Description
・表皮の厚さ、並びに表皮及び角質層の両方の細胞ターンオーバー速度が低下し、表皮分化が変化する。
・コラーゲン、エラスチン、及びグリコサミノグリカンを含む、主要な構造分子の量が減少するにつれて、真皮はより薄くなる。紫外線損傷した皮膚における弾性網状組織は、破壊され、凝集し、種々の構造タンパク質が、グリケーションによって修飾され得る。メタロプロテイナーゼ活性は、紫外線損傷した皮膚において増加し、コラーゲン及びエラスチンの分解に寄与する。
・真皮−表皮接合部の回旋(乳頭間隆起)は、加齢に伴い扁平化し、機械的強度の損失をもたらす。これはまた、真皮上層への微小循環の減少をもたらし、したがって、表皮への栄養の減少をもたらす。
・加齢に関係した細胞間及び細胞内シグナル伝達の変化は、コラーゲン合成の減少をもたらす。
・皮膚内のヒアルロン酸含有量の変化は、加齢に伴って生じる。ヒアルロン酸は、皮膚内の天然保湿剤であり、最大でその重量の1000倍の水に結合する。加齢に関係した低下は、保湿及び堅さの低下をもたらす。
・ATP及びNADHを含む細胞内エネルギー源の減少は、皮膚細胞が若々しい皮膚生化学を維持する能力の低下をもたらし、それにより、皮膚が若々しい皮膚構造を維持及び修復する能力を低下させる。
・表皮において、エストロゲンの不足は、基礎ケラチノサイトの活性を鈍化させ、その結果として、表皮萎縮をもたらす。この萎縮性の脆弱な皮膚は、全ての人が年をとるにつれて経験する皮脂分泌の緩やかな減少のため、脂質の正常な表面膜によってあまり保護されない。角質層バリアは効果が低下し、皮膚は、特にスキンケアが不十分であったか、又は攻撃的過ぎたかの刺激に対して反応を起こし得る。
・角質層バリアを修復するのに必要な時間バリアは、加齢に伴い大幅に増加し、皮膚細胞の置換は、30歳前後の人よりも75歳の人に対して約2倍長くかかる。
図2、4、及び5を参照すると、摂動因子と、皮膚の老化組織の状態と、皮膚の老化組織の状態と関連する遺伝子との間の関連性の同定で使用するための、本発明によるシステム及びデバイスのいくつかの実施例がここに記載される。システム10は、コンピュータデバイス12、14、コンピュータデバイス12と関連するコンピュータ可読媒体16、及び通信ネットワーク18のうちの1つ以上を備える。
いくつかの実施形態では、本発明の方法は、複数の遺伝子発現プロファイリング実験から得られるデータを含む、複数のインスタンス(例えば、22、24、26)を、少なくとも第1のデジタルファイル20に追加する工程を含んでもよく、実験のうちの1つ以上は、真皮線維芽細胞及び/又はケラチノサイト細胞(又はヒト皮膚同等培養物、若しくはエクスビボ培養ヒト皮膚等の他の皮膚細胞)を、少なくとも1つの摂動因子に曝露する工程を含む。考察を簡単にするために、以下に考察される遺伝子発現プロファイリングは、マイクロアレイ実験を背景とする。
本発明のいくつかの方法は、対象となる皮膚の老化状態と関連する上方制御及び下方制御された遺伝子を表す、遺伝子発現シグネチャーを同定する工程を含む。皮膚の老化状態は、典型的には、多くの既知及び未知の外因性及び内因性要因を伴う複雑なプロセス、並びに比較的短い期間ではわずかであるが、より長い期間ではわずかではない、かかる要因への応答を伴う。これは、薬物スクリーニング法において典型的に観察されるものと対照的であり、特異的な標的、遺伝子、又は作用機構が対象となる。皮膚の老化状態と関連する独特のスクリーニング課題によって、対象となる状態を表す遺伝子発現シグネチャーの質は、実際に摂動因子への反応と関連する遺伝子発現データと、バックグラウンド発現データとの間の区別に重要であり得る。皮膚老化遺伝子発現シグネチャーの開発における一課題は、選択される遺伝子の数が、主要かつ重要な生態を反映するために正確であるが、偶然によって統計的有意性レベルに達した多くの遺伝子を含むようにあまり大きくはなく、かつ無情報である必要があることである。したがって、予測値が遺伝子発現シグネチャーの質によって決まり得るため、クエリシグネチャーは、慎重に得られるべきである。
図6及び図7を参照すると、1つ以上の皮膚老化遺伝子シグネチャーで複数のインスタンスにクエリを行うための方法がここに記載される。概して、本方法は、1つ以上の皮膚老化遺伝子シグネチャーで複数のインスタンスにクエリを行う工程と、シグネチャー遺伝子がインスタンス中の制御された遺伝子とどれほど強く一致するかを決定するために、統計的方法を適用する工程とを含む。正の関連性は、上方制御されたシグネチャーリスト中の遺伝子が、インスタンス中の上方制御された遺伝子の間で強化され、下方制御されたシグネチャーリスト中の遺伝子が、インスタンス中の下方制御された遺伝子の間で強化される時に生じる。一方で、シグネチャーの上方制御された遺伝子が、インスタンスの下方制御された遺伝子の間で主に見つかった場合、及びその逆の場合、これは、負の関連性として記録される。図6は、シグネチャー90と、プローブID 102を含むインスタンス104との間の正の関連性の極端な例を概略的に示し、インスタンスのプローブIDは、最も上方制御されたものから最も下方制御されたものへと順序付けられる。この例では、上方リスト97及び下方リスト99を含む、遺伝子シグネチャー90のプローブID 100(例えば、X1、X2 X3、X4、X5、X6、X7、X8)は、インスタンス104の最も上方制御及び下方制御されたプローブID 102のそれぞれと、1対1の正の一致を有する。同様に、図7は、シグネチャー94と、プローブID 90を含むインスタンス88との間の負の関連性の極端な例を概略的に示し、インスタンスのプローブIDは、最も上方制御されたものから最も下方制御されたものへと順序付けられる。この例では、上方リスト93(例えば、X1、X2、X3、X4)のプローブIDは、インスタンス88の最も下方制御された遺伝子と正確に一致し、下方リスト95(例えば、X5、X6、X7、X8)のプローブIDは、インスタンス88の最も上方制御されたプローブIDに正確に一致する。図8は、中立的関連性の極端な例を概略的に示し、インスタンスの上方及び下方制御された遺伝子の間に、シグネチャーの上方及び下方制御された遺伝子の一貫した強化が、正又は負のいずれにおいても存在しない。したがって、遺伝子シグネチャー108(上方リスト107及び下方リスト109を含む)のプローブID 106(例えば、X1、X2、X3、X4、X5、X6、X7、X8)は、インスタンス112のプローブID 110を有するランクに対して分散され、インスタンスのプローブIDは、最も上方制御されたものから下方制御されたものへと順序付けられる。上記の実施形態が、遺伝子シグネチャーが、皮膚状態の最も有意に上方及び下方制御された遺伝子を代表する、上方リスト及び下方リストの両方を含む、プロセスを示す一方で、遺伝子シグネチャーは、対象となる状態と関連する主要な生態が、主に一方向への遺伝子制御を示す時に、上方リスト又は下方リストのみを含んでもよいことが考えらえる。
種々の化粧品の皮膚抗老化の利益を消費者に提供することが望ましいため、本明細書に記載されるスクリーニング方法のうちの1つ以上によって同定される試験薬剤又は化合物を、皮膚への局所適用に好適な化粧組成物に組み込むことが有益であり得る。つまり、化粧組成物中に成分として試験薬剤を含むことが望ましくあり得る。ある特定の実施形態では、化粧組成物は、皮膚科学的に許容できる担体、試験薬剤、及び提供される特定の化粧組成物に一般的に含まれる種類の1つ以上の任意の成分を含んでもよい。
インスタンスの生成
個々の実験(バッチと称される)は概して、Affymetrix GeneChip(登録商標)技術プラットフォームを使用して解析される、30〜96のサンプルを含み、溶媒対照(例えば、DSMO)の6の複製、使用される細胞型において強い再現可能な効果をもたらす、陽性対照の2の複製サンプル(例えば、線維芽細胞に対するオールトランス型レチノイン酸)、及び試験物質/摂動因子のサンプルを含有する。試験物質の複製は、バッチ効果の理由から別々のバッチで行われる。GeneChip(登録商標)解析に十分なRNA(2〜4μgの合計RNA収量/ウェル)を提供するために、インビトロ試験を6ウェルプレートで実施した。
紫外線老化遺伝子発現シグネチャーを得る
日光によってもたらされる皮膚老化(紫外線老化)と関連する遺伝子発現パターンの調査で使用するための生検標本を得るための臨床的調査研究を実施した。紫外線老化と関連する遺伝子発現プロファイルを検査するために、若年及び高齢の女性からの、日光から保護された皮膚、及び日光に曝露された皮膚において、基準遺伝子発現パターンを検査した。10人の若年女性(18歳〜20歳)、及び10人の高齢女性(60歳〜67歳)のそれぞれからの、日光から保護された(臀部)、及び日光に曝露された(前腕)身体部位から、合計で3つの全層皮膚生検(〜4mm)を採取した。中程度から重度の前腕の紫外線による損傷を有するように、高齢女性を選択した。生検を液体窒素中で急速冷凍し、RNA単離まで−80℃で保管した。
a.非存在/限界/存在コールに基づくフィルタリング。このフィルタは、遺伝子発現シグネチャー中に含むための潜在的遺伝子のリストを作成する。例えば、好適なフィルタは、1つの処置群中のサンプルの少なくとも50%が、各プローブセットに対して存在コールを有しなければならないようなものであってもよい。存在コールは、生のGeneChipデータの処理から得られ、プローブセットに相補的な遺伝子転写物が生体サンプル中で実際に発現するという証拠を提供する。全サンプルからの非存在であるプローブは、単にノイズの測定値である可能性が高い。この工程は、シグネチャーにとって重要なデータに寄与しないプローブセットをフィルタリングして除去するために重要である。紫外線老化及び内因性老化遺伝子発現シグネチャーの両方に対して、Affymetrix MAS 5ソフトウェアによって提供される、少なくとも10%の存在コールを有する、プローブセットに対して、データをフィルタリングした。
b.統計的尺度によるフィルタリング。例えば、好適な統計的尺度は、t検定、ANOVA、相関係数、又は他のモデルベース解析からのp値であってもよい。一例として、p値は、統計的尺度として選択されてもよく、p=0.05のカットオフ値が選択されてもよい。シグネチャーリストを、適切な対照と比較して、統計的有意性に対して何らかの妥当なカットオフに適合する遺伝子に限定することが、対象となる生物学的状態の特徴である遺伝子の選択を可能にするために重要である。これは、倍数変化値の使用より好ましく、それは、測定値周囲のノイズを考慮しない。シグネチャー中のプローブセットを選択するために、t統計量を使用し、それは、符号付けられ、遺伝子発現変化の指向性(すなわち、上方又は下方制御される)、並びに統計的有意性の表示を提供する。
c.プローブセットのソーティング。全てのプローブセットは、統計的尺度を使用して、上方制御及び下方制御されたセットのセットにソートされる。例えば、p値を計算するために、t検定を使用した場合、p値は常に正であるため、リストをソートするために、t統計量の値(正及び負)が使用される。ソートされたt統計量により、最も有意なp値を有するセットがリストの最上部及び最下部に、有意ではないものを有するセットが中間付近に配置される。

内因性老化遺伝子発現シグネチャーを得る
経時的(内因性)皮膚老化と関連する遺伝子発現パターンの調査で使用するための生検標本を得るための臨床的調査研究を実施した。内因性老化と関連する遺伝子発現プロファイルを検査するために、若年及び高齢の女性からの、日光から保護された皮膚において、基準遺伝子発現パターンを検査した。10人の若年女性(18歳〜20歳)、及び10人の高齢女性(60歳〜67歳)のそれぞれからの、日光から保護された(臀部)身体部位から、合計で3つの全層皮膚生検(〜4mm)を採取した。生検を液体窒素中で急速冷凍し、RNA単離まで−80℃で保管した。
(i)表A及びBのそれぞれに記載される、高齢の臀部を若年の臀部と比較する、100の最も有意な上方制御された、及び100の最も有意な下方制御されたプローブセットを含む、「内因性老化200」シグネチャー(I200)。

線維芽細胞及びケラチノサイトインスタンスに対する紫外線老化遺伝子発現シグネチャー及び内因性老化遺伝子発現シグネチャーの比較
本発明者らは、紫外線老化遺伝子シグネチャー(例えば、実施例2)が、BJ線維芽細胞において試験される摂動因子インスタンスに対して強い選択的関連性を有することを観察した。換言すれば、紫外線老化シグネチャーでのクエリから得られる、トップランクインスタンス(すなわち、高い関連性スコア)間で、BJ線維芽細胞インスタンスは、強度に過剰表現された。この効果は、紫外線によって老化した皮膚において上方制御された遺伝子を、クエリシグネチャーとして使用した時に、最も劇的であった。同様の結果であるが、それほど強くないものが、内因性老化シグネチャー(例えば、実施例3)で得られた。複数のインスタンスの紫外線老化600シグネチャーとの比較の結果の要約を表1に示す。これらの結果は、i)皮膚老化表現型に寄与することが予測される種々の細胞型を含む、全層生検から得られる、遺伝子シグネチャー、及びii)皮膚老化が、皮膚の組織及び構造のほとんどにおける変化を伴う複雑な多因子状態であることを考慮すると、完全に予想外であり、驚くべきものである。

加齢に関係した遺伝子発現シグネチャーのテーマ解析
BJ線維芽細胞インスタンスを使用して得られる結果をよりよく理解するためのツールとして、テーマ解析を使用した。テーマ解析は、遺伝子発現プロファイリングデータ中の生物学的パターンを検出するための、統計解析ベースの方法である。本方法は、遺伝子製品と関連する生物学的プロセス、分子機能、及び細胞成分を記載するGene Ontology(GO)Consortium[Ashburner,M.et al.(2000)Gene ontology:tool for the unification of biology.The Gene Ontology Consortium.Nat Genet,25,25〜29]によって開発された統制用語のオントロジーを使用する。解析は、特定のGO用語に注釈付けられる遺伝子が、制御されたリスト中で有意に強化されているかを決定するために、遺伝子の制御されたリストと、全ての発現した遺伝子のより大きい参照リストとの統計的比較を伴う。この解析は、所与のGO用語と関連する複数の遺伝子が、偶然で予測されるよりも大きい頻度で、制御されたリスト上で生じる時の、生物学的パターンを明らかにする。各オントロジー用語に対するp値を計算する、テーマ抽出装置の独占的ソフトウェア及びアルゴリズムを使用して、かかる解析を実施した。フィッシャーの直接確率検定によって、統計的有意性に対してデータを解析した。ここで使用される方法及び統計的方法は、文献[Subramanian,A.et al.(2005)Gene set enrichment analysis:a knowledge−based approach for interpreting genome−wide expression profiles.Proc.Natl.Acad Sci U.S.A,102,15545〜15550]に記載されている、遺伝子セット強化解析と非常に似ている。
アーティチョーク葉抽出物及びイナゴマメ種子抽出物に対する関連性マップ結果
本実施例では、抗老化顔面効果研究に対する候補であった20の物質を、線維芽細胞及びケラチノサイトにおいて試験した。各サンプル化合物に対してマイクロアレイ解析を実施し、プローブセットIDの順序付きリストを、データベース中にインスタンスとして記憶した。インスタンスに対して紫外線老化及び内因性老化遺伝子発現シグネチャーを比較し、関連性スコアを得た。

アーティチョーク葉抽出物及びイナゴマメ種子抽出物に対するインビトロ及びインビボ結果
Cマップは、老化皮膚を改善することができる薬剤の同定に対するその予測性が、臨床試験によって実証されるべきである、仮説を生むツールである。アーティチョーク葉抽出物及びイナゴマメ種子抽出物を臨床試験に供する前に、証拠の重みを構築するためにインビトロ試験に供した。皮膚同等培養物を以下に記載する物質で処理し、プロコラーゲン、ヒアルロン酸、フィブロネクチン、及びマトリックスメタロプロテアーゼ1(MMP1)活性の阻害を含む、化粧品の効果に関する終点に対してアッセイした。
ヒト皮膚同等培養物(EFT−400全層皮膚モデル、MatTek Corporation,Ashland MA)を、37℃、及び5% CO2で、一晩平衡化した。培養物を24時間、40μLの試験物質で局所的に処理した。評価した試験物質は、3.0%の加水分解されたイナゴマメ種子抽出物の市販調製物、又は3.5%のアーティチョーク葉抽出物の市販調製物の水溶液であった。対照培養物を水溶媒のみで処理した。試験物質での処理後、培養物をPBSで洗い流した。プロコラーゲン及びヒアルロン酸測定のために、5mmのパンチ生検サンプルを採取し、各培養物の残量を、細胞生存(MTT)解析のために使用した。
研究設計は、中程度から重度(moderate to moderately-severe)の紫外線老化した顔面皮膚を有する、40〜65歳の女性の小皺及び皺、フィッツパトリック皮膚型I〜IIIを評価するための13週間、ランダム化、二重盲検、溶媒制御、分割面研究であった。研究の持続期間は、溶媒生成物による1週間の前提条件調整、続いて、12週間の顔面皮膚への毎日2回の試験生成物の適用を含んだ。対象の顔面皮膚の高密度デジタル画像を、60mmのNikonレンズを有するFuji S2 ProデジタルSLRカメラで撮った。基準画像、4、8、及び12週目の画像を撮った。各被験者に対する一致基準画像と比較して、4、8、及び12週目の目の周りの小皺及び皺における変化の程度を決定するために、熟練した評価者による0〜8の評価尺度で、符号化画像を評価した。

(c)(a)及び(b)において差次的に発現した遺伝子セットを含む、遺伝子発現シグネチャーを決定するために、(a)と(b)の発現プロファイルを比較する工程と、 (d)1つ以上の遺伝子発現シグネチャーリストを作成するために、識別子を、遺伝子発現シグネチャーを構成する各遺伝子に割り当て、差次的発現の方向に従って、識別子を順序付ける工程と、
(e)1つ以上の遺伝子発現シグネチャーリストを、少なくとも1つのコンピュータ可読媒体上に記憶する工程と、を含む、方法。
63.ヒト皮膚細胞が、皮膚の表皮又は真皮層のいずれかに由来し、被験者が、約18歳〜約80歳である、第62節に記載の方法。

(a)複数のインスタンスと、少なくとも1つの皮膚老化遺伝子発現シグネチャーとを記憶した、少なくとも1つのコンピュータ可読媒体であって、インスタンス及び遺伝子発現シグネチャーが、ヒト真皮線維芽細胞から得られ、各インスタンスが、差次的に発現した遺伝子のランク付けられた識別子のインスタンスリストを含み、少なくとも1つの皮膚老化遺伝子発現シグネチャーが、皮膚の老化状態と関連する差次的に発現した遺伝子を表す、識別子の1つ以上の遺伝子発現シグネチャーリストを含む、少なくとも1つのコンピュータ可読媒体と、
(b)プログラム可能コンピュータに以下:
(i)コンピュータ可読媒体上に記憶される、複数のインスタンス及び少なくとも1つの皮膚老化遺伝子発現シグネチャーにアクセスする工程、
(ii)少なくとも1つの皮膚老化遺伝子発現シグネチャーを、複数のインスタンスと比較する工程であって、この比較が、遺伝子発現シグネチャーリスト中の各識別子を、複数のインスタンスのそれぞれに対するインスタンスリスト中の同一の識別子の位置と比較する、工程、
(iii)複数のインスタンスのそれぞれに、関連性スコアを割り当てる工程、のうちの1つ以上を実行させる、コンピュータ可読命令を含む、プログラム可能コンピュータと、を備える、システム。
69.ヒトケラチノサイト細胞から得られる、複数のインスタンス及び少なくとも1つの遺伝子発現シグネチャーを更に含む、第68節に記載のシステム。
70.ヒト真皮線維芽細胞及び/又はヒトケラチノサイト細胞を含むサンプルを受容するための、マイクロアレイスキャナーと、スキャナーから第1のプログラム可能コンピュータに、遺伝子発現データを伝送するための、第2のプログラム可能コンピュータと、を更に備える、第68節に記載のシステム。
71.真皮線維芽細胞及びケラチノサイト細胞に適用するための、摂動因子のアレイを更に含む、第70節に記載のシステム。

Claims (14)
- 1つ以上の皮膚の老化状態と関連する、摂動因子と遺伝子との間の関連性に基づき化粧品剤を同定するための、データアーキテクチャを構築するための方法であって、
(a)対照ヒト真皮線維芽細胞に対する遺伝子発現プロファイルを提供する工程と、
(b)少なくとも1つの摂動因子に曝露されるヒト真皮線維芽細胞に対する、遺伝子発現プロファイルを生成する工程と、
(c)(a)及び(b)の前記遺伝子発現プロファイルを比較することによって、前記少なくとも1つの摂動因子に反応して差次的に発現した遺伝子を同定する工程と、
(d)前記差次的に発現した遺伝子を表す識別子を含む、順序付きリストを作成する工程であって、前記識別子が、前記遺伝子の前記差次的発現に従って順序付けられる、工程と、
(e)少なくとも1つのコンピュータ可読媒体上に、線維芽細胞インスタンスとして、前記順序付きリストを記憶する工程と、
(f)(a)〜(e)を繰り返すことによって、記憶された線維芽細胞インスタンスのデータアーキテクチャを構築する工程であって、工程(b)の前記少なくとも1つの摂動因子が、各線維芽細胞インスタンスに対して異なる、工程と、
(g)対照ヒトケラチノサイト細胞に対する遺伝子発現プロファイルを提供する工程と、
(h)少なくとも1つの摂動因子に曝露されるヒトケラチノサイト細胞に対する、遺伝子発現プロファイルを生成する工程と、
(i)(g)及び(h)の前記遺伝子発現プロファイルを比較することによって、前記少なくとも1つの摂動因子に反応して差次的に発現した遺伝子を同定する工程と、
(j)前記差次的に発現した遺伝子を表す識別子を含む、順序付きリストを作成する工程であって、前記識別子が、(i)において同定される前記遺伝子の前記差次的発現に従って順序付けられる、工程と、
(k)前記少なくとも1つのコンピュータ可読媒体上に、ケラチノサイトインスタンスとして、工程(j)において作成される前記順序付きリストを記憶する工程と、
(l)(g)〜(k)を繰り返すことによって、記憶されたケラチノサイトインスタンスのデータベースを構築する工程であって、工程(h)の前記少なくとも1つの摂動因子が、各ケラチノサイトインスタンスに対して異なる、工程と、
を含み、
前記摂動因子が、化粧剤、化学物質、活性物質、天然産物、抽出物、薬物、小分子、紫外線、熱、浸透圧ストレス、pH、微生物、ウイルス、低分子干渉RNA、およびそれらの組み合わせからなる群から選択されることを特徴とする方法。 - 工程(c)、(d)、(e)、(f)、(i)、(j)、(k)、及び(l)のうちの1つ以上を実施するために、プログラム可能コンピュータを使用する工程を含む、請求項1に記載の方法。
- 前記生成する工程が、前記少なくとも1つの摂動因子に曝露されるヒト真皮線維芽細胞または前記少なくとも1つの摂動因子に曝露されるヒトケラチノサイト細胞から生体サンプルを抽出し、前記生体サンプルをマイクロアレイ解析に供することによって実施される、請求項1〜2のいずれか一項に記載の方法。
- 前記マイクロアレイ解析が、大域的マイクロアレイ解析又は特異的マイクロアレイ解析を含み、前記特異的マイクロアレイ解析が、細胞表現型に対する遺伝子発現シグネチャーに対応する遺伝子にハイブリッド形成するオリゴヌクレオチドを含む、請求項3に記載の方法。
- 老化した皮膚を治療するために有効な化粧剤を同定するのに役立つ関連性を生成するために、請求項1〜4のいずれか一項に記載の方法により構築される前記データアーキテクチャを応用するための方法であって、
少なくとも1つの皮膚老化遺伝子発現シグネチャーで前記データアーキテクチャにクエリを行う工程を含み、クエリを行う工程が、前記少なくとも1つの皮膚老化遺伝子発現シグネチャーを、各記憶された線維芽細胞インスタンスと比較する工程を含み、
前記皮膚老化遺伝子発現シグネチャーが、少なくとも1つの皮膚の老化状態と関連して差次的に発現した遺伝子を表す、
方法。 - 前記少なくとも1つの皮膚老化遺伝子発現シグネチャーが、(i)対照と比較して、前記少なくとも1つの皮膚の老化状態において上方制御された発現を有する遺伝子を同定する工程と、(ii)対照と比較して、前記少なくとも1つの皮膚の老化状態において下方制御された発現を有する遺伝子を同定する工程と、(iii)(i)及び(ii)において同定される複数の前記遺伝子に対応する識別子を含む、前記少なくとも1つの皮膚老化遺伝子発現シグネチャーと関連する、1つ以上の遺伝子発現シグネチャーリストを作成する工程と、前記少なくとも1つのコンピュータ可読媒体上に、前記1つ以上の遺伝子発現シグネチャーリストを記憶する工程と、を含む方法によって構築される、請求項5に記載の方法。
- 少なくとも1つの皮膚サンプルが、前記少なくとも1つの皮膚の老化状態を示す被験者から採取され、生体サンプルが、前記皮膚サンプルから抽出され、前記少なくとも1つの皮膚サンプルの遺伝子発現プロファイルが、前記工程(i)及び(ii)のうちの少なくとも1つの前に生成される、請求項6に記載の方法。
- 前記皮膚サンプルが、前記被験者の表皮及び真皮層からの細胞を含む、請求項7に記載の方法。
- 前記比較する工程が、複数のインスタンスのそれぞれに関連性スコアを割り当てる工程を更に含む、請求項5〜8のいずれか一項に記載の方法。
- 皮膚科学的に許容可能な担体と少なくとも1つの摂動因子とを含む、スキンケア組成物を配合する工程を更に含み、前記少なくとも1つの摂動因子と関連する前記複数のインスタンスのそれぞれに割り当てられた前記関連性スコアが、負相関を有する、請求項9に記載の方法。
- ヒト真皮線維芽細胞及び/又はヒトケラチノサイト細胞を含むサンプルがハイブリッド形成されたマイクロアレイをスキャンするための、マイクロアレイスキャナーを提供する工程と、前記スキャナーから、前記少なくとも1つのコンピュータ可読媒体を含有する第2のプログラム可能コンピュータに、遺伝子発現データを伝送するために、第1のプログラム可能コンピュータを構成する工程と、を更に含む、請求項1〜4のいずれか一項に記載の方法。
- 前記少なくとも1つの皮膚老化遺伝子発現シグネチャーが、表A及びBに記載される前記遺伝子から選択される遺伝子からなる、請求項5〜10のいずれか一項に記載の方法。
- 前記少なくとも1つの皮膚老化遺伝子発現シグネチャーが、表C及びDに記載される前記遺伝子から選択される遺伝子からなる、請求項5〜10のいずれか一項に記載の方法。
- 1つ以上の遺伝子発現シグネチャーリストであって、前記1つ以上の遺伝子発現シグネチャーリストのそれぞれが、50〜600の識別子を含む1つ以上の遺伝子発現シグネチャーリストを作成し、
各識別子が、表A〜Dのいずれかに記載の遺伝子を表す、マイクロアレイプローブセットID、ヒト遺伝子名、ヒト遺伝子記号、及びそれらの組み合わせからなる群から選択される、
請求項5〜10、12、および13のいずれか一項に記載の方法。
