Abstract
【解決手段】矩形の設置面積を有し、気管空間118によって隔てられた多重の細胞培養チャンバ111を有する、多層細胞培養装置100であって、細胞培養チャンバのそれぞれが、汚染の危険性を低減しつつ、細胞培養装置の内外への流体の移動が可能になるように構造化され、配置された、ポート120およびポートカバー121または外部マニホールドを有する装置と、該装置の使用方法。各細胞培養チャンバが少なくとも1つのポートを有し、少なくとも3つの細胞培養チャンバと少なくとも2つの1体化した気管空間と、3つの細胞培養チャンバの少なくとも1つのポートと連結する構造化され、配置された取り外し可能なマニホールドとを備えている多細胞培養装置。

Description
本発明の1つの実施の形態では、多数の細胞増殖チャンバ111は、多層フラスコ100の本体101と一体となり、細胞増殖用の栄養培地で十分に満たされうることから、個別の細胞増殖チャンバ111は、ガス透過性の膜113上での細胞増殖を可能にする。多層フラスコ100を貫く一連の気管空間118は、ガス透過性表面113上で増殖する細胞117間、多層フラスコ内部の個別の細胞増殖チャンバ111の培地127、および外部環境にガス伝達を提供する。気管空間118は、ガス透過性表面113を通じて、細胞増殖チャンバ111内に位置する培地の酸素供給を可能にする。さらには、気管チャンバ118は空隙または空間の形をとって差し支えなく、液体の侵入が起きないようにする。結果として、気管空間118と交互に存在する多数の増殖チャンバ111を有する剛性の細胞培養多層フラスコ100は、大量の細胞117へのガスの均等分配の利益をもたらすように、協同的に構成される。
図17を参照すると、多層フラスコ100の2つの側に、入口ポート1740および出口ポート1750でありうる、ポート1720が存在しうる。1つの実施の形態では、入口ポート1740に挿入されたマニホールド1700のカニューレ1710を通じて、流体が多層フラスコ100に入る際に、行き場がなくなった流体またはガスがフラスコのもう1つの側にある出口ポート1750を通じて多層フラスコ100から流出しうる。流体は、例えばマニホールドまたはフラスコに取り付けたポンプまたは真空を使用することによって、マニホールドに陽圧または陰圧を提供することにより、マニホールドを通じて移動させることができる(図20参照)。多層フラスコ100の細胞培養チャンバが流体で完全に満たされたら、マニホールド1700のカニューレ1710を入口ポート1740から取り外すことができ、入口ポート1740と出口ポート1750の両方をポートカバー1721で塞ぐことができる。入口および出口ポートの両方がポートカバーによって閉鎖されたこの配置では、多層フラスコ100は閉鎖された多層細胞培養系である。次に、この閉鎖された多層フラスコを、細胞増殖チャンバ内の細胞増殖表面が最大となる(すなわち、底板を下にして置く)ように回転させて、インキュベーターなどの細胞培養に適切な場所に置くことができる。さらなる実施の形態では、マニホールド1700はカニューレを有さず、ポートと直接液密封止を形成してもよい(例えば、図10参照)。
Claims (2)
- 多層細胞培養装置であって、
a.各細胞培養チャンバが少なくとも1つのポートを有する、少なくとも3つの細胞培養チャンバと、
b.少なくとも2つの一体化した気管チャンバと、
c.前記少なくとも3つの細胞培養チャンバの前記少なくとも1つのポートと連結するように構造化され、配置された、取り外し可能なマニホールドと、
を備え、
d.前記取り外し可能なマニホールドが、前記少なくとも3つの細胞培養チャンバの前記少なくとも1つのポートと、開放可能な液密封止を形成するように構造化され、配置されたことを特徴とする多層細胞培養装置。 - 前記取り外し可能なマニホールドがバルブを含むことを特徴とする請求項1記載の多層細胞培養装置。
