本発明による有用な遺伝子および毒素には、開示される完全長配列だけでなく、本明細書で具体的に例示される毒素の特徴的な殺虫活性を保持するこれらの配列の断片、変異体、突然変異体、および融合タンパク質も含まれる。本明細書で用いるように、遺伝子の「変異体」または「変形形態」という用語は、同じ毒素をコードするか、殺虫活性を有する同等毒素をコードするヌクレオチド配列を指す。本明細書で用いるように、用語「同等毒素」は、標的有害生物に対して特許請求する毒素と同じか事実上同じである生物的活性を有する毒素を指す。
本発明の特定の毒素が、本明細書で具体的に例示された。これらの毒素は本発明の毒素の例示にすぎないので、本発明が例示毒素と同じか類似した殺虫活性を有する変異体または同等毒素(および同等毒素をコードするヌクレオチド配列)を含むことは容易に明らかなはずである。同等毒素は、例示される毒素とのアミノ酸相同性を有する。このアミノ酸相同性は、一般的に75%を超え、好ましくは90%を超え、最も好ましくは95%を超える。アミノ酸相同性は、生物的活性を担う毒素の重要領域、又は最終的に生物的活性を担う3次元構造の決定に関与する毒素の重要領域で最も高くなる。この点に関して、それらの置換が活性に重要でない領域にあるか、分子の3次元構造に影響を及ぼさない保存的アミノ酸置換であるならば、特定のアミノ酸置換が許容可能であり、期待できる。例えば、アミノ酸は以下のクラスに入れることができる:無極性、無電荷極性、塩基性および酸性。1つのクラスのアミノ酸が同じ種類の別のアミノ酸で置換される保存的置換は、その置換が化合物の生物的活性を実質的に変更しない限り本発明の範囲内である。下記は、各クラスに属するアミノ酸の例のリストである。
本発明の毒素をコードする遺伝子は、多種類の微生物または植物宿主に導入することができる。毒素遺伝子の発現は、直接または間接的に、殺虫剤の細胞内での生成および維持をもたらす。本発明の両毒素を発現するBt株を作製するために、接合移入および組換え移入を用いることができる。他の宿主生物体を毒素遺伝子の一方または両方で形質転換し、次に相乗効果を達成するために用いることもできる。適する微生物宿主、例えばシュードモナス(Pseudomonas)で、微生物を有害生物の位置へ施用することができ、そこでそれらは増殖して摂取される。この結果が有害生物の防除である。あるいは、毒素遺伝子を受け入れる微生物は、毒素の活性を長引かせ、細胞を安定させる条件の下で処理することができる。毒性活性を保持する処理細胞は、次に標的有害生物の環境に施用することができる。
Bt毒素を発現するバチルス・チューリンゲンシス(Bacillus thuringiensis)または組換え体の細胞は、毒素活性を長引かせ、細胞を安定させるために処理することができる。形成される殺虫剤マイクロカプセルは、安定化された細胞構造の中にBt毒素(複数可)を含み、このマイクロカプセルが標的有害生物の環境へ施用されるときに毒素を保護する。適する宿主細胞には、原核生物または真核生物のいずれかが含まれてよく、通常、哺乳動物などの高等生物体に有毒である物質を生成しない細胞に限定される。しかし、毒性物質が不安定であるか、哺乳動物宿主への毒性のいかなる可能性も避けるために施用量が十分に低い場合、高等生物体にとって有毒である物質を生成する生物体が用いることもできる。宿主としては、原核生物および真菌類などの下等真核生物が特に興味深い。
Bt殺虫性遺伝子(複数可)を含む細胞宿主は、DNA構築物が選択有利性を提供し、細胞の実質的に全てまたは全てがBt遺伝子を保持するように選択培地を提供する、任意の便利な栄養培地で増殖させることができる。これらの細胞は、従来の方法に従って次に収穫することができる。あるいは、収穫する前に細胞を処理することができる。
誘引剤ならびにBt分離株の胞子、結晶および毒素、または本明細書で開示されるBt分離株から入手できる遺伝子を含む組換え体微生物を含む製剤化された餌粒剤は、土壌に施用することができる。製剤化された製品は、種子コーティングまたは作物サイクルの後期段階で根部処理もしくは植物全体処理として施用することもできる。Bt細胞の植物および土壌処理は、様々な不活性の材料、例えば無機鉱物(フィロシリケート、炭酸塩、硫酸塩、リン酸塩など)または植物材料(粉末状の穂軸、籾殻、クルミ殻など)と混合することによって、水和剤、粒剤または粉剤として使用することができる。製剤は、展着助剤、安定化剤、他の殺虫性添加剤または界面活性剤を含むことができる。液体製剤は水性または非水性であってよく、フォーム、ゲル、懸濁液、乳剤などとして使用することができる。これらの成分には、レオロジー調整剤、界面活性剤、乳化剤、分散剤またはポリマーが含まれてよい。
本発明の殺虫性タンパク質の生成のための好ましい組換え体宿主は、形質転換された植物である。本明細書で開示されるようなBt毒素タンパク質をコードする遺伝子は、当技術分野で周知である様々な技術を用いて植物細胞に挿入することができる。例えば、大腸菌(Escherichia coli)の複製系および形質転換細胞の選択を可能にするマーカーを含む多数のクローニングベクターが、高等植物への外来遺伝子の挿入のための調製のために利用できる。ベクターとしては、例えば、とりわけpBR322、pUCシリーズ、M13mpシリーズ、pACYC184が挙げられる。したがって、Bt毒素タンパク質をコードする配列を有するDNA断片は、適する制限部位でベクターに挿入することができる。生じたプラスミドは、大腸菌(E. coli)への形質転換のために用いられる。大腸菌(E. coli)細胞は適する栄養培地で培養され、次に収穫され、溶解される。プラスミドを回収する。配列分析、制限酵素解析、電気泳動および他の生化学的分子生物学的方法が、分析方法として一般に実行される。各操作の後、用いたDNA配列を切断して次のDNA配列に連結することができる。各プラスミド配列は、同じか他のプラスミドにクローニングすることができる。植物への所望の遺伝子の挿入の方法によっては、他のDNA配列が必要なこともある。例えば、植物細胞の形質転換のためにTiまたはRiプラスミドが用いられるならば、TiまたはRiプラスミドT−DNAの少なくともライトボーダー、しかし多くの場合はライトボーダーとレフトボーダーとが、挿入される遺伝子の隣接領域として連結されなければならない。植物細胞の形質転換のためのT−DNAの使用は集中的に研究されており、EP120516、Lee and Gelvin (2008)、Hoekema (1985)、Fraley et al., (1986)およびAn et al., (1985)で十分に記載され、当技術分野でよく確立されている。

Cry毒素のヨウ素化。
Iodo−BeadまたはIodo−gen(Pierce)を用いて、精製されたトランケーション化Cry毒素をヨウ素化した。簡潔には、2つのlodo−Beadを500μlのリン酸緩衝食塩水、PBS(20mMリン酸ナトリウム、0.15M NaCl、pH7.5)で2回洗浄し、鉛シールドの裏で1.5ml遠心管に入れた。これに、100μlのPBSを加えた。フード内で、および適切な放射能取扱い技術を用いることにより、0.5mCi Na125I(17.4Ci/mg、Lot0114、Amersham)をIodo−Beadを有するPBS溶液に加えた。構成成分を室温で5分間反応させ、次に高度に純粋なトランケーション化Cryタンパク質の2〜25μgをその溶液に加え、さらなる3〜5分間反応させた。Iodo−Beadから溶液を取り出し、それをPBSで平衡させた0.5mlの脱塩Zebaスピンカラム(InVitrogen)に加えることによって、反応を停止した。Iodo−Beadを各々10μlのPBSで2回洗浄し、洗浄溶液も脱塩カラムに加えた。放射性溶液を1,000×gで2分間の遠心によって脱塩カラムを通して溶出した。Cry1Daの場合、放射能標識法を行うためにlodo−gen法を用いた。この手法を用いて、100mMリン酸緩衝液(pH8)中のCry毒素を小さな0.5mlポリミキシンカラムに複数回通すことによって、リポポリサッカライド(LPS)を先ず除いた。Iodo−genチューブ(Pierce Chem.Co.)に20μgの無LPSのCry1Da毒素を加え、次に0.5mCiのNa125Iを加えた。反応混合液を25℃で15分間振盪させた。溶液をチューブから取り出し、50μlの0.2Mの非放射性標識NaIを加えて反応をクエンチした。緩衝液を3回交換してタンパク質をPBSに対して透析し、未結合の125Iを除去した。
可溶性BBMVの調製および分画。
終齢スポドプテラ・フルギペルダ(Spodoptera frugiperda)、オストリニア・ヌビラリス(Ostrinia nubilalis)またはヘレオチス・ゼア(Heleothis zea)幼虫を一晩絶食させ、次いで、翌朝、氷の上で15分間冷やしたのちに解剖した。中腸組織を体腔から取り出し、外皮に接着した後腸を残した。中腸を、仕入先の推奨通りに希釈したプロテアーゼ阻害剤カクテル1(SigmaP−2714)を補充した、9倍容量の氷冷ホモジナイゼーション緩衝液(300mMマンニトール、5mM EGTA、17mMトリス、塩基、pH7.5)に入れた。15ストロークのガラス組織ホモジナイザーで組織をホモジナイズした。Wolfersberger(1993)のMgCl2沈殿法によってBBMVを調製した。簡潔には、300mMマンニトール中の等量の24mM MgCl2溶液を中腸ホモジネートと混合し、5分間撹拌し、氷上で15分間静置させた。4℃で15分間、2,500×gで溶液を遠心分離した。上清を保存し、元の容量の0.5倍希釈のホモジナイゼーション緩衝液にペレットを懸濁して再び遠心分離した。2つの上清を合わせ、4℃で30分間、27,000×gで遠心分離してBBMV分画を形成した。ペレットを10mlホモジナイゼーション緩衝液に懸濁し、プロテアーゼ阻害剤を追加し、4℃で30分間、27,000×gで再び遠心分離してBBMVを洗浄した。生じたペレットをBBMV保存緩衝液(10mM HEPES、130mM KCl、10%グリセロール、pH7.4)に懸濁して、約3mg/mlタンパク質の濃度にした。タンパク質濃度は、標準としてウシ血清アルブミン(BSA)を用いてBradford法(1976)を用いることによって測定した。製造業者の説明書に従ってSigmaアッセイを用いて、試料を凍結させる前にアルカリ性ホスファターゼの測定を行った。BBMV分画中のこのマーカー酵素の比活性は、中腸ホモジネート分画で見られるものと比較して一般的に7倍増加した。BBMVを250μlの試料に等分し、液体N2で瞬間冷凍し、−80℃で保存した。
(1カクテル構成成分の最終濃度(μΜ)は、AEBSF(500)、EDTA(250mM)、ベスタチン(32)、E−64(0.35)、ロイペプチン(0.25)およびアプロチニン(0.075)である。)
BBMVへのl25I Cryタンパク質の結合。
結合アッセイで用いるBBMVタンパク質の最適量を決定するために、飽和曲線を作成した。125I放射性標識Cryタンパク質(0.5nM)を、結合緩衝液(8mM NaHPO4、2mM KH2PO4、150mM NaCl、0.1%ウシ血清アルブミン、pH7.4)中に0〜500μg/mlの様々な量のBBMVタンパク質と、28℃で1時間インキュベートした。全容量は、0.5mlであった。1.5ml遠心管から500μl遠心管に反応混合液の150μlを3つ組で採取し、室温で6分間、14,000×gで試料を遠心分離することによって、結合した125I Cryタンパク質を未結合のものから分離した。上清を静かに除去し、氷冷結合緩衝液でペレットを3回静かに洗浄した。ペレットを含む遠心器の底を切り取り、13×75mmガラス培養試験管に入れた。試料をガンマ計数器でそれぞれ5分間計数した。試料に含まれる計数をバックグラウンド計数(タンパク質なしでの反応)から差し引き、BBMVタンパク質濃度に対してプロットした。用いるタンパク質の最適量は、0.15mg/mlのBBMVタンパク質であると決定された。
図1は、非標識の相同的Cry1Ab(◆)および非相同的Cry1Be(●)による競合に対する、ECB由来BBMVにおける125I Cry1Ab(0.5nM)の特異的結合の百分率を示す。Cry1Abによる相同的競合の置換曲線は、約0.5nMのCry1Abで放射性リガンドの50%の置換を示すS字形の曲線をもたらす。Cry1Beは125I Cry1Beをその結合部位から置換もするが、125I Cry1Abの50%をその結合部位から置換するために、約40nMの濃度(Cry1Abによって必要とされるより80倍高い)を必要とする。
図2は、非標識の相同的Cry1Ab(◆)および非相同的Cry1Be(●)による競合に対する、FAW由来BBMVにおける125I Cry1Ab(0.5nM)の特異的結合の百分率を示す。Cry1Abによる相同的競合の置換曲線は、約0.3nMのCry1Abで放射性リガンドの50%の置換を示すS字形の曲線をもたらす。Cry1Beは約300nMの濃度で125I Cry1Abを50%置換し、すなわちCry1Abによって必要とされるよりも約1,000倍多い。エラーバーは、2つ組の測定から得られた値域を表す。
Claims (23)
- Cry1Ab殺虫性タンパク質をコードするDNAおよびCry1Be殺虫性タンパク質をコードするDNAを含むトランスジェニック植物。
- Cry2A、Cry1IおよびDIG−3からなる群から選択される第三の殺虫性タンパク質をコードするDNAをさらに含む、請求項1に記載のトランスジェニック植物。
- 殺虫性Cry1Faタンパク質をコードするDNAと、Cry1Fa、Vip3Ab、Cry1CaおよびCry1Eからなる群から選択される第四の殺虫性タンパク質をコードするDNAとをさらに含む、請求項3に記載のトランスジェニック植物。
- 請求項1から3のいずれかに記載の植物の種子。
- 非Bt緩衝帯植物および請求項1から3のいずれかに記載の複数の植物を備える植物の圃場であって、前記緩衝帯植物は前記圃場の全ての作物植物の40%未満を構成する、圃場。
- 前記緩衝帯植物が前記圃場の全ての作物植物の30%未満を構成する、請求項5に記載の植物の圃場。
- 前記緩衝帯植物が前記圃場の全ての作物植物の20%未満を構成する、請求項5に記載の植物の圃場。
- 前記緩衝帯植物が前記圃場の全ての作物植物の10%未満を構成する、請求項5に記載の植物の圃場。
- 前記緩衝帯植物が前記圃場の全ての作物植物の5%未満を構成する、請求項5に記載の植物の圃場。
- 前記緩衝帯植物がブロックまたは帯状地にある、請求項5に記載の植物の圃場。
- 非Bt緩衝帯植物からの緩衝帯種子および請求項4に記載の複数の種子を含む種子混合物であって、前記緩衝帯種子は混合物の全ての種子の40%未満を構成する、種子混合物。
- 前記緩衝帯種子が混合物の全ての種子の30%未満を構成する、請求項11に記載の種子混合物。
- 前記緩衝帯種子が混合物の全ての種子の20%未満を構成する、請求項11に記載の種子混合物。
- 前記緩衝帯種子が混合物の全ての種子の10%未満を構成する、請求項11に記載の種子混合物。

- トウモロコシ、ダイズおよびワタからなる群から選択される、請求項1から3のいずれかに記載の植物。
