Abstract

本発明は、ヒト及び獣医薬における使用に適した高純度なアルファウイルス・レプリコン粒子(ARP)又はウイルスの調製品の製造方法であって、高収量、GMP互換性、商業的に実行可能な方法を提供する。本発明は、生弱毒化アルファウイルス・ワクチン及び同じ物を含む免疫原組成物の製造にも適用可能であり、ここで、米国特許第5,643,576号において記載されるように、弱毒化されたアルファウイルスは、ワクチンにおいて発現させるために、異種遺伝子を持つこともあるし又は持たないこともある。本発明の方法は、以下のステップ

A. 種々の細胞型におけるキャップなしヘルパーRNAの使用
293T細胞

HIV-gagをコードするレプリコン・ベクターRNA及びVEEキャプシド及び糖タンパク質遺伝子をコードする2個のヘルパーRNAを使用する実験において、in vitro転写反応を、プロメガ・コーポレーションから市販されるキット(Madison, WI; カタログ番号P1300)を使用して行った。調製されたキャップ有りヘルパー及びレプリコンRNAに対して、キャップ付き-Gを転写反応に加え、そしてrGTP濃度を、別のrNTPに比較して1.4mM分低減した。キャップなしヘルパーRNAを調製するために、4個のrNTPを等モル濃度で使用した。RNA精製は、プロメガSVトータルRNAアイソレーション・システム(Promega SV total RNA isolation System)(Promega Corp., Madison, WI;カタログ番号Z3100、シリカ・レジン)を使用し、そしてRNAをRNAse-フリー水中に溶出することにより達成した。例えばサイズ排除クロマトグラフィーなどのRNA精製の別の方法もまた適切であるが、キーパラメーターは、かなり純粋で、濃縮されたRNA溶液、つまり少なくとも0.5μg/μl、好ましくは少なくとも2〜3μg/μlを作り出す方法を使用することである。分光光度計を使用して精製RNAの濃度を測定し、一方、既知のRNA量の一定量とともに1μlの量をホルムアルデヒドゲル中で泳動することにより、未精製RNAの濃度を見積もった。
レプリコン及びヘルパーRNAを、実施例1に記載するように調製した。電気穿孔では、30μgの各レプリコン及びヘルパーRNAを、RNaseフリーのマイクロ遠心チューブ中で混合した。コントロールのチューブは、精製済み又は未精製のRNAのみを有し、一方残りのチューブは、5×転写反応緩衝液[400mM・HEPES-KOH(pH7.5);120mM・MgCl2;10mMスペルミジン;200mM・DTT]、NotI制限酵素、T7酵素混合液(T7・RNA依存性RNAポリメラーゼ、RNase阻害剤、ピロホスファターゼ)、またはこれら3種の組み合わせをさらに入れられた。各転写反応要素の終濃度は、未精製RNAの等体積における終濃度とおおよそ同じであった。1.2×107ベロ細胞を各マイクロ遠心チューブに加え、そして混合液を0.4cmギャップ電気穿孔キュベット中に移した。細胞を580Vかつ25μFde4回パルスをかけ、そして室温で10分間回復させた。電気穿孔された細胞を10%ウシ胎児血清及び抗体を伴う25ml・HyQMEMを含むT75フラスコ中に蒔いた。電気穿孔効率の分析のため、一定量を96ウェル・プレート中に蒔いた。
電気穿孔するためのある細胞系列、例えば293T及びCEF細胞を電気穿孔する最初の実験において、電気穿孔混合液中の細胞濃度を増加し、一方入力核酸を同じ量に維持することにより、高いARP収量が得られるということが観測された。
A. 実験1
30μgレプリコンRNA(癌腫瘍抗原をコードする)、26.8μgVEEキャプシドRNAヘルパー、及び55.6μgVEE糖タンパク質RNAヘルパーを伴うPBS中で、0.4cmギャップ・キュベット中に、ベロ細胞を指示された濃度に再懸濁した。電気穿孔に続いて、1.4×105細胞/cm2の成長領域の密度で、1cm2あたり約0.3mlの成長培地で、必要に応じ細胞を1以上のフラスコ中に蒔いた。電気穿孔後24時間において、培地を各フラスコから回収し、そして細胞単層を約10mlの無血清培地で洗浄した。この洗浄溶液を各フラスコから回収された培地中に加えた。各フラスコにおいて、10分間0.5M・NaClを使用することにより塩洗浄を行った。塩洗浄液を回収し、そして別々に分析した。指示された場合、2回目の塩洗浄を行い、そして別々に分析した。ARP収量を一回のキュベット電気穿孔から得られた粒子の全数として、表3中に載せた。
ベロ細胞を指示された細胞密度でInVitrus無血清培地中で懸濁した。30μgのVEE・HIV−gagレプリコンRNA、及びCMVプロモーターの下でVEE構造タンパク質の全てを発現する100〜150μgのVEE・DNAヘルパーを使用して、細胞を電気穿孔した。DNAヘルパーを精製し、そして電気穿孔での使用前に少なくとも5mg/mlに濃縮した。
コーニング・セル・キューブ(Corning Cell Cube)(Corning, Inc., Acton, MA)を使用して、大量の細胞を育てた。1cmのギャップ電気穿孔キュベットにおいて5×108ベロ細胞を使用して、一回の電気穿孔を行った。バイオ-ラド電気穿孔器(Gene Pulser II, BioRad Laboratories, Inc., Hercules, CA; カタログ番号165-2105)上で1150V、25μFで4回のパルスを使用して、各150μgのVEEレプリコン(HIV-gapをコードする)、VEEキャプシド・ヘルパー、及びVEE糖タンパク質ヘルパーを細胞中へと電気穿孔した。電気穿孔後、細胞を100mlのオプティプロSFM(Invitrogen, Carlsbad, CA; カタログ番号12309019)を含む5T-300フラスコ中へと蒔いた。電気穿孔後24時間に、培地を核フラスコから回収し、そしてタイターを調べるために混合した。各フラスコにおいて20mlの1.0M・NaClを使用して塩洗浄を各フラスコにおいて行い、そして塩洗浄液をタイターを調べるために混合した。培地中のARPの全収量は1×108i.u.であり;塩洗浄液中のARPの全収量は、2.2×1011i.u.であった。
A. 電気穿孔後の成長培地
ARP生成の以前のレポートにおいて、電気穿孔後に10%FBSを含む培地中に細胞を蒔いた。ARP収量に関する血清の効果を試験するため、種々の濃度の血清を含む異なる培地中に細胞を蒔いた。電気穿孔後の最適な培地は、血清の使用を必要とせず、かつ低タンパク質濃度を有する。低濃度血清を有する培地並びに3種の無血清培地は、高い細胞密度における電気穿孔での使用を試験された。オプティプロ無血清培地(Gibco Cell Culture/Invitrogen, San Diego, CA); VP-SFM, VP無血清培地(Gibco Cell Culture/Invitrogen, San Diego, CA); Ex-Cell 505(JRH Biosciences, Lenexa, KS); 及びInVitrus化学的に規定された細胞培養培地(InVitrus chemically defined cell culture medium)(Cell Culture Technologies GmbH, Zurich, CH; カタログ番号IVT)。これらの培地の全てはpH7.0〜7.4に緩衝されたpHを有する。ベロ細胞をヘルパー及びレプリコンRNAで電気穿孔した。電気穿孔混合液を異なる培地を含む8個のT−75フラスコに分配した。細胞を37℃で18〜24時間培養した。ARP収量の分析により、EMEM中の血清レベルが、ARP収量の有意な低下をもたらさずに10%から1%へと減少されうるということが明らかにされた。また、オプティプロ培地は、EMEM+10%FBSと同等のARP収量を与えた。オプティプロは、ヒト及び/又は動物タンパク質を含まず、いくつかの異なるARPsについて、EMEM+10%FBSを使用して得られる収量と同等のARP収量を与える。電気穿孔後の成長及びARP生成に有用である無血清培地は、いくつかの市販の培地を含む。
ARP収量に関する電気穿孔後の培地のpHの効果を試験した。ベロ細胞をペトリ皿電極装置中で電気穿孔し、そして一定分量を関心のpHに調節された完全成長培地中に蒔かれた。結果を図4に示した。

電気穿孔された細胞は、ポリスルホン・ファイバー、例えばファイバーセルHFカートリッジ(Fibercell Systems, Inc.; Frederick, MO;カタログ番号:4300-C2011)に素早く接着することが発見された。電気穿孔された細胞をホローファイバーに配置することにより、ホロー・ファイバーにおける細胞により生成されるARPsを回収する利点を与える。ARPsが溶出される終体積は、最低40ml(109細胞まで保持することができる小さいカートリッジの使用)〜100ml(5×1010細胞まで保持することができる大きなカートリッジの使用)でありうる。
1×108ベロ細胞を使用して、0.4cmギャップ・キュベット中で電気穿孔を行った。各電気穿孔は、各30μgのHIV-gag・VEEレプリコンRNA及びVEE糖タンパク質ヘルパーを含んだ。VEEキャプシド・ヘルパーの濃度は、指示されるように変化させた。以下の条件:580V、25μFで4回のパルス(パルスの長さ:約0.8ms)を使用してバイオ-ラド装置上で電気穿孔を行った。指示したように、細胞をT175フラスコ中の50ml・EMEM+FBS又はオプティプロに蒔いた。電気穿孔後24時間において、各フラスコからの培地を収集し、そしてろ過した。5mlの1M・NaClを各フラスコに加え、そして接着細胞と5分間インキュベーションさせた。塩洗浄溶液を次に取り出し、そして以前に回収された培地に使用されたのと同じフィルターを通してろ過された。
典型的に、RNAヘルパーに使用される量及び条件に比較して、多くのDNA及びいくらか異なる電気穿孔条件は、DNAヘルパーをベロ細胞中へ効率的に電気穿孔をするために必要とされる。例えば、DNAヘルパーは、250V、950μF又は250V800μFで2〜3回のパルス(30m秒)を使用して電気穿孔される一方、RNAヘルパーは、580V25μF(4回のパルス)を使用して電気穿孔される。
電気穿孔効率に関する細胞周期G2/M期へ細胞を同期化する効果が試験された。細胞を植え付けた2時間後に、DMSO中の1μg/mlのアフィジコリン(Sigma Chemical Co., St. Louis MO)で20時間処理し、4時間回復させ、そして電気穿孔用に収集した(Golzio et al., (2002) Biochem. Biophys. Acta 1563:23-28)。(レプリコンRNAと共に)ヘルパーDNA電気穿孔を本明細書中に記載されるように行った。処理過程を通して、細胞を成長及び細胞形態学について観察した。細胞の各群において、DMSO処理のコントロールを含むフラスコは、アフィジコリン/DMSO処理フラスコと共に行われた。試験又はコントロール・フラスコにおける処理の間、細胞の一般的な健康に関して、マイナスの効果は無かった。同期化プロトコルを開始する前の2時間のインキュベーションのうちにコンフルエントな状態を得ることは有利であるということが測定された。アフィジコリン処理の開始前に、90〜100%で細胞を敷き詰めることが最良である。何故なら更なる細胞分裂は、この処理により妨げられるからである。ベロ細胞を使用するとき、4.0×105細胞/cm2の密度は高すぎるということが分かり;細胞密度1.1×104細胞/cm2は、90%の密集度を二時間のうちにもたらす。
電気穿孔された細胞によりARPが生成されたのち、細胞を細胞洗浄溶液で過度に洗浄して、ARPsを収集する前に余分な物質を取り除いた。細胞洗浄液の選択は、この前洗浄ステップの間に放出されるARPsの数に影響する。
A. 塩組成
5×108ベロ細胞、150μgづつのHIV-gag・VEEレプリコンRNA、VEEキャプシド・ヘルパーRNA、及びVEE糖タンパク質ヘルパーRNAを使用して、10mmギャップ・キュベット中で電気穿孔を行った。1150V、25μFで4回のパルスを使用して電気穿孔を行った。電気穿孔された細胞を40個のT-75フラスコに均等に蒔いた。1のフラスコを16時間で収集し、新たな培地で再び栄養を与え、そして電気穿孔後24時間で収集した。他のフラスコを24時間で収集した。培地を各フラスコから取り除き、そして5mlの指示された塩溶液を各フラスコに加え、そして5分間インキュベーションした。塩洗浄溶液を次に取り出し、そしてタイターを調べた。これらの異なる洗浄溶液を使用して得られた結果を図5に示した。細胞から成長培地へと放出されたARPsの量(タイターにより計測されるとき)は、pH7.0〜8.0の範囲内で穏やかに影響をうけた(図3を参照のこと)。
ARPは、種々のレジンを使用してアフィニティークロマトグラフィーにより精製されうる。一般的に、溶出条件の範囲は、選んだレジンに左右され、ARPを約6以下のpHにかけないように気をつけながら使用されうる。レプリコンRNAによりコードされる抗原は、通常ARPs中に含まれ;該抗原の性質は、精製の間のARPsの行動に影響しうるということが知られている。当業者は、そうした影響の認識の仕方、そしてそれによるARP精製方法の改変の仕方を知っている。
塩洗浄溶液中のARPを、5mMリン酸ナトリウム、pH7.4で、塩化ナトリウムの含有量が0.12M以下になるまで希釈した。溶液を次に、ヘパリン・セファロース・ファスト・フロー・レジン(Amersham, Pharmacia Biotech, Inc, Piscataway, NJ; カタログ番号17-0998-01)又はヘパリン・ハイパーD・M(Biosepra, Mariboro, MA)レジンを含むカラム上に、線速度100cm/hr以下でロードした。例えば、VEE3014ARPsは、約0.3M、pH7.4の塩化ナトリウム濃度で溶出した。VEE3014ARPsを回収し、そして直接希釈又はダイアフィルトレーションにより処方した。該方法により精製されるARPの典型的収量は、70%である。ステップ又は直線的塩勾配は、ARPsを溶出するために使用されうる。
塩洗浄溶液中のARPsを、5mMリン酸ナトリウム、pH7.4で、塩化ナトリウム含有量が0.25M以下になるまで希釈した。この溶液を次に、塩化ナトリウム含有量増加勾配における硫酸セルフィン・レジン(Millipore)を含むカラム中へとロードした。pH7.4、約0.7M塩化ナトリウム濃度で、ARPsは溶出した。ARPsを回収し、そして直接希釈又はダイアフィルトレーションにより処方した。この方法により精製されるARPsの典型的な収量は、85%である。
塩洗浄溶液中のARPsを、5M・NaCl/20mMリン酸ナトリウムpH7.4で、終NaCl濃度3Mまで希釈する。ARPsを、線速度100cm/hrでトヨピール・フェニル(Toyopearl phenyl 650-Mレジンを含むカラム上にロードし、そして3M〜0M塩化ナトリウムの線勾配で溶出する。この方法によるARPs回収率は、約77%である。
ARPsを直接、又は適切な希釈の後に、アニオン交換レジン、例えばトヨピール・スーパーQ(Toyopearl super Q)またはアマシャムQセファロース(Amersham Q Sepharose)、又はアニオン交換膜、例えばムスタングQ(Mustang Q)(Pall Trincor, Exton, PA)上にロードした。Qクロマトグラフィー物質は、該物質を通過する物質上の酸性基に結合する4級アミンに頼る。ロード条件は、ARPsの多くの型への結合又は流出をもたらすように操作される。ここで結合性質は、アルファウイルス・レプリコン・ベクターによりコードされ、そしてARPsが生成される細胞内で発現される関心のタンパク質の発現により影響される。特定の場合では、アニオン交換レジンは、一回の生成ステップで十分であり、血清タンパク質、宿主細胞タンパク質、及びDNAの低減をもたらす。規定のレジン領域機能上のARP結合性質は、選ばれたレジン、ARP種、及びARP標本を含む溶液のpH及び塩濃度の関数である。
アルファバックス(AlphaVax)WCBp146細胞を、PBS中に1.6×108細胞/mlへと再懸濁した。電気穿孔1〜4用に、レプリコンRNA(ヘルペスウイルスgDコード配列を有する)をG-キャップ付加し、そしてサイズ排除クロマトグラフィーにより精製した。電気穿孔1〜3用に、ヘルパーRNAのキャップを外し、そしてLiCl沈殿により精製した。電気穿孔4用に、ヘルパーRNAのキャップを外し、そしてサイズ排除クロマトグラフィーにより精製した。700μlの細胞(1.1×108細胞)を、RNAと混合し、そして0.4cmキュベット中で電気穿孔した(BioRad電気穿孔器、580V、25μFd、4回のパルス)。細胞を、各々100mlオプティプロ培地を含む2個のT300cm2フラスコ中に蒔いた。電気穿孔後約18時間で、ARPsを30mlの0.5M・NaCl/10mM・NaPO4中において収集し、そして0.2μmフィルターを通してろ過した。
Claims (30)
- アルファウイルス・レプリコン粒子(ARPs)の製造方法であって、該方法が以下のステップ:
(a) アルファウイルス・レプリコン核酸を宿主細胞中に導入し、該レプリコン核酸は少なくともウイルス・パッケージング・シグナル及び該アルファウイルス・レプリコン核酸中で発現できる少なくとも1の異種性コード配列を含み、ここで改変宿主細胞を作り出すために該宿主細胞が少なくとも1のヘルパー機能を含み;
(b) 上記少なくとも1のヘルパー機能の発現を許容し、上記アルファウイルス・レプリコン核酸の複製及び該アルファウイルス・レプリコン核酸のパッケージングを許容する条件下で上記改変宿主細胞を培養して、ARPsを形成し;
(c) ステップ(b)の後に上記改変宿主細胞を、0.2M〜5Mのイオン強度を有する水溶液と接触して、上記ARPsを該水溶液中に放出させて、ARPを含む溶液を作り出し;
(d) ステップ(c)のARPを含む溶液からARPsを回収するステップ
を含む前記方法。 - 1mlあたり5×107〜5×108のステップ(a)に記載の宿主細胞における前記少なくとも1のヘルパー機能が、該宿主細胞のゲノム中に安定して組み込まれた核酸配列によりコードされる、請求項1に記載の方法。
- 前記宿主細胞中の前記少なくとも1のヘルパー機能が、前記アルファウイルス・レプリコン核酸を結合できるキャプシドタンパク質をコードする少なくとも1のヘルパー核酸、及び少なくとも1のアルファウイルス糖タンパク質に基いて導入され、ここで該アルファウイルス糖タンパク質が、該アルファウイルス・レプリコン核酸及び該キャプシドタンパク質と会合し、ここで該少なくとも1のヘルパー核酸分子が該アルファウイルス・レプリコン核酸と共に該宿主細胞中へと導入される、請求項1に記載の方法。
- 前記少なくとも1のヘルパー機能が、少なくとも2個のヘルパー核酸分子によりコードされ、ここで該2個のヘルパー核酸分子の各々が、少なくとも1のウイルス・ヘルパー機能をコードする、請求項1に記載の方法。
- 前記イオン強度が0.5M〜5Mである、請求項1に記載の方法。
- 前記少なくとも1のヘルパー機能が、DNA分子中にコードされる、請求項1に記載の方法。
- 前記アルファウイルス・レプリコン核酸が、電気穿孔により前記宿主細胞中に導入される、請求項1に記載の方法。
- 前記宿主細胞が、1mlあたり5×107〜5×108の濃度で電気穿孔混合液中に存在する、請求項8に記載の方法。
- 前記宿主細胞が、1mlあたり5×107〜1.5×108の濃度で電気穿孔混合液中に存在する、請求項9に記載の方法。
- 請求項1に記載のステップ(c)の前に、細胞洗浄ステップをさらに含む、請求項1に記載の方法。

- アルファウイルス・レプリコン粒子の製造方法であって、アルファウイルス・レプリコン・ベクター及び1以上のヘルパー核酸分子を、電気穿孔を介してアルファウイルス-許容状態細胞へと導入することを含み、ここで、電気穿孔の間の培養培地中の該アルファウイルス許容状態細胞の濃度が、5×107〜5×108細胞/mlであり、かつ電気穿孔前に該細胞に加えられた該アルファウイルスRNAレプリコン・ベクターの濃度が、約35μg/mlである、前記方法。
- 前記電気穿孔が、電気穿孔チャンバー内で行われ、ここで電極間のギャップが0.4〜1.0cmである、請求項11に記載の方法。
- 前記ヘルパー機能が、全てのアルファウイルス構造タンパク質をコードするシングルDNAヘルパー分子内にコードされる、請求項11に記載の方法。
- 前記DNAヘルパー分子の濃度が、少なくとも100μg/mlである、請求項15に記載の方法。
- 前記アルファウイルス-許容状態細胞が、ベロ細胞である、請求項1〜18のいずれか1項に記載の方法。

- 前記アルファウイルスが、弱毒化アルファウイルスである、請求項1〜17のいずれか1項に記載の方法。
- 前記アルファウイルスが、ベネズエラ・ウマ脳炎ウイルス(VEE)である、請求項17に記載の方法。
- 前記塩洗浄ステップにおいて前記塩が、NaCl、KCl、MgCl2、CaCl2、NH4Cl、(NH4)2SO4、酢酸アンモニウム、及び炭酸水素アンモニウムである、請求項1〜21のいずれか1項に記載の方法。
- アルファウイルス・レプリコン粒子(ARPs)の製造方法であって、該方法が以下のステップ:
(a) アルファウイルス・レプリコン核酸を宿主細胞中へ導入し、該レプリコン核酸が、少なくともウイルス・パッケージング・シグナルと該アルファウイルス・レプリコン核酸中に発現する少なくとも1の異種性コード配列を含み、ここで改変宿主細胞を作り出すために該宿主細胞が少なくとも1のヘルパー機能を含み、ここで上記レプリコン核酸が、1mlあたり5×107〜5×108細胞濃度で、宿主細胞を電気穿孔することにより該宿主細胞中に導入され;
(b) 上記少なくとも1のヘルパー機能の発現を許容し、上記アルファウイルス・レプリコン核酸の複製及び該アルファウイルス・レプリコン核酸のパッケージングを許容する条件下で上記改変宿主細胞を培養して、ARPsを形成し;
(c) ステップ(b)の後に上記改変宿主細胞を、0.2M〜5Mのイオン強度を有する水溶液と接触して、上記ARPsを該水溶液中に放出させて、ARPを含む溶液を作り出し;
(d) ステップ(c)のARPを含む溶液からARPsを回収するステップ
を含む前記方法。 - 前記宿主細胞が、1mlあたり5×107〜1.5×108の濃度で、電気穿孔混合液中に存在する、請求項9に記載の方法。
- 前記アルファウイルスが、ベネズエラ・ウマ脳炎ウイルス(VEE)である、請求項17に記載の方法。
- 前記ヘルパー核酸が、キャップなしRNA分子である、請求項26に記載の方法。
- 請求項1〜29のいずれか1項に記載の方法により製造されるアルファウイルス・レプリコン粒子調製品。
