Description
<1>二酸化塩素混合物Aの調製
以下の(1)〜(5)を混合し、二酸化塩素混合物Aを調整した。
(1)セイバーケミカルエンジニアリング社製の二酸化塩素希釈液3600ppm、900ml
(2)アミノ酸銀(株式会社CSL製)0.025%、90ml
(3)アパタイトチタン20%希釈液(株式会社CSL社製)0.1ml
(4)エタノール10ml
(5)グリセリン10ml
<2>二酸化塩素混合物Bの調製
以下の(1)〜(5)を混合し、二酸化塩素混合物Bを調整した。
(1)セイバーケミカルエンジニアリング社製の二酸化塩素希釈液3600ppm、900ml
(2)アミノ酸亜鉛(株式会社CSL製)0.025%、90ml
(3)アパタイトチタン20%希釈液(株式会社CSL社製)0.1ml
(4)エタノール10ml
(5)グリセリン10ml
<3>二酸化塩素混合物Cの調製
以下の(1)〜(6)を混合し、二酸化塩素混合物Cを調整した。
(1)セイバーケミカルエンジニアリング社製の二酸化塩素希釈液3600ppm、900ml
(2)アミノ酸銀(株式会社CSL製)0.03%、45ml
(3)アミノ酸亜鉛(株式会社CSL社製)0.03%、45ml
(4)アパタイトチタン20%希釈液(株式会社CSL社製)0.1ml
(5)エタノール10ml
(6)グリセリン10ml
<4>二酸化塩素混合物Dの調製
以下の(1)〜(6)を混合し、二酸化塩素混合物Dを調整した。
(1)セイバーケミカルエンジニアリング社製の二酸化塩素希釈液3600ppm、900ml
(2)アミノ酸銀(株式会社CSL製)0.05%、45ml
(3)アミノ酸亜鉛(株式会社CSL社製)0.05%、45ml
(4)アパタイトチタン20%希釈液(株式会社CSL社製)0.1ml
(5)エタノール10ml
(6)グリセリン10ml
<5>二酸化塩素混合物Eの調製
固形分であるトリクロロイソシアヌル酸30%と、炭酸水素ナトリウム69%に、アパタイトチタンに金属イオンを被覆した固形分(上記混合物A〜Dとほぼ同量のアパタイトチタンおよび金属イオンを含む)を混合し、圧力成型し固形化して、二酸化塩素混合物Eを調製した。
1.試験方法
(1)スギの雄花から採取した花粉をスライドガラスに散布したものを9枚用意した。
(2)シャーレに、実施例1で調整した二酸化塩素混合物A(3500ppm)と、精製水をそれぞれ30ml入れた。また、何も入っていない空のシャーレも用意した。
(3)シャーレの上に、スライドガラスを固定して蓋付きの容器に入れた。
(4)インキュベーター内温度を25℃に設定し、インキュベートを開始した。
(5)経過時間毎(インキュベート開始0時間後、6時間後、12時間後)にスライドがガラスを取り出し、花粉の形態を顕微鏡観察し、花粉分子の形態変化が1分子以上確認された場合は(+)と判定し、顕著な形態変化が確認された場合は(++)と評価した。
結果を図1に示す。図1に示したように、二酸化塩素混合物A(3500ppm)が自然気化した場合、6時間経過後および12時間経過後に、顕著な花粉の破壊が確認された。一方、精製水を入れた場合および何も入れなかった場合(DRY)には、顕著な花粉の破壊は確認されなかった。
スギ花粉アレルゲン2種(Cryj-1,Cryj-2)の免疫染色(蛍光抗体法)を行い、抗原性への影響を確認した。具体的には、以下の方法で試験を行った。
<A>自然気化による接触
(1)スギの雄花から採取した花粉を、2区画に分けたスライドガラス上に載せたものを4枚用意した。
(2)二酸化塩素混合物A(3500ppm)を20mlと、対照検体として精製水20mlをそれぞれビーカーに入れた。
(3)ビーカー上にスライドガラスを固定し、蓋付き容器に入れた。
(4)インキュベーター内温度を25℃に設定し、蓋付き容器を入れ、インキュベートを開始した。
(5)0時間、12時間経過後のスライドガラスを取り出し、さらに以下の操作を行った。
(6)無蛍光シールを用いて、(5)のスライドガラス上の花粉を回収した。
(7)回収した花粉を洗浄・ブロッキング後、抗Cryj-1抗体、Cryj-2抗体を用いて一次抗体反応を行った。
(8)洗浄後、Alexa488ラベル抗体を用いて二次抗体反応を行った。
(9)蛍光顕微鏡(Axiovert(カールツァイス))を用いて、顕微鏡観察を行った。抗原性に変化がない(免疫染色による陽性部位の蛍光強度に変化がない)場合は(−)、変化が認められる場合に(+)、消失あるいは顕著な変化が認められる場合は(++)と評価した。
図2は、二酸化塩素混合物を配置してから0時間後の、Cryj-1およびCryj-2の状態を示す顕微鏡写真である。Cryj-1およびCryj-2について、図中の右側は、蛍光顕微鏡による写真であり、白く映る部分は抗原性がある部分を示している。
Claims (7)
- 二酸化塩素ガス発生成分、アパタイト系化合物および金属イオンを含むことを特徴とする花粉アレルゲン不活性化剤。
- 金属イオンは、銀イオン、チタンイオン、亜鉛イオン、銅イオンのうちの1種または2種以上であることを特徴とする請求項1の花粉アレルゲン不活性化剤。
- 固形化されていることを特徴とする請求項1または2の花粉アレルゲン不活性化剤。
- 請求項1から3のいずれかの花粉アレルゲン不活性化剤を含む機能性物品。
- 請求項1から3のいずれかの花粉アレルゲン不活性化剤を含む化粧品または医薬品。
- 飛散する花粉のアレルゲン不活性化方法であって、請求項1から3のいずれかの花粉アレルゲン不活性化剤からの気体によって花粉アレルゲンを不活性化させることを特徴とする花粉アレルゲン不活性化方法。
- アレルゲン不活性化剤からの気体には、二酸化塩素および金属イオンが含まれることを特徴とする請求項6の花粉アレルゲン不活性化方法。
