Brocksら(Immunotechnology 3(3) 173-184 (1997))は、TNF受容体拮抗性の一価および二価のscFv誘導体について記載している。
Armourら(European Jounal of Immunology 29(8) 2613-2624 (1999))は、FcγRIへの結合を減少させるための突然変異を有する組換え型ヒトIgG1分子について記載している。
好ましくは、これは、Fc領域を糖鎖工学によりエフェクタ機能を下方制御することによって達成される。
該特に好ましい結合エピトープは、huTNFR1のN末端領域に1〜70個のアミノ酸によって表されている。
好ましくは、該抗体は、少なくとも2つの結合部位によってhuTNFR1に特異的に結合する。特に、該抗体は、少なくとも二価である(すなわち、2つの結合価によって同じ抗原もしくはエピトープに結合する)か、または二重特異性である(2種類の異なる抗原もしくはエピトープに結合する)。
一側面によれば、本発明に係る抗体と、薬学的に許容される担体とを含む医薬品が提供される。
好ましくは、該発現系は、CHO産生細胞株を用いる。具体的な側面によれば、抗TNF治療薬を用いる治療に代わるものとして、作動性TNFR1シグナル伝達複合体を形成することなくTNF拮抗薬として使用されるIgG1型の抗huTNFR1抗体が提供される。生物学的TNF拮抗薬としてもみなされているそのようなTNF拮抗薬は典型的に、関節、皮膚および腸の慢性非伝染性炎症の発症へのTNF機能の生物学的関連性が分かっている治療的使用のために提供される。
具体的には、該抗体は、自己免疫疾患、関節リウマチ、乾癬、乾癬性関節炎、若年性関節炎、強直性脊椎炎、クローン病、多発性硬化症、鬱血性心不全、代謝性疾患、サイトカイン放出症候群、敗血症性ショック、脳卒中、アルツハイマー病およびパーキンソン病を含む急性および慢性神経変性疾患、または癌の治療における使用のために提供される。

本発明のさらに別の態様では、huTNFR1抗体は、生物学的に許容される化合物との特異的相互作用を可能にするさらなるタグを含む。huTNFR1抗体のhuTNFR1への結合またはヒトに投与された場合の免疫原性応答に対して悪影響が全くないか容認できるものである限り、本発明で使用可能なタグに関して特定の制限はない。好適なタグの例としては、Hisタグ、Mycタグ、FLAGタグ、Strepタグ、カルモジュリンタグ、GSTタグ、MBPタグおよびSタグが挙げられる。

機能的に活性なFc変異体は、上記配列を変えることによって得てもよく、かつ抗体を安定化させるか長期の半減期を与える能力などの各配列によって呈されるものに類似した生物学的活性を有することを特徴とする。本発明に係る抗体で使用される好ましいFc変異体は、Fcエフェクタ機能を低下させるための突然変異体を含む。
本発明に従って使用されているFc領域は、特異的突然変異または発現系の選択に応じて、グリコシル化されていてもグリコシル化されていなくてもよい。
本発明に係る抗体の産生および特性評価方法は、当該技術分野でよく知られている。好ましい態様では、抗体変異体を産生し、かつhuTNFR1を発現する細胞を用いる1種以上の細胞系アッセイまたは生体内アッセイを用いて、所定の特性についてスクリーニングする。そのようなアッセイでは、拮抗活性および作動活性を測定するために、例えばTNFαの存在および非存在下で細胞を結合することができるように、通常は本抗体を外から加える。これらのアッセイは、典型的に免疫グロブリンの機能に基づいており、すなわち、抗体がhuTNFR1に結合し、かつ、例えば競合的結合アッセイにおけるTNFαの前記細胞への結合の遮断、TNF/受容体結合の阻害、TNF(具体的にはIL6またはIL8などの炎症性インターロイキン)の存在または非存在下でのサイトカイン発現の減少およびアポトーシスなどのいくつかの生化学的事象を媒介する能力に基づいている。
本発明に係る好ましい医薬組成物は、治療的有効量の上に定義したhuTNFR1抗体と、任意に、薬学的に許容される担体、薬学的に許容される塩、補助剤、安定化剤、希釈剤および溶媒またそれらの任意の組み合わせからなる群から選択される1種以上のさらなる成分とを含む。
具体的には、本発明の医薬組成物は、自己免疫疾患、関節リウマチ、乾癬、乾癬性関節炎、若年性関節炎、強直性脊椎炎、クローン病(Morbus Crohn)、多発性硬化症、鬱血性心不全、代謝性疾患、サイトカイン放出症候群、敗血症性ショック、脳卒中、アルツハイマー病およびパーキンソン病などの急性および慢性神経変性疾患を治療するのに適している。さらなる適当な適応症としては、潰瘍性大腸炎および他の慢性炎症性および/または自己免疫疾患、急性劇症ウイルス性もしくは細菌性感染症、代謝性疾患、急性神経変性疾患、慢性神経変性疾患、原因となる病的メディエータとしてTNF/TNFR1を有する遺伝性疾患(好ましくは周期症候群およびケルビム症から選択される)および癌が挙げられる。
西洋ワサビペルオキシダーゼ(HRP)に結合した抗マウスIgG(Fc特異的)抗体、HRPに結合した抗ヒトIgG(全分子、Fc特異的、Fab特異的)抗体をそれぞれ、Sigma社(タウフキルヘン、ドイツ)から購入した。PE標識した抗マウス(全分子)および抗ヒトIgG(γ鎖特異的)抗体をそれぞれ、Sigma社(タウフキルヘン、ドイツ)から購入した。TNFR1−Fas(MEF−TNFR1−Fas)およびTNFR2−Fas(MEF−TNFR2−Fas)が形質移入されたマウス胚性線維芽細胞(MEF)をそれぞれ、RPMI1640培地(5%FCS、2mMのL−グルタミン、2μg/mlのピューロマイシン)で培養した。ヒト横紋筋肉腫細胞株Kym−1を、RPMI1640培地(10%FCS、2mMのL−グルタミン)で培養し、HT1080wt細胞およびHeLa細胞を、RPMI1640培地(5%FCS、2mMのL−グルタミン)で培養した。
CHO細胞での産生のために最適化されたIgκシグナル配列およびコドンを含む、ATROSABの軽鎖および重鎖をコードするDNAを、国際公開第2008/113515A2号の配列情報を用いて合成により産生した(Geneart社、レーゲンスブルク、ドイツ)。軽鎖(LC)DNAを、BamHI/NotIフラグメントとしてシャトルベクターpCV072(Celonic社、ユーリッヒ、ドイツ)にクローン化し、重鎖(HC)DNAを、KasI/NheIフラグメントとしてpFUSE(InvivoGen社、トゥールーズ、フランス)にクローン化した。pFUSEによってコードされるFc領域を変化させて、正しい好ましいアロタイプG1m1,17(E356D、M358L、G431A)を再構成した。pFUSE−HCをSmiI(SwaI)で消化し、全HC発現カセットを含む得られた平滑末端フラグメントを、PsiIで消化したpCV072−LCにクローン化した。このバイシストロン性発現カセットでは、軽鎖はPhEF1−HTLVプロモータの制御下にあり、重鎖遺伝子はPCMVの増強プロモータによって制御される。
UniProtKB(Swiss−Prot)エントリP19438(ヒト(学名:Homo sapiens)TNFR1)の配列情報を用い、個々のドメイン間に適当な制限部位を導入して、pSecTagL1−Fc(pSecTag−FcHisから修飾、(Muller et al. J. Immunol. Methods (2008) 339(1): 90-8))にクローン化して、ヒトTNFR1(aa29〜211)、アカゲザルTNFR1(aa27〜209)およびマウスTNFR1(aa30〜212)の細胞外領域をコードするDNAを合成により産生した(Geneart社、レーゲンスブルク、ドイツ)。ヒトTNFR1−FcとマウスTNFR1−Fcとの間の異なる領域を交換して、キメラのヒト/マウスTNFR1−Fc融合タンパク質を産生した。リポフェクタミン(Invitrogen社、カールスルーエ、ドイツ)を用いて、HEK293細胞にプラスミドDNAを形質移入し、Muller et al. J. Biol. Chem (2007) 282(17):12650-60に記載されているように、ゼオシンの存在下で安定に形質移入されたクローンを選択した。細胞を、90%の集密状態になるまで、RPMI(5%FCS、2mMのL−グルタミン)で培養した。タンパク質の産生のために、培地をOpti-MEM I(Invitrogen社、カールスルーエ、ドイツ)で交換し、上清を3〜4日毎に回収した。タンパク質Aクロマトグラフィにより、タンパク質を細胞培養上清から精製した。簡潔に言うと、上清を、1/10体積の1MのTrisHCl(pH8.0)を添加してpH8に調整し、タンパク質AセファロースCL−4Bカラム(Sigma社、タウフキルヘン、ドイツ)に充填した。結合されたタンパク質を、100mMのグリシン(pH3.0)で溶離し、1/10体積の1MのTrisHCl(pH8.0)を添加して中和し、画分を含むタンパク質をPBSに対して透析した。タンパク質濃度を測光法で測定し、SDS−PAGEと、HRPに結合した抗IgG(Fc特異的)抗体(Sigma社、タウフキルヘン、ドイツ)を用いる免疫ブロット法で、純度を分析した。
BioSuite(商標)250高解像度SEC(5μm)(Waters社、エシュボルン、ドイツ)を用いるHPLCで、サイズ排除クロマトグラフィ(SEC)を行った。以下の標準的なタンパク質:アポフェリチン(443kDa)、β−アミラーゼ(200kDa)、ウシ血清アルブミン(67kDa)、炭酸脱水酵素(29kDa)、アプロチニン(6.5kDa)を使用した。
抗体の親和性を、水晶振動子マイクロバランス法(QCM、Attana A-100 C-Fastシステム、ストックホルム、スエーデン)で測定した。0.005%Tween20のPBS溶液中で、25〜35μl/分の流速で結合実験を行い、温度を20℃に制御した。製造業者の手順書に従って、50μg/mlの濃度でアミン結合によりTNFR1−Fc融合タンパク質をAttanaカルボキシルセンサチップに化学的に固定すると、約200Hzのシグナルの増加(周波数の変化)が生じた。62.5〜3.9nMの濃度で抗体を分析した(濃度ごとに4回の測定)。10mMのグリシン−HCl(pH3.0)でチップを再生した。Attester評価での基準として使用するために、各試料注入前に緩衝液注入を行った。データをAttester3.0(バージョン3.1.1.8、Attana社、ストックホルム、スエーデン)で収集し、ClampXP(MyszkaおよびMorton、1998)で分析した。物質輸送モデル(Myszka、1997)をデータに当てはめた。
組換え型ヒトTNFR1−Fc融合タンパク質を、96ウェルプレート(50ng/ウェル、PBS溶液)に4℃で一晩固定した。2%(w/v)の粉ミルク/PBSで2時間ブロッキングした後、組換え型抗体フラグメントを二つ組で滴定し、RTで1時間インキュベートした。TMB基質(1mg/mlのTMB、酢酸ナトリウム緩衝液(pH6.0)、0.006%のH2O2)を用いて、HRPに結合した抗ヒトIgG(Fab特異的)抗体およびHRPに結合した抗マウスIgG(Fc特異的)で検出を行った。50μlの1MのH2SO4で反応を停止させた。ELISAリーダーで、吸光度を450nmで測定した。
フローサイトメトリにより、TNFR1−FasまたはTNFR2−Fasが形質移入されたMEF細胞への結合を分析した。細胞(2×105)を連続希釈した抗体と共に4℃で4時間インキュベートした。次いで、細胞をPBSで洗浄し、結合された抗体を、PEで標識したヤギの抗マウスもしくは抗ヒト抗体で検出した。細胞をフローサイトメトリ(Cytomics FC 500、Beckman−Coulter社、クレーフェルト、ドイツ)で分析した。データを、WinMDIプログラム(バージョン2.9)で評価し、GraphPrismソフトウェア(ラホヤ、米国)を用いて、3種類の独立した結合曲線を当てはめた。
Kym−1細胞(1.5×104細胞/100μl)を、96ウェルプレートで一晩培養した。培地で、抗体(図に示されている濃度)と共に三つ組で1時間プレインキュベートした後に、一定量のヒト可溶性TNFα(培地に1.25ng/ml)を加えた。7時間後、細胞をクリスタルバイオレット(20%メタノール、0.5%クリスタルバイオレット)で15分間染色した。ウェルをH2Oで洗浄し、空気乾燥した。染料をメタノールで15分間溶解し、550nmで光学濃度を測定した(Tecan infinite M200、クライルスハイム、ドイツ)。
HT1080細胞(2.0×105細胞/100μl)を、96ウェルプレートで一晩培養した。翌日、培地を交換して、構成的に産生されたIL−8を除去し、細胞を連続希釈したヒト可溶性TNFと共に二つ組でさらに18時間インキュベートした。製造業者の手順書に従って、IL−8サンドイッチELISA(ImmunoTools社、フリーゾイテ、ドイツ)により、IL−8の産生および培養上清中への分泌の誘導を測定した。さらに、細胞を、TNFα(一定の1ng/ml)の存在下で連続希釈した抗体と共にインキュベートし、18時間インキュベートした後に、IL−8の分泌について分析した。同じ方法で、製造業者の手順書に従い、IL−6サンドイッチELISA(ImmunoTools社、フリーゾイテ、ドイツ)を用いて、HeLa細胞からのIL−6のTNF媒介性分泌に対する抗体の阻害効果を分析した。TNFの非存在下で作動活性を測定した。
標準的なC1q結合アッセイでも、ATROSABの作動活性の欠如を実証した。C1qは、補体活性化の古典経路のC1複合体の第1の亜成分であり、細胞溶解にも関与している。従って、化合物がC1qに結合することにより、補体活性化と、その後に細胞溶解が生じる。行った実験では、免疫グロブリンであるハーセプチン(トラスツズマブ、Roche社)を陽性対照として使用し、免疫グロブリンG2aを陰性対照として使用した。ハーセプチンは、細胞溶解を誘導することが知られており、かつ転移性乳癌および胃癌の治療に使用されている。図11に示すように、ATROSABは、C1qに有意に結合せず、これは、本調査で比較物として使用した非選択的TNF受容体拮抗薬であるレミケード(インフリキシマブ、Centocor社)とは明らかに対照的である。これらの結果により、ATROSABは補体依存性細胞傷害性を誘導しないことが分かる。
IZI-06.1 IgG(ATROSAB)の産生および結合活性
エフェクタ機能が失われた重鎖(InvivoGen社(カリフォルニア州サンディエゴ)製のIgG1e3工学処理済Fc;E233P/L234V/L235A/ΔG236およびA327G/A330S/P331S置換を含むヒトFc(配列番号9)の配列を有するhIgG1e3−Fc1配列)を用いて、ヒト化抗ヒトTNFR1 Fab IZI-06.1(国際公開第2008/113515号)をヒトIgG1に転換した。上記配列情報を、図9(配列番号10〜18)に示す。
固定化TNFR1−Fcを用いた水晶振動子マイクロバランス法により、ATROSABのTNFR1に対する親和性を測定した。H398のヒトTNFR1−FcおよびアカゲザルTNFR1−Fcに対する親和性と同様に、ATROSABは、nM以下の親和性でヒトおよびアカゲザルTNFR1−Fcに結合した(図4、表1)。より早いKoff速度により、一価のscFv IZI-06.1では、約10倍で減少した親和性が測定され、これは、ATROSABおよびH398の二量体TNFR1−Fc融合タンパク質への結合が結合活性効果による影響を受けることを示している。
ATROSABは、Kym−1細胞のTNF誘導性アポトーシスを用量依存的に阻害した(図5)。このアッセイでは、90%の細胞傷害性を生じさせるTNF濃度を使用した。約最大半値の細胞傷害性、すなわち55%の生存細胞が、ATROSABでは60nM、H398では8nMでそれぞれ観察された。次いで、HeLa細胞からのIL−6とHT1080細胞からのIL−8のTNF誘導性分泌に対するATROSABの効果をそれぞれ調べた。TNFは、HeLa細胞からのIL−6の強い分泌を用量依存的に誘導し、4nMのTNF(200ng/ml)と共に18時間インキュベートした後に約700pg/mlのIL−6に到達した。同様に、TNFは、HT1080細胞からのIL−8の分泌を誘導し、4nMのTNFと共に18時間インキュベートした後に約7000pg/mlに到達した(図6c、d)。ATROSABおよびH398は、20pMのTNF(1ng/ml)によって用量依存的に誘導されるHeLa細胞からのIL−6およびHT1080細胞からのIL−8の放出を阻害した(図6a、b)。これらのアッセイでは、IL−6放出の阻害(図6a)およびIL−8放出の阻害(図6b)のためのIC50値はそれぞれ、ATROSABでは60nM、H398では6nMであった。HeLa細胞またはHT1080細胞を(TNFの非存在下で)ATROSABおよびH398のそれぞれと共にインキュベートすると、非常に狭い用量範囲でサイトカインの放出が僅かに誘導された。約10nMの濃度でのみ、比較され得るTNF濃度(4nM)で3〜4.5%の応答に相当するIL−6レベルの僅かな上昇が観察された(未処理の細胞の15pg/mlに対して40〜60pg/ml、すなわち25〜45pg/mlの誘導)。IL−8では、そのレベルが、未処理の細胞の80pg/mlから、抗体とのインキュベート後に、同等のTNF応答の約2%に相当する約200pg/mlまで上昇した。陰性対照として含めたヒトIgGは、サイトカインの放出に対して全く効果を有していなかった。
CD1マウスへの単回用量の静脈内注射後に、ATROSABおよびH398の半減期を測定した。7日間の期間にわたって抗体の濃度をELISAで測定し、すなわち機能性抗体分子を検出した。どちらの抗体も、ATROSABでは10.5±2.8d(n=2)、H398では8.1±1.5d(n=3)の消失半減期で、血液からの類似した消失を示した(図7)。
H398およびATROSABはマウスTNFR1に結合しないため、エピトープマッピングのためにドメイン交換戦略を適用した(図8)。これらのキメラTNFR1−Fc分子への結合をELISAで分析した。全ての構築物は、ヒトTNFに結合することができたが、構築物3および4は、キメラ分子がリガンド結合活性を保持していることが実証されているにも関わらず、他の構築物と比べて僅かな結合の減少を示した。ヒトのCRD1および2が対応するマウスのドメイン(構築物3)またはCRD1のみが対応するマウスのドメイン(構築物4)によって置換されたキメラの分子では、ATROSABおよびH398への結合は全く認められなかった。さらに、ヒトTNFR1のCRD1のA1ドメインのみが対応するマウスの配列(構築物5)で交換されている場合に、結合は全く認められなかった。ヒトのCRD1(構築物7)またはヒトのCRD1のA1ドメイン(構築物6)を含むマウスTNFR1についても、結合は大きく減少し、これは、完全な結合のためにさらなる領域が必要であることを示していた。CRD2のサブドメインA1を含めるようにヒト部分を拡大すると、H398およびATROSABがそれに対して強い結合を示すキメラTNFR1が得られた(構築物8)。従って、TNFR1のN末端領域のエピトープ残基は、残基1〜70を含む。この領域では、ヒトTNFR1とマウスTNFR1との間では15個の残基が異なっているが、ヒトTNFR1とアカゲザルTNFR1との間では1つの残基のみが異なっている(図8b)。この残基(Ile21)は、アカゲザルおよびマウスTNFR1ではバリンによって置換されている。ヒトTNFR1とマウスTNFR1との間で異なる残基のいくつかは、Pro23、Gln24、Tyr30、Asn31、Ser57、Ser59、His66およびHis69を含む受容体とTNFとの相互作用部位に露出されている。エピトープをさらに狭めるために、CRD1のサブドメインA1に位置しているP23およびGln24をキメラTNFR1 h1-2A1/m2B2-4内の対応するマウス残基で交換した(構築物10、図8a,b)。これらの突然変異により、適用したアッセイ条件では、ATROSABおよびH398の結合は完全に失われた。
ここで、ヒト化TNFR1特異的拮抗性モノクローナル抗体のIgG1誘導体(ATROSAB)の産生について述べる。その長い半減期、安定的な産生および二価による結合の上昇を理由に、IgG型を選択した。ATROSABは、ADCCおよびCDCの活性化が不十分なFc領域を有する。

- Fc媒介性エフェクタ機能が欠損している修飾されたFc領域を有する、IgG1型の抗huTNFR1抗体。
- 該Fc領域が、該エフェクタ機能を下方制御するための突然変異を含む、請求項1に記載の抗体。
- 該Fc領域が、該エフェクタ機能を下方制御するために糖鎖工学処理されている、請求項1または2に記載の抗体。
- 該Fc領域が、E233P、L234V、L235A、ΔG236、A327G、A330SおよびP331Sからなる群から選択される少なくとも1つの突然変異を有する重鎖を含む、請求項1〜3のいずれかに記載の抗体。
- huTNFR1の膜遠位CRD1と、CRD2のサブドメインA1とを含むエピトープに特異的に結合する、請求項1〜4のいずれかに記載の抗体。
- huTNFR1のN末端領域のアミノ酸1〜70によって表わされるエピトープに特異的に結合する、請求項1〜5のいずれかに記載の抗体。
- 該H398抗体によって認識される該エピトープに特異的に結合する、請求項1〜6のいずれかに記載の抗体。
- 少なくとも2つの結合部位によってhuTNFR1に特異的に結合する、請求項1〜7のいずれかに記載の抗体。
