前記VEGF結合断片(例えば、VIG2またはこれを含むポリペプチドなど)は前記抗c−Met抗体以外の他の抗c−Met抗体と結合する場合にも抗体効能を増進させるだけではなく、副作用(agonism)を顕著に減少させることが確認された(図5参照)。
1.1.c−Metに対するマウス抗体「AbF46」の産生
1.1.1.マウスの免疫化
ハイブリドーマ細胞株の開発に必要な免疫化されたマウスを得るために、5匹のマウスに1匹当たりに100μgのヒトのc−Met/Fc融合タンパク質(R&Dシステムズ社製)と同量の完全フロインドアジュバント (Freund'sadjuvant)を混合して4−6週齢のBALB/cマウス(日本SLC社製)の腹腔内に注射した。2週後に、上記の方法と同様にして、前記抗原として用いられたヒトのc−Met/Fc融合タンパク質を先に注射した量の半分である50μgを同量の不完全フロインドアジュバントと混合してマウスの腹腔内に注射した。一週間後に最後の追加免疫が行われ、3日後に前記マウスの尻尾から採血して血清を得た後、1/1000でPBSに希釈してELISAでc−Metを認知する抗体の力価(titer)が増大されることを確認した。上記の結果をもって抗体の量が十分に得られるマウスを選別して下記の細胞融合過程を行った。
細胞融合実験3日前に50μgのリン酸緩衝生理食塩水(PBS:phosphate buffered solution)にヒトのc−Met/Fc融合タンパク質混合物をBALB/cマウス(日本SLC社製)の腹腔内に注射し、免疫化されたマウスを麻酔した後に身体の左側に位置する脾臓を摘出した。摘出した脾臓をメッシュで磨って細胞を分離し、培養培地(ダルベッコ改変イーグル培地(DMEM)、GIBCO、インビトロゲン社製)と混合して脾臓細胞懸濁液を作成した。前記懸濁液を遠心分離して細胞層を回収した。前記得られた脾臓細胞1×108個と骨髓腫細胞(Sp2/0)1×108個を混合した後、遠心分離して細胞を沈殿させた。前記遠心分離された沈殿物を徐々に懸濁し、37℃で1分間DMEM中の45%ポリエチレングリコール(PEG)(1mL)で処理した後、1mLの培養培地(DMEM)に懸濁した。次いで、細胞を10mLのDMEMに10分間以上浸し、その後37℃で5分間放置した。そして、DMEMで50mLにし、再び遠心分離した。細胞沈殿物を分離培地(HAT培地)に約1〜2×105/mLで再懸濁し、96ウェルプレートに0.1mLずつ分注した後、37℃の二酸化炭素培養器において培養してハイブリドーマ細胞群を製作した。
前記参考例1.1.2に従い製造されたハイブリドーマ細胞群から、c−Metタンパク質にのみ特異的に反応するハイブリドーマ細胞を選別するために、ヒトのc−Met/Fc融合タンパク質とヒトのFcタンパク質を抗原としたELISA分析方法を用いてスクリーニングした。
前記参考例1.1.3において得られたハイブリドーマ細胞を無血清培地において培養し、培養液からモノクローナル抗体を産生および精製した。
一般に、マウス抗体は、治療目的でヒトに注入されたときに免疫拒絶反応を示す可能性が高い。そのため、これを解決するために、前記実施例1に従い製作されたマウス抗体AbF46から、抗原結合に関連する変異領域(variable region)を除く不変領域(constant region)をヒトIgG1抗体の配列で置換したキメラ型抗体chAbF46を製作した。
1.3.1.重鎖のヒト化(Heavy chain humanization)
H1−heavyおよびH3−heavyの2種のデザインのために、先ず、Ig Blast(http://www.ncbi.nlm.nih.gov/igblast/)を介して前記参考例1.2において精製されたマウス抗体AbF46のVH遺伝子と最も相同性が高いヒトの生殖細胞系遺伝子を分析した。その結果、VH3−71がアミノ酸レベルにおいて83%の相同性を有することを確認し、マウス抗体AbF46のCDR−H1、CDR−H2、CDR−H3をKabat番号付けで定義し、マウス抗体AbF46のCDR部分がVH3−71の骨格(フレームワーク)に取り込まれるようにデザインした。このとき、30番(S→T)、48番(V→L)、73番(D→N)、78番(T→L)のアミノ酸は、元のマウスAbF46抗体のアミノ酸配列に復帰突然変異した。次いで、H1はさらに83番(R→K)と84番(A→T)のアミノ酸に突然変異を与えて最終的にH1−heavy(配列番号40)とH3−heavy(配列番号41)を構築した。
H1−light(配列番号43)およびH2−light(配列番号44)の2種のデザインのために、Ig Blast(http://www.ncbi.nlm.nih.gov/igblast/)を介して、マウス抗体AbF46のVL遺伝子と最も相同性が高いヒト生殖細胞系遺伝子を分析した。その結果、VK4−1がアミノ酸レベルにおいて75%の相同性を有することを確認し、マウス抗体AbF46のCDR−L1、CDR−L2、CDR−L3をKabat番号付けで定義し、マウス抗体AbF46のCDR部分がVK4−1のフレームワークに取り込まれるようにデザインした。このとき、H1−lightは、36番(Y→H)、46番(L→M)、49番(Y→I)の3個のアミノ酸を復帰突然変異し、H2−lightは、49番のアミノ酸(Y→I)1個のみを復帰突然変異して構築した。
huAbF46抗体の重鎖可変領域および軽鎖可変領域を用いてhuAbF46抗体のscFvを製作するための遺伝子をデザインした。それぞれの重鎖可変領域および軽鎖可変領域を「VH−リンカー−VL」の形にし、前記リンカーは「GLGGLGGGGSGGGGSGGSSGVGS」(配列番号54)のアミノ酸配列を有するようにデザインした。このようにしてデザインされたhuAbF46抗体のscFvをコードするポリヌクレオチド(配列番号55)をバイオニア社に依頼して合成し、これを発現させるためのベクターを配列番号56に示す。
1.5.1.標的CDRの選定およびプライマの製作
huAbF46抗体の親和性成熟のために6個の相補性決定部位(complementary determining region、CDR)を前記製作されたマウス抗体AbF46から「Kabat番号付け」によって定義し、それぞれのCDRは下記の表1に示す通りである。
CDRのランダムな配列の取り込みを用いた抗体ライブラリー遺伝子の構築は、前記参考例1.5.1などの方法により製作されたプライマーを用いて行った。鋳型としてhuAbF46抗体のscFvを含むポリヌクレオチドを用いて、図1に示す方法と同様にして2個のPCR断片を製作し、これをオーバーラップ・エクステンションPCR(overlap extension PCR)方法を用いて、所望のCDRのみがそれぞれ突然変異されたhuAbF46抗体のscFvライブラリー遺伝子を確保して製作された6個のCDRをそれぞれ標的とするライブラリーを構築した。
前記構築されたライブラリーからc−Metへのライブラリーの結合力を向上させた後、それぞれの個別クローンからscFvの遺伝子配列を分析した。確保された遺伝子配列はそれぞれ下記の表2に示す通りであり、これをIgG形式に変換した。下記のクローンのうちから、L3−1、L3−2、L3−3、L3−5から産生された4種の抗体を選別して後続の実験を行った。
選別された4種の抗体の重鎖をコードするポリヌクレオチドは「EcoRI−signal sequence−VH−NheI−CH−XhoI」(配列番号38)から構成され、重鎖の場合、親和性成熟後に抗体のアミノ酸が変更されなかったため、huAbF46抗体の重鎖をそのまま用いた。但し、ヒンジ領域はヒトIgG1のヒンジではないU6−HC7ヒンジ(配列番号57)で置換した。軽鎖は「EcoRI−signal sequence−VL−BsiWI−CL−XhoI」から構成されるようにそれぞれデザインして遺伝子を合成し、親和性成熟後に選別された前記4種の抗体の軽鎖可変領域を含んでコードするポリヌクレオチド(配列番号58〜配列番号61)をバイオニア社に依頼して合成した。次いで、インビトロゲン社製のOptiCHOTM抗体発現キット(Cat no.12762−019)に含まれているpOptiVECTM−TOPO TAクローニングキットに前記重鎖に相当するヌクレオチド配列を含むDNA断片(配列番号38)を、pcDNATM3.3−TOPO TAクローニングキット(Cat no.8300−01)に前記軽鎖に相当するヌクレオチド配列を含むDNA断片(L3−1由来のCDR−L3を含むDNA断片:配列番号58、L3−2由来のCDR−L3を含むDNA断片:配列番号59、L3−3由来のCDR−L3を含むDNA断片:配列番号60、L3−5由来のCDR−L3を含むDNA断片:配列番号61)をそれぞれEcoRI(NEB、R0101S)とXhoI(NEB、R0146S)の制限酵素を用いてクローニングすることにより、親和性成熟された抗体の発現のためのベクターを構築した。
前記参考例1.7において選別された4種の抗体のうち、c−Metとの結合親和性が最も高く、Aktリン酸化およびc−Met分解の度合いが最も低く測定されたhuAbF46−H4−A1を対象として、ヒンジ領域または不変領域およびヒンジ領域が置換された抗体を製作した。
前記参考例1において製作された抗c−Met抗体を基に重鎖C末端にリンカーが連結されるように融合した。次いで、リンカーの末端にVEGF受容体1を構成するアミノ酸のうちIg2ドメイン、すなわち、129番目から229番目までのアミノ酸を連続して融合してcMetとVEGFに同時に結合可能な抗体を製作した(図1参照)。

前記製作されたc−Met抗体の重鎖とIg2ドメイン(VIG2)を連結するために、GGGGSが繰り返される構造の「GGGGS」(G4S)(配列番号116)、「GGGGSGGGGS」((G4S)2)(配列番号117)、または「GGGGSGGGGSGGGGSGGGGS」((G4S)4)(配列番号118)の3種類のリンカーをデザインし、これをc−Met抗体とVEGF受容体1のIg2ドメイン(VIG2)との間に位置させた。前記抗c−Met抗体の軽鎖をコードする遺伝子、および前記抗c−Met抗体の重鎖、前記リンカー、および前記VEGF結合断片をコードする遺伝子をバイオニア社に依頼して合成し、このとき、最終遺伝子の最後に終止コドン(TGA)を挿入した。このようにして合成された遺伝子をpOptivecベクター(インビトロゲン社)にEcoRI/XhoIクローニングサイトを用いて重鎖発現ベクターを製作した。軽鎖発現ベクターは、前記L3−1Yの製作時に用いられたベクターをそのまま用いた。
2.1.親和性検査
前記製作された二重特異性融合タンパク質の結合速度をBiacore T100インスツルメント(GEヘルスケア社製)を用いて分析した。ヒトFab結合剤(GEヘルスケア社製)をCM5チップ(GEヘルスケア社製、Cat.No.BR−1005−30)の表面に製造者の説明書に従って固定化した。約90〜120RUのL3−1YまたはMV10AYを捕獲し、様々な濃度のc−Met(R&Dシステムズ社製、Cat.No.358−MT/CF)を前記捕獲された抗体に注入した。ここに10mMグリシン−HCl(pH1.5)溶液を注入して前記表面を再生させた。結合速度を測定するために、前記実験において得られたデータをBIAevaluationソフトウェア(GEヘルスケア社製、Biacore T100 evaluation software)を用いて適用した。
相対的なc−Metの総量は、抗体がc−Metに結合した場合、抗体の内在化によって、c−Metの分解が起こることを利用して、c−Metの総量の増減を把握して抗体の影響を評価するものである。c−MetとHGFの結合ががん細胞の成長を促すことは既に知られており、したがって、合計のc−Met量が減少すれば、がん細胞の成長も低下することに着目したものである。
治療用抗体における安全性と効能をメカニズム基盤実験で検証した。安全性を調べるために、AKTというキナーゼのリン酸化の度合いを定量的ELISA方法により測定した。AKTによって調節されている細胞作用は、細胞増殖、細胞生存、細胞の大きさの調節、可用栄養分への反応性、中間代謝過程、血管新生(angiogenesis)、組織浸潤浸潤(tissue invasion)などを含む。これらのあらゆる過程はがんの特徴を代表するものであり、数多くの腫瘍発生タンパク質(oncoprotein)と腫瘍発生抑制物質(tumor suppressor)はAKT経路上において相互交差的に影響を及ぼし、シグナル伝達と古典的な代謝調節の連結点において細胞の作用を微細に調節する機能を行う。このため、AKTの活性型であるリン酸化されたAKTの度合いが高いほど、がん細胞が一層高い活性を示し、この実施例においては、抗体が陽性対照抗体5D5を処理したときに比べてAKTのリン酸化をどれほど阻害できるかについて検証した。
ヒト胃がん細胞株MKN45(JCRB0254)をヘルス・サイエンス・リサーチ・リソース・バンク(HSRRB、日本、新宿)から入手した。前記細胞株を10%(v/v)のウシ胎児血清(FBS)(GIBCO、Cat.#16000−044)と1%(v/v)のペニシリン/ストレプトマイシン(GIBCO、Cat.#15410−122)を含むRPMI1640培地(GIBCO、Cat.#11875−119)において培養した。細胞株を5%のCO2を含む湿潤大気下の37℃において培養し、細胞が培養容器内で過密状態(confluence)になる前に継代培養した。CEDEX Analyzer(ロシュ・ダイアグノスティックス社製)を用いて細胞数を測定した。抗体処理(in vitro)による腫瘍細胞の増殖を調べるために、Celltiter Glo(CTG:プロメガ社製)発光分析法(ルミネセンス測定法)を用いた。
c−Met単独標的抗体がc−Metに結合する場合、c−Metによる不所望のアゴニズム(拮抗)現象が見られる。これは抗体の安全性と関連するため、二重特異性を有することに伴い、c−Met単独標的抗体の不所望のアゴニズム性質が増大されるか否かを評価するためにNCI−H441細胞株を用いて試験した。NCI−H441細胞株は、c−Met単独標的抗体の投与時に、下記の特定の条件下でアゴニズムによる細胞分裂を示すという性質がある。
HuVEC細胞株(ATCC)をHGF(R&Dシステムズ社製)0.4μg/mlとともにEGM2 bulletkit培地(Lonza Cat.CC−3162)において培養した。細胞を5%のCO2を含む湿潤大気下の37℃でインキューベーティングした後に細胞が培養容器内で過密状態(confluence)になる前に継代培養した。CEDEX Analyzer(ロシュ・ダイアグノスティックス社製)を用いて細胞数を測定した。抗体処理(in vitro)による腫瘍細胞成長阻害の度合いを確認するためにXcelligence Real time cell analyzer(ロシュ社製)を用いてリアルタイム細胞分析法を行った。前記分析法は製造者の説明書に従って行った。略述すると、前記培養されたHuVEC細胞を16−ウェルE−プレートの上に1ウェル当たりに1×104cellsの濃度で分注し、24時間後にインキュベーション培地を除去し、無血清培地(EBM培地、ロンザ社製)で希釈された抗体(100uL、最終的な抗体濃度10μg/mL)を処理した。発生するインピーダンス値をリアルタイムにて測定した。リアルタイム分析は、48時間かけて行った。
3.1.親和性(affinity)検査
VEGFを標的とする二重特異性抗体の結合動態をBiacore T100インスツルメント(GEヘルスケア社製)を用いるSPR方法によって測定した。hVEGF(GEヘルスケア社製、28−9583−25)をCM5チップの表面に製造者の説明書に従って固定化した。約90RUの様々な濃度(6種類の濃度)のMV10AYを捕獲した。1M NaCl+30mM NaOHを30μl/分の流速で60秒間注入することにより、前記表面を再生した。結合動態を決定するために、BIAevaluationソフトウェア(GEヘルスケア社製)を用いて前記実験において得られたデータをフィットした(KD=0.45nM)。
前記得られた結果を表5に示す。
Xcelligenceリアルタイムセルアナライザ(ロシュ社製)を用いて抗体処理(in vitro)によるHUVEC増殖の度合いを調べて血管新生抑制効果を調べてみた。前記分析は、製造者の説明書に従って行った。略述すると、HuVEC細胞株(ATCC)をVEGF(R&Dシステムズ社製)0.4μg/mlとともにEGM2 bulletkit培地(Lonza Cat.CC−3162)において培養した。前記培養されたHuVEC細胞を16−ウェルE−プレートの上に各ウェル当たりに1×104cellsの濃度で分注し、24時間後にインキュベーション用培地を除去し、無血清培地(EBM培地、ロンザ社製)で希釈された抗体(100μL、最終濃度10μg/mL)を処理した。発生するインピーダンス値をリアルタイムにて測定した。リアルタイム分析は96時間かけて行った。

抗体処理(in vitro)による細胞浸潤能の阻害有無を確認するために、上部チャンバと下部チャンバに分けられて上部チャンバにコラーゲンがコードされた細胞浸潤能分析用プレート(BioCoat Growth Factor Reduced MATRIGEL Invasion Chamber;BDサイエンス社製、Cat#.354483)を用いて細胞浸潤能(invasion)を試験した。分析は、製造者の説明書に従って行った。略述すれば、上部チャンバにはHUVEC細胞(ATCC;5×104cells/well)を分注し、下部チャンバには抗体(10μg/ml)を処理し、37℃で培養した(VEGFは、下部チャンバに添加、0.4μg/ml)。24時間後に、上部チャンバのコラーゲン層を貫いて下部チャンバに下降した細胞をカルセインで染色して蛍光顕微鏡(ツァイス社製)で観察した。
抗体処理(in vitro)による細胞浸潤能の阻害有無を確認するために、上部チャンバと下部チャンバに分けられて上部チャンバにコラーゲンがコードされた細胞浸潤能分析用プレート(BioCoat Growth Factor Reduced MATRIGEL Invasion Chamber;BDサイエンス社製、Cat#.354483)を用いて細胞浸潤能を試験した。分析は、製造者の説明書に従って行った。
c−MetとVEGFは両方とも細胞成長だけではなく、がんの転移にも関連する因子である。それぞれの阻害剤を併用投与するときにがん細胞転移能と関連する細胞の移動に相乗効果を示すか否かを試験した。抗体処理(in vitro)による転移能阻害能を評価するために下記の実験を行った。細胞の移動はがんの転移に直接的に関与するため、下記の分析法は細胞の転移能を評価する方法として汎用される方法である。
c−Met/HGFとVEGFをそれぞれ抑制するときに抗がん効果が得られると知られているU87−MG脳がん細胞株を用いて、生体内抗がん効果試験を行った。U87−MG脳がん細胞株(中国科学院の細胞銀行から購入;ATCC Cat.No.HTB−14)2.5×106個(100μL)をBALB/cヌードマウス(雄性、6−7週齢、20−25gram)に皮下注射した後、1週間が経過した後に治療群(MV10AY(■)、MV10AY U3 HC9/IgG1(□)、またはMV10AY U3 HC9/IgG2(○);投与量:それぞれ0.2mg/kg(点線)または1mg/kg(実線);投与時期:3−4日おきにまたは10日おきに;投与経路:静脈注射)と対照群(ビヒクル:PBS 0.2mL、週1回静脈注射で投与)に分けて薬物投与を始めた。各群あたりのマウスは15匹にした。腫瘍の大きさを1週につき2回測定し、腫瘍の大きさ(tumor volume)は{(長径(mm))×(短径(mm))2}/2によって計算した。治療を始めてから30日後にマウスを安楽死させた後に腫瘍を摘出し、腫瘍の重さを測定して、前記測定された腫瘍の大きさとともに抗がん効果の評価に用いた。
c−Met単独標的抗体に比べてc−MetとVEGFを同時に標的とする二重特異性抗体がより向上した抗がん効果を示すか否かを調べるために、NCI−H441細胞株を用いて生体内抗がん効果実験を行った。NCI−H441は、c−Met標的による抗がん効果とアゴニズム効果を両方とも示す細胞株であり、包括的な抗がん効果を示すのに最適な細胞株であると決定した。NCI−H441肺がん細胞株(ATCCから購入、Cat.# HTB−174)5×106個(100uL)をBALB/cヌードマウス(雄性、5−6週齢、20−25g)に皮下注射した後、1週間が経過した後に治療群(単独標的抗体L3−1Y、二重特異性抗体それぞれ5mg/kg、週1回投与)と対照群(ビヒクル:PBS 0.2mL、週1回静脈注射で投与)に分けて薬物投与を始めた。各群当たりのマウスは15匹にした。腫瘍の大きさを1週につき2回測定し、腫瘍の大きさは{(長径(mm))×(短径(mm))2}/2により計算した。治療を始めてから26日後にマウスを安楽死させた後に腫瘍を摘出し、腫瘍の重さを測定して、前記測定された腫瘍の大きさとともに抗がん効果評価に用いた。
前記得られた結果を図4Bに示す。図4Bの上部は、経時的な腫瘍の大きさを示し、下部は、実験終了後に最終的に摘出された腫瘍の重さを示す。図4Bに示すように、投与を始めてから1週間後から両治療群においていずれも統計的に有意的な抗がん効果が観察された。特に、L3−1Yに比べてMV10AY治療時よりも強力な抗がん効果が得られた(上部グラフは、分散分析法(ANOVA)により繰り返し測定した結果を示し、****(p<0.0001)、下部グラフは、ダネットの多重比較検定を用いた一元配置分散分析法(1−way ANOVA Dunnett''multiple comparison test)によるビークル群に対する分析結果を示す、*(p<0.05)、***(p<0.0001))。
VIG2を融合した場合、上記の例において示すL3−1やL3−1Y、L3−1YIgG2に加えて他のCDRを有する抗c−Met抗体の効能も改善されるか否かを確認した。

配列番号68のアミノ酸配列、配列番号68の21番目から240番目までのアミノ酸配列、配列番号70、配列番号70の21番目から240番目までのアミノ酸配列、または配列番号108のアミノ酸配列を含む軽鎖とからなる抗体である、請求項7〜9のいずれか1項に記載の融合タンパク質。 - 前記抗c−Met抗体は、配列番号112または配列番号114のアミノ酸配列を含む重鎖および配列番号113または配列番号115のアミノ酸配列を含む軽鎖を含む、請求項1〜10のいずれか1項に記載の融合タンパク質。
- 前記抗c−Met抗体は、モノクローナル抗体である、請求項1〜11のいずれか1項に記載の融合タンパク質。
- 前記抗c−Met抗体は、マウス由来の抗体、マウス−ヒトキメラ抗体、ヒト化抗体またはヒト抗体である、請求項1から請求項11のうちのいずれか1項に記載の融合タンパク質。
- 前記抗c−Met抗体の抗原結合断片は、scFv、(scFv)2、Fab、Fab'およびF(ab')2からなる群から選ばれる、請求項1から請求項11のうちのいずれか1項に記載の融合タンパク質。
- 請求項1から請求項11のうちのいずれか1項に記載の融合タンパク質を含む、c−MetとVEGFの二重特異性抗体。
- 請求項15に記載のc−MetとVEGFの二重特異性抗体を有効成分として含む、c−MetまたはVEGF誘発疾病の予防または治療用組成物。
- 前記c−MetまたはVEGF誘発疾病は、がん、妊娠糖尿病、糖尿病性網膜症、黄斑変性である、請求項16に記載の予防または治療用組成物。
- 請求項15に記載のc−MetとVEGFの二重特異性抗体を含む、c−MetまたはVEGF誘発疾病の診断用組成物。
- 抗c−Met抗体またはその抗原結合断片にVEGF結合断片を連結する工程を含む、融合タンパク質の製造方法。
- 前記VEGF結合断片は、配列番号110のVIG2、または配列番号109のアミノ酸のうち配列番号110のVIG2を含んで連続する102〜1338個のアミノ酸からなるポリペプチドである、請求項19に記載の製造方法。
- 前記抗c−Met抗体またはその抗原結合断片にVEGF結合断片を連結する工程において、c−Met標的抗体またはその断片とVEGF結合断片とがリンカーを介して結合する、請求項19または20に記載の製造方法。
- 前記リンカーは、1〜100個のアミノ酸からなるペプチドリンカーである、請求項21に記載の製造方法。
