ドナー皮膚
本明細書において使用するのに好適なドナー皮膚は、外科手術廃棄物(例えば、ヒト皮膚)及び畜殺場(例えば、ブタを含む、家畜)等の、任意の好適な源から得られてもよい。幾つかの実施形態において、エックスビボ皮膚は、機械的分離によって幾つかのサンプルへと分割されてもよい。必要とはされないが、皮膚の比較的脂肪質の皮下組織層は、剥離又は切断によって等の、当該技術分野で既知の任意の手段によって除去されてもよく、この除去は、通常、皮膚を、直径が数ミリメートルほどにも小さいサイズを含む、任意のサイズであり得る個別的なサンプルに分割する前に遂行される。皮下組織層の除去は、培養の間、残りの皮膚組織(即ち、表皮及び真皮)内への培地の吸収を改善し得、それによって炎症の候補モジュレータの局所送達又は培地送達のいずれにも好適である皮膚モデルを提供する。皮膚サンプルが0.5cm2〜cm2の範囲であるとき、複数のサンプルを同じ皮膚源から得ることができ、これにより、異なる候補モジュレータ間の又は候補モジュレータと対照との間の比較試験が可能となる。
ヒト生物学等の真核生物学の調査は、しばしば、エックスビボ物質に依存する。頻繁に、エックスビボ生体物質は、培養物中で継続的に成長する特性を示す、不死化細胞種である。一次細胞培養物は、細胞の限定された継代を弱点とし得、これが次に、着手可能な研究を限定する。不死化細胞株を使用する欠点は、インビボ環境でのほとんどの健康な真核細胞が長期的成長を示さず、しばしば、長時間維持することさえもできないため、これらの細胞がインビボ挙動を正確に反映しない場合があるということである。
エックスビボ組織の寿命は、好適な湿度を選択することによって改善することができる。典型的な真核細胞及び組織培養は、約95%湿度の環境でインキュベートされる。エックスビボヒト皮膚サンプルを支持するのに必要とされる条件についての幾つかの研究を通じて、器官培養の湿度を低下させることが、インビボ挙動からの有意な外見上の逸脱を伴わずに、37℃で皮膚サンプルの長期的維持及び/又は成長を可能にするであろうことが発見された。下記の実施例において実証されるように、湿度を約50%まで低減することは、特にそのようなサンプルが少なくとも10日間インキュベートされたとき、エックスビボ皮膚サンプルの生存性の著しい増加をもたらした。好適な湿度を選択することによって、エックスビボ皮膚サンプルを、湿度を低減することにより更により高い温度で培養し、かつ依然としてエックスビボ皮膚モデルの生存性及び/又は一貫した遺伝子調節を維持することが可能であり得る。
本明細書におけるエックスビボ皮膚サンプルのためのインキュベーションの時間は、数時間から10日間以上まで異なり得る。大規模なスクリーニングの取り組みのために、短いインキュベーション時間が有益であり得る。エックスビボ皮膚サンプルは、生体物質が培養条件に順応し、炎症応答のモジュレータに応答性であるのと同様に炎症応答のインデューサに応答性となるのに十分な時間にわたって、インキュベートされるべきである。エックスビボ皮膚サンプルは、しばしば、培地中で少なくとも数時間インキュベートされる。少なくとも幾つかのマーカー遺伝子の発現パターンが、少なくとも4、若しくは少なくとも7、若しくは少なくとも10日間、又は少なくとも19日間インキュベートされたサンプルを必要とし得ることから、10日間を超えて存続する皮膚サンプルを有することが望ましい。マーカー遺伝子は、当然のことながら、炎症を起こしていない組織の細胞と対比して、炎症を起こした組織の細胞内で異なる発現パターンを示す。炎症の指標となるために数日間を必要とするか、又はその期間にわたって炎症の指標であり続ける、いずれかの例示のマーカー遺伝子には、セラミドシンターゼ3及びHMG補酵素A受容体(HMGCR)等の、脂質代謝に関与する遺伝子産物をコードする遺伝子が含まれる。
インビボ皮膚のような挙動を示し、したがって皮膚炎症のモジュレータを特定するためのより効率的で費用効果の高いシステムを提供するであろう、エックスビボ皮膚モデルを確立することの1つの難題は、温度である。37℃のインビボ温度は、この比較的高い温度が、より低い温度よりもインビボ挙動のより急速な喪失につながって、皮膚サンプルが異常な生理的応答をより迅速に示すため、このモデルシステムに単純に課することができない。従来のエックスビボ皮膚モデルにおいて、エックスビボ皮膚サンプルの成長及び維持を可能にするために約33℃の温度を維持することは珍しくないが、この温度は、インビボ条件とは異なる生理的環境をもたらす。故に、アッセイの少なくとも一部分又は更には全てに対して、インビボ条件により近い温度(例えば、37℃)を維持することが望ましいであろう。インビボ条件とより緻密に整合した条件を確立するために、幾つかの環境特性が変動させられてもよい。驚くべきことに、湿度をおよそ50%まで低下させることが、37℃でエックスビボ皮膚培養物の成長及び維持を延長する一助となる。ほとんどのエックスビボヒト皮膚サンプルが37℃で培養及び/又は維持されることが望ましい場合があるが、そのような皮膚サンプルのためのインキュベーション温度は、使用されている皮膚炎症のモジュレータを特定する方法の詳細に応じて、37℃から変動し得る(例えば、37℃〜33℃)ことが企図される。
本明細書に開示される炎症のモジュレータを特定する方法には、候補モジュレータを皮膚における炎症応答のモジュレータとして特定するための、階層化されたスクリーニング戦略に組み入れた方法が含まれる。例えば、第1階層のスクリーニングは、候補抗炎症化合物を特定するためのインシリコの取り組みを伴い得、続いて第2階層としてインビトロの細胞ベースのアッセイが行われる。本エックスビボヒト皮膚モデルの使用を含む、皮膚炎症のモジュレータを特定する方法は次いで、モジュレータを特定するための第3階層として、多階層化されたアプローチに組み込まれてもよい。
炎症の候補モジュレータは、小分子、生体分子、及び化学的重合体を含む、任意の生化学又は化学化合物であり得る。候補モジュレータは、任意のサイズの化合物ライブラリにおいて組織化されてもよく、起源が合成であっても、又は化合物若しくはより複合の構造としての天然源に由来してもよい。その上、候補モジュレータは、合理的薬物設計プログラム等の合理的設計の取り組みから生じる化合物であってもよく、そのような取り組みがコンピュータベースであるか否かに関わらない。本明細書における候補モジュレータの構造又は機能に対して明示的に課される限定以外には、何の限定も企図されない。故に、皮膚モジュレータを特定するための方法において使用することができる、物理的物質を含む任意の化合物又は物質、並びに化学及び/又は生化学分子及びより複合の構造が、候補モジュレータとして企図される。
本明細書に開示される皮膚炎症のモジュレータを特定する方法の効率及び経費節減を最大限にするために、エックスビボ皮膚サンプルは、インビボ皮膚の挙動を可能な限り緻密にたどる挙動を呈するべきである。本方法の目的のために、哺乳動物における皮膚炎症の重要な特性は、炎症応答が調節可能である(調節できる)ことである。多様な炎症の既知の阻害剤を使用して、本明細書における方法において炎症を阻害してもよい。既知の炎症阻害剤の幾つかの非限定的な例としては、クロベタゾール、1,10−フェナントロリン、アピゲニン、βメタゾン、ヒドロコルチゾン、ケトプロフェン、アーラトン、ジンクピリチオン(ZPT)、及び硫化セレンが挙げられる。少なくとも幾つかの既知の炎症阻害剤は、エックスビボ皮膚サンプルにおいて、内因性炎症及び誘導性炎症の両方(例えば、クロベタゾール)を阻害することができる一方で、幾つかは、内因性炎症(例えば、アピゲニン)又は誘導性炎症(例えば、ZPT)を他方の種類の炎症よりも有効に阻害することにおいて、選択性を示す。幾つかの実施形態において、テトライソパルミチン酸アスコルビルエステル(INCI名称:アスコルビン酸テトラヘキシルデシル)形態でのビタミンCの安定化型である、BV−OSC等の、他の化合物を使用して、炎症を阻害してもよい。
炎症のいずれの既知の阻害剤も、エックスビボ皮膚サンプルにおいて炎症のモジュレータを特定するための、本明細書における方法において使用するのに好適であり得る。幾つかの既知の炎症インデューサは、炎症誘発性のサイトカインであり、全てのそのような分子が開示される方法の範囲内に含まれる。そのようなサイトカインの特定の例は、インターロイキン17(IL−17)、インターロイキン22(/IL−22)、及びインターロイキン1β(IL−1β又はIL−1β)である。また、IL−17及びIL−22の組み合わせを含む、炎症誘発性のサイトカイン等の既知の阻害剤の組み合わせも明示的に企図される。
本明細書に開示されるヒトエックスビボ皮膚モデル、並びに本エックスビボ皮膚モデルを使用する、皮膚炎症のモジュレータを特定するための方法は、より正確に、ヒト皮膚のインビボ挙動及び生理学を反映する。皮膚炎症のモジュレータを特定する本方法は、任意の既知の炎症マーカー遺伝子又はコードされたタンパク質の使用を企図する。幾つかの好適な炎症マーカー遺伝子には、分泌されたサイトカイン、抗菌タンパク質、脂質生合成タンパク質、及び本明細書に明示的に開示される炎症マーカーによって表される遺伝子/タンパク質の種類をコードする遺伝子(及びコードされた産物)が含まれるが、これらに限定されない。例示の炎症マーカーには、発現レベルが、炎症を起こした状態と炎症を起こしていない状態との間で異なるという点で、炎症に関連することが知られている、マーカー遺伝子の次のカテゴリが含まれる。IL1A、IL1B、IL5、IL7、IL8、IL9、IL10、IL12、IL17A、IL17C、IL17F,IL19、IL21、IL23、IL27、IL31、及びIL33等であるが、これらに限定されないインターロイキンが企図され、IL10RA、IL10RB、IL1R1、IL5RA(CD125)、及びIL9Rを含むが、これらに限定されないインターロイキン受容体も同様に企図される。また、C5、Eotaxin、MCP−4、TARC、MCP−1、MIP−3A、CCL22、CCL23、MIP−1B、RANTES、MCP−3、MCP−2、CX3CL1、IL8RA、INP10、L8RB、及びCXCL3を含むが、これらに限定されないケモカイン、並びにCCL13(MCP−4)、CCR1、CCR2、CCR3、CCR4、CCR5、CCR6、CCR8、CX3CR1、CXCR1、及びCXCR2を含むが、これらに限定されないケモカイン受容体も本開示によって理解される。加えて、MCP−1、GM−CSF、TNFSF5、MCSF、GCSF、TNFSF6、IFNA2、IFNG、TNFB、MIF、NAMPT、TNF、TRAIL、IFNA1、IFNG、及びTNF等であるが、これらに限定されない他のサイトカイン、並びにCD4、CD40、TNFSF5、FASLG、JAK2、JNK1、NFκB1、RAG1、STAT1、s100ファミリー、及びβデフェンシンを含むが、これらに限定されない炎症に関与する他の遺伝子も企図される。
本明細書における方法は、エックスビボヒト皮膚サンプルの内因性炎症及び/又はエックスビボヒト皮膚サンプルの誘導性炎症を利用して、炎症のモジュレータを特定してもよい。内因性炎症は、生物から物理的に取り出され、かつ/又は個々のサンプルへと分離された皮膚サンプルにおいて生じ、炎症の既知の原因である皮膚創傷に類似させられてもよい。分子レベルで、内因性炎症は、比較的特徴付けられていない。対照的に、誘導性炎症は、使用されている炎症のインデューサに関連する分子経路を伴うので、分子レベルで、内因性炎症よりも大きな程度に特徴付けられている炎症のより具体的な形態と見なすことができる。炎症のインデューサに曝露されたエックスビボ皮膚サンプルはまた、内因性炎症も示し得るが、誘導性炎症応答のはるかにより大きい規模により、エックスビボ皮膚サンプルの誘導性炎症応答を測定又は監視することが可能となる。
前に考察されたように、ヒトドナー組織サンプルは、外科手術廃棄物組織に由来し得、膜上で培養され得るか、又は皮下脂肪層を含むか若しくは皮下脂肪層が除去されたサンプルを含むかのいずれかである。ヒトドナー組織サンプルが少なくとも第1及び第2のヒトドナー組織サンプルへと分割される場合の実施形態において、皮下脂肪層は、分割前に除去され得る。組織分割サンプルのうちの幾つかは、陽性対照を投薬されてもよく、分割された組織サンプルのうちの幾つかは、1つ以上の候補モジュレータを投薬されてもよく、分割された組織サンプルのうちの幾つかは、無処理の比較対象として使用されてもよい(陽性対照又は目的とする候補モジュレータのいずれでも処理されていないことを意味する)。
皮膚炎症と闘う種々の化粧品組成物を提供することの望ましさから、本明細書に記載される方法及びモデルのうちの1つ以上によって特定される皮膚炎症の候補モジュレータは、皮膚への局所適用に好適な化粧品組成物へと調合される。ある特定の実施形態において、化粧品組成物は、皮膚科学的に許容される担体、候補モジュレータ、及び提供されている特定の化粧品組成物に一般的に含まれる種類の1つ以上の任意の成分を含んでもよい。
エックスビボ皮膚サンプル調製
この実施例は、本明細書に開示される方法に従って使用するためのエックスビボ皮膚サンプルを調製する際に使用される、物質及び方法を記載する。開示されるスクリーニング方法において使用するためのエックスビボ皮膚サンプルの調製に使用される物質は、ダルベッコ変法イーグル培地(DMEM)及びGlutamax、抗生物質及び抗真菌剤、1Xリン酸緩衝食塩水(PBS)、150×15mm滅菌培養皿、滅菌ゲージ4×4、使い捨て安全外科用メス、4mm使い捨て生検パンチ、6ウェル細胞培養インサート、消毒済みピンセット、綿棒、6ウェル培養皿、罫線付き消毒済みまな板(12×12)、スクレーパーハンドル、組織凍結培地、生検カセット、凍結組織凍結容器、10%ホルマリン、固定組織用の輸送用容器、及びバイオハザード/鋭利物容器を、含んだ。培地をエックスビボ皮膚の到着前に調製した。皮膚は滅菌培養皿中の滅菌DMEM浸漬ガーゼ上で真皮(脂肪)側を下にして到着した。培養皿はテープで閉じられ、バイオハザードジップロック袋に入れられ、二次容器内の氷のう上にあった。
培養エックスビボ皮膚サンプル
6ウェルプレートの各ウェル中、2.5mLの培地又は処理(例えば、候補モジュレータ、炎症インデューサ、及び/又は炎症の阻害剤)を伴う培地を含む、培養プレートを調製した。1つのMillicell培養インサートを各ウェルに添加し、100μLの1xDMEMを各インサート膜の中心に置いた。皮膚組織の角を無作為に選択し、膜上の培地の上の、インサートの各々の中に配置した(1インサート当たり1片の皮膚)。プレートを、培地を毎日新たに交換しながら33℃で保管した。対照として、ベースライン組織測定値を余剰の廃棄物から集めた(急速凍結のために低温を使用した)。ベースライン生検試料採集は、MTT比色分析アッセイ(下に記載される)用に2つの4mmパンチを含み、このうち1つの4mmパンチをOCT組織化合物中で急速凍結し、1つの4mmパンチを10%ホルマリン中に固定し、パラフィン包埋に回した。炎症マーカー遺伝子の発現測定を、PCR及び目的とする遺伝子(複数可)の既知の配列に適切なプライマーを使用して行った。

この方法を使用して、Meso Scale Discovery(「MSD」)の標準ELISAビーズアッセイプロトコル及びMSDのサイトカイン特異的試薬を用いて、サイトカインの存在を測定することができる。組織に物質供給する前に培養培地を毎日収集し、アッセイするまでおよそ−80℃で凍結保管する。アッセイ前に、標準物を調製し、希釈過程を実行して分析のために適切な希釈を確立する。サンプルを抗サイトカインビーズと共に3時間インキュベートし、Bio−Rad Bioplex 100計器を使用して実行する。
mRNAの凍結調製及び抽出のために、以下の装置を得、準備した:2mL丸底管に1mLのTRIZOL及び各サンプルにつき5mmのステンレススチールビーズを準備した。ドライアイスバケツを得、液体窒素をそのバケツに入れた。生検パンチをドライアイス上に維持し、サンプルを凍結したままにした。凍結袋及びcovaris cryoprepをサンプルの凍結破砕において使用した。サンプルを凍結破砕するために、生検を凍結袋(サンプル当たり2つ)に入れ、液体窒素中に浸し、その後、cryoprep(設定4)に入れた。サンプル袋を取り出し、窒素中に再び浸した。平坦なディスクを除去し、TRIZOLを有する対応する2mL管に入れた。ディスクをTRIZOL中で凍結したまま維持した。管を閉じ、急速凍結し、管全体を液体窒素中に配置した。このプロセスを全てのサンプルで反復した後、サンプルを箱の中に配置し、更なるプロセシングに必要になるまで−80℃で保管した。
RNA定量ゲルを、Agilent Chip RNA−Nanoを使用して調製した。試薬を10分間、室温まで温めた。550μLのAgilent Red(ゲル)をフィルターカラムに移し、1,500gで10分間、室温で遠心分離した。Agilent Blue(色素)を10秒間、ボルテックスした。65μLのフィルターを通したゲルを管の中に移し、1μL Agilent Blue(色素)を添加し、その管を13,000gで10分間、遠心分離した。次に、Agilentチップを9μLのゲル−色素混合物と共にプライミングステーションにロードした。プライミングステーションを30秒間閉じ、続いてプランジャを引いた。プライミングステーションの残りの2Gウェルに9μLのゲル−色素混合物をロードした。ウェルに以下をロードした:各ウェル中に5μLのAgilent Green、ラダーウェルに1μLのAgilent Yellow(Nano Ladder)、及びサンプルウェルに1μLのサンプル。チップ振とう器中でボルテックスを1分間行い、その時間の後、Agilent Analyzerを製造業者の仕様書に従って使用した。
cDNAをmRNAサンプルから合成した。cDNA反応物を、最初にマスターミックス、次いでサンプル及び水を用いて、0.2mLのPCRストリップ管中で調製した。管をサーマルサイクラー中に配置し、製造業者によって推奨されるcDNAサーマルサイクリングプロトコルに従った。サンプルは必要になるまで−20℃で保管した。
炎症マーカー遺伝子の発現レベルを、ROXを用いるQuanta Perfecta Sybr Greenマスターミックスを使用したPCRによって決定した。PCRプレートにプロジェクト番号を付した。PCRに必要なプライマーを全てのプレートに事前に入れた。試薬を混合し、反応混合物を室温のプレート中、1ウェル当たり20μLでプレートにわたってアリコートした。プレートを12×8のパターンで準備、即ち、プレートの縦列別に8個のサンプル及び横列別に12個の遺伝子を用いた。PCR分析は、StepOneソフトウェアを利用した。
エックスビボ皮膚サンプルにおける炎症のインデューサ
炎症誘発性のサイトカイン等の種々の既知の炎症のインデューサを、エックスビボ皮膚モデルシステムを使用して試験した。IL−17、IL−22、IL−1β、及びこれらの組み合わせを含む、種々の炎症誘発性のサイトカインは、エックスビボ皮膚サンプルにおいて炎症を誘導することが示された。
エックスビボ皮膚モデルシステムにおいて使用するためのIL−17及びIL−22の検査
IL−17及びIL−22を、エックスビボ皮膚サンプルの細胞内の炎症誘導効果について検査した。IL−17及びIL−22の両方が、特に組み合わされると、エックスビボ皮膚サンプルにおいて炎症を誘導することが見出された。これらの分子が皮膚細胞に対して有する効果をより十分に理解するため、及び皮膚炎症のモジュレータを特定するための開示の方法において使用する量を最適化するために、これらのサイトカインの追加の研究を行った。
炎症の阻害剤−エックスビボ皮膚サンプルにおける内因性又は誘導性炎症
クロベタゾールがエックスビボ皮膚サンプルにおいて内因性及び誘導性炎症を阻害し、それが炎症応答を調節する皮膚組織及び細胞の能力を破壊しないことを示すデータが、本明細書に開示される。故に、炎症の他の既知の阻害剤の、炎症を起こしたエックスビボ皮膚サンプルに対する効果を比較するための実験を行った。
内因性及び誘導性炎症の阻害剤
前述の実施例は、とりわけ、クロベタゾールが、エックスビボ皮膚モデルにおいて内因性及び誘導性炎症の阻害剤として良好に機能することを確立した。実験を、本明細書に開示される他の炎症阻害剤によって阻害される炎症の種類を特徴付けるように設計した。当初は、応答がエックスビボ皮膚モデルにおいて作り出されるときに、所与の阻害剤が内因性炎症か、誘導性炎症か、それとも両方の種類の炎症を阻害するかは明確でなかった。この知識は、炎症の種類と阻害剤との間のいずれのミスマッチも回避され得るという点で、炎症のモジュレータを特定するための方法の設計を補助することが予想された。加えて、この情報は、特定の種類の炎症に対して有効なモジュレータの特定への、より状態に合わせたアプローチを容易にすることが予想された。
炎症マーカー遺伝子
皮膚炎症のモジュレータを特定する方法、及びそのようなモジュレータを特定するためのエックスビボ皮膚モデルの使用は、皮膚細胞及び組織内の炎症応答を監視する能力に依存する。炎症性状態に応答性であることが見出された、選択される遺伝子、即ち、炎症を起こした組織内の細胞において、炎症を起こした組織の一部でないその同じ細胞又はより現実的には同じ種類の細胞における発現とは検出可能に異なる発現レベルを有する遺伝子についての発現レベルの使用が、本明細書に開示される。本明細書に開示される実験に基づいて、次の炎症マーカー遺伝子の組が特定されてきた:IL−8、IL−6、S100A7、DEFB4、KRT10、KRT6A、TNF−α、FLG、CALML5、セラミドシンターゼ3、及びHMGCR。発現レベルは、転写RNA、総タンパク質産生、又は少なくとも1つのタンパク質活性の測定を伴い得る。
炎症マーカー遺伝子としての脂質代謝遺伝子の使用
脂質代謝に関与する遺伝子をまた、それらがエックスビボ皮膚モデルシステムにおける皮膚炎症に対する有用なマーカーであり得るかどうかを決定するために検査した。本明細書に開示される炎症インデューサのうちの3つ、即ち、IL−17、IL−22、及びIL−1βを使用して、エックスビボ皮膚サンプルにおける炎症を誘導し、2つの脂質代謝遺伝子の発現レベルを10日間の時間経過にわたって測定した。
Claims (14)
- 化粧品において使用するための炎症応答のモジュレータを特定する方法であって、
(a)表皮層及び真皮層を含む第1のエックスビボヒト皮膚サンプルを、候補モジュレータと接触させる工程であって、前記第1のエックスビボヒト皮膚サンプルが、内因性で炎症を起こしたエックスビボヒト皮膚サンプル及び誘導性の炎症を起こしたエックスビボヒト皮膚サンプルからなる群から選択される、工程と、
(b)前記第1のエックスビボヒト皮膚サンプルにおいて炎症のマーカーのレベルを測定する工程と、
(c)対照レベルを提供する工程と、
(d)前記第1のエックスビボヒト皮膚サンプルにおける前記炎症のマーカーの前記レベルが、前記対照レベルと比べた炎症応答の変化に対応するとき、前記候補モジュレータを炎症応答のモジュレータとして特定する工程と、
を含む、方法。 - 前記対照レベルを提供する工程が、第2のエックスビボ皮膚ヒト皮膚サンプルの前記マーカーの前記レベル炎症を測定することを含む、請求項1に記載の方法。
- 前記対照レベルを提供する工程が、第2のエックスビボ皮膚サンプルを対照物質と接触させることと、前記炎症のマーカーの前記レベルを測定することと、を含む、請求項1に記載の方法。
- 前記第2のエックスビボヒト皮膚サンプルが、誘導性の炎症を起こしたエックスビボヒト皮膚サンプルであり、前記対照物質が、クロベタゾール及びアピゲニンのうちの少なくとも1つである、請求項3に記載の方法。
- 前記エックスビボヒト皮膚サンプルが、誘導性の炎症を起こしたエックスビボヒト皮膚サンプルであり、炎症が、IL−17、IL−22、IL−1β、及びそれらの組み合わせからなる群から選択される有効量の化合物を、前記エックスビボヒト皮膚サンプルに送達することによって誘導される、請求項1〜4のいずれか1項に記載の方法。
- 工程(a)〜(d)を、異なる候補モジュレータを用いて反復することを更に含む、請求項1〜5のいずれか1項に記載の方法。
- 工程(a)〜(d)が、階層化されたスクリーニング方法における第2のアッセイを含み、前記候補モジュレータが、前記第2のアッセイを行う前に、前記階層化されたスクリーニング方法の第1のアッセイを行う結果として選択される、請求項1〜6のいずれか1項に記載の方法。
- 前記炎症のマーカーが、分泌されたサイトカイン、抗菌タンパク質、脂質生合成タンパク質、及び皮膚炎症に応答性の遺伝子に対応するmRNAからなる群から選択される、請求項1〜7のいずれか1項に記載の方法。
- 前記炎症のマーカーが、IL−6、IL−8、IL−10、IL−12、及び腫瘍壊死因子αからなる群から選択される分泌されたサイトカインである、請求項8に記載の方法。
- 前記炎症のマーカーが、S100A7及びDEFB4からなる群から選択される抗菌タンパク質である、請求項8に記載の方法。
- 前記炎症のマーカーが、セラミドシンターゼ3及び3−ヒドロキシ−3−メチル−グルタリル−CoAレダクターゼからなる群から選択される脂質生合成タンパク質である、請求項8に記載の方法。
- 工程(b)が、前記エックスビボヒト皮膚サンプルを前記候補モジュレータと接触させた3〜6日後に行われ、
前記候補モジュレータが前記脂質生合成タンパク質の発現を阻害するとき、前記候補モジュレータが、炎症応答のモジュレータとして特定される、
請求項11に記載の方法。 - 工程(b)が、前記エックスビボヒト皮膚サンプルを前記候補モジュレータと接触させた少なくとも7日後に行われ、
前記候補モジュレータが前記脂質生合成タンパク質の発現を増加させるとき、前記候補モジュレータが、炎症応答のモジュレータとして特定される、
請求項11に記載の方法。 - 前記炎症のマーカーが、IL6、IL8、IL10、IL12、TNFα、KRT10、KRT6A、FLG、CALML5、S100A7、DEFB4、セラミドシンターゼ3、及びHMGCRからなる群から選択される皮膚炎症に応答性の遺伝子に対応するmRNAである、請求項8に記載の方法。
