実験用マウスを、肺の調製前にイソフルラン/酸素混合物およびケタミン/ロンパン混合物(1.33:1)100μl/マウスで気管内処置した。麻酔後、静脈カテーテルをマウスに埋め込んだ。次に、肺の透過性亢進を誘発するため、肺に25μlの液体を微細シリンジで噴霧した。該液体は、0.9%生理食塩水溶液または250ngの毒素ニューモリシン、もしくは250ng/mlのニューモリシン/50μg/mlのペプチド配列番号1を含んでいた。

材料
ヒト肺上皮細胞のタイプH441はATTC社から得た。
細胞培養
上皮ナトリウムチャンネルの発現の確認
50μg/mlのペプチド配列番号3を加えると、ナトリウムチャンネルENaCの発現の実質的な増加はみられない。
ペプチド配列番号3とペプチド配列番号1の違いは、ペプチド配列番号1のアミノ酸Thr (6)、Glu (8)、およびGlu (11)がペプチド配列番号3ではAla (6)、Ala (8)、およびAla (11)に置き換わっていることである。
以下の動物試験群について、ウイルス肺感染症に対するペプチド配列番号1の効果を試験した:
グループ1. 陰性対照 (経鼻PBS)。
グループ2. 陽性対照 (経鼻で約2,000単位のインフルエンザAウイルスを感染)。
グループ3. 試験グループ(経鼻で約2,000単位のインフルエンザAウイルスを感染および10μgのペプチド配列番号1を気管内投与)。
各群、BALB/cマウス6匹を使用した。
処置第0日:
グループ1:PBSを経鼻投与。
グループ2:インフルエンザAウイルスでマウスを経鼻感染。
グループ3:インフルエンザAウイルスでマウスを経鼻感染およびペプチド配列番号1を投与。
グループ1:PBSを気管内投与。
グループ2:PBSを気管内投与。
グループ3:ペプチド配列番号1を気管内投与。
試験動物の体温、体重、および生存率を毎日観察。
材料と方法
管轄動物保護委員会の承認を得て、2頭のブタ(25kg)に全身麻酔下で界面活性剤枯渇(surfactant depletion)(4回気管支肺胞洗浄、各30ml/kg体重)により肺損傷を誘導した。次に、1 mg/kg体重のAP301 (ペプチド配列番号1)を気管内適用した。動物1 (1)には全用量の深部気管注射を行い、動物2 (2)には、同用量の噴霧を30分間かけて行った。次に、5時間の換気期間を設けた。動脈酸素分圧(paO2)を、予め確認した大動脈内リアルタイム測定プローブ(FOXY、Ocean Optics、USA)を用いて記録した。肺活量測定および血行動態を恒久的に記録し、PiCCO技術を用いた測定を半時間間隔で行った。

- 上皮細胞および内皮細胞の透過性亢進を予防および治療するための7〜17隣接アミノ酸からなる、ヘキサマーTX1EX2X3Eを含むペプチド(ここで、X1、X2、およびX3はあらゆる天然もしくは非天然のアミノ酸であり得る)であって、TNFレセプター結合活性を持たず、環化されているペプチド。
- 上皮細胞および内皮細胞の透過性亢進を予防および治療するための7〜17隣接アミノ酸からなる、ヘキサマーTPEGAEを含むペプチドであって、TNFレセプター結合活性を持たず、環化されているペプチド。
- 上皮細胞および内皮細胞の透過性亢進を予防および治療するための医薬を製造するための、
- QRETPEGAEAKPWY
- PKDTPEGAELKPWY
- CGQRETPEGAEAKPWYC、および
- CGPKDTPEGAELKPWYC
からなる群から選ばれる連続アミノ酸、および
その少なくとも7アミノ酸であってヘキサマーTPEGAEを含む断片
の配列からなる環化ペプチド。 - 肺炎、急性肺損傷、急性呼吸窮迫症候群 (ADRS)、または細菌またはウイルス性肺疾患、特に、Listeria monocytogenes、Streptococcus pneumoniae、SARSウイルス、RSV、またはインフルエンザウイルスによる感染を治療するための請求項1〜3のいずれかに記載のペプチド。
- アミノ酸配列CGQRETPEGAEAKPWYCを含み、C残基を介して環化することを特徴とする請求項1〜4のいずれかに記載のペプチド。
- 該C残基間のジスルフィド架橋を介して環化することを特徴とする請求項5記載のペプチド。
- 上皮細胞および内皮細胞を反応性酸素分子によって引き起こされる透過性亢進から保護するために用いることを特徴とする請求項1〜6のいずれかに記載のペプチド。
- 上皮細胞および内皮細胞を細菌毒素によって引き起こされる透過性亢進から保護するために用いることを特徴とする請求項1〜6のいずれかに記載のペプチド。
- ミオシン軽鎖のリン酸化を阻害するために用いることを特徴とする請求項1〜6のいずれかに記載のペプチド。
- プロテインキナーゼCの活性を阻害するために用いることを特徴とする請求項1〜6のいずれかに記載のペプチド。
- 上皮ナトリウムチャンネルの発現を増加するために用いることを特徴とする請求項1〜6のいずれかに記載のペプチド。
- 反応性酸素分子、細菌毒素、グラム陽性微生物、または肺ウイルス感染症によって引き起こされる透過性亢進を治療するために用いることを特徴とする請求項1〜6のいずれかに記載のペプチド。
- 請求項1〜12のいずれかに記載のペプチドおよび医薬担体を含む医薬組成物。
