を含む、請求項4記載のポリペプチド。 - ラルストニア(Ralstonia)由来合成転写アクチベーター様エフェクター(TALE)タンパク質を含む単離されたポリペプチドであって、
該TALEタンパク質が、R1はKである、QはYである、QはSである、QはRである、R2はWである、R2はGである、R2は欠失している、SはRである、SはHである、SはAである、SはNである、およびSはTであることから選択される1以上の突然変異を除けば、配列番号8(IVDIAR1QR2SGDLA)に示すN −1 ヘアピンにおけるアミノ酸配列を含むN末端ドメイン(NTD)を有する、単離されたポリペプチド。 - 前記NTDが、IVDIARQWSGDLA(配列番号9)、IVDIARYRGDLA(配列番号10)、IVDIARSRSGDLA(配列番号11)、IVDIARYHGDLA(配列番号12)およびIVDIARRGAGDLA(配列番号13)から選択されるアミノ酸配列を含む、請求項7記載のポリペプチド。

- 前記リコンビナーゼが、Gin、Hin、Tn3、Sin、Beta、Pin.Min、DinおよびCinならびにGinの突然変異タンパク質、Hinの突然変異タンパク質、Sinの突然変異タンパク質、Betaの突然変異タンパク質、Pinの突然変異タンパク質、Minの突然変異タンパク質、Dinの突然変異タンパク質、Cinの突然変異タンパク質、Tn3の突然変異タンパク質からなる群から選択される、請求項9記載のポリペプチド。
- 請求項1〜10のいずれか1項記載のポリペプチドをコードする単離された核酸分子。
- 請求項11記載の核酸分子を含むベクター。
- 請求項11記載の核酸分子または請求項12記載のベクターで形質転換またはトランスフェクトされた宿主細胞。
- 部位特異的組換えのための方法であって、
(a)請求項1記載のポリペプチドと特異的に相互作用するための少なくとも2つの結合部位を含むDNA配列を提供することと、
(b)該DNA配列を該ポリペプチドと反応させることと
を含み、該ポリペプチドと特異的に相互作用する2つの部位の間で該DNA配列の鎖の両方が切断される部位特異的組換え事象を該ポリペプチドが触媒する、方法。 - 転写アクチベーター様エフェクター(TALE)タンパク質N末端ドメイン(NTD)の製造方法であって、
