Description

本発明の核酸は、SEQ ID NO: 1〜16に示される少なくとも一つ、その特定の断片、変異体、又は共通配列の少なくとも70〜100%を保持する連続アミノ酸をコードするヌクレオチド配列である。本発明に記載される方法または当該分野で公知の方法によって、少なくとも一つの抗TNF抗体をコードする本発明の核酸分子を得ることができる。
ヒト化抗体の重鎖可変領域配列のVH遺伝子断片を含むプラスミド(pHu-VH)をテンプレートとして、5'プライマーFVHX(5’-CGCGCAAG-CTTCCTCGAG-3’ SEQ ID NO: 17) と3' プライマーRVCG (5’-CGATGGGCCCTTGGTGGA-3’ SEQ ID NO: 18)を用いて、ヒト化抗体の重鎖可変領域(VH)とヒトIgG1重鎖定常領域(Cγ1)の5'末端の7個のアミノ酸との遺伝子を含む5'断片を得る。一方、ヒト白血球から調製したRNAを用いて、適当な5'プライマーHuCGF (5’-ACCAAGGGCCCATCGGTCTTC-3’; SEQ ID NO: 19)と3'プライマーHUCGE (5’-CGGAATTCTCATTTACCCGGAGACAGGGA 3’, SEQ ID NO: 20)で逆転写及びPCRによって、ヒトIgG1重鎖定常領域(Cγ1)をコードする遺伝子を得る。最後に、5'プライマー(FVHX, SEQ ID NO: 17)と3'プライマー(HUCGE, SEQ ID NO: 20)を用いて、ヒト化抗体の重鎖可変領域(VH)を含む断片と前記ヒトCγ1遺伝子とをPCRで連結させることにより、重鎖をコードする配列を含む、長さが約1400bpの遺伝子断片を得る。該遺伝子断片を、エンドヌクレアーゼHind IIIとEcoR1で処理後、ベクター、例えばPUC19に挿入する(Yanisch-Perron, C., Vieira, J. and Messing, J. 本発明は、遺伝子技術で作製した、組み換えベクターを含んだ宿主細胞、及び当分野の公知の組み換え技術で生成した少なくとも一つの抗TNF抗体にも関する。
本発明の抗腫瘍壊死因子ヒト化モノクローナル抗体は、組換えヒト腫瘍壊死因子(rhTNFα、ターゲット分子)のみに対し特異的親和性を持つが、ほかのタンパク質分子に結合しない。ターゲット分子に対する親和性の競合アッセイにおいて、本発明の抗腫瘍壊死因子ヒト化モノクローナル抗体はヒュミラ(アダリムマブモノクローナル抗体)に近い親和性を有することが測定された。
抗hTNFα抗体はhTNFα関連疾患の治療・予防に用いられる。hTNFα関連疾患として、例えば、膿血症、自己免疫病、悪性腫瘍、肺機能障害、移植による拒絶反応、細菌性髄膜炎、脳マラリア、エイズ及びエイズ関連症候群(ARC)、移植後のサイトメガロウイルス感染症が挙げられる。以下、本発明の抗体、及びhTNFα関連疾患の治療への抗体部分の用途を更に検討する。
腫瘍壊死因子は、膿血症病理学上の作用が確定され、その生物学的効果が低血圧、心筋阻害、血管漏出症候群、臓器壊死などを含む(例えば、米国特許第5,231,024号を参照)。従って、本発明のヒト化抗体および抗体部分は、膿血症ショック、内毒素ショック、グラム陰性膿血症および毒性ショック症候群を含む、任意的な臨床バックグランドの膿血症を治療するのに用いることができる。
腫瘍壊死因子は、様々な自己免疫病の病態生理学上で役割を果たすことが発見された。例えば、TNFαは、組織の炎症を活性化させ、リウマチ様関節炎における関節破損を引き起こすことに関与する(例えば、米国特許第5,231,024号;Moeller, A.らの (1990) Cytokine 2:162-169を参照)。また、TNFαは、糖尿病において膵島細胞死を促進し、希突起神経膠細胞に対する細胞毒性を仲介し、炎症斑を誘起することにも関与する。
腫瘍壊死因子は悪性腫瘍において悪液質の誘導、腫瘍成長の刺激、転移能の増強および細胞毒性の仲介に関与することが発見された。従って、本発明の抗体および抗体部分は、悪性腫瘍を治療し、腫瘍成長または転移を阻害し、および/または悪性腫瘍続発の悪液質を緩和するのに用いることができる。該抗体または抗体部分は、全身的にまたは局所的に腫瘍部位に投与することができる。
腫瘍壊死因子は白血球−内皮細胞活性化の刺激、肺細胞への細胞毒の指向、および血管漏出症候群の誘導を含む、成人呼吸困難症候群(ARDS)の病態生理学に関与することが知られている。従って、本発明の抗体および抗体部分は、成人呼吸困難症候群、慢性肺炎、肺サルコイドーシス、肺線維症およびケイ肺症を含む肺機能障害を治療するのに用いることができる。
本発明のヒト化抗体および抗体部分は、限局性回腸炎および潰瘍性大腸炎との二つの症候群を含む自発性炎症性腸疾患などの腸機能障害を治療するのに用いることができる。
腫瘍壊死因子は、同種移植拒絶や移植片対宿主病(GVHD)の中枢的メジエーターであり、T細胞受容体のCD3複合体へ導かれたラット抗体OKT3を腎移植体の拒絶を阻害するのに用いたときに観察された副作用を仲介することにも関与することが発見された(例えば、Suthanthiran, M., and Strom, T.B. (1994) New Engl, J.Med.331 :365-375を参照)。従って、本発明の抗体および抗体部分は、同種移植および異種移植の拒絶を含む移植拒絶およびGVHDを阻害するのに用いることができる。
本発明の抗体および抗体部分は、細菌性髄膜炎、脳マラリア、エイズやエイズ関連症候群(ARC)、および移植後のサイトメガロウイルス感染を含む伝染病の治療に用いることができる。また、本発明の抗体および抗体部分は、感染(例えばインフルエンザ)による発熱や筋肉痛、および感染続発の悪液質(例えばエイズまたはARC続発の悪液質)を含む、伝染病に付随する徴候を緩和するのに用いることもできる。
本発明の抗体は、任意の公知の分析方法、例えば、競合的結合分析、直接または間接サンドイッチ分析および免疫沈降分析などに用いることができる。Zola, 「モノクローナル抗体:技術マニュアル」 (Monoclone Antibodies; A Manual of Techniques), 第147〜158頁 (CRC Press, Inc., 1987)。
本発明の抗体は、アフィニティー精製試薬として使用することができる。この方法では、抗体は当該分野で公知の方法によってセファデックス樹脂や濾紙などの担体に固定される。精製しようとするhTNFα含有サンプルを固定された抗体に接触させ、適当な溶媒を用いてキャリアを洗浄する。固定された抗体に結合されたhTNFαを除いてサンプル中のほかの物質は実質的にすべて前記溶媒により除去される。
本発明の抗体および抗体部分は、被験者への投与に適した医薬組成物に添加することもできる。該医薬組成物は、本発明の抗体及び薬学的に許容される賦形剤を含むものである。薬学的に許容される賦形剤としては、任意の生理学的に適用できる溶媒、分散媒体、抗菌剤、抗真菌剤、等張化剤、コーティング、吸収遅延剤などが挙げられる。本発明の医薬組成物は、液体、半固体および固体製剤などの様々な形態としても良い。
1)注射剤
本発明の別の態様では、前記疾患の治療に使用される材料を含む製品を提供する。該製品は容器とラベルを含む。適切な容器としては、普通の瓶、薬瓶、注射器、実験管などが挙げられる。容器はガラスやプラスチックなど様々な材料で作られる。容器に疾患の治療に有効な組成物が含まれ、無菌の入り口がある(例えば、容器は、皮下注射針で透過できる栓のある静脈内輸液バッグや薬瓶)。該組成物中の活性成分は抗hTNFα抗体である。容器上又は容器に付着するレベルは、組成物が治療できる特定病症を説明するものである。また、製品はリン酸塩バッファー、リンゲル(Ringer)溶液、グルコース溶液のような薬学的に許容されるバッファーを含む別の容器を有しても良い。さらに、製品は、商業上のニーズやユーザーのニーズに応じて、ほかの材料、例えば、ほかのバッファー、希釈剤、フィルター、針、シリンジ、および、取扱説明が添付された包装マニュアルを含んでも良い。
本発明の抗腫瘍壊死因子ヒト化モノクローナル抗体は、徐放性製剤の製造に使用することができる。適宜な徐放性製剤としては、例えば、前記抗体を含む固体疎水性ポリマーの半透過性マトリックスなどが挙げられる。前記マトリックスは形のあるものであり、例えば、フィルムまたはマイクロカプセルが挙げられる。適宜な徐放性マトリックスとしては、ポリエステル、ヒドロゲル(例えば、ポリ(メタクリル酸2-ヒドロキシエチル)、またはポリビニルアルコール)、ポリラクチド(米国特許第3,773,919号)、L-グルタミン酸とL-グルタミン酸エチルの共重体、非分解性のエチレンビニルアセテート、Lupron DepotTM(乳酸-グリコール酸共重体とleuprolide acetateから構成される注射用ミクロスフェア)のような分解性の乳酸-グリコール酸共重体およびポリ-D-(-)-3-ヒドロキシ酪酸が挙げられる。エチレンビニルアセテートおよび乳酸-グリコール酸などのポリマーは分子の放出を100日以上に続けることができるが、一部のヒドロゲルは短期間内にタンパク質を放出する。カプセル化された抗体は、長期間に体内に保持される場合、37℃で水との接触で変性または凝集するため、生物学的活性が低下し、免疫原性が変化する可能性もある。合理的な安定化方法は、メカニズムに合わせて設計することができる。例えば、安定化は、スルフィド−ジスルフィドの交換反応により分子間S−S結合を形成し凝縮になる場合、チオール残基の修飾、酸性溶液の凍結乾燥、水分含有量の制御、適当な添加剤の使用、又は特別なポリマーマトリックス組成物の設計によって達成することができる。
本発明の抗体は、便利を図るために、所定量の試薬と診断分析用のマニュアルとを組み合わせたキットの形で提供することもできる。抗体は酵素で標識される場合、キットには、酵素に必要とする基質や補因子(例えば、検出可能な発色団および蛍光団を提供する基質前駆体)が含まれている。また、キットには、例えば、安定剤、バッファー(例えば、ブロッキングバッファーまたは分解用バッファー)などほかの添加剤が含まれても良い。提供した試薬濃度で最高の分析感度を達成させるために、試薬の相対量の差異が大きい。具体的に、試薬は、通常に凍結乾燥粉末のような形の乾燥粉末とすることができ、賦形剤を含んでもよく、溶かされた際に適当な濃度を有する試薬溶液を形成する。

7〜11週齢の雌Balb/cマウス数匹に組換えhTNF(rhTNFα:Pepro Tech Inc.より購入)でIPまたはID免疫した。これらの組換えヒトTNFαは、最終容量が100〜400μLとなるように等容量のTITERMAXまたは完全フロイントアジュバントで乳化された。1〜7、5〜12、10〜18、17〜25及び/又は21〜34日後に、等容量のTITERMAXまたはフロイント不完全アジュバントで乳化されたTNFを用いて、各マウスにIP(1〜400μg)とSC(1400μg×2)免疫した。12〜25日後と25〜40日後に非抗凝の条件下で眼底穿刺によりマウスから採血した。次に室温で血液を1時間凝固させて血清を収集し、公知の方法に従ってTNFαELAで血清の力価を測定した。繰返して注射しても力価が増加しない時に融合を実施した。その時点で、生理食塩水100μLで希釈されたTNFα1~400μgをマウスに最終のブースター注射してもよい。3日後に頸部脱臼によりマウスを安楽死させ、脾臓を無菌の条件下で摘出し、ペニシリン100U/mL、ストレプトマイシン100μg/mL及びアンホテリシンB(PSA)0.25μg/mLを含んだ冷リン酸バッファー(PBS)10mLに浸漬した。脾臓をPSA-PBSで無菌の条件下で灌流することにより脾細胞を回収した。脾細胞をトリパンブルー染色排除法によりカウントし、25mM Hepesを含んだRPMI 1640培地に再懸濁した。
プレートを2μg/mLのTNFα PBS溶液で一晩コーティングした。0.02%(v/v)Tween 20を含んだ0.15M食塩水で洗浄後、1%(w/v)BSAを含んだ PBS溶液で 200μL/ウェルの量でウェルを室温にて1時間ブロックした。このプレートをすぐに使用してもよく、−20℃で凍結し将来の使用に備えてもよい。TNFαをコーティングしたプレートにマウス血清希釈液50μL/ウェルを室温で1時間インキュベートした。プレートを洗浄した後、1%BSA-PBSで1:30000に希釈されたHRP標識IgGFc 50μL/ウェルを室温にて1時間プローブした。プレートを再び洗浄し、クエン酸塩−リン酸塩基質溶液(0.1Mクエン酸と0.2Mリン酸ナトリウム、0.01% H2O2及び1mg/mL OPD)100μL/ウェルを室温で加え、15分間反応させた。その後、停止溶液(4N硫酸)25μL/ウェルを加え、自動プレート分光光度計により490nmのODを読み取った。
1:1~10:1の比率で、血清アッセイにより検出された高レベルの抗hTNFα抗体血清を有するマウスの生存脾細胞をマウス骨髄腫細胞と融合させた。非限定的な例として、下記の方法が挙げられる。即ち、脾細胞と骨髄腫細胞を一緒にペレット化した後、ペレットを50%(w/v)PEG/PBS溶液(PEG分子量1,450,Sigma)1mLに37℃にて30秒間以上かけて再懸濁した。次に25mM Hepes(37℃)を含んだRPMI 1640培地10.5mlを1分間以上かけてゆっくりと加えることにより融合を停止させた。融合細胞を500〜1500rpmで5分間遠心した。続いて細胞をHAT培地(25mM Hepes、10% Fetal Clone I血清(Hyclone)、1mMピルビン酸ナトリウム、4mM L-グルタミン、10μg/mLゲンタマイシン、2.5% Origen培養サプリメント(Fisher)、10%653馴化RPMI 1640/Hepes培地、50μM 2-メルカプトエタノール、100μMヒポキサンチン及び16μMチミジン、を含んだRPMI 1640培地)に再懸濁した後、200μL/ウェルで96ウェル平底組織培養プレート15枚にプレーティングした。次にプレートを5%CO2と95%空気を含有する加湿の37℃インキュベーターにいれ、7〜10日インキュベートした。
抗hTNFα抗体の定性は2種類の測定法がある。一つは抗体がヒュミラとhTNFαに対する競合的結合を測定するものであり、もう一つは抗体がL929細胞毒性測定においてhTNFαを中和する能力を測定するものである。以下は、該2種類の方法及び実験結果をそれぞれ説明する。
西洋ワサビペルオキシダーゼ(HRP)で標識された抗hTNFαヒト抗体であるヒュミラを試薬として使用した。rhTNF(50μL、0.05g/mL)でELISAプレートを室温で一晩コーティングした。コーティング溶液を捨て、リン酸塩バッファー(PBS)に溶解した1%脱脂ミルクで各ウェルを0.5時間ブロックし、0.05%Tween 20を含んだPBSでウェルを洗浄した。次に増殖培地50μlとHRPで標識されたヒュミラ50μlとの混合液を、各ウェルに加えた。未標識のヒュミラと、抗体を含まない培地を、それぞれポジティブとネガティブコントロールとした。この方法により、rhTNFαへのHRP標識ヒュミラの結合を高レベルで阻害するマウスモノクローナル抗体を選別することができる。rhTNFαへのHRP標識ヒュミラの結合を阻害するウェルを増幅及びサブクローニングし、阻害効果を示している複数のマウスモノクローナル抗体を続いて分析し、最終的にハイブリドーマ細胞を選別した。該ハイブリドーマ細胞を増幅培養し、上清を取って精製し、マウスモノクローナル抗体TM2-11-12とTM2-6-3を得た。精製したマウスモノクローナル抗体TM2-11-12とTM2-6-3を用いて、競合的結合測定を行った。マウス抗体TM2-6-3は、高濃度1μg/mLの場合でも、ただ約50%のヒュミラをhTNFαに結合させなくなった。一方、もう一つのマウス抗体TM2-11-12は、標識のないヒュミラと同等の優れた競合能力を示し、濃度約0.05μg/mL(3×10-10Mに相当)の場合、約50%のヒュミラをhTNFαに結合させなくなった。
抗hTNFαマウス化抗体及びキメラ抗体でhTNFαを中和する生物活性は、両方ともL929細胞毒性アッセイによって測定することができ、具体的な方法は下記のように行った。合わせて7.5×103のL929細胞(ATCC)(105/mL、75μL)を96ウェル培養プレートの各ウェルに入れ、37℃、5%CO2のインキュベーター内に24時間放置した。L929細胞の増殖培地は5%ウシ胎児血清を含んだRPMI-1640(GIBCO)であった。別の96ウェル培養プレートを用いて、抗hTNFα抗体を含む溶液をRPMI増殖培地2部で1/2になるように連続的に希釈し、各サンプルウェルにおけるrhTNFαの最終濃度が5ng/mLとなるようにrhTNFαを添加し、混合液を有する培養プレートを37℃、5%CO2のインキュベーター内に2時間放置し、次に各行の抗体濃度が0.001から2μg/ mLとなる順で抗体とrhTNFαを含有する混合液を各L929細胞ウェルに入れた。該培養プレートを37℃、5%CO2のインキュベーター内に放置した。3日後、生存細胞数を定量した時、2.5mg/mLの3-(4,4-ジメチルチアゾール-2-イル)-2,5-ジフェニル-テトラゾリウム ブロミド(MTT、Sigma Biochemicalsより購入)のPBS溶液20μlを添加し37℃で4時間インキュベートし、更に10%ドデシル硫酸ナトリウム(SDS)の0.01 N HCl溶液100μlを加えて一晩放置した。そして各ウェルに対して吸光度を540/690nmで測定した。吸光度と抗体濃度との曲線を描いた。結合曲線より、IC50、即ちL929細胞に対するrhTNFα毒性の50%を中和した抗体濃度を得た。従って、IC50値は、hTNFα細胞毒性を阻害する抗体の能力を比較するために使用することができる。複数の抗hTNFαマウス化抗体(TM2-11-12、TM2-10-20、TM2-2-2)のIC50値及びヒュミラのIC50値は、全て0.01〜0.04μg/mLの範囲内にあり、即ち、全てはrhTNFαによって引き起こされたL929細胞毒性を中和する類似な能力を持っている。抗hTNFαマウス化抗体TM2-11-12は、キメラ抗体の作製のために選定した。
1)TM2-11-12マウス化抗体の重鎖可変領域のクローニング
マウス化抗体をヒト化させるには、まず、マウス化抗体TM2-11-12の重鎖と軽鎖の可変領域をコードする配列を含むDNA断片を取得しなければならない。RNA精製キット(Invitrogen社)を用いてTM2-11-12マウスハイブリドーマ細胞からRNAを単離することにより、cDNA(GeneRacerキット、Invitrogen社)を作製した。5'プライマー(5'-CGACTGGAGCACGAGGACACTGA-3', SEQ ID NO: 24)と、マウスIgG1の重鎖定常領域に対して相同でアンチセンスである3'プライマー(5'-TCCAGGGGCCAGTGGATAGAGAGA-3', SEQ ID NO: 25)を用いて、ポリメラーゼ連鎖反応(PCR)によってcDNAから重鎖可変領域のDNA断片を単離した。これらのDNA断片をTOPO TAベクター(Invitrogen社)にクローニングし、塩基配列を測定した。重鎖可変領域のアミノ酸配列はSEQ ID NO:34であり、相補性決定領域のアミノ酸残基は、CDR-H1 (SEQ ID NO: 3)、CDR-H2 (SEQ ID NO: 4) およびCDR-H3 (SEQ ID NO: 5)である。相補性決定領域の定義は、Kabat E.らの「Sequences of Proteins of Immunological Interest」,5th Edition, U.S. Department of Health and Human Services, NIH Publication No.91-3242を参考してください。
重鎖可変領域のクローニング法と同様に、SEQ ID NO: 29の5'プライマーと、マウス免疫グロブリンのκ軽鎖定常領域に対して相同でアンチセンスである3'プライマー(5'-CACTGGATGGTGGGAAGATGGATA-3'、SEQ ID NO:26)を用いて、PCR法によりcDNAから軽鎖可変領域のDNA断片を単離し、これらのDNA断片をTOPO TAベクターにクローニングし、塩基配列を測定した。その結果、2種類のクローンを確認した。クローンの約3/4は、ヌクレオチド配列の一部が読み取り可能なアミノ酸配列(その配列が示されない)に翻訳できないものであった。このクローンは、機能的抗体の軽鎖タンパク質をエンコードできない変異軽鎖メッセンジャーRNAである。クローンの約1/4は完全に読み取り可能なアミノ酸配列に翻訳できるヌクレオチド配列を示した。このクローンは、機能のある軽鎖メッセンジャーRNAに由来する。これらのDNA断片をTOPO TAベクター(Invitrogen社)にクローニングし、塩基配列を測定した。軽鎖可変領域のアミノ酸配列はSEQ ID NO:35であり、相補性決定領域のアミノ酸残基は、CDR-L1(SEQ ID NO:6)、CDR-L2(SEQ ID NO:7)およびCDR-L3(SEQ ID NO:8)である。このアミノ酸配列は軽鎖のヒト化設計に用いられた。
抗原結合活性を保持するために、ヒト化させる段階ですべての軽鎖及び重鎖の可変領域内のアミノ酸残基がTM2-11-12マウス化抗体の配列を保持するようにした。ヒト化設計は、ヒト化抗体の配列に従ってフレームワーク領域内のアミノ酸残基を変更し、様々な修飾のあるヒト化抗体の重鎖可変領域と軽鎖可変領域を設計し、抗体の結合親和性を増加したり、抗体の免疫原性を低減するように、コンピュータシミュレーション技術によって抗体の重鎖及び軽鎖の可変領域配列のオリゴヌクレオチド部位を定方向突然変異させるものである。
1)ヒト化抗体軽鎖遺伝子の構築
まず人口合成法によりヒト化抗体の軽鎖可変領域(F001VL)の遺伝子断片を調製した。具体的な方法として、先に軽鎖可変領域のアミノ酸配列に対応する遺伝コドンから逆翻訳することによってヌクレオチド配列を得て、さらに5'末端にKozak配列および軽鎖リーダー配列を導入し、その後人口合成法により軽鎖可変領域の遺伝子断片を調製した。該遺伝子断片を適当なベクターに導入し、pHu-VL1プラスミドを得た。次はpHu-VL1プラスミドをテンプレートとして、5'プライマーFVHX (SEQ ID NO: 17)と3'プライマーVKCKO(SEQ ID NO: 21)を用いて、ヒト化抗体の軽鎖可変領域(VL)及びヒトκ軽鎖定常領域(Cκ)の5'末端からの7個アミノ酸の遺伝子を含む5'断片を得た。一方、ヒト白血球から調製したRNAを用いて、適当な5'プライマーHuCKF (SEQ ID NO: 22)と3'プライマーHUCKB (SEQ ID NO: 23)で、逆転写及びPCRによって、ヒトκ軽鎖定常領域(Cκ)をコードする配列を含む遺伝子を取得した。最後に、5'プライマー(FVHX, SEQ ID NO: 17)と3'プライマー(HUCKB, SEQ ID NO: 23)を用いて、ヒト化抗体の軽鎖可変領域(VL1)を含有する断片と前記ヒトCκ遺伝子とをPCRで連結させることにより、軽鎖をコードする配列を含む、長さが約700bpの遺伝子断片を得た。ヒト化抗体の軽鎖タンパク質を発現するために、該遺伝子断片をエンドヌクレアーゼHind IIIとBam H1で処理後、PUC19の如くベクターに挿入した(Yanisch-Perron, C., Vieira, J. and Messing, J. (1985) Gene, 33, 103-119を参照)。DNAシークエンサーによって該遺伝子断片の配列が正しいと確認した。
まず人口合成法によりヒト化抗体の重鎖可変領域(E001VH)の遺伝子断片を調製した。具体的な方法として、先に重鎖可変領域のアミノ酸配列に対応する遺伝コドンから逆翻訳することによってヌクレオチド配列を得て、さらに5'末端にKozak配列および重鎖リーダー配列を導入し、その後人口合成法により重鎖可変領域の遺伝子断片を調製した。該遺伝子断片を適当なベクターに導入し、pHu-VL1プラスミドを得た。次はpHu-VL1プラスミドをテンプレートとして、5'プライマーFVHX (SEQ ID NO: 17)と3'プライマーRVCG(SEQ ID NO: 18)を用いて、ヒト化抗体の重鎖可変領域(VH1)及びヒトIgG1重鎖定常領域(Cγ1)の5'末端からの7個アミノ酸の遺伝子を含む5'断片を得た。一方、ヒト白血球から調製したRNAを用いて、適当な5'プライマーHuCGF(SEQ ID NO: 19)と3'プライマーHUCGE(SEQ ID NO: 20)で、逆転写反応及びPCRによって、ヒトIgG1重鎖定常領域(Cγ1)をコードする配列を含む遺伝子を取得した。最後に、5'プライマー(FVHX, SEQ ID NO: 17)と3'プライマー(HUCGE, SEQ ID NO: 20)を用いて、ヒト化抗体の重鎖可変領域(VH)を含有する断片と前記ヒトCγ1遺伝子とをPCRで連結させることにより、重鎖をコードする配列を含む、長さが約1400bpの遺伝子断片を得た。ヒト化抗体の重鎖タンパク質を発現するために、該遺伝子断片をエンドヌクレアーゼHind IIIとEcoR1で処理後、PUC19の如くベクターに挿入した(Yanisch-Perron, C., Vieira, J. and Messing, J. (1985) Gene, 33, 103-119を参照)。DNAシークエンサーによって該遺伝子断片の配列が正しいと確認した。
前記方法で得られた重鎖と軽鎖をコードするcDNAをベクターpcDNA3(Invitrogen USA, Carlsbad, CA, U.S.A.より購入)に挿入し、ヒト化発現ベクターpHu_anti-H1L1-TNFαを構築した。該発現ベクターに、哺乳動物細胞内において高いレベルの発現に必要なサイトメガロウイルス初期遺伝子プロモーター・エンハンサーが含まれる。また、ベクターに、細菌でアンピシリン耐性、哺乳動物細胞でG418耐性を付与する任意のメーカー遺伝子が含まれる。更に、ベクターにDHFR遺伝子が含まれ、これにより、適当な宿主細胞においてメトトレキサート(Methotrexate MTX, Sigma)を用いてキメラ抗体遺伝子とDHFR遺伝子を共同で増幅する可能である(例えば、Axel, R.らの米国特許第5,179,017号;Kaufman,R. と Sharp,P., J.Mol. Biol. 159:601-621,1982を参照)。
抗hTNFαヒト化抗体を発現させるために、前記構築した組換え発現ベクターを哺乳動物宿主細胞にトランスフェクショントした。高レベルの発現を安定させる点で、好ましい宿主細胞は、ジヒドロ葉酸レダクターゼ(DHFR)欠損チャイニーズハムスター卵巣(CHO)細胞(例えば、Chasin, L.らの米国特許第4,818,679号を参照)である。好ましいトランスフェクション法はエレクトロポレーション法であるが、リン酸カルシウム共沈法、脂質トランスフェクション法、プロトプラスト融合法などの方法を使用することもできる。エレクトロポレーション法では、250Vの電界と960μFdのコンデンサにしたGene Pulser (Bio-Rad Laboratories)を用いて、キュベットに、2×107個の細胞が懸濁したPBS 0.8mlおよびPvuI(Takara)で線形化された10μgの発現ベクターDNAを加えた。トランスフェクション2日後、0.2mg/mLのG418および200nMのメトトレキサート(methotrexateまたはMTX)を添加した。高レベルの発現を達成するために、トランスフェクションされたヒト化抗体遺伝子をMTX薬物によって阻害されるDHFR遺伝子で共同増幅させた。極限まで希釈されたサブクローニングのトランスフェクタントで各細胞株の分泌率を測定することにより、高レベルでヒト化抗体を発現する細胞株をスクリーニングした。
L929細胞をトリプシンで処理・遠心分離し、10%FCSを入れた1640培地に再懸濁・カウントした後、特定の濃度で96ウェルプレートの第1〜11列に加え、更に96ウェルプレートの第1〜10列に適切濃度のTNFαを加えた。続いて、ヒュミラ(Abbott Laboratories社より購入)、アミノ酸の修飾されないキメラ抗体AT(CE)-1、及び実施例5と6で調製されたヒト化抗体AT132、AT135、AT143、AT151、AT164を、第1位のウェル濃度0.4μg/mLで2倍の勾配希釈で、高い濃度から順次(第1〜9列)に、96ウェルプレートの第A、B、C、D、E、F、GとH行にそれぞれ添加した。第10列はTNFα対照であり、第11列は細胞対照であり、第12列は培地対照であった。添加完了後、37℃の二酸化炭素インキュベーターに放置しインキュベートした。インキュベート終了後、発色試薬を加え、インキュベートし、吸光度をマイクロプレートリーダーで検出した。結果は表2に示す。
調子の良いU937細胞をカウントし、3.75×104個/ウェルになるように10%FCSをいれた1640培地で細胞濃度を調整し、96ウェルプレートに75μl/ウェルを添加した。120ng/mLのTNFαを含む細胞培養液を用いて、第1位のウェル濃度600ng/mLで1.5倍の希釈勾配でAT132標準試料と実験試料を勾配希釈した後、25μl/ウェルで96ウェルプレートに添加し、37℃の二酸化炭素インキュベーターで40時間インキュベートした。インキュベート終了後、各ウェルにCCK8発色試薬10μlを添加し、3時間インキュベートし、490nm/630nm二波長でマイクロプレートリーダーにより検出し、4パラメーターでカーブフィッティングを作って、標準試料と実験試料のED50を統計し、比活性を算出した(計算式:100%×標準試料のED50/実験試料のED50)。
図1の結果よりわかるように、AT132濃度は極めて低い場合はTNFαが細胞毒性を誘導するが、AT132濃度の増加に従って、TNFαの作用が徐々に阻害され、AT132濃度は約80ng/mLの時点で、TNFαの細胞毒性が実質的に完全に阻害された。よって、阻害効果は使用量に依存することが明らかであり、複数回の実験結果によれば、30ng/mLのTNFαを中和するAT132の平均半数有効濃度は24.1ng/mLであった。
AT132の親和性はBiacore X100により測定し、Biacore X100 kinetics/affinity解析ソフトウェアによって分析を行い、間接的な捕獲法でAmine Coupling Kitによりヤギ抗ヒトIgG Fcポリクローナル抗体をCM5チップ表面にカップリングして捕獲分子とし、計算よりそれぞれに特定の濃度に希釈されたAT132及びコントロールとするヒュミラをリガンドとし、TNFαを分析物とした。分析物を5段階の濃度に希釈し、一つの濃度を一つのサイクルとし、まずHBS−EPバッファーで3サイクルを実行し、そして分析物の濃度が0にしたもので2サイクルを実行し、最後に繰り返した分析物濃度で1サイクルを実行した。全過程で11サイクルを実行し、各サイクルごとに一つの曲線を描い、Biacore X100 kinetics/affinity解析ソフトウェアによって抗体とヒュミラのkinetics/affinityデータを得た。
組換えヒトTNFα 5ng/ウェル、マウスTNFα 25ng/ウェル、サルTNFα 5ng/ウェルでコーティングし、室温で1時間ブロッキングし、洗浄してから、初期濃度250ng/mLで1.8倍で勾配希釈されたAT132を添加し、25℃で2時間インキュベートし、洗浄後、HRPで標識された抗ヒトFc抗体を添加し、室温で1時間インキュベートし、洗浄後、100μl/ウェルの基質溶液を添加し、37℃で暗所に30分間反応し、発色試薬を加えた順番で0.2M H2SO4を50μl/ウェル加えて反応を停止させた。反応終了後5分間内に、マイクロプレートリーダーにより450nm/630nmのOD値を測定し、4パラメーターのフィッティングによって半数有効濃度を得て、異なる属由来のTNFαとのAT132の結合活性を比較した。
サルTNFαとの結合活性の比較研究結果より、サルTNFαとの結合活性はヒトTNFαとの結合活性の約50%であることがわかった。
次のようにAT132、AT135の注射製剤を調製した。
1)20Lのバッファーの調製(20.180kgに相当。溶液の密度:1.009g/mL)
マンニトール240.0g、クエン酸一水和物26.1g、クエン酸ナトリウム6.1g、リン酸水素二ナトリウム二水和物30.6g、リン酸二水素ナトリウム二水和物17.2g、塩化ナトリウム123.3g、無水ポリオキシエチレン(20)ソルビタンオレイン酸エステル20.0g、及び水19715.7〜19716.1gを量った。
そして、前記予め量ったマンニトール、クエン酸一水和物、クエン酸ナトリウム、リン酸水素二ナトリウム二水和物、リン酸二水素ナトリウム、塩化ナトリウムおよび無水ポリオキシエチレン(20)ソルビタンオレイン酸エステルを約90%の注射用水に溶かしてバッファーを作った。
濾過されたバッファーを以下の方法で調製した解凍・合併の抗体濃縮液(前記医薬製剤の有効成分)に加えた。前記医薬製剤調製の直前に水浴中に前記抗体(濃縮液)を解凍した。タンパク質2.0kgに相当する、濃度が60mgタンパク質/mLタンパク質濃縮液、密度が1.0262g/mLである抗体濃縮液34.207gを使用した。タンパク質濃縮液中のタンパク質濃度が50〜80mg/mLに相当する、25.655〜37.316範囲のタンパク質濃縮液を使用することができる。撹拌しながら、前記バッファーを溶液の最終量に達するまで添加した。
その後、すべての成分を含む製剤を、2層の無菌0.22μmメンブレンフィルターで濾過することを除き、前記方法に準じて濾過した。製剤を消毒した後、バイアル瓶または予め充填したシリンジで使用できるように包装した。
実験試料:20mg/ボトルのAT132凍結乾燥粉末、バッファーコントロール:AT132バッファー(ヒスチジンとトレハロースを含む)、溶媒:注射用滅菌水。実験動物のグループおよび投与量:4-6週齡の昆明(KM)マウス60匹、18〜22g、オスメス各半数、SPF級。前記動物を、オスメス各半数で各グループが20匹になるようにランダムに3グループに分けた。一回投薬し、14日観察した。投与量および投与経路は表3に示す。
体重が20.0±2.0gのC57BL/6マウス51匹を6グループに分けた(表4に示す)。第2グループのマウスへの投薬量がAT132を基準として5.2μg/匹、第3グループのマウスへの投薬量がAT132を基準として26μg/匹、第4グループのマウスへの投薬量がAT132を基準として52μg/匹、第5グループのマウスへの投薬量がAT132を基準として26μg/匹となるように、第2〜5グループの各マウスの腹腔内にそれぞれAT132溶液0.25mLを注入した。第1グループの各マウスの腹腔内にpH5.63のクエン酸バッファー0.25mLを注射した。HuIgG1を基準として26μg/匹となるように、第6グループの各マウスの腹腔内にヒトIgG1(HuIgG1、ネガティブコントロール)0.25mLを注射した。30分間後、各グループのマウス(第5グループを除く)の腹腔内にrhTNFα(Primegene,ロット番号1030109021)とD-ガラクトサミンとの混合溶液0.25mlを注射し、第5グループのマウスの腹腔内にバッファー0.25mLを注射した。48時間以内に死亡したマウスの数を観察し、生存率を算出した。結果は表4に示す。
Wistar系雌ラット50匹を、各グループが10匹になるようにランダムに4グループに分け、それぞれ、コントロールグループ、炎症性コントロールグループ(モデル群)、1mg/kgのAT132投与グループ、及び5mg/kgのAT132投与グループとした。
コントロールグループを除いて、他のグループのラットはそれぞれ背中の皮内にII型コラーゲン免疫原を注射した。炎症性コントロールグループは0.1mol/Lの酢酸および完全または不完全フロイントアジュバント乳剤(Sigma社、ロット番号129K8701)を注射した。
1mg/kgのAT132投与グループ及び5mg/kgのAT132投与グループは、注射免疫の当日から腹腔内の注射投与を始めた。第1回目免疫の1週間後、同じ方法で強化のため再免疫し、合わせて28日間投与した。投与前後の異なる時点で、ラット足関節の腫脹値をYLS-7B足つま先ボリューム測定器により測定した。実験終了後、足関節を病理検査のために採取した。腫脹率と阻害率を算出し、グループ間の差別をt検定により比較した。計算式は下記に示す。
本実験では、Tg197トランスジェニックマウス(Cyagen Biosciencesより購入)を採用し、各グループが10匹、オスメス各半数となるように6グループに分けた。具体的に、第1グループ:1mg/kgのAT132、第2グループ:溶剤(クエン酸及び塩化ナトリウムを含むバッファー)、第3グループ:30mg/kgのAT132、第4グループ:10mg/kgのAT132、第5グループ:10mg/kgのヒュミラ、第6グループ:3mg/kgのAT132であった。また、Tg197マウス4匹をコントロールグループとした。
関節炎程度を評価するため、毎週マウスの関節形態変化に対し得点を評価した。関節炎の採点基準は下記の通りである。
0.5=発症した関節炎(関節と爪がわずかな腫脹を示し、外観が正常、体重を支えることができ、全体的な柔軟性/避難能力が正常、握力が最も大きい)
1.0=軽度の関節炎(関節が腫れや変形を示し、爪が赤い腫れを示し、外観が正常、体重を支えることができ、全体的な柔軟性/避難能力が正常、握力が最も大きい)
1.5=軽度から中程度の関節炎(関節と爪が腫れ、変形する+最後の手指が内側へ変形し、上体の重さを支える時間が短く、全体的な柔軟性が減少し、握力が減少する)
2.0=中程度の関節炎(関節、爪および手指が重度に腫れ、足関節が変形し、上体の重さを支えることができず倒れ、全体的な柔軟性が消え、握力が消え、はって進みや採食に影響がある)
2.5=中程度から重度の関節炎(関節、爪および手指が重度に腫れ、足関節が変形し、上体の重さを支えることができず倒れ、全体的な柔軟性が消え、握力が消え、はって進みや採食に影響がある+手指と爪が変形し、マウスの活動に支障がある)
3.0=重度の関節炎(関節がこわばり、曲げと重度な活動支障が見られ、マウスが瀕死の状態となる)。
Tg197マウスの関節炎点数は図3Aに示す。
疾患の状態を監視するために、本実験では、同腹のTg197マウス4匹(番号Con1〜Con4)を3週齢時に犠牲させ、治療開始時点の関節サンプルとした。受験マウスを10週齢時に犠牲させ、足首関節スライスを採取し、ヘマトキシリン/エオシンで染色後、盲検法により関節炎の顕微鏡組織病理学得点を評価し、下記のように0〜4を採点した。
1 = 滑膜増殖および多形核白血球浸潤
2 = パンヌスと線維組織の形成、感染巣軟骨の下骨びらん
3 = 軟骨破壊および骨びらん
4 = 拡大した軟骨破壊および骨びらん
Tg197マウスの組織病理学分析は図3Bおよび3Cに示す。
1)ヒト組織へのAT132モノクローナル抗体の交差反応
ヒト鼻ポリープのパラフィン切片および4つの正常なヒト組織(ドナーA、B、C、D,中国食品医薬品検定研究院、国家薬物安全性評価監測センターより提供)のパラフィン切片を、実験グループ(ビオチン標識のAT132)、ポジティブコントロールグループ(ビオチン標識のヒュミラ)、ネガティブコントロールグループ(PBSバッファー)の3グループに分け、組織交差反応後の組織切片の着色状況を観察した。
ヒト鼻ポリープのパラフィン切片および3つの正常なニクイザル組織(ドナーA、B、C,中国食品医薬品検定研究院、国家薬物安全性評価監測センターより提供)のパラフィン切片を、実験グループ(ビオチン標識のAT132)、ポジティブコントロールグループ(ビオチン標識のヒュミラ)、ネガティブコントロールグループ(PBSバッファー)の3グループに分け、組織交差反応後の組織切片の着色状況を観察した。
Claims (11)
- SEQ ID NO: 1に示されるアミノ酸配列またはそれに対し少なくとも75%の相同性を保持するアミノ酸配列を有する重鎖と、SEQ ID NO: 2に示されるアミノ酸配列またはそれに対し少なくとも75%の相同性を保持するアミノ酸配列を有する軽鎖と、を含む抗腫瘍壊死因子ヒト化モノクローナル抗体。
- SEQ ID NO: 1に示されるアミノ酸配列またはそれに対し少なくとも75%の相同性を保持するアミノ酸配列を有する重鎖と、SEQ ID NO: 2に示されるアミノ酸配列またはそれに対し少なくとも75%の相同性を保持するアミノ酸配列を有する軽鎖と、を含み、
前記重鎖および/または軽鎖が1〜5個のアミノ酸残基の挿入、欠失または保守的置換により修飾されるものである抗腫瘍壊死因子ヒト化モノクローナル抗体。 - 前記の重鎖可変領域の相補性決定領域CDR-H1はSEQ ID NO: 3に示されるアミノ酸配列であり、CDR-H2はSEQ ID NO: 4に示されるアミノ酸配列であり、CDR-H3はSEQ ID NO: 5に示されるアミノ酸配列であり、
フレームワーク領域FR-H1において、第16位のアミノ酸AがEで置換でき、第17位のアミノ酸SがTで置換でき、第20位のアミノ酸IがVで置換でき(SEQ ID NO: 11);FR-H2において、第3位のアミノ酸KがRで置換でき、第9位のアミノ酸GがSで置換でき(SEQ ID NO: 12);FR-H3において、第3位のアミノ酸TがVで置換でき、第7位のアミノ酸EがDで置換でき、第10位のアミノ酸VがTで置換でき、第14位のアミノ酸FがYで置換でき、第19位のアミノ酸SがTで置換でき、第27位のアミノ酸TがVで置換でき(SEQ ID NO: 13)、
該ヒト化抗体の軽鎖の可変領域の相補性決定領域CDR-L1はSEQ ID NO: 6に示されるアミノ酸配列であり、CDR-L2はSEQ ID NO: 7に示されるアミノ酸配列であり、CDR-L3はSEQ ID NO: 8に示されるアミノ酸配列であり、

- 請求項4に記載の核酸配列を含むベクター。
- 発現を容易にするために核酸に操作可能に連結されるプロモーターをさらに含む請求項5に記載のベクター。
- 請求項5または6に記載のベクターを含む宿主細胞。
- hTNFαの診断性解析のための医薬の製造における、請求項1〜3のいずれかに記載の抗腫瘍壊死因子ヒト化モノクローナル抗体の使用。
- hTNFα関連疾患の治療のための医薬の製造における、請求項1〜3のいずれかに記載の抗腫瘍壊死因子ヒト化モノクローナル抗体の使用。
- 前記hTNFα関連疾患が、膿血症、自己免疫病、悪性腫瘍、肺機能障害、移植による拒絶反応、細菌性髄膜炎、脳マラリア、エイズ及びエイズ関連症候群(ARC)、移植後のサイトメガロウイルス感染症である、請求項9に記載の使用。
- 請求項1〜3のいずれかに記載の抗腫瘍壊死因子ヒト化モノクローナル抗体を含む医薬組成物であり、抗hTNFαヒト化抗体および薬学的に許容される賦形剤を含む医薬組成物。
