストラクトを作製した。NTHi772ゲノムDNA(HI0175からHI0178)を含有するpUC18を鋳型として使用した。TaqDNAポリメラーゼはRoche(Manheim、Germany)からのものであり、PCR条件は、製造者により推奨されるものであった。4種類の予想されるタンパク質、HI0175からHI0178のオープンリーディングフレームをクローニングしたが、ここでは、HID(HI0178)のクローニングを述べる手順のみを記載した。pE22−160[pE(A)と称する。]は、アミノ酸残基グルタミン21を含む内在性シグナルペプチドを欠き、制限酵素部位、BamRI及びHindIIIを導入するプライマー5’−ctcaggatccaaaggctgaacaaaatgatgtg−3’及び5’−ggtgcagattaagcttttttttatcaactg−3’を用いてPCRにより増幅した。発現ベクターによりコードされる6個のヒスチジン残基を融合させるために、pE22−160終止コドンを突然変異させた。pe遺伝子の得られた417bpオープンリーディングフレームをpET26(+)(Novagen、Darmstadt、Germany)に連結した。推定される毒性を避けるために、得られたプラスミドを最初に宿主E .コリDH5αに形質転換した。その後、pE及びpE(A)をコードするプラスミドを発現宿主BL21(DE3)に形質転換した。全長pE及びpE(A)に加えて、一連の短縮型pE変異体を製造した。概略を図8で示す。BamHI及びHindIIIを含有するプライマーを全コンストラクトに対して使用した。短縮型変異体に対する手順は上述のとおりであった。BigDye Terminator Cycle Sequencing v .2 .0 Ready reaction配列決定キット(Perkin−Elmer、Foster City、Ca)を用いて、全コンストラクトの配列決定を行った。
DNeasy Tissue kit(Qiagen、Hilden、Germany)を用いてH .インフルエンザ臨床分離株からのゲノムDNAを単離した。TaqDNAポリメラーゼは、Roche(Mannheim、Germany)からのものであり、PCR条件は製造者により推奨されるものであった。pE遺伝子を単離するために、プライマーペア5’−gcatttattaggtcagtttattg−3’及び5’−gaaggattatttctgatgcag−3’(これらは、それぞれ、隣接遺伝子HI077、HI0179にアニーリングする。)を使用した。プライマー5’−cttgggttacttaccgcttg−3’及び5’−gtgttaaacttaacgtatg−3’に加えて上述のプライマーを用いて遺伝子歩行により、得られたPCR産物(948bp)の配列決定を行った。ダイターミネーターサイクル配列決定キット(CEQ DTCSキット、Beckman Coulter、Stockholm、Sweden)を用いてBeckman CEQ2000においてキャピラリー電気泳動を行った。PHRED(CodonCode、Deadham、USA)及びSEQUENCHER(MedProbe、Oslo、Norway)を用いて、得られたDNA配列の編集及びアラインメントを行った。
NTHi 772から単離されたゲノムDNAを鋳型として使用した。2つのカセットにおいて特異的な取り込み配列に加えて、それぞれ制限酵素部位XhoIとEcoRI、及びEcoRIとSpEIを導入するDyNAzymeTMII DNAポリメラーゼ(Finnzymes、Espoo、Finland)を用いて、遺伝子HI0177及びHI0179の一部を含むpeの5’−及び3’−末端を2つのカセット(それぞれ815bp及び836bp)として増幅した(18)。得られたPCR断片を消化し、pBluescript SK(+/−)にクローニングした。EcoRIに対する制限酵素部位を用いてpUC4Kからカナマイシン耐性遺伝子カセット(1282bp)を得た。消化後、HI0177及びHI0179遺伝子の一部を含有する短縮型pe遺伝子断片にPCR産物を連結した。HeriottらのM−IV法(19)に従い、H .インフルエンザ株Eagan及びRM804を形質転換した。得られた突然変異体をPCRにより確認し、ウエスタンブロット及びフローサイトメトリーによりpE発現を分析した。
利用可能なH .インフルエンザKW20ゲノムと得られた配列を比較した(http://www .tigr .org)(15)。SignalP Vl .1 World Wide Web Prediction Server Center for Biological Sequence Analysis(http://www .cbs .dtu .dk/services/SignalP/)(16)を用いてシグナルペプチドを推定した。Kyte及びDoolittleの方法(17)により、pEの疎水性プロファイルを分析した。
体重200から250gの健常な雄Sprague−Dawleyラットを使用した。動物は全て、手術前に耳顕微鏡で観察したところ、中耳感染はなかった。診療において、ラットをメトヘキシタール(Brietal(R)、Elli Lilly and Company、Indianapolis、NI)で静脈内投与により、又は抱水クロラール(apoteksbolaget、Lund、Sweden)で腹腔内投与により麻酔した。動物実験のための細菌を上述のように培養した。遠心により回収した後、NTHi 3655及び対応するpE突然変異体の両方に対して、新鮮な培地中で2x1010コロニー形成単位(cfu)の濃度になるように細菌を懸濁した。調製物を使用まで氷上で保存した。急性中耳炎(AOM)を誘発するために、頸部を腹側正中切開して中耳に到達するようにし、細菌懸濁液およそ0 .05mLを中耳腔に染み込ませた。術後第3日及び第5日に耳顕微鏡により観察を行った。第3日に化膿性滲出液、即ち不透明な滲出液、及びしばしば著しい器官の膨張がある中耳炎をAOMとした。
IgD(λ)骨髄腫血清により検出されるH .インフルエンザタンパク質(pE)の抽出及び分離
以前、型別不能 H .インフルエンザ(NTHi)はIgDに結合せず(19)、一方、莢膜性H .インフルエンザはIgDに強く結合すると考えられてきた。発明者らは、IgD(λ)骨髄腫血清がNTHiにも特異的に結合することを発見した。NTHi772の典型的なフローサイトメトリープロファイルを図1で示す。IgDなしの対照と比較して、IgD(λ)骨髄腫タンパク質存在下で強いシフト及び蛍光の増強が見られた。
分離後、2−D分析でタンパク質を全く検出することができなかったので、型別不能H .インフルエンザ(NTHi)株772を用いてH .インフルエンザDNAライブラリを構築した。2から7kbpの範囲の断片を含有するゲノムDNAをpUC18に連結し、続いてE .コリJM83へと形質転換した。ヒトIgD(λ)及びHRP結合抗−ヒトIgDポリクローナル抗体からなるコロニー免疫アッセイを用いて、IgD結合について形質転換体を分析した。試験した20,000個のコロニーから3個の陽性のコロニーを見出し、IgD(λ)を用いた第2回目のスクリーニングに供した。発明者らは、陽性コロニーのうち1個の配列決定を行い、H .インフルエンザKW20(15)の物理的地図に従う4個のタンパク質HI0175からHI0178をコードするDNAを含有する3 .55kb挿入断片を見出した。IgD(λ)との特異的な相互作用をさらに確認するために、選択した形質転換体をフローサイトメトリーによっても分析した。図2Bで見ることができるように、空のベクターのみで形質転換した陰性対照E .コリと比較した際に、HI0175からHI0178をコードする配列に対応するH .インフルエンザ772ゲノムDNAを有するE .コリJM83をIgD(λ)により検出した(図2A)。
株NTHi 772からのpE1−160のDNA及びアミノ酸配列を図6で概説する。オープンリーディングフレーム(ORF)は160アミノ酸長であり、予想されるシグナルペプチドの長さは20アミノ酸である。コンピュータ分析から、シグナルペプチダーゼがアミノ酸残基アラニン18からリジン22を認識し、残基イソロイシン20とグルタミン21との間で切断することが示唆された(38)。平行して、HIDハイドロパシープロファイル(39)から、pEが疎水性シグナルペプチドを有し、一方、その分子の残りの部分は主に親水性であることが分かる(図7)。
タンパク質のB−細胞エピトープは主にその表面に局在する。タンパク質EポリペプチドのB−細胞エピトープを予測するために、2D−構造予想及び抗原性指数予想の2種類の方法を組み合わせた。2D−構造予想は、PSIPREDプログラム(David Jones、Brunei Bioinformatics Group、Dept .Biological Sciences、Brunei University、Uxbridge UB8 3PH、UKより)を用いて行った。抗原性指数は、Jameson及びWolf(CABIOS 4:181−186[1988])によって述べられた方法に基づいて計算した。このプログラムで使用したパラメーターは抗原性指数及び抗原性ペプチドに対する最小限度の長さであった。最小で5個の連続アミノ酸に対する0 .9という抗原性指数をプログラムにおいて閾値として使用した。良好で可能なB−細胞エピトープを含むペプチドを表3で挙げる。ntHi感染を予防するためのワクチン組成物において、これらは有用であり得(好ましくは、より大きいタンパク質に共役又は組み換え連結されている。)、保存的突然変異(表3の配列に対して好ましくは70、80、85、95、99又は100%同一)又はそれらからの5以上(例えば、6、7、8、9、10、11、12、15、20又は25)のアミノ酸を含む短縮型又は、タンパク質Eポリペプチドに対して宿主において免疫反応を誘発できる有効なエピトープが保存されている、タンパク質E(上記表2で示されるような配列番号1又はその天然の相同体)ポリペプチドの天然の状態からのペプチドのN及び/又はC末端に例えば、1、2、3、5、10個のさらなるアミノ酸を含む延長型を含む、類似のペプチドも同様である。
表面露出ヘモフィルス外膜タンパク質pEは、NTHi誘発性AOMに対する決定的な毒性因子であり、規定の集団において高い免疫原性があり、従って、様々なヒト疾患に対する非常に適切なワクチン候補である。

- ヘモフィルス・インフルエンザ(Haemophilus influenzae)中で検出され得、配列番号1に記載されるアミノ酸配列を有する表面露出タンパク質又はその、断片、相同体、機能的同等物、誘導体、変性物又はヒドロキシル化、スルホン化もしくはグリコシル化産物又はその他の二次的プロセシング産物。
- ヘモフィルス・インフルエンザ中で検出され得る、請求項1に記載の表面露出タンパク質の免疫原性断片又は天然に生じるかもしくは人工的に修飾したその変異体。
- 配列番号1の位置1から21のアミノ酸が欠失しているか又は1つ以上のアミノ酸により置換されている、請求項1に記載のタンパク質に基づく組み換え免疫原性タンパク質。

- 請求項1から13の何れか一項に記載のタンパク質、断片又はペプチドをコードする核酸配列、ならびにその相同体、多形体、変性物及びスプライシング変異体。
- 少なくとも1つの別の遺伝子に融合された、請求項25に記載の核酸配列を含む組み換え核酸配列。
- 請求項25又は26に記載の核酸配列を含む、プラスミド又はファージ。
- 少なくとも1つの請求項27に記載のプラスミドを含み、請求項1から13の何れか一項に記載のタンパク質、断片又はペプチドを産生することができる、非ヒト宿主(該宿主は、細菌、酵母及び植物の中から選択される。)。
- E .コリである、請求項28に記載の宿主。

- 請求項30に記載の融合タンパク質もしくはポリペプチド又は請求項31に記載の融合産物を含む、請求項18に記載の薬剤又は請求項19から24の何れか一項に記載のワクチン組成物。
- 請求項1から13の何れか一項に記載のタンパク質、断片又はペプチドの単離の方法であり、
a)該タンパク質、断片又はペプチドをコードするDNAを含むヘモフィルス・インフルエンザ(Haemophilus influenzae)又はE .コリを増殖させ、細菌を回収し、及び外膜又は封入体を単離し;
b)強い可溶化剤で封入体を可溶化し;
c)再生剤を添加し;並びに
d)得られた懸濁液を8から10のpHの緩衝液に対して透析する
段階を含む、前記方法。 - 可溶化剤がグアニジン塩酸塩である、請求項34に記載の方法。
- 再生剤がアルギニンである、請求項34又は35に記載の方法。
- 請求項18に記載の薬剤又は請求項19から23の何れか一項に記載のワクチン組成物の医薬的有効量を投与することを含む、個体において感染を予防又は治療する方法。
- 配列番号1の全長にわたり配列番号1のアミノ酸配列と少なくとも85%の同一性を有するアミノ酸配列を含む、単離ポリペプチド。
- アミノ酸配列が、配列番号1のアミノ酸配列と少なくとも95%の同一性を有する、請求項38に記載の単離ポリペプチド。
- 配列番号1のアミノ酸配列を含む、請求項38に記載のポリペプチド。
- 配列番号1の単離ポリペプチド。
- 配列番号1がシグナルペプチド(アミノ酸1−20)を欠く、請求項40又は41に記載のポリペプチド。

- 配列番号11のポリヌクレオチドを含む単離ポリヌクレオチド。
- 配列番号11の配列又はその断片を有する標識プローブとのストリンジェントなハイブリッド形成条件下で適切なライブラリをスクリーニングすることにより得ることができる、配列番号1のポリペプチドをコードするヌクレオチド配列を含む、単離ポリヌクレオチド。
- 請求項59から66の何れか一項に記載の単離ポリヌクレオチドを含む、発現ベクター又は生きている組み換え微生物。
- 請求項67に記載の発現ベクターを含む生きている組み換え微生物。
- 請求項67の発現ベクターを含む宿主細胞。
- 配列番号1のアミノ酸配列と少なくとも85%の同一性を有するアミノ酸配列を含む単離ポリペプチドを発現する、請求項66に記載の宿主細胞の膜。
- 請求項38から58のポリペプチド又は免疫原性断片を産生させるための方法であり、該ポリペプチド又は免疫原性断片の産生に十分な条件下で請求項69の宿主細胞を培養すること、及び培地からポリペプチドを回収することを含む、前記方法。
- 請求項59から66の何れか一項のポリヌクレオチドを発現させるための方法であり、該ポリヌクレオチドの少なくとも1つを含む発現ベクターにより宿主細胞を形質転換すること、及び該ポリヌクレオチドの何れか1つの発現に十分な条件下で該宿主細胞を培養することを含む、前記方法。

- 請求項38から58の何れか一項に記載のポリペプチド又は免疫原性断片に対して生成された抗体。
- ヘモフィルス・インフルエンザ(Haemophilus influenzae)感染を診断する方法であり、かかる感染を有する疑いのある動物由来の生体試料内に存在する、請求項38から58の何れか一項に記載のポリペプチド又は免疫原性断片又は該ポリペプチドに免疫特異的である抗体を同定することを含む、前記方法。
- 動物において免疫反応を生じさせることにおける使用のための薬剤の調製における、請求項38から58の何れか一項に記載のポリペプチド又は免疫原性断片の免疫学的有効量を含む組成物の使用。
- 動物において免疫反応を生じさせることにおける使用のための薬剤の調製における、請求項59から66の何れか一項に記載のポリヌクレオチドの免疫学的有効量を含む組成物の使用。
- 請求項38から58に記載のポリペプチド又は免疫原性断片に対する少なくとも1つの抗体及び適切な医薬賦形剤を含む、ヘモフィルス・インフルエンザ(Haemophilus influenzae)疾患を有するヒトを治療することに有用な治療用組成物。
