用語「同種ペア」は、単一細胞の中に含まれる、あるいは単一細胞に由来する目的の非隣接核酸の原初のペアを表す。好ましい実施態様において、同種ペアは、ともに結合タンパク質可変ドメインをコードし、同一の細胞に由来する2つの可変領域をコードする配列を含む。このように、完全結合タンパク質、またはその安定的断片のいずれかとして発現された場合、それらは、この細胞から原始的に発現した結合タンパク質の結合親和性および特異性を保存する。同種ペアは、例えば、同一の細胞からの可変軽鎖をコードする配列と結合した抗体可変重鎖をコードする配列、あるいは同一の細胞からのβ鎖をコードする配列と結合したT細胞受容体α鎖をコードする配列でありうる。同種ペアのライブラリーは、このような同種ペアの収集物である。
本発明に従い連結してもよい目的のヌクレオチド配列は、その発現産物がタンパク質又はタンパク質の部分である異なるサブユニット又はドメインをコードする配列から選択することができる。特に、コードされるタンパク質又はその部分は、異種タンパク質、すなわち少なくとも2つの非同一サブユニットを含むタンパク質である。このようなタンパク質が属するいくつかのクラスは、例えば、酵素、インヒビター、構造タンパク質、毒素、チャネルタンパク質、Gタンパク質、受容体タンパク質、免疫グロブリンスーパーファミリータンパク質、輸送タンパク質などである。このような異種タンパク質をコードするヌクレオチド配列は、非隣接であり、これは、それらが、例えば、異なる遺伝子、あるいは異なるmRNA分子に由来することを意味する。しかしながら、本発明の文脈において使用される非隣接は、同一のタンパク質のドメインをコードするヌクレオチド配列をも意味してもよく、ここで、ドメインは、目的でないヌクレオチド配列によって分離される。
本発明は、単離単一細胞(それぞれの個別の細胞は、単一ウェル又は他の容器に位置する)、同系細胞の集団、又は単一容器に分けられなかった遺伝的に多様な細胞集団に由来するヌクレオチド配列の接続を可能にする。
本発明は、1より多いプライマーセット(例えば、可変領域をコードする配列を増幅するために必要な全てのプライマー)を同一の反応に含めることによって、2又はそれ以上の標的配列を同一の容器において同時に増幅する変形PCRを利用する。一般的に、この取り組みは、多重ポリメラーゼ鎖反応(多重PCR)として知られる。本発明に従い多重PCRによって増幅される標的配列は、例えば、増幅プロセスにきわめて密接に接近して、重複伸長PCRによって、連結される。特に、抗体可変領域をコードする配列の同種ペアは、このプロセスによって連結される。
本発明のプライマー混合物は、2つずつでプライマーセットを形成し、少なくとも2つの異なる目的の標的配列の増幅をすることができる少なくとも4つのプライマーを含む。プライマーセットは、遺伝子ファミリー変異体を増幅するように設計した1又は複数のプライマーペアを含む。このようなプライマーペア又はプライマーセットの2又はそれを超える混合物は、多重プライマーミックスを構成する。ニワトリにおける抗体多様性は、遺伝子変換を通して達成され、このプロセスによって、重鎖(HC)及び軽鎖(LC)可変領域の上流の偽遺伝子が、組換えによって単一VH及びVL遺伝子に挿入される配列のドナーとして機能する。これは、全ての可変領域が、原理的には、VHのための単一のプライマーペア、及びVLのための単一のプライマーペアによって増幅されることを意味する。好ましい実施態様において、単一のVH及びVL5’プライマーは、多重反応において1又は複数の3’定常領域プライマーとともに用いられ、他方、ネストPCR反応は、単一JHプライマー及び単一JLプライマーで実行される。好ましくは、多重プライマーミックスは、少なくとも、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19又は20のプライマーペアを含み、例えば、少なくとも、30、40、50、60、70、80、90、100、110、120、130、140又は150のプライマーペアを含む。特に、可変領域をコードする配列の増幅にとって、多重プライマーミックスのなかの個別のプライマーセットは、2を超えるプライマーペアを含んでもよい。好ましくは、個別のプライマーセットは、少なくとも、3、4、5、6、7、8、9、10、11、12、13、14、15、20、25、30、35、45、50、60、70、80、90、100、110、120、130、140、150、160、170、180、190、200、220、240、260、280又は300のプライマーを含む。好ましくは、多重プライマーミックスにおけるプライマーの全体数は、少なくとも、4、5、6、7、8、9、10、11、12、13、14、15、20、25、30、35、45、50、60、70、80、90、100、125、150又は200であり、最大で225、250、275、300、325、350、375又は400プライマーである。

2段階及び1段階手順の両方の多重重複伸長PCR段階のパラメーターは、さまざまなパラメーター(例えば、Henegariu, O. et al.1997.BioTechniques 23,504−511; Markoulatos,P.et al.2002.J.Clin.Lab.Anal.16,47−51を参照のこと)に最適化することができる。外側及び内側プライマーの割合は、このような反応において、重要性が低いけれども、一般的には、同一の最適化パラメーターが、多重RT−PCRに適用される。
a. プライマー濃度
重複伸長尾部を有するプライマー(例えば、VH及びVLプライマー)の濃度は、好ましくは、重複伸長尾部を持たない外側プライマー(例えば、JH及び軽鎖プライマー)の濃度より低い。
多重PCR反応は、DNAを弛緩させてテンプレート変性をより容易にするDMSO、グリセロール、ホルムアミド又はベタインのようなPCR添加物を使用することによって、顕著に改善することができる。
デオキシヌクレオシド三リン酸(dNTP)品質及び濃度は、多重重複伸長PCRにとって、重要である。最善のdNTP濃度は、それぞれのdNTP(dATP、dCTP、dGTP及びdTTP)につき200と400μMの間であり、これを超えると、増幅は、急激に阻害される。より低いdNTP濃度(それぞれのdNTPにつき100μM)は、PCR増幅を達成するのに十分である。dNTPストックは、解凍/冷凍サイクルに敏感である。このような3〜5のサイクルの後、多重PCRは、しばしば、十分に働かなくなる。このような問題を避けるため、dNTPの少量アリコートを作ることができ、−20℃で冷凍保存することができる。
一般的には、KClベースの緩衝液は、多重重複伸長PCRに十分である。しかしながら、(NH4)2SO4、MgSO4、Tris−Cl、あるいはこれらの組み合わせなどの他の成分をベースとする緩衝液も、多重重複伸長PCRで機能するように最適化されてもよい。より短い生成物の増幅に関与するプライマーペアが、より高い塩濃度(例えば、80〜100mM KCl)でより良く機能する一方で、より長い生成物の増幅に関与するプライマーペアは、より低い塩濃度で(例えば、20〜50mM KCl)より良く機能する。1Xの代わりに2Xへ緩衝液濃度を上昇させることにより、多重反応の効率が改善するかもしれない。
本発明は、Taqポリメラーゼにより例示される。その代わりに、例えば、Pfu、Phusion、Pwo、Tgo、Tth、Vent又はDeep−ventを含む、他のタイプの熱耐性DNAポリメラーゼを用いても良い。3’から5’エキソヌクレアーゼ活性を持たない又は持つポリメラーゼを、単独で、又はそれぞれを組み合わせてのいずれかで用いてもよい。
本発明による目的のヌクレオチド配列の接続は、免疫グロブリンの変領域をコードする連結されたヌクレオチド配列を含むヌクレオチド部分を創出する。さらに、このような連結された核酸配列のライブラリー、特に、ヒト定常領域(重鎖及び軽鎖)配列に連結した又は接合した非ヒト可変領域をコードする配列のライブラリー、又はヒト定常領域配列に連結された、トランスジェニックニワトリ又は他のトランスジェニック鳥に由来するヒト可変領域をコードする配列のライブラリーが、本発明の方法によって作製される。
本発明のさらなる実施態様において、連結された可変領域をコードする配列の同種ペアの前記ライブラリーは、本明細書に説明する段階を含む方法によって得ることができる。このライブラリーも、親ライブラリーと呼ばれる。
本発明の方法の1つを用いて、ドナーから単離された連結された可変領域をコードする配列のペアの親ライブラリーは、特に、コンビナトリアルライブラリーにおいて、いくつかは無関係である(すなわち、所望の標的への結合でない)多様な結合タンパク質を示すことが予想される。したがって、本発明は、特定の標的に対する結合特異性の多様なサブセットをコードするサブライブラリーの富化及びスクリーニングを包含する。
本発明のライブラリーは、目的の連結した核酸配列からコードされたタンパク質、特に、可変領域含有タンパク質又はその断片の発現及び産生に適したベクターに移行することができる。このようなベクターは、ベクター及びライブラリーの節において説明されており、例えば、選択の種の完全長抗体、Fab断片、Fv断片、scFv、膜結合又は可溶性TcR又はTcR断片の発現を提供する。
診断、処置及び予防における組換えモノクローナル抗体の使用が、よく知られている。本発明により産生される組換えモノクローナル及びポリクローナル抗体は、既存の技術により産生された抗体生成物と同様に用いることができるであろう。特に、有効成分としてポリクローナル組換え抗体を(ここで、特に、ポリクローナル組換え抗体は、可変領域をコードする配列の同種ペアを含む)、少なくとも1つの医薬的に許容される賦形剤と組み合わせて含む医薬組成物は、本発明の手段によって作製することができる。ポリクローナル組換え抗体組成物は、所定の疾患標的に特異的又は反応性であることができ、このようにして、組成物は、ヒト、家畜、ペットなどの哺乳動物における、腫瘍、感染症、炎症性疾患、アレルギー、ぜんそく及び他の呼吸器疾患、自己免疫疾患、免疫性機能不全、心血管疾患、中枢神経系疾患、代謝内分泌疾患、移植拒絶反応、不要な妊娠などの疾患の処置、改善、又は予防に用いることができる。
この実施例は、リンパ球特異的細胞表面マーカーの組み合わせを用いる蛍光活性化細胞分離(FACS)を用いる細胞分離及び蛍光色素接合抗体染色による、ニワトリまたはメンドリ(ここでは、今後、単にニワトリ(Gallus gallus;Isa Warren strain)と呼ぶ)からの抗体産生B細胞の単離のための異なるゲート及び分離戦略を実証する。Bu−1は、よく知られた抗体産生細胞への成熟の間にB細胞上に存在する特異的ニワトリB細胞表面抗原であり、形質細胞への分化の間に失われる(Rothwell et al.(1996)Vet.Immunology Immunopathology 55:225−34)。さらに、抗体分泌細胞が、細胞表面におけるIgYの存在に基づき検出された。IgYの細胞表面存在が形質細胞への分化の間に失われるという事実にもかかわらず、この抗体発現の直接マーカーは、哺乳動物IgG発現システムに観察されるように、IgYの膜濃度が単一細胞分離を可能とするという仮定に基づく分離戦略に含まれる(Wiberg et al.(2006)Biotechnol.Bioeng.94(2):396−405)。T細胞細胞は検出され、CD3抗原の存在によって、分離された集団から除去された。この研究において使用されたニワトリは、破傷風トキソイド(TT)抗原で免疫付与した。それゆえ、ビオチニル化破傷風トキソイドも、TT特異的細胞の上昇した集団を染色し選別するために、蛍光色素標識ストレプトアビジンとの組み合わせにおいて使用された。ELISpotアッセイによって、抗体産生B細胞および特異的抗TT抗体産生細胞の両方について、選別されたB細胞集団をアッセイした。細胞集団の由来として用いられた脾臓は、大半の分化B細胞を含み、そのため、高含有量の抗体分泌細胞を含んでいた(Mansikka et al.,1989,Scand.J.Immunol.29(3):325−331))。
6つの23週齢メスニワトリであるLohmann Brown Lite株を、完全フロインドアジュバント(CFA)中の0.5mg破傷風トキソイド(TT)の皮下投与し、第一次免疫付与の14、21および28日後に、不完全フロインドアジュバント中の0.5mgTTで繰り返し追加免疫をすることによって免疫付与した。免疫付与したニワトリからの脾臓を、最後の追加免疫付与の2、7および10日後に取り出し、脾細胞を直ちに、回収した。
ニワトリを安楽死させ、脾臓を直ちに回収した。脾臓を、一時的に1% ペニシリン/ストレプトマイシン(P/S)(Invitrogen,CA,US)を有する10mlの4℃RPMI1640(Invitrogen,CA,US)中に保存し、氷上に放置した。脾臓組織を50mlチューブ中の70μm細胞ろ過器(BD Falcon(商標)352350)に移した。ろ過器を通して細胞を柔らかくするために10ml 注射器プランジャーの背部を用い、手順の間、4℃の完全培地(10%ウシ胎仔血清(FCS)および1%P/Sを有するRPMI 1640)で一定間隔ろ過器をすすいだ。懸濁液中の細胞を、300xg、4℃、5分間での遠心分離によって、採取し、続いて、50ml 4℃ FACS緩衝液(リン酸緩衝生理食塩水(PBS)中の2%FCS)における懸濁によって洗浄し、前に説明したように遠心分離した。最後に、細胞を4℃のFACS緩衝液に希釈し、FACSに用い、あるいは冷凍培地(10%DMSO、90%ウシ胎仔血清)中に−140℃で保存する前に、50μmFACSフィルター(BD340603)に通した。
下のリストに示される50μlのそれぞれのネズミ抗ニワトリ抗体を、1ml 4℃FACS緩衝液中の1x108細胞に加え、暗室で、20分間、4℃でインキュベートし、第一次染色の後に、二度洗浄し、第二次染色の後に三度洗浄した。

この例は、IgY−特異的及びTT−特異的 ELISpotアッセイを用いることにより、実施例1における分離B細胞集団の間から抗体産生B細胞集団を同定することができることを実証する。
洗浄緩衝液 (1xPBS,0.05% Tween):

PBS中に希釈したいずれも10μg/mlの100μlの抗IgY抗体 (Abcam ab 6872)または破傷風トキソイドでPVDF底プレート(Multiscreen−HTS,Millipore,MSIP S45 10)を被覆し、4℃で一晩インキュベートした。陰性コントロールとして、PBSのみで被覆したウェルを用いた。プレートをPBSで3回洗浄し、続いて、200μlのブロッキング緩衝液で、4℃で少なくとも2時間ブロックした。そして、緩衝液を取り除き、50μl完全RPMIに取り替えた。
・まず、特異的な定常領域プライマーによって重鎖及び軽鎖cDNA合成が開始するRT反応を行う。
・次に、重複伸長によって結合したVH及びVLの形成を促進する相補的突出を備えたVH及びVL5’領域プライマーを用いて、多重PCR反応を実行する。3’プライマーは、重鎖及び軽鎖配列の定常領域に位置する。
・ヒトラムダ軽鎖及びIgG1重鎖定常領域の重複伸長による後の追加のための突出を備えたJH及びJLプライマーを用いて接合したVH及びVKのみを増幅するネストPCR反応を実行する。Symplex(商標)PCR生成物は、CDRのサイズに依存する約700のヌクレオチドからなる。
・ヒトIgG1及びラムダ定常領域は、重複伸長により付加される。
・最終生成物は、5’末端が5’末端に接合しリンカーで連結された、ヒトIgG1定常領域に結合したニワトリVH、及びヒトラムダ定常領域に結合したニワトリVLからなる。リンカー領域は、哺乳動物細胞プロモーターリーダー断片の挿入のための制限酵素サイトを含み、隣接サイトがクローニングを促進する。
・連結したキメラLC−HC断片が、ベクターバックボーンへクローニングされ、哺乳動物細胞プロモーターリーダー断片が挿入される。
・ プライマーhCHC−F及びhCHC−R(表4)を用いてPhusion(登録商標)ポリメラーゼ(Finnzymes)を用いて、抗体発現プラスミドからヒトIgG1定常重鎖領域cDNA断片(発現を改善するように最適化したコドン)を増幅した。hCHC−Fプライマーは、ネスト反応に用いられるプライマーCH−JHに相補的な5’部位を含む。プライマーhCHC−Rは、重複バンドのクローニングに用いられる隣接PacIサイトを導入する。約1000kbの定常領域断片を1%アガロースゲル上で精製した。
・ プライマーhL−F及びhL−R(表4)及び抗体発現プラスミドをテンプレートとして用いてヒトラムダ軽鎖定常領域断片を増幅した。hL−Fプライマーは、ネスト反応に用いられるプライマーCH−JLに相補的な5’部分を含む。hL−Rプライマーは、重複バンドのクローニング用いられる隣接NotIサイトを導入する。約350kb定常領域断片を、1%アガロースゲル上で精製した。
・ 精製VH−VL、ヒトラムダ及びヒトIgG1定常領域バンドを混合し(それぞれ、25:12.5:25ng)、重複伸長PCRを、プライマーhCHC−R及びhL−R(表4)を用いるPhusion(登録商標)ポリメラーゼを用いて実行した。(約2kbの重複バンドを有する)反応生成物を図11に示す。
・ 3つの抗体(図13において上から下へ1、2及び5番目)のVH領域は、1つのヌクレオチドの相異を除いて、同一であったが、他方、残りの2つは、非常に異なった。
・ 3つの抗体(図14において上から下へ1、2及び3番目)のVL領域は、2ヌクレオチド塩基を除き、大半の配列(フレームワーク、CDR1及びCDR2)において同一であり、これは、PCR導入変異のためでありえ、他方、1つのクローン(図14において上から1番目)は、他の2つとCDR3配列において異なり、このことは、遺伝子変換によるこの領域におけるさらなる分化を示す。残りのクローン(図14において上から下へ4および5番目)のVL領域は、VH領域と同様に、非常に異なった。

- 接続された可変領域をコードする配列を含む同種ペアのライブラリーを製造する方法であって、
a) 鳥類起源のドナーからのリンパ球含有細胞分画の提供、
b) 前記細胞分画から細胞を個別に複数の容器に分配することによる、単離単一細胞の集団の取得(ここで、少なくとも該細胞の亜集団は、免疫グロブリン遺伝子、及び所望により任意の鳥類B細胞マーカー抗原を発現する)、及び
c) 多重分子増幅手順において単離単一細胞又は同系細胞の集団由来のテンプレートを用いて目的のヌクレオチド配列を増幅し、そして増幅された目的のヌクレオチド配列の接続を達成することによる、前記単離単一細胞の集団に含まれる可変領域をコードする配列の増幅及びそれらの接続の達成

- 細胞の亜集団が、IgY+ CD3−、例えば、IgY+ CD3− Bu−1−である、請求項3に記載の方法。
- 増幅及び接続を実行する前に、単一細胞の集団における個別の単離単一細胞を同系細胞の集団に拡大する、請求項1〜4のいずれかに記載の方法。
- リンパ球含有細胞分画が、脾細胞、全血、骨髄、単核細胞、又は白血球、好ましくは、脾細胞又は骨髄、より好ましくは脾細胞を含む、請求項1〜5のいずれかに記載の方法。
- リンパ球含有細胞分画又はBリンパ球系列が、形質細胞、形質芽球又は記憶B細胞に富む、請求項1〜6のいずれかに記載の方法。
- 目的のヌクレオチド配列が、免疫グロブリン可変領域をコードする配列を含み、そして接続が、重鎖可変領域をコードする配列に結合した軽鎖可変領域をコードする配列の同種ペアを創出する、請求項1〜7のいずれか1項に記載の方法。
- 複数の目的の非隣接ヌクレオチド配列をランダムに接続する方法であって、
a)多重分子増幅手順において遺伝的に多様な細胞の集団由来のテンプレートを用いた、目的のヌクレオチド配列の増幅(ここで、前記遺伝的に多様な細胞は、鳥類起源のドナーからのリンパ球含有細胞分画に由来し、少なくとも該細胞の亜集団は、免疫グロブリン遺伝子、及び所望により任意の鳥類B細胞マーカー抗原を発現する)、及び
b)段階a)において増幅された目的のヌクレオチド配列の接続の達成
を含む方法。 - 細胞の集団が、次のいずれかによって特徴づけられる、請求項9に記載の方法。
・IgYの発現(IgY+)、
・IgYの発現、及びCD3陰性(IgY+ CD3−)、
・IgYの発現、Bu−1の非又は低発現、及びCD3陰性(IgY+ Bu−1− CD3−)、
・Bu−1及びIgYの発現(Bu−1+ IgY+)、
・Bu−1及びIgYの発現、並びにCD3陰性(Bu−1+ IgY+ CD3−)、
・Bu−1の発現、及び単球マーカーの非発現(Bu−1+、単球−)、
・Bu−1の発現、及びIgMの非又は低発現レベル(Bu−1+ IgM−)、又は
・Bu−1 及びBAFFの発現(Bu−1+ BAFF+)。 - 細胞の亜集団が、IgY+、好ましくは、IgY+ CD3−、例えば、IgY+ CD3− Bu−1−である、請求項10に記載の方法。
- 細胞の集団が、溶解している、請求項9〜11のいずれかに記載の方法。
- 目的のヌクレオチド配列が可変領域をコードする配列を含み、そして接続が可変領域をコードする配列のペアのコンビナトリアルライブラリーを作製する、請求項9〜12のいずれかに記載の方法。
- 目的のヌクレオチド配列が免疫グロブリン可変領域をコードする配列を含み、そして接続が軽鎖可変領域と重鎖可変領域をコードする配列のペアのコンビナトリアルライブラリーを作製する、請求項13に記載の方法。
- 多重分子増幅の前に、リンパ球含有細胞の集団が、検出可能なレベルのIgY、所望により、検出可能なレベルのIgY及びBu−1を発現する細胞を含むことを評価することをさらに含む、請求項1〜14のいずれかに記載の方法。
- 多重分子増幅の前に、IgYを発現する、所望により、IgY及びBu−1を発現するリンパ球集団のためにリンパ球含有細胞分画を富化することをさらに含む、請求項1〜15のいずれかに記載の方法。
- 多重分子増幅の前に、リンパ球含有集団から、免疫グロブリン遺伝子、好ましくは、IgY、所望により、IgY及びBu−1を発現する細胞を単離することをさらに含む、請求項1〜16のいずれかに記載の方法。
- 富化又は単離が、自動分離手順を含む、請求項1〜17のいずれかに記載の方法。
- 自動分離手順が、MACS又はFACSである、請求項18に記載の方法。
- 鳥類が、ニワトリである、請求項1〜19のいずれかに記載の方法。
- 鳥類が、アヒル、ガチョウ、ハト又は七面鳥である、請求項1〜19のいずれかに記載の方法。
- ニワトリが、トランスジェニックであり、ヒト免疫グロブリン配列を発現する、請求項20に記載の方法。
- 多重分子増幅手順が、多重RT−PCR増幅である、請求項1〜22のいずれかに記載の方法。
- 多重RT−PCR増幅が、多重PCR増幅の前に、別個の逆転写(RT)段階を含む2段階プロセスである、請求項23に記載の方法。

- 接続されたヌクレオチド配列又は同種ペアのライブラリーをベクターに挿入することをさらに含む、請求項1〜29のいずれかに記載の方法。
- ベクターが、クローニングベクター、シャトルベクター、ディスプレイベクター及び発現ベクターから選択される、請求項30に記載の方法。
- 接続されたヌクレオチド配列または同種ペアのライブラリーの個別のメンバーが、軽鎖可変領域をコードする配列と結合した免疫グロブリン重鎖可変領域をコードする配列を含み、前記配列が、1又は複数の免疫グロブリン定常ドメインまたはその断片をコードする配列を含むベクターにインフレームで挿入されている、請求項30または31に記載の方法。
- 所望の標的特異性を有する結合タンパク質をコードする連結された可変領域配列の同種ペアのサブセットを選択し、それにより、可変領域をコードする配列の標的特異的同種ペアのライブラリーを作製することによってサブライブラリーを製造することをさらに含む、請求項30〜32のいずれか1項に記載の方法。
- 同種ペア又は可変領域をコードする配列の標的特異的同種ペアのライブラリーを哺乳動物発現ベクターに転写することをさらに含む、請求項32又は33に記載の方法。
- 哺乳動物発現ベクターが、ヒト免疫グロブリンクラスIgA1、IgA2、IgD、IgE、IgG1、IgG2、IgG3、IgG4、IgM、カッパ軽鎖及びラムダ軽鎖から選ばれる1又は複数の定常領域ドメインをコードする、請求項34に記載の方法。
- a)接続されたヌクレオチド配列の部分をコードするベクターの宿主細胞への導入
b)発現に適合した条件下での前記宿主細胞の培養、及び
c)前記宿主細胞に挿入されたベクターから発現したタンパク質生成物の取得
の段階をさらに含む、請求項30〜35のいずれか1項に記載の方法。 - タンパク質生成物が、重鎖可変領域に結合した軽鎖可変領域の同種ペアを含む抗体である、請求項36に記載の方法。
- 大多数のウェルにおいて、
・鳥類ドナーからのリンパ球含有細胞分画に由来する1つの細胞(ここで、前記細胞は、IgY及び/又はBu−1抗原を含む免疫グロブリン遺伝子を発現する)、及び、
・mRNAの逆転写を実行するために、及び重鎖及び軽鎖可変コード領域を増幅するために必要な緩衝液及び試薬、
を含む多重ウェルプレート。 - ヒト定常領域及び非ヒト可変領域を有するキメラ抗体をコードするベクターを作製する方法であって、
a) 鳥類起源のドナーからのリンパ球含有細胞分画の提供;
b) 前記細胞分画から細胞を個別に複数の容器に分配することによる、単離単一細胞の集団の取得;
c) 多重分子増幅手順において単離単一細胞又は同系細胞の集団由来のテンプレートを用いて単離単一細胞の集団に含まれる可変領域をコードする核酸を増幅し、そして増幅された重鎖及び軽鎖の可変領域をコードする核酸の接続を達成することによる、前記核酸の増幅及びそれらの接続の達成;
d) ヒト定常領域への増幅された可変領域の接続の達成;及び
e) ベクターへの取得された核酸の挿入
を含む方法。 - ドナーが、ヒト抗体可変重鎖及び軽鎖に由来するか又はそれと顕著な類似性を有する免疫グロブリンを産生することができるヒト免疫グロブリン配列を有するトランスジェニックのニワトリである、請求項39に記載の方法。
- 多重分子増幅手順が、多重RT−PCR増幅である、請求項30または40に記載の方法。
- 多重RT−PCR増幅が、多重PCR増幅の前に、別個の逆転写(RT)段階を含む2段階プロセスである、請求項41に記載の方法。
- 多重RT−PCR増幅が、逆転写(RT)及び多重PCR増幅の両方を実行するのに必要な全ての成分を単一の容器に最初に添加することを含む単一段階において実行される、請求項41に記載の方法。
- 目的のヌクレオチド配列の接続が、多重分子増幅と同一の容器において実行される、請求項39〜43のいずれかに記載の方法。
- 目的のヌクレオチド配列の接続が、多重重複伸長プライマーミックスを利用する多重PCR増幅に関連して達成される、請求項41〜44のいずれかに記載の方法。
- 目的のヌクレオチド配列の接続が、ライゲーションにより達成される、請求項39〜44のいずれかに記載の方法。
- 接続された目的の核酸配列の増幅に適したプライマーミックスを利用する付加的分子増幅が実行される、請求項39〜46のいずれかに記載の方法。
- PCR生成物が、発現ベクターに挿入される、請求項41に記載の方法。
- 二重プロモーターカセットが発現コンストラクトに挿入され、二重プロモーターカセットが重鎖及び軽鎖の同時発現を指揮することができ、好ましくは、二重プロモーターカセットが双方向性である、請求項48に記載の方法。
- 二重プロモーターカセットがさらに二重シグナルペプチドをコードする核酸配列を含む、請求項49に記載の方法。
- 発現ベクターが、ヒト定常軽鎖をコードする配列若しくはその断片及び/又はヒト定常重鎖をコードする配列若しくはその断片を含むバックボーンを含む、請求項48に記載の方法。
- 順にニワトリVH鎖、リンカー、ニワトリVL鎖、及びヒト定常軽鎖を含むコンストラクトを増幅することができるプライマーセットとともに、可変軽鎖への接続を提供することができるオーバーラップを有するヒト定常軽鎖をコードするポリヌクレオチド又はその断片をPCR混合物に添加するさらなる増幅段階を含む、請求項39〜51のいずれか1項に記載の方法。
- 順にヒト定常重鎖、ニワトリVH鎖、リンカー、及びニワトリVL鎖を含むコンストラクトを増幅することができるプライマーセットとともに、可変重鎖への接続を提供することができるオーバーラップを有するヒト定常重鎖をコードするポリヌクレオチド又はその断片をPCR混合物に添加するさらなる増幅段階を含む、請求項39〜51のいずれか1項に記載の方法。
- それぞれのキメラ抗体が、ニワトリ免疫グロブリン可変領域をコードする配列及びヒト免疫グロブリン重鎖及び軽鎖定常領域からなる、キメラ抗体をコードするベクターのライブラリー。
- ベクターが請求項1〜37又は39〜53のいずれかに記載の方法により得られる、請求項54に記載のライブラリー。
- ニワトリ免疫グロブリン可変領域をコードする配列が、ヒト抗体可変重鎖及び軽鎖に由来するか又はそれと顕著な類似性を有する免疫グロブリンを産生することができるヒト免疫グロブリン配列を有するトランスジェニックニワトリに由来する、請求項54に記載のライブラリー。
- 軽鎖定常領域がカッパ又はラムダ定常領域である、請求項54に記載のライブラリー。
- ベクターが発現ベクターである、請求項54に記載のライブラリー。
- 可変領域が可変重鎖及び軽鎖の同種ペアである、請求項54に記載のライブラリー。

- 鳥起源免疫グロブリン可変領域をコードする配列のライブラリーを製造する方法であって、
a)鳥類起源のドナーからのリンパ球含有細胞分画の提供、
b)前記細胞分画から細胞を個別に複数の容器に分配することによる、単離単一細胞の集団の取得(ここで、少なくとも該細胞の亜集団は、免疫グロブリン遺伝子、例えばIgY、及び所望により少なくとも1つの鳥類B細胞マーカー抗原を発現する)、及び
c)多重分子増幅手順において単離単一細胞又は同系細胞の集団由来のテンプレートを用いて目的のヌクレオチド配列を増幅することによる、前記単離単一細胞の集団に含まれる可変領域をコードする配列の増幅
を含む方法。 - 同種ペアのライブラリーを得るように、さらに、重鎖及び軽鎖可変領域をコードする配列の接続を達成することを含む、請求項63に記載の方法。
