(1)BMPファミリータンパク質を含有し、血清を含まない培養液を用いて接着培養する工程、
(2)VEGFを含有し、血清を含まない培養液を用いて接着培養する工程、
(3)造血因子を含有し、血清を含まない培養液を用いて接着培養する工程、および
(4)血清を含まない培養液を用いて浮遊培養する工程。
Description
件で樹状細胞を製造することに成功し、本発明を完成するに至った。
本発明の一つの態様は、以下の工程により多能性幹細胞を培養することを含む、多能性幹細胞より樹状細胞を製造する方法を提供することである。
(1)BMPファミリータンパク質を含有し、血清を含まない培養液を用いて接着培養する工程、
(2)VEGFを含有し、血清を含まない培養液を用いて接着培養する工程、
(3)造血因子を含有し、血清を含まない培養液を用いて接着培養する工程、および
(4)血清を含まない培養液を用いて浮遊培養する工程。
本発明の他の態様は、前記工程(1)から工程(3)における接着培養が、細胞外基質をコーティングしたディッシュで行われる、前記方法を提供することである。
本発明の他の態様は、前記細胞外基質がマトリゲルである、前記方法を提供することである。
本発明の他の態様は、BMPファミリータンパク質がBMP4である、前記方法を提供することである。
本発明の他の態様は、前記工程(2)において、培養が、さらにbFGFおよび/またはSCFを含む培地において行われる、前記方法を提供することである。
本発明の他の態様は、前記造血因子が、SCF、TPO、IL-3、Flt3-ligand、GM-CSFおよびM-CSFから成る群より選択される少なくとも一つの造血因子である、前記方法を提供することである。
本発明の他の態様は、前記工程(3)において、SCF、TPO、IL-3、Flt3-ligand を含む培養液で培養した後、さらにGM-CSF、M-CSFおよびFlt3-ligandを含む培養液で培養する、前記方法を提供することである。
本発明の他の態様は、前記工程(3)において、SCF、TPO、IL-3、Flt3-ligand を含む培養液で培養した後、さらにSCF、Flt3-ligand、GM-CSFおよびM-CSFを含む培養液で培養する、前記方法を提供することである。

本発明で使用可能な多能性幹細胞は、生体に存在するすべての細胞に分化可能である多能性を有し、かつ、増殖能をも併せもつ幹細胞であり、それには、以下のものに限定されないが、例えば胚性幹(ES)細胞、核移植により得られるクローン胚由来の胚性幹(ntES)細胞、精子幹細胞(「GS細胞」)、胚性生殖細胞(「EG細胞」)、人工多能性幹(iPS)細胞などが含まれる。好ましい多能性幹細胞は、ES細胞、ntES細胞、およびiPS細胞である。
ES細胞は、ヒトやマウスなどの哺乳動物の初期胚(例えば胚盤胞)の内部細胞塊から樹立された、多能性と自己複製による増殖能を有する幹細胞である。

液を使用し、37℃、2% CO2/98% 空気の湿潤雰囲気下でヒトES細胞を維持することができる(O. Fumitaka et al. (2008), Nat. Biotechnol., 26:215-224)。また、ES細胞は、3〜4日おきに継代する必要があり、このとき、継代は、例えば1mM CaCl2及び20% KSRを含有するPBS中の0.25% トリプシン及び0.1mg/mlコラゲナーゼIVを用いて行うことができる。
精子幹細胞は、精巣由来の多能性幹細胞であり、精子形成のための起源となる細胞である。この細胞は、ES細胞と同様に、種々の系列の細胞に分化誘導可能であり、例えばマウス胚盤胞に移植するとキメラマウスを作出できるなどの性質をもつ(M. Kanatsu-Shinohara et al. (2003) Biol. Reprod., 69:612-616; K. Shinohara et al. (2004), Cell, 119:1001-1012)。神経膠細胞系由来神経栄養因子(glial cell line-derived neurotrophic factor (GDNF))を含む培養液で自己複製可能であるし、またES細胞と同様の培養条件下で継代を繰り返すことによって、精子幹細胞を得ることができる(竹林正則ら(2008),実験医学,26巻,5号(増刊),41〜46頁,羊土社(東京、日本))。
胚性生殖細胞は、胎生期の始原生殖細胞から樹立される、ES細胞と同様な多能性をもつ細胞であり、LIF、bFGF、幹細胞因子(stem cell factor)などの物質の存在下で始原生殖細胞を培養することによって樹立しうる(Y. Matsui et al. (1992), Cell, 70:841-847; J.L. Resnick et al. (1992), Nature, 359:550-551)。
人工多能性幹(iPS)細胞は、特定の初期化因子を、DNA又はタンパク質の形態で体細胞に導入することによって作製することができる、ES細胞とほぼ同等の特性、例えば分化多能性と自己複製による増殖能、を有する体細胞由来の人工の幹細胞である(K. Takahashi and S. Yamanaka (2006) Cell, 126:663-676; K. Takahashi et al. (2007), Cell, 131:861-872; J. Yu et al. (2007), Science, 318:1917-1920; Nakagawa, M.ら,Nat. nt ES細胞は、核移植技術によって作製されたクローン胚由来のES細胞であり、受精卵由来のES細胞とほぼ同じ特性を有している (T. Wakayama et al. (2001), Science, 292:740-743; S. Wakayama et al. (2005), Biol. Reprod., 72:932-936; J. Byrne et al.
(2007), Nature, 450:497-502)。すなわち、未受精卵の核を体細胞の核と置換することによって得られたクローン胚由来の胚盤胞の内部細胞塊から樹立されたES細胞がnt ES(nuclear transfer ES)細胞である。nt ES細胞の作製のためには、核移植技術(J.B. Cibelli et al. (1998), Nature Biotechnol., 16:642-646)とES細胞作製技術(上記)との組み合わせが利用される(若山清香ら(2008),実験医学,26巻,5号(増刊), 47〜52頁)。核移植においては、哺乳動物の除核した未受精卵に、体細胞の核を注入し、数時間培養することで初期化することができる。
本発明において、樹状細胞とは、細胞突起を有し樹状あるいは樹枝状の形態を呈する細胞であって、T細胞への抗原提示能を有する免疫細胞であって、骨髄系樹状細胞、形質細胞様樹状細胞、ランゲルハンス細胞、指状嵌入細胞、ヴェ−ル細胞および真皮内樹状細胞等を含む。好ましくは、骨髄系樹状細胞であって、CD11a、MHC-classII(例えば、ヒトの場合、HLA-DR、HLA-DPおよびHLA-DQ)、CD40 、CD80およびCD86からなる表面抗原の少なくともいずれか一つのマーカーを発現する細胞である。より好ましくは、HLA-DRおよびCD83陽性ならびにCD14陰性の細胞である。

として、最小必須培養液(MEM)、ダルベッコ改変イーグル培養液(DMEM)、イスコフ改変ダルベッコ培養液(IMDM)、StemPro-34SFM (インビトロジェン社)、Stemline II(Sigma-Aldrich社)等へITSを添加した培養液もしくは血清代替物を予め含有した霊長類ES細胞培養用培養液(霊長類ES/iPS細胞用培養液、リプロセル社)、無血清培地(mTeSR、Stemcell Technology社)が例示される。より好ましい血清を含まない培養液は、ITSを含有したStemline IIである。
上記工程(2)の好ましい態様において、培地はさらに、bFGFおよび/またはSCFを含んでもよい。
うことができ、特に限定されないが、CD14 MicroBeads(#130-050-201, Miltenyi Biotec)を用いてautoMACS pro(Miltenyi Biotec)またはフローサイトメーターにより精製することができる。また、工程(4)に用いる浮遊細胞は、フローサイトメーターまたはLymphoprep(AXIS-SHIELD PoC AS)等を用いた密度勾配遠心法により死細胞を除去して用いることが望ましい。

ng/mlから70 ng/mlであり、より好ましくは、50 ng/mlである。TPOの場合は、1 ng/mlから25 ng/mlであり、好ましくは、1 ng/mlから10 ng/mlであり、より好ましくは、5 ng/mlである。Flt3-ligandの場合は、10 ng/mlから100 ng/mlであり、好ましくは、30 ng/mlから70 ng/mlであり、より好ましくは、50 ng/mlである。GM-CSFの場合は、5 ng/mlから100
ng/mlであり、好ましくは、10 ng/mlから50 ng/mlであり、より好ましくは、25 ng/mlである。M-CSFの場合は、5 ng/mlから100 ng/mlであり、好ましくは、30 ng/mlから70 ng/mlであり、より好ましくは、50 ng/mlである。IL-4の場合は、5 ng/mlから100 ng/mlであり、好ましくは、10 ng/mlから70 ng/mlであり、より好ましくは、40 ng/mlである。TNF-αの場合は、0.05 ng/mlから1 ng/mlであり、好ましくは、0.1 ng/mlから0.5 ng/mlであり、より好ましくは、0.2 ng/mlである。LPSの場合は、0.01 μg/mlから10 μg/mlであり、好ましくは、0.1 μg/mlから1 μg/mlであり、より好ましくは、0.2μg/mlである。
下記の方法により、ヒトiPS細胞(253G4)をCD14陽性の単球を経て樹状細胞へと分化誘導した後、フローサイトメトリーを用いて分析したところ、CD14陰性HLA-DR陽性細胞が含有されていることが確認された(図1)。
1. iPS細胞コロニーを1ウェルあたり5個以下のコロニー数になるようにgrowth factor-reduced Matrigelでコートした6ウェルプレート上へ播種し、mTeSR1を用いて各コロニーの直径が約1mmになるまで培養した。
2. 培養液をInsulin-Transferrin-Selenium-X Supplement (ITS)(#51500-056; Invitrogen)および20 ng/mLのBMP4(#314-BP-010; R&D Systems)を含有するStemline II serum-free hematopietic stem cell expansion medium(Stemline II)(#S0192; Sigma-Aldrich)へ交換し、4日間培養した。
3. 培養液をITS 、50 ng/mLのVEGF(#293-VE-050; R&D Systems)および50 ng/mLのSCF(#255-SC-050; R&D Systems)を含有するStemline IIへ交換し、2日間培養した。
4. 培養液をITS 、50 ng/mLのSCF、50ng/mLのIL-3(#203-IL-050; R&D Systems)、5 ng/mLのTPO(#288-TPN-025; R&D Systems)および50 ng/mLのFlt-3 ligand(#308-FK-025; R&D Systems)を含有するStemline IIへ交換し、7〜9日間培養した。
5. 培養液をITS 、50 ng/mLのFlt-3 ligand 、25 ng/mL のGM-CSF、50 ng/mLのM-CSFを含有するStemline IIへ交換し、15日間培養した。この時、5日おきに新しい培養液へ交換した。
6. 培養上清中の浮遊細胞を回収し、Lymphoprep(#1114740; AXIS-SHIELD PoC AS)を用いて密度勾配遠心法により死細胞を除去した後、CD14 MicroBeads(#130-050-201, Miltenyi Biotec)を用いてautoMACS pro(Miltenyi Biotec)によりCD14陽性細胞を分離した。3×104個の分離したCD14陽性細胞をUltra-Low Attachment Surface (#3471; CORNING)でコートした6ウェルディッシュを用いて、GM-CSF(50 ng/mL) +IL-4(40 ng/mL) とITSを含有する Stemline II中で6日間浮遊培養を行った。
下記の方法により、ヒトiPS細胞(253G4)を樹状細胞へ分化誘導を行い、メイ・ギムザ染色を行ったところ、得られた細胞は末梢血由来の樹状細胞と類似した形状であることが確認された(図2)。
1. iPS細胞コロニーを1ウェルあたり5個以下のコロニー数になるようにgrowth factor-reduced Matrigelでコートした6ウェルディッシュ上へ播種し、mTeSR1を用いて各コロニーの直径が約1mmになるまで培養した。
2. 培養液をITSおよび20 ng/mLのBMP4を含有するStemline IIへ交換し、4日間培養した。
3. 培養液をITS 、50 ng/mLのVEGFおよび50 ng/mLのSCFを含有するStemline IIへ交換し、2日間培養した。
4. 培養液をITS 、50 ng/mLのSCF、50ng/mLのIL-3、5 ng/mLのTPOおよび50 ng/mLのFlt-3 ligandを含有するStemline IIへ交換し、7〜9日間培養した。
5. 培養液をITS 、50 ng/mLのFlt-3 ligand、25 ng/mL のGM-CSFおよび50 ng/mLのM-CSFを含有するStemline IIへ交換し、15日間培養した。
6. 培養上清中の浮遊細胞を回収し、Lymphoprepを用いて密度勾配遠心法により死細胞を除去した後、CD14 MicroBeadsを用いてautoMACS proによりCD14陽性細胞を分離した。3×104個の分離したCD14陽性細胞をUltra-Low Attachment Surfaceでコートした6ウェルディッシュを用いて、ITS, 25 ng/mL のGM-CSFおよび40 ng/mL のIL-4を含有するStemline II中で5日間浮遊培養を行った。さらに、培養液へ0.2ng/mL のTNF-α と1.0 μg/mlのLPSを添加し、2日間培養を続けた。
下記の方法により、ヒトiPS細胞(253G4)をCD14陽性の単球を経て樹状細胞へと分化誘導した後、フローサイトメトリーを用いて分析した(図3および4)。その結果、工程6にてTNF-αおよびLPSを添加した方が、HLA-DRおよびCD83陽性細胞がより多く含まれていた。つまり、TNF-αおよびLPSを含む培養液で培養する工程により樹状細胞が成熟することが示唆された。
さらに、ナイーブT細胞を、iPS細胞から分化させた単球、未成熟樹状細胞または成熟樹状細胞と接触させた後の、増殖活性を3Hチミジン取り込みアッセイで調べた(図5)。その結果、iPS細胞から分化させた成熟樹状細胞はナイーブT細胞に対して増殖刺激能を有することがわかった。
1. iPS細胞コロニーを1ウェルあたり5個以下のコロニー数になるようにgrowth factor-reduced Matrigelでコートした6ウェルディッシュ上へ播種し、mTeSR1を用いて各コロニーの直径が約1mmになるまで培養した。
2. 培養液をITSおよび20 ng/mLのBMP4を含有するStemline IIへ交換し、4日間培養した。
3. 培養液をITS 、50 ng/mLのVEGF 25ng/ml bFGF (#133-FB-025; R&D Systems)および50
ng/mLのSCFを含有するStemline IIへ交換し、2日間培養した。
4. 培養液をITS 、50 ng/mLのSCF、50ng/mLのIL-3、5 ng/mLのTPOおよび50 ng/mLのFlt-3 ligandを含有するStemline IIへ交換し、7〜9日間培養した。
5. 培養液をITS、50 ng/mLのFlt-3 ligand 、25 ng/mL のGM-CSFおよび50 ng/mLのM-CSFを含有するStemline IIへ交換し、15日間培養した。
6. 培養上清中の浮遊細胞を回収し、Lymphoprepを用いて密度勾配遠心法により死細胞を除去した後、CD14 MicroBeadsを用いてautoMACS proによりCD14陽性細胞を分離した。1.5×106個の分離したCD14陽性細胞(単球として使用)を、Ultra-Low Attachment Surfaceでコートした6ウェルディッシュを用いて、(i)ITS, 25 ng/mL のGM-CSFおよび40 ng/mL のIL-4を含有するStemline IIで7日間浮遊培養(未成熟樹状細胞として使用)、または(ii)ITS, 25 ng/mL のGM-CSF、40 ng/mL のIL-4を含有するStemline II中で5日間浮遊培養し、続いて0.2ng/mL のTNF-αおよび1 μg/mLのLPSを加えてさらに2日間浮遊培養した(成熟樹状細胞として使用)。
分化効率が変化するかどうかについて他の基礎培地を評価した。その結果、次の方法でもiPS細胞から樹状細胞を得ることができた。
1. ES細胞またはiPS細胞コロニーを1ウェルあたり5個以下のコロニー数になるようにgrowth factor-reduced Matrigelでコートした6ウェルディッシュ上へ播種し、mTeSR1を用いて各コロニーの直径が約1mmになるまで培養した。
2. 培養液を80 ng/mLのBMP4を含有するStemPro-34SFM (#10639-011; Invitrogen)へ交換し、4日間培養した。
3. 培養液を100ng/mLのVEGF、25ng/ml のbFGFおよび10 ng/mLのSCFを含有するStemPro-34SFMへ交換し、2日間培養した。
4. 培養液を50 ng/mLのSCF、50ng/mLのIL-3、5 ng/mLのTPOおよび50 ng/mLのFlt-3 ligandを含有するStemPro-34SFMへ交換し、7〜9日間培養した。
5. 培養液を50 ng/mLのFlt-3 ligand、25 ng/mL のGM-CSF、50 ng/mLのM-CSFを含有するStemPro-34SFMへ交換し、3日から15日間培養した。この時、5日おきに新しい培養液へ交換した。
6. 培養上清中の浮遊細胞を回収し、Lymphoprepを用いて密度勾配遠心法により死細胞を除去した後、CD14 MicroBeadsを用いてautoMACS proによりCD14陽性細胞を分離した。1.5×106個の分離したCD14陽性細胞をUltra-Low Attachment Surfaceでコートした6ウェルディッシュを用いて、25 ng/mL のGM-CSFおよび40 ng/mL のIL-4を含有するStemPro-34SFM中で浮遊培養を行った。浮遊培養開始から5日目に、培養液へ1 μg/mL のLPSおよび0.2ng/mL のTNF-α を添加し、さらに2日間培養を続けた。
Claims (16)
- 以下の工程により多能性幹細胞を培養することを含む、多能性幹細胞より樹状細胞を製造する方法;
(1)BMPファミリータンパク質を含有し、血清を含まない培養液を用いて接着培養する工程、
(2)VEGFを含有し、血清を含まない培養液を用いて接着培養する工程、
(3)造血因子を含有し、血清を含まない培養液を用いて接着培養する工程、および
(4)血清を含まない培養液を用いて浮遊培養する工程。 - 前記工程(1)から工程(3)における接着培養が、細胞外基質をコーティングしたディッシュで行われる、請求項1に記載の方法。
- 前記細胞外基質がマトリゲルである、請求項2に記載の方法。
- 前記BMPファミリータンパク質が、BMP4である、請求項1に記載の方法。
- 前記工程(2)において、培養が、さらにbFGFおよび/またはSCFを含む培地において行われる、請求項1に記載の方法。
- 前記造血因子が、SCF、TPO、IL-3、Flt3-ligand、GM-CSFおよびM-CSFから成る群より選択される少なくとも一つの造血因子である、請求項1に記載の方法。
- 前記工程(3)において、SCF、TPO、IL-3、Flt3-ligand を含む培養液で培養した後、さらにGM-CSF、M-CSFおよびFlt3-ligandを含む培養液で培養する、請求項6に記載の方法。
- 前記工程(3)において、SCF、TPO、IL-3、Flt3-ligand を含む培養液で培養した後、さらにSCF、Flt3-ligand、GM-CSFおよびM-CSFを含む培養液で培養する、請求項6に記載の方法。
- 前記工程(4)の浮遊培養において、GM-CSFおよびIL-4を含有する培養液を用いる、請求項1に記載の方法。
- 前記工程(4)の浮遊培養において、さらにLPSおよびTNF-αを含む培養液で培養する工程を含む、請求項9に記載の方法。
- 前記樹状細胞が、HLA-DR陽性ならびにCD14陰性の樹状細胞である、請求項1に記載の方法。
- 前記樹状細胞が、さらにCD83陽性である、請求項11に記載の方法。
- 前記工程(1)が、4日間である、請求項1に記載の方法。
- 前記工程(2)が、2日間である、請求項1に記載の方法。
- 前記多能性幹細胞が、ヒトiPS細胞である、請求項1に記載の方法。
- 請求項1〜15のいずれかに記載の方法で製造された樹状細胞。
