Abstract
Description
フェルスター共鳴エネルギー移動(FRET)は、近接して存在する2つの適当に選ばれた蛍光分子の間の強く距離に依存した(6乗に反比例する)非放射性エネルギー移動機構である[Foerster, T (1948) Ann Physik 2:55-75]。共鳴エネルギー移動(RET)は、ドナーとアクセプター蛍光団がFoerster半径(典型的な値は4〜7nm)内にあり、ドナーの発光スペクトルとアクセプターの吸収スペクトルが重なるときに実用的効率となる。RETは、通常、ドナーとアクセプターの近接から生じるドナー発光強度の減少またはアクセプター発光強度の増加(感作アクセプター発光として知られる)[Selvin, PR (1995) Biochem Spectroscopy 246: 300-334]のいずれかを測定することにより観測される。非蛍光アクセプター(消光剤として知られる)の場合、ドナー発光強度の変化が観測される。
従来の蛍光に基づく技術の検出感度は、生物学的試料マトリクスの自己蛍光、散乱励起光および吸収により制限され、アクセプターに基づく共鳴エネルギー移動に基づくアッセイにおいては、アクセプター特異的発光波長でのドナー発光のクロストークやドナー特異的励起波長でのアクセプターの直接励起によっても制限される。生物学的液体または血清に存在する多くの化合物やタンパク質は、もともと蛍光を発し、従来の蛍光団(fluorophore)の使用によりその感度が甚だしく制限される[Soini E and Hemmilae I (1979) Clin Chem 25: 353-361; Wu P and Brand L (1994) Anal Biochem 218:1-13]。強度測定に基づくホモジニアスな蛍光技術を使用した場合の別の主な問題は、内部フィルター効果および試料の光学特性のばらつきである。この欠点を是正するために試料希釈が使用されているが、常に分析感度が犠牲となる。分析応用における蛍光共鳴エネルギー移動の実行可能性は、長寿命発光と大きいストークシフトを有する蛍光ランタニドクリプタートおよびキレートが1990年代にドナーとして用いられると、著しく改善した[Mathis G (1993) Clin Chem 39:1953-1959; Wu P and Brand L (1994) Anal Biochem 218, 1-13; Selvin PR et al. 異なる光輝性ランタニドに基づくレポーターを利用する2つの新しい共鳴エネルギー移動に基づく方法[Mathis, G (1993) Clin Chem 39: 1953-1959; Blomberg, K et al. (1999) Clin Chem 45: 855-861]が、従来のFRETに基づくホモジニアスアッセイに関連した主要な問題を大幅に解決するために導入された。これらの方法は両方、従来の方法と比べて顕著な利点を提供するが、シグナル発生における特異性は、特に標識プローブが高濃度に存在する場合、放射性エネルギー移動(ドナー発光の吸収)によりなお制限される(たとえば、高ダイナミックレンジに達するか、または弱い相互作用の場合、結合を促進する)[H. Bazin, M. et al. (2001) Spectrochim. Acta, Part A, 57]。過剰な非結合アクセプターは、アクセプター特異的波長で放射性バックグラウンドシグナルのゆっくりとした減衰を生じるが、測定波長でのドナークロストークも、充分なスペクトル分解能が用いられない限り、バックグラウンドシグナルを増加できる。ランタニドキレートドナーと重複しないアクセプター(非重複FRET)の利用は[Hemmilae, I. and Laitala, V. (2005) Anal. Chem. 77:1483-1487; Laitala, V. and Hemmilae I. (2005) Analytica Chimica Acta 551: 73-78]、ドナー発光の再吸収を通じて可能性のあるバックグラウンドをさらに排除することができる。
該方法においては、
a)該ランタニドイオンキャリアキレートが、該バイオアッセイ法の条件において該ランタニドに充分強く結合し、結果として、該バイオアッセイ法の条件において、遊離のランタニドイオンが本質的には存在しない、すなわち1nmol/Lより少ない、好ましくは10pmol/Lより少ない遊離のランタニドイオンが存在する、または
b)該ランタニドイオンキャリアキレートは、該バイオアッセイ法の条件において該ランタニドに強く結合し、結果として、遊離のランタニドが本質的には存在しない、すなわち1nmol/Lより少ない、好ましくは10pmol/Lより少ない遊離のランタニドが存在し、かつ該ランタニドイオンを錯化する薬剤が少なくとも1pmol/Lの濃度でさらに用いられる、そして
該アンテナ配位子は、該ランタニドイオンに弱く結合する、すなわち該アンテナ配位子は単座、二座、三座または四座のいずれかである;ならびに
該第1のグループの第1認識エレメントおよび該第2のグループの第2認識エレメントによる該検体の認識が、
i)キレート相補性、すなわち該ランタニドを担持する該ランタニドイオンキャリアキレートと該アンテナ配位子との相補性による混合ランタニドキレート錯体の形成、およびそれによる蛍光の増加;または
ii)キレート脱相補性、すなわち該ランタニドを担持する該ランタニドイオンキャリアキレートが該アンテナ配位子から分離され、それによる蛍光の減少
のいずれかを生じる
バイオアッセイ法を提供する。
用語「蛍光」および「発光」は、マイクロ秒またはミリ秒蛍光寿命を有する遅延した蛍光、イオン性フォトルミネッセンス、アップコンバージョンに基づく抗−ストークフォトルミネッセンスおよびりん光などのフォトルミネッセンス、すなわち光により励起される発光、蛍光におよぶとして理解されるものとする。さらに、この用語は、電気発光および電気化学発光も対象とするものとする。
本発明により検体を検出するため、および/または検体の濃度を定量するための典型的なバイオアッセイ法は、ランタニドイオンキャリアキレートおよび第1認識エレメントを含み、該ランタニドイオンキャリアキレートがランタニドイオンキャリア配位子およびランタニドイオンを含む第1のグループ;アンテナ配位子および第2認識エレメントを含む第2のグループを用い、
a)該ランタニドイオンキャリアキレートが、該バイオアッセイ法の条件において、該ランタニドに、遊離のランタニドイオンが本質的には存在しない、すなわち1nmol/Lより少ない、好ましくは10pmol/Lより少ない遊離のランタニドイオンが存在するという結果となるのに充分強く結合する;または
b)該ランタニドイオンキャリアキレートは、該バイオアッセイ法の条件において、遊離のランタニドが本質的には存在しない、すなわち1nmol/Lより少ない、好ましくは10pmol/Lより少ない遊離のランタニドが存在するという結果となるのに充分に強く該ランタニドに結合し、該ランタニドイオンを錯化する薬剤が少なくとも1pmol/Lの濃度でさらに用いられる;そして
該アンテナ配位子は、該ランタニドイオンに弱く結合する、すなわち該アンテナ配位子は単座、二座、三座または四座のいずれかである;ならびに
該第1のグループの該第1認識エレメントおよび該第2のグループの第2認識エレメントによる該検体の認識が、
i)キレート相補、すなわち該ランタニドを担持するランタニドイオンキャリアキレートの該アンテナ配位子との相補性による混合ランタニドキレート錯体の形成、およびそれによる蛍光の増加;または
ii)キレート脱相補、すなわち該ランタニドを担持する該ランタニドイオンキャリアキレートが該アンテナ配位子から分離され、それによる蛍光の減少
のいずれかの結果となる。
a)logKLnL1は少なくとも12、好ましくは18以上であって、KLnL1は、イオンキャリア配位子とランタニドイオンとの錯体の溶液中での安定度定数を意味する;または
b)該ランタニドイオンを錯化する薬剤をさらに用いる場合、
i)logKLnL2は少なくとも12であって、KLnL2は、イオンキャリア配位子とランタニドイオンとの錯体の溶液中での安定度定数を意味する;および
ii)logKLnL3は少なくとも8であって、KLnL3は、該ランタニドイオンを錯化する該錯化剤とランタニドイオンとの溶液中での安定度定数を意味する。
ホモジニアスな近接プローブに基づくハイブリダイゼーションアッセイ
合成標的DNAオリゴヌクレオチド(5’−GATGCAGTAGCAGGAAGAGGATCGTAGCAATG−3’;配列番号1)、アミノ修飾プローブAオリゴヌクレオチド(5’−CATTGCTACGATCC(C6dT)C−3’;配列番号2)およびアミノ修飾プローブBオリゴヌクレオチド(5’−T(C2dT)CCTGCTACTGCATC−3’;配列番号3)は、シグマ−アルドリッチ社(セントルイス、MI)から購入した。プローブAは、3’−端付近に位置する第一級アミノ基修飾で、Eu3+イオンキャリアキレート、(N1−(4−イソチオシアネートベンジル)ジエチレントリアミン−N1,N2,N3,N3−テトラキス(アセテート)ユーロピウム(III)[Mukkala, V.-M. et al., (1989) Anal. Biochem., 176: 319]、Eu3+−N1)により標識し、そしてプローブBは、5’−端付近で、集光性アンテナ配位子(4−((イソチオシアネートフェニル)エチニル)ピリジン−2,6−ジカルボン酸、3d−アンテナ)により標識した。プローブA(25nmol)を、50mM炭酸塩緩衝液中、pH9.8、+37℃で20倍モル過剰のEu3+−N1と一晩インキュベートした。標識化反応の全量は50μLであった。プローブBの3d−アンテナによる標識化については、3d−アンテナをN,N−ジメチルホルムアミド(シグマ−アルドリッチ社)中に溶解し、水に溶解したオリゴヌクレオチドと混合し、そしてその後、炭酸塩緩衝液(pH9.8)を50mMの濃度まで添加した。標識化反応において、3d−アンテナのモル過剰は全量110μLで50倍であった。反応物をゆっくりと回転させながら+50℃で一晩インキュベートした。
シグナル発生における特異性
非相補的標的ヌクレオチド(5’−CTGCTCTATCCACGGCGCCCGCGGCTCCTCTC−3’;配列番号:4)は、Biomers.net社(ウルム、ドイツ)から購入した。実施例1に記載した実験を、標的オリゴヌクレオチドを非相補的標的オリゴヌクレオチドに代えて繰り返した。相補的標的オリゴヌクレオチドを可変濃度の非相補的32−merオリゴヌクレオチドに置換したことにより、相補的標的オリゴヌクレオチドの非存在下におけるのと同じ蛍光シグナルを生じ;可変濃度の非相補的標的オリゴヌクレオチドの存在下ではゼロ濃度の標的オリゴヌクレオチド;すなわちブランク対照と比較して、検出可能なシグナル差は観察されなかった。これは、本発明のシグナル発生機序は、高度に特異的であり、かつ2つの同時に起こる生体分子認識事象に依存するということを示す。
発光スペクトルおよび蛍光寿命
追加的プローブ、アミノ修飾プローブCオリゴヌクレオチド(5’−CATTGCTACGATCC(C2dT)C−3’;配列番号:5)は、シグマ−アルドリッチ社(セントルイス、MI)から購入し、本来蛍光である2,2’,2’’,2’’’−[[4−[(4−イソチオシアネートフェニル)エチニル]ピリジン−2,6−ジイル]ビス(メチレンニトリロ)]テトラキス(アセテート)ユーロピウム(III)(Eu3+−7d;図8cに概略構造)により標識した。プローブC(5nmol)を、20倍モル過剰のEu3+−7dと50mM炭酸塩緩衝液(pH9.8)中、+37℃で一晩インキュベートし、実施例1にA−Eu3+−N1について記載したように精製した。

アミノ修飾プローブオリゴヌクレオチド(プローブA、5’−CATTGCTACGATCC(C6dT)C−3’;配列番号2、およびプローブB、5’−T(C2dT)CCTGCTACTGCATC−3’;配列番号3)は、シグマ−アルドリッチ社(セントルイス、MI)から購入した。プローブAは、N1−(4−イソチオシアネートベンジル)ジエチレントリアミン−N1、N2、N3、N3−テトラキス(アセテート)ユーロピウム(III)[V.-M. Mukkala et al. (1989) Anal. Biochem., 176: 319](Eu3+−N1;図8aに概略構造)および本来蛍光である2,2’,2’’,2’’’−[[4−[(4−イソチオシアネートフェニル)エチニル]ピリジン−2,6−ジイル]ビス(メチレンニトリロ)]テトラキス(アセテート)ユーロピウム(III)[H. Takalo et al. (1994) Bioconjugate Chem, 5, 278](Eu3+−7d;図8cに概略構造)で標識し、プローブBは、4−((イソチオシアネートフェニル)エチニル)ピリジン−2,6−ジカルボン酸(アンテナ;図8bに概略構造)で標識した。プローブAの25nmolおよび5nmolを、それぞれ20倍モル過剰のEu3+−N1およびEu3+−7dと50mMの炭酸塩緩衝液(pH9.8)中、+37℃で一晩インキュベートした。標識化反応の全量は50μlであった。プローブBのアンテナによる標識化については、アンテナをN,N−ジメチルホルムアミド(シグマ−アルドリッチ社)に溶解し、水に溶解したオリゴヌクレオチドと混合し、その後、炭酸塩緩衝液(pH9.8)を50mMの濃度まで加えた。標識化反応において、アンテナのモル過剰は全量110μlで50倍であった。反応物は+50℃でゆっくりと回転させながら一晩インキュベートした。
ストレプトアビジンおよびアビジンのためのホモジニアスな近接プローブに基づくアッセイ
N−(6−アミノヘキシル)−5−(2−オキソヘキサヒドロ−1H−チエノ[3,4−d]イミダゾール−4−イル)ペンタンアミド((+)−ビオチニル−ヘキサンジアミン)を、4−((イソチオシアネートフェニル)エチニル)ピリジン−2,6−ジカルボン酸(アンテナ)にカップリングした。カップリング反応の生成物の概略構造は、図12aに説明されている。カップリングのために、(+)−ビオチニル−ヘキサンジアミンおよびアンテナをN,N−ジメチルホルムアミド(シグマ−アルドリッチ社、セントルイス、MO)に溶解し、混合し、その後、炭酸緩衝液(pH9.8)を50mMの濃度まで添加した。カップリング反応において、(+)−ビオチニル−ヘキサンジアミンのモル過剰は、全量270μlで3倍であった。反応物を+50℃でゆっくり回転させながら一晩インキュベートした。
テルビウム(III)イオンを用いるホモジニアスなハイブリダイゼーションアッセイ
合成標的DNAオリゴヌクレオチド(5’−GATGCAGTAGCAGGAAGAGGATCGTAGCAATG−3’;配列番号1)およびアミノ修飾プローブAオリゴヌクレオチド(5’−CATTGCTACGATCC(C2dT)C−3’;配列番号2)は、シグマ−アルドリッチ社(セントルイス、MI、USA)から購入し、アミノ修飾プローブBオリゴヌクレオチド(5’−T(C2dT)CCTGCTACTGCATC−3’;配列番号3)はサーモ サイエンティフィック社(ウォルサム、MA、USA)から購入した。プローブAは、3’端付近に位置する第一級アミノ基修飾で、Tb3+イオンキャリアキレート、(N1−(4−イソチオシアネートベンジル)ジエチレントリアミン−N1,N2,N3,N3−テトラキス(アセテート)テルビウム(III)、Tb3+−N1)により標識し、そしてプローブBは、5’端付近で、集光性アンテナ配位子(4−(3−(4−イソチオシアネートフェネチル)−2,4,6−トリメトキシフェニル)ピリジン−2,6−ジカルボン酸、TMP−アンテナ)により標識した。プローブA(10nmol)を20倍モル過剰のTb3+−N1と、50mM炭酸塩緩衝液(pH9.8)中、+37℃で一晩インキュベートした。標識化反応の全量は50μLであった。プローブBのTMP−アンテナによる標識化については、TMP−アンテナを75%N,N−ジメチルホルムアミド(シグマ−アルドリッチ)中に溶解し、オリゴヌクレオチドと混合し、そしてその後、炭酸塩緩衝液(pH9.8)を50mMの濃度まで添加した。標識化反応において、TMP−アンテナのモル過剰は全量70μLにおいて50倍であった。反応物はゆっくりと回転させながら+50℃で一晩インキュベートした。
テルビウム(III)イオンによる発光スペクトルおよび発光寿命
プローブA−Tb3+−N1およびプローブB−TMP−アンテナの標的オリゴヌクレオチド指向錯体の蛍光スペクトルおよび発光寿命をVarian Cary Eclipse 蛍光分光光度計(バリアン サイエンティフィック インスツルメント社、マルグレーブ、オーストラリア)を用いて測定した。標的オリゴヌクレオチド(0または10nM)をプローブA−Tb3+−N1(50nM)およびプローブB−TMP−アンテナ(50nM)とアッセイ緩衝液中で混合し、測定前にRTで60分間インキュベートした。
オリゴヌクレオチド指向キレート相補アッセイ(OCCA)
検出技術の性能は、閉管リアルタイムPCRにおいて、合成鋳型(5’CTTCAGCGCTACACACGCTCAAATCATCGAGGAAAACCGTATGAGAAACGGATCTAAGCTTGTCATTTGATAAAGCATCATGCAACATTAACCCGAGATACGATTTGTCCATATCTTTGATACGACGCCGCAAAAGCTCTTCCCAAGCCGAGTCTACAG3’;配列番号7;サーモ サイエンティフィック社、USA)の0〜105分子を増幅することにより調べた。リアルタイムPCRは、光学キャップ(MicroAmp(登録商標)、光学8−キャップストリップ、アプライド バイオシステムズ社、USA)を閉めた96ウェルPCRプレート(Thermo-Fast(登録商標)96 Robotic PCR Plate、サーモ サイエンティフィック社)を用いて行った。各40μlPCR反応は、500nMプライマー(5’プライマー 5’CTGTAGACTCGGCTTGGGAAGAGC3’;配列番号8および3’プライマー 5’AAGCCTTCCCTTTATACGCTCAAGC3’;配列番号9;サーモ サイエンティフィック社)、(2,2’,2’’−(10−(3−イソチオシアネートベンジル)−1,4,7,10−テトラアザシクロドデカン−1,4,7−トリイル)三酢酸;図8d)の非蛍光Eu3+イオンキャリアキレートで標識されたプローブA(5’AATCGTATCTCGGGTTAATG[AmC7];配列番号10;サーモ サイエンティフィック社)50nM、集光性アンテナ配位子(4−((4−イソチオシアネートフェニル)エチニル)ピリジン−2,6−ジカルボン酸)[U. Karhunen et al., Anal Chem, 82 (2010) 751]で標識されたプローブB(5’T(AmC2dT)GCATGATGCTTTATCAAA3’3’リン酸塩;配列番号11;サーモ サイエンティフィック社)50nM、30μM DTPA、400μM dNTPs、0.6μl PhireホットスタートDNAポリメラーゼ(Finnzymes社、フィンランド)Phire反応緩衝液(Finnzymes社)および各種量の合成一本鎖オリゴヌクレオチド鋳型を含む。熱サイクルは、98℃で2分の初期変性およびポリメラーゼ活性化ステップ、その後、98℃で15秒、60℃で20秒および72度で15秒を8サイクル;98℃で15秒、60℃で20秒、72℃で15秒、98℃で15秒、60℃で20秒、72℃で15秒、94℃で15秒および30℃で30秒を17サイクルから構成された。その熱サイクルは、PTC−200サーマルサイクラー(MJ リサーチ、USA)を用いて行われ、時間分解蛍光はサイクル9で開始して、340nm励起フィルター、615nm発光フィルター、400μs遅延時間および400μs測定時間を用い、1000測定サイクル計測することにより、Victor1420Multilabel Counter(パーキンエルマー ライフ サイエンス社、フィンランド)で30℃で測定した。各蛍光測定のために、PCRプレートは、PTC−200サーマルサイクラーからVictor1420Multilabel Counterへ一時的に移された。
当然のことながら、本発明の方法は、多種多様な態様の形態で組み込まれることができ、本明細書に開示されているのはそのうちの数例のみである。他の実施態様が存在し、本発明の精神から逸脱しないことは当業者には明らかである。したがって、記載された実施態様は、説明のためのものであり、制限的なものとして解釈されるべきものではない。
Claims (15)
- ランタニドイオンキャリアキレートおよび第1認識エレメントを含み、該ランタニドイオンキャリアキレートがランタニドイオンキャリア配位子とランタニドイオンを含む第1のグループ;アンテナ配位子および第2認識エレメントを含む第2のグループを用いる、検体を検出および/または定量するためのバイオアッセイ法であって、
該方法においては、
a)該ランタニドイオンキャリアキレートが、該バイオアッセイ法の条件において該ランタニドに充分強く結合し、結果として、該バイオアッセイ法の条件において、遊離のランタニドイオンが本質的には存在しない、すなわち1nmol/Lより少ない、好ましくは10pmol/Lより少ない遊離のランタニドイオンが存在する;または
b)該ランタニドイオンキャリアキレートが、該バイオアッセイ法の条件において該ランタニドに充分強く結合し、結果として、遊離のランタニドが本質的には存在しない、すなわち1nmol/Lより少ない、好ましくは10pmol/Lより少ない遊離のランタニドが存在し、かつ該ランタニドイオンを錯化する薬剤が少なくとも1pmol/Lの濃度でさらに用いられる;そして
該アンテナ配位子が該ランタニドイオンに弱く結合する、すなわち該アンテナ配位子は単座、二座、三座または四座のいずれかである;ならびに
該第1のグループの該第1認識エレメントおよび該第2のグループの該第2認識エレメントによる認識が、結果として、
i)キレート相補性、すなわち該ランタニドを担持する該ランタニドイオンキャリアキレートと該アンテナ配位子との相補性による混合ランタニドキレート錯体の形成、およびそれによる蛍光の増加;または
ii)キレート脱相補性、すなわち該ランタニドを担持する該ランタニドイオンキャリアキレートが該アンテナ配位子から分離され、それによる蛍光の減少
のいずれかを生じる
バイオアッセイ法。 - a)logKLnL1は少なくとも12、好ましくは18以上であって、KLnL1は、イオンキャリア配位子とランタニドイオンとの錯体の溶液中での安定度定数を意味する;または
b)該ランタニドイオンを錯化する薬剤をさらに用いる場合、
i)logKLnL2は少なくとも12であって、KLnL2は、イオンキャリア配位子とランタニドイオンとの錯体の溶液中での安定度定数を意味する;および
ii)logKLnL3は少なくとも8であって、KLnL3は、該ランタニドイオンを錯化する該錯化剤とランタニドイオンとの溶液中での安定度定数を意味する
を特徴とする請求項1記載のバイオアッセイ法。 - 前記イオンキャリアキレートが五座、六座、七座または八座であり、好ましくは六座、七座または八座であることを特徴とする請求項1または2記載のバイオアッセイ法。
- 前記イオンキャリアキレートのランタニドイオンは、プラセオジム(III)、ネオジム(III)、サマリウム(III)、ユーロピウム(III)、テルビウム(III)、ジスプロシウム(III)、ホルミウム(III)、エルビウム(III)、ツリウム(III)およびイッテルビウム(III)からなる群より選択されることを特徴とする請求項1〜3のいずれか1項に記載のバイオアッセイ法。
- 第1および第2認識エレメントが互いに独立してオリゴヌクレオチド、アプタマー、ペプチド、タンパク質、ハプテンおよびオリゴ糖からなる群より選択されることを特徴とする請求項1〜4のいずれか1項に記載のバイオアッセイ法。
- アンテナ配位子が四座、三座、二座または単座、好ましくは三座または二座であることを特徴とする請求項1〜5のいずれか1項に記載のバイオアッセイ法。
- logKLnL1が少なくとも20、好ましくは22以上であることを特徴とする請求項1〜6のいずれか1項に記載のバイオアッセイ法。
- ランタニドイオンを錯化する薬剤が用いられる請求項1〜7のいずれか1項に記載のバイオアッセイ法であって、該錯化剤がCDTA、EDTA、DOTA、DTPA、EGTA、HBED、HEDTA、NOTA、NTA、TETAおよびTTHAからなる群より選択されることを特徴とするバイオアッセイ法。
- ランタニドイオンを錯化する薬剤が用いられる請求項1〜8のいずれか1項に記載のバイオアッセイ法であって、該錯化剤が、アンテナ配位子よりも該ランタニドイオンの強力な結合剤、すなわちlogKLnL3>logKLnL4であって、KLnL4は溶液中での該アンテナ配位子と該ランタニドイオンとの錯体の安定度定数を意味し;そして好ましくはイオンキャリアキレートよりも弱いランタニドイオンの結合剤、すなわちlogKLnL3>logKLnL2であることを特徴とするバイオアッセイ法。
- 前記ランタニドイオンキャリア配位子がEDTA、DTPA、NOTAまたはDOTAから誘導されるか、または図7i、図7iiおよび図7iiiに示される構造a)〜p)から選択されることを特徴とする請求項1〜9のいずれか1項に記載のバイオアッセイ法。
- 前記アンテナ配位子が、図5i、図5ii、図5iiiおよび図5ivに示される構造a)〜z)からなる群より選択される集光構造を含むことを特徴とする請求項1〜10のいずれか1項に記載のバイオアッセイ法。
- 検体の認識が結果として蛍光の増加または減少を生じる請求項1〜11のいずれか1項に記載のバイオアッセイ法であって、該蛍光が400および1600nmの間の波長で測定されることを特徴とするバイオアッセイ法。
- 検出または定量される検体が、ストレプトアビジン、タンパク質、ハプテン、核酸配列、細胞、ウイルス、核酸増幅反応産物およびポリメラーゼ連鎖反応産物からなる群より選択されることを特徴とする請求項1〜12のいずれか1項に記載のバイオアッセイ法。
- 検体の認識が結果として蛍光の増加または減少を生じる請求項1〜13のいずれか1項に記載のバイオアッセイ法であって、該蛍光が長い蛍光寿命、すなわち寿命が>1μsであることを特徴とするバイオアッセイ法。
- 検体の認識が結果として蛍光の増加または減少を生じる請求項1〜14のいずれか1項に記載のバイオアッセイ法であって、該蛍光がアップコンバージョン蛍光、すなわち発光が励起よりも短い波長で検出される抗−ストークフォトルミネッセンスであることを特徴とするバイオアッセイ法。
