本明細書で開示される化合物および医薬組成物は、溶液、乳濁液、ゲルまたは気泡体を含む種々の形態で、調製および投与することができる。従って、本明細書において企図されている医薬組成物には、本明細書に記載の、薬剤的に許容できる担体または賦形剤および一つまたは複数の化合物が含まれる。「溶液」とは、通例の意味において、化合物(例えば、本明細書に記載の化合物)が少なくとも部分的に溶解しており、好ましくは完全に溶解しており、液体として投与することができる、液体医薬組成物を意味する。「乳濁液」とは、通例の意味において、2つ以上の不混和液の混合物を意味し、一方の化合物(例えば、本明細書に記載の化合物またはその溶液)が、その他の化合物(例えば、本明細書に記載の担体)を通して分散している。「ゲル」とは、通例の意味において、粘稠性の半硬質流体をもたらす、連続的な液相内の化合物の高粘稠性の溶液、乳濁液、またはコロイド懸濁液を意味する。「コロイド」とは、通例の意味において、重力の影響下では定着しない小粒子が全体に分散している連続的な媒体を含む組成物を意味する。「気泡体」とは、通例の意味において、気体(例えば、空気)がそれを通して分散している連続的な媒体(すなわち、溶液、乳濁液、ゲル等)を含む組成物を意味する。
本明細書で提供される医薬組成物には、活性成分(例えば、式(I)、(II)、(III)、(IV)、(IIa)、(IIb)、(IIc)、(IIIa)、(IIIb)、(IIIc)、(IIId)、(IVa)、(IVb)、もしくは(IVc)の構造を有する化合物またはその誘導体、異性体もしくは鏡像異性体、およびその実施形態を含む)が、治療有効量で含有されている組成物も含まれる。特定の適用に有効な実際の量は、とりわけ、治療される疾患、症状または状態によって決まる。
特定の化合物に関する毒性と治療効果の比がその治療指数であり、LD50(集団の50%に致死的である化合物の量)とED50(集団の50%に有効である化合物の量)の間の比率として表現することができる。高い治療指数を示す化合物が好ましい。細胞培養アッセイおよび/または動物試験から得られた治療指数データを、ある範囲の投与量をヒト用に製剤化する際に使用することができる。そのような化合物の投与量は、毒性がほとんどないED50を含む、ある範囲の血漿中濃度の範囲内であることが好ましい。投与量は、採用される剤形および利用される投与経路に依存して、この範囲内で変化し得る。例えば、Fingl et al., In: THE PHARMACOLOGICAL BASIS OF THERAPEUTICS, Ch.1, p.1, 1975を参照されたい。個々の医師は、患者の状態および化合物が使用される具体的な方法を考慮に入れて、正確な製剤、投与経路、および投与量を選択することができる。
別の態様では、発毛(例えば、毛形成)を誘導するための方法が提供される。本方法は、それを必要とする対象に、治療有効量の本明細書で提供される化合物(例えば、式(I)、(II)、(III)、(IV)、(IIa)、(IIb)、(IIc)、(IIIa)、(IIIb)、(IIIc)、(IIId)、(IVa)、(IVb)、もしくは(IVc)の構造を有する化合物、またはそれらの誘導体、異性体もしくは鏡像異性体、並びにそれらの実施形態も含まれる)を投与することを含む。本化合物は、本明細書に記載の医薬組成物の一部として提供され得る。
以下の実施例は、本発明のいくつかの実施形態を解説することを目的としたものであり、本発明の範囲を限定しない。本明細書に記載されている合成のための出発物質は、市販されているか、または当技術分野で公知の方法および/もしくは本明細書に記載の方法で合成できる。
実施例1a.15−tert−ブチルジメチルシリルオキシ−ビマトプロストの合成
アセトニトリル(1.5ml)とテトラヒドロフラン(1ml)の混合物中の15−tert−ブチルジメチルシリルオキシ−11−プロピオニル−ビマトプロスト(60mg;1.02ミリモル)溶液に、0℃で、フッ化水素酸水溶液(48〜51%;0.1ml)を加えた。この溶液を室温まで温めた。30分後に水(5ml)を加えて、ジクロロメタン(2×10ml)で混合物を抽出した。合わせた有機層を硫酸ナトリウムで乾燥させ、真空中で蒸発させた。残留物のシリカゲルクロマトグラフィーによりヘキサン中50〜100%の酢酸エチルで溶出させた後、透明油状物として11−プロピオニルビマトプロスト(43mg;89%)を得た。
15−tert−ブチルジメチルシリルオキシ−11−ブチリル−ビマトプロスト(41mg;0.068ミリモル)から、実施例1cに記載の方法と類似の方法で、蒼白色油状物として化合物11−ブチリル−ビマトプロスト(30mg;91%)を合成した。
無水ジクロロメタン(1ml)中のビマトプロスト(90mg;0.22ミリモル)およびn−ブチルボロン酸(28mg;0.27ミリモル)溶液を40分間還流撹拌し、冷却し、真空中で蒸発させて、無水ベンゼン(3×2ml)との共沸混合物を形成した。残留物に無水ジクロロメタン(1ml)を加えてから、氷浴上で、トリエチルアミン(0.11ml;0.79ミリモル)、4−ジメチルアミノピリジン(15mg;0.12ミリモル)、およびプロピオン酸無水物(42μl;0.33ミリモル)を加えた。得られた混合物を周囲温度で一晩撹拌してから、酢酸エチルと10%クエン酸水溶液の間で分配した。有機層を分離し、ブラインで洗浄し、硫酸ナトリウムで乾燥させ、真空中で蒸発させた。残留物をメタノール(2ml)中で2時間撹拌し、真空中で蒸発させた。残留物のシリカゲルクロマトグラフィーによりヘキサン中50%酢酸エチル〜酢酸エチル中5%メタノールの勾配で溶出させた後、透明油状物として15−プロピオニルビマトプロスト(90mg;88%)を得た。
ビマトプロスト(90mg;0.22ミリモル)および無水酪酸(53μL;0.32ミリモル)から、実施例3に記載の方法と類似の方法で、透明油状物として化合物15−ブチリルビマトプロスト(63mg;59%)を合成した。
11,15−ジ−tert−ブチルジメチルシリルオキシ−9−イソブチリル−ビマトプロスト(145mg;0.20ミリモル)のテトラヒドロフラン(0.25ml)中溶液に、フッ化テトラブチルアンモニウム(THF中1M;0.2ml)を加えた。6時間後、反応混合物を真空中で蒸発させた。残留物のシリカゲルクロマトグラフィーにより酢酸エチルで溶出させた後、化合物9−イソブチリル−ビマトプロスト(48mg;49%)を得た。
ビマトプロスト(180mg;0.43ミリモル)およびn−ブチルボロン酸(53mg;0.52ミリモル)の無水ジクロロメタン(1ml)中溶液を4時間還流撹拌し、冷却し、真空中で蒸発させた。残留物に無水ピリジン(0.9ml)を加え、次いで、0℃で塩化アセチル(0.05ml;0.70ミリモル)を加えた。反応混合物を3時間撹拌してから、酢酸エチルと10%クエン酸水溶液の間で分配した。有機層を分離し、ブラインで洗浄し、硫酸ナトリウムで乾燥させ、真空中で蒸発させた。残留物をメタノール(5ml)中で4時間撹拌し、真空中で蒸発させた。残留物のシリカゲルクロマトグラフィーによりジクロロメタン中10%メタノールで溶出させた後、化合物15−アセチル−ビマトプロスト(150mg;76%)を得た。
実験計画
本明細書に開示される化合物の経時的安定性を、温度とpHに相関させて製剤形態で測定した。活性薬剤のターゲット濃度を0.03%として、本明細書に記載の製剤Aのいくつかのバリエーションを使用し、pHはpH4、5、6、および7に変化させた。実験中、温度を25℃、40℃、および60℃に維持した。ゼロ時間時および80日後にサンプルを評価した。
化合物IVa、IIIb、およびIVbの安定性を評価した。

これらの試験は、化合物IVa、IIIb、およびIVbとして例示される本明細書に記載の化合物が製剤状態で安定していることを明らかにしている。
実験計画
本明細書に記載の化合物に関して、インビトロの刺激スクリーニングアッセイを行った。試験システムは、当技術分野で公知の再生ヒト表皮(RHE)である。単回投与を採用し、24時間までの時間との相関関係でエンドポイントを評価した。エンドポイントとは、組織生存率とIL−1αの放出である。
試験化合物には、化合物IVa(0.03%)、IIIb(0.03%)、IVb(0.03%)、ビマトプロスト(0.03%)、ミノキシジル(5%、泡状)、およびミノキシジル(5%、溶液)が含まれていた。化合物IVa、IIIb、IVb、およびビマトプロストの投与には、製剤Aのビヒクルを使用した。上記表1を参照のこと。ミノキシジルの投与には、20%H2O、50%プロピレングリコール、30%エタノールで配合したミノキシジルビヒクルを使用した。
RHEシステムにおいて、24時間までの時間中、どの試験化合物にも組織生存率の有意な減少は一切観察されなかった。同様に、同じ時間中、IL−1α放出の有意な増加は一切観察されなかった。
化合物IVa、IIIb、およびIVbは、濃度≦0.03%において、ヒト皮膚の刺激作用の可能性の徴候を一切示さなかった。刺激作用の可能性は、当技術分野で公知のミノキシジル市販製剤に匹敵する。
実験計画
雌性ラットに対し、1日1回、ビヒクルまたは化合物IVa、IIIb、IVbのいずれかを8日間投与した。投与量は3mg/kg/日、濃度は3mg/mLであった。観察したデータは、当技術分野で知られる、体重、生存率、摂食量、および皮膚所見(皮膚反応分類)である。ビヒクルは製剤Aであった。上記表1を参照のこと。
試験動物に病的状態および予定外死亡は一切見られなかった。さらに、毒性の臨床徴候も一切観察されなかった。皮膚所見に関しては、浮腫の観察は一切なかった。2〜4日目に、化合物IVaとIIIbで微小かつ一過性の刺激症状があった。
実験計画
化合物IVa、IIIb、およびIVbを、当技術分野で公知のmicroAmesスクリーンにかけた。
microAmesスクリーンの条件下では、代謝活性化の有無によらず、化合物IVa、IIIb、およびIVbに変異原性はないと判定された。
実験計画
ビマトプロストおよび化合物IVc、IVbを用いた真皮乳頭(すなわちヒト毛乳頭細胞(HHDPC))のアッセイにおいて、細胞誘電分光分析(CDS:cellular dielectric spectroscopy)を行った。当技術分野で知られている通り、CDSとは、電気細胞インピーダンスを測定するリアルタイム、非侵襲的、標識不要、高速処理、細胞ベースのアッセイである。いかなる理論にも拘束されることを望むものではないが、GPCRの活性化が細胞形態、細胞間相互作用、および細胞接着の変化を誘発し、この変化が細胞インピーダンスの変化として現れるものと考えられる。
HHDPCに対するビマトプロスト、化合物IVc、および化合物IVbの機能試験を行った。ビマトプロストに関しては、EC50値で判定した場合に、3.0〜6.3×10-7mol/Lの範囲でマイクロモル以下の活性が観察された。対照的に、化合物IVcとIVbは、10-4〜10-13mol/Lの範囲ではEC50を測定できなかった。当技術分野で理解されている通り、「EC50」とは、最大活性の50%を示す効果濃度をいう。
いかなる理論にも拘束されることを望むものではないが、本明細書に開示される化合物(例えば化合物IVc、IVb)は、HHDPCを活性化しないと考えられる。他方、ビマトプロストはHHDPCを活性化する。したがって、本明細書に記載の化合物は、加水分解を経てビマトプロストを産生でき、次いで、このビマトプロストがHHDPCを活性化することができる。
実験計画
毛形成試験には、7週齢のC57 Black/6J雌性マウス(n=10)を使用した。背部は剪毛した(面積は〜2cm×4cm)。1日に1回(QD)、活性薬剤を局所投与により14日間投与した。42日間観察し、新しい毛成長の発生と毛成長完了の日を測定した。化合物は、a)50%プロピレングリコール、30%エタノール、20%水、b)上記製剤Aの配合物、のいずれかとして製剤化した。図4A〜4B、5A〜5B、6A〜6B、8A〜8B、および9A〜9Bで報告している実験は、50%プロピレングリコール、30%エタノール、20%水で配合したビヒクルを用いて実施した。図7A〜7Bで報告している実験は、上記製剤Aのビヒクルを用いて実施した。
図4A、5A、6A、7A、8A、9Aの各々は、毛成長の発生日をヒストグラムで示す。図4B、5B、6B、7B、8B、9Bの各々は、完全な毛成長の達成日をヒストグラムで示す。これらの図の「70」という記載は、試験の末時点(すなわち42日目)には完全な毛成長が観察されなかったことを表す。
本明細書に開示される化合物の毛成長誘発能力を試験した。完全な毛成長の回復に必要な時間中に最良の活性を有すると見られるのは、15−プロプリオニル(proprionyl)ビマトプロスト化合物IVbである。なお、0.03%ビマトプロストは、市販の育毛製品であるLATISSE(登録商標)に見られるものと同一の活性薬剤である。図6Aを参照すると、0.03%ビマトプロストが毛成長の開始に22日を要したのに対し、15−プロプリオニル(proprionyl)ビマトプロストである化合物IVbは、わずか16日で毛成長を開始した。また、0.03%ビマトプロストが完全な毛成長の達成に44日を要したのに対し、15−プロプリオニル(proprionyl)ビマトプロストである化合物IVbは、わずか32日で完全な毛成長を達成した。
実験計画
実施例12に記載の試験で使用した被験体の組織サンプルを、病理評価用として取得した。
図10A〜10E(それぞれビヒクル、ビマトプロスト、化合物IVa、化合物IIIb、化合物IVを表す)が示す通り、活性薬剤で処置したマウスは、42日目に毛周期が休止期に戻っていたのに対し、ビヒクル対照はまだ成長期の段階にあった。42日目のマウス皮膚に副作用はまったく観察されなかった。
実験計画
雌性C57BL/6Jマウス(n=2)を、処置群ごとに各時点にアッセイした。処置群:(1)ビヒクル、(2)0.03%化合物IVa、(3)0.03%化合物IIIb、(4)0.03%化合物IVb、(5)0.03%ビマトプロスト。処置投与:〜60uLの試験化合物溶液を1日に1回経皮投与する。1日目(投与から1、4、8、24時間後)、2日目(投与から24時間後)、14日目(投与から24時間後)、および42日目(14日目の投与から28日後)の各時点に、血液および皮膚の薬物動態を取得した。バイオ分析にはLC/MS−MSを使用した。
図11Aは、ビマトプロストおよび化合物IVa、IIIb、IVbの皮膚内平均濃度を示す。図11Bは、同じ時点のビマトプロストの平均濃度を示す。すべての測定時点で、ビマトプロストおよび化合物IVa、IIIb、IVbは体循環内では最も検出可能性が低かった。
化合物IVa、IIIb、IVbの変換物から観察された皮膚内平均ビマトプロスト濃度は、ビマトプロスト処置群で観察されたレベルより75%超低かった。図11Bを参照のこと。いかなる理論にも拘束されることを望むものではないが、化合物IVa、IIIb、IVbの効力が増大する理由の少なくとも一部分は、毛包を標的とした分布にあるものと考えられる。
実験計画
ヒト死体皮膚において、化合物IVa、化合物IVbからのビマトプロストの形成速度を測定した。
図12に示す通り、化合物IVa、化合物IVbのどちらも、ヒト死体皮膚と共にインキュベートするとビマトプロストを産生した。形成速度:化合物IVa(1.32pmol/分/mg、化合物IVb(5.59pmol/分/mg)。
実験計画
検査物質:皮膚採取されたエキソビボのヒト死体後方体幹皮膚を当技術分野で公知の1.0cm2フランス拡散チャンバー上に設置。検査サイズ:皮膚ドナー3名(40歳アフリカ系アメリカ人、60歳白人、72歳白人。ニューヨーク・ファイアーファイターズ・スキンバンクから取得)、各製剤、各ドナーにつき3回繰り返し。検査製剤:製剤A中に0.03%活性薬剤(化合物IVa、化合物IIIb、化合物IVb)、対照製剤:製剤A中に0.03%ビマトプロスト。投与計画:2、4、24、48時間時にレセプター液、48時間時に皮下/表皮および真皮。サンプル分析:LC/MS−MSによりビマトプロストおよび化合物IVa、IIIb、IVbを検出。
図13Aに示す通り、実験中、各活性薬剤が皮膚を浸透した。ビマトプロストと比べて、化合物IVa、IIIb、IVbの浸透性は著しく大きかった。図13Bに示す通り、化合物IVa、IIIb、IVbの浸透流束は、ビマトプロストと比べて大きかった。
図14A〜14Cは、レセプターチャンバー液内の化合物の累積濃度をヒストグラムで表したものであり、これらの図が明白に示す通り、化合物IVa、IIIb、IVbの各々がレセプターチャンバー液内でビマトプロストに転換することが観察された。
図15Aに示す通り、化合物IVa、IIIb、IVbのいずれも、ビマトプロストで観察されるのと同様に、総量が皮膚層へと分布した。さらに、化合物IVa、IIIb、IVbは皮下/表皮/真皮層内でビマトプロストに転換している。図15Bは、真皮における薬剤別の皮膚保持量をヒストグラムで表したものである。真皮の上部3分の1にある活性薬剤(化合物IVa、IIIb、IVb)の量が、ビマトプロストで観察される量より少ないことが観察された。
驚いたことに、化合物IVa、IIIb、IVbはビマトプロストより透過性が高く、レセプターチャンバー液でビマトプロスト濃度が上昇している。化合物IVa、IIIb、IVbはビマトプロストと同程度に皮下/表皮/真皮上層に分布しているが、真皮の上部3分の1では、これらの活性薬剤はビマトプロストより保持量が少ないことを示している。
理論的根拠
いかなる理論にも拘束されることを望むものではないが、ビマトプロストとそのプロドラッグが半液体の皮脂相を通って毛包まで浸透するとすれば、ビマトプロストより大きな皮脂−水分配を有するプロドラッグは、ビマトプロストで観察される毛成長効力より大きな効力を達成できると考えられる。
1〜20mgの人工皮脂を含む水溶液と含まない水溶液(1mL、2〜20ug/mL)に、ビマトプロスト、化合物IVa、化合物IIIbの各化合物を加えた。37℃で16時間、サンプルを振盪した。次に、サンプルを室温で15分間遠心分離(8000RPM)してから、水溶液を回収した。人工皮脂を含まない化合物の濃度(すなわちCo)と、人工皮脂を含む化合物の濃度(すなわちCi)について、質量分析によりサンプルを分析した。人工皮脂と水の分配係数(Ksebum)を、人工皮脂1g中の薬剤濃度を水溶液1g中の薬剤濃度で割った値として表した。計算方法は次の通りである。Ksebum=(Co−Ci)Waqueous/(Ci×Wsebum)(式1)(ここで、Wは重量を表す)
人工皮脂を次のように調製した:15%(w/w)スクアレン、15%鯨蝋、10%ヤシ油、1.5%オレイン酸、5%パルミチン酸、2.4%オレイン酸コレステロール、10%パラフィン蝋、10%オリーブ油、25%綿実油、5%パルミトレイン酸、1.2%コレステロール。半液体の融点は37℃であった。
n=4(試行回数)とする下の表6に示す通り、実験条件下では、Ksebumは、当技術分野で周知の方法で計算したLogPと相関関係があることが観察される。いかなる理論にも拘束されることを望むものではないが、特に、化合物の親油性が高いほど、人口皮脂への分配が大きいことが観察される。さらに、ビマトプロストおよび化合物IVa、IIIbの過半数が水相に残っていることが観察される。
理論的根拠。経皮投与時のビマトプロストおよび本明細書に開示される化合物の浸透、蓄積、および/または差動分布を測定するため、局所経皮投与後のマウス皮膚の外植サンプルに対して、MALDI−MS(マトリックス支援レーザー脱離イオン化質量分析)イメージングを行った。
ビマトプロストまたは本明細書に開示される化合物を1日1回、3日間投与した後、マウスの細長い皮膚片に対してMALDI−MSイメージングを実施した。3日目の化合物投与から4時間後に、マウス皮膚サンプルを回収した。ビヒクル、0.03%ビマトプロスト、3%ビマトプロスト、0.03%化合物IVa、および3%化合物IVaを用いて経皮投与した。
MALDI−MSイメージングを実施することにより、マウス皮膚に3.0%化合物IVaを局所経皮投与した後の化合物IVaとビマトプロストを選択的に検出することができる。いかなる理論にも拘束されることを望むものではないが、MALDI−MSイメージングは、3%(w/w)投与量で局所経皮投与した際の、ビマトプロストおよび本明細書に開示される化合物を検出するのに十分な感度、あるいはさらに高い感度を有すると考えられる。実際、0.03%の化合物IVaを投与した後、このアッセイシステムで化合物IVaが検出可能であったのに対し、ビヒクル対照サンプルは化合物IVaのシグナルをまったく示さなかった。
