Description
7−(6−(2−ヒドロキシプロパン−2−イル)ピリジン−3−イル)−1−((トランス)−4−メトキシシクロヘキシル)−3,4−ジヒドロピラジノ[2,3−b]ピラジン−2(1H)−オンの組成物、その固体形態、その同位体族、および病気、障害、または症状の治療のためのその使用方法が本明細書に提供される。

以下の実施例は実例を示すためであり、限定を示すためのものではない。
6.1.1 分析方法論
6.1.1.1 X線粉末回折(XRPD)
固体形態スクリーンで生成された全ての固体形態サンプルを、XRPDによって分析した。XRPD分析は、Bruker AXS C2 GADDSまたはBruker AXS D8 Advance X線粉末回析測定装置において行った。
変調DSCデータを、50ポジションの自動試料採取装置を備えたTA Instruments Q2000において収集した。サファイアを用いて熱容量を校正し、認証インジウムを用いてエネルギーおよび温度を校正した。ピンホールアルミニウム皿に置いた典型的には3〜1.5mgの各サンプルを、−80℃から300℃まで2℃/分で加熱した。サンプルに対する50mL/分での乾燥窒素の除去を維持した。2℃/分の基本加熱速度、および60秒(ピリオド)ごとの±1.272℃(振幅)の温度調節パラメータを用いて、変調温度DSCを行った。装置制御ソフトウェアは、Advantage for Q Series v2.8.0.39:2およびThermal Advantage v4.8.3であり、Universal Analysis v4.4Aを用いてデータを解析した。
TGAデータを、34ポジションの自動試料採取装置を装備したMettler TGA/SDTA 851eにおいて収集した。認証インジウムを用いて装置の温度を校正した。典型的には5−15mgの各サンプルを、予め重量を測定したアルミニウムるつぼに入れて、室温から350℃まで10℃/分で加熱した。サンプルに対する50mL/分での乾燥窒素の除去を維持した。装置制御およびデータ解析ソフトウェアはSTARe v9.20であった。
画像記録のためのデジタルビデオカメラを備えたライカLM/DM偏光顕微鏡において、サンプルを分析した。少量の各サンプルをスライドガラスに載せて、個々の粒子が可能な限り文意されるように、浸オイル中に固定し、ガラススリップによって覆った。λフォールスカラーフィルターを通じて、適当な倍率および部分的な偏光を用いてサンプルを観察した
6.1.1.5 重量法水蒸気吸着(GVS)
DVS Intrinsic Controlソフトウェアv1.0.0.30によって制御されるSMS DVS Intrinsic水分吸着計測器を用いて、吸着等温線を得た。機器制御によってサンプルの温度を25℃に維持した。総流速が200mL/分の、乾燥窒素と湿った窒素との気流の混合によって湿度を制御した。サンプルの近くに配された校正ロトロニックプローブ(1.0〜100%RHのダイナミックレンジ)によって相対湿度を測定した。%RHの関数としてのサンプルの重量変化(質量応力)を微量天秤(精度±0.005mg)によって常時測定した。典型的には5〜20mgの各サンプルを室温条件のもとで風袋メッシュステンレスバスケットに入れた。40%RHかつ25℃(典型的な室内条件)でサンプルを置きかつ取り出した。25℃かつ10%RH間隔の0〜90%RHの範囲で標準等温線を行った。マイクロソフトエクセルにおいてDVS Analysis Suite v6.0.0.7を用いてデータ分析を行った。
多形スクリーンに用いた溶媒は、トルエン、MTBE(メチルtert−ブチルエーテル)、DIPE(ジイソプロピルエーテル)、THF(テトラヒドロフラン)、DME(ジメトキシエタン)、IPAc(酢酸イソプロピル)、EtOAc(酢酸エチル)、MIBK(メチルイソブチルケトン)、アセトン、IPA(イソプロピルアルコール)、エタノール、ACN(アセトニトリル)、ニトロメタン、またはIPA:水(たとえば95:5)を含むHPLCグレードまたは試薬グレードのいずれかであった。
アモルファス化合物A(実験あたり〜10mg)を前述の溶媒で処理した。室温で溶媒をゆっくり蒸発させ、残った固体をXRPDによって分析した。懸濁液を16時間の加熱/冷却サイクル(50℃/室温、8時間サイクル)に供し;その後に溶媒を蒸発させ、残った固体をXPRDによって分析した。
6.1.3.1 XPRD、TGA、およびDSC特徴付け
形態Aは図1に示す結晶XRPDパターン、および図2に示す不規則プレート晶癖を有する。化合物Aの形態AのXRPDパターンは形態Aが結晶であることを示す。結晶形態AのいくつかのXRPDピークを表2に要約する。
形態Aの吸湿性を、湿気の吸着と脱着とによって決定した。形態Aの湿気吸着/脱着の性質をDVSによって決定し、その結果を図4に要約した。0%と80%との間の相対湿度において、形態Aは有意な水の取り込みを示さなかった(<0.1%w/w)、このことは形態Aが吸湿性ではないことを示す。完全な吸着/脱着サイクルを受けた後に、サンプルのXRPDディフラクトグラムは、物質が最初の形態Aから変化していないことを示した。この特徴付けの結果に基づいて、形態Aが無水物であり、かつ非吸湿性の結晶物質であることが分かった。
調製1:化合物AをIPAおよび水(3x:5x容量)中でBHT(0.001当量)と混合した。混合物を65℃に加熱し、この温度で維持して、65℃に加熱された水(5x容量)を加えた。65℃に加熱された水中の少量の標記化合物(0.02当量)を加えた。混合物を2時間保ち、4時間かけて室温まで冷却し、さらに2時間攪拌した。得られた固体をろ過によって収集して、20%IPA水溶液によって洗浄し、乾燥させて、化合物Aを白色から黄色の固体として得た。1H NMR(400MHz、DMSO−d6)δ(ppm)9.03(d、J=1.56Hz、1H)、8.28(s、1H)、8.24(dd、J=2.34、8.20Hz、1H)、7.74(d、J=7.81Hz、1H)、7.61(s、1H)、5.26(s、1H)、4.90(tt、J=3.71,12.10Hz、1H)、4.13(s、2H)、3.28(s、3H)、3.20(tt、J=4.00、10.84Hz、1H)、2.58(qd、J=2.93、12.82Hz、2H)、2.14(d、J=10.15Hz、2H)、1.68(d、J=10.93Hz、2H)、1.47(s、6H)、1.17〜1.35(m、2H);MS(ESI)m/z398.3[M+1]+。201.9℃でのDSC吸熱。XRPDディフラクトグラム(topピーク、±0.5°)2θ角度(°):8.0、9.0、12.0、13.0、16.5、17.5、18.2、21.5、22.5、25.0、26.5。

化合物A、ピナコール(2.4当量)、およびTHF(5x容量)を混合し、45〜50℃に加熱して、トルエン(1x容量)を加えた。溶液を40〜45℃の間の温度に保ちながら、4x容量まで減圧下(300〜350Torr)で蒸留した。溶液を冷却して、15%THFのトルエン溶液となるまで、減圧下(300〜350Torr)で溶媒を連続して除去する間にトルエン(5x容量)を加えた。25℃でバッチにピナコール共結晶(0.02当量)を入れて、バッチを72時間保管した。固体をろ過して、THF/トルエンで洗浄し、45〜50℃の減圧下で乾燥させ、化合物Aピナコール共結晶を得た(1H NMRによる71%収率、20wt%ピナコール)。119.0℃でのDSC融点。XRPDディフラクトグラム(topピーク、±0.5°)2θ角度(°):5.0、6.0、12.5、14.0、15.0、15.5、17.5、18.5、22.5。
化合物AをIPA溶液および水(3x:5x容量)の中でBHT(0.001容量)と混合した。混合物を55℃に加熱して、水(5x容量)を加えた。水中の少量の標記化合物(0.02当量)を加えた。1時間かけて混合物を室温まで冷却して、さらに室温で48時間攪拌した。得られた固体をろ過によって収集し、20%IPA水溶液で洗浄し、化合物A水和物をピンク色の固体として得た。固体は111.3℃のDSC吸熱、164.9℃の発熱、および201.6℃の吸熱を有した。TGA分析によれば、質量消失は6.4%であり、開始温度は50℃であった。XRPDディフラクトグラム(topピーク、±0.5°)2θ角度(°):6.0、7.0、8.0、10.0、12.0、14.0、17.0、18.0、20.0、20.5、22.5、24.5。
調製1:化合物AをMeOH(10x容量)中でBHT(0.001当量)と混合した。混合物を容積が減少(5x)するまで蒸留し、追加の50mLの蒸留液を収集するまでIPAを添加して、さらに蒸留した。そして溶液を室温まで冷却した。得られた固体をろ過によって収集し、IPA(2x容量)で洗浄し、乾燥させ、化合物Aをオフホワイトの固体として得た。該固体のDSC分析によって、161℃の吸熱および200℃の吸熱が示された。XRPDディフラクトグラム(topピーク、±0.5°)2θ角度(°):6.5、9.0、10.0、14.5、16.5、19.0、23.0、23.5。
化合物AをMeOH(20x容量)中でBHT(0.001当量)と混合して、65℃に加熱した。溶液を室温まで冷却し、更に18時間攪拌した。得られた固体をろ過によって収集して、洗浄し、40〜45℃で乾燥させて、化合物Aをピンク色の固体として得た。固体は98.3℃のDSC吸熱、159.3℃の発熱、および200.6℃の吸熱を有した。TGA分析によれば、質量消失は7.4%であり、開始温度は80℃であった。XRPDディフラクトグラム(topピーク、±0.5°)2θ角度(°):6.0、7.5、8.0、9.0、10.0、12.5、14.5、16.5、19.0、19.5、20.5、23.0。
6.2.1 化合物Aの大規模合成
6.2.1.1 合成1
化合物Aの代謝産物を以下のように合成した:
6.3.1 14Cを豊富に含む化合物Aの合成
14C−放射標識化合物Aを下記のように調製した。
化合物10および11(1.1当量)、THF、およびK2CO3水溶液(2.5当量K2CO3)をPdCl2(Amphos)2(0.02当量)と混合して、反応完了まで(約18時間)70〜75℃に加熱した。混合物を冷却して、EtOAcおよびブラインで処理し、層を分離した。Na2SO4を用いて有機層を乾燥させ、ろ過し、残留物になるまで濃縮した。その残留物をシリカゲルを用いたカラムクロマトグラフィー(CH2Cl2:EtOAc 1:3;その後に、MeOH:EtOAc 2:98)によって精製して、残留物になるまで濃縮した。残留物をその後に0.015MのKH2PO4およびMeCNを用いた予備的なHPLCによって精製した。集められたフラクションをEtOAcによって抽出して、Na2SO4を用いて乾燥させ、ろ過し、濃縮して、14C標識化合物Aを固体として得た。

重水素を豊富に含む化合物A代謝産物を以下のように調製した。
6.4.1 錠剤
化合物Aを、活性のある薬剤成分として約5mg、20mg、および50mgの化合物Aを含む錠剤として、調製した。錠剤の調製に用いた賦形剤および担体を、それらの意図された機能と共に、表3に要約する。
セルロース(NF))と混合/調合した。次に、混合物を18メッシュの篩を用いてふるいにかけた。ふるいにかけた混合物を、Globepharma 4−8’’Bin Blenderを用いて、さらに混合/調合した。潤滑剤(たとえばステアリン酸(NF)および/またはステアリン酸マグネシウム(NF))を30メッシュの篩を用いてふるいにかけた後、その潤滑剤を混合物に加えた。得られた混合物を、次に、Globepharma 4−8’’Bin Blenderを用いて、混合/調合した。この混合物を、次に、Globepharma Korsch XL100を用いて錠剤へと圧縮し、次に、Ohara 8’’パン中でコーティングした。このように調製された錠剤を対象に、それらの粉末特性、錠剤特性、薬物産物光安定性/短期安定性、および製造工程について評価した。
最終濃度が28.6mg/mLになるように、適量の50:50(v:v)のEtOH:PEG400、[14C]−化合物A、および化合物Aを用いて、溶液を調製した。その溶液のアリコートを、投与のために、白色のSize 00 Capsugel(登録商標)V Caps Plus Hypromelloseカプセルに移した。予備的な安定性データによれば、製造過程のバルク溶液は、冷蔵条件で、かつ遮光して保管された場合、少なくとも48時間安定である。
6.5.1 健康な男性成人被検者に、1回の経口服用錠剤およびカプセル調合物の投与した後における、化合物Aの薬物動態を評価するための、第1相試験、非盲検、無作為抽出、交差研究。
Claims (32)
- 有効量の7−(6−(2−ヒドロキシプロパン−2−イル)ピリジン−3−イル)−1−((トランス)−4−メトキシシクロヘキシル)−3,4−ジヒドロピラジノ[2,3−b]ピラジン−2(1H)−オンまたはその薬剤的に許容される塩、同位体族、代謝産物、もしくは固体形態、ステアリン酸、およびラクトース一水和物を含む薬剤組成物。
- 約0.1〜5重量%のステアリン酸を含む請求項1に記載の薬剤組成物。
- 約0.4重量%のステアリン酸を含む請求項2に記載の薬剤組成物。
- 約40〜60重量%のラクトース一水和物を含む請求項1に記載の薬剤組成物。
- 約49.2重量%のラクトース一水和物を含む請求項4に記載の薬剤組成物。
- 微晶質セルロースをさらに含む請求項1に記載の薬剤組成物。

- 約15重量%の7−(6−(2−ヒドロキシプロパン−2−イル)ピリジン−3−イル)−1−((トランス)−4−メトキシシクロヘキシル)−3,4−ジヒドロピラジノ[2,3−b]ピラジン−2(1H)−オンまたはその薬剤的に許容される塩、同位体族、もしくは固体形態を含む請求項14に記載の薬剤組成物。
- 7−(6−(2−ヒドロキシプロパン−2−イル)ピリジン−3−イル)−1−((トランス)−4−メトキシシクロヘキシル)−3,4−ジヒドロピラジノ[2,3−b]ピラジン−2(1H)−オンの形態Aを含む請求項15に記載の薬剤組成物。
- ステアリン酸マグネシウムをさらに含む請求項1に記載の薬剤組成物。
- 約0.5〜3重量%からのステアリン酸マグネシウムを含む請求項17に記載の薬剤組成物。
