Abstract
Description

本発明は、産生のための組換えDNA技術および細胞培養技術に基づいて、ワクチン、診断ツールおよびR&Dツールとして使用するための新たなウイルス様粒子を産生することを目的とする。本発明の粒子は、可能性のあるパンデミックインフルエンザの大流行と戦うに適したワクチンへの要求を充足する。本発明の組換えウイルス様粒子は、(a)同じウイルスの異なるウイルス株および/または(b)同じウイルスの異なる血清型および/または(c)異なる宿主に特異的な異なるウイルス株のいずれかから選択される、いくつかの、特に2個以上、例えば2個、3個、4個もしくは5個、またはまた3の倍数、例えば6個、9個もしくは12個の異なるエピトープまたはエピトープを含む異なるタンパク質を取り込んだ、ポリペプチド鎖によってアセンブルされている。その後、これらのエピトープは粒子表面上にディスプレイされる。異なる株、異なる血清型および/または異なる宿主に特異的なウイルスに由来するエピトープの選択により、天然で起こるような、例えば2009年4月のブタインフルエンザの大流行中に観察されたようなウイルスの天然の変化を模倣した多機能性のウイルス様粒子が得られる。当技術分野において最新技術のウイルス様粒子は、単一のタンパク質から構成されるか、または同じウイルス株に由来する3個までの異なるエピトープを含むかのいずれかである。本発明の粒子は、昆虫細胞、細菌細胞および哺乳動物細胞などの宿主細胞における産生のために使用される、単一のDNAベクター(ウイルスまたはプラスミドのいずれかに基づく)によってコードされる。好ましい態様において、DNAベクターはバキュロウイルスベクターであり、そして宿主細胞は昆虫細胞である。
(1)同じウイルスの2個または3個の異なる株または血清型に由来する、同じ型の表面タンパク質;
(2)異なるウイルス株に由来する、例えばインフルエンザウイルスH5N1株およびH1N1株に由来する、2個を超える異なる表面タンパク質の混合物;
(3)異なる宿主に特異的なウイルス、例えばブタ、ヒトおよび/またはトリ宿主に特異的なインフルエンザウイルスの合わせられた、異なる表面タンパク質の混合物
を含む。
E.coli細胞における増殖を可能とするMultiBacまたはYFPMultiBac(WO 2005/085456)と名付けられたバキュロウイルスベクター、およびCAP(登録商標)技術を、従来のシステム(Fitzgerald et al., Nature Methods, 3, 1021, 2006)を使用して組換えAcNPVs(Autographa californica核多角体病ウイルス(Autographa californica nuclear polyhedrosis virus)、バキュロウイルス)を生成するために使用する。10ngの多重遺伝子ベクターを、DH10MultiBacおよび/またはDH10YFPMultiBacコンピテント細胞において形質転換する。青色/白色スクリーニングおよびPCRによって陽性クローンを選択する。対応するMultiBac bacmidDNAを、Birnboim & Doly法を使用して単離する。1μgの多重遺伝子MultiBac bacmidを、製造業者のプロトコールに従って、トランスフェクション試薬Fugene(Roche)を使用して0.9×106個のSf21(Invitrogen)細胞中にトランスフェクションすることによって、組換えAcNPVsを生成する。ウイルスの増殖は、以前に記載されている通りに行なう(Fitzgerald et al., Nature Methods, 3, 1021, 2006)。全ての組換えAcNPVの力価を、Bac-to-Bac-マニュアル(Invitrogen)に記載のプラークアッセイによって決定する。感染効率(MOI)、細胞数(TOI)および収集時刻(TOH)のようなタンパク質産生パラメーターを、小規模な発現研究を用いて分析する。
種々の構築物を製造するために、導入ベクターpFLに存在する増殖モジュールMを(WO 2005/085456およびCAP(登録商標)技術)、WO 2005/085456に記載の方法に従って使用する。エピトープのDNAを、元のウイルスからウイルスRNAを単離し、その後、PCRと合わせた逆転写を行なうことによって(インフルエンザウイルスエピトープについて)、または遺伝子合成によって(Geneart社によって提供)のいずれかで得る。逆転写を、製造業者のプロトコールに従って、RevertAid(登録商標)H Minus First strand cDNA合成キット(Fermentas)を使用して実施する。cDNA(2μL)をPCR反応のための鋳型として使用する。以下の条件を、製造業者のプロトコールに基づいて使用する。50μLの全容量の反応のために、0.2mM dNTP(NEB)、1.2%DMSO、0.5μM逆方向および正方向プライマー(Microsynth)、10μlの5×Phusion GC反応緩衝液、並びに2UのPhusion Hot Startポリメラーゼ(Finnzyme)を使用する。多重遺伝子会合のために適切な制限酵素部位(BstZ17I、SpeI、PmeI、AvrII)をPCRを使用して導入する。PCRフラグメントを制限酵素で切断し、その後、ライゲーションおよび形質転換プロセスを行ない、導入ベクターに増殖モジュールを組み込む。ライゲーションを、500ngの鎖状導入ベクター(pFL)、4μLのPCR産物および1UのT4−DNAリガーゼ(Fermentas)を使用して4℃で一晩かけて行なう。プラスミドを生成するために、4μLのライゲーション溶液を、50μlのコンピテントなDH5α細胞に加え、そして氷上で30分間インキュベーションする。42℃で30秒間の熱ショックおよび4℃で2分間の寒冷ショックの後、200μlのLB培地を加え、そして37℃および220rpmで1時間インキュベーションする。その後、80μlの細胞懸濁液を、適切な抗生物質(この場合、100μgのアンピシリンおよび100μgのゲンタマイシン)を含むLB寒天プレート上に播く。全手順を、全エピトープが導入ベクターに導入されるまで繰り返す。
このウイルスは、複数のエピトープをその表面上に提示するウイルス様粒子またはウイルスシミュレーターを生成するための、前記の複数のエピトープを含む。ウイルス様粒子を、種々の適用のために、例えばインフルエンザ分野におけるワクチンとして使用することができる。AcNPV由来バキュロウイルスは、2008/2009−VLPワクチン接種キャンペーンのためにWHOによって推奨されるウイルス株に由来する複数の異なるエピトープを含む。導入ベクターの全ての遺伝子は、WO2005/085456のプロトコールによる、MultiBac細胞中への部位特異的相同的組換えによって置き換えられる。
最善の発現構築物の決定の後、細胞株、細胞数(TOI)、組換えウイルス接種材料の量(感染効率、MOI)および収集時刻(TOH)のようなバイオテクノロジー産生パラメーターを、50mLのバイオリアクター中において決定する。異なるTOI、MOIおよびTOHのマトリックスを、Eibl、RiesenおよびJohn (Bioforum 03/2009)およびFriesen J. (Bachelor thesis, University of Applied Science, Esslingen, Germany)に従って設計する。分泌または細胞内多重エピトープウイルス様粒子の発現を、毎日の試料採取により6〜8日間観察する。細胞内粒子(例えばHPV)のために、細胞ペレットを、50mMトリスHCl、pH7.6、100mM NaCl、0.1%TritonX100を用いて溶解し、そして4℃および8000×gで10分間遠心分離にかける。上清中に存在するウイルス様粒子のエピトープを、特異的抗体を用いて、ドットブロット装置(Biometra)、次いでウェスタンブロットを使用してさらに確認する。最も高い収量が得られる条件を発現パラメーターとして規定し、3日間から4日間の収集時刻が好ましい。これらの規定されたパラメーターに従って、ウイルス様粒子を、振とうフラスコまたは揺動培養バッグ中のいずれかのfall armywormのSpodoptera frugiperda細胞Sf9およびSf21において産生する。多重エピトープインフルエンザウイルス様粒子発現のための、Sf21細胞は、以下の条件を用いて選択される:1.5×106個の細胞/mL、MOI 0.05および収集時刻は感染から4日後。細胞を、27℃で二酸化炭素の非存在下、ウシ胎児血清を補充することなく、増殖させる。規定の収集時刻に従って、分泌されたウイルス様粒子を、4℃で500〜1000×gで20分間の遠心分離によって回収する。粒子を含む上清容量を、100kDaのカットオフを有するカセット(Sartocon-Slice 200、SartoriusおよびCentramateOS, PALL)を使用したタンジェンシャルフローろ過による精製のために減少させる。ウイルス様粒子の精製を、計れるクロマトグラフィー法およびスクロース勾配超遠心分離法を用いて実施する。
異なるエピトープの存在を確認するために、精製した材料をSDS−PAGE、その後、クーマシー染色またはウェスタンブロットによって分析する。150μlの異なるクロマトグラフィー画分を、4〜12%Bis−Tris NuPAGEゲル(Invitrogen)にのせ、150Vで15分間および175Vで45分間流し、そしてSimplyBlueSafestain (Invitrogen)を使用してクーマシー染色する。イムノブロットのためにタンパク質をニトロセルロース膜(BioRAD)に19Vで40分間かけて半乾燥装置(BioRAD)を使用して転写する。5%無脂肪ドライミルク−トリスCl−Tween20(0.1%)溶液を用いて30分間かけて非特異的結合部位を遮断した後、膜を、HA、NAおよびマトリックスタンパク質に対する抗体と共に一晩かけて4℃でインキュベーションする。膜を数回、トリスCl−Tween20(0.1%)緩衝液を用いて洗浄する。一次抗体の起源に依存して、二次抗体は、検出のためのアルカリホスファターゼまたはセイヨウワサビペルオキシダーゼと接続された抗マウスまたは抗ウサギのいずれかである。これらのタンパク質の同じ場所における局在は、1つの発現ベクターおよび1つのバキュロウイルスの会合およびそれからの産生を示す(図3B、配列番号1から)。この同じ場所における局在はまた、遺伝子HA、NAおよび両方のマトリックスタンパク質M1およびM2(その膜アンカーを含む)を含む発現構築物についても示され得る。
VLPがその表面に赤血球凝集素タンパク質(HA)を正しく組み込んだかどうかを分析するために、ニワトリ由来の赤血球(RBC)を使用する標準的な赤血球凝集アッセイを実施する(図4、配列番号3のVLP)。精製VLPの2倍連続希釈を、V底96ウェルプレートにおいてPBS(1×)を用いて行なう。等量の赤血球(1%溶液)を加え、そして1時間4℃でインキュベーションする。ウェルの底上のRBC凝集体の外見は、赤血球凝集がないことを示す。力価は、RBCを凝集させる精製VLP溶液の最も高い希釈度の逆数として表現される。得られた結果は、VLPがニワトリ赤血球を凝集させることができ、そしてVLP表面上におけるHAの存在を間接的に示す。陰性対照(PBS)は凝集を全く示さない。
VLP(配列番号3から調製)の免疫原性(免疫系の刺激)をin vivoにおいて、それぞれ0週間目および3週間目にプライム・ブースト計画で皮下免疫化を受ける2群のマウスを使用して試験する。免疫化を、50または100μl(50または100ng)のVLP懸濁液を使用して実施する。VLP単独の免疫応答の品質を決定するために、アジュバントを全く使用しない。マウスを3週間目および6週間目に出血させ、そして血清を、免疫化VLPに対する抗体応答について調べるために分析する。得られた結果は、VLPが、抗体免疫応答を刺激するのに効果的であることを示す。最善の結果は、免疫化を100μlを用いて実施した場合に得られ、このことは用量依存的な免疫応答を示す。抗VLP抗体の量の明瞭な増加が、ブースト後に観察される。予期した通り、ナイーブな動物は免疫応答を全く示さない。
細胞株、細胞数(TOI)、組換えウイルス接種材料の量(感染効率、MOI)および収集時刻(TOH)のようなバイオテクノロジー産生パラメーターを、Eibl et al. (Bioforum 3/2009)に従って50mLのバイオリアクター(図7)中で決定する。これらの規定されたパラメーターに従って、ウイルス様粒子を、振とうフラスコまたは揺動培養バッグのいずれかの中のfall armywormのSpodoptera frugiperda細胞Sf9およびSf21において産生する。多重エピトープパピローマウイルス様粒子は、2×106個の細胞/mL、MOI 0.5および感染から3日後の収集時刻を使用して、Sf21細胞において配列番号2から発現される。細胞を、27℃で二酸化炭素の非存在下、ウシ胎児血清を補充することなく、増殖させる。規定の収集時刻に(感染から3日後)、細胞内ウイルス様粒子を500〜1000×gで20分間4℃での遠心分離によって回収する。細胞を低浸透圧リン酸緩衝液、その後、超音波を使用することによって溶解する。4℃および2000×gでの遠心分離工程後、上清を回収する。ウイルス様粒子の精製を、計れるクロマトグラフィー法およびスクロース勾配超遠心分離法を用いて実施する。クロマトグラフィー精製は、陽イオン交換クロマトグラフィー、陰イオン交換クロマトグラフィーおよびゲルろ過クロマトグラフィーを使用する多段階精製である。上清を、50mMリン酸緩衝液(pH7.4)中のFPLC−システム(AEKTA purifier, GE Healthcare)に接続したCaptoDEAEカラムにのせる。粒子を、50mMリン酸、1M NaCl、pH7.4を使用する線形勾配での漸増塩濃度で溶出する(図8A)。粒子を含む画分をプールし、そしてさらに、ヒドロキシアパタイトカラムを使用して精製する。結合を、20mMリン酸緩衝液(pH7.0)中で実施し、その後、500mMリン酸、150mM NaCl、pH7.0を用いての線形勾配溶出を実施する。多重エピトープ粒子を磨くために、ゲルろ過クロマトグラフィーを実施する。精製を、HighLoad Superdex200pgカラムを使用して50mMリン酸、150mM NaCl、pH7.4緩衝液中で実施する。全てのクロマトグラフィー工程をSDS−PAGEによって、その後、クーマシー染色およびイムノブロットによって分析する。
精製された材料における異なるエピトープの存在を確認するために、配列番号2から調製したVLPをSDS−PAGEにより、その後、クーマシー染色および(図8B)およびウェスタンブロット(図8C)によって分析する。イムノブロットのために、異なる血清型のL1タンパク質に対する抗体を使用する。150μlの異なるクロマトグラフィー画分を4〜12%Bis−トリスNuPAGEゲル(Invitrogen)にのせ、150Vで15分間および175Vで45分間流し、そしてSimplyBlueSafestain (Invitrogen)を使用してクーマシー染色する。イムノブロットのために、タンパク質を、半乾燥装置(BioRAD)を使用して19Vで40分間かけてニトロセルロース膜(BioRAD)上に転写する。5%無脂肪ドライミルク−トリスCl−Tween20(0.1%)溶液を用いて非特異的な結合部位を30分間かけて遮断した後、膜を、L1エピトープに対する抗体と共に一晩かけて4℃でインキュベーションする。Camvir抗体(SantaCruz)をHPV16検出のために使用し、そしてHPV18検出のために抗HPV18ab(Abcam)を使用する。膜を、トリスCl−Tween20(0.1%)緩衝液を用いて数回洗浄する。膜を、検出のためのアルカリホスファターゼと接続された抗マウス抗体と共に1時間インキュベーションする。エピトープを、BCIP/NPT溶液によって検出する。これらのタンパク質の同じ場所での局在は、1つの発現ベクターおよび1つのバキュロウイルスのアセンブルおよびそれからの産生を示す。
配列番号2から調製したヒトパピローマウイルス様粒子がその表面にL1タンパク質を正しく組み込んだかどうかを分析するために、標準的なELISAアッセイを実施する。精製VLPの2倍連続希釈を、V底96ウェルプレートにおいてPBS(1×)を用いて行なう。等量の血清型特異的抗体(濃度1:1000)を加え、そして1時間37℃でインキュベーションする。L1タンパク質への抗体の適切な結合を、セイヨウワサビペルオキシダーゼを有する二次抗体および化学発光検出システムを使用して検出する。得られた結果は、用量依存的な組換え発現エピトープへの抗体の結合を示す。陰性対照(PBS)は全く結合を示さなかった。
Claims (16)
- (a)同じウイルスの異なるウイルス株および/または(b)同じウイルスの異なる血清型および/または(c)異なる宿主に特異的な異なるウイルス株のいずれかから選択される、2個以上の異なるエピトープまたはエピトープを含む異なるタンパク質を含む、組換えウイルス様粒子。
- 3個以上の異なるエピトープまたはエピトープを含む異なるタンパク質を含む、請求項1記載の組換えウイルス様粒子。
- 4個以上の異なるエピトープまたはエピトープを含む異なるタンパク質を含む、請求項1記載の組換えウイルス様粒子。
- 6個、9個または12個のエピトープまたはエピトープを含む異なるタンパク質を含む、請求項1記載の組換えウイルス様粒子。
- エピトープが、3個以上の異なるウイルス株または血清型に由来する、請求項1〜4のいずれか記載の組換えウイルス様粒子。
- B細胞エピトープおよび/またはT細胞エピトープをさらに含む、請求項1〜5のいずれか記載の組換えウイルス様粒子。
- 完全なウイルス様表面を形成するタンパク質並びに場合によりカプシドおよび/またはヌクレオポアタンパク質をさらに含む、請求項1〜6のいずれか記載の組換えウイルス様粒子。
- 蛍光タンパク質、粒子の精製目的のためにもしくは標識を付着するために有用なタンパク質、および/または輸送プロセスのために必要とされるタンパク質性構造をさらに含む、請求項1〜7のいずれか記載の組換えウイルス様粒子。
- ウイルスが、インフルエンザウイルスである、請求項1〜8のいずれか記載の組換えウイルス様粒子。
- ウイルスが、ヒトパピローマウイルスである、請求項1〜9のいずれか記載の組換えウイルス様粒子。
- (a)同じウイルスの異なるウイルス株および/または(b)同じウイルスの異なる血清型および/または(c)異なる宿主に特異的な異なるウイルス株のいずれかから選択される、異なるエピトープまたはエピトープを含む異なるタンパク質をコードする2個以上のポリヌクレオチドを含む、ベクター。
- 請求項1〜10のいずれか記載の組換えウイルス様粒子をコードする、請求項11記載のベクター。
- バキュロウイルスベクターである、請求項11または12記載のベクター。
- 配列番号1〜5から選択されるポリヌクレオチド配列を含む、請求項11記載のベクター。
- 請求項11〜14のいずれか記載のベクターを含む、宿主細胞。
