Description

を含む、生物学的に活性なワクシニアウイルスの精製方法に向けられる。
・高いポアサイズを持ち(可能な限り多くのリガンドがワクシニアウイルスにアクセスできるようにするため)、
・速い流速に備えて堅い構造を持ち、
・リガンドの(例えば結合又はカップリングによる)直接的又は間接的取付けが可能な形態で入手することができ、
・例えば照射を使用することにより、定置滅菌用に応用できるか、又は滅菌済ユニットとして利用できる。
1)固相へのワクシニアウイルス又はワクシニア組換えウイルスの負荷;
2)夾雑物を除去するための固相の洗浄;及び
3)単離されるべきワクシニアウイルス又は組換えウイルスの溶出
が含まれる。
例えばヘパラン硫酸又は他のGAG若しくはGAG様構造がリガンドとして取付けられている固相への負荷は、バッチ法、カラム法、又はメンブレン法で行うことができる。メンブレン法は、とりわけ大きな生体分子、特にワクシニアウイルスのような大きなウイルスにとって、いくつかの利点を持ちうる。例えば、メンブレンはポアサイズが大きく、表面上のリガンドが利用可能であることにより、大きなウイルス粒子でも、高い結合容量が可能になる。したがってメンブレン法は本発明の好ましい実施形態である。上述の全ての実施形態では、単離されるべきワクシニアウイルス又は組換えウイルスが液相に存在する。ワクシニアウイルス又は組換えウイルスがGAG又はGAG様リガンドに接近すると、ワクシニアウイルスはGAGリガンドに特異的に結合し、又はGAGリガンド「によって捕捉され」、それにより、ワクシニアウイルス又は組換えワクシニアウイルスを一時的に固相上に固定化することができ、その一方で、夾雑物は液相中に留まることになる。
リガンドへの生物学的に活性なワクシニアウイルス又は組換えウイルスの結合が十分に進行すると、ワクシニアウイルスが結合される固相を、適当な洗浄媒質で洗浄することによって、液相に留まる宿主細胞夾雑物(特に宿主細胞DNA及びタンパク質)を除去することができる。
1)例えばGAGリガンドとワクシニアウイルス上のA27L表面タンパク質との間の特異的相互作用を特異的に破壊する剤(これを特異的剤と呼ぶ)、又は
2)例えば負に帯電したGAGリガンドと正に帯電したA27L表面タンパク質との間の静電相互作用を非特異的に破壊する剤(これを非特異的剤と呼ぶ)
によって行うことができる。
前処理
固相に負荷する前に、ウイルス懸濁液の前処理を、とりわけ液相培養中のワクシニアウイルスから夾雑物を除去するために行うことができる。前処理は、単独の、又は組合わされた、以下のステップの一つ以上であることができる:
1)宿主細胞のホモジナイゼーション
・超音波処理
・凍結/融解
・低浸透圧溶解
・高圧処理
2)細胞残渣の除去
・遠心分離
・濾過
3)宿主細胞DNAの除去/低減
・ベンゾナーゼ処理
・陽イオン交換
・陽イオン界面活性剤による選択的沈殿。
ワクシニアウイルス又は組換えウイルスの溶出に使用した剤に応じて、ウイルス調製物の純度を高めるために、後処理を行うことができる。後処理は、不純物及び/又は溶出に使用した特異的若しくは非特異的剤をさらに除去するための限外濾過/ダイアフィルトレーションであることができる。ウイルスの効率的な精製を達成するには、本発明による精製を、一つ以上のさらなる精製ステップ(例えばイオン交換によるもの)と組合わせることも好ましい。その場合はイオン交換を後処理ステップとして行うこともできる。
a)固相マトリックス(この固相マトリックスは、例えばウイルスを可逆的に結合することなどによって、ウイルスと相互作用するのに適したリガンドを含む)に液相ウイルス調製物を負荷するステップ、
b)マトリックスを洗浄するステップ、及び
c)ウイルスを溶出させるステップ。
a.一つ以上の適当なウイルス結合リガンドを含むカラム、メンブレン、フィルター又は類似の固相マトリックスに、液相ウイルス調製物を負荷するステップ、
b.夾雑物を除去するためにマトリックスを適当な溶媒で洗浄するステップ、及び
c.純度が高く生物学的に活性で安定なウイルス調製物を得るために、適当な溶媒でワクシニアウイルスを溶出させるステップ。
a.一つ以上の適当なグルコサミングリカン(GAG)又はGAG様ウイルス結合リガンドを含むカラム、メンブレン、フィルター又は類似の固相マトリックスに液相ウイルス調製物を負荷するステップ、
b.夾雑物を除去するためにマトリックスを適当な溶媒で洗浄するステップ、及び
c.純度が高く生物学的に活性で安定なウイルス調製物を得るために、NaCl、H+などの非特異的溶離剤の濃度勾配、或いはGAG様化合物又は/及びA27Lペプチド若しくはペプチドフラグメントなどの特異的溶離剤の濃度勾配をもたらす溶媒でワクシニアウイルスを溶出させるステップ。
a.ヘパリン(HP)で置換されたカラム、メンブレン、フィルター又は類似の固相マトリックスに、生理的塩濃度(約150mM NaCl)を持つ中性緩衝液(pH6.5〜8.5、好ましくは≧pH7.5)に溶解したワクシニアウイルス調製物を負荷するステップ、
b.全ての非結合ワクシニアウイルス粒子及び非結合夾雑物の完全な溶出が保証されるように、十分な量の負荷緩衝液でマトリックスを洗浄するステップ、及び
c.ワクシニアウイルス粒子より弱いアフィニティを持つ夾雑物をまず最初に除去し、生物学的に活性なワクシニアウイルス粒子を最後に溶出させるために、0.15M NaClから2.0M NaClまでの、濃度が増加するNaClで、ワクシニアウイルスを溶出させるステップ。

iii)ウイルスを溶出させること
を含む、生物学的に活性なワクシニアウイルスの精製方法。
2.工業規模プロセスである、上記1に記載の方法。
3.無菌である、上記1又は2に記載の方法。
4.溶出されたワクシニアウイルスが10 8 ウイルス粒子あたり10ng未満の宿主細胞DNAを含有する、上記1〜3のいずれ一項に記載の方法。
5.ワクシニアウイルスが組換えワクシニアウイルスである、上記1〜4のいずれか一項に記載の方法。
6.ワクシニアウイルスがMVA又は組換えMVAである、上記1〜5のいずれか一項に記載の方法。
7.固相マトリックスがメンブレンを含むか、又はメンブレンである、上記1〜6のいずれか一項に記載の方法。
8.固相マトリックスがセルロースメンブレンを含むか、又はセルロースメンブレンである、上記7に記載の方法。
9.前記マトリックスが0.25μmより大きいポアサイズを含む、上記1〜8のいずれか一項に記載の方法。

18.ワクシニアウイルスを0.15M〜2.0Mの範囲の増加するNaCl濃度勾配によって溶出させる、上記17に記載の方法。
19.イオン交換による精製ステップを追加して含む、上記1〜18のいずれか一項に記載の方法。
20.ウイルス調製物のpH値が4.0〜11.0の範囲のpHに調節される、上記1〜19のいずれか一項に記載の方法。
21.医薬組成物として使用するための、上記1〜20のいずれか一項に記載の方法によって得られる溶出されたワクシニアウイルス。
22.ワクチンとして使用するための、上記21に記載のウイルス。
23.がん及び/又は感染性疾患を処置及び/又は予防するための、上記1〜20のいずれか一項に記載の方法によって得られる溶出されたワクシニアウイルス。
24.上記1〜20のいずれか一項に記載の方法によって得られる溶出されたワクシニアウイルスと、場合により、薬学的に許容できる担体及び/又は希釈剤とを含む、医薬組成物。
25.ワクチンである、上記24に記載の医薬組成物。
26.がん及び/又は感染性疾患を処置及び/又は予防するための、上記24又は25に記載の医薬組成物。
27.がん及び/又は感染性疾患を処置及び/又は予防するための医薬組成物、好ましくはワクチンを調製するための、上記1〜20のいずれか一項に記載の方法によって得られる溶出されたワクシニアウイルスの使用。
28.上記1〜20のいずれか一項に記載の方法によって得られる溶出されたワクシニアウイルスを、ヒトを含む動物に投与することを含む、免疫応答を生じさせるための方法。29.動物が哺乳動物である、上記28に記載の方法。
30.哺乳動物がヒトである、上記29に記載の方法。
Claims (30)
- i)リガンドが取付けられた固相マトリックスに、液相培養に含有されるワクシニアウイルスを負荷すること;
ii)マトリックスを洗浄すること、及び
iii)ウイルスを溶出させること
を含む、生物学的に活性なワクシニアウイルスの精製方法。 - 工業規模プロセスである、請求項1に記載の方法。
- 無菌である、請求項1又は2に記載の方法。
- 溶出されたワクシニアウイルスが108ウイルス粒子あたり10ng未満の宿主細胞DNAを含有する、請求項1〜3のいずれ一項に記載の方法。
- ワクシニアウイルスが組換えワクシニアウイルスである、請求項1〜4のいずれか一項に記載の方法。
- ワクシニアウイルスがMVA又は組換えMVAである、請求項1〜5のいずれか一項に記載の方法。
- 固相マトリックスがメンブレンを含むか、又はメンブレンである、請求項1〜6のいずれか一項に記載の方法。
- 固相マトリックスがセルロースメンブレンを含むか、又はセルロースメンブレンである、請求項7に記載の方法。
- 前記マトリックスが0.25μmより大きいポアサイズを含む、請求項1〜8のいずれか一項に記載の方法。
- リガンドがグルコサミングリカン(glucosamine glycan)(GAG)又はGAG様リガンドである、請求項1〜9のいずれか一項に記載の方法。
- リガンドが負に帯電したスルフェート基を含む、請求項1〜10のいずれか一項に記載の方法。
- リガンドがヘパラン硫酸又はヘパリンである、請求項9〜11のいずれか一項に記載の方法。
- 夾雑物が液相培養中のワクシニアウイルスから除去される、請求項1〜12のいずれか一項に記載の方法。
- ワクシニアウイルスをGAG若しくはGAG様リガンド又はその一部で溶出させる、請求項1〜13のいずれか一項に記載の方法。
- ワクシニアウイルスをA27LのGAG結合ドメイン又はその一部で溶出させる、請求項1〜14のいずれか一項に記載の方法。
- ワクシニアウイルスを0-グリコシド結合切断酵素で溶出させる、請求項1〜15のいずれか一項に記載の方法。
- ワクシニアウイルスを塩化ナトリウム(NaCl)で溶出させる、請求項1〜16のいずれか一項に記載の方法。
- ワクシニアウイルスを0.15M〜2.0Mの範囲の増加するNaCl濃度勾配によって溶出させる、請求項17に記載の方法。
- イオン交換による精製ステップを追加して含む、請求項1〜18のいずれか一項に記載の方法。
- ウイルス調製物のpH値が4.0〜11.0の範囲のpHに調節される、請求項1〜19のいずれか一項に記載の方法。
- 医薬組成物として使用するための、請求項1〜20のいずれか一項に記載の方法によって得られる溶出されたワクシニアウイルス。
- ワクチンとして使用するための、請求項21に記載のウイルス。
- がん及び/又は感染性疾患を処置及び/又は予防するための、請求項1〜20のいずれか一項に記載の方法によって得られる溶出されたワクシニアウイルス。
- 請求項1〜20のいずれか一項に記載の方法によって得られる溶出されたワクシニアウイルスと、場合により、薬学的に許容できる担体及び/又は希釈剤とを含む、医薬組成物。
- ワクチンである、請求項24に記載の医薬組成物。
- がん及び/又は感染性疾患を処置及び/又は予防するための、請求項24又は25に記載の医薬組成物。
- がん及び/又は感染性疾患を処置及び/又は予防するための医薬組成物、好ましくはワクチンを調製するための、請求項1〜20のいずれか一項に記載の方法によって得られる溶出されたワクシニアウイルスの使用。
- 請求項1〜20のいずれか一項に記載の方法によって得られる溶出されたワクシニアウイルスを、ヒトを含む動物に投与することを含む、免疫応答を生じさせるための方法。
- 動物が哺乳動物である、請求項28に記載の方法。
